The phylogenetic positions of Japanese Abies species (A. firma, A. homolepis, A. veitchii, A. sachalinensis, and A. mariesii) were revealed based on nucleotide sequences of chloroplast DNA. First, rbcL sequences of 24 Abies species worldwide were analyzed to clarify the phylogenetic position of the Japanese Abies within this genus. While 4 of the Japanese species formed a monophyletic group with almost no sequence divergence, A. mariesii was placed into a clearly different group with North American species. Second, to clarify the relationships among the species in the monophyletic group, sequences of rbcL, matK, and six spacer regions (total ca. 5 kb) were analyzed for 12 species of the group, with A. mariesii as an outgroup. Although a unique sequence and some species-specific sequences were detected in A. mariesii and in A. firma and A. homolepis, respectively, A. veitchii and A. sachalinensis gave identical sequences for all the sequenced regions.  相似文献

Cortical essential oil from Abies sachalinensis, A. veitchii, A. mariesii, A. firma, A. homolepis, and A. Kawakamii was analyzed for its mono and sesquiterpenoid constituents. Qualitatively the essential oils were similar to those from the New World firs. Using percent composition of monoterpenoid hydrocarbons, it was possible to completely separate most of the mentioned taxa from each other. A method was developed for calculation of similarity coefficients and construction of taxonomic structure on the basis of biosynthetic relationships between individual components of cortical oleoresin. The resulting chemosystematic dendrograms reflected similarity in material flow through various biosynthetic intermediates and paralleled in many features the morphological similarity relationships between the species studied.  相似文献

Sub-alpine Abies veitchii and A. homolepis are distributed in the central part of Honshu Island, Japan, and their habitats are segregated vertically. These species sometimes form a mixed forest in the overlapping area of the two species, that is, in the upper limit of the A. homolepis habitat and the lower limit of A. veitchii. These species have been considered to be distantly related because they were classified into different sections by most conventional classifications. No natural hybridization has been reported between the two species. The aim of this study was to demonstrate, through the use of molecular markers, whether natural hybridization takes place between these two species at two experimental sites on Mt. Fuji, where the species occur naturally. DNA markers from paternally inherited chloroplast DNA (cpDNA), maternally inherited mitochondrial DNA (mtDNA) and biparentally inherited nuclear DNA (nDNA), were used for this study. As organelle DNA markers, polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) markers were developed to determine the maternal and paternal species for each individual. Two of 334 individuals possessed a cpDNA haplotype derived from A. homolepis and a mtDNA haplotype from A. veitchii. Furthermore, the nDNA of these two individuals was analysed using the random amplified polymorphic DNA (RAPD) assay to investigate their genomic composition. RAPD analysis indicated that the nuclear genomes of the two individuals were derived from both species. We conclude that A. veitchii and A. homolepis produce natural hybrids, and that their systematic relationship should be re-evaluated.  相似文献

Abstract Two types of isometric virus-like particles (VLPs) with diameters of c. 20 and 35 nm were detected in partially purified extracts from Abies alba Mill., A. homolepis Sieb. et Zucc., and five other Abies species. An immunoelectron microscopical test was developed by means of which the large VLPs could often be detected even in crude sap preparations. They were to be widespread in A. alba and A. homolepis in the Federal Republic of Germany and Southern France. Theirpresence in glasshouse-grown seedlings of A. alba and A. homolepis shows that they are seed-transmissible. The small particles were detected only in partically purified preparations. Neither type of VLP was found in coniferous genera otherthan Abies.  相似文献

Essential oils were isolated from the leaves of Platycladus orientalis growing in 16 areas of China. The essential oils were analyzed by gas chromatography and gas chromatography-mass spectrometry. In total, 98 volatile compounds were identified. Chemical variability in essential oil composition was evaluated using cluster analysis and principal component analysis. The two analyses led to the identification of four chemotypes: α-pinene, α-pinene/3-carene, cedrol, and cedrol/terpinyl acetate. Geographically, the populations growing in close proximity had similar essential oil composition. The chemical variability could possibly be attributed to genetic and environmental factors.  相似文献

Citrus aurantium L. var. amara Engl., is a better species of sour oranges. There are essential oils in the flowers, the peels, the leaves and the branches of C. aurantium. The flower oil can be used in the preparation of perfumes of high quality. The peel oil is used mainly for the flavor-endowing of soft drinks, alcoholic drinks, bread, confectionaries and cakes. In order to control the quality of the essential oils and to improve them, we have systema- tically studied the chemical constituents of the flowers, the leaves and the peals of C. aurantium with our preparation. 12 main components were separated by silica gel column chromatography. The following 33 chemical components were identified by IR, GC-MS and GC retention index: α-thujene, α-pinene, camphene, β-pinene, myrcene, limonene, β-ocimene, trans-linalooloxide (furanoid), cis-linalooloxide (furanoid), linalool, 1,4-p-methadien-7-ol, trans-pinocarveol, camphor, terpinen-4-ol α-terpineol, nerol, citral-b, geraniol, linalylacetate, citrala, trans-linalooloxide (pyranoid), methyl anthranilate, terpinyl acetate, cis-linalooloxide (pyranoid), neryl acetate. geranyl acetate, nonanal, β-caryophyllene, α-humulene, γ-muurolene, β-nerolidol, farnesol, α- nerolidol. GC retention index of 33 compounds were measured. A fast method for routine determination is presented.  相似文献

水翁的花营和鲜叶经水蒸汽蒸馏得到一种淡黄色的精油,前者出油率为0.18%。后者为0.08%。我们应用毛细管气相色谱,气相色谱/质谱联用,红外光谱和紫外光谱等方法,对两种精油进行化学分成分析,分别鉴定出35个和27个已知化学成分。两者相同的化学成分有:β-罗勒烯(Z)、β-罗勒烯(E)、α-蒎烯、β-蒎烯、月桂烯、小茴香烯、香叶醇、顺式-丁香烯、橙花叔醇等23个化学成分,分别占花营精油全油的90%和叶精油95%以上。  相似文献

Plants of damiana(Turnera diffusa Willd.) are important to industry and traditional medicine in semi-arid climates. Although all populations are wild, no reports have been made previously of their different phenotypes. Here, we investigated various micromorphological characteristics and the levels of essential oils in two phenotypes. Oils were extracted from fresh leaves via hydrodistillation and analyzed by gas chromatography-mass spectrometry. Morphological analyses were conducted under a stereoscopic microscope and with a scanning electron microscope. In all, 56 compounds were identified, enabling us to distinguish separate phenotypes. DL1 plants mainly contained 1,8-cineole, 10-epi y eudesmol, and guaiol; whereas those of DL2 primarily constituted ß-pinene, ß-caryophyllene oxide, cadinene, and α-cadinol. These two phenotypes also differed in their morphologies, with DL1 leaves showing elevated essential oil concentrations, but lacking trichomes. In contrast, the DL2 plants had lower contents of essential oils but did possess trichomes on their abaxial and adaxial leaf surfaces. This documentation of individual damiana phenotypes is the initial process toward validating the quality of essential oils from this species as well as inherent structural variations.  相似文献

The chemical composition of the essential oils obtained by hydrodistillation from leaves, branches and female cones of Cupressus arizonica Greene cultivated in Tunisia was determined by GC and GC/MS analysis. Significant differences were found between the constituent percentages of the different oils. Among the 87 identified components α-pinene (60.5% in female cones), umbellulone (18.4% in leaves), δ-3-carene (15.6% in branches) and cis-muurola-4(14),5-diene (9.4% in leaves) were found to be the major ones.Composition of essential oils extracted from different organs of C. arizonica Greene growing in Tunisia showed remarkable differences from the same species cultivated in Algeria, Argentina, Iran, Italy, France and Texas based on a comparison with published results. The in vitro antibacterial activity of the essential oils samples was evaluated against some Gram positive and negative bacteria.  相似文献

Twenty seven chemical constituents of oils from sweet leaves, flowers and fruits of Foeniculum vulgare Mill. are examined by GC and GC-MS with both different chromatographic columns. They are 1,1-diethoxyethane, α-thujene, α-pinene, camphene, sabinene, β-pinene, myrcene, α- phellandrene, p-cymene, limonene, cineole, γ-terpinene, fenchone, camphor, terpinen-4-ol, α-terpineol, estragole, verbenone, fenchol acetate, carveol, trans-fenchol acetate, carvone, anethole, anisaldehyde, trans-anethole, methoxyphenyl acetone and benzoic acid, 4-methoxy-, othylester. The limonene is 57.8% in the essential oil from leaves, 34.2% from flowers, 13.1% from fruits, The trans-anethole is 21.8% in the essential oil from leaves, 41.2% from flowers, 63.4% from fruits.  相似文献

Leaves of seven species of the genus Eucalyptus L'Hér., viz., E. cladocalyx F. Muell., E. citriodora Hook., E. diversicolor F. Muell., E. fasciculosa F. Muell., E. grandis W. Hill, E. ovata Labill., and E. botryoides Sm., were harvested from Zerniza arboreta (region of Sejnene, northwest of Tunisia) in June 2007. Of the latter species, leaves were collected from trees having two origins, Morocco and Italy. Hydrodistillation of the dried leaves provided essential oils in yields varying from 0.4±0.0 to 3.3±0.1%, according to the species. E. citriodora had the highest mean percentage of essential oil amongst the species examined, whereas the lowest one was obtained for E. botryoides originating from Morocco. Analysis by GC (RI) and GC/MS allowed the identification of 140 compounds, representing 92.5 to 99.4% of the total oil composition. The contents of the different samples varied according to the species. The main components were 1,8-cineole (2), followed by α-pinene (1), p-cymene, borneol, α-terpineol, cryptone, spathulenol, trans-pinocarveol (4), bicyclogermacrene (5), caryophyllene oxide, and β-phellandrene. Principal components analysis and hierarchical cluster analysis separated the eight Eucalyptus leaf essential oils into five groups, each constituting a chemotype.  相似文献

Mitochondrial DNA polymorphism of 40 populations of five Abies species was investigated using PCR-amplified coxI and coxIII gene probes. Using four combinations of probe and restriction enzyme, we detected three major haplotypes and 15 total haplotypes. We also found varied levels of gene diversity for the different species: 0.741, 0.604, 0.039, 0.000, and 0.292 for A. firma, A. homolepis, A. veitchii, A. mariesii, and A. sachalinensis, respectively. The marginal and southern populations of A. firma and A. homolepis have unique haplotypes, especially the Kyushu, Shikoku, and Kii Peninsula populations, which inhabit areas coinciding with probable refugia of the last glacial period and possess high levels of mtDNA genetic diversity. The haplotypes in some populations suggested mtDNA capture also occurred between species through introgression/hybridization. The strong mtDNA population differentiation in Abies is most likely due to the maternal inheritance of mitochondria and restricted seed dispersal. A phenetic tree based on the genetic similarity of the mtDNA suggests that some species are polyphyletic. Based on mtDNA variation, the five Abies species could be divided roughly into three groups: (1) A. firma and A. homolepis, (2) A. veitchii and A. sachalinensis, and (3) A. mariesii. However, we found that all these Abies species, except A. mariesii, are genetically very closely related according to an analysis of their cpDNA sequences. This showed that the chloroplast rbcL gene differed by only one base substitutions among the four species. We believe that the mtDNA variation and cpDNA similarity clearly reflect relationships among, and the dissemination processes affecting these Abies species since the last glacial period.  相似文献

The species differentiation between Chamaecyparis formosensis, C. obtusa var. formosana, and C. obtusa, based on the composition of the leaf essential oils, was studied. The characterization of the oils by GC-FID and GC/MS analyses showed remarkable differences between these three essential oils. Cluster analysis (CA) and principal-component analysis (PCA) distinguished three groups of essential oils. The C. formosensis oil was dominated by α-pinene while those isolated from C. obtusa var. formosana and C. obtusa were characterized by high levels of (-)-thujopsene and α-terpinyl acetate, respectively. Moreover, the phylogenetic relationships of the genus Chamaecyparis were in agreement with previous findings based on morphological and molecular evidence. In addition, the essential oils from C. obtusa var. formosana could be classified into three chemical types, according to their different characteristic main compounds (β-elemol, (-)-thujopsene, and cis-thujopsenal). The biochemical correlations between the major constituents of the Chamaecyparis species were examined and their relationship is discussed.  相似文献

Abies nebrodensis (Lojac .) Mattei (Pinaceae) is a species occurring in a very small population only in a restricted area of Sicily. Its taxonomic classification as different species has been object of discussion. In this work the chemical composition of the essential oil from the leaves is presented for the first time and compared to the essential oils from other euroasiatic species reported in literature. The qualitative and quantitative composition of the essential oils obtained from the inflorescences of Achyrocline flaccida (Asteraceae) has been investigated for the first time. Plant material was collected from eleven locations in Argentina. The essential oils were obtained by hydrodistillation (0.1–0.8% v/w, dried material) and analyzed by GC–FID–MS. Eighty-three compounds were identified representing more than the 90% of the oils. The major components were α-pinene and β-caryophyllene. Statistical analysis was performed in order to evaluate the variability of the essential oils analyzed. Two groups were formed reflecting only quantitative differences in the content of major compounds. The chemical pattern of essential oils observed for A. flaccida is similar to other Achyrocline species studied, except Achyrocline hyperchlora.  相似文献

The aim of current work was to determine essential oils (EOs) composition from three Eucalyptus species, including E. citriodora, E. camaldulensis and E. globulus and assess their α-glucosidase inhibitory activity. The EOs were collected using the hydrodistillation technique and characterized by GC/MS, GC-FID and NMR. The isolated EOs from leaves parts of Eucalyptus species varied from 0.56 to 1.0 % on fresh weight basis. The content of the EOs was distinct according to the species. The most abundant metabolites were identified as citronellal (0–83.0 %), 1,8-cineole (0.2–44.8 %), spathulenol (0.4–16.1 %) α-pinene (0.4–15.9 %), p-cymene (3.7–11.9 %), citronellol (0–8.6 %), β-eudesmol (5.3–8.6 %) and β-pinene (0–7.1 %). The EOs obtained from targeted samples exhibited strong α-glucosidase inhibitory activity. These results are encouraging and underline that the EOs of Eucalyptus species may be a promising alternative source of natural antidiabetic.  相似文献

Cortial essential oil from 395 typical Abies grandis and Abies grandis-Abies concolor intermediate trees collected in 48 locations was analyzed for composition of its monoterpenoids. The monoterpenoids were composed primarily of α-pinene, camphene, β-pinene, β-phellandrene and bornyl acetate, with tricyclene, 3-carene, myrcene and limonene in secondary quantities. Camphene, present in larger amounts in A. grandis and in smaller amounts in A. concolor var. lowiana, was used as a marker for studying intergradation of the two species in western and northwestern United States. A. grandis from northern Montana, Idaho and Washington, from British Columbia and Oregon and from Washington coastal areas could be classified as typical while the rest of the populations showed various degrees of intermediacy. The variability patterns agreed with the hypothesis that introgressive hybridization was the main reason for the intermediacy. Compared with morphological evaluations, the intermediate populations were chemically significantly more A. concolor-like. Typical A. grandis exhibited little population-to-population variability, in spite of its large geographic range. The results are correlated with and explained on the basis of existing paleobotanical records.  相似文献

The essential oil of the fresh leaves of Platycladus orientalis (L.), grown in four different biogeographic zones of Jordan,- (the Mediterranean, Irano-Turanian, Saharo-Arabian, and Sudanian penetration) -, were obtained by hydrodistillation and analysed by gas chromatography (GC) and gas chromatography-mass-spectrometry (GC/MS). The actual composition of the spontaneous emitted volatiles was obtained using the solid-phase-micro-extraction (SPME) method and investigated using the same chromatographic and spectroscopic methods. Hydrocarbon monoterpenes dominated the hydrodistilled oils and emissions of all regions. Bicyclic monoterpenes (sabinene, α-pinene, and α-thujene) and monocyclic α-terpinene were detected as the major constituents of the oils and emissions. Additionally, hierarchical cluster analysis (HCA) revealed that the clustering is based on the region of collection rather than the applied methodology. Differences were observed in the quantity of the obtained oils (P-values <0.01); the highest amount of volatile oil was obtained from samples grown in the Irano-Turanian biogeographic zone.  相似文献
