Carbohydrate and abscisic acid (ABA) metabolism were investigated in normal and phenotypically small 'Hass' avocado ( Persea americana Mill.) fruit in an attempt to link alterations in sugar and ABA content with changes in 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR, EC 1.1.1.34) activity and fruit size. The small-fruit phenotype was characterized by reduced seed HMGR activity, increased seed insoluble acid invertase ( β - d -fructofuranosidase, EC 3.2.1.26), decreased sucrose synthase (SS; UDP- d -glucose: d -fructose-2- α -glucosyl-transferase, EC 2.4.1.13) activity, decreased sucrose content, and increased glucose as a proportion of the total soluble sugar. Sucrose phosphate synthase (SPS; UDP- d -glucose: d -fructose 6-phosphate 2- α - d -glucosyltransferase, EC 2.4.1.14) activity was unaffected in seed but reduced in mesocarp of the small fruit. In addition, the small-fruit variant displayed enhanced respiration and both seed and mesocarp tissue showed increased ABA metabolism. Applied ABA caused an increase in insoluble acid invertase activity in seed tissue of normal fruit while mevastatin reduced HMGR activity in this tissue, caused sucrose depletion and increased the proportion of glucose from 5 to 57% of total soluble sugars. Exogenous glucose suppressed HMGR activity in seed tissue whereas in mesocarp tissue, HMGR activity was reduced to 38% of the control after 6 h but enhanced by 46% by 48 h. Glucose increased ABA biosynthesis and turnover in competent tissues. These results suggest that ABA turnover is mediated, in part, by carbohydrate content and composition which also affects HMGR activity. It is proposed that sugar and ABA signals act in concert to modulate expression and/or activity of HMGR in the control of 'Hass' avocado fruit growth and final fruit size.  相似文献

Sugar content largely determines watermelon fruit quality. We compared changes in sugar accumulation and activities of carbohydrate enzymes in the flesh (central portion) and mesocarp of elite sweet watermelon line 97103 (Citrullus lanatus subsp. vulgaris) and exotic non-sweet line PI296341-FR (C. lanatus subsp. lanatus) to elucidate the physiological and biochemical mechanisms of sugar accumulation in watermelon fruit. The major translocated sugars, raffinose and stachyose, were more unloaded into sweet watermelon fruit than non-sweet fruit. During the fruit development, acid α-galactosidase activity was much higher in flesh of 97103 than in mesocarp of 97103, in flesh and mesocarp of PI296341-FR fruit. Insoluble acid invertase activity was higher in 97103 flesh than in 97103 mesocarp, PI296341-FR flesh or mesocarp from 18 days after pollination (DAP) to 34 DAP. Changes in soluble acid invertase activity in 97103 flesh were similar to those in PI296341-FR flesh and mesocarp from 18 DAP to full ripening. Sucrose synthase and sucrose phosphate synthase activities in 97103 flesh were significantly higher than those in 97103 mesocarp and PI296341-FR fruits from 18 to 34 DAP. Only insoluble acid invertase and sucrose phosphate synthase activities were significantly positively correlated with sucrose content in 97103 flesh. Therefore, phloem loading, distribution and metabolism of major translocated sugars, which are controlled by key sugar metabolism enzymes, determine fruit sugar accumulation in sweet and non-sweet watermelon and reflect the distribution diversity of translocated sugars between subspecies.  相似文献

Sugar content and composition are major criteria used in judging the quality of netted muskmelon (Cucumis melo L. var reticulatus) fruit. Sugar composition and four enzymes of sucrose metabolism were determined in `Magnum 45' muskmelon fruit at 10-day intervals beginning 10 days after pollination (DAP) until full-slip (35 DAP). Sugar content increased in both outer (green) mesocarp and inner (orange) mesocarp between 20 and 30 DAP. The major proportion of total increase in sugar was attributed to sucrose accumulation. The large increase in sucrose relative to glucose and fructose was accompanied by a dramatic decrease in acid invertase activity, which was highest in both tissues at 10 and 20 DAP, and increases in sucrose phosphate synthase (SPS) and sucrose synthase activities. The green tissue had a lower proportion of total sugar as sucrose, greater invertase activity, and less SPS activity than the orange tissue. Changes in relative sucrose content were highly correlated with changes in enzyme activity. The results strongly suggest that increases in the proportion of sucrose found in melon fruit were associated with a decline in acid invertase activity and an increase in SPS activity approximately 10 days before full-slip. Therefore, these enzymes apparently play a key role in determining sugar composition and the quality of muskmelon fruit.  相似文献

Sucrose accumulation in developing peach fruit   总被引:35,自引:0,他引:35  
Uptake of 14C-sugars and activities of sucrose metabolizing enzymes were determined in order to study the mechanism(s) of sucrose accumulation in developing peach fruit. Mesocarp of young peach fruit contained glucose and fructose but little sucrose. Starting 88 days after anthesis (DAA) the sucrose concentration increased greatly. The mechanism of sucrose accumulation was studied by measuring 14C-sucrose and 14C-glucose uptake rates at three different stages of fruit development, and by assaying weekly the activity of enzymes involved in the hydrolysis and/or synthesis of the soluble sugars. Uptake of 0.5–100 m M 14C-sucrose and 14C-glucose by mesocarp tissue slices showed a complex pattern at the first stage of fruit development (62 DAA). During the subsequent growth stages the pattern of sugar uptake changed and was approximately monophasic at the third stage of fruit development.
At 10 m M , glucose was taken up more rapidly than sucrose at the first and second stage of fruit development. Uptake was partially inhibited by the uncoupler carbonylcyanide m -chlorophenylhydrazone (CCCP) at 25 μ M. These results, together with the presence of a putative extracellular invertase, suggest an apoplastic route for sucrose uptake which is dependent, at least in part, on energy supply.
Activities of sucrose hydrolyzing enzymes (insoluble acid invertase, soluble acid invertase, neutral invertase, sucrose synthase) were high in young fruits and declined sharply with fruit development concomitantly with accumulation of sucrose. The storage of the sugar was not accompanied by a rise in synthetic activities (sucrose synthase, sucrose phosphate synthase), suggesting that sucrose could, at least in part enter the carbohydrate pool directly.  相似文献

Malaysia is blessed with abundant natural resources and bears a favorable climate for commercial cultivation of crops such as oil palm. In Malaysia the total plantation area of oil palm was 4,487,957 ha in 2008. It has been reported that in 2005 there was a total of 423 palm oil mills having production capacity of approximately 89 million tonnes of fresh fruit bunch (FFB) per year. Waste from the oil palm mill process include palm oil mill effluent (POME), generated mainly from oil extraction, washing and cleaning up processes. POME contains cellulosic material, fat, oil, and grease. Discharging untreated effluent into water streams may cause considerable environmental problems. The solid wastes generated are mainly decanter cake, empty fruit bunches, seed shells and fibre from the mesocarp. POME as well as the solid wastes may rapidly deteriorate the surrounding environment if not dealt with properly. Hence there is an urgent need for a sustainable waste management system to tackle these wastes. As these wastes are organic in origin, they are rich in plant nutrients. Composting of waste generated from palm oil mills can be good practice as it will be helpful in recycling useful plant nutrients. This review deals with various aspects of waste management practices in palm oil mills and the possibility of composting the wastes.  相似文献

To better understand lipid biosynthesis in oil palm mesocarp, in particular the differences in gene regulation leading to and including de novo fatty acid biosynthesis, a multi-platform metabolomics technology was used to profile mesocarp metabolites during six critical stages of fruit development in comparatively high- and low-yielding oil palm populations. Significantly higher amino acid levels preceding lipid biosynthesis and nucleosides during lipid biosynthesis were observed in a higher yielding commercial palm population. Levels of metabolites involved in glycolysis revealed interesting divergence of flux towards glycerol-3-phosphate, while carbon utilization differences in the TCA cycle were proven by an increase in malic acid/citric acid ratio. Apart from insights into the regulation of enhanced lipid production in oil palm, these results provide potentially useful metabolite yield markers and genes of interest for use in breeding programmes.  相似文献

The date palm Phoenix dactylifera has played an important role in the day-to-day life of the people for the last 7000 years. Today worldwide production, utilization and industrialization of dates are continuously increasing since date fruits have earned great importance in human nutrition owing to their rich content of essential nutrients. Tons of date palm fruit wastes are discarded daily by the date processing industries leading to environmental problems. Wastes such as date pits represent an average of 10% of the date fruits. Thus, there is an urgent need to find suitable applications for this waste. In spite of several studies on date palm cultivation, their utilization and scope for utilizing date fruit in therapeutic applications, very few reviews are available and they are limited to the chemistry and pharmacology of the date fruits and phytochemical composition, nutritional significance and potential health benefits of date fruit consumption. In this context, in the present review the prospects of valorization of these date fruit processing by-products and wastes’ employing fermentation and enzyme processing technologies towards total utilization of this valuable commodity for the production of biofuels, biopolymers, biosurfactants, organic acids, antibiotics, industrial enzymes and other possible industrial chemicals are discussed.  相似文献

Protoplasts were enzymatically prepared from the mesocarp of two species of oil palm (Elaeis guineensis Jacq. and E. oleifera HBK and Cortes) 16–20 weeks after anthesis and from rapidly multiplying embryogenic cultures of E. guineensis. The protoplasts were purified by density gradient centrifugation in 20% (w/v) sucrose. Radioactive incorporation studies showed that the protoplasts metabolized [1-14C]acetate to lipids, water-soluble compounds and 14CO2. The [14C]fatty acids obtained consisted mainly of C16: 0, C18: 0 and C18: 1. C16: 1, a very minor fatty acid in palm oil, was also labelled and accounted for 8–39% of total fatty acids synthesized by the mesocarp and embryogenic culture protoplasts. The ratio of labelled C18: 0 to C18: 1 was found to vary with the age of the fruit from which the protoplasts were prepared. Thin layer chromatography (TLC) of the labelled lipids showed the presence of all neutral acylglycerol classes. However the distribution of radiolabel in the various classes differed from those previously reported for oil palm mesocarp [K.C. Oo et al. Lipids, 20 (1985) 205] and embryoid tissue slices [E. Turnham and D.H. Northcote, Phytochem., 23 (1984) 35]. Ozonolysis showed that all the labelled C18: 1 acid was vaccenic acid.  相似文献

A common modern cultivation practice is bagging the fruit bunch if date palm (Phoenix dactylifera L.) which may influence fruit maturity and nutraceutical quality. Exposure of fruits to photooxidative stress induces changes in the endogenous concentrations of plant hormones and other metabolites, which may cause accelerated fruit maturity. This study was conducted to examine the effect of exposure to direct and indirect sunlight on date palm fruit development. The indirect sunlight treatment was simulated by fruit bunch bagging, a common practice in modern date production. The exposure of date palm fruits to direct sunlight-induced photooxidative stress causing an increased concentration of ascorbic acid and decreased content of chlorophyll, anthocyanins, carotenoids, and phenols compared to the fruit bagging treatment. Direct sunlight also reduced the concentration of phytohormones, including indoleacetic acid, gibberellin, and zeatin, but increased abscisic acid accumulation. The directly-exposed fruits reached a partially-mature stage (Rutab) in August, whereas the bagged fruits remained at the immature stage (Khalal). This study is the first to describe the biochemical basis of the observed improvement of date palm fruit development in response to reduced light intensity. Besides, it provides insights into controlling date palm fruit maturity and subsequently prolonging the shelf life dates on the tree; thus, extending the marketing period for the benefit of the farmers.

The motacú palm (Attalea phalerata) is widespread in Bolivia and is an economically valuable species as a source of vegetable oil. In Bolivia it is utilized as construction material, food, medicine, cosmetic, and edible oil. Oil extracted from the kernel (60–70% of dry weight) is high in lauric and myristic oils and compares favorably with other tropical oil crops. The palms reach reproductive maturity in 7–10 years and potential oil production from cultivated stands is 1.1-2.4 tons/ha/yr.  相似文献

Changes in the carbohydrate profiles in the mesocarp, endocarp, and seeds of maturing cucumber (Cucumis sativus, L.) fruit were analyzed. Fruit maturity was measured by a decrease in endocarp pH, which was found to correlate with a loss in peel chlorophyll and an increase in citric acid content. Concentrations of glucose and fructose (8.6-10.3 milligrams per gram fresh weight, respectively) were found to be higher than the concentration of sucrose (0.3 milligrams per gram fresh weight) in both mesocarp and endocarp tissue. Neither raffinose nor stachyose were found in these tissues. The levels of glucose and fructose in seeds decreased during development, but sucrose, raffinose, and stachyose accumulated during the late stages of maturation. Both raffinose and stachyose were found in the seeds of six lines of Cucumis sativus L. This accumulation of raffinose saccharides coincided with an increase in galactinol synthase activity in the seeds. Funiculi from maturing fruit were found to be high in sucrose concentration (4.8 milligrams per gram fresh weight) but devoid of both raffinose and stachyose. The results indicated that sucrose is the transport sugar from the peduncle to seed, and that raffinose saccharide accumulation in the seed is the result of in situ biosynthesis and not from direct vascular transport of these oligosaccharides into the seeds.  相似文献

Citrate production and degradation during the last stage of fruit development were modelled by representing the fluxes through the enzymes of the citrate cycle and the malic enzyme, the transport of metabolites between the cytosol and the mitochondria, and the stoichiometry equations that relate these reactions. After solving the corresponding system of equations, the rate of citrate synthesis (or degradation) was expressed as a simple function of temperature, mesocarp weight, and respiration. The model was applied to peach fruit, and its parameters were estimated from the data of a 2-year field experiment. The predictions of the model were in agreement with experimental data. Simulations were made to analyse the responses to variations of temperature and fruit growth. Increasing fruit growth before stone hardening stimulated citrate production, while increasing fruit growth after stone hardening reduced it. Delaying the date at which the maximum growth rate was reached enhanced citrate production during most of the period. In the last weeks before harvest, increasing temperature depressed citrate production, while, at the beginning of the period studied, it enhanced it.  相似文献

Flooding the roots of greenhouse-grown muskmelon (Cucumis meloL. cv. Noy Yizreel) plants for 4 days reduced sucrose accumulation36% in the inner mesocarp and 88% in the outer mesocarp of developingfruit. Concentration of the translocated sugars raffinose andstachyose were also lower in fruit on flooded plants than inthose from nonflooded plants. In contrast, fruit hexose concentrationwas similar in both flooded and nonflooded plants. There wasno alteration in activities of enzymes associated with sucrosemetabolism in the fruit which could explain the decreased sucroseconcentration. Four days of root flooding caused no reductionin leaf carbon exchange rate or assimilate export rate, indicatingthat the reduction in fruit sucrose accumulation was not dueto source limitation. Root respiration, measured as CO2 evolution,was approximately 30% lower in anaerobic roots than in aerobicroots. When viewed as carbohydrate consumed, a doubling of glycolyticactivity occurred in the anaerobic root mass. Increased demandfor carbohydrates by anaerobic roots may lead to a reductionin translocated carbohydrates available for sucrose biosynthesisin the developing fruit.
