Tuco-tucos (small subterranean rodents of the genus Ctenomys) that inhabit sandy soils of the area under the influence of the second largest wetland of South America, in Northeastern Argentina (Corrientes province), are a complex of species and forms whose taxonomic status were not defined, nor are the evolutionary relationships among them. The tuco-tuco populations of this area exhibit one of the most ample grades of chromosomal variability within the genus. In order to analyze evolutionary relationships within the Corrientes group and its chromosomal variability, we completed the missing karyotypic information and performed a phylogenetic analysis. We obtained partial sequences of three mitochondrial markers: D-loop, cytochrome b and cytochrome oxidase I. The Corrientes group was monophyletic and split into three main clades that grouped related karyomorphs. The phylogeny suggested an ancestral condition of the karyomorph with diploid number (2n) 70 and fundamental number (FN) 84 that has evolved mainly via reductions of the FN although amplifications occurred in certain lineages. We discuss the relationship between patterns of chromosomal variability and species and groups boundaries. From the three main clades the one named iberá exhibited a remarkable karyotypic homogeneity, and could be considered as an independent and cohesive evolutionary lineage. On the contrary, the former recognized species C. dorbignyi is a polyphyletic lineage and hence its systematic classification should be reviewed.  相似文献

The tuco-tucos [Ctenomys) are subterranean rodents that are widespread in southern South America. On the basis of its 60 living species, Ctenomys is one of the most speciose mammalian genera and displays great chromosomal variation. In order to study the mode of speciation in Ctenomys and to evaluate the role of chromosomes in cladogenesis, it is essential to generate an accurate phylogeny of the genus. From such a phylogeny it should be possible to identify particular species lineages worthy of further study. Following the success in 1998 of Lessa & Cook in generating a phylogeny of 10, mostly Bolivian, species from mitochondrial cytochrome b sequences, we sequenced a further 18, mostly Argentinian, species. By combining our datasi't with that of Lcssa & Cook's we were able to confirm six species lineages within Ctenomys. These lineages correspond well to taxonomic groups suggested on the basis of morphology and biogeographv. At least two of the species groups (the Boliviano-Matogrossense and Chaco) arc very variable chromosomally, and are worthwhile systems to examine the possibility of chromosomal spec iation. While, in general, multiple specimens of the named species did form monophyletic groups on the basis of the cytochrome b analysis, there were some exceptions. Thus, C. bergi and C yolandae did not form separate clades in the phylogenetic analysis, nor did C. roigi and C. dorbignyi. The molecular analysis was also very informative with regards species designations for C. boliviensis and related forms.  相似文献

Genetic variability measured by allozymic electrophoresis has been studied in several species of the subterranean rodent genus Ctenomys (Octodontidae). The study was carried out with the main purpose of analysing a special group known as the 'Corrientes group' which inhabits the province of Corrientes in Argentina. The members of the group are, with high probability, isolated reproductively due to their karyotypic differences diploid numbers are between 42 and 70). To evaluate whether the chromosomal differences were the causes of speciation or if they arose a posteriori, we compared the level of the genetic distance among taxa within the orrientes group with all those measured between intraspecific populations in Ctenomys . The results indicate that the distances among the populations of the Corrientes group are at or below the level of those measured among traspecific populations in Ctenomys . Thus, it is unlikely that genetic differentiation triggered speciation. It is shown that this low level of genetic differentiation is not in contradiction with the high levels expected for species originating from one of the possible mechanisms of chromosomal speciation named, chromosomal transilience. Although for geographical reasons it seems obviousthat gene flow is precluded among the members of the Corrientes group, the Slatkin method for estimating Nm values was also used. Because high values of Nm exist but no isolation by distance could be detected, it is suggested that reproductive and geographical isolation are very ecent. Finally, analysis of population variability suggests that the high levels of heterozygosity observed (a) can be explained by the population structure, and (b) are within the range of expected values if bottlenecks have occurred in the recent history of the Corrientes group. As a general conclusion, the results indicate that in the Corrientes group the genetic data support a putative causal role for chromosomes in speciation.  相似文献

The genus Ctenomys comprises about 70 species with great chromosome diversity. The Corrientes group is one of the most chromosomally variable lineages in the genus, where the diploid number (2n) varies from 41 to 70. In this group, three nominal species and numerous polymorphic and polytypic populations have been described. In order to get insight into the chromosomal evolution of this species complex, we applied different banding and molecular cytogenetic techniques. The results were interpreted in an evolutionary context, based on mitochondrial cytochrome b analyses. Studied samples are representative of the broad chromosomal variability in the group, including specimens with 2n?=?42 to 2n?=?70. Heterochromatin was scarce but concentrated in a few chromosomes. Centromeric DAPI-negative heterochromatin was observed in some autosomal pairs, which differed among populations. Location and amount of DAPI-neutral heterochromatin within the Y chromosome varied among populations. The variable distribution of heterochromatin indicates its dynamic behavior. NORs were detected in one pair of autosomes, which also differed among some populations. Telomeric FISH signals were observed in all complements only at the chromosome ends. The Corrientes group belongs to a clade that also includes C. pearsoni, C. lami, C. minutus, C. ibicuiensis and C. torquatus. Almost all of these species are variable at the chromosomal level, suggesting that this is the ancestral condition of the clade. Within the Corrientes group, the observed low genetic divergence, in contrast with its high chromosomal variability, is indicative of decoupling between the rates of chromosomal and mitochondrial evolution.  相似文献

We investigated the relationship between satellite copy number and chromosomal evolution in tuco-tucos (genus Ctenomys), a karyotypically diverse clade of rodents. To explore phylogenetic relationships among 23 species and 5 undescribed forms, we sequenced the complete mitochondrial cytochrome b genes of 27 specimens and incorporated 27 previously published sequences. We then used quantitative dot-blot techniques to assess changes in the copy number of the major Ctenomys satellite DNA (satDNA), named RPCS. Our analysis of the relationship between variation in copy number of RPCS and chromosomal changes employed a maximum-likelihood approach to infer the copy number of the satellite RPCS in the ancestors of each clade. We found that amplifications and deletions of RPCS were associated with extensive chromosomal rearrangements even among closely related species. In contrast, RPCS copy number stability was observed within clades characterized by chromosomal stability. This example reinforces the suspected role of amplification, deletion, and intragenomic movement of satDNA in promoting extensive chromosomal evolution.  相似文献

The Ozark madtom, Noturus albater , is distributed in upland streams in the upper White, Black, and St. Francis rivers of Missouri and Arkansas, USA. Fixed chromosomal and biochemical differences between populations in the upper White River and the Black and St. Francis rivers led earlier researchers to suggest that the Ozark madtom comprised two distinct species. The current study reviewed previous data, and examined osteology, pigmentation, morphometrics and DNA sequences from the mitochondrial cytochrome b gene and the nuclear Recombination Activating Gene 2. Sequence data were analysed using parsimony and Bayesian methods to construct a phylogenetic hypothesis of relationships within Noturus and examine relationships of populations of Ozark madtoms. Results of the molecular analyses were consistent with earlier chromosomal and biochemical studies. Morphology could not distinguish between populations from the upper White River and populations from the Black and St. Francis rivers. However, karyotypes, allozyme variation, and DNA sequences all provide diagnostic characters demonstrating the presence of two species consistent with the phylogenetic species concept. We describe the populations from the Black and St. Francis rivers as Noturus maydeni , a new cryptic species of madtom catfish.  相似文献

To investigate the molecular phylogenetic status of the Korean goral, Nemorhaedus caudatus raddeanus, and Japanese serow, Capricornis crispus, we determined partial sequences of the mitochondrial cytochrome b gene of twelve Korean gorals and sixteen Japanese serows, and compared them with those of the major lineages of Rupicaprini species including two other Nemorhaedus species and two other Capricornis species. The Korean gorals examined possessed two haplotypes with only one nucleotide difference between them, while the Japanese serows showed slightly higher sequence diversity with five haplotypes. Genetic distances and molecular phylogenetic trees indicated that there is considerable genetic divergence between the Korean goral and N. caudatus (the Chinese goral) [Groves and Shields (1996)], but virtually none between Korean and Russian gorals. The Korean and Russian gorals may therefore be distinct from the Chinese goral. The data highlight the importance of conservation of the goral populations of these regions, and the need to reconsider the taxonomic status of Korean and Russian gorals. Our study also clearly demonstrated sufficient genetic distance between serows and gorals to justify their assignment to separate genera. Of the three species of Capricornis, the Formosan serow, C. swinhoei is more closely related to C. sumatraensis than to the Japanese serow, suggesting that the Formosan serow is a distinct species. Preliminary data on intraspecific genetic variation in the Japanese serow are also presented.  相似文献

The chromosomes of subterranean rodents of the South American genus Ctenomys are highly variable with diploid numbers ranging from 10 to 70. The phylogenetic relationships of this group have been analysed cladistically using G-banded karyotypes as have the chromosomal rearrangements involved in its karyotypic differentiation. One group, called the 'Corrientes group', has very variable chromosomes but low allozymic and morphological differentiation among its members. This group has been analysed with respect to chromosomal speciation. Using a member of another subfamily (Octodontomys gliroides) as an outgroup, the results indicate that karyotypes with low diploid and fundamental numbers are plesiomorphic. The range of diploid numbers studied here is between 22 and 70, while the fundamental numbers are between 40 and 86. It was found that the main chromosomal rearrangement that transforms karyotypes towards higher diploid and fundamental numbers is the acquisition of new chromosomal material via unknown mechanisms, followed by pericentric inversions that generate new chromosomal arms, centric fusions and centric fissions. In spite of their low differentiation regarding allozymic and morphological features, it was found that the karyomorphs of the Corrientes group have enough chromosomal differentiation to consider them as distinct species. Beside the range of diploid and fundamental numbers of this group (42–70 and 80–84 respectively), their pairwise chromosomal differences are high. The most closely related of them differ in one nonhomologous arm, one Robertsonian change and a whole chromosome duplication. The most differentiated taxa differ in 20 arms with lack of homology, 12 Robertsonian changes (one with monobrachial homology), six pericentric inversions and the above mentioned probable arm duplication. For these reasons, it is probable that some kind of chromosomal speciation has occurred in the Corrientes group.  相似文献

Patterns of mitochondrial DNA (mtDNA) variation were examined in 133 mole-rats constituting all four chromosomal species (2n = 52, 2n = 54, 2n = 58, and 2n = 60) of the Spalax ehrenbergi superspecies in Israel, as well as the peripheral isolates of 2n = 60. In the main range of the complex, a total of 28 mtDNA haplotypes were found in 64 mole-rats, with most haplotypes being unique to either a single chromosomal species or population. mtDNA divergence increased from low to high diploid number in a north-to-south direction in Israel. Overall levels of mtDNA diversity were unexpectedly the highest in the 2n = 60, the youngest species of the complex. The mtDNA haplotypes can be separated into two major groups, 2n = 52-54 and 2n = 58-60, and a phylogenetic analysis for each group revealed evidence of a few haplotypes not sorted by diploid number. The overall patterns of mtDNA divergence seen within and among the four chromosomal species are consistent with the parapatric mode of speciation as suggested from previous studies of allozyme and DNA hybridization. In a separate data set the patterns of mtDNA variation were examined across the main geographic range and across peripheral semi-isolates and isolates of the 2n = 60 chromosomal species. Fifteen haplotypes were found in 69 mole-rats. High levels of mtDNA diversity characterized the main range, semi-isolated, and even some desert isolated populations. The peripheral isolates contain much mtDNA diversity, including novel haplotypes.   相似文献

Background

Taxonomical studies of the neotropical Peripatidae (Onychophora, velvet worms) have proven difficult, due to intraspecific variation and uniformity of morphological characters across this onychophoran subgroup. We therefore used molecular approaches, in addition to morphological methods, to explore the diversity of Epiperipatus from the Minas Gerais State of Brazil.

Methodology/Principal Findings

Our analyses revealed three new species. While Epiperipatus diadenoproctus sp. nov. can be distinguished from E. adenocryptus sp. nov. and E. paurognostus sp. nov. based on morphology and specific nucleotide positions in the mitochondrial cytochrome c oxidase subunit I (COI) and small ribosomal subunit RNA gene sequences (12S rRNA), anatomical differences between the two latter species are not evident. However, our phylogenetic analyses of molecular data suggest that they are cryptic species, with high Bayesian posterior probabilities and bootstrap and Bremer support values for each species clade. The sister group relationship of E. adenocryptus sp. nov. and E. paurognostus sp. nov. in our analyses correlates with the remarkable morphological similarity of these two species. To assess the species status of the new species, we performed a statistical parsimony network analysis based on 582 base pairs of the COI gene in our specimens, with the connection probability set to 95%. Our findings revealed no connections between groups of haplotypes, which have been recognized as allopatric lineages in our phylogenetic analyses, thus supporting our suggestion that they are separate species.

Conclusions/Significance

Our findings suggest high cryptic species diversity and endemism among the neotropical Peripatidae and demonstrate that the combination of morphological and molecular approaches is helpful for clarifying the taxonomy and species diversity of this apparently large and diverse onychophoran group.  相似文献

Using the data on complete sequences of cytochrome b gene, phylogenetic relationships were studied among the Stenocephalemys s. lat. (Stenocephalemys ssp. + Praomys albipes) murine rodents, inhabiting adjacent altitudinal belts of the isolated Ethiopian mountain massifs, and among the related Praomys s. lat. species. Extremely low resolution of the relationships among the main Praomys s. lat. lineages hampered identification of the nearest sister group for the Stenocephalemys s. lat. "Ethiopian" clade, monophyly of which was strongly supported. Sister relationships between P. albipes and S. griseicauda (implying "accelerated" morphological and chromosomal evolution upon the formation of the former species), as well as between S. albocaudata and the recently described novel chromosomal form of Stenocephalemus sp. A (2n = 50; NFa = 56) were demonstrated. Definite discordance between the rates of their molecular, chromosomal, and morphological evolution was revealed. Based on phylogenetic reconstructions and the estimates of the divergence time, obtained by use of molecular clock method, an attempt to draw a phylogenetic scenario for the group examined was made. The obtained data were compared to those for analogous Sigmodontinae species complexes, distributed across a marked altitudinal gradient on the Andean slopes. It was shown that molecular genetic data on the rodents from mountain tropics did not support the gradient model of diversification, based on the possibility of morphological diversification prior to their achievement of the species status (without interruption of the gene flow between the forms) due to differently directed selection across a strong environmental gradient.  相似文献

Oak gallwasps are cyclically parthenogenetic insects that induce a wide diversity of highly complex species- and generation-specific galls on oaks and other Fagaceae. Phylogenetic relationships within oak gallwasps remain to be established, while sexual and parthenogenetic generations of many species remain unpaired. Previous work on oak gallwasps has revealed substantial intra-specific variation, particularly between regions known to represent discrete Pleistocene glacial refuges. Here we use statistical phylogenetic inference methods on sequence data for a fragment of the mitochondrial cytochrome b gene to reconstruct the relationships among 62 oak gallwasp species. For 16 of these we also include 23 additional cytochrome b haplotype sequences from different Pleistocene refuge areas to test the effect of intra-specific variation on inter-specific phylogeny reconstruction. The reconstructed phylogenies show good intra-generic resolution and identify several conserved clades, but fail to reconstruct either very recent or very ancient divergences. Nine of the 16 species represented by multiple haplotypes are not monophyletic. The apparent discordance between the recovered gene tree and the current taxonomic classification can be explained through: (a) collapsing of some species currently known only from either a sexual or a parthenogenetic generation into a single cyclically parthenogenetic entity; (b) sorting of ancestral polymorphism in diverging lineages, and (c) horizontal transfer of haplotypes, perhaps due to hybridization within glacial refuges. Our conclusions emphasise the need for careful intra-specific sampling when reconstructing phylogenies for radiations of closely related species and imply that for certain taxonomic groups full phylogenetic resolution (using molecular markers) may not be attainable.  相似文献

We used banding techniques to analyze the karyotypes of 17 individuals ofCtenomys dorbignyi Contreras and Contreras, 1984, from populations in Corrientes (Mbarigüí, the type locality, and Sarandicito) belonging to the previously known range of the species, and Entre Ríos (Tiro Federal and Paso Vera) representing a new geographically isolated group of populations. All known populations have 2n=70 although the extreme geographic isolation of the different localities suggested the possibility of chromosomal divergence. We found three different karyomorphs; two of them (Mbarigüí and Sarandicito) had 6 pairs of biarmed autosomes and 28 pairs of telocentric autosomes, with an FN=80. Pair 20 (telocentric) was the carrier of the only detected NOR. However, pair 2 was clearly different in each of these two, being metacentric in Sarandicito and submetacentric in Mbarigüí. The third fixed karyomorph differed significantly from the others, showing 8 pairs of biarmed autosomes and 26 pairs of telocentric autosomes, with an FN=84. Pair 3 (biarmed) was the carrier of the single NOR detected. The NOR carrier also showed a marked heteromorphism (Corrientes vs Entre Ríos) evidenced by differences in the length of the short arm. These observations revealed a chromosomal polytypism that was expected owing to the disjunct geographic distribution of the species.  相似文献

真江蓠(Gracilaria vermiculophylla)是中国近海潮间带生态系统结构组成和功能维持的重要支撑物种, 但有关其群体遗传结构和多样性分布模式的研究目前仍较缺乏。本研究利用线粒体cox1序列对我国近海19个真江蓠地理群体进行了系统发育和群体遗传分析。461个长度为641 bp的cox1序列片段共含有21个多态位点, 产生15个单倍型。基于cox1序列的系统进化分析、单倍型分析和主成分分析显示, 19个真江蓠群体分化为南北两个类群, 其中浙江嵊泗以北的13个群体形成北方类群, 福建厦门以南的6个群体形成南方类群。遗传距离和分子方差分析显示真江蓠南北各类群内的遗传分化较小, 南北类群间的遗传分化达到亚种水平。南北类群间的差异是我国近海真江蓠群体遗传变异的主要来源。  相似文献

The details of mitochondrial DNA (mtDNA) phylogenetic structure of the northern grasshopper mouse Onychomys leucogaster were examined using populations from a postulated area of endemism that includes three arid regions (Colorado Plateaus, Interior Plains, and Wyoming Basins) in western North America. Fifteen tetra- and heptanucleotide restriction enzymes were used to assay restriction-site variation in a 2150-bp PCR-amplified fragment of mtDNA representing the ND2 and part of the COI gene regions. A total of 18 mtDNA haplotypes were detected. Although overall genetic divergence among these haplotypes was low (average = 1.1%), phylogeographic structuring was apparent. Notably, a clear phylogenetic split separated one group of haplotypes restricted to the Wyoming Basins from all others. This phylogenetic split was further corroborated by examination of nucleotide sequence variation from a 270-bp stretch of the mtDNA cytochrome b gene. Overall geographic and phylogenetic patterns suggest a complex history of geographic structuring and subsequent mixing of populations of grasshopper mice throughout the late Pleistocene. These patterns of variation are evaluated relative to alternative hypotheses about biotic responses to Quaternary climatic oscillations in western North American arid regions.  相似文献

The phylogenetic relationships among 23 individuals representing 14 species of underground hystricognath rodents of the genus Ctenomys were studied by analyzing variation of complete cytochrome b gene sequences. Maximum parsimony, neighbor joining, and maximum likelihood analyses were performed, using the octodontine genera Octodon and Tympanoctomys as outgroups. Our analyses support previous studies based on chromosomes and skull morphology that suggested a clade comprised of Argentinean and Uruguayan populations of C. rionegrensis. This clade is closely related to one comprised of C. flamarioni and the C. mendocinus species complex. Our analyses provide evidence that the symmetric sperm morph, which is common to other South American hystricognath rodents, is the plesiomorphic character state in Ctenomys and in Hystricognathi. Our analyses do not support the hypothesis that the sperm morphs define two major lineages of tuco-tuco species, because species with asymmetric sperm are diphyletic on the basis of cytochrome b sequences, and this morphology appears to have evolved twice in Ctenomys.  相似文献

African mole-rats (Bathyergidae, Rodentia) of the (eu)social genus Fukomys are one of the most speciose mammal genera endemic to Sub-Saharan Africa. Fukomys distributed in the Zambezian phytochorion is characterized by extreme chromosomal variation (2n=40-78). We inferred a molecular phylogeny of Zambezian Fukomys to resolve the interrelationships and the evolutionary history of the known chromosomal races. We sequenced the entire cytochrome b gene (1140bp) for a total of 66 specimens representing 18 karyotypical races from Zambia. An additional 31 sequences were retrieved from GenBank including data on all other chromosomal races. The haplotypes belonging to a small chromosomal race from Salujinga cluster with the Fukomys mechowii (Giant mole-rat) haplotypes. Differential degrees of chromosomal variation are observed among the major mole-rat clades, which is most pertinent when comparing the central Zambezian Fukomys micklemi and the northern Zambezian Fukomys whytei clades. The karyotypically hyper-diverse (12 known chromosomal races) Fukomys micklemi clade shows low levels of cytochrome b sequence divergence. Within the F. whytei clade we find a more conservative pattern of chromosomal diversification (three known chromosomal races) while the levels of sequence divergence are much higher then in the F. micklemi clade. Our results suggest that chromosomal changes may drive phyletic divergence and, eventually, speciation. The observed cladogenetic events during the Plio-Pleistocene within the F. mechowii, F. whytei, F. damarensis and F. micklemi clades appear to coincide with climatically mediated speciation bursts in other savannah dwelling mammals, including hominids. Based on the molecular data presented, combined with morphological and chromosomal data, the taxonomic implication seems to be that Fukomys may contain several (undescribed) cryptic species.  相似文献

Six Australian species of Coptotermes are traditionally recognized, but recent cuticular hydrocarbon studies suggest that some of these may represent more than one species. An understanding of the phylogenetic diversity of Australian Coptotermes, particularly the pest species, is likely to be important for the improvement of termite management strategies. A study of phylogenetic relationships among species of this genus was performed, based on the mitochondrial cytochrome oxidase (COII) gene, comparing the data with recent data from Asian species. Representatives of the species C. lacteus (Froggatt), C. frenchi Hill and C. michaelseni Silvestri were each found to form closely related monophyletic groups, however representatives of C. acinaciformis (Froggatt) were not. For C. acinaciformis, representatives from northern mound-building populations were found to form a distinct group to southern, tree-nesting forms. Among southern C. acinaciformis, two Western Australian representatives were found to be divergent from other populations. The results suggest that C. acinaciformis probably represents a complex of species rather than one, as has been suggested previously. One unidentified Coptotermes sp. taxon from Melbourne was found to be divergent from other taxa. Notably, some Australian species were more closely related to Asian species than other Australian species.  相似文献

We examined intra-specific phylogenetic relationships in leatherside chub, Gila copei. The complete mitochondrial (mt) cytochrome b gene (1140 bp) was sequenced for 30 individuals from 10 populations that span the geographical distribution of this species. Traditional phylogenetic analyses revealed two deeply divergent and evolutionarily distinct mtDNA clades that are geographically separated in northern and southern drainage basins. Interpopulation sequence variation between clades ranged from 7.7 to 8.1%. The northern clade was genetically more similar and phylogenetically more closely related to the selected out-group Lepidomeda m. mollispinus than to the southern clade, suggesting that the taxonomy of this species may require revision. Sequence variation among populations within clades ranged from 0 to 0.3% in the north and from 0 to 0.7% in the south. Statistical parsimony was used to construct phylogenetic networks of haplotypes within clades. Nested clade analysis revealed that geographical fragmentation has played an important role in genetic structuring within northern and southern clades.  相似文献
