In a succession of seed harvests of carrot, the highest percentage and most rapid germination was obtained from seed harvested 51 days after anthesis (DAA) when dried by conditioning at 25°C and 60% RH for one week and from seeeds harvested 65 or 79 DAA with or without conditioning treatment. Seed from these harvests had reached maximum weight when dried, had embryos of maximum length and were considered mature. The germinaton of seed from these treatments was unaffected by a mixture of the gibberellins A4 and A7 (GA4/7) applied in the incubation medium. Seed harvested 37 DAA also gave maximum percentage germination when it was both conditioned and incubated in GA4/7 solution. Seed harvested earlier than this germinated poorly. Germination times of both mature and immature seed were reduced after storage for 18 months but there was no response to GA4/7.Abbreviations ABA abscisic acid - DAA days after anthesis - GA gibberellin  相似文献

Summary We identified two S-allele-associated proteins (S-proteins) in a self-compatible cultivar of Petunia hybrida based on their segregation in F1 hybrids between P. hybrida and its self-incompatible relative, Petunia inflata (with S2S2 genotype), and in selfed progeny of P. hybrida. These two S-proteins, designated Sx-protein (24 kDa) and So- protein (31 kDa), are pistil specific, and their expression follows a temporal and spatial pattern similar to that of S-proteins characterized in self-incompatible solanaceous species. Their amino-terminal sequences also share a high degree of similarity with those of solanaceous S-proteins. Selfing of P. hybrida yielded plants with SoSo, SxSo, and SxSx genotypes in an approximately 1:2:1 ratio, indicating that the Sx- and So-alleles, though expressed in the pistil, failed to elicit a self-incompatibility response. The S2-allele of P. inflata is expressed in all the F1 hybrids, rendering them capable of rejecting pollen bearing the S2-allele. The So-allele is not functional in the F1 hybrids, because all the F1 progeny with S2So genotype are self-compatible. However, in F1 hybrids with S2Sx genotype, approximately half are self-incompatible and half are self-compatible, indicating that the function of the Sx-allele depends on the genetic background. These results strongly suggest that the presence of functional S-alleles alone is not sufficient for expression of a self-incompatibility phenotype, and reaffirm the multigenic nature of gametophytic self-incompatibility suggested by earlier genetic studies.  相似文献

Current evidence in citrus indicates that gibberellins (GAs) are main determinants of early fruit set while subsequent growth of developing fruits is mostly dependent upon carbohydrate availability. In this work, branch girdling performed at anthesis in Satsuma mandarin (Citrus unshiu (Mak.) Marc.) cv. Okitsu transitorily reduced early abscission rates (12–32 days after anthesis, DAA) delaying initially the process of natural fruitlet drop. The effects of girdling on growth, gibberellin (GA) and carbohydrate concentrations in developing ovaries and fruitlets were assessed during this initial growth stage (0–69 DAA). In girdled branches, abscission rate reduction was preceded by elevated concentrations of carbohydrate and GA in developing ovaries and fruitlets. Girdling at anthesis stimulated higher hexose (21 DAA) and starch (6–20 DAA) concentrations and also higher GA1 (6 DAA), GA19 (13–20 DAA) and GA20 (6–20 DAA). The results established a relationship between the reduction of early abscission rates and higher concentrations of carbohydrates and GAs induced by girdling in developing fruitlets. These findings revealed that girdling certainly increased GA concentration and strongly suggested that its effect on early fruitlet abscission delay is likely mediated by both GA and carbohydrates.  相似文献

The S-allele-associated proteins (S-proteins) in the styles of the Japanese pear (Pyrus serotina Rehd. var. culta Rehd.) were purified by cation exchange chromatography. Their inhibitory action on the growth of incompatible pollen tubes (pollen tubes bearing the same S- allele as in the style from which the S-proteins were prepared) was characterized in vitro. Germination and tube growth of self-pollen (pollen from the same cultivar from which the S-proteins were prepared) decreased dose-dependently when the S-protein was added to the medium. Tube length was reduced to 10% that of compatible pollen tubes (pollen tubes bearing the S-allele different from that in the style from which the S-proteins were prepared) at 1.5 μg μl1. S-proteins from Shinsui (S 4 S 5 ) also inhibited growth of cross-incompatible Kosui (S 4 S 5 ) pollen tubes, but not of compatible Chojuro (S 2 S 3 ) pollen tubes. After inactivation of RNase of the S- protein, the inhibitory action of the S-protein disappeared. These results indicate that the S-protein acts directly to inhibit growth of incompatible pollen tubes in Japanese pear styles, and that the RNase activity of the protein is essential for the biological function. However, small amounts of proteins that co-migrated with the S-protein may also play some roles in the inhibition. A defective S-allele, S 0, and a functional S-allele, S x, have previously been found to be retained in an F1 hybrid of a self-compatible commercial cultivar of Petunia hybrida. Pistil proteins associated with these two alleles have also been identified. Their amino-terminal sequences have been found to share a high degree of similarity with those of S-proteins characterized from self-incompatible solanaceous species. Here we report the isolation and sequencing of cDNAs encoding S 0- and S x-proteins. Their deduced amino acid sequences contain all the consensus primary structural features of S-proteins from self-incompatible solanaceous species. Both proteins also have ribonuclease activity. The implications of these findings are discussed in relation to the presumed function of the S-protein in the self-incompatibility interaction.  相似文献

Summary Gamma ray treatments at various doses were applied to flowers after cross-compatible pollinations (S1S4 × S12S13) and self-incompatible pollinations (S1S4 × S1S4). After 200 kR treatment (highest dose) a high percentage of pollen became non-viable, and tube growth of all the germinated pollen was inhibited at the first third, or just before half the length of the style. Observations on the ultrastructural aspects revealed that the pollen tubes were destroyed by a precise degradation process which led to the disappearance of the inner wall, lysis of the tube, accumulation of several bipartite particles and alteration of endoplasmic reticulum into a whorl of concentric circles. These results indicated that the high dose of gamma rays probably interferes with protein synthesis. The ultrastructural aspects of compatible pollen tubes after gamma irradiation were similar to those of self-incompatible tubes and therefore it is suggested that cessation of protein synthesis might also be a result of incompatibility.Electron microscopy observations after treatment with 200 kR gamma rays on flowers after self-incompatible pollination (S1S4 × S1S4) showed that gamma irradiation affects the self-incompatibility reaction; but the results obtained so far are difficult to explain.  相似文献

This study was conducted to investigate the influence of soil water potential, depth of N placement, timing, and cultivar on uptake of a small dose of labeled N applied after anthesis by wheat (Triticum aestivum L.) Understanding postanthesis N accumulation should allow better control of grain protein concentration through proper manipulation of inputs. Two hard, red spring-wheat cultivars were planted in early and late fall each yr of a 2-yr field experiment. Less than 1 kg N ha–1 as K 15NO3 was injected into the soil at two depths: shallow (0.05 to 0.08 m) and deep (0.15 to 0.18 m). In both years an irrigation was applied at anthesis, and injections of labeled N were timed 4, 12, and 20 days after anthesis (DAA). Soil water potential was estimated at the time of injection. Mean recovery of 15N in grain and straw was 57% of the 15N applied. Recovery did not differ between the high-protein (Yecora Rojo) and the low-protein (Anza or Yolo) cultivars. Mean recovery from deep placement was 60% versus only 54% from shallow placement (p < 0.01). Delaying the time of injection decreased mean recovery significantly from 58% at 4 DAA to 54% at 20 DAA. This decrease was most pronounced in the shallow placement, where soil drying was most severe. Regressions of recovery on soil water potential of individual cultivar x yr x planting x depth treatments were significant only under the driest conditions. Stepwise regression of 15N recovery on soil water potential and yield parameters using data from all treatments of both years resulted in an equation including soil water potential and N yield, with a multiple correlation coefficient of 0.64. The translocation of 15N to grain was higher (0.89) than the nitrogen harvest index (0.69), and showed a highly significant increase with increase in DAA. This experiment indicates that the N uptake capacity of wheat remains reasonably constant between 4 and 20 DAA unless soil drying is severe.  相似文献

Stylar proteins of 13 almond (Prunus dulcis) cultivars withknown S-genotypes were surveyed by IEF and 2D-PAGE combinedwith immunoblot and N-terminal amino acid sequence analysesto identify S-RNases associated with gametophytic self-incompatibility(SI) in this plant species. RNase activities corresponding toSa and Sb, two of the four S-alleles tested, were identifiedby IEF and RNase activity staining. The Sa-RNase band reactedwith the anti-S4serum prepared from Japanese pear (Pyrus serotina);no reaction with the antiserum was observed with the sbRNaseband. When the sa-RNase band was excised from an IEF gel stainedfor RNase activity, subjected to SDS-PAGE, and detected by immunoblotting,it appeared that this band consisted of a single protein thatreacted with the anti-s4serum with Mr of about 28 kDa. With2D-PAGE and silver staining of the stylar extracts, all fourS-proteins could be successfully distinguished from each otherin the highly basic zone of the gel. Although Sb-, Sc-, andSdproteins had roughly the same Mr of about 30 kDa, the Sc-proteinseemed to be slightly smaller than the Sb-protein and slightlylarger than the Saprotein. In 2D-PAGE profiles as well, theSa-protein had Mr of about 28 kDa, apparently smaller than theother three proteins. A bud sport, in which one of the two S-allelesof the original cultivar is impaired, was visualized as a lossof Scprotein, which is consistent with the previous pollinationstudy. All four S-proteins reacted with the anti-S4serum, probablybecause of the differing conformations of these S-proteins inthe IEF and 2D-PAGE gels. The Sa-protein in 2D-PAGE appearedto be identical to Sa-RNase in IEF; both bad the same Mr andwere reactive with the anti-S4-serum. N-terminal amino acidsequence analysis of the four 5-proteins revealed that theywere highly homologous to each other and similar to the 5-RNasesof Malus, Pyrus, Scrophulariaceae, and Solanaceae. Taken together,RNases in the style are strongly suggested to be associatedwith the gametophytic SI of al- mond. This is the first reportidentofiying and characterizing S-RNase in almond. Summary A cross between a 0% pseudo-self-compatible (PSC) plant (S3.3) and a 100% PSC plant (S1.1) yielded an F1 population which, when selfed, produced a high mean seed set which was not significantly different than that produced when the F1 was backcross pollinated by the 100% PSC parent. Backcross pollinating the F1 with the 0% PSC parent yielded no seed. No S3.3 plants were recovered in the F2 populations, indicating that pollen tubes containing the S3 allele were inhibited during pollen tube growth of the selfed F1 plants. Apparently stylar-conditioned PSC does not remove all discriminatory power from these petunia styles. Crossing the F1 (S1.3) with an self-incompatible (SI) plant (S2.2) produced plants which were used for computation of a standard linkage test. An approximate map distance of 28 units was found between the S specificity locus and the major gene(s) which influenced its expression. Antiserum to a protein fraction of an extract of mature styles of P. avium cv. Lambert (S 3 S 4) was raised in rabbits. Two major antigenic components of the style extracts were detected by immunoelectrophoresis and immunodiffusion. The presence of one antigen (S-antigen) correlated with a particular S-genotype (S 3 S 4). This antigen is restricted to mature styles of P. avium. The second antigen (P-antigen) was detected in styles of all Prunus species examined, but not in styles of other species of the Rosaceae. The S-antigen is positively charged and the P-antigen negatively charged at pH 8.8.  相似文献

Most cultivars of Japanese pear (Pyrus pyrifolia Nakai) exhibit gametophytic self-incompatibility controlled by a single S-locus with multiple S-haplotypes. A self-compatible (SC) cultivar, ??Osanijisseiki?? (S 2 S 4 sm ), arising by a bud mutation of ??Nijisseiki?? (S 2 S 4 ), has a stylar-part mutant S 4 sm -haplotype, which lacks the pistil S 4 gene, which is the S 4 -RNase gene. To efficiently breed SC cultivars, we selected ??Nashi Chuukanbohon Nou 1 Gou?? (??NCN1??) harboring homozygous S 4 sm from a self-progeny of Osanijisseiki and crossed it with ??Okusankichi?? (S 5 S 7 ), ??Hakkou?? (S 4 S 5 ), or ??Ri-14?? (S 1 S 2 ). Fruit set (%) was compared after self-pollination of the trees in the three progenies. All trees derived from the three progenies were predicted to be SC, except for the S 4 S 4 sm trees in the progeny of NCN1 × Hakkou. However, S 1 S 4 sm trees in the progeny of NCN1 × Ri-14 proved to be self-incompatible (SI). The pollen from Osanijisseiki was incompatible with ??Doitsu?? (S 1 S 2 ), but that from Nijisseiki was compatible, suggesting a possibility that the S 4 sm pollen was rejected by S 1 -harboring pistils. This possibility was clarified by crossing the pollen from NCN1 (S 4 sm S 4 sm ) to Doitsu, ??Imamuraaki?? (S 1 S 6 ), or ??Hougetsu?? (S 1 S 7 ), all of which proved incompatible. On the other hand, S 4 sm pollen was accepted by pistils harboring the S 2 , S 3 , S 5 , S 6 , S 7 , S 9 , and S k haplotypes. The dual recognition of S 1 and S 4 pistils by S 4 sm pollen can be attributed to a mutation of the pollen S 4 gene(s).  相似文献

Illus. 1962.—A diallel cross involving 11 self-incompatible and 3 self-compatible species of Solanum was made to study the genetic basis of interspecific incompatibility. Interspecific incompatibility was not limited to crosses in which a self-compatible species was used as the male parent onto a self-incompatible species (unilateral incompatibility). A number of crosses between self-incompatible species were incompatible. In one cross, Q vernei X verrucosum, a self-compatible species was successful as a pollen parent with a self-incompatible species. Unlike other hybrids between self-compatible and self-incompatible species, which are self-incompatible, these F1 hybrids were self-fertile, and cross-fertile among themselves and with both parents. The self-fertile S. polyadenium was cross-incompatible as a female as well as a male parent with all other species. It is suggested that the unilateral incompatibility is a property of the allele SC which originated as a consequence of one kind of breakdown of the SI gene; the SC allele produces “bare” pollen growth substances which are inactivated in an incompatible style. It is proposed that the failure of the principle of unilateral interspecific incompatibility in solanaceous species may be due to the action of alleles at the second incompatibility locus revealed in certain Mexican species. It is assumed that the South American species are selected intraspecifically only for the action of S alleles but that in certain interspecific crosses and rarely in intraspecific crosses the alleles at the second locus may be expressed, thus interfering with the usual action of S alleles. The F1 hybrids Q verrucosum (self-fertile) X simplicifolium (self-sterile) were self-incompatible at the tetraploid as well as the diploid level, and their cross-compatibility behavior was consistent with the expected activity of the SC and SI alleles of the 2 parents respectively.  相似文献

We investigated the effect of gibberellic acid (GA3) in the fertilization of Clementine mandarin cv. ‘Clemenules’ (Citrus clementina Hort. ex Tan.), a parthenocarpic variety that produces seedless fruit due to its self-incompatible nature, but yields seedy fruits when grown under cross-pollination conditions.Experiments were conducted with on-tree ‘Clemenules’ flowers and ‘Fortune’ mandarin pollen (C. clementina Hort. ex Tan. × C. reticulata Blanco), which is sexually compatible with the former. Preanthesis treatment at −2 days after anthesis (−2 DAA) enhanced ovule abortion in both unpollinated and cross-pollinated (at +2 DAA) flowers. In the latter, the number of pollen tubes reaching the ovules was significantly reduced although pollen grains were not treated; thus, fertilization was partially avoided and seed set was reduced. When GA3 was applied at anthesis (0 DAA) at the time of pollination, ovule abortion was again enhanced, and pollen tube growth was completely arrested; thus, fertilization was prevented and seed set was impeded. When GA3 was applied 24 h after pollination (+1 DAA in flowers pollinated at anthesis), pollen tube growth was impaired but not arrested and ovule abortion was enhanced; therefore, fertilization was not prevented but impaired.We conclude that, when applied the days around anthesis, GA3 (10 mg l−1) impairs fertilization by either enhancing ovule abortion or reducing pollen tube growth, in ‘Clemenules’ flowers under cross-pollination conditions. The intensity of the response depends on the physiological flower state at the moment of treatment.  相似文献

Summary S-alleles of self-incompatibility were isolated from a wild population of Brassica campestris growing at Balcesme, Turkey. Out of 88 plants observed, 73 were self-incompatible and 4 were self-compatible. In certain families, selfed progenies from a self-incompatible plant segregated into fewer than three incompatibility classes, which is consistent with a one-locus sporophytic genetic control of self-incompatibility. Out of 25 combinations of S-alleles tested, dominance interactions were observed in 6 of them on the pollen side and on 5 of them on the stigma side. The 35 S-homozygotes thus isolated consisted of 18 independent S-alleles. The number of S-alleles in this population was estimated to be more than 30. The S-locus glycoproteins (SLGs) corresponding to the respective S-alleles were identified by iso-electric focusing (IEF)-gel immunoblotting with a polyclonal antiserum against SLG8. SLGs in a stigma were generally composed of several bands, one major and a few minor ones, whose molecular weight was similar to each other, and the major and minor bands were heritable in correlation with each other. SDS-PAGE analysis of SLGs differentiated a few juxtaposed bands between 50 and 60 kDa, and the variations in these bands were considered to be due to differences in the number of polysaccharide residues. General features of the variation of S-genes and their SLGs between the populations in Balcesme, Turkey and Oguni, Japan, were comparatively similar to one another, despite the different surroundings and history of these populations.  相似文献

Self-compatibility in almond (Prunus dulcis) is attributed to the presence of the S f haplotype, allelic to and dominant over the series of S-alleles controlling self-incompatibility. Some forms of the S f haplotype, however, are phenotypically self-incompatible even though their nucleotide sequences are identical. DNA from leaves and styles from genetically diverse almond samples was cloned and sequenced and then analyzed for changes affecting S f -RNase variants. Epigenetic changes in several cytosine residues were detected in a fragment of 4,700 bp of the 5′ upstream region of all self-compatible samples of the S f -RNases, differentiating them from all self-incompatible samples of S f -RNases analyzed. This is the first report of DNA methylation in a Rosaceae species and appears to be strongly associated with inactivation of the S f allele. Results facilitate an understanding of the evolution of self-compatibility/self-incompatibility in almond and other Prunus species, and suggest novel approaches for future crop improvement.  相似文献

 The self-incompatibility in yellow passion fruit was previously described as homomorphic sporophytic with monofactorial inheritance. Five progenies were obtained by bud-selfing. The plants of these progenies were selfed, reciprocally crossed within each progeny and crossed with known incompatible phenotypes to identify their phenotypic group. Fruit set was evaluated at the 7th day after pollination. Two progenies consisted of two self-incompatible groups, the other three formed three suck groups. The groups were identified as S1, S2, S3, S4, S5 and S6. The results provide evidence that the self-incompatibility of passion fruit is controlled by two loci, the S-gene and another, whose expression needs to be investigated. Sk2 protein is the most abundant member of the pistil-specific proteins of Solanum tuberosum. Sk2 protein has been localized by use of a polyclonal antibody (anti-Sk2) in the pistils of four clones of Solanum tuberosum. In the stigmas Sk2 protein accumulates to a high level in the cytoplasm of the internal secretory cells underlying the papillae one day prior to anthesis. In styles, the intercellular matrix of the transmitting tissue cells is intensely labelled by anti-Sk2. Sk2 protein is present in all four clones and shows the same labelling pattern. The possible role of the Sk2 protein in pollen tube growth is discussed.  相似文献

Summary AnS 1.1 self-incompatible (SI) petunia plant which showed atypical seed set was found in an I7 population. This plant showed a strong SI reaction when selfed but produced varying amounts of seed when used as the seed parent in crosses with unrelated individuals homozygous for the sameS allele. Reciprocal crosses yielded no seed indicating that the reaction was a stylar response. Self seed obtained by high temperature treatments produced 18 plants, all of which exhibited the parental characteristics, the ability to reject self pollen but accept, to varying degrees, pollen bearing the sameS allele from unrelated plants. Several petunias homozygous forS 1, and exhibiting various levels of PSC as determined by self seed set, progeny tests and temperature treatments, were used as pollen parents. The mean seed set of these crosses produced a ranking of the pollen parents which reflected the PSC levels obtained by other methods. The behavior of the F1 and F2 populations suggests that the pollen discriminating ability may be a simply inherited, dominant character in these plants.
