The effects of elevated dietary calcium (as CaCO3) and acute waterborne Cd exposure (50 microg/l) on whole body uptake, tissue uptake, and internal distribution of newly accumulated Cd, Ca2+, and Na+ in juvenile rainbow trout were examined. Fish were fed with three diets (mg Ca2+/g food): 20 (control), 30 and 60 for 7 days before fluxes were measured with radiotracers. The highest dietary Ca2+ elevation reduced waterborne whole body Ca2+ uptake, but did not protect against inhibition of waterborne Ca2+ uptake by waterborne Cd. Both Ca2+-supplemented diets reduced newly accumulated Ca2+ in the gills in relation to the control treatment, but did not prevent the Cd-inhibiting effect against accumulation of new Ca2+ in most compartments. Fish fed with Ca2+-supplemented diets showed markedly lower rates of whole body uptake and internalization (in some tissues) of waterborne Cd, illustrating that, while dietary Ca2+ supplementation did not protect against the impact of waterborne Cd on waterborne Ca2+ uptake, it did protect against the uptake of Cd. Waterborne Cd had no effect on Na+ fluxes, total Cl-, and in most body compartments, newly accumulated Na+ and total Na+ were also not affected. Dietary supplementation with CaCO3 had the same protective effect as demonstrated by dietary supplementation with CaCl2 in an earlier study. Thus, the reduction of waterborne Cd uptake and internalization by dietary Ca2+ was specifically due to Ca2+ and not to the anion.  相似文献

The recent increased prevalence of uterine prolapses in sows around parturition has led to inferences that the prolapses may be associated with hypocalcemia. However, limited data are available to support that hypocalcemia occurs in sows. Hypocalcemia in dairy cows is associated with feeding excess dietary Ca during late gestation. The excess Ca is assumed to suppress homeostatic mechanisms critical to maintain serum Ca concentrations as the Ca demand increases during the early stages of lactation. In this experiment, sows were fed diets with excess Ca during late gestation and early lactation to assess the potential development of hypocalcemia in the peripartum period. Twelve crossbred (Large White × Landrace) multiparous gestating sows were fed a control diet (CON), 0.65% Ca to 0.38% standardized total tract digestible P (STTD P) and 0.67% Ca to 0.38% STTD P in gestation and lactation diets, respectively) or a high Ca diet (HCa, 1.75% Ca to 0.46% STTD P and 1.75% Ca to 0.45% STTD P in gestation and lactation diets, respectively). The diets were fed from gestation day 86 þ ± 1 until the end of lactation (27 þ ± 2 days period). On day 112 of gestation, indwelling venous catheters were placed in each sow. Blood samples were collected at 15-min intervals within four designated times (0700, 1000, 1300 and 1700 h) on gestation day 113 and lactation days 1, 3 and 5. Venous blood pH, gases (pO2, pCO2 and HCO3), electrolytes (K+, Na+ and Cl), ionized Ca (iCa), metabolites (glucose and lactate), plasma total Ca (tCa), and P were analyzed. Overall, sows fed HCa diet had greater (P < 0.001) concentrations of blood iCa and plasma tCa than sows fed CON diets. No clinical signs of Ca metabolism disorders were observed. Unexpectedly, concentrations of plasma P in sows fed HCa diets were lower (P < 0.001) than in sows fed CON diets. Plasma P tended to decrease (P = 0.057) as day of lactation increased. Differences between dietary treatments for blood pH, gases, electrolytes and metabolites were not detected (P > 0.05). No evidence for hypocalcemia was detected in peripartum sows fed CON or HCa diets. These data imply that excess Ca in late gestation diets did not result in hypocalcemia during the peripartum period. Future experiments should focus on factors other than hypocalcemia to identify causes of uterine prolapses in sows.  相似文献

This study investigated the effects of dietary Ca2+ on branchial Ca2+ and Zn2+ uptake, new and total zinc accumulation in target tissues (gill, liver and kidney), calcium and zinc homeostasis, and acute tolerance to waterborne zinc in fish chronically exposed to waterborne zinc. Juvenile rainbow trout (Oncorhynchus mykiss) were maintained on a calcium-enriched diet [41.2 mg vs. 21.2 mg (control) calcium/g dry wt. of food] and chronic waterborne zinc exposure (2.3 micromol/L), both separately and in combination, for 28 days. Calcium-supplemented diet in the absence of waterborne zinc significantly reduced branchial Ca2+ and Zn2+ influx rates, and new and total zinc accumulations in target tissues relative to control. However it did not protect against the acute zinc challenge. In contrast, waterborne zinc exposure significantly increased branchial Ca2+ and Zn2+ influx rates, new and total zinc concentrations in target tissues, and acute zinc tolerance relative to control. Interestingly, no such changes in any of these parameters were recorded in fish treated simultaneously with elevated dietary Ca2+ and waterborne zinc, except acute zinc tolerance which was highest among all the treatments. Thus, we conclude that the interactions between elevated dietary Ca2+ and waterborne zinc can protect freshwater fish against waterborne zinc toxicity.  相似文献

We evaluated the differential nature of interactions between waterborne Ca and Cd transport in the gills of yellow perch (Perca flavescens) and rainbow trout (Oncorhynchus mykiss), two species with a more than 400-fold difference in acute waterborne Cd tolerance. The Jmax (maximum rate of uptake) and Km (inverse of affinity) for Ca uptake, in the absence of Cd, were significantly lower in yellow perch (120.48 nM g–1 wet wt h–1 and 92.17 M, respectively) relative to rainbow trout (188.68 nM g–1 wet wt h–1 and 243.90 M, respectively). Similarly, the Jmax for Cd uptake, at the lowest waterborne Ca level (100 M) tested, was significantly lower in yellow perch (0.27 nM g–1 wet wt h–1) relative to rainbow trout (0.40 nM g–1 wet wt h–1), but no significant difference was observed in the Km values between the two species (yellow perch: 32.47 nM; rainbow trout: 31.27 nM). Waterborne Cd (0–890 nM) as well as waterborne Ca (100–1,000 M) competitively inhibited branchial uptake of each other in both species. However, analyses of inhibitor constants for branchial Ca uptake by waterborne Cd ( ) revealed that the inhibition was about 1.8 times more potent in rainbow trout compared to yellow perch. In contrast, analyses of inhibitor constants for branchial Cd uptake by waterborne Ca ( ) indicated that the inhibition was more than three fold more potent in yellow perch than in rainbow trout. Higher branchial Ca uptake and more potent inhibition by Cd as well as higher branchial Cd uptake and less potent inhibition by Ca were also reflected in whole-body measurements of Ca and Cd influx in trout relative to perch. Overall, whole-body effects were in accord with the branchial kinetic analyses. These results further strengthen the conclusion that branchial influxes of Ca and Cd occur through common pathways. Rainbow trout (Oncorhynchus mykiss) were fed either a control diet containing fish oil or an essential fatty acid (EFA) deficient diet containing only hydrogenated coconut oil and palmitic acid as lipid source (93.4% saturated fatty acids) for 14 weeks and the fatty acid compositions of individual phospholipid classes from skin and opercular membrane (OM) determined. The permeability of skin and OM to water and the production of eicosanoids in skin and gills challenged with the Ca2+ ionophore A23187 were also measured. Phospholipid (PL) fatty acid compositions were substantially modified in EFA-deficient fish, with increased saturated fatty acids and decreased polyunsaturated fatty acids (PUFA), especially arachidonic acid (AA) and eicosapentaenoic acid (EPA), while docosahexaenoic acid (DHA) was largely retained. The onset of EFA deficiency was shown by the appearance of n-9 PUFA, particularly 20:3n-9. The main effects of EFA deficiency on phosphatidylcholine (PC) and phosphatidylethanolamine (PE) were to increase saturated fatty acids and monoenes, especially 16:1 and 18:1, and to decrease EPA and DHA. The content of DHA in phosphatidylserine (PS) was high in control animals (40% in skin and 35% in opercular membrane) and was mostly retained in EFA deficient animals. Arachidonic acid (AA) was the most abundant PUFA esterified to phosphatidylinositol (PI) and was significantly reduced in EFA deficient animals (from 31% to 13% in skin), where a large amount of 20:3n-9 (9% in skin) was also present. Influxes and effluxes of water through skin and opercular membrane were measured in vitro. No differences were detected between rainbow trout fed the control or the EFA deficient diet. 12-Hydroxyeicosatetraenoic acid (12-HETE), 12-hydroxyeicosapentaenoic acid (12-HEPE) and 14-hydroxydocosahexaenoic acid (14-HDHE) could not be detected in skin from control or EFA deficient fish. There was no difference between control and EFA deficient trout in the levels of leukotriene C4 (LTC4) and leukotriene C5 (LTC5) in skin cells challenged with the calcium ionophore A23187, and of prostaglandin F (PGF), 12-HETE and 12-HEPE in gill cells challenged similarly. Prostaglandin F (PGF) production by ionophore stimulated gill cells was significantly reduced in fish fed the EFA-deficient diet. 14-HDHE produced by gill cells was 3.3 fold higher in EFA deficient fish compared to controls.  相似文献

A suite of respiratory, acid-base, ionoregulatory, hematological, and stress parameters were examined in adult rainbow trout (Oncorhynchus mykiss) after chronic exposure to a sublethal level of dietary Cd (500 mg/kg diet) for 45 days and during a subsequent challenge to waterborne Cd (10 microg/L) for 72 h. Blood sampling via an indwelling arterial catheter revealed that dietary Cd had no major effects on blood gases, acid-base balance, and plasma ions (Ca(2+), Mg(2+), K(+), Na(+), and Cl(-)) in trout. The most notable effects were an increase in hematocrit (49%) and hemoglobin (74%), and a decrease in the plasma total ammonia (43%) and glucose (49%) of the dietary Cd-exposed fish relative to the nonexposed controls. Dietary Cd resulted in a 26-fold increase of plasma Cd level over 45 days (approximately 24 ng/mL). The fish exposed to dietary Cd showed acclimation with increased protection against the effects of waterborne Cd on arterial blood P(aCO2) and pH, plasma ions, and stress indices. After waterborne Cd challenge, nonacclimated fish, but not Cd-acclimated fish, exhibited respiratory acidosis. Plasma Ca(2+) levels declined from the prechallenge level, but the effect was more pronounced in nonacclimated fish (44%) than in Cd-acclimated fish (14%) by 72 h. Plasma K(+) was elevated only in the nonacclimated fish. Similarly, waterborne Cd caused an elevation of all four traditional stress parameters (plasma total ammonia, cortisol, glucose, and lactate) only in the nonacclimated fish. Thus, chronic exposure to dietary Cd protects rainbow trout against physiological stress caused by waterborne Cd and both dietary and waterborne Cd should be considered in determining the extent of Cd toxicity to fish.  相似文献

The 109Cd binding assay of Eaton & Toal was critically evaluated and then used to assess the induction of cytosolic metal-binding ligands in rainbow trout exposed to Zn in the diet and/or in the water for 16 weeks. With purified rabbit Cd-Zn metallothionein (MT), 109Cd binding and total Cd recovery in the assay were linear up to 5 μg of protein; gel chromatography revealed a single peak. With heat-denatured extracts of gill, liver and intestine from control and Cd- and Zn-injected trout, 109Cd binding was generally linear with sample size. Gel chromatography demonstrated that 109Cd was bound by a protein with the same apparent weight as MT (∼ 11000 daltons), but significant binding occurred also at three other regions [molecular weight (mol wt) >70 000, 30000 and <3000]. In the dietary/waterborne Zn exposure, induced 109Cd-binding activity occurred not in the MT peak but in the low mol wt peak (< 3000). Activity in the gill rose in response to both dietary and waterborne Zn, but the liver did not respond. The maximum five-fold elevation in the gill was primarily a waterborne effect. In the intestine, the maximum rise was 25-fold due to both factors. The thresholds for induction were > 39 μg Zn| in water, and > 90 mg kg | in the diet, but only when waterborne Zn was also high. There was no correlation between 109Cd binding and acid soluble thiol levels, which tended to decline at higher Zn exposures.  相似文献

Porphyra dioica meal was added at levels of 5, 10 and 15% to a diet for rainbow trout formulated to be isonitrogenous and isolipidic. The control diet was a commercial trout diet without seaweed meal. The experimental groups were fed in triplicate for 12.5 weeks, during which fish weight increased on average from 107–261 g. Seaweed meal inclusion did not affect significantly weight gain (WG), specific growth rate (SGR), feed efficiency (FE), protein efficiency ratio (PER) and apparent digestibility coefficient of the dry matter (ADCdm) for any of the diets. Voluntary feed intake (VFI) increased for all seaweed diets compared to the control diet but not significantly (P?>?0.05). Final weight (FW) was significantly smaller for the 15% P. dioica inclusion and hepatosomatic index (HSI) for the 10% and 15% inclusion. Carcass protein content increased for all three experimental diets, and was significantly higher for the diet with 10% seaweed inclusion. Rainbow trout fed with Porphyra meal presented a dark orange pigmentation of the flesh at the end of the trial, compared to the whitish color from the control fish. These results suggest that P. dioica can effectively be included in diets for rainbow trout up to 10% without significant negative effects on weight gain and growth performance. The pigmentation effect of the fish flesh by adding P. dioica meal to the feed is of a considerable interest to the organic salmon-farming industry.  相似文献

The effects of supplementing diets with acetone extract (1% w/w) from four medicinal plants (Bermuda grass Cynodon dactylon, H1, beal Aegle marmelos, H2, winter cherry Withania somnifera, H3 and ginger Zingiber officinale, H4) on growth, the non‐specific immune response and ability to resist pathogen infection in tilapia Oreochromis mossambicus were assessed. In addition, the antimicrobial properties of the extract were assessed against Vibrio alginolyticus, Vibrioparahaemolyticus, Vibrio mimicus, Vibrio campbelli, Vibrio vulnificus, Vibrio harveyi and Photobacterium damselae. Oreochromis mossambicus were fed 5% of their body mass per day for 45 days, and those fed the experimental diets showed a greater increase in mass (111–139%) over the 45 days compared to those that received the control diet (98%). The specific growth rate of O. mossambicus fed the four diets was also significantly greater (1·66–1·93%) than control (1·52%) diet‐fed fish. The blood plasma chemistry analysis revealed that protein, albumin, globulin, cholesterol, glucose and triglyceride levels of experimental fish were significantly higher than that of control fish. Packed cell volume of the blood samples of experimental diet‐fed fish was also significantly higher (34·16–37·95%) than control fish (33·0%). Leucocrit value, phagocytic index and lysozyme activity were enhanced in fish fed the plant extract‐supplemented diets. The acetone extract of the plants inhibited growth of Vibrio spp. and P. damselae with extracts from W. somnifera showing maximum growth inhibition. A challenge test with V. vulnificus showed 100% mortality in O. mossambicus fed the control diet by day 15, whereas the fish fed the experimental diets registered only 63–80% mortality at the end of challenge experiment (30 days). The cumulative mortality index for the control group was 12 000, which was equated to 1·0% mortality, and accordingly, the lowest mortality of 0·35% was registered in H4‐diet‐fed group.  相似文献

Juvenile rainbow trout (Oncorhynchus mykiss) were exposed to waterborne Cu (22 microg/l) in moderately hard water for up to 28 days. Relative to control fish kept at background Cu levels (2 microg/l), Cu-preexposed fish displayed decreased uptake rates of waterborne Cu via the gills but not of dietary Cu via the gut during 48-h exposures to (64)Cu-radiolabeled water and diet, respectively. At normal dietary and waterborne Cu levels, the uptake rates of dietary Cu into the whole body without the gut were 0.40-0.90 ng. g(-1). h(-1), >10-fold higher than uptake rates of waterborne Cu into the whole body without the gills, which were 0.02-0.07 ng. g(-1). h(-1). Previously Cu-exposed fish showed decreased new Cu accumulation in the gills, liver, and carcass during waterborne (64)Cu exposures and in the liver during dietary (64)Cu exposures. A 3-h gill Cu-binding assay showed downregulation of the putative high-affinity, low-capacity Cu transporters and upregulation of the low-affinity, high-capacity Cu transporters at the gills in Cu-preexposed fish. Exchangeable Cu pools in all the tissues were higher during dietary than during waterborne (64)Cu exposures, and previous Cu exposure reduced waterborne exchangeable Cu pools in gill, liver, and carcass. Overall, these results suggest a quantitatively greater role for the dietary than for the waterborne route of Cu uptake, a key role for the gill in Cu homeostasis, and important roles for the liver and gut in the normal metabolism of Cu in fish.  相似文献

Two semisynthetic diets differing in the levels of pantothenic acid (PA) supplementation (0 or 40 mg kg−1 diet) were fed to rainbow trout, Salmo gairdneri Richardson, initially weighing 0.7 g fish−1. Each diet was fed to two tanks of fish and, at the start of the feeding trial, each tank contained 200 animals. The experiment was conducted for 28 days during which time, every 2 days, gill tissue was sampled and food consumption was determined. Lamellar hyperplasia was first detected in the gills of deficient fish on the 12th day sample, while reduced feed intake was first manifest at 16 days. Hyperplasia first appeared in the distal regions of the gill filaments, but the lesion rapidly progressed in a proximal direction and 35 of 40 fish examined on days 22–28 exhibited hyperplasia on more than 75% of the filament surface. Gill lamellar hyperplasia is a sensitive indicator of PA deficiency in the rainbow trout. Moreover, the lesion is specific to PA deficiency since its developmental pattern differs histologically from the lamellar hyperplasia of the non-nutritional gill diseases.  相似文献

Effects of Hilyses®, fermented Saccharomyces cerevisiae (S. cerevisiae), on growth, body composition and skin mucus immune components in rainbow trout were quantified. Ninety rainbow trout (105 ± 5 g) were randomly assigned to 2 groups in triplicates and fed dietary Hilyses® (5 g kg?1) or control diet without Hilyses® for 50 days. Results of this study demonstrated that growth performance increased significantly by the dietary yeast supplement; however body composition was not affected in treatment group. At the 45th and 50th day of feeding trial, results of mucus samples demonstrated that yeast supplementation in treatment group significantly promoted enzyme activities, namely lysozyme, protease, alkaline phosphatase and esterase compared to control group. Significant increases were also observed in hemagglutination and antibacterial activity against Yersinia ruckeri in fish fed treatment diet. The present study suggests that fermented S. cerevisiae may effectively promote the growth performance and skin non-specific immune parameters in rainbow trout.  相似文献

The effects of dietary Ergosan on the growth performance and mucosal immunity in rainbow trout skin were investigated. 60 rainbow trout (100-110 g) were randomly assigned to 2 groups in triplicates and fed one of the experimental diet formulated with 5 g kg−1 Ergosan or control diet for 50 days. Results showed that on the 45th day of feeding trial, Ergosan supplementation significantly enhanced the growth performance compared to control group. Various enzyme activities, namely lysozyme, protease, alkaline phosphatase and esterase in treatment group were also enhanced on the 45th and 50th day. Skin mucus in Ergosan-fed fish showed the agglutination of erythrocytes while in control group, no visible agglutination was shown. In addition, skin mucus in treatment group showed strong antibacterial activity against Yersinia ruckeri. In conclusion, the major immune components of rainbow trout mucus that are involved in the non-specific immunity were enhanced by administration of Ergosan in 5 g kg−1.  相似文献

Renal function was examined in adult rainbow trout (Oncorhynchus mykiss) after chronic exposure to a sublethal level of dietary Cd (500 mg/kg diet) for 52 d and during a subsequent challenge to waterborne Cd (10 microg/L) for 72 h. Dietary Cd had no major effects on UFR (urine flow rate) and GFR (glomerular filtration rate) but caused increased renal excretion of glucose, protein, and major ions (Mg(2+), Zn(2+), K(+), Na(+), Cl(-) but Ca(2+)). However, dietary Cd did not affect any plasma ions except Na(+) which was significantly elevated in the Cd-acclimated trout. Plasma glucose and ammonia levels fell by 25% and 36% respectively, but neither plasma nor urine urea was affected in Cd-acclimated fish. Dietary Cd exposure resulted in a remarkable increase of Cd load in the plasma (48-fold, approximately 22 ng/mL) and urine (60-fold, 8.9 ng/mL), but Cd excretion via the kidney was negligible on a mass-balance basis. Clearance ratio analysis indicates that all ions, Cd, and metabolites were reabsorbed strongly (58-100%) in both na?ve and dietary Cd exposed fish, except ammonia which was secreted in both groups. Mg(2+), Na(+), Cl(-) and K(+) reabsorption decreased significantly (3-15%) in the Cd-exposed fish relative to the control. Following waterborne Cd challenge, GFR and UFR were affected transiently, and only Mg(2+) and protein excretion remained elevated with no recovery with time in Cd-acclimated trout. Urinary Ca(2+) and Zn(2+) excretion rates dropped with an indication of renal compensation towards plasma declines of both ions. Cadmium challenge did not cause any notable effects on urinary excretion rates of metabolites. However, a significant decrease in Mg(2+) reabsorption but an increase in total ammonia secretion was observed in the Cd-acclimated fish. The study suggests that dietary Cd acclimation involves physiological costs in terms of renal dysfunction and elevated urinary losses.  相似文献

This study aimed to assess the protective roles of polysaccharides from Agaricus blazei Murill (ABP) against cadmium (Cd)-induced damage in chicken livers. A total of 80 Hy-Line laying chickens (7 days old) were randomly divided into four groups (n = 20). Group I (control) was fed with a basic diet and 0.2 ml saline per day, group II (Cd-treated group) was fed with a basic diet containing 140 mg/kg cadmium chloride (CdCl2) and 0.2 ml saline per day, group III (Cd + ABP-treated group) was fed with a basic diet containing 140 mg/kg CdCl2 and 0.2-ml ABP solution (30 mg/ml) per day via oral gavage, and group IV (ABP-treated group) was fed with 0.2-ml ABP solution (30 mg/ml) per day via oral gavage. The contents of Cd and malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), the messenger RNA (mRNA) levels of inflammatory cytokines and heat shock proteins (HSPs), the protein levels of HSPs, and the histopathological changes of livers were evaluated on days 20, 40, and 60. The results showed that Cd exposure resulted in Cd accumulating in livers and inhibiting the activities of antioxidant enzymes (SOD and GSH-PX). Cd exposure caused histopathological damage and increased the MDA content, the mRNA levels of inflammatory cytokines (TNF-α, IL-1β, and IL-6) and HSPs (HSP27, HSP40, HSP60, HSP70, and HSP90) and the protein levels of HSPs (HSP60, HSP70, and HSP90). ABP supplementation during dietary exposure to Cd reduced the histopathological damage and decreased the contents of Cd and MDA and the expression of inflammatory cytokines and HSPs and improved the activities of antioxidant enzymes. The results indicated that ABP could partly ameliorate the toxic effects of Cd on chicken livers.  相似文献

In the enclosed fresh-water environmsnt of Hanningfield Reservoir, Essex, England, Anisakis sp. larvae (parasites of marine fish) were found in 55 per cent of 40 brown trout and in 26·2 per cent of 61 rainbow trout. Experimental infection by intubating larvae into the stomach was more successful in brown trout (50·6 per cent recovery rate) than in rainbow trout (27 per cent recovery rate). Some larvae reached the body-cavity as early as 2 h after infection. They penetrated the region between the oesophagus and intestine immediately posterior to the caecal openings. Fewer larvae successfully penetrated the gut wall of brown trout within 24 h at 8°C than at 15 ± 1°C. It appears that the reservoir trout acquired Anisakis by being fed as juveniles on untreated marine fish offal containing live larvae.  相似文献

Separate laboratory experiments were conducted to examine if incorporation of Zn into the otoliths of juvenile pink snapper (Pagrus auratus Forster) was related to levels in the food and/or water. In the first experiment, fish were fed a regular diet (600 mg Zn kg− 1 dw, control group) or a Zn-enriched diet (6000 mg Zn kg− 1 dw or 9000 mg Zn kg− 1 dw) for 35 days. In the second experiment, fish were exposed to waterborne Zn concentrations of < 0.005 µg L− 1 (control), 50 µg L− 1, 100 µg L− 1 and 200 µg L− 1 for 35 days. The sagittal otoliths were analysed using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Juvenile fish exposed to higher concentrations of waterborne Zn did not display increased Zn levels in their otoliths. However, Zn levels in the otoliths of fish consuming the Zn-enriched diet were significantly higher relative to control fish. This study clearly demonstrated that dietary Zn was the major source of Zn incorporated into the otoliths by this marine fish.  相似文献

Natural substances are now generally preferred over chemical and synthetic compounds for the growth and immune enhancement of aquatic organisms. The aim of this study was to evaluate the effect of Saccharomyces cerevisiae extract and hydrolyzed powder on immunity, hematological parameters and body composition in rainbow trout, Oncorhynchus mykiss. Six hundred rainbow trout (50 ± 5 g mean weight) were acclimated to laboratory conditions and then randomly divided into four groups of triplicate tanks. The first group was fed with a commercial diet (control) without supplementation. The second and third groups were given a diet supplemented with 1% of yeast extract and hydrolyzed powder, respectively. The fourth group was also fed with a basal diet supplemented with 0.5% of both substances. Fish were cultured in 300‐L polyethylene tanks for 60 days; immune and hematological parameters, fillet composition and disease resistance were analyzed at days 0, 30 and 60. Results showed that a combination of Saccharomyces cerevisiae extract and hydrolyzed powder could improve the immunity and alter hematological parameters of the rainbow trout compared to the control. Mortality rates of fish fed yeast extract and hydrolyzed powder were also lower than in fish fed the control diet after challenging with Yersinia ruckeri. There were no significant changes in rainbow trout fillet composition compared to the control. It can be concluded that fish diet supplementation with a mixture of yeast extract hydrolyzed powder is preferable compared to each one used alone.  相似文献

Transport of calcium (Ca) and cadmium (Cd) was examined along the gastro-intestinal tract (GIT) of freshwater and seawater Oncorhynchus mykiss irideus (FWT and SWTies respectively) using in vitro and in vivo experiments. Based on known physiological differences between FWT and SWT which aid in regulating ion levels and osmolarity, we hypothesized that SWT would have lower rates of Ca uptake. Also, we predicted that Cd rates would also be lower because Cd is known to share a common transport mechanism with Ca. Kinetics of Ca and Cd transport were determined using mucosal salines of varying concentrations [1, 10, 30, 60, and 100 (mmol L− 1 for Ca, μmol L− 1 for Cd)]. Linear and saturating relationships were found for Ca for FWT and SWT, but overall SWT had lower rates. Linear and/or saturating relationships were also found for Cd uptake, but rates varied little between fish types. Elevated Ca had no inhibitory effect on Cd transport, and Ca channel blockers nifedipine and verapamil had little effect on Ca or Cd uptake. However, lanthanum reduced Ca transport into some compartments. A 21 day in vivo feeding experiment was also performed where FWT and SWT were exposed to control diets or Cd-spiked diets (552 μg Cd g− 1 food). Whole body Cd uptake between fish types was similar, but the majority of Cd in SWT remained in the posterior intestine tissue, while FWT transported more Cd through their gut wall. Overall it appears that large differences in Ca and Cd uptake between FWT and SWT exist, with SWT generally having lower rates.  相似文献

Dietary Na does not reduce dietary Cu uptake by juvenile rainbow trout   总被引:2,自引:0,他引:2  
Rainbow trout Oncorhynchus mykiss fry in moderately hard water were exposed to control or high levels of dietary Cu (c. 6 and 580 ug Cu g food?1) at one of three levels of Na (1·5, 3·0 or 4·5%) in the diet, i.e. six experimental groups. Fish were fed a 4% body mass ration daily for 28 days and 10 individuals from each group were sampled every 7 days. Concentrations of Cu and Na were measured in the gills, liver, gut and remaining carcass of sampled fish. Growth was not affected and no consistent differences were found in mass, total lengths (LT) or indices of body condition among any of the groups on any sampling day. Copper concentration was significantly higher in tissues of Cu‐exposed groups, although within treatment types (control Cu v. high Cu diet), it did not differ consistently among groups that received different levels of dietary Na. Tissue Na concentration did not differ among any of the groups and did not show any marked changes over time. In Cu‐exposed groups, the proportion of total body Cu burden contained in the liver approximately doubled over time, from c. 30% on day 7 to c. 60% on day 28. In unexposed fish, the liver maintained c. 25% of the total Cu burden throughout the experiment. In contrast, the proportion of the total body Cu burden contained in the gut decreased somewhat over time in Cu‐exposed fish, from c. 40% on day 7 to c. 30% on day 28, and remained fairly stable at c. 25–30% in control groups, i.e. approximately equal to liver values. In all groups, the carcass contained by far the largest portion of the total Na content (>80%). Measurements made 36 h post‐feeding indicated that all six groups had much higher Na efflux relative to influx, suggesting that the fish were eliminating excess Na taken up from the diet, and differences in Na influx rates were small. Na efflux rate was significantly higher in the high Cu and high Na group than in the high Cu and low Na group. The results indicate that at the concentrations used in this experiment, dietary Na has little effect on dietary Cu uptake by juvenile rainbow trout, and dietary Cu has little effect on Na homeostasis.  相似文献
