【目的】探明茄二十八星瓢虫Henosepilachna vigintioctopunctata(Fabricius)成虫的天花板爬行行为,为气流控制技术防治农业害虫提供技术参数。【方法】本研究通过采用录像观察法对茄二十八星瓢虫雌雄成虫的天花板爬行行为进行观测,明确其成虫的运动行为参数,并分析了其步速分别与步距、步频和负荷因数的关系。【结果】茄二十八星瓢虫雌雄成虫天花板爬行时的速度为3.77~19.32 mm/s,步距为2.63~4.51 mm,步频范围为1.32~4.74 Hz。步距和步频均与速度之间存在极显著线性关系,速度的增加同时依靠步距和步频的增加,但步距的变化相对步频的变化小;前足、中足和后足的负荷因数均随着速度的提高而降低,且负荷因数均与速度存在显著相关性;一个步态周期内,足的摆动时间均远小于支撑时间。【结论】茄二十八星瓢虫成虫天花板爬行时速度的增加主要是依靠步频的提高来实现。  相似文献

为了探索快速鉴定马铃薯瓢虫Henosepilachna vigintioctomaculata(Motschulsky)和茄二十八星瓢虫Henosepilachna vigintioctopunctata(Fabricius)的分子生物学方法,本研究在随机扩增多态性DNA(random amplified polymorphic DNA,RAPD)的基础上,分别设计了可以鉴别两个物种的序列特征扩增区域(sequence characterized amplified regions,SCAR)标记。从随机合成的60条引物中筛选出来2条特异性引物(分别为OPI-6和OPJ-15),引物OPI-6在马铃薯瓢虫中扩增出约750 bp的特异性条带,引物OPJ-15在茄二十八星中扩增出约750 bp的特异性条带,根据测序结果设计了两对SCAR引物对筛选结果进行验证,发现根据OPI-6的测序结果所设计的SCAR引物(OPI-6 test)仅能在马铃薯瓢虫中扩增出645 bp的条带,而根据OPJ-15的测序结果所设计的SCAR引物(OPJ-15 test)仅能在茄二十八星瓢虫中扩增出436 bp的条带。这两对SCAR引物能够准确、稳定且快速地区分马铃薯瓢虫与茄二十八星瓢虫,对这两种害虫的精准防控具有重要意义。  相似文献

RNAi抗虫技术,是一种新型绿色环保的害虫防控方法,具有广阔的应用前景.茄二十八星瓢虫Henosepilachna vigintioctopunctata是茄科植物上的重要害虫,对作物生产造成了严重的经济损失.本研究用饲喂法RNAi探究了沉默凋亡抑制蛋白1(inhibitor of apoptosis protein 1,IAP1)表达对茄二十八星瓢虫生长发育的影响.结果显示,HvIAP1在茄二十八星瓢虫的每个发育阶段均表达,1龄幼虫和3龄幼虫的表达量最高,成虫的表达量最低.取食200 ng/μL dsHvIAP1浸泡处理的叶片2 d后,可导致92%的茄二十八星瓢虫1龄幼虫死亡,同时在取食dsHvIAP124 h后,HvIAP1基因的表达量下降了2.40倍;另外,单头取食200 ng的dsHvIAP1可导致70%的4龄幼虫死亡,同时在取食48 h后,HvIAP1基因的表达量下降了2.58倍.并且RNAi处理后1龄幼虫和4龄幼虫大部分个体在48 h内出现急性取食障碍现象,基本不取食直至死亡.上述结果表明,HvIAP1基因在茄二十八星瓢虫的生长发育过程中起重要作用,当该基因表达受阻,会直接影响茄二十八星瓢虫的取食、抑制其生长发育而死亡.这一研究表明HvIAP1基因可作为潜在的防治茄二十八星瓢虫的RNAi靶标基因.  相似文献

纳米材料被视为重要的RNA干扰增效物质。层状双氢氧化物(LDH)是具有生物相容性的可降解纳米材料,可以显著提高dsRNA防控黄瓜花叶病毒的效果。为明确LDH能否提高昆虫RNA干扰的效果,以期促进RNA生物农药在农业害虫防治中的应用和推广,本研究选择茄二十八星瓢虫Henosepilachna vigintioctopunctata的EcR基因进行RNA干扰介导的致死效应研究。将LDH与dsEcR组装形成复合物LDH-dsEcR,利用琼脂糖凝胶电泳确定LDH-dsEcR的最佳装载比例。通过静置法检测LDH-dsEcR的稳定性,借助透射电子显微镜验证其组装状态。采用浸叶法测定LDH-dsEcR的增效性,使用体视镜观察其对试虫形态的影响。结果表明:LDH与dsEcR可形成稳定的复合物,其最佳装载比为16∶1。LDH对茄二十八星瓢虫无毒性,但经LDH与dsEcR处理,部分试虫虫体呈黑褐色,或无法蜕皮化蛹,或维持在预蛹状态。遗憾的是LDH-dsEcR复合物的增效作用不明显,推测昆虫肠道的酸碱度可能是影响LDH-dsRNA增效的因素之一。本研究首次为利用LDH研究昆虫RNA生物农药提供了可观的理论基础。  相似文献

The outbreak of Henosepilachna vigintioctopunctata (F.) was noticed on a medicinal plant, Withania somnifera Dunal during 2004-05. The population level of the pest was reached its peak in August. The parasitoid Pediobius foveolatus caused 51.94%±12.20% parasitism. The pest completed its life cycle in 20.15±1.50 days on W. somnifera. The longevity of the male and female adults of the pest was 22.07±3.71 and 31.07±4.38 days, respectively. The gravid females of H. vigintioctopunctata laid an average of 287.64±33.38 eggs during their oviposition period of 10.40±2.80 days.  相似文献

An entomopathogenic bacterium was isolated from tomato leaves and used as a microbial agent to control larvae of phytophagous ladybird beetles Epilachna vigintioctopunctata. The isolate was identified as Pseudomonas fluorescens KPM-018P on the basis of its bacteriological characteristics. KPM-018P produced extracellular chitinase to form a transparent zone around their colonies by hydrolyzing chitin in a minimal medium. Pale-yellow colonies turned red after a change of incubation temperature. These characteristics were availed as markers for tracking KPM-018P. The bacteria produced biosurfactants that enabled the bacteria to stably colonize the hydrophobic leaf surface; they were recovered without any considerable decrease even after a suspension of KPM-018P was sprayed onto leaves. KPM-018P, transformed with the gfp gene and observed with fluorescence microscopy, stably dwelled in the junctions of epidermal cells of bacteria-sprayed leaves. Ingestion of KPM-018P-sprayed leaves by the larvae caused prompt death of these insects to eventually suppress their pupation. This method is thus effective for decreasing the population of larvae and adult insect pests in the subsequent generation. The study provides an experimental basis for the biocontrol of herbivorous insect pests using a leaf-inhabiting, entomopathogenic strain of P. fluorescens.  相似文献

【目的】茄二十八星瓢虫Henosepilachna vigintioctopunctata是茄科(Solanaceae)植物上的重要害虫。昆虫体内几丁质脱乙酰酶1(chitin deacetylase 1, CDA1)催化N-乙酰氨基-D-葡萄糖胺脱去乙酰基,促使几丁质转化为壳聚糖,控制昆虫体内几丁质纤维有序堆积,并维持角质层结构的完整性。抑制虫体中CDA1基因的表达会抑制壳聚糖的合成,影响昆虫表皮结构的形成,使昆虫不能正常发育而亡。【方法】利用RT-qPCR方法测定HvCDA1基因在茄二十八星瓢虫不同发育阶段(卵、1-4龄幼虫和预蛹)和4龄幼虫不同组织(表皮、脂肪体、中肠和马氏管)中的表达模式。通过饲喂茄二十八星瓢虫1龄幼虫不同浓度dsHvCDA1溶液浸泡处理1 min的茄子叶片后及直接饲喂4龄幼虫不同浓度dsHvCDA1溶液,探究沉默茄二十八星瓢虫HvCDA1基因对其幼虫存活和发育以及HvCDA1基因表达量的影响。【结果】发育表达谱结果表明,HvCDA1在茄二十八星瓢虫的各发育阶段均有表达,但在1龄末和2龄末幼虫中的表达量最高。组织表达谱结果显示,在茄二十八星瓢虫4龄幼虫的表皮中H...  相似文献

In interspecific crosses, a mismatch in internal physiological conditions between two species can reduce sperm viability in the interval from insemination to fertilization, leading to gametic isolation. Two closely related Japanese phytophagous ladybird beetles, Henosepilachna vigintioctomaculata and H. pustulosa, show several isolating barriers, including reduction in the number of heterospecific sperm in the female reproductive tract and low egg‐hatching rates in interspecific matings. However, the mechanisms of these two potential isolating barriers and the association between them are unknown. Here we investigated temporal changes in the number of sperm stored in the female reproductive tract and egg‐hatching rates in inter‐ and intraspecific crosses between these species. Although the number of sperm decreased after both inter‐ and intraspecific crosses, the reduction was more drastic in inter‐ than in intraspecific crosses for females of both species. Most of the sperm reduction occurred early on, during sperm transfer from the bursa copulatrix to the paired ampullae of the common oviduct (the sperm storage organs). These two species also demonstrated stably low egg‐hatching rates in interspecific crosses. Since the degree and timing of the sperm reduction did not correlate with egg‐hatching rates, the reduction in heterospecific sperm in interspecific crosses may not directly cause the low hatching rates. These two isolating barriers could be different expressions of the physiological mismatch and/or genetic incompatibility between gametes of these species.  相似文献

This study reports genome size (C‐value) estimates for seven species of ladybird beetles (Coleoptera: Coccinellidae) in Japan using flow cytometry. The results demonstrated genome sizes of 1.0–1.4 Gb in four closely related phytophagous ladybird beetles belonging to the Henosepilachna vigintioctomaculata species complex. These values were approximately two times larger than that of a congeneric phytophagous ladybird beetle H. vigintioctopunctata (0.66 Gb), and of two very distantly related common carnivorous ladybird beetles, Harmonia axyridis (0.46 Gb) and Coccinella septempunctata (0.42 Gb). These lines of evidence suggest that rapid and large genome size increase occurred just after the branching of the common ancestor of the H. vigintioctomaculata species complex from other ladybird species.  相似文献

The telotrophic ovary of Epilachna vigintioctopunctata is composed of 32-40 ovarioles, each with an apical germarium and a basal vitellarium. The germarium encloses mononucleate and binucleate trophocytes, prefollicular tissue and oogonia, while the vitellarium contains 2-5 oocytes arranged in order of maturity. Definite nutritive cords are absent. When females are exposed to 75 mg 4,4,6-trimethyl-1h, 4H-pyrimidine-2-thiol by contact, the trophocytes and the follicular epithelial cells disintegrate to form dark-staining clumps and thus fail to supply nourishment to the developing oocytes, which consequently remain yolk-less and are ultimately reduced to shrunken masses.  相似文献

Extraction, thin-layer chromatography, and gas chromatography–mass spectrometry of leaf surface waxes of Momordica charantia L. (Cucurbitaceae) revealed that 20 n-alkanes between n-C15 and n-C36, except n-C34 and n-C35, commonly occur in young, mature, and senescent stages. Hentriacontane, hentriacontane, and hexatriacontane were the predominant compounds in young, mature, and senescent leaves, respectively. The cuticular alkanes from young, mature, and senescent leaves attracted the female insect, Epilachna dodecastigma (Wied.), at 25–400, 25–400, and 100–400 μg concentrations, respectively, whereas the mixtures of synthetic alkanes mimicking cuticular alkanes of young, mature, and senescent leaves showed attraction at 100–400, 100–400, and 200–400 μg concentrations, respectively, in Y-shaped glass tube olfactometer bioassay. The difference in insect attraction is probably due to the absence of branched-chain alkanes in the synthetic mixtures. Individual synthetic heptacosane, nonacosane, and hentriacontane at 28.19–56.90, 32.04–64.08, and 60.44–120.88 μg, respectively, elicited attraction of the insect. A synthetic blend of 4.82, 4.91, 5.71, 6.74, 56.39, 7.94, 62.42, 120.88, and 36.33 μg of nonadecane, eicosane, heneicosane, pentacosane, heptacosane, octacosane, nonacosane, hentriacontane, and tritriacontane, respectively, was most attractive to the insect.  相似文献

In planta RNAi against essential insect genes offers a promising route to control insect crop pests, but is constrained for many insect groups, notably phloem sap-feeding hemipterans, by poor RNAi efficacy. This study conducted on the phloem-feeding whitefly Bemisia tabaci reared on tomato plants investigated the causes of low RNAi efficacy and routes to ameliorate the problem. Experiments using tomato transgenic lines containing ds-GFP (green fluorescent protein) revealed that full-length dsRNA is phloem-mobile, ingested by the insects, and degraded in the insect. We identified B. tabaci homologs of nuclease genes (dsRNases) in other insects that degrade dsRNA, and demonstrated that degradation of ds-GFP in B. tabaci is suppressed by administration of dsRNA against these genes. dsRNA against the nuclease genes was co-administered with dsRNA against two insect genes, an aquaporin AQP1 and sucrase SUC1, that are predicted to protect B. tabaci against osmotic collapse. When dsRNA constructs for AQP1, SUC1, dsRNase1 and dsRNase2 were stacked, insect mortality was significantly elevated to 50% over 6 days on artificial diets. This effect was accompanied by significant reduction in gene expression of the target genes in surviving diet-fed insects. This study offers proof-of-principle that the efficacy of RNAi against insect pests can be enhanced by using dsRNA to suppress the activity of RNAi-suppressing nuclease genes, especially where multiple genes with related physiological function but different molecular function are targeted.  相似文献

RNA interference already proved its usefulness in functional genomic research on insects, but it also has considerable potential for the control of pest insects. For this purpose, the insect should be able to autonomously take up the dsRNA, for example through feeding and digestion in its midgut. In this review we bring together current knowledge on the uptake mechanisms of dsRNA in insects and the potential of RNAi to affect pest insects. At least two pathways for dsRNA uptake in insects are described: the transmembrane channel-mediated uptake mechanism based on Caenorhabditis elegans’ SID-1 protein and an ‘alternative’ endocytosis-mediated uptake mechanism. In the second part of the review dsRNA feeding experiments on insects are brought together for the first time, highlighting the achievement of implementing RNAi in insect control with the first successful experiments in transgenic plants and the diversity of successfully tested insect orders/species and target genes. We conclude with points of discussion and concerns regarding further research on dsRNA uptake mechanisms and the promising application possibilities for RNAi in insect control.  相似文献

Bt WZ-9 strain, containing a single Cry7Ab3 toxin, had effective insecticidal activity against larvae of Henosepilachna vigintioctomaculata. By incubation with larvae midgut homogenate and trypsin in vitro, 130 kDa Cry7Ab3 protoxin was degraded into the ~75 kDa proteinase-resistant fragments. In vivo analysis, 130 kDa Cry7Ab3 protoxin was also processed into ~75 kDa fragment. Histopathological observations indicated that Cry7Ab3 ingestion by H. vigintioctomaculata larvae causes acceleration in the blebbing of the midgut epithelium cells into the gut lumen and eventual lysis of the epithelium cells resulting in larval death. A ligand blotting experiment demonstrated that Cry7Ab3 toxin bound a 220 kDa BBMV protein. This receptor protein was identified as cadherin by matrix assisted laser desorption-time of flight-mass spectrometry (MALDI-TOF-MS). The cadherin protein may be the receptor of Cry7Ab3. The data obtained may contribute to a better understanding of the mechanism of Cry7Ab3 toxin against H. vigintioctomaculata larvae.  相似文献

In this study, I tested an artificial diet, Insecta LFS, for rearing the 28-spotted ladybird beetle, Henosepilachna vigintioctopunctata (Fabricius). H. vigintioctopunctata larvae could not be reared through all of the larval stages on the artificial diet. However, initially they could be reared on tomato leaves up to the second or third instar, and thereafter exclusively on the artificial diet. The larval and pupal periods were not significantly different from those reared only on tomato leaves. For females reared by the diet-switching method, the preoviposition period was significantly longer and the number of eggs laid significantly lower than for those on tomato leaves. However, these differences did not become a hindrance to laboratory rearing. This rearing method saves labor involving obtaining fresh plant leaves and produces a reliable supply of the insects.  相似文献

The oligophagous ladybird beetle Henosepilachna yasutomii Katakura (Coleoptera: Coccinellidae) shows inter-population differences in its host-use. In this beetle, populations using the deciduous tree Pterostyrax hispida Sieb. et Zucc. (Styracaceae) were recently rediscovered in the Kanto districts of central Honshu, Japan. In the present study, the ability to utilize P. hispida and other host plants was compared among four populations of H. yasutomii occurring on P. hispida and Scopolia japonica Maxim. (Solanaceae), Chelidonium japonicum Thunb. (Papaveraceae), and Solanum tuberosum L. (Solanaceae). As regards the feeding habits of adult beetles, only the populations occurring on P. hispida accepted this plants leaves, which differed distinctly from the feeding habits of the other populations, although the differences among the four populations were not readily apparent with respect to the leaf amounts consumed. The larvae from the populations occurring on P. hispida showed significantly higher survivorship on this plant than did the larvae from the other populations. Considering the host use patterns and the life cycles of beetles under natural conditions, the large abundance of P. hispida leaves throughout the season may have played an important role in selection for the ability to utilize P. hispida observed in the H. yasutomii populations occurring on this woody host.  相似文献

Geographical variation in the elytral spot patterns of a phytophagous ladybird, Epilachna vigintioctopunctata, was studied in the Province of Sumatera Barat, Indonesia. Populations of E. vigintioctopunctata were divided into four major groups (I–IV) by the incidence of spot pattern variations. Group I, occurring in the coastal plains and inland lowlands, and Group IV, confined to the highlands, were the extremes of the spot pattern variations, the latter had many more non-persistent spots and confluences with larger body size and advanced melanism than the former. These two groups were connected with each other via the intermediate groups. A positive relationship was detected between the elevations of sample sites and the average number of non-persistent spots per elytron. Consequently, present results favor the view that the two previously recognized forms of E. vigintioctopunctata (formae A and B inKatakura et al., 1988) represent a complicated intraspecific variation rather than two distinct sibling species. Elytral spot pattern variations were not different between the sexes or between the beetles collected from different kinds of host plants.  相似文献
