Fully expanded leaves of Erythroxylum coca var. coca Lam. (Erythroxylaceae)35-70-d-old were harvested from 21-month-old plants grown undergreenhouse conditions. Each harvested leaf was placed on a plexiglasssilhouette according to its dimensions and divided into fourprimary sections (petiole, base, mid and anterior) and threesubsections (lamina periphery, false mid-rib, and true mid-rib)to determine the distribution and content of hygrine, cuscohygrine,trans-cinnamoylcocaine, cis -cinnamoylcocaine, tropacocaine,tropinone, methyl ecgonine and cocaine. The leaf sub-sectionsand petiole were extracted and analysed for alkaloid content(%) by HPLC (cocaine alkaloid only) gas chromatography and GC/MS.Cocaine, methyl ecgonine and hygrine were highest in the laminaperiphery with a content of 0·48, 0·46 and 0·32%,respectively. Trans-cinnamoylcocaine was pre-eminent of thecinnamoylcocaines and was most abundant in the petiole at acontent of 0·24%. Larvae of the lymantriid moth Eloria noyesi, which are obligate feeders on Erythroxylum coca, excrete most of the ingested cocaine as unchanged base. Cocaine, analysed by mass fragmentography, is readily, detectable in the blood of larvae and is presumably sequestered during larval feeding, since it is present in the bodies of adult moths that do not feed on E. coca. Cocaine is an effective feeding deterrent for the ant Monomorium pharaonis when present at a concentration below that found in the leaves of E. coca.  相似文献

Net photosynthesis rate (Pn), stomatal conductance to CO2 andresidual conductance to CO2 were measured in the last six leaves(the sixth or flag leaf and the preceding five leaves) of Triticumaestivum L. cv. Kolibri plants grown in Mediterranean conditions.Recently fully expanded leaves of well-watered plants were alwaysused. Measurements were made at saturating photosynthetic photonflux density, and at ambient CO2 and O2 levels. The specificleaf area, total organic nitrogen content, some anatomical characteristics,and other parameters, were measured on the same leaves usedfor gas exchange experiments. A progressive xeromorphic adaptation in the leaf structure wasobserved with increasing leaf insertion levels. Furthermore,mesophyll cell volume per unit leaf area (Vmes/A) decreasedby 52·6% from the first leaf to the flag leaf. Mesophyllcell area per unit leaf area also decreased, but only by 24·5%.However, nitrogen content per unit mesophyll cell volume increasedby 50·6% from the first leaf to the flag leaf. This increasecould be associated to an observed higher number of chloroplastcross-sections per mm2 of mesophyll cell cross-sectional areain the flag leaf: values of 23000 in the first leaf and 48000in the flag leaf were obtained. Pn per unit leaf area remainedfairly constant at the different insertion levels: values of33·83±0·93 mg dm–2 h–1 and32·32±1·61 mg dm–2 h–1 wereobtained for the first leaf and the flag leaf, respectively.Residual conductance, however, decreased by 18·2% fromthe first leaf to the flag leaf. Stomatal conductance increasedby 41·7%. The steadiness in Pn per unit leaf area across the leaf insertionlevels could be mainly accounted for by an opposing effect betweena decrease in Vmes/A and a more closely packed arrangement ofphotosynthetic apparatus. Lolium temulentum seedlings were grown on a nutrient mediumcontaining NH4NO2 at 0, 0·1, 0·5, 1·0 and4·3 mmoll–1 as the sole N source. Relative andabsolute extension rates, maximal leaf size, duration of extensiongrowth, rate of leaf appearance and plastochron index were determinedfrom the parameters of Richards functions fitted to lengthsof laminae measured at intervals after sowing. The final lengthof leaf I was relatively insensitive to N whereas mean relativeextension rate was increased and duration of growth decreasedwith increasing NH4NO2 concentration. Leaves 2 and 3 enlargedprogressively with N at concentrations up to 1·0 mmoll–1but were unresponsive thereafter. There was no significant correlationbetween final length and mean relative extension rate for leaves1 to 3. Leaves 4 to 6 continued to show increasing length beyond1·0 mmoll–1 N and final length was significantlycorrelated with mean relative extension rate. Increasing N increasedthe rate of leaf appearance by decreasing the duration of leafextension and plastochron. These results are discussed in relationto the control of leaf and N turnover. This study was conducted to test the hypothesis that photoperiodor its rate of change significantly affects the rate of leafappearance (RLA) and final number of leaves (FNL) in wheat,as suggested from several time-of-sowing experiments. Two wheatcultivars (Condor and Thatcher) were sown in the field on 2Sep. 1992 at Melbourne (38°S). Photoperiod was extendedartificially to give five treatments up to terminal spikeletinitiation (TS) viz.: natural photoperiod (rate of change ofphotoperiod = 2 min d-1), two faster rates of change (8·5and 13·3 min d-1) and two constant photoperiods of 14·0and 15·5 h. After TS, the two constant photoperiods wereextended to 15·0 and 16·5 h, respectively, andtreatments were re-randomised, i.e. some plots received differentphotoperiod regimes before and after TS. The rate of leaf appearance maintained strong linear relationshipswith thermal time. It was greater for Condor [0·012-0·013(°C d)-1] than for Thatcher [0·011-0·012 (°Cd)-1] and did not alter during plant development or in responseto the change in photoperiod at TS. Rate of leaf appearanceon the main culm was not influenced by the rate of change ofphotoperiod nor by the average photoperiod. Cultivar and photoperiod significantly affected FNL on the mainculm. Condor produced more leaves than Thatcher under long butnot under short photoperiods. The rate of change of photoperioddid not affect FNL independently of the effect of average photoperiod.Most of the variation in FNL due to photoperiod resulted fromdifferences in duration of leaf initiation. The lack of effects of the photoperiod treatments on RLA contrastwith previous reports of its effects on the rate of phasic developmentfrom seedling emergence to double ridge. Therefore, the numberof visible leaves on the main culm (NL) at double ridge andat TS were not constant. There is controversy as to whether the mature fruit of Erythroxylumcoca var. coca Lam. contains the cocaine alkaloid (benzoylmethylecgonine).In the present study, cocaine was monitored to determine ifit was present in embryos and endosperms of mature fruit ofE. coca var. coca Lam., and if present, the time required forde novo synthesis in imbibing seed. Seeds from mature fruitof E. coca were dissected to separate the embryos from the endosperms.The separated embryos and endosperms were analysed for cocaine.Subsequently, endosperms and embryos from seed imbibed. undera light and dark treatment were separated on days 3, 6, 9, 12and 15 and analysed for cocaine. Cocaine was present in embryos(0.005% of d. wt) and endosperms (0–001% of d. wt) ofmature fruit of E. coca. De novo synthesis of cocaine occurredonly in embryos of seed imbibed under light after day 9 of imbibition. A controlled environment experiment investigated whether thered:far-red (R:FR) ratio of light at the apical bud of the mainstolon could alter plant morphogenesis in clonal cuttings ofwhite clover (Trifolium repens L.) The apical bud included theapical meristem, five to six developing leaf primordia withassociated axillary bud primordia and stipules and the firstemerged folded leaf until development was greater than 0·3on the Carlson scale. Three light regimes were imposed on theapical bud by collimating light from R or FR light-emittingdiodes so that the R:FR ratio of light incident at the apicalbud was set at 0·25, 1·6 or 2·1, withoutsignificantly altering photosynthetically active radiation.The effect of these light regimes on white clover seedling growthwas also tested. At a low R:FR ratio seedling hypocotyl and cotyledon lengthswere significantly longer. However, with the cuttings, the lighttreatments did not alter node appearance rate or internode lengthof the main stolon, petiole length, area of leaves or totalshoot dry matter. There was one significant photomorphogeneticresponse in the cuttings, a delay of 0·5 of a phyllochronin the appearance of branches from axillary buds in the lowR:FR ratio treatment relative to the other treatments. Wherebranch appearance was delayed plants had fewer branches. Thisdifference could be ascribed solely to a delay in branch appearanceas there were no significant treatment effects on either theinitiation of axillary bud primordia within the apical bud,the probability of branching or on the rate of growth of branchesafter appearance. Epidermal Cell Division and the Coordination of Leaf and Tiller Development   总被引:7,自引:2,他引:5  
Initiation and development of grass leaves and tillers are oftendescribed individually with little attention to possible interrelationshipsamong organs. In order to better understand these interrelationships,this research examined epidermal cell division during developmentaltransitions at the apical meristem of tall fescue (Festuca arundinaceaSchreb.). Ten seedlings were harvested each day for a 9-d period,and lengths of main shoot leaves and primary tillers were measured.In addition, numbers and lengths of epidermal cells were determinedfor 0·5 mm segments along the basal 3 mm of each leafand tiller. Primordia development and onset of rapid leaf elongationwere characterized by an increase in the number of cells perepidermal file with mean cell length remaining near 20 µmper cell. After the leaf had lengthened to 1-1·5 mm,cells near the leaf tip ceased dividing and increased in length,at which time leaf elongation rate increased rapidly. Liguleformation, marking the boundary between blade and sheath cells,occurred prior to leaf tip emergence above the whorl of oldersheaths, while the earliest differentiation between blade andsheath cells probably began when leaves were < 1 mm long.Major transitions in leaf and tiller development appeared tobe synchronized among at least three adjacent nodes. At theoldest node, cessation of cell division in the leaf sheath wasaccompanied by initiation of cell division and elongation inthe associated tiller bud. At the next younger node the ligulewas being initiated, while at the youngest node cell divisioncommenced in the leaf primordium, as elongation of a new leafblade began. Leaf sections of Browallia viscosa and B. speciosa were placedon Murashige and Skoog (1962) salts and vitamins medium (MS)containing auxins and cytokinins, singly or in combination,to elicit morphogenetic responses. B. viscosa developed extensiveroots in 4 weeks on media supplemented with indolebutyric acid(IBA), indol-3-yl acetic acid (IAA) or naphthalene acetic acid(NAA) (0·01, 0·1, 1·0, 5·0 and 10·0mg–1), but with 2, 4-D (0·1 mg–1) only lightyellow friable callus was obtained. Shoot initiation and elongationoccurred consistently in 4–6 weeks on leaf sections inthe presence of 6---dimethylallyl amino purine (2iP). Similarly,shoot regeneration from leaf-derived callus, initiated and sub-culturedon MS + benzyladenine (BA) + NAA only induced callus on leafexplants of both species. B. speciosa did not respond exceptfor moderate and prolific callus formation on MS + BA + NAAand Uchimiya and Murashige (1974) media respectively. Leaf age effects on the leaf conductance to water vapour diffusionof the adaxial and abaxial leaf surfaces were measured in themorning and in the afternoon on 17 different plantain and banana(Musa spp.) genotypes. The irradiance levels increased three-foldwhile leaf to air vapour pressure deficit levels increased two-to four-fold from morning to afternoon during the sampling periodin a field site located in the humid forest-moist savanna transitionzone of Nigeria. Conductance values were reduced in older, andsenescing leaves relative to the young and mature leaves bothin the morning and in the afternoon. Conductances were higherfor the abaxial leaf surfaces than the adaxial surface and higherin the afternoon than in the morning, with some genotypic differences.Lower values of leaf conductance to water vapour in the afternoonunder a short dry spell was sufficiently variable (P 0·05)among the test genotypes to indicate potential adaptation totransient dry conditions. Differential and relative leaf conductanceadjustments were noted among genotypes experiencing a shortdry spell versus non-limiting soil moisture conditions. Significantgenotypic differences were observed for leaf conductance amongthe 17 genotypes during the afternoon on the lower leaf surfaceof younger leaves. Longitudinal elongation contributes most to leaf area expansionof grasses and its rate is known to be strongly affected byN. Our objective was to determine the effect of two N regimes(N0and N+) on the gradient of leaf tissue formation in meristemsof two contrasting tall fescue (Festuca arundinacea Schreb.)genotypes. Proportions of epidermal, mesophyll and vasculartissue as well as intercellular air space were determined throughoutthe base of actively elongating leaves. The area of leaf transversesections nearly doubled between the ligule and the distal endof the growth zone (about 30 mm), and was mainly associatedwith lateral epidermal and mesophyll cell division in the proximal5.0–7.5 mm. Further increase in transverse area was dueto the formation of intercellular airspace and transverse expansionof epidermal cells. Depending on genotype and N treatment themesophyll, epidermis, vascular bundles and air space comprised45–54%, 20–28%, 6–9%, and 17–21%, respectively,of transverse leaf area in the distal part of the growth zone.After a slight increase close to the leaf base, the area ofvascular tissue remained constant throughout the growth zone.The proportion of air space to mesophyll space was higher atN0than at N+ because mesophyll area was enhanced by N+ to agreater degree than by N0. In the genotype with slow leaf elongation,the increase in cross-sectional leaf area was due to an increasein both leaf width and leaf thickness. In the genotype whichhad faster leaf elongation and wider leaves, only leaf thicknesswas enhanced by N+. Studies of in vivo nitrate reductase activity (NRA) in Eichhorniacrassipes showed that adding 5% isopropanol gave highest NRAin all parts of the plant, but it was also necessary to flushthe assay media containing leaves with N1 to obtain the maximalactivity. There were differences in the pH optima for NRA, theroot optimum being 6·5 leaf 7·5–8·0,and the petiole showing a major peak at 6·5 and a minorpeak at 7·.5–8·0. Electrical impedance spectra (100 Hz–800 kHz) were measuredin leaves of Peperomia obtusifolia L. (a succulent) and Brassicaoleracea L. (cabbage). By measuring impedances at three or moreinter-electrode distances in a single leaf, electrode impedanceand specific tissue impedance were separated. Analysis of impedance data from B. oleracea leaves in relationto an equivalent circuit model showed that leaf developmentwas accompanied by increases in extracellular resistance, cytoplasmicresistance and vacuole interior resistance, together with decreasesin plasma membrane capacitance and tonoplast capacitance. AfterB. oleracea leaves were subjected to a –6 °C freeze-thawstress, extracellular resistance, cytoplasmic resistance andvacuole interior resistance decreased, but plasma membrane capacitanceand tonoplast capacitance did not change. These results indicatethat useful measurements of leaf parameters can be obtainedby this technique. Examination of the electrode impedance spectrum showed thatelectrode insertion produced a damaged collar, 0·4–0·5mm wide, around the electrode. The effect of tetraploidy on leaf characteristics and net gasexchange was studied in diploid (2x ) and autotetraploid (4x) ‘Valencia’ sweet orange (Citrus sinensis (L.)Osb.) and ‘Femminello’ lemon (Citrus limon (L.)Burm. f.) leaves. Comparisons between ploidy levels were madeunder high irradiance (I) in a growth chamber or low total Iin a glasshouse. Tetraploids of both species had thicker leaves,larger mesophyll cell volume and lower light transmittance thandiploids regardless of growth I. Mesophyll surface area perunit leaf area of 2x leaves was 5–15% greater than on4x leaves. Leaf thickness and mesophyll cell volume were greaterin high I leaves than low I leaves. In high I, average leafarea was similar for 2x and 4x leaves, whereas in low I it was30% greater in 4x than in 2x leaves. Nitrogen and chlorophyllconcentration per cell increased with ploidy level in both growthconditions. The ratio of chlorophyll a:b was 25% greater in2x than in 4x leaves. When net CO2assimilation rate (ACO2) wasbased on leaf area, 4x orange leaves had 24–35% lowerACO2than their diploids. There were no significant differencesin ACO2between 2x and 4x orange or lemon leaves when expressedon a per cell basis. Overall, lower ACO2per unit leaf area oftetraploids was related to increase in leaf thickness, largermesophyll cell volume, the decrease in mesophyll area exposedto internal air spaces, and the lower ratio between cell surfaceto cell volume. Such changes probably increased the resistanceto CO2diffusion to the site of carboyxlation in the chloroplasts. Baldy, P. 1986. Comparison between growth chamber and field-grownZea mays plants for photosynthetic carboxylase activities andother physiological characteristics with respect to leaf position.—J.exp. Bot. 37: 309–314. The lengths and fresh weights of leaves, and the amounts ofchlorophyll and protein per leaf, were higher in maize grownin a controlled environment than for field-grown maize harvestedat a similar stage of growth with the seventh leaf just emerging.However, the fresh weight and chlorophyll per unit leaf areawere not different and more protein was present per Unit leafarea in maize cultivated in the field, particularly in the 4thleaf. Except in the seventh leaf the youngest present, the PEP carboxylaseactivity was 2-fold higher in field-grown maize than in plantsfrom the controlled environment. The RuBP carboxylase activitywas not significantly affected by growth conditions. The maximumactivities of the carboxylases were found in the 5th leaf formaize grown in the controlled environment and the 4th leaf forfield maize; in these two leaves the ratios of the PEP: RuBPcarboxylasc activities were 3·0 and 4·4 respectively. The results are used to justify the choice of the fully expanded4th leaf of l9-d-old plants grown in a controlled environmentfor studies of the enzymes involved in the photosynthetic carbonmetabolism of this C4 plant. It was confirmed that the leaves of Acer platanoides containan antifungal inhibitory substance. Low concentrations of sterilecold water extracts inhibited the germination of the sporesof Cladosporium herbarum (three isolates), Cladosporium sphaerospermumand Cylindrocarbon radiclcola. In the concentration range 0·06–0·125per cent (w/v) of leaf material the inhibitory response wasdemonstrated to increase linearly as the concentration of leafmaterial increased logarithmically. Inhibitory activity wasfound in leaf samples collected during a period from July toOctober but activity had disappeared from leaves collected inthe following January. The inhibitory activity was located intwo components of the water extract by bioassay tests followingether extraction and separation by chromatography. One of theactive components has been identified as gallic acid by gaschromatography. Gallic acid has also been detected in dew collectedfrom leaf surfaces where it is suggested that it may play animportant part in the colonization of the leaves by fungi.  相似文献

The dried leaves of 31 species and two varieties of the genusErythroxylum were quantitatively analysed for cocaine and bothcis- and trans-cinnamoylocaine, using a stable-isotope dilutionmethod and gas chromatography-mass spectrometry selected-ionmonitoring. Both cocaine and cinnamoylcocaine were detectedin all four varieties of cultivated coca (E. coca var. coca,E. coca var. ipadu, E. novogranatense var. novogranatense andE. novogranatense var.truxillense). Cinnamoylcocaines (cis andtrans) were found in much higher concentrations in both varietiesof E. novogranatense than in either variety of E. coca. Amazoniancoca (E. coca var. ipadu) contained consistently lower cocainelevels than the montane variety (E. coca var. coca). Twenty-ninewild species of Erythroxylum, selected to represent morphological,ecological and taxonomic diversity, were analysed for the samealkaloids; cocaine was detected in 13 neotropical species, representingfive sections of the genus. No cocaine was detected in the OldWorld species. Two wild species from Venezuela, E. recurrensand E. steyermarkii, contained cocaine levels comparable tothose found in the commercially cultivated species.
