The effect of irradiance on changes in photosynthesis, free amino acids and polyamines was investigated. Two-week-old maize ( Zea mays L.) plants were chilled at 5°C in the light (250 μmol m−2 s−1 PAR) or dark. The chlorophyll fluorescence ratio, Fv/Fm, decreased in the light by ca 50% but did not change in the dark. Similarly to the Fv/Fm, there was no change in the transpiration rate or the stomatal conductance in the dark, while these parameters decreased by ca 55% in the light. The net photosynthesis rate declined in both cases, but to a far greater extent in the light (73%) than in the dark (40%). The intercellular CO2 partial pressure increased by ca 50% in all cases. The free amino acid contents increased compared to the control during the cold treatment. In most cases this increase was more pronounced in the light than in the dark. There was a continuous increase in the putrescine level, which was more pronounced in the light than in the dark. The spermidine content increased one and a half times after one day in the light but decreased by 70% in the dark compared to the control values. From the second day a 50% decline in the spermidine content was observed in the light and an 80% decline in the dark. These results suggest that light has an influence not only on the photosynthetic processes during chilling stress but also on other stress markers such as polyamines and free amino acids.  相似文献

The activities NADH and NADPH dehydrogenases were measured with ferricyanide as electron-acceptor (NADH-FeCN-ox and NADPH-FeCN-ox, respectively) in mitochondria-free chloroplasts of barley leaf segments after receiving various treatments affecting senescence. NADPH-FeCN-ox declined during senescence in the dark, in a way similar to chlorophyll and Hill reaction, and increased when leaf segments were incubated at light. These results suggest that NADPH-FeCN-ox is related to some photosynthetic electron transporter activity (probably ferredoxin-NADP+ oxidoreductase). In contrast, NADH-FeCN-ox is notably stable during senescence in the dark and at light. This activity increased during incubation with kinetin or methyl-jasmonate (Me-JA) but decreased when leaf segments were treated with abscisic acid (ABA). The effects of the inhibitors of protein synthesis cycloheximide and chloramphenicol suggest that the changes of NAD(P)H dehydrogenase activities may depend on protein synthesis in chloroplasts. The activities of nitrate reductase (EC1.6.6.1), nitrite reductase (EC 1.6.6.4), glutamine synthetase (EC6.3.1.2), glutamate synthase (EC1.4.7.1) and NAD(P)H-dependent glutamate dehydrogenase (EC 1.4.1.3) were investigated in mesophyll and bundle sheath cells of maize leaves (Zea mays L.). Exogenous spermidine (Spd) and methylglyoxal bis(guanylhydrazone) (MGBG), a putative inhibitor of Spd synthesis, improved somatic embryo formation of Scots pine (Pinus sylvestris L.). The induced maturation due to MGBG and Spd was accompanied by significantly retarded proliferation growth and by reduction in the concentration of free polyamines compared to the control cultures. The action of MGBG revealed that it has a non-specific effect on the whole polyamine metabolism of Scots pine. Furthermore, at certain concentrations it may induce plant differentiation as well.  相似文献

In callus cultures of Nicotiana tabacum L. cv. Burley 21 we have examined the effect of two auxin concentrations (1 and 11.5 μ M α-naphthaleneacetic acid) in the culture medium on K+, putrescine and nicotine levels and activities of putrescine-biosyn-thetic enzymes l -arginine decarboxylase (EC 4.1.1.19) and l -ornithine decarboxylase (EC 4.1.1.17). The calli grown on the low-auxin medium (with optimal auxin concentration for nicotine synthesis) had significantly lower concentrations of K+ and higher concentrations of nicotine than those grown on the high-auxin medium (with a supraoptimal auxin concentration). Furthermore, in the calli grown on both culture media, there was a positive correlation between the levels of HCIO4-soluble free putrescine and nicotine, as well as a negative correlation between those of HCIO4-soluble bound putrescine and the alkaloid. The results suggest that in tobacco callus K+ uptake, the accumulation of HCIO4-soluble free putrescine and nicotine synthesis are related processes that depend upon the concentration of auxin in the culture medium; a concentration of 1 μ M NAA would increase HCIO4-soluble free putrescine level to a greater degree than that of 11,5 μ M NAA, and consequently lead to a higher production of the alkaloid. Although both putrescine-biosynthetic enzymes are active in our callus cultures, ornithine decarboxylase activity was considerably greater. This interpretation is supported by the enhancement of the 35.5 kDa band and 38.9 kDa band (detected by SDS-PAGE) which showed ornithine and arginine decarboxylase activity, respectively.  相似文献

Vascular development is a central theme in plant science. However, little is known about the mechanism of vascular development in monocotyledons (compared with dicotyledons). Therefore, we investigated sequential processes of differentiation into various different vascular cells by carrying out detailed observations using serial sections of the bases of developing leaves of rice and maize. The developmental process of the longitudinal vascular bundles was divided into six stages in rice and five stages in maize. The initiation of differentiation into procambial progenitor cells forming the commissural vein arose in a circular layer cell that was adjacent to both a metaxylem vessel and one or a few phloem cells in stage V longitudinal vascular bundles. In most cases the differentiation of ground meristem cells into procambial progenitor cells extended in one direction, toward the next longitudinal vascular bundle, and subsequent periclinal divisions and further differentiation produced a vessel element, two companion cells and a sieve element to form a commissural vein. These results suggest the presence of an intercellular signal(s) that induces differentiation of the circular layer cell and the ground meristem cells into procambial progenitor cells, forming a commissural vein sequentially.  相似文献

Effects of cadmium (Cd(2+)) on photosynthetic and antioxidant activities of maize (Zea mays L.) cultivars (3223 and 32D99) were investigated. Fourteen-day-old cultivar seedlings were exposed to different Cd concentrations [0, 0.3, 0.6 and 0.9mM Cd(NO(3))(2).4H(2)O] for 8 days. The results of chlorophyll fluorescence indicated that different levels of Cd affected photochemical efficiency in 3223 much more than that in 32D99. In parallel, the level of Cd at 0.9mM caused oxidative damage but did not indicate cessation of PSII activity of the cultivars; plant death was not observed at highly toxic Cd levels. Additionally, the increase in Cd concentration caused loss of chlorophylls and carotenoid and membrane damage in both cultivars, but greater membrane damage was observed in 3223 than in 32D99. Depending on Cd accumulation, a significant reduction in dry biomass was observed in both cultivars at all Cd concentrations. The accumulation of Cd was higher in roots than in leaves for both cultivars. Nevertheless, cultivar 3223 transferred more Cd from roots to leaves than did 32D99. On the other hand, our results suggest that there were similar responses in SOD, APX and GR activities with increasing Cd concentrations for both cultivars. However, POD activity significantly increased at highly toxic Cd levels in 32D99. This result may be regarded as an indication of better tolerance of the Z. mays L. cultivar 32D99 to Cd contamination.  相似文献

Nannochloropsis oculata (strain CCAP 849/1) was sampled at least every 12 h over a 26-d period of batch culture growth in a 12 h/12 h light/dark illumination cycle. Exponential cell-specific growth rate was 0.5 d–1. Cell division occurred during the dark phase, while ammonium uptake, pigment synthesis and cell volume increase occurred mainly during the light. Stationary phase cells were on average larger that the largest exponentially growing cells. The lag phase prior to cell division was short with the C/N ratio returning to 6.25 (from 28) within 2 d of refeeding with ammonium. Significant Chl.a synthesis commenced after this period; net synthesis of Chl.a ceased on exhaustion of the N-source with a 40% fall in levels by the end of the stationary phase. Levels of carotenoids per cell also declined during N-deprivation although per ml of culture levels remained constant. Ammonium-refeeding of N-deprived cells resulted in a very rapid rise in glutamine (Gln) and very high ratios of glutamine/glutamate (Gln/Glu peaking at 35 within 1 h); peak Gln/Glu was lower in cells refed in the dark or after a shorter period of N-deprivation. The major intracellular amino acids during exponential phase were Glu, Gln, alanine and arginine, but on exhaustion of the N-source, levels of Gln fell rapidly (Gln/Glu falling to below 0.1 from 0.5–0.9 in the light and 0.3 in darkness during exponential growth). During N-deprivation tyrosine accumulated within the cells. We have previously reported that aseptically cultured mesophyll protoplasts of Vigna divide rapidly and regenerate into complete plants, while mesophyll protoplasts of Avena divide only sporadically and senesce rapidly after isolation. We measured polyamine titers in such cultures of Vigna and Avena, to study possible correlations between polyamines and cellular behavior. We also deliberately altered polyamine titer by the use of selective inhibitors of polyamine biosynthesis, noting the effects on internal polyamine titer, cell division activity and regenerative events.In Vigna cultures, levels of free and bound putrescine and spermidine increased dramatically as cell division and differentiation progressed. The increase in bound polyamines was largest in embryoid-forming callus tissue while free polyamine titer was highest in root-forming callus. In Avena cultures, the levels of total polyamines decreased as the protoplast senesced. The presence of the inhibitors -difluoromethyl-arginine (specific inhibitor of arginine decarboxylase) and dicyclohexylamine (inhibitor of spermidine synthase) reduced cell division and organogenesis in Vigna cultures. Addition of low concentration of polyamines to such cultures containing inhibitors or removal of inhibitors from the culture medium restored the progress of growth and differentiation with concomitant increase in polyamine levels.  相似文献

Entry of the divalent cations Ni2+, Co2+ and Zn2+ into cells of maize ( Zea mays L. cv. Dekalb XL 85) root tissue is accompanied by an acidification of the incubation medium, a decrease in both the pH of the cell sap and the level of malate in the cells, and by an inhibition of dark fixation of CO2. K+, on the contrary, induces only a very low acidification of the incubation medium, does not change either the pH of the cell sap or the malate level in the cells, and induces an increase in CO2 dark fixation. Different mechanisms are postulated for the stimulation of proton extrusion by divalent cations and K+.  相似文献

A total of 280 laboratory colonies of Reticulitermes urbis were raised for 3 years, in order to observe caste differentiation and overall colony growth, and to investigate whether the initial composition (0, 2 or 4 nymphs; 0, 5, 10, 20 or 50 workers) affected colony survival, population growth and other colony dynamics. We also wanted to determine the minimum number of individuals needed to establish a vital colony. As a result, every colony beginning with less than 20 workers died within 60 weeks, while colonies beginning with at least 20 workers survived until the end of the tests, with varied survival rates dependent upon the initial size and the time of formation (summer or autumn). The number of nymphs did not affect colony survival and colony growth. Reproductives developed from nymphs within 2 weeks and later from workers. Many of the reproductives derived from nymphs were killed by workers. The majority of the colonies contained two reproductives 3 years after initiating the tests. In addition, a few individual workers were still alive at the end of the 3-year tests. At the end of the tests, the proportion of soldiers ranged anywhere from 5.23 to 7.69% of the total population. The number of viable “juveniles” from each colony was relatively low throughout the tests and the overall population growth was not sufficient enough to replace dead workers or increase the colony size considerably. These results indicate that the potential to establish viable and sustainable colonies for groups of R. urbis composed of 5–50 workers is very low.  相似文献

Application of a peat-based powder inoculant of Azospirillum brasilense, as well as a granular inoculant (each containing 0.5–1.0×107 Azospirillum/g moist peat), in the seed furrows of Zea mays resulted in significantly increased yields (11 to 14%) in light soils at low rates of N fertilization. In general, there was no effect of inoculation on plant yields in heavier soils nor when N fertilization was high. Pre-emergence application of granular inoculant and inoculation associated with irrigation were more efficient in increasing yield than inoculation post-emergence or seed coating.E. Water-stressed maize (Zea mays L.) leaves showed a large decrease in leaf conductance during photosynthesis. Net CO2 uptake and evaporation declined fast at mild stress (=–0.6 to –1.0 MPa) and slower at more severe stress (=–1.0 to -1.2 MPa), whereas the CO2 concentration in the intercellular spaces (Ci) did not drop to the CO2 compensation point. The activities of the enzymes of photosynthetic carbon metabolism tested in this study dropped by approx. 30% at =-1.2 MPa. Glutamine synthetase activity was unaffected by water stress, whereas the activity of nitrate reductase was almost completely inhibited. The decline of enzyme activities in relation to was correlated with a concomitant decrease in the content of total soluble protein of the stressed leaves. The total leaf pools of malate, pyruvate and oxaloacetate decreased almost linearly in relation to , thus obviously contradicting the almost constant Ci. Maize ( Zea mays L. hybrid ZP-704) and black pine ( Pinus nigra Arn.) were grown for five days at low fluence rate (0.4–4.0, μmol m–2 s−1) in blue or red light. Compared to red light of the same fluence rate, blue light effects in maize were repressive for the accumulation of Chita, b , carotenoids and light-harvesting complex-2 (LHC-2) proteins. The maximal reduction of proteins bound to the light-harvesting complex of photosystem 2 and pigments was attained at different fluence rate levels. In black pine, blue light compared to the red of the same fluence rate level either activated or reduced accumulation of pigments and LHC proteins, the effect being dependent on its fluence rate level. At fluence less than 3.0 μmol m−2 s−1 blue light was more efficient for the synthesis of Chi a, b and carotenoids, hut for LHC-2 complexes, fluence rates between 0.4 and 1.5 [μmol m−2 s−1 were more effective. In pine the effects of the two lights on the accumulation of pigments and LHC proteins were demonstrated separately and were dependent on fluence rate level. This suggests irradianoe-controlled activation/deactivation of the photoreceptor at the level of the cell.  相似文献

Characteristic steady-state mRNA level oscillations were monitored for the chlorophyll a/b-binding (cab) protein in tomato plants grown under the natural day/night (light/dark) regime as well as under constant environmental conditions. This typical expression pattern was altered when plants were transferred to different light/dark regimes. For example, by shifting the light phase by six hours, a change of the time points of maximum and minimum of expression level was monitored, while the principal oscillation pattern remained the same. It appeared that the transition from dark to light is involved in determining the time points of minima and maxima of mRNA accumulation.After exposing tomato plants to an abnormal light/dark periodicity (e.g. six hours of alternating light/dark) an altered oscillation pattern was determined: within 24 hours two maxima of cab mRNA levels were detected. However, this entrained abnormal rhythm was not manifested at the molecular level and the circadian pattern reappeared under constant environmental conditions (e.g. darkness). This result favours the hypothesis that the oscillation pattern of the cab mRNA in tomato plants is not only endogenous but also hereditary.  相似文献

Summary In Nassarius reticulatus the nuclei and nucleoli undergo important morphological changes from the zygote to the 16-cell stage. In the zygote and in the trefoil (2-cell) and 4-cell stage, several agranular, fibrillar nucleolus-like bodies 1 m diameter are present in the interphase nucleus. Granular nucleoli first appear at the 8-cell stage. These nucleoli have fibrillar and granular regions. The granular regions are made up predominantly of ribosome-like osmiophilic granules. From the 16 and 32-cell stage onwards, the one or two spherical nucleoli of each nucleus measure 2.5 m in diameter and show a concentric organization with a very dense central region surrounded by a broad peripheral zone containing numerous granules, possibly of ribonucleoprotein. At the same time the number of ribosome-like particles increase in the karyoplasm and that of ribosomes in the cytoplasm. These findings are surprising because in eggs with radial cleavage, which have been subjected to more detailed analysis, the first granular nucleoli appear after the end of the cleavage, at the blastula or gastrula stage. The early appearance of granular nucleoli which seem to be characteristic of several eggs with spiral cleavage is discussed in connection with biochemical data on RNA-synthesis.This investigation was supported by the Deutsche Forschungsgemeinschaft and the Stiftung VolkswagenwerkWe wish to thank Mrs. C. Nannochloropsis gaditana was grown in semicontinuous culture with a circadian light:dark cycle in a flat-panel photobioreactor. The microalga had a maximal protein content (3 pg cell–1) after 6 h light and then only storage compounds were accumulated that were consumed during the dark phase. Carbohydrates reached their maximum value after 8 h (0.8 pg cell–1) and lipids after 12 h light (2.5 pg cell–1). The results demonstrated that young or adult microalgae might be obtained according to the time of day.  相似文献
