A method of analysis for comparing the variability of two samples drawn from two populations has been developed. The method is also suitable for the nonnumeric form of data. A test based on ordered observations for testing the null hypothesis of equality of two variances has been given. The test statistic is a function of the sum of ranks assigned to smaller size sample. Ranking procedure has been modified to depict the variability in the data by the sum of ranks. The null distribution of the test-statistic has been worked out for small samples and it turns out to be chi-square distribution for large samples. The analytical procedure has been explained by a numerical example on the productivity and production of rice and wheat in India from 1950–51 to 1983–84.  相似文献

The spermatozoon of the polychaete Tomopteris helgolandica is of an aberrant type with two flagella, each measuring about 40μm. The nucleus is roughly conical and weakly bent. At the anterior end it is rounded and covered only by the nuclear and plasma membranes. Membraneous, electron-dense structures are applied laterally to the nucleus. These structures may have a helical arrangement. The middle piece contains about ten mitochondria, two centrioles, and two centriolar satellite complexes. The centriolar regions are connected with the posterior part of the nucleus. The axonemes of the two tail flagella lack the usual central complex with central tubules, radial spokes, or related structures. No arms seem to be present on the A tubules of the doublets. In the middle piece the tail flagella are surrounded by invaginations of the plasma membrane forming flagellar canals. The sperm has a bilateral symmetry whereas the primitive sperm has a radial symmetry. The occurrence of two tail flagella in this spermatozoon has no phylogenetical connection with biflagellate spermatozoa in other animal groups. A series of mutations has resulted in the development of two flagella emerging from the two centrioles, the lack of a central complex in the axoneme, and the lack of a typical acrosome. In the Polychaeta, sperm structure is generally more related to function that to phylogenetics. During swimming the spermatozoon of Tomopteris rotates around its longitudinal axis.  相似文献

A variant (LeuRS-A) of Escherichia coli leucyl-tRNA synthetase (LeuRS) carrying a 40-residue duplication in its connective peptide 1 (CP1) has a 3-fold lower specificity for than for, whereas wild-type LeuRS has the same specificity for these two isoacceptors. The replacement of the acceptor stem of with yields a chimeric tRNA(Leu) for which wild-type LeuRS has the same specificity as it does for the two normal isoacceptors mentioned, but for which LeuRS-A has a reduced specificity similar to that for, indicating a difference between these two acceptor stems. LeuRS-A is slightly less stable than the native enzyme. Wild-type LeuRS and LeuRS-A have almost same K(d) value for their interaction with as determined by fluorescence quenching. No difference was detected between these two proteins by CD and fluorescence spectroscopy. These results show that LeuRS-A can discriminate between the two isoacceptors of tRNA(Leu).  相似文献

The Johnston's organ of the female mosquito, Aedes aegypti, has only three types of scolopidia: types A, B, and C. It lacks the type D scolopidium of the male's organ. The basic structure and the location of each type in the female are similar to the counterparts in the male's organ. A single scolopidium is composed of a scolopale cell, an envelope cell, a long cap, and a third sheath, in addition to the two electron-dense scolopales produced inside the cytoplasm of two satellite cells. Each scolopidium has either two (type A) or three (type B) sensory cells. A type C scolopidium, mononematic in contrast to the amphinematic types A and B scolopidia, has two sensory cells, a microtubular cap cell, two microtubular accessory cells, and a scolopale cell with an intracellular scolopale. Even though the female Johnston's organ has all the components of the male's organ except for the single type D scolopidium, the female's organ shows relatively poorer organization and development. The female has a smaller and thinner basal plate, shorter and thicker prongs, fewer type A sensory cells, and a shorter flagellar flange, in addition to the overall smaller size of the pedicel.The probable function of each scolopidial type is discussed, especially in connexion with the probable identification of a single auditory sensillum in the male.  相似文献

Hemocyanin has been used as a protein marker for Crustacea speciation. The specificity of the hemocyanin fractions of two Idotea species has been confirmed by mono and bidirection electrophoresis. Four protein fractions have been found in the two Idotea species. However, the molecular weight of each Idotea chelipes fraction is slightly higher. The two fractions of high molecular weight are strongly associated. We assumed a molecular weight of 240,000 for I. chelipes and 230,000 for I. balthica basteri. The two other protein fractions are well separated and have a weight of 130,000 and 77,000 in I. chelipes; 120,000 and 75,000 for I. b. basteri. In contrast, the subspeciation of I. chelipes has not been clearly demonstrated by electrophoresis of the hemocyanin.  相似文献

Since the collapse of the Soviet bloc, the term “Europe” has reached a new dimension. The fall of the Berlin Wall opened the East to the West and, in 1989, the European space moved up from two entities symbolically separated by a wall to one territorial unity. But we cannot say that this unity is a political one. This dichotomy is not based on two different political orientations, but on the adhesion or the non-adhesion to economic, political and especially moral principles. Thus two conceptions of the European space are competing. The term “Europe” has two major significations: firstly it refers to the European Union, and secondly to a Great Europe, including the states which have belonged to the former Soviet bloc. How can we define how this Great Europe perceived itself and what are its limits? The objective of this paper is to demonstrate that the two visions of the European space are under competition through two institutions. They convey two representations of this European space and imagine it in two perspectives. Therefore, the collapse of the Soviet bloc has altered in depth the perception of the European space and created a new dichotomy.  相似文献

Structures at steady states have been investigated for an enzyme reaction in a continuous stirred tank reactor that has a random bi-uni reaction mechanism, using the imperfect bifurcation theory via singularities. The analysis has shown that two types of singular points exist. One of these points has the two types of transition states characterized by the hysteresis and double-limit points. It is obtained when the derivative of the steady-state equation with respect to the bifurcation parameter does not vanish. When the derivative vanishes, the other type of singular point is obtained. This point has the two transition states of hysteresis and bifurcation points.  相似文献

Two-dimensional crystalline sheets of Na, K-ATPase were studied in the vitrified, frozen-hydrated state by electron microscopy and image processing. The technique of correlation averaging was used to determine the projected structure. The projection map shows asymmetry between the pair of "alpha beta" protomers comprising a dimer of Na, K-ATPase molecules. The two protomers differ in overall density as well as in shape. One protomer has an oblong shape, whereas the other with higher density has a head and a hook region. Such an asymmetry has not been reported by other laboratories. This asymmetry may either be due to the coexistence of two different conformations of the enzyme in the dimeric form or due to the simultaneous existence of two molecular species of Na, K-ATPase.  相似文献

Summary A gene coding for an endoglucanase from the anaerobic cellulolytic bacteriumClostridium cellulolyticum has been cloned by direct selection inEscherichia coli, using the carboxymethyl cellulose-Congo Red assay. The cloned gene has been subcloned in the two possible orientations in pUC plasmids. One of the two resulting constructs exhibited a higher level of expression, which was associated with a high level of plasmid instability. The enzyme synthesized inE. coli from the cloned gene has been characterized by two procedures, maxicells and gel filtration chromatography, as a polypeptide of approximately 40 kilodaltons.  相似文献

The differences in geometry at the metal centres in the two known [Fe-4S] proteins rubredoxin (Rd) and desulforedoxin (Dx) are postulated to be a result of the different spacing of the C-terminal cysteine pair in the two proteins. In order to address this question, two mutants of Desulfovibrio gigas Dx with modified cysteinyl spacing were prepared and their solution structures have been determined by NMR. Mutant 1 of Dx (DxM1) has a single glycine inserted between the adjacent cysteines (C28 and C29) found in the wild type Dx sequence. Mutant 3 (DxM3) has two amino acid residues, -P-V-, inserted between C28 and C29 in order to mimic the primary sequence found in Rd from Desulfovibrio gigas. The solution structure of DxM1 exists, like wild type Dx, as a dimer in solution although the single glycine inserted between the adjacent cysteines disrupts the stability of the dimer resulting in exchange between a dimer state and a small population of another, probably monomeric, state. For DxM3 the two amino acid residues inserted between the adjacent cysteines results in a monomeric protein that has a global fold near the metal centre very similar to that found in Rd.  相似文献

Quinacrine fluorescence has been used to examine variant and abnormal human Y chromosomes, at interphase and mitosis. The length difference in variant Y chromosomes has been shown to involve the highly fluorescent segment only. Eight abnormalities of the Y chromosome have been positively identified, two isochromosomes of the long arms of the Y, five pericentric inversions, and a dicentric Y chromosome with two long arms. Contributory evidence for two further abnormalities, a ring Y and a dicentric with two short arms, is also given.  相似文献

The mitochondrial tyrosine tRNA from Saccharomyces cerevisiae has been sequenced. It has two interesting structural features: (i) it lacks two semi-invariant purine residues in the D-loop which are involved in tertiary interactions in the yeast cytoplasmic tRNAPhe; (ii) it has a large variable loop and therefore resembles procaryotic tRNAsTyr rather than eucaryotic cytoplasmic ones.  相似文献

Seedlings of Brassica napus L. 2–11 days after germination were used. However, the most investigation was concentrated on the 6-day old seedlings. The primary root has a diarch protostele, the two groups of primary phloem alternate with the primary xylem. At higher level, the metaxylem is gradually differentiated in a lateral direction. Being coincident with this changes of the metaxylem, the groups of phloem cell are extended. The stele of the lower hypocotyl is root-like and has no pith. In the middle hypocotyl, there is a further lateral differentiation of the metaxylem. At the higher level, four metaxylem arms appear and the groups of phloem are extended circumferentially to form two crescent shaped sectors. In the upper hypocotyl below 0.2 cm of the cotyledonary node, a central pith has been formed which separates the differentiating primary xylem into two distinct units. At a slightly higher level, each primary phloem divides into two small groups, at this time, each xylem unit and the two adjacent groups of phloem constitute a cotyledonary trace. The foliar traces of the first two foliage leaves appear in the inter-cotyledonary plane between the vascular elements of the cotyledonary traces. At this level, the vascular tissue of the hypocotyl forms a siphonostele made up of two cotyledonary traces and the two foliage leaves, where the root-stem transition has nearly been completed, while the endarch condition is not attained in the hypocotyl. At incresing distances from the cotyledonary node upwards, in the cotyledonary petiole, the protoxylem occupies a more and more adaxial position and the metaxylem a more and more abaxial direction and, thus, the endarch condition is attained. The primary system of the root, hypocotyl, and cotyledons forms a complete circular system, the plumular vascular elements are directly connected by secondary elements formed by the cambium in the region of the hypocotyl. As for the results mentioned above, the authers have not detected that the primary xylem has a rotation of 180˚, as described by Van Tieghem.  相似文献

The reaction of copper-free lentil seedlings amine oxidase with substrates has been studied. While devoid of catalytic activity, this enzyme preparation is still able to oxidize two moles of substrate and to release two moles of aldehyde and two moles of ammonia per mole of dimeric protein. The same stoichiometry has been determined on the native enzyme in the absence of oxygen. Although copper is essential for the reoxidation of the reduced enzyme, a binding of oxygen to the copper-free protein has been demonstrated.  相似文献

The change of the conformational stability and quaternary structure of the 7S globulin from french beans (phaseolin) has been investigated in the pH range 2.0-11.0 using the high-sensitivity differential scanning microcalorimetry technique. It has been established that each polypeptide chain of phaseolin consists of two thermodynamically unequivalent cooperative domains. The number and type of the side-chain hydrogen bonds which participate in the stabilization of the folded structure of each domain have been determined. The more stable domain contains six side-chain hydrogen bonds: four of the carboxylate-tyrosyl type and two of the carboxylate-histidyl type. The less stable domain contains four side-chain hydrogen bonds: two of the carboxylate-tyrosyl type and two of the carboxylate-histidyl type. All these side-chain hydrogen bonds appear to be localized within the hydrophobic interior of the domains. It has been found that the 3S form of phaseolin that is a product of the complete phaseolin dissociation at extreme pH values does not undergo any cooperative transition at heating. Consequently, this form probably has a conformation of 'molten globule' type.  相似文献

The rat liver soluble catechol-O-methyltransferase (EC 2.1.1.6.) has been purified utilizing a combination of conventional chromatography and HPLC. The purified enzyme has a molecular mass of 25 kDa, a pI of 5.1, and exists in two forms which differ in the nature of their intramolecular disulfide bonds. This difference causes these two protein forms to behave differently in reversed phase chromatography.  相似文献

The grasshopper Oedaleonotus enigma possesses a neo-XY sex system and the rest of the chromosome complement is highly polymorphic, suggesting recent chromosomal evolution. It contains a fusion between two telocentrics, a centric shift and an extra heterochromatic segment, all of which are polymorphic, and in addition it has B-chromosomes. The fusion polymorphism has a preponderance of heterokaryotypes, and despite frequent inbreeding the other two polymorphisms have binomial frequencies of heterokaryotypes. Male individuals with two neo-X chromosomes and no neo-Y (at least in the germ line) are discussed.  相似文献

The inactivation of beef red cell catalase by fast deuterons, heat, and a combination of the two has been reported. The data have been analyzed quantitatively in terms of the target theory of ionizing radiation and the theory of absolute reaction rates. Evidence for the existence of at least two forms of stable catalase has been presented. The data are shown to indicate two possible alternative molecular weights (about 250,000 and 130,000) for catalase.  相似文献

The syntheses of two nucleosides with additional nucleobases in the 2'-position are presented. The nucleosides have two- and one-carbon linkers to the additional nucleobase, respectively. The two nucleosides are synthesized from different strategies. Preparation and partial characterization of two forms of bovine thrombin.   总被引:8,自引:0,他引:8  
A chromatographic procedure has been developed for the separation and purification of bovine α- and β-thrombin. α-Thrombin has a specific activity of 2400–3000 NIH U/mg while β-thrombin has only approximately 100 NIH U/mg. When assayed with an ester substrate, the two forms have equivalent activity while β-thrombin has only 30% of the activity of α-thrombin toward an anilide substrate. Previous studies have suggested that the degradation of α-thrombin produces a species in which a peptide fragment containing a disulfide bridge is lost. The amino acid composition determined in the present study indicates that the content of cysteine is identical in the two forms of the enzyme thus permitting the proposal of a structure for β-thrombin which differs from that currently in the literature. It is suggested that changes in the environment of the active site histidine residue in the two species is largely responsible for the observed changes in the catalytic activity.  相似文献
