Eight novel and two heterologous microsatellite pairs of primers are presented for the Austral hake (Merluccius australis), representing the first microsatellite markers available for this species. Loci were characterized for 50 individuals from two populations in South America (Argentinean and Chilean coasts). All loci were polymorphic within M. australis (5 to 30 alleles per locus; observed heterozygosity between 0.320 and 0.840), and therefore useful for population genetic studies within the species. Cross-species transferability was tested for 100 individuals from four additional species within the Merluccius genus (M. albidus, M. bilinearis, M. gayi and M. hubbsi), and results indicate that most of these primers pairs will likely be useful for population genetic studies on Merluccius species.  相似文献

The ontogeny of the developmental stages of the hake Merluccius hubbsi is described. Fish larvae and post-transitional juveniles were collected in the Nor-Patagonian area from 1989 to 2004. The opening of the mouth and the pigmentation of the eyes are coincident with yolk resorption, finishing the yolk-sac stage. This species presents pigmentation on the head, trunk and tail typical of gadiform larvae. Pectoral fin development is completed during the transformation stage. The post-transitional juvenile stage begins when the fin-ray complements are complete and squamation begins. The fins become fully formed in the following sequence: pelvic fins, first dorsal fin, second dorsal and anal fins together, caudal fin and pectoral fins. The caudal complex is totally developed in larvae of 22·0–23·0 mm standard lengths ( L S) and all vertebral elements are first observed in larvae of 8·5 mm L S. The rate of development of M. hubbsi observed in this study could be faster than the rates reported for other species of Merluccius by different authors.  相似文献

Morphologically, both classic and landmark-based morphometry and meristic analyses of 241 specimens of Merluccius, along with the re-examination of six paratype specimens of Merluccius hubbsi, the holotype and three paratypes of Merluccius patagonicus and the syntype of Merluccius australis revealed the presence of only two species of Merluccius in Argentinean waters. Internal structures (hyomandibula, urohyal and sagitta otolith) of M. hubbsi were compared to those reported for M. patagonicus and were shown to have identical morphology. Type specimens of M. patagonicus showed a complete overlap in morphometric and meristic characters with M. hubbsi, whereas M. australis had a greater number of second dorsal and anal-fin rays, and more lateral-line scales. In addition, M. australis had a smaller eye and longer snout than M. hubbsi and M. patagonicus. The results indicate that there is no evidence for a third hake species in Argentinean waters. Merluccius patagonicus is a synonym of M. hubbsi.  相似文献

The utility of 15 new and 17 previously published microsatellite markers was evaluated for species identification and stock delimitation in the deep‐water hake Merluccius paradoxus and the shallow‐water hake Merluccius capensis. A total of 14 microsatellites were polymorphic in M. paradoxus and 10 in M. capensis. Two markers could individually discriminate the species using Bayesian clustering methods and a statistical power analysis showed that the set of markers for each species is likely to detect subtle genetic differentiation (FST < 0·006) that will be valuable to delimit and characterize genetic stocks.  相似文献

Phylogeny of the genus Merluccius based on mitochondrial and nuclear genes   总被引:2,自引:0,他引:2  
Genus Merluccius is considered one of the most important groups within the Teleostei, and comprises 12 extant species distributed along the coasts of America, Europe and Africa, being its fisheries very important in these continents. Despite their noticeable economical importance for humans, to date the phylogeny of hakes has not been clearly established. In this study we used mitochondrial sequences (the ribosomal genes 12S rDNA and 16S rDNA, the coding gene cytochrome b and the control region) and the nuclear 5S rDNA conserved region in order to determine the phylogenetic and biogeographical relationships within the genus Merluccius. This is the first time that all the species of this genus recognized by the FAO are included in a phylogeny. Maximum Parsimony, Maximum Likelihood and Bayesian analyses of the mitochondrial sequences suggest that the geographical origin of the genus was the North Atlantic Ocean, and indicate that two main clades, early separated in the evolution, exist within the genus: one American (7 species) and one Euro-African (5 species). Among the American species, M. bilinearis seems to be the most ancient one, and the rise of the Panama Isthmus could act as a physical barrier leading to further processes of speciation. Within the Euro-African clade, successive events of geographical differentiation could explain the observed pattern of species distribution. Therefore, we propose both vicariant speciation and geographical dispersion as main mechanisms to explain the evolutionary history of the genus Merluccius.  相似文献

The diet composition and feeding strategy of the Argentine hake Merluccius hubbsi in the San Matías Gulf were analysed in order to use this information for the sustainable management of the fishery. Merluccius hubbsi behaved as an opportunistic predator. Small M. hubbsi consumed planktonic crustaceans, whereas medium and large fish ate numerous prey taxa with low frequency of occurrence and variable specific abundance. Intra- and intercohort cannibalism were detected in all size groups and were particularly significant in large M. hubbsi. Medium-sized M. hubbsi consumed small conspecifics and large-sized M. hubbsi consumed both small and medium M. hubbsi. These results indicate that the removal of large M. hubbsi by fishing may increase the risk of overfishing by two combined effects: a direct effect of recruitment-overfishing and an indirect effect of growth-overfishing through an enhanced cannibalism of medium M. hubbsi on small M. hubbsi. Intra- and intercohort cannibalism and other trophic relationships in the M. hubbsi should therefore be considered explicitly in stock assessment models.  相似文献

The present study evaluated the possible occurrence of cryptic species among Merluccidae from Argentina by examining sequences of cytochrome c oxidase subunit I (coI) mtDNA. This approach can discriminate Merluccius hubbsi and Merluccius australis; specimens with morphological diagnostic characters of Merluccius patagonicus formed a cohesive cluster with M. hubbsi specimens. The Cape hake species, Merluccius capensis and Merluccius paradoxus are the most important resource of the South African and Namibian demersal fishery, but it is unclear whether there is a single population of each shared by both countries. We analysed the population structure and evolutionary history of these two species using the variable 5' region of the mtDNA control region for 311 specimens of M. capensis and 333 specimens of M. paradoxus sampled between Lüderitz (southern Namibia) to south of Cape Point (South Africa). 107 haplotypes for M. capensis and eight haplotypes for M. paradoxus were recovered. AMOVA and pairwise Phi(st) analyses revealed no structure in M. capensis, however significant genetic differentiation between Namibian and South African 'populations' was detected for M. paradoxus. This was only restricted to mature fish older than 3 and 4 years and not for juvenile fish younger than 3 years. Analyses reveal that M. capensis has undergone population expansion (Fu's Fs=-26.65, P<0.001), possibly within the last 4500-23,000 years, whereas M. paradoxus has not. Our study highlights the utility of genetic markers to unravel the evolutionary history of sympatric species, as well as addressing management issues within regions where commercially valuable fish stocks are shared between nations.  相似文献

Sulfite oxidase (sulfite:oxygen oxidoreductase, EC 1.8.3.1) was purified 482-fold from liver of the Pacific hake Merluccius productus. The molecular weight of the enzyme was found to be 120 000 by gel exclusion chromatography on Sephadex G-100. Electrophoretic analysis on sodium dodecyl sulfate (SDS)-polyacrylamide gel revealed that the enzyme was composed of two subunits whose molecular weight was estimated to be 60 000. The pH optimum of the enzyme was 8.7; Ks for sulfite, 2.5 x 10(-5) M; and that for cytochrome c, 3.6 x 10(-7) M. The enzyme elicited an EPR signal at g = 1.97 characteristic of pentavalent molybdenum. Colorimetric analysis also disclosed that the enzyme contained 2 mol each of heme and molybdenum per mol of protein. This fish liver homogenate in isotonic sucrose solution was fractionated by differential centrifugation into nuclei, mitochondria, microsomes and supernatant (100 000 X g). The major portion of sulfite oxidase activity was found in mitochondria. The sulfite oxidase activity was markedly high in liver and kidney, as compared with that in heart, spleen, muscle, gill and eye.  相似文献

This study investigated the somatic growth and energy allocation strategy of two sympatric hake species (Merluccius polli and Merluccius senegalensis), coexisting under the strong influence of the Mauritanian upwelling. The results revealed that ontogeny, bathymetry, geography and reproduction shaped the differences found between the condition dynamics of the two species. Aside from species‐specific differences, individuals were observed in better condition in the northernmost area (more influenced by the permanent upwelling) and in the deepest waters, probably the most favourable habitat for Merluccius spp. Both species also displayed contrasting trade‐offs in energy allocation probably due to the dissimilarity of their habitats, which favours the existence of divergent adaptive strategies in response to different ontogenic requirements. It was hypothesized that M. polli invests in mass and energy reserves while sacrificing growth, as larger sizes may not provide an ecological advantage in a deeper and more stable environment. Moreover, M. senegalensis capitalizes on a steady growth without major disruptions, enabling earlier spawning at the expense of a lower somatic mass, which is fitting to a less stable shallower environment. This study sheds new light on differences in the biological traits and life strategies of Merluccius spp., which permit their overlap in a complex upwelling system and may contribute to the long‐lasting scientific‐based management of these species.  相似文献

Genetic variation and divergence among samples of Chilean hake Merluccius gayi, from three localities off the coast of Chile and one locality off the coast of northern Peru, were assessed using sequences from the control region of mitochondrial DNA. Homogeneity tests revealed occurrence of at least three distinct genetic stocks of M. gayi within the region sampled. Factors potentially contributing to genetic divergence among M. gayi probably include hydrodynamics and behaviour.  相似文献

The genetic structure of Chilean hake Merluccius gayi gayi, was analyzed using starch gel protein electrophoresis. Samples were collected from four localities along the coast off Chile. A total of 1500 specimens sampled from Coquimbo, San Antonio, Talcahuano and Puerto Montt were used in the study. Genetic information was obtained for six allozyme loci Pgi-1, Pgi-2, Pgm, Idh-1, Idh-2 and Aat. The results of the analysis showed no significant allelic differences among samples from various localities, with an average FST value of 0.007 for all loci and samples. A heterogeneity test for all loci and specimens from the four localities showed only two significant values. Thus, Chilean hake populations along the coast of Chile appear to be genetically homogeneous.  相似文献

The anacrosomal aquasperm of the gadiform Merluccius merluccius is ultrastructurally similar to the advanced type II spermatozoa (perciform‐type sperm) typically found in most Perciformes. The perciform‐type spermatozoon is characterized by the lateral insertion of the flagellum and the location of the centrioles outside the nuclear fossa. Apart from these characteristics, the spermatozoon of M. merluccius is remarkable because of the mutually parallel arrangement of the centrioles, a rare feature among fishes, which is considered an apomorphic condition for animal sperm cells. Within the superorder Paracanthopterygii, which contains a large diversity of sperm patterns resulting from a high number of apomorphies, a perciform‐type sperm is present only in the order Gadiformes. The significance of the presence of perciform‐type spermatozoa in the three investigated gadiform families is discussed in a phylogenetic context.  相似文献

A pioneering experiment of archival tagging of European hake Merluccius merluccius provided evidence of a diel vertical migration pattern which was analysed using an automatic method, developed and validated through time-frequency analyses. Frequent vertical movements across the thermocline were observed and fish experienced rapid temperature changes >7° C. These tagging-recapture data also suggested a homing behaviour to feeding grounds.  相似文献

The interannual distribution of early life stages of Pacific hake Merluccius productus , within the southern part of the California Current (32–23° N) from 1951 to 2001, was examined to describe the relationship between spawning habitat and environmental conditions. Mean annual abundance was affected by different factors along the west coast of the Baja California Peninsula. In the northern areas (Ensenada and Punta Baja), reduced abundance of larvae coincided with the El Niño and a North Pacific Ocean climatic regime shift, but in the southern areas (San Ignacio to Bahía Magdalena), the drastic reductions suggested a fishery effect for large adults of the coastal migratory population, starting in 1966. Two spawning stocks, coastal and dwarf, were evident in comparisons of latitudinal differences in occurrence of early stages and differences in temperature preferences that seemed to break at Punta Eugenia.  相似文献

The cloning and sequencing of the small subunit (SSU) ribosomal DNA gene from Kudoa rosenbuschi (myxosporean species associated with post-mortem myoliquefaction process in the Argentinean hake Merluccius hubbsi) is reported. The SSU rDNA was found to contain 1740 bp with a single polymorphic site with either a C or T at position 221. The sequence data obtained in this study and those known sequences of Kudoa species deposited in the GenBank were all analyzed to construct a phylogenetic tree. Nucleotide sequences showed the highest degree of identity with K. funduli, followed by K. miniauriculata, K. clupeidae and K. dianae. Phylogenetic analysis placed K. rosenbuschi in the same branch of K. clupeidae and K. funduli, and showed it to be closely related to K. dianae, K. paniformis and K. miniauriculata.  相似文献

General linear models (GLMs) were used to determine the relative importance of interspecific, ontogenetic and spatial effects in explaining variability in stable isotope ratios of nitrogen (δ15N) and carbon (δ13C) of the co‐occurring Cape hakes Merluccius capensis and Merluccius paradoxus off South Africa. Significant GLMs were derived for both isotopes, explaining 74 and 56% of observed variance in Merluccius spp. δ15N and δ13C, respectively. Spatial effects (west or south coast) contributed most towards explaining variability in the δ15N model, with Merluccius spp. off the west coast having higher (by c. 1·4‰) δ15N levels than Merluccius spp. off the south coast. Fish size and species were also significant in explaining variability in δ15N, with both species showing significant linear increases in δ15N with size and M. capensis having higher (by c. 0·7‰) δ15N values than M. paradoxus. Species and coast explained most and similar amounts of variability in the δ13C model, with M. capensis having higher (by c. 0·8‰) δ13C values than M. paradoxus, and values being lower (by c. 0·7‰) for fishes off the west coast compared with the south coast. These results not only corroborate the knowledge of Merluccius spp. feeding ecology gained from dietary studies, in particular the ontogenetic change in trophic level corresponding to a changing diet, but also that M. capensis feeds at a slightly higher trophic level than M. paradoxus. The spatial difference in Merluccius spp. δ15N appears due to a difference in isotopic baseline, and not as a result of Merluccius spp. feeding higher in the food web off the west than the south coast, and provides new evidence that corroborates previous observations of biogeographic differences in isotopic baselines around the South African coast. This study also provides quantitative data on the relative trophic level and trophic width of Cape hakes over a large size range that can be used in ecosystem models of the southern Benguela.  相似文献

The patterns were reproducible within each species and distinguishable from one another. 3. The frequency distribution of numbers of vertebrae and of gillrakers were obtained for the same fish. 4.
