为研究荷叶黄酮对乳酸杆菌和双歧杆菌抗氧化能力的影响,0.25 mg/m L的荷叶黄酮添加至培养基培养保加利亚乳杆菌(Lactobacillus bulgaricus A105)、动物双歧杆菌(Bifidobacterium animalis B211),并提取细胞裂解液,依次从自由基清除能力、抗脂质过氧化能力及抗氧化物质活性三方面评估其抗氧化能力。结果显示,0.25mg/m L的荷叶黄酮提高了动物双歧杆菌(54.9%)及保加利亚乳杆菌(14.8%)对O-·2清除能力;其对保加利亚乳杆菌清除DPPH自由基能力稍有提升,对动物双歧杆菌清除DPPH自由基能力稍有抑制;其对保加利亚乳杆菌抗脂质过氧化能力提升率达658%,具有显著的协同增效作用。同时发现:经添加0.25 mg/m L的荷叶黄酮至培养基后,动物双歧杆菌和保加利亚乳杆菌胞内超氧化物歧化酶(SOD)酶活及谷胱甘肽(GSH)活力得到显著的提高。结果表明,荷叶黄酮的添加对保加利亚乳酸杆菌和动物双歧杆菌的抗氧化活性具有促进作用,且对保加利亚乳酸杆菌抗脂质过氧化能力具有明显的协同促进作用,可能与其胞内SOD酶活及GSH活力得到的显著的提升有关,具有潜在的应用价值。  相似文献

采用单因素和正交试验法对热水浸提、超声波辅助和微波辅助提取灰树花多糖的工艺进行研究,分别获得了3种方法的最佳条件,并发现微波辅助法最佳,其最优条件为:微波功率800 W、微波时间3 min,浸提2h,多糖得率为13.05%,比热水浸提法提高了51.22%.利用提取的灰树花多糖进行抗氧化性研究,发现灰树花多糖对O2-·和·OH都具有一定的清除作用,且多糖浓度与其对O2-·的清除作用存在量效关系,但不如·OH明显.  相似文献

通过响应面法优化提取发酵麸皮多糖的工艺,并评价其体外益生和抗氧化活性。以发酵麸皮多糖的得率为响应值,采用纤维素酶酶解与水浴浸提相结合的方法提取发酵麸皮多糖,以纤维素酶添加量、料液比、水浴浸提温度、水浴浸提时间为试验因素建立数学模型,筛选最佳提取工艺条件。通过测定还原力、DPPH和·OH自由基的清除能力对比发酵和未发酵麸皮多糖的体外抗氧化活性,并通过测定嗜酸乳杆菌、植物乳杆菌、两歧双歧杆菌的生长对比发酵和未发酵麸皮多糖的体外益生活性。结果表明,发酵麸皮多糖最佳提取工艺为:料液比1∶16(w/v),酶添加量1 000 U/g,水浴浸提温度90℃,水浴浸提时间60 min,在此条件下发酵麸皮多糖的得率实测值为73. 35%。发酵麸皮多糖具有较强的DPPH和·OH自由基的清除能力,可促进嗜酸乳杆菌、植物乳杆菌和两歧双歧杆菌的生长。  相似文献

目的:对夏枯草色素的抗氧化作用进行研究.方法:以蒸馏水为溶剂,从夏枯草中提取色素;采用比色法测定夏枯草色素对DPPH·、O2-·和·OH 3种自由基的清除作用以及还原能力.结果:夏枯草色素对三种自由基都有明显的清除作用,清除能力为DPPH·>·OH> O2-·;对Fe3+也具有较强的还原能力.结论:清除率与浓度存在明显的量效关系,当色素浓度为0.13 mg/mL时,其清除率O2-·为4.1%、DPPH·为28.3%、·OH为17.9%;还原Fe3+能力随色素的浓度增加而增强.  相似文献

在超声辅助下从"黑美人"马铃薯中提取原花青素,提取浓缩液经AB-8大孔吸附树脂填充柱纯化后,所得产物中原花青素含量为93.59%,得率为4.3%。抗氧化性研究表明,其对DPPH·自由基、·OH自由基和O~(-·)_2自由基具有显著的清除效果,清除率与其浓度成正相关,且不输于V_C,尤其是对·OH自由基和O~(-·)_2自由基的清除作用明显高于V_C。表明"黑美人"马铃薯原花青素有较强的抗氧化性,这为进一步开发利用"黑美人"马铃薯提供必要的理论支持。  相似文献

混菌发酵芝麻粕制备芝麻多肽及其体外抗氧化活性研究   总被引:1,自引:0,他引:1  
活性氧自由基(ROS)是细胞有氧代谢过程中产生的一类活性物,利用食源蛋白制备具有抗氧化能力的天然活性肽是当前的研究热点。本研究以机械冷榨芝麻粕为原料,比较了解淀粉芽孢杆菌、植物乳杆菌单菌及其混菌发酵芝麻粕制备芝麻多肽的过程,以DPPH·与·OH清除能力为评价指标,研究其体外抗氧化活性。数据表明,解淀粉芽孢杆菌、植物乳杆菌单菌及其混菌发酵芝麻粕36 h,芝麻多肽提取液中芝麻多肽的浓度分别达到9.841 mg/m L、11.620 mg/m L、14.412 mg/m L,DPPH·清除率分别是71.378%、77.013%、87.622%,·OH清除率分别是68.427%、82.507%、92.460%。因此,芝麻多肽具有较强的抗氧化活性,芝麻粕是制备抗氧化活性肽的良好原料;微生物发酵制备芝麻多肽时,解淀粉芽孢杆菌与植物乳杆菌混菌发酵显著优于单菌发酵,其多肽产量及DPPH·与·OH清除能力均优于相应单菌发酵。  相似文献

为探讨唐古特白刺(Nitraria tangutorun Bobr.)果实花色苷体外清除自由基活性,研究了其对超氧阴离子(O-2·)体系、羟基自由基(·OH)体系和二苯代苦味酰基自由基(DPPH·)体系的清除效果,并与抗坏血酸(vc)进行了比较.结果表明,该花色苷对O-2·、·OH、DPPH·均具有清除作用,且与浓度呈量效关系.对·OH和DPPH·清除效果相对较好,优于Vc.  相似文献

对甜杏仁的有效成份进行了分析并对其多糖的抗氧化性进行了研究,结果表明甜杏仁中脂肪、黄酮、多糖、蛋白质量分数分别为:46.53%、0.04%、1.73%、31%;抗氧化性研究表明0.2 g.L-1多糖液与同质量浓度Vc对羟自由基的清除相当,且对羟自由基.OH的抑制率随着质量浓度的增加而增强,质量浓度达到1.5 g.L-1时,清除率可达到88.8%。多糖液对超氧阴离子O2-.有很强的清除作用。  相似文献

Seahorse, Hippocampus kuda (SH) a marine teleost fish, is well known not only for its special medicinal composition and used as one of the most famous and expensive materials of traditional Chinese medicine. It was extracted with water (SHW), methanol (SHM), and ethanol (SHE), respectively and evaluated by various antioxidant assays. The including reducing power, total antioxidant, DPPH radical scavenging, hydroxyl radical scavenging, superoxide anion radical scavenging, alkyl radical scavenging, and protective effect on DNA damage caused by hydroxyl radicals generated. Further, the ROS level was detected using a fluorescence probe, 2′,7′-dichlorofluorescin diacetate (DCFH-DA), which could be converted to highly fluorescent dichlorofluorescein (DCF) with the presence of intracellular ROS on mouse macrophages, RAW264.7 cell and inhibited myeloperoxidase (MPO) activity in human myeloid, HL60 cells, respectively. Those various antioxidant activities were compared to standard antioxidants such as α-tocopherol. Among SHM exhibited the highest antioxidant activity in linoleic acid system, effective reducing power, DPPH radical scavenging, hydroxyl radical scavenging, superoxide radical scavenging, alkyl radical scavenging, inhibitory intracellular ROS, and inhibited MPO activity. Furthermore, MTT assay showed no cytotoxicity on mouse macrophages cell (RAW264.7) and human cell lines (MRC-5, HL60, U937). This antioxidant property depends on concentration and increasing with increased amount of extracts. The results obtained in the present study indicated that the see horse (Hippocampus kuda Bleeker) is a potential source of natural antioxidant.  相似文献

通过DPPH自由基清除测定、还原能力、总抗氧化能力、羟自由基清除测定,评价有柄石韦Pyrrosia petiolosa醇提物的抗氧化活性。结果显示,有柄石韦醇提物清除DPPH·的IC50为75.82 μg·mL-1,清除·OH的IC50为46.30 μg·mL-1。有柄石韦醇提物清除DPPH· 的能力强于抗坏血酸,清除·OH的能力弱于抗坏血酸;同时,其还原能力和总抗氧化能力均强于抗坏血酸。该结果说明有柄石韦醇提物具有较强的抗氧化活性。  相似文献

Antioxidant activity in α- and β-chitosan at a wide range of molecular weight (Mw) and chitosan concentration (CS) was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, reducing ability, chelating ability, and hydroxyl radical scavenging activity. The form of chitosan (FC) had significant (P <0.05) effect on all measurements except DPPH radical scavenging activity, and antioxidant activity was dependent on Mw and CS. High Mw (280–300 kDa) of β-chitosan had extremely lower half maximal effective concentrations (EC50) than α-chitosan in DPPH radical scavenging activity and reducing ability. The 22–30 kDa of α- and β-chitosan showed significantly (P <0.05) higher activities in DPPH radical scavenging, reducing ability, and hydroxyl radical scavenging than samples at other Mw, while chelating ability was the highest in 4–5 kDa chitosan. CS had significant effect on all measurements and the effect was related to Mw. The antioxidant activity of 280–300 kDa chitosan was affected by coil-overlap concentrations (C1) in the CS range of 4–10 mg/mL, forming entanglements. Reducing ability and hydroxyl radical scavenging activity were more predominant action in antioxidant activity of chitosan as shown by the lower EC50 values than those in other antioxidant measurements.  相似文献

The methanolic extract of Musa ABB cv Pisang Awak was investigated for the polyphenolic contents and antioxidant activity. The total phenol and flavonoid contents of the fruit extract were found to be 120 mg gallic acid equivalents (GAE) and 440 mg quercetin equivalents (QE)/100 g of sample dry weight, respectively. The antioxidant activity of the Pisang Awak methanol extract (PAME) (20-500 microg/ml) was determined using 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing capacity, 2-2'-azinobis-3-ethylbenzothiozoline-6-sulfonic acid (ABTS) radical cation decolourization and hydroxyl radical scavenging capacity (OH*). The EC50 values of DPPH, ABTS and OH* activities of the PAME and butylated hydroxy toluene (BHT) were found to be 65 and 9 microg/ml, 29 and 6 microg/ml, 36 and 42 microg/ml respectively. The reducing capacity increased with increasing concentration (31.5-1000 mg/ml) of the fruit extract and the activity was comparable with the standard BHT. The high performance thin layer chromatography (HPTLC) analysis of the extract revealed the presence of polyphenols. The strong and positive correlations were obtained between total phenol/flavonoid contents (R2 = 0.693-1.0) and free radical scavenging ability was attributed to the polyphenols as the major antioxidants.  相似文献

利用DPPH自由基清除法、羟基自由基(OH.)清除法、铁离子鳌合能力及测定还原能力等方法对鸡油菌Cantharellus cibarius、变绿红菇Russula virescens、蜜环菌Armillaria mellea和棕灰口蘑Tricholoma myomyces等4种食用菌的粗多糖进行了抗氧化活性评价。结果显示,4种真菌粗多糖均不同程度地具有抗氧化活性。在DPPH自由基清除试验中,棕灰口蘑和蜜环菌表现出很强的活性,其EC50值分别为1.35 g/L、1.53 g/L;棕灰口蘑和蜜环菌对羟基自由基的清除能力也要优于其他2种食用菌,其EC50值分别为0.65 g/L、0.78 g/L;棕灰口蘑的铁离子螯合能力明显优于其他3种测试菌,其EC50值为1.69 g/L;在还原力方面同样是棕灰口蘑活性最强,蜜环菌次之,其EC50值为1.05 g/L、1.37 g/L。  相似文献

以野生百合渥丹、山丹和传统食用的兰州百合为研究对象,对其鳞茎中多酚类物质、11种单体酚的含量及抗氧化活性(ABTS自由基、超氧阴离子、羟自由基的清除能力,铜离子还原能力以及抑制脂质过氧化活性)进行了分析。结果表明:两种野生百合鳞茎中的多酚类物质含量及抗氧化活性均显著高于兰州百合。3种百合鳞茎中单体酚的种类也有所不同,但均含有没食子酸、矢车菊素-3-芸香糖苷、儿茶素、表儿茶素、杨梅酮、芦丁、对香豆酸、山奈酚。相关性分析显示,除对羟自由基的清除力外,各酚类物质总量与不同抗氧化指标之间呈显著正相关关系。试验结果认为,野生百合鳞茎可作为天然抗氧化资源应用于食品和医药业,具有一定的开发应用前景。  相似文献

对印度块菌Tuber indicum子实体的提取物,包括55%乙醇粗提物(ECE)、石油醚提取物(PEF)和乙酸乙酯提取物(EAF)的清除DPPH自由基和羟基自由基能力、铁离子鳌合能力以及各提取物的总酚含量等进行了研究和测定,结果显示,3种提取物的清除自由基能力和铁离子鳌合能力具有显著差异(P0.05);ECE对DPPH自由基的清除活性最高,其EC50值为1.61g/L;EAF对羟基自由基及铁离子表现出较强的清除或螯合的能力,其EC50值分别为3.31g/L和0.70g/L;EAF的总酚含量(2.964mg GAE/g提取物)最高,其次是ECE,总酚含量为(2.618mg GAE/g提取物);PEF的清除自由基和铁离子鳌合能力较差,其总酚含量也最低(1.124mg GAE/g提取物);总酚含量与印度块菌提取物清除自由基以及鳌合铁离子的能力密切相关。  相似文献

研究了乳清分离蛋白(WPI)酶解物对DPPH.、超氧阴离子自由基和羟基自由基的清除效果,同时用还原法研究了其抗氧化活性。结果表明,WPI酶解物在体外具有较强的抗氧化能力。木瓜蛋白酶酶解物和胰蛋白酶酶解物对DPPH.、超氧阴离子自由基、羟基自由基的清除能力和还原能力强于胰凝乳蛋白酶酶解物和胃蛋白酶酶解物。  相似文献

The antioxidant potency of chitin derivative-glucosamine hydrochloride was investigated employing various established in vitro systems, such as superoxide (O2*-)/hydroxyl (*OH)-radical scavenging, reducing power, and ferrous ion chelating potency. As expected, we obtained several satisfying results, as follows: first, glucosamine hydrochloride had pronounced scavenging effect on superoxide radical. For example, the O2*- scavenging activity of glucosamine hydrochloride was 83.74% at 0.8 mg/mL. Second, the *OH scavenging activity of glucosamine hydrochloride was also strong and was about 54.89% at 3.2 mg/mL. Third, the reducing power of glucosamine hydrochloride was more pronounced. The reducing power of glucosamine hydrochloride was 0.632 at 0.75 mg/mL. However, ferrous ion-chelating potency was soft. Furthermore, ferrous ion-chelating potency, the scavenging rate of radical, and the reducing power of glucosamine hydrochloride increased with their increasing concentration, and they were concentration dependent. The multiple antioxidant activity of glucosamine hydrochloride was evident as it showed considerable reducing power, superoxide/hydroxyl-radical scavenging ability. These in vitro results suggest the possibility that glucosamine hydrochloride could be effectively employed as an ingredient in health or functional food, to alleviate oxidative stress.  相似文献

Date palm (P. dactylifera) plays a vital role in ethnomedicinal practices in several parts of the world. There are over 2000 cultivars of date palm that differ in chemical composition and extent of bioactivity. The present study was undertaken to comparatively evaluate the antioxidant potential of three cultivars of date palm (Ajwah, Safawy and Sukkari) from Saudi Arabia and analyze their phenolic constituents in order to draw a rationale for their activity. Antioxidant activities of the date cultivars were evaluated by different quantitative methods including 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical scavenging assay, total antioxidant capacity, reducing power, total phenolic (TPC), flavonoid (TFC) and tannin content (TTC), while qualitative phenolic composition was determined using ultra performance liquid chromatography coupled to quadropole time of flight mass spectrometry (UPLC-QTOF-MS). All the three date extracts showed good DPPH radical scavenging (IC50 103–177 μg/mL) and hydroxyl radical scavenging (IC50 1.1–1.55 mg/mL) activity and total antioxidant capacity (IC50 87–192 μg/mL). The reducing power was also comparable to that of ascorbic acid, used as standard in above experiments. All the three samples contain significant amount of major antioxidant components (phenolic, flavonoid and tannin) that successfully correlates with the results of radical scavenging assays. UPLC-QTOF-MS revealed a total of 22 compounds in these date cultivars classified into common phenolics, flavonoids, sterols and phytoestrogens. Significant variation in the degree of antioxidant activity of these three date cultivars can be attributed to the difference in the content and composition of phenolic compounds.  相似文献
