This study examines the oviposition response of a specialist weevil (Mogulones larvatus) to patches of its host, the noxious weed Paterson"s curse/salvation Jane (Echium plantagineum). We simultaneously examined the effect of patch size and plant density (and their interaction), on the recorded oviposition
patterns. Our results show that oviposition first occurred on the largest patches with the highest number of plants. However,
there was no significant effect of patch size or number of plants per patch at the end of the experiment. At this time the
level of attack per plant was negatively correlated with plant density. This negative effect of density on the level of oviposition
was not mediated by a reduction in plant size at higher densities. The pattern observed may indicate a risk-spreading strategy
by females.

As one of large amphibian group, there are a total of 45 species of Amolops in the world. However, the antimicrobial peptides (AMPs) existing in this genus has not been extensively studied. In this study, cDNAs encoding five novel AMP precursors were cloned by screening the skin-derived cDNA library of Amolops ricketti, a frog species that exists in southern and western parts of China. Protein sequence analysis led to the identification of five deduced peptides, three belonging to the brevinin-1 family and two belonging to the brevinin-2 family of amphibian AMPs. Thus, they were named as brevinin-1RTa (FLPLLAGVVANFLPQIICKIARKC), brevinin-1RTb (FLGSLLGLVGKVVPTLFCKISKKC), brevinin-1RTc (FLGSLLGLVGKIVPTLICKISKKC), brevinin-2RTa (GLMSTLKDFGKTAAKEIAQSLLSTASCKLAKTC), and brevinin-2RTb (GILDTLKEFGKTAAKGIAQSLLSTASCKLAKTC), respectively. The purification of brevinin-1RTa, brevinin-1RTb, and brevinin-2RTb was carried out by RP-HPLC, and confirmed by the LC-MS/MS-based proteomics approach. All of the peptides displayed different antimicrobial potency against a variety of microorganisms. In addition, brevinin-2RTa and brevinin-2RTb were found to have relatively low hemolytic activity (>400μg/ml) against mammalian red blood cells in vitro, which could potentially be as candidates for developing novel anti-infection agents. 相似文献

In order to identify sequence-tagged sites (STSs) appropriate for recombinant inbred lines (RILs) of barley cultivars ‘Azumamugi’ × ‘Kanto
Nakate Gold’, a total of 43 STS primer pairs were generated on the basis of the terminal sequences of barley restriction fragment
length polymorphism (RFLP) clones. Forty one of the 43 primer pairs amplified PCR products in Azumamugi, Kanto Nakate Gold,
or both. Of these, two showed a length polymorphism and two showed the presence or absence of polymorphism between the parents.
PCR products of the remaining 37 primers were digested with 46 restriction endonucleases, and polymorphisms were detected
for 15 primers. A 383.6-cM linkage map of RILs of Azumamugi×Kanto Nakate Gold was constructed from the 19 polymorphic STS
primer pairs (20 loci) developed in this study, 45 previously developed STS primer pairs (47 loci), and two morphological
loci. Linkage analysis and analysis of wheat-barley chromosome addition lines showed that with three exceptions, the chromosome
locations of the STS markers were identical with those of the RFLP markers.

Summary Selection of sites for oviposition when the risk of predation by green frogs was variable studied in tandems of the damselfly Pyrrhosoma nymphula. In the absence of predators the number of tandems landing increased with the size of the oviposition site and with the number of pairs already present. Pairs approaching an oviposition site landed promptly and preferred locations near other ovipositing pairs. Pairs which had landed near others left the landing site and flew to another water plant after a shorter period than those landing alone. They stayed in groups for only 16.5% of the total duration of stay at the oviposition site. With predators present fewer tandems landed. There was no relationship between the landing site of solitary tandems and the locations of the perching frogs. Pairs landing in groups were disturbed by frogs after a shorter period than pairs landing alone. Damselfly aggregation did not affect the predation success of the frogs, but the predation risk to individual tandem pairs was reduced. 相似文献

In Idotea baltica, a marine isopod that lives and feeds on the brown alga Fucus vesiculosus, microhabitat choice differs between sexes so that males are found more often than females on the light-coloured and exposed apical parts of the alga. We investigated how the requirements of avoiding visual predators and feeding were related to microhabitat choice in relation to diurnal and life-cycle stage in males and females. Faced with a choice between an apical and a basal piece of the alga, females spent more time than males on the basal piece, but this difference was not due to food choice. Faced with a choice between a dark, concealing and a light, exposing background, the preference for a dark background was stronger at day than at night, and stronger in females than in males. This suggests that a sex difference in the importance of avoiding visual predators can explain the sex difference in microhabitat choice. Further, the preference for a dark background and night feeding both increased with age, suggesting that feeding is increasingly subordinated to the need to avoid visual predators. Our experiment found no effect of the presence of the opposite sex on microhabitat choice. Our results support the hypothesis that the sexes trade off feeding against predation risk differently, presumably because growth is more important to males than to females, which have more to gain by protection and therefore spend more time on the lower parts of the alga. In insects, habitat selection should be most distinctly effected by females during oviposition, and this choice should directly affect the performance of the less mobile offspring stages. To date, this behaviour has been poorly investigated in carabid beetles. This study examines sexual differences in microhabitat selection as well as egg-laying behaviour and its consequences for offspring in the stenoecious beetle Carabus clatratus. Laboratory experiments with adults and different offspring stages were undertaken to test the impact of soil moisture and substrate type on microhabitat selection and immature fitness. Only females changed their substrate selection behaviour after copulation, revealing a distinct preference for wet peat for oviposition. Egg mortality was less than 7% under both wet and dry soil conditions. In contrast to other investigations, egg mortality in C. clatratus was therefore of minor importance. Larval mortality was also low, but mortality of third-instar larvae and the pupal stage increased strongly under dry conditions. Furthermore, beetles that developed under dry conditions showed a significantly higher proportion of exoskeleton deformations, and immature adults were smaller when reared under low moisture conditions. The importance of a strong oviposition site selection for wet peat to ensure sufficient moisture conditions for offspring in the stenoecious species C. clatratus is discussed. The use of zinc(II) to probe iron binding and oxidation by the ferritin (EcFtnA) of Escherichia coli

The ferritin of Escherichia coli (EcFtnA) is similar to human H-chain ferritin (HuHF) in having 24 subunits, each containing a dinuclear site at which two
iron atoms can be oxidised (the diiron centre). In EcFtnA, unlike HuHF, fluorescence quenching of Trp122, located near site
A of the dinuclear centre, can be used to monitor metal binding (this tryptophan is absent from HuHF). Metal binding also
perturbs the UV absorbance spectrum of Trp122 and that of Tyr24 (a conserved residue near site B of the dinuclear centre).
Using UV-difference spectroscopy and fluorescence quenching it is shown that Fe(II) and Zn(II) bind at the same sites, A and
B. Sequential stopped-flow studies of Fe(II) binding and oxidation also show that Zn(II) is an effective competitor of Fe(II)
binding and an inhibitor of its oxidation.

A genetic map covering 615 cM in 12 linkage groups was assembled based on 92 RFLP and AFLP markers segregating in a population
of 107 doubled haploid lines (DH lines) of Brassica oleracea. The DH-line population was obtained through microspore culture from the of two homozygous parents: DH-line Bi derived from the cabbage landrace Bindsachsener, and DH-line Gr from broccoli cv ‘Greenia’.
Sixty-five percent of the loci, and in some cases complete linkage groups, displayed distorted segregation ratios, a frequency
much higher than that observed in populations of the same species. DH-line Bi was resistant to clubroot, which is caused by a Dutch field isolate of Plasmodiophora brassicae. Resistance in the DH-line population was determined in two ways: by assigning symptom grades to each plant, and by measuring
the fresh weights of the healthy and affected parts of the root system of each plant. Using a multiple QTL mapping approach
to analyze the fresh weight data, we found two loci for clubroot resistance; these were designated pb-3 and pb-4. The additive effects of these loci were responsible for 68% of the difference between the parents and for 60% of the genetic
variance among DH-line means. Also, indications for the presence of two additional, minor QTLs were found. Analysis of symptom
grades revealed the two QTLs pb-3 and pb-4, as well as one of the two minor QTLs indicated by analysis of the fresh weight data.

Around 40 species of Hylarana amphibians are distributed in tropical and subtropical Asia, and Chinese broad-folded frog, Hylarana latouchii (Boulenger, 1899) is one of them. In this study, six different cDNAs encoding four novel antimicrobial peptide precursors were cloned by screening the cDNA library of the Chinese broad-folded frog skin. The protein sequence analysis demonstrated that two deduced peptides belong to the brevinin-1 family, and the other two belong to temporin family of amphibian antimicrobial peptides. Thus, they were named as brevinin-1LT1 (FMGSALRIAAKVLPAALCQIFKKC), brevinin-1LT2 (FFGSVLKVAAKVLPAALCQIFKKC), temporin-LT1 (FLPGLIAGIAKML–NH2) and temporin-LT2 (FLPIALKALGSIFPKIL–NH2), respectively. Furthermore, brevinin-1LT1 and temporin-LT1 were purified by HPLC from the skin secretion of H. latouchii. Predator-induced morphological changes in an amphibian: predation by dragonflies affects tadpole shape and color 总被引:4,自引:0,他引:4
Predator-induced defenses are well studied in plants and invertebrate animals, but have only recently been recognized in
vertebrates. Gray treefrog (Hylachrysoscelis) tadpoles reared with predatory dragonfly (Aeshnaumbrosa) larvae differ in shape and color from tadpoles reared in the absence of dragonflies. By exposing tadpoles to tail damage
and the non-lethal presence of starved and fed dragonflies, we determined that these phenotypic differences are induced by
non-contact cues present when dragonflies prey on Hyla. The induced changes in shape are in the direction that tends to increase swimming speed; thus, the induced morphology may
help tadpoles evade predators. Altering morphology in response to predators is likely to influence interactions with other
species in the community as well.

Vagrant non-breeding ravens frequently attract conspecifics to rich ephemeral food sources. There, grouping may allow them to overcome the defence of territorial breeders. Here, we focus on ravens making use of regular food supplies in a game park, where they divert food from the provision of park animals. We investigated if ravens foraging in the Cumberland game park (Grünau, Austria) are attentive towards one another when they experience some unpredictability in food provisioning. We confronted a group of 30–50 ravens with two different treatments. Ten minutes ahead of the feeding of either wolves or wild boars we showed buckets containing pieces of meat to the ravens flying overhead. In the reliable cue treatment (RCT), the meat was placed next to one of the two enclosures, whereas in the unreliable cue treatment (UCT), the buckets were placed simultaneously in front of both enclosures though only in one of the enclosures were the animals fed 10 min later. Thus, during RCT but not during UCT, ravens could predict where food would become available. Only during UCT, ravens moved in large groups between the two feeding sites. Many ravens moving at the same time in the same direction may indicate some co-ordination in space and time, which is most likely achieved by social attraction among individuals. Furthermore, the number of ravens approaching and leaving, respectively, a feeding site cross-correlated with a temporary increase in the rate of a food-associated call, the yell. This suggests that in addition to watching each other, calling may have contributed to group formation. Possible benefits of group formation during food inspection are discussed. Previous studies have demonstrated that 2-hydroxy-1-naphthaldehyde isonicotinoyl hydrazone (NIH) and several other aroylhydrazone
chelators possess anti-neoplastic activity due to their ability to bind intracellular iron. In this study we have examined
the structure and properties of NIH and its FeIII complex in order to obtain further insight into its anti-tumour activity. Two tridentate NIH ligands deprotonate upon coordination
to FeIII in a meridional fashion to form a distorted octahedral, high-spin complex. Solution electrochemistry of [Fe(NIH–H)2]+ shows that the trivalent oxidation state is dominant over a wide potential range and that the FeII analogue is not a stable form of this complex. The fact that [Fe(NIH–H)2]+ cannot cycle between the FeII and FeIII states suggests that the production of toxic free-radical species, e.g. OH
.
or O2
.
–, is not part of this ligand's cytotoxic action. This suggestion is supported by cell culture experiments demonstrating that
the addition of FeIII to NIH prevents its anti-proliferative effect. The chemistry of this chelator and its FeIII complex are discussed in the context of understanding its anti-tumour activity.

To test the hypothesis that the contribution of phosphoribulokinase (PRK) to the control of photosynthesis changes depending
on the light environment of the plant, the response of transgenic tobacco (Nicotiana tabacum L.) transformed with antisense PRK constructs to irradiance was determined. In plants grown under low irradiance (330 μmol m−2 s−1) steady-state photosynthesis was limited in plants with decreased PRK activity upon exposure to higher irradiance, with a
control coefficient of PRK for CO2 assimilation of 0.25 at and above 800 μmol m−2 s−1. The flux control coefficient of PRK for steady-state CO2 assimilation was zero, however, at all irradiances in plant material grown at 800 μmol m−2 s−1 and in plants grown in a glasshouse during mid-summer (alternating shade and sun 300–1600 μmol m−2 s−1). To explain these differences between plants grown under low and high irradiances, Calvin cycle enzyme activities and metabolite
content were determined. Activities of PRK and other non-equilibrium Calvin cycle enzymes fructose-1,6-bisphosphatase, sedoheptulose-1,7-bisphosphatase
and ribulose-1,5-bisphosphate carboxylase-oxygenase were twofold higher in plants grown at 800 μmol m−2 s−1 or in the glasshouse than in plants grown at 330 μmol m−2 s−1. Activities of equilibrium enzymes transketolase, aldolase, ribulose-5-phosphate epimerase and isomerase were very similar
under all growth irradiances. The flux control coefficient of 0.25 in plants grown at 330 μmol m−2 s−1 can be explained because low ribulose-5-phosphate content in combination with low PRK activity limits the synthesis of ribulose-1,5-bisphosphate.
This limitation is overcome in high-light-grown plants because of the large relative increase in activities of sedoheptulose-1,7-bisphosphatase
and fructose-1,6-bisphosphatase under these conditions, which facilitates the synthesis of larger amounts of ribulose-5-phosphate.
This potential limitation will have maintained evolutionary selection pressure for high concentrations of PRK within the chloroplast.

The mitochondrial DNA of various carrot lines was characterized by random amplified polymorphic DNA (RAPD) analysis, and six
sequence-tagged sites (STSs) led to identification of the petaloid type of cytoplasmic male sterility (CMS). Using six STS
primer combinations, we were able to classify five CMS lines into two groups and eight fertile carrots into six groups. Both
the STS1 and the STS4 primer combinations differentiated CMS cytoplasms from the fertile cytoplasms, and the STS2 primer combination
revealed two different types of CMS cytoplasms – of Wisconsin Wild and Cornell origins. Cybrid carrot lines with petaloid
flowers which had been obtained by asymmetric cell fusion could also be separated from fertile cybrids by the STS1 primer
combination. The STS1 fragment contained a homologous sequence with the orfB gene. DNA gel blot analysis indicated that homologous regions to the STS1 fragment existed in fertile types as well as the
CMS types, although the restriction fragment size patterns differed. These observations demonstrate that rearrangements involving
this region occurred in the mitochondrial genome. The STS4 fragment had a more complicated gene structure, including retrotransposon-like
sequences and small segments of chloroplast genome.

How the allocation of searching investiment by Ural owls (Strix uralensis) in a patchy and fluctuating environment is affected by prior information was experimentally studied. The owls searched among
four patches, two with prey and two without prey. During the five days, the positions of two prey patches were randomly assigned
daily (random treatment) and for the other five days, they were fixed (fixed treatment). In experiment I, the sequence of
treatments was from random to fixed. In experiment II, the sequence of treatments was reversed. The choice of prey present
patch was affected by acquired information which owls acquired during one night (short term), a treatment period (medium term),
and throughout the experiment run (long term). They predicted the positions of the prey patches, to some extent, in the first
choice in one night in the fixed treatment by the experience of previous days. The searching time per visit was also affected
by previous information. Owls searched prey patches longer than empty patches. In the random treatment, the giving up time
in prey patches was variable, while in the fixed treatment, it was longer and fixed. Although owls could use information on
the environment where they had searched to make a decision of foraging behavior, they were not able to choose prey patches
accurately. The information which was acquired during the treatment period and throughout the experiment run affected the
gain from prey. The changes in gain were not caused by the encounter rate but by the proportion of attack success. 相似文献

The conservation of 90 microsatellite-flanking sequences from chickpea in 39 accessions of eight annual and 1 accession of a perennial species of the genus Cicer was investigated. All of the primer sequences successfully amplified microsatellites in related species, indicating the conservation of microsatellite-flanking sequences in chickpea’s relatives. However, the degree of conservation of the primer sites varied between species depending on their known phylogenetic relationship to chickpea, ranging from 92.2% in C. reticulatum, chickpea’s closest relative and potential ancestor, down to 50% for C. cuneatum. A phylogenetic tree revealed that chickpea and the other members of its crossability group were more closely related to the perennial C. anatolicum than to other annual species of the genus. Considerable variation in size and number of amplification products between and within species was observed. Sequence analysis of highly divergent amplification products proved that variation is either due to large differences in the number of microsatellite repeats or to the amplification of a locus unrelated to the one amplified from chickpea. Sequence information and bootstrapping using PAUP suggested that STMSs derived from chickpea may be efficiently and reliably used for synteny studies in chickpea’s crossability group, including C. anatolicum. However, care should be taken when applying these markers to other species of the genus. Considering the data presented here and the known historical record, the age of section Monocicer, including chickpea, is estimated to be about 100,000 years. The protein named T1, present in Triticum tauschii, was previously characterized as a high-molecular-weight (HMW) glutenin subunit with a molecular size similar to that of
the y-type glutenin subunit-10 of Triticum aestivum. This protein was present along with other HMW glutenin subunits named 2t and T2, and was considered as part of the same allele at the Glu-D
t
1 locus of T. tauschii. This paper describes a re-evaluation of this protein, involving analyses of a collection of 173 accessions of T. tauschii, by SDS-PAGE of glutenin subunits after the extraction of monomeric protein. No accessions were found containing the three
HMW glutenin subunits. On the other hand, 17 lines with HMW glutenin subunits having electrophoretic mobilities similar to
subunits 2t and T2 were identified. The absence of T1 protein in these gel patterns has shown that protein T1 is not a component of the
polymeric protein. Rather, the T1 protein is an ω-gliadin with an unusually high-molecular-weight. This conclusion is based
on acidic polyacrylamide gel electrophoresis (A-PAGE), sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE),
and two-dimensional gel electrophoresis (A-PAGE+ SDS-PAGE), together with analysis of its N-terminal amino-acids sequence.
The inheritance of ω-gliadin T1 was studied through analyses of gliadins and HMW glutenins in 106 F2 grains of a cross between synthetic wheat, L/18913, and the wheat cv Egret. HMW glutenin subunits and gliadins derived from
T. tauschii (Glu-D
t
1 and Gli-D
t
1) segregated as alleles of the Glu-D1 and Gli-D1 loci of bread wheat. A new locus encoding the ω-gliadin T1 was identified and named Gli-DT1. The genetic distance between this new locus and those of endosperm proteins encoded at the 1D chromosome were calculated.
The Gli-DT1 locus is located on the short arm of chromosome 1D and the map distance between this locus and the Gli-D1 and Glu-D1 loci was calculated as 13.18 cM and 40.20 cM, respectively.

In California’s Sacramento-San Joaquin estuary, environmental protection and habitat restoration efforts directed at a threatened native osmerid, the delta smelt (Hypomesus transpacificus), are complicated by the presence of a morphologically similar non-native congener, the wakasagi (H. nipponensis), transported to the estuary from upstream reservoirs. In order to better define delta smelt critical habitat and to evaluate the potential for habitat overlap by these two species, we compared the tolerances of the two species to temperature, salinity, and water velocity, environmental factors that vary spatially and temporally within the estuary. For fishes acclimated to 17°C and fresh water (0 ppt), we measured critical thermal maxima and minima, chronic upper salinity tolerance limits, and critical swimming velocities. Wakasagi had higher critical thermal maxima (29.1°C vs. 25.4°C for delta smelt), lower critical thermal minima (2.3°C vs. 7.5°C for delta smelt), higher upper salinity tolerances (26.8 ppt vs. 19.1 ppt for delta smelt), and swam faster (for 6–6.9 cm SL fish, 43.3 cm s–1 vs. 28.2 cm s–1 for delta smelt) than delta smelt. This suggests that the wide seasonal and year-to-year fluctuations in temperature, salinity, and flow typical in the estuary would not exclude wakasagi, although their eggs and larvae may be less tolerant. With respect to these factors, the native delta smelt may be at a physiological disadvantage, particularly in habitats with suboptimal environmental conditions, and may be excluded from shallow-water habitat restoration sites, which are characterized by poor circulation, low flows, and more environmentally extreme conditions. The low abundance of wakasagi in the estuary recorded to date may indicate that factors other than temperature, salinity, and flow determine wakasagi distribution. Seeds of Cichorium intybus L. var. foliosum cv. Flash were sown in acid-washed vermiculite and grown in a controlled-environment growth chamber. After 1 month of growth,
plantlets did not contain sucrose:sucrose 1-fructosyltransferase (1-SST), the key enzyme in fructan biosynthesis. No fructan
could be observed. Some of the plants were submitted to drought for 2 weeks. Glucose, fructose and sucrose concentrations
increased in roots and leaves of stressed plants and the fructan concentration in roots and leaves was ten times higher than
in control plants. The onset of fructan synthesis coincided with the increase in 1-SST activity in roots. Expression of the
1-SST gene could be observed in roots and leaves of stressed plants.

Summary. L- and D-aspartic acids (L-Asp and D-Asp) are present in the majority of nervous systems. In phylogeny, significant levels
have been reported in mollusc brains, particularly cephalopods. To examine the role of L- and D-Asp on a cephalopod receptor,

voltage clamp, application of L-glutamate (L-Glu; 1–30 mM), but not D-glutamate (D-Glu), or L- or D-Asp, evoked an inward
current of 0.1 nA. L- or D-Asp (200 μM) applied with 20 mM L-Glu, slowed the time course of activation and inactivation of
the L-Glu gated current (time constant increased from 1 s (L-Glu alone) to 3 s (D-Asp and L-Glu) and to 19 s (L-Asp and L-Glu)).
Our results suggest that in molluscan systems, aspartic acid could act as a neuromodulator during glutamatergic transmission
and could significantly alter synaptic integration by slowing glutamate receptor gating. 相似文献
