To assess the applicability of latex cell coatings as an ‘off‐the‐shelf’ biocatalyst, the effect of osmoprotectants, temperature, humidity and O2 on preservation of H2 production in Rhodopseudomonas palustris coatings was evaluated. Immediately following latex coating coalescence (24 h) and for up to 2 weeks of dry storage, rehydrated coatings containing different osmoprotectants displayed similar rates of H2 production. Beyond 2 weeks of storage, sorbitol‐treated coatings lost all H2 production activity, whereas considerable H2 production was still detected in sucrose‐ and trehalose‐stabilized coatings. The relative humidity level at which the coatings were stored had a significant impact on the recovery and subsequent rates of H2 production. After 4 weeks storage under air at 60% humidity, coatings produced only trace amounts of H2 (0–0.1% headspace accumulation), whereas those stored at < 5% humidity retained 27–53% of their H2 production activity after 8 weeks of storage. When stored in argon at < 5% humidity and room temperature, R. palustris coatings retained full H2 production activity for 3 months, implicating oxidative damage as a key factor limiting coating storage. Overall, the results demonstrate that biocatalytic latex coatings are an attractive cell immobilization platform for preservation of bioactivity in the dry state.  相似文献

Sulfide utilization by purple nonsulfur bacteria   总被引:1,自引:0,他引:1
Summary The purple nonsulfur bacteria Rhodospirillum rubrum SMG 107, Rhodopseudomonas capsulata SMG 155, Rps. sphaeroides SMG 158 and Rps. palustris SMG 124 were tested for a possible utilization of sulfide. The first three strains were found to oxidize sulfide to extracellular elemental sulfur only, whereas Rps. palustris SMG 124 converted sulfide into sulfate without intermediate accumulation of elemental sulfur. Growth ceased at lower sulfide concentrations than usually found with purple sulfur bacteria. In consequence of the low sulfide tolerance information on the specific growth rates obtainable with sulfide as photosynthetic electron donor could not be provided by cultivation in batch cultures. Sulfide-limited chemostat cultures of Rps. capsulata SMG 155 showed that the maximum specific growth rate was close to 0.14 h-1 (doubling time 5 h). Sulfide was converted into extracellular elemental sulfur at all dilution rates tested. The maximum specific growth rate of Rps. palustris SMG 124 was found to be much lower (less than 0.03 h-1). Sulfate was the only product of the conversion of sulfide. These data show that at least some purple nonsulfur bacteria may play a role in the dissimilatory sulfur cycle in nature. Taxonomic implications of our results are discussed.Abbreviation SMG Sammlung für Mikroorganismen, Göttingen  相似文献

This study describes the relationships between dinitrogen (N2) fixation, dihydrogen (H2) production, and electron transport associated with photosynthesis and respiration in the marine cyanobacterium Trichodesmium erythraeum Ehrenb. strain IMS101. The ratio of H2 produced:N2 fixed (H2:N2) was controlled by the light intensity and by the light spectral composition and was affected by the growth irradiance level. For Trichodesmium cells grown at 50 μmol photons · m?2 · s?1, the rate of N2 fixation, as measured by acetylene reduction, saturated at light intensities of 200 μmol photons · m?2 · s?1. In contrast, net H2 production continued to increase with light levels up to 1,000 μmol photons · m?2 · s?1. The H2:N2 ratios increased monotonically with irradiance, and the variable fluorescence measured using a fast repetition rate fluorometer (FRRF) revealed that this increase was accompanied by a progressive reduction of the plastoquinone (PQ) pool. Additions of 2,5‐dibromo‐3‐methyl‐6‐isopropyl‐p‐benzoquinone (DBMIB), an inhibitor of electron transport from PQ pool to PSI, diminished both N2 fixation and net H2 production, while the H2:N2 ratio increased with increasing level of PQ pool reduction. In the presence of 3‐(3,4‐dichlorophenyl)‐1,1‐dimethylurea (DCMU), nitrogenase activity declined but could be prolonged by increasing the light intensity and by removing the oxygen supply. These results on the coupling of N2 fixation and H2 cycling in Trichodesmium indicate how light intensity and light spectral quality of the open ocean can influence the H2:N2 ratio and modulate net H2 production.

Studies on the photoautotrophic growth of new isolated nonsulfur purple bacteria at the expense of molecular hydrogen
Summary By incubating samples of mud in a mineral salt medium containing 0.1% NH4Cl and 0.01% yeast extract under an atmosphere of 90% H2 and 10% CO2 in the light, eight strains of nonsulfur purple bacteria capable of photoautotrophic growth at the expense of H2-CO2 were enriched and isolated. The bacteria were characterized by their microscopical features and their ability to use various organic compounds for growth. Three strains of Rps. capsulata need thiamine as a growth factor and two strains of R. rubrum biotin. The Rps. gelatinosa strain is dependent on thiamine and nicotinic acid. One of two Rps. palustris strains grows slowly in a medium completely free of growth factors, the other strain is dependent on the presence of 0.01% yeast extract. The growth of all strains is markedly stimulated by small amounts (0.01%) of amino acids or yeast extract. The three Rps. capsulata strains differ significantly from the other strains with regards to their fast photoautotrophic growth (doubling time: 17–23 h in a mineral salt medium with thiamine, 10.5–11 h in a medium with 0.01% yeast extract). The hydrogenase activities of all strains are strongly dependent on the culture conditions. The lowest activities are obtained after photoheterotrophic growth in a medium with NH4Cl as the nitrogen source, moderate activities after photoheterotrophic growth in a medium with glutamate as the nitrogen source and maximum activities after photoautotrophic growth at the expense of H2-CO2. Maximum rates of CO2-fixation are also obtained after photoautotrophic growth.

In laboratory experiments, the antifouling (AF) properties of zinc oxide (ZnO) nanorod coatings were investigated using the marine bacterium Acinetobacter sp. AZ4C, larvae of the bryozoan Bugula neritina and the microalga Tetraselmis sp. ZnO nanorod coatings were fabricated on microscope glass substrata by a simple hydrothermal technique using two different molar concentrations (5 and 10?mM) of zinc precursors. These coatings were tested for 5?h under artificial sunlight (1060?W?m?2 or 530?W?m?2) and in the dark (no irradiation). In the presence of light, both the ZnO nanorod coatings significantly reduced the density of Acinetobacter sp. AZ4C and Tetraselmis sp. in comparison to the control (microscope glass substratum without a ZnO coating). High mortality and low settlement of B. neritina larvae was observed on ZnO nanorod coatings subjected to light irradiation. In darkness, neither mortality nor enhanced settlement of larvae was observed. Larvae of B. neritina were not affected by Zn2+ ions. The AF effect of the ZnO nanorod coatings was thus attributed to the reactive oxygen species (ROS) produced by photocatalysis. It was concluded that ZnO nanorod coatings effectively prevented marine micro and macrofouling in static conditions.  相似文献

Although sea‐ice represents a harsh physicochemical environment with steep gradients in temperature, light, and salinity, diverse microbial communities are present within the ice matrix. We describe here the photosynthetic responses of sea‐ice microalgae to varying irradiances. Rapid light curves (RLCs) were generated using pulse amplitude fluorometry and used to derive photosynthetic yield (ΦPSII), photosynthetic efficiency (α), and the irradiance (Ek) at which relative electron transport rate (rETR) saturates. Surface brine algae from near the surface and bottom‐ice algae were exposed to a range of irradiances from 7 to 262 μmol photons · m?2 · s?1. In surface brine algae, ΦPSII and α remained constant at all irradiances, and rETRmax peaked at 151 μmol photons · m?2 · s?1, indicating these algae are well acclimated to the irradiances to which they are normally exposed. In contrast, ΦPSII, α, and rETRmax in bottom‐ice algae reduced when exposed to irradiances >26 μmol photons · m?2 · s?1, indicating a high degree of shade acclimation. In addition, the previous light history had no significant effect on the photosynthetic capacity of bottom‐ice algae whether cells were gradually exposed to target irradiances over a 12 h period or were exposed immediately (light shocked). These findings indicate that bottom‐ice algae are photoinhibited in a dose‐dependent manner, while surface brine algae tolerate higher irradiances. Our study shows that sea‐ice algae are able to adjust to changes in irradiance rapidly, and this ability to acclimate may facilitate survival and subsequent long‐term acclimation to the postmelt light regime of the Southern Ocean.  相似文献

A computerized oxygen electrode Astern was used to make rapid and accurate measurements of photosynthetic light and dissolved inorganic carbon (DIC) response cures with a macroalga. Ulva rotundata Blid. was grown in an outdoor, continuous flow system in seawater under sunlight or 9% of sunlight at Beaufort, North Carolina. The light compensation points in the shade- and sun-grown plants, measured in seawater, were at photon flux densities (PFDs) of 16 and 27 μmol. Photons·m?2·s?1, respectively but the quantum yield of O2 evolution was not significantly different. Rates of photosynthesis in seawater per unit area of thallus under saturating light and rates of dark respiration were about 1.5-fold higher in sun- than in shade-grown plants. The concentration of DIC in seawater (approximately 2 mM) limited photosynthesis at absorbed PFDs above 60–70 μmol photons·m?2·s?1 Addition of 20 mM inorganic carbon had no effect on quantum yield but caused about a 1.5-fold increase in the light-saturated photosynthetic rate in both shade- and sun-grown Ulva. The effect of DIC supplementation was greatest in plants grown in October and least in plants grown in June. The light- and DIC-saturated rate of photosynthesis in seawater was similar to the maximum rate obtained by exposing Ulva to 10% CO2, in the gas phase. The carbon isotope values (δ13C, reflecting the 13C/12C ratio compared to a standard) of Ulva grown in the same seawater supply were dependent on light and agitation. Samples from Beaufort Inlet were more negative (δ13C value, ?20.03‰) than those grown in bright light with agitation (δ13C value, ?17.78‰ outdoors; ?17.23‰ indoors), which may indicate DIC supply limited carbon uptake in seawater.  相似文献

The possible protective role of endogenous isoprene against oxidative stress caused by singlet oxygen (1O2) was studied in the isoprene‐emitting plant Phragmites australis. Leaves emitting isoprene and leaves in which isoprene synthesis was inhibited by fosmidomycin were exposed to increasing concentrations of 1O2 generated by Rose Bengal (RB) sensitizer at different light intensities. In isoprene‐emitting leaves, photosynthesis and H2O2 and malonyldialdehyde (MDA) contents were not affected by low to moderate 1O2 concentrations generated at light intensities of 800 and 1240 µmol m?2 s?1, but symptoms of damage and reactive oxygen accumulation started to be observed when high levels of 1O2 were generated by very high light intensity (1810 µmol m?2 s?1). A dramatic decrease in photosynthetic performance and an increase in H2O2 and MDA levels were measured in isoprene‐inhibited RB‐fed leaves, but photosynthesis was not significantly inhibited in leaves in which the isoprene leaf pool was reconstituted by fumigating exogenous isoprene. The inhibition of photosynthesis in isoprene‐inhibited leaves was linearly associated with the light intensity and with the consequently formed 1O2. Hence, physiological levels of endogenous isoprene may supply protection against 1O2. The protection mechanisms may involve a direct reaction of isoprene with 1O2. Moreover, as it is a small lipophilic molecule, it may assist hydrophobic interactions in membranes, resulting in their stabilization. The isoprene‐conjugated double bond structure may also quench 1O2 by facilitating energy transfer and heat dissipation. This action is typical of other isoprenoids, but we speculate that isoprene may provide a more dynamic protection mechanism as it is synthesized promptly when high light intensity produces 1O2.  相似文献

In this work, plasmonic Au/SnO2/g‐C3N4 (Au/SO/CN) nanocomposites have been successfully synthesized and applied in the H2 evolution as photocatalysts, which exhibit superior photocatalytic activities and favorable stability without any cocatalyst under visible‐light irradiation. The amount‐optimized 2Au/6SO/CN nanocomposite capable of producing approximately 770 μmol g?1 h?1 H2 gas under λ > 400 nm light illumination far surpasses the H2 gas output of SO/CN (130 μmol g?1), Au/CN (112 μmol g?1 h?1), and CN (11 μmol g?1 h?1) as a contrast. In addition, the photocatalytic activity of 2Au/6SO/CN maintains unchanged for 5 runs in 5 h. The enhanced photoactivity for H2 evolution is attributed to the prominently promoted photogenerated charge separation via the excited electron transfer from plasmonic Au (≈520 nm) and CN (470 nm > λ > 400 nm) to SO, as indicated by the surface photovoltage spectra, photoelectrochemical IV curves, electrochemical impedance spectra, examination of formed hydroxyl radicals, and photocurrent action spectra. Moreover, the Kelvin probe test indicates that the newly aligned conduction band of SO in the fabricated 2Au/6SO/CN is indispensable to assist developing a proper energy platform for the photocatalytic H2 evolution. This work distinctly provides a feasible strategy to synthesize highly efficient plasmonic‐assisted CN‐based photocatalysts utilized for solar fuel production.  相似文献

Rhodopseudomonas palustris, a purple non-sulfur bacterium, was recently found throughout the water column in Lake Kinneret. It was demonstrated to be of a versatile nature, growing under both aerobic and anaerobic conditions at different light intensities. A comparison of C-acetate uptake byR. palustris andChlorobium phaeobacterioides, a green sulfur bacterium, showed that, under identical growth conditions, C-acetate assimilation byR. palustris was greater. Furthermore, C-acetate uptake forR. palustris was greater than C−CO2 uptake at all light intensities. Depending on the prevailing conditions, acetate can be used byR. palustris as both an electron donor and carbon source. Malate synthase was used as an indicator of activity of the glyoxylic acid cycle. It was found that enzyme activity was higher (i.e., acetate was used mainly as a carbon source) under anaerobic conditions, in the dark, or in the absence of HCO 3 . Acetate was used preferably as an electron donor under photosynthetic microaerophillic conditions.  相似文献

2D materials are of particular interest in light‐to‐heat conversion, yet challenges remain in developing a facile method to suppress their light reflection. Herein, inspired by the black scales of Bitis rhinoceros, a generalized approach via sequential thermal actuations to construct biomimetic 2D‐material nanocoatings, including Ti3C2Tx MXene, reduced graphene oxide (rGO), and molybdenum disulfide (MoS2) is designed. The hierarchical MXene nanocoatings result in broadband light absorption (up to 93.2%), theoretically validated by optical modeling and simulations, and realize improved light‐to‐heat performance (equilibrium temperature of 65.4 °C under one‐sun illumination). With efficient light‐to‐heat conversion, the bioinspired MXene nanocoatings are next incorporated into solar steam‐generation devices and stretchable solar/electric dual‐heaters. The MXene steam‐generation devices require much lower solar‐thermal material loading (0.32 mg cm?2) and still guarantee high steam‐generation performance (1.33 kg m?2 h?1) compared with other state‐of‐the‐art devices. Additionally, the mechanically deformed MXene structures enable the fabrication of stretchable and wearable heaters dual‐powered by sunlight and electricity, which are reversibly stretched and heated above 100 °C. This simple fabrication process with effective utilization of active materials promises its practical application value for multiple solar–thermal technologies.  相似文献

Simultaneous aerobic treatment of COD, phosphate, nitrate and H2S in a synthetic sewage wastewater was carried out using porous ceramic immobilized photosynthetic bacteria, Rhodobacter sphaeroidesS, Rb. sphaeroidesNR-3 and Rhodopseudomonas palustris. In the batch treatment, effective simultaneous removal of COD (89%), phosphate (77%), nitrate (99%) and H2S (99.8%) was observed after 48 h. In semi-continuous treatments with dilution rates of 0.17 to 0.75 day–1under aerobic conditions, simultaneous removal of these four components was also observed after about one month.  相似文献

Environmental controls on carbon dynamics operate at a range of interacting scales from the leaf to landscape. The key questions of this study addressed the influence of water and nitrogen (N) availability on Pinus palustris (Mill.) physiology and primary productivity across leaf and canopy scales, linking the soil‐plant‐atmosphere (SPA) model to leaf and stand‐scale flux and leaf trait/canopy data. We present previously unreported ecophysiological parameters (e.g. Vcmax and Jmax) for P. palustris and the first modelled estimates of its annual gross primary productivity (GPP) across xeric and mesic sites and under extreme drought. Annual mesic site P. palustris GPP was ~23% greater than at the xeric site. However, at the leaf level, xeric trees had higher net photosynthetic rates, and water and light use efficiency. At the canopy scale, GPP was limited by light interception (canopy level), but co‐limited by nitrogen and water at the leaf level. Contrary to expectations, the impacts of an intense growing season drought were greater at the mesic site. Modelling indicated a 10% greater decrease in mesic GPP compared with the xeric site. Xeric P. palustris trees exhibited drought‐tolerant behaviour that contrasted with mesic trees' drought‐avoidance behaviour.  相似文献

The susceptibility to photoinhibition of tree species from three different successional stages were examined using chlorophyll fluorescence and gas exchange techniques. The three deciduous broadleaf tree species were Betula platyphylla var. japonica, pioneer and early successional, Quercus mongolica, intermediate shade‐tolerant and mid‐successional, and Acer mono, shade‐tolerant and late successional. Tree seedlings were raised under three light regimes: full sunlight (open), 10% full sun, and 5% full sun. Susceptibility to photoinhibition was assessed on the basis of the recovery kinetics of the ratio of vaviable to maximum fluorescence (Fv/Fm) of detached leaf discs exposed to about 2000 μmol m?1 s?1 photon flux density (PFD) for 2 h under controlled conditions (25 to 28 °C, fully hydrated). Differences in susceptibility to photodamage among species were not significant in the open and 10% full sun treatments. But in 5% full sun, B. platyphylla sustained a significantly greater photodamage than other species, probably associated with having the lowest photosynthetic capacity indicated by light‐saturated photosynthetic rate (B. platyphylla, 9·87, 5·85 and 2·82; Q. mongolica, 8·05, 6·28 and 4·41; A. mono, 7·93, 6·11 and 5·08 μmol CO2 m?1 s?1for open, 10% and 5% full sun, respectively). To simulate a gap formation and assess its complex effects including high temperature and water stress in addition to strong light on the susceptibility to photoinhibition, we examined photoinhibition in the field by means of monitoring ΔF/Fm on the first day of transfer to natural daylight. Compared with ΔF/Fm in AM, the lower ΔF/Fm in PM responding to lower PFD following high PFD around noon indicated that photoinhibition occurred in plants grown in 10 and 5% full sun. The diurnal changes of ΔF/Fm showed that Q. mongolica grown in 5% full sun was less susceptible to photoinhibition than A. mono although they showed little differences both in photosynthetic capacity in intact leaves and susceptibility to photoinhibition based on leaf disc measurements. These results suggest that shade‐grown Q. mongolica had a higher tolerance for additional stresses such as high temperature and water stress in the field, possibly due to their lower plasticity in leaf anatomy to low light environment.  相似文献

The distribution of photosynthetic bacteria in Lake Kinneret and their contribution to primary productivity was investigated. Both Chlorobium phaeobacteroides and Rhodopseudomonas palustris were detected from mid-June through October in 1987 and reached a maximum of 7 × 106 cells/ml in the metalimnion by mid-August. Chlorobium density was greater towards the end of the bloom period, when the thermocline was exposed to higher sulfide concentrations and lower light intensities. Rhodopseudomonas peaked earlier in the season, when less sulphide was present and light irradiation was higher. This was the first time that R. palustris was observed in Lake Kinneret; in addition to its presence at the thermocline, it was found also at the epilimnion and hypolimmon. The contribution of photosynthetic bacteria to the total primary productivity of the lake was small (∼1%). Low but perceptible rates of N2 fixation in situ were also demonstrated by photosynthetic bacteria present in the metalimnion.  相似文献

Crop leaves are subject to continually changing light levels in the field. Photosynthetic efficiency of a crop canopy and productivity will depend significantly on how quickly a leaf can acclimate to a change. One measure of speed of response is the rate of photosynthesis increase toward its steady state on transition from low to high light. This rate was measured for seven genotypes of soybean [Glycine max (L.) Merr.]. After 10 min of illumination, cultivar ‘UA4805’ (UA) had achieved a leaf photosynthetic rate (Pn) of 23.2 μmol · m?2 · s?1, close to its steady‐state rate, while the slowest cultivar ‘Tachinagaha’ (Tc) had only reached 13.0 μmol · m?2 · s?1 and was still many minutes from obtaining steady state. This difference was further investigated by examining induction at a range of carbon dioxide concentrations. Applying a biochemical model of limitations to photosynthesis to the responses of Pn to intercellular CO2 concentration (Ci), it was found that the speed of apparent in vivo activation of ribulose‐1:5‐bisphosphate carboxylase/oxygenase (Rubisco) was responsible for this difference. Sequence analysis of the Rubisco activase gene revealed single nucleotide polymorphisms that could relate to this difference. The results show a potential route for selection of cultivars with increased photosynthetic efficiency in fluctuating light.  相似文献

The growth and photosynthesis of Alexandrium tamarense (Lebour) Balech in different nutrient conditions were investigated. Low nitrate level (0.0882 mmol/L) resulted in the highest average growth rate from day 0 to day 10 (4.58 × 102 cells mL?1 d?1), but the lowest cell yield (5420 cells mL?1) in three nitrate level cultures. High nitrate‐grown cells showed lower levels of chlorophyll a‐specific and cell‐specific light‐saturated photosynthetic rate (Pmchl a and Pmcell), dark respiration rate (Rdchla and Rdcell) and chlorophyll a‐specific apparent photosynthetic efficiency (αchla) than was seen for low nitrate‐grown cells; whereas the cells became light saturated at higher irradiance at low nitrate condition. When cultures at low nitrate were supplemented with nitrate at 0.7938 mmol/L in late exponential growth phase, or with nitrate at 0.7938 mmol/L and phosphate at 0.072 mmol/L in stationary growth phase, the cell yield was drastically enhanced, a 7–9 times increase compared with non‐supplemented control culture, achieving 43 540 cells mL?1 and 52 300 cells mL?1, respectively; however, supplementation with nitrate in the stationary growth phase or with nitrate and phosphate in the late exponential growth phase increased the cell yield by no more than 2 times. The results suggested that continuous low level of nitrate with sufficient supply of phosphate may facilitate the growth of A. tamarense.  相似文献

We studied the growth and photosynthetic characteristics of a toxic (CS506) and a nontoxic strain (CS509) of the bloom‐forming cyanobacterium Cylindrospermopsis raciborskii grown under identical experimental conditions. When exposed to light‐saturating growth conditions (100 μmol photons · m?2 · s?1), values for maximal photosynthetic capacity (Pmax) and maximum quantum yield (Fv/Fm) indicated that both strains had an equal ability to process captured photons and deliver them to PSII reaction centers. However, CS506 grew faster than CS509. This was consistent with its higher light requirement for saturation of photosynthesis (Ik). Greater shade tolerance of CS509 was indicated by its higher ability to harvest light (α), lower photosynthetic light compensation point (Ic), and higher chlorophyll a to biovolume ratio. Strain‐specific differences were found in relation to non‐photochemical quenching, effective absorption cross‐sectional area of PSIIα‐centers (σPSIIα), and the antenna connectivity parameter of PSIIα (JconPSIIα). These findings highlighted differences in the transfer of excitation from phycobilisome/PSII to PSI, on the dependence on different pigments for light harvesting and on the functioning of the PSII reaction centers between the two strains. The results of this study showed that both performance and composition of the photosynthetic apparatus are different between these strains, though with only two strains examined we cannot attribute the performance of strain 506 to its ability to produce cylindrospermopsins. The emphasis on a strain‐specific light adaptation/acclimation is crucial to our understanding of how different light conditions (both quantity and quality) can trigger the occurrence of different C. raciborskii strains and control their competition and/or dominance in natural ecosystems.  相似文献
