肝细胞性肝癌(HCC)是具有高度侵袭性和丰富血供的恶性肿瘤,极易肝内外转移,死亡率高。叉头框蛋白质M1(forkheadbox protein M1,Fox M1)作为一种致癌因子,在肝癌的增殖、血管生成和转移等病理生理过程中发挥着重要作用。上皮-间充质转换(epithelial-mesenchymal transition,EMT)是指具有极性的上皮细胞转换为具有运动能力的间充质细胞并获得侵袭和迁移能力的生物学过程。目前研究发现,EMT在肝癌的侵袭和转移过程中扮演着重要角色。本文旨在对Fox M1通过作用于EMT在肝癌转移中的作用进行简要综述。  相似文献

肺癌是具有高度侵袭性的一类恶性肿瘤,传统以手术结合放化疗的治疗手段对肺癌的治疗效果不佳,5年生存率仅为20%~30%,其中重要原因之一是肺癌容易转移与复发。在与肺癌长期斗争的过程中,人们发现肺癌组织中存在一小群肺癌干细胞(lung cancer stem cells,LCSCs),它们是肺癌难以治愈和复发的根本原因。从LCSCs的发现、相关的生物学特性和信号转导通路,以及LCSCs与表皮-间质转化(epithelialmesenchymal transition,EMT)之间的关系等方面对LSCSs进行综述。  相似文献

过去近半个世纪的研究表明,间充质干细胞(MSCs)作为肿瘤微环境中细胞成份的重要成员之一,它在肿瘤细胞生长增殖、凋亡调控、侵袭性生长和转移、血管新生、能量代谢以及治疗抗性等诸多恶性生物学特性中均发挥重要的生物学作用,其相关的分子机制也逐步被揭示;相应地,基于这些发现的基因工程修饰和物理(化学)预处理的MSCs在特定肿瘤综合治疗中临床应用研究也取得了令人振奋的结果。本文就目前MSCs在恶性肿瘤发生发展中所起的生物学作用及其分子机制研究的相关进展作一综述,并就未来MSCs相关的新研究方向及其肿瘤综合治疗中可能的临床应用作一简单的展望。  相似文献

近年来,肿瘤干细胞(cancer stem cell,CSC)学说研究认为CSC与肿瘤发生、发展、转移和复发关系极为密切。研究还发现CSC具有明显的异质性,即CSC可分为增生、耐药、侵袭和转移等行为不同的亚群细胞,其中具有转移生物学特性的CSC亚群细胞称为肿瘤转移干细胞(migrating cancer stem cell,MCSC)。目前认为,上皮-间质转变、趋化因子和靶器官微环境可能在肿瘤转移过程中起着重要作用。针对MCSC及其相关机制的靶向治疗有望能更有效地遏制肿瘤的转移。  相似文献

非小细胞肺癌(non-small cell lung cancer,NSCLC)是全球发病率最高的恶性肿瘤之一,大多数患者在确诊时已是中晚期,预后极差。外泌体是细胞主动分泌的直径为40~100 nm的膜小体,包含丰富的蛋白质、microRNAs(miRNAs)等物质,参与细胞间的物质交换和信息交流。非小细胞肺癌源性的外泌体miRNA参与调节肿瘤细胞的发生发展、侵袭及转移等过程,在肿瘤细胞的生理、病理过程中扮演了重要角色。本文将系统阐述外泌体的生物学特性、生物学功能以及外泌体miR-145和miR-21在调控非小细胞肺癌进展、早期诊断、治疗靶点筛选及预后中的作用,为外泌体miRNA基础研究与临床应用提供参考。  相似文献

趋磁细菌(Magnetotactic bacteria,MTB)是一种可在外磁场作用下沿磁场线定向运动的革兰氏阴性菌,其体内的磁小体(Bacterial magnetosomes,BMs)是由MTB经过生物矿化合成的。BMs在MTB体内成链状排列,由外层膜和内部磁铁矿晶体构成。BMs具有大小均一,单磁畴,大的比表面积,良好的生物相容性,超顺磁性等特点被广泛应用于医疗领域。目前,基于MTB的改造方法相对较少且主要偏向于通过改变BMs的形态、组成等进一步达到改造MTB的目的。BMs的功能化策略相对较多,主要分为化学修饰和生物修饰两种。综述了MTB和BMs的基本特性及筛选技术,并着重介绍了MTB的改造方法和BMs的功能化策略,最后讨论了MTB改造和BMs功能化在实际应用中的意义以及存在的问题。展望了MTB改造和BMs功能化的发展前景,可能面临的机遇及挑战,以期能够进一步促进MTB和BMs在实际中的应用。  相似文献

肿瘤的侵袭转移是恶性肿瘤患者死亡的主要原因。目前对胃癌侵袭转移的分子机制尚未查明。胃癌侵袭转移过程中涉及许多特殊基因,包括促进转移的转移基因和控制转移的转移抑制基因。转移基因有:细胞粘附分子(cAM)基因、基质金属蛋白酶(MMPs)基因、肝素酶(heparanase)基因等;转移抑制基因有:nm23、组织基质金属蛋白酶抑制因子(TIMPs)基因等。本文对胃癌侵袭转移相关基因及其产物的结构、功能及其在侵袭转移过程中作用的研究进展作一综述。  相似文献

脑胶质瘤是侵袭能力极强的恶性肿瘤,目前治疗策略主要是手术治疗辅助放射治疗、化学治疗和免疫治疗等多种治疗,但疗效有限。肾上腺素能受体(adrenergic receptor, AR)是体内一种重要的应激激素受体,AR通过激活下游不同信号转导通路广泛参与调节多种肿瘤的发生与发展进程。最近研究显示,AR在脑胶质瘤细胞以及组织中表达失调,在脑胶质瘤的发生、侵袭和转移等一系列生物学行为中发挥着重要作用。本文对近年来AR在脑胶质瘤发生和发展中的研究进展进行综述,为以AR为靶点的脑胶质瘤的预防和治疗提供理论依据。  相似文献

目的研究奥沙利铂对人子宫内膜癌细胞HEC-1A侵袭转移的影响。方法体外稳定培养HEC-1A细胞株系,用不同浓度奥沙利铂(40、80、160μg/ml)给药72h后,采用Transwell法测定奥沙利铂对HEC-1A侵袭能力的影响,重组基底膜试验测定奥沙利铂对HEC-1A粘附能力的影响,划痕试验检测奥沙利铂对HEC-1A迁移能力的影响。结果与未处理对照组比较,奥沙利铂(40、80、160μg/ml)明显抑制HEC-1A侵袭性,提高侵袭抑制率(P0.01),显著降低肿瘤细胞的迁移能力(P0.01),降低HEC-1A粘附程度(P0.01)。结论奥沙利铂有效抑制子宫内膜癌细胞继发性侵袭及转移,从而发挥抗癌作用。  相似文献

An investigation of the invasiveness of avian neural crest cells and neural crest-derived melanocytes through a human amniotic basement membrane (BM) was undertaken. Avian neural tube explants or derived melanocyte populations were seeded directly onto BMs in membrane invasion culture system (MICS) chambers for periods of 24, 48, and 72 h. In 36 experimental trials for each group, neither neural crest nor neural crest-derived melanocytes were observed to have invaded the BMs. In concert with these studies, coculturing of B16F10 murine melanoma cells with avian neural crest-derived melanocytes was performed in MICS chambers. Under these experimental conditions, the neural crest-derived melanocytes were able to successfully invade the BMs and to a greater extent than the B16F10 tumor cells. These data suggest that neural crest cells and neural crest-derived melanocytes do not have the ability to invade the BM alone; however, they can be induced to be invasive when cocultured in the presence of B16F10 cells. Alternatively, the B16F10 cells may create weaknesses within the BM that facilitate migration of the pigmented crest cells.  相似文献

目的:探讨甲状腺功能亢进(甲亢)、甲状腺功能减低(甲低)与肝纤维化指标的关系及其可能的机制。方法:采用放射免疫分析法(R1A)检测57例甲亢患者、43例甲低患者、39例甲亢治疗后甲状腺激素正常者和50例健康成人的血清Ⅳ型胶原(1V-C)、Ⅲ型前胶原(PCⅢ)、透明质酸(HA)、层粘连蛋白(LN)、T3、T4、FT3、FT4、TSH、TGA、TMA含量。结果:甲亢患者组血清中1V-C、PCⅢ含量比正常对照组及甲低患者组显著性增高(P〈0.05);治疗后甲状腺激素下降,1V-C、PCⅢ含量也随之下降(与治疗前比较P〈0.01);HA、LN在四组中无显著性差异(P〉0.05),在甲亢治疗前后无显著性差异(P〉0.05)。各项肝纤维化指标与TGA、TMA的阳性率无关。结论:甲亢患者可有不同程度的肝功能损害,血清中甲状腺激素水平增高时,1V-C、PCⅢ水平也增高,在用1V-C、PCⅢ判断肝纤维化时应注意有无甲状腺疾病特别是甲亢。未发现HA、LN含量与甲状腺激素水平的关系。  相似文献

The current basement membrane (BM) model proposes a single-layered extracellular matrix (ECM) sheet that is predominantly composed of laminins, collagen IVs and proteoglycans. The present data show that BM proteins and their domains are asymmetrically organized providing human BMs with side-specific properties: A) isolated human BMs roll up in a side-specific pattern, with the epithelial side facing outward and the stromal side inward. The rolling is independent of the curvature of the tissue from which the BMs were isolated. B) The epithelial side of BMs is twice as stiff as the stromal side, and C) epithelial cells adhere to the epithelial side of BMs only. Side-selective cell adhesion was also confirmed for BMs from mice and from chick embryos. We propose that the bi-functional organization of BMs is an inherent property of BMs and helps build the basic tissue architecture of metazoans with alternating epithelial and connective tissue layers.  相似文献

Nidogen 1 and 2 are ubiquitous basement membrane (BM) components. They show a divergent expression pattern in certain adult tissues with a prominent localization of nidogen 2 in blood vessel BMs. Deletion of either nidogen 1 or 2 in mice had no effect on BM formation, suggesting complementary functions. However, studies in these mice revealed isoform-specific functions with nidogen 1-deficient mice showing neurological abnormalities and wound-healing defects not seen in the absence of nidogen 2. To investigate this further nidogen 1- or 2-deficient mice were intravenously injected with B16 murine melanoma cells, and lung metastasis was analyzed. The authors could show that loss of nidogen 2, but not of nidogen 1, significantly promotes lung metastasis of melanoma cells. Histological and ultrastructural analysis of nidogen 1- and 2-deficient lungs did not reveal differences in morphology and ultrastructure of BMs, including vessel BMs. Furthermore, deposition and distribution of the major BM components were indistinguishable between the two mouse strains. Taken together, these results suggest that absence of nidogen 2 might result in subtle changes of endothelial BMs in the lung, which would allow faster passage of tumor cells through these BMs, leading to a higher metastasis rate and more larger tumors.  相似文献

基底膜是一种特化的细胞外基质,是肿瘤转移过程中必须穿越的物理屏障。基底膜的组成成分通过和细胞表面受体整合素相互作用,在调节肿瘤转移的过程中发挥了重要作用。另一方面,肿瘤细胞通过分泌基质降解酶类破坏基底膜的组织结构,同时调节细胞外基质受体整合素的表达,为穿过:基底膜和在靶器官粘附、增殖创造有利条件。了解细胞和基底膜的相互作用可以为抗转移药物的研发提供新的策略。  相似文献

Glioma cell infiltration of brain tissue often occurs along the basement membrane (BM) of blood vessels. In the present study we have investigated the role of laminins, major structural components of BMs and strong promoters of cell migration. Immunohistochemical studies of glioma tumor tissue demonstrated expression of alpha2-, alpha3-, alpha4- and alpha5-, but not alpha1-, laminins by the tumor vasculature. In functional assays, alpha3 (Lm-332/laminin-5)- and alpha5 (Lm-511/laminin-10)-laminins strongly promoted migration of all glioma cell lines tested. alpha1-Laminin (Lm-111/laminin-1) displayed lower activity, whereas alpha2 (Lm-211/laminin-2)- and alpha4 (Lm-411/laminin-8)-laminins were practically inactive. Global integrin phenotyping identified alpha3beta1 as the most abundant integrin in all the glioma cell lines, and this laminin-binding integrin exclusively or largely mediate the cell migration. Moreover, pretreatment of U251 glioma cells with blocking antibodies to alpha3beta1 integrin followed by intracerebral injection into nude mice inhibited invasion of the tumor cells into the brain tissue. The cell lines secreted Lm-211, Lm-411 and Lm-511, at different ratios. The results indicate that glioma cells secrete alpha2-, alpha4- and alpha5-laminins and that alpha3- and alpha5-laminins, found in brain vasculature, selectively promote glioma cell migration. They identify alpha3beta1 as the predominant integrin and laminin receptor in glioma cells, and as a brain invasion-mediating integrin.  相似文献

During tumor progression, the extracellular matrix (ECM) and particularly the basement membrane (BM) appear to be dynamic structures that are not only degraded but also deposited around tumor clusters. In this study we examined by immunohistochemistry the localization of three chains of Type IV collagen (alpha1, alpha3 and alpha5), Type VII collagen, and laminin 5 at different stages of bronchopulmonary cancers. In normal tissues, alpha1(IV) chain was detected in all BMs (bronchial, vascular, alveolar, and glandular), alpha5(IV) chain was present only in vascular BM, and laminin 5 and Type VII collagen were co-localized in bronchial and glandular BMs, whereas alpha3(IV) immunolabeling was totally absent from normal bronchi. In well-differentiated carcinomas, alpha3(IV) chain staining was found in some neosynthetized BMs interfacing the tumor cell and the stromal compartment, contrasting with the total absence of labeling in normal tissues. alpha1(IV) chain showed strong reactivity in all BM. Laminin 5 and Type VII collagen were also detected in neosynthetized BM. In poorly differentiated invasive cancers, alpha3(IV) chain and Type VII collagen were not found, whereas laminin 5 and alpha1(IV) chain persisted. The most important modifications in BM composition during tumor progression therefore appear to be the appearance of the alpha3 (IV) chain in well-differentiated carcinomas and its subsequent disappearance in poorly differentiated carcinomas, together with the loss of type VII collagen. alpha5(IV) chain distribution was restricted in vascular BM of well- and poorly differentiated carcinomas. These results show that the composition of BM is modified during the progression of bronchopulmonary tumor, emphasizing that the BM represents a dynamic element in tumor progression and has an important role in tumor cell invasiveness.  相似文献

Scinderin是一种依赖Ca2+的肌动蛋白丝(F-actin)切割蛋白,在细胞分泌过程中发挥着重要作用。目前,针对scinderin在人类疾病尤其是肿瘤中的生物学功能研究报道的并不多。该实验通过构建scinderin—shRNA慢病毒载体并感染人胃癌细胞株SGC-7901,于荧光显微镜下观测感染效率,利用RT-qPCR和Western blot实验证实scinderin的沉默效果。运用实时细胞分析4K(RTCA)检测细胞的增殖能力,流式细胞术检测细胞周期变化,Transwell小室检测细胞的迁移能力。结果显示,将构建好的病毒载体成功转入了胃癌细胞SGC-7901。感染scinderin—shRNA病毒载体后,scinderin的mRNA和蛋白质表达水平均受到不同程度的抑制(P〈0.01),细胞的增殖和迁移能力均显著降低(P〈0.05),细胞周期阻滞在G2/M期。该研究表明,胃癌细胞SGC-7901中scinderin低表达能有效抑制细胞的增殖和转移能力,这也为scinderin在胃癌演化过程中的机制研究奠定了实验基础。  相似文献

The ability to metastasize represents the most important characteristic of malignant tumors. The biological details of the metastatic process remain somewhat unknown, due to difficulties in studying tumor cell behaviour with high spatial and temporal resolution in vivo. Several lines of evidence involve transglutaminases (TGs) in the key stages of tumor progression cascade, even though the molecular mechanisms remain controversial. TG expression and activity display a different role in the primary tumor or in metastatic cells. In fact, TG expression is low in the primary tumor mass, but augmented when cells acquire the metastatic phenotype. Nevertheless, in other cases, the use of inducers of TG transamidating activity seems to contrast tumor cell plasticity, migration and invasion. In the following review, the function of TGs in cancer cell migration into the extracellular matrix, adhesion to the capillary endothelium and its basement membrane, invasion and angiogenesis is discussed.  相似文献
