The aflagellate spermatozoa of the gamasid mite Megisthanus floridanus are characterized by a large vacuole which contains a cytoplasmic column protruding into the vacuole from the region defined as the posterior part of the cell. The membrane of the column and the inner membrane of the posterior part of the cytoplasmic mantle (outer sheath) surrounding the vacuole bear numerous so-called cellular processes. However, most of the outer sheath is reduced and represented solely by a very thin membrane-like envelope. The posterior part of the cell bears extensive folds. The cell, or, more precisely, the column, shows a deep posterior invagination. This invagination contains extracellular material composed of thin filaments or strands. Peripheral folds emerging from the posterior rim of the cell form a thin-walled tube that contains the same material as the invagination. The elongated nucleus is attached to a peculiar acrosomal complex consisting of a flat acrosomal cisterna that parallels most of the cell membrane, an attachment cone, and a short acrosomal filament which is embedded in a narrow canal within the nucleus. The spermatozoa of M. floridanus represent a peculiar version of the vacuolated type of sperm known to be plesiomorphic within the anactinotrichid Acari. Some details of spermiogenesis are described and consequences for phylogenetic and systematic considerations are discussed.  相似文献

The accessory muscle of the walking leg of the horseshoe crab, Tachypleus gigas, was examined electron microscopically. The muscle fibers vary in size but are small in diameter, when compared with other arthropod skeletal muscles. They are striated with A, I, Z and poorly defined H bands. The sarcomere length ranges from 3-10 μm with most sarcomeres in the range of about 6 μm. The myofilaments are arranged in lamellae in larger fibers and less well organized in the smaller ones. Each thick filament is surrounded by 9-12 thin filaments which overlap. The SR is sparse but well organized to form a fenestrated collar around the fibrils. Individual SR tubules are also seen among the myofibrils. Long transverse tubules extend inward from the sarcolemma to form dyads or triads with the SR at the A-I junction. Both dyads and triads coexist in a single muscle fiber, a feature believed to have evolutionary significance. The neuromuscular relationship is unique. In the region of synaptic contact, the sarcolemma is usually elevated to form a large club-shaped structure containing no myofilaments and few other organelles. The axons or axon terminals and glial elements penetrate deep into the club-shaped sarcoplasm and form synapses with the fiber. As many as 13 terminals have been observed within a single section. Synaptic vesicles of two types are found in the axon terminals.  相似文献

We describe sperm ultrastructure and acrosome differentiation during spermiogenesis in Crassostrea gigas (Mollusca Bivalvia). The sperm cell is a uniflagellated cell of the primitive type. The head region contains a rounded or conical nucleus surmounted by small acrosome. This organelle consists of a membrane-bound acrosomal granule, the contents of which have a homogeneous density, except in the anterior region, which is positive for PTA. The acrosome also surrounds the perforatorium, which includes oriented fibrillar elements: this is the axial body. The middle piece contains four mitochondria encircling two perpendicular centrioles. The distal centriole is provided with a system of mechanical fixation to the plasma membrane, consisting of nine fibers in radial arrangement. The spermatozoon of Hemirhamphodon pogonognalhus shows modifications that are frequent though not obligate in internally fertilizing sperm, notably elongation of the nucleus and extension of the mitochondria of the midpiece as an elongate sheath around the proximal region of the axoneme. These similarities to poecilid and jenynsid sperm are considered homoplasic. As in the mature sperm of all but one investigated teleost, an acrosome is absent. The elongate, blade-shaped, electron-dense nucleus has a mean length of 3.2 μm; its basal implantation fossa, less than one-tenth of the length of the nucleus, houses the anterior half of the distal and only centriole (of triplet construction with satellite rays), a centriolar plug, and a mass connecting the centriole to the wall of the fossa. A unilateral putative centriole adjunct is present. The anterior region of the axoneme is surrounded by a mitochondrial sleeve, and internal to this, separated by a cisterna, by a submitochondrial sleeve. The mitochondrial sleeve unites posteriorly with the submitochondrial sleeve. Between the submitochondrial sleeve and the axoneme is a space, the cytoplasmic canal, that is open to the exterior posteriorly. The discrete, cristate mitochondria, in their sleeve, are unique in investigated atherinomorph sperm in being bilateral, grouped on only two opposing sides of the axoneme, with an arc-shaped ‘intermitochondrial link’ between. The 9 + 2 flagellum is unique for the Animalia in having 23 radial subplasmalemmal rods, repeated longitudinally (periodicity 0.025 pm) in a quasicrystalline array. Internal fertilization is deduced to have arisen in the Exocoetoidei independently of that in the Cyprinidcntiformes.  相似文献

The general organization of the male genital system, the spermatogenesis and the sperm structure of the proturan Acerella muscorum have been described. At the apex of testis apical huge cells are present; their cytoplasm contains a conventional centriole, a large amount of dense material and several less electron-dense masses surrounded by mitochondria. Spermatocytes have normal centrioles and are interconnected by cytoplasmic bridges. Such bridges seem to be absent between spermatid cells and justify the lack of synchronization of cell maturation. Spermatids are almost globular cells with a spheroidal nucleus and a large mass of dense material corresponding to the centriole adjunct. Within this mass a centriole is preserved. Mitochondria of normal structure are located between the nucleus and the plasma membrane. The spermatids are surrounded by a thick membrane. No flagellar structure is formed. Sperm have a compact spheroidal nucleus, a large cap of centriole adjunct material within which a centriole is still visible. A layer of mitochondria is located over the nucleus. The cytoplasm is reduced in comparison to spermatids; many dense bodies are interspersed with sperm in the testicular lumen. The sperm are small, immotile cells of about 2.5-3 μm in diameter.  相似文献

The sperm of Siphonaria algesirae (Gastropoda, Pulmonata), a species with internal fertilization, was studied by light and electron microscopy. The spermatozoon is a very long, uniflagellate cell composed of a conical head with an apical acrosome, a midpiece with a helically coiled external sheath containing a complex mitochondrial derivative with a wavelength of ~ 5.5 μm, and an endpiece. There are no axonemal microtubules. Instead, nine homogeneous coarse fibers with transverse striations in the apical zone project toward the anterior section of the midpiece. In the posterior zone of the midpiece the coarse fibers are differentiated in a common microtubular axoneme. The complex mitochondrial derivative of the midpiece shows an organized group of 100 Å diameter spherical particles. Externally the midpiece is surrounded along its length by a cylinder formed by two membranes. A complex structure separates the transitional zone between the midpiece and the endpiece.  相似文献

The ultrastructure of spermatozoa in Hydrolagus colliei is described. Basic similarities of structure to the sperm of the related elasmobranch fish are noted. The most significant features of sperm structure in Hydrolagus that differ from those of elasmobranch fish occur in the tail. The axoneme is eccentrically located and forms a double helix with a single longitudinal column. A second longitudinal column is reduced to a short remnant at the base of the tail. Microtubules within the axoneme are also helically disposed, a feature that is consistent with the rotating motion of the sperm. Abundant glycogen reserves are stored along the length of the tail.  相似文献

Spermatozoa of the hagfishes Eptatretus burgeri and Eptatretus stouti, caught in the sea near Japan and North America, respectively, were found to undergo the acrosome reaction, which resulted in the formation of an acrosomal process with a filamentous core. The acrosomal region of spermatozoa of E. stouti exhibited immunofluorescent labeling using an actin antibody. The midpiece also labeled with the antibody. The acrosomal region showed a similar labeling pattern when sperm were probed with tetramethylrhodamine isothyocyanate (TRITC)-phalloidin; the midpiece did not label. Following induction of the acrosome reaction with the calcium (Ca2+) ionophore ionomycin, TRITC-phalloidin labeling was more intense in the acrosomal region, suggesting that the polymerization of actin occurs during formation of the acrosomal process, as seen in many invertebrates. The potential for sperm to undergo acrosomal exocytosis was already acquired by late spermatids. During acrosomal exocytosis, the outer acrosomal membrane and the overlying plasma membrane disappeared and were replaced by an array of vesicles; these resembled an early stage of the acrosome reaction in spermatozoa of higher vertebrates in which no formation of an acrosomal process occurs. It is phylogenetically interesting that such phenomena occur in spermatozoa of hagfish, a primitive vertebrate positioning between invertebrates and high vertebrates.  相似文献

Horseshoe crabs (order Xiphosura) are often referred to as an ancient order of marine chelicerates and have been considered as keystone taxa for the understanding of chelicerate evolution. However, the mitochondrial genome of this order is only available from a single species, Limulus polyphemus. In the present study, we analyzed the complete mitochondrial genomes from two Asian horseshoe crabs, Carcinoscorpius rotundicauda and Tachypleus tridentatus to offer novel data for the evolutionary relationship within Xiphosura and their position in the chelicerate phylogeny. The mitochondrial genomes of C. rotundicauda (15,033 bp) and T. tridentatus (15,006 bp) encode 13 protein-coding genes, two ribosomal RNA (rRNA) genes, and 22 transfer RNA (tRNA) genes. Overall sequences and genome structure of two Asian species were highly similar to that of Limulus polyphemus, though clear differences among three were found in the stem-loop structure of the putative control region. In the phylogenetic analysis with complete mitochondrial genomes of 43 chelicerate species, C. rotundicauda and T. tridentatus were recovered as a monophyly, while L. polyphemus solely formed an independent clade. Xiphosuran species were placed at the basal root of the tree, and major other chelicerate taxa were clustered in a single monophyly, clearly confirming that horseshoe crabs composed an ancestral taxon among chelicerates. By contrast, the phylogenetic tree without the information of Asian horseshoe crabs did not support monophyletic clustering of other chelicerates. In conclusion, our analyses may provide more robust and reliable perspective on the study of evolutionary history for chelicerates than earlier analyses with a single Atlantic species.  相似文献

The Indian horseshoe crab, Tachypleus gigas (Müller), is a benthic feeder which subsists mainly on molluscs, decayed organic matter and polychaetes, in order of prevalence. A strong and positive preference was recorded for molluscs over other food organisms. The intensity of feeding was maximum during the north-east monsoon (November to January) in females and during the intermonsoon season (February to May) in males. Morphology and fine structure of Barbus barbus L 1758 spermatozoa were studied using scanning (SEM) and transmission (TEM) electron microscopy. The results confirm that spermatozoa exhibit morphological features typical to all teleost fishes. They are differentiated into a head, a midpiece and a flagellum with the typical '9 + 2' pairs of microtubules. Both dynein arms are present in the flagellum. The spermatozoa have spherical nuclei, 4–6 mitochondria located in the postnuclear cytoplasmic region and centriolar complex (proximal and distal centrioles). Total length, head width, length of midpiece and length of flagellum were measured to be 56.35 ± 7.42, 1.80 ± 0.06, 0.48 ± 0.14 and 54.30 ± 6.97 μm, respectively. Highly significant linear correlation was observed between posterior and anterior width of midpiece (P < 0.01). Principal component analysis (PCA) was used to explore which parameters can explain the individual variation of sperm morphology. About 44% of the total accumulated variance was absorbed by the analysis of the two first components, distinguishing different groups of parameters related to head and midpiece. The lengths of flagellum and head are more isolated; indicating that the individual variation of sperm morphology depends on these two parameters. Comparing the results of this study with information on cyprinids spermatozoa reveals that the number of mitochondria and the length of the flagellum are good characters to characterize spermatozoa of the Cyprinidae in a phylogenetic arrangement.  相似文献

Membrane cofactor protein (MCP; CD46) is a complement regulator widely expressed as four isoforms that arise via alternative splicing. On human spermatozoa, MCP is expressed on the inner acrosomal membrane and alterations of spermatozoa MCP may be associated with infertility. In rodents, expression of MCP is largely restricted to the testes. MCP on human spermatozoa has a unique M(r) pattern that we have investigated. We also characterized MCP expression in mice transgenic (tg) for human MCP. Human MCP expression in the tg mice mimics the human pattern in that it is located on the inner acrosomal membrane and has a faster M(r) than MCP expressed elsewhere. Sequencing of RT-PCR products from the testis indicates that there is not a unique male reproductive tissue specific cytoplasmic tail. Instead, human spermatozoa express MCP bearing cytoplasmic tail two, which is also utilized in most other tissues and contains several signaling motifs. Further, using N-glycosidases, we demonstrate that the unique lower molecular weight of MCP on spermatozoa is secondary to a modification in the N-linked sugars. Specifically, as the spermatozoa mature, but before they reach the epididymis, the three N-linked sugars of MCP are trimmed to less complex structures. While the purpose of this deglycosylation is unknown, we propose that it is a common feature of proteins expressed on the plasma and inner acrosomal membranes of spermatozoa and hypothesize that it is a spermatozoa specific event critical for facilitating sperm-egg interactions.  相似文献

Oyster granular amebocytes were characterized by electron microscopy to be of two types. Acidophiles contained relatively small, spherical, amorphous, osmiophilic cytoplasmic granules with a maximum dimension of 0.4–0.56 μ. Basophiles contained large, cytoplasmic granules, approximately 0.7–1.2 μ in diameter. The granules resembled hollow, spherical structures in which an outer membrane and a layer of closely associated granular material enclosed an electron-transparent interior.  相似文献

Summary In this study the fine structure of the cerebral ganglion of Sagitta setosa (Chaetognatha) is investigated. The ganglion is flat and superficially positioned dorsally, below the basal lamina of the cephalic epidermis. It is surrounded by a specifically differentiated sheath. This sheath is made up of cells, which are interpreted as representing glial cells, and can be divided into an outer and an inner zone. The outer zone is composed of flat sheath cells with pale nuclei and few organelles. The inner zone consists of densely packed, extremely thin lamellar cellular processes. These attenuated lamellae, which still contain cytoplasm, resemble the myelin sheath of vertebrate axons. The intercellular space between the lamellae contains electron-dense material. In the sheath specialized intercellular contacts occur. The inner zone of this sheath extends at definite points into the centre of the ganglion and separates a zone of perikarya from the neuropil, as well as the single perikarya from each other. The perikarya are relatively uniform and do not form a cortex, but are concentrated mainly in lateral parts of the cerebral ganglion. Within the neuropil are axonal endings which have synaptic contacts with several postsynaptic elements. These anatomical findings are discussed with respect to their functional significance.  相似文献

The sperm structure of several ladybird species belonging to different subfamilies of Coccinellidae was studied. Three main sperm types were clearly recognized, and were characterized by differences in acrosomal length, the presence of a dense coat around the acrosome, the length of the basal body, the amount of the centriole adjunct material, and the diameter of the mitochondrial derivatives. However, the whole group shares a pattern of the posterior sperm region uncommon for insects, in which the axoneme and other flagellar components are running parallel with the nucleus. As a general conclusion, this study has revealed an inconsistency between the sperm structure and the systematics of the group, indicating that the generic concepts within the group do not reflect a natural classification, a statement also shared by molecular studies.  相似文献

Early spermatids of the carb, Pinnixia sp., are characterized by a large nuclear/cytoplasmic ratio. During early spermiogenesis an organelle, composed of pentalaminar membranes derived from the endoplasmic reticulum and nuclear membranes, is formed. Portions of this organelle become incorporated within a cylinder-shaped invagination of the acrosome. Portions of the nuclear membrane disappear beneath the acrosome resulting in intermingling of nucleoplasm, centrioles and mitochondria. The nuclear membrane elsewhere is found as a pentalaminar membrane underlying the plasma membrane. Centrioles are found in mature spermatozoa at the base of the cylinder-shaped acrosomal invagination, and mitochondria are found intermingled with nuclear remanants surrounding the acrosome. This datum is compared to previously described events in spermiogenesis of other decapod crustacea and arthropods exhibiting similar modes of spermiogenesis. It is concluded that the differences and/or similarities exhibited by centrioles and mitochondria in these forms could be significant in terms of subsequent zygote differentiation.  相似文献

为阐明F-肌动蛋白在优雅蝈螽Gampsocleis gratiosa Brunner von Wattenwyl精子形成过程中的动态变化, 本研究利用微分干涉相衬技术和免疫荧光技术首次对优雅蝈螽精子形成过程中的F-肌动蛋白进行了细胞定位, 利用透射电镜技术从超微水平观察了优雅蝈螽精子顶体复合体的结构。结果显示: 精子形成早期, F-肌动蛋白富集于亚顶体区域, 形态由“球状”转变为“棒锥状”; 精子形成中期, F-肌动蛋白呈“倒Y型”分布于亚顶体区域和细胞核前端两侧; 精子形成后期, 亚顶体区域的F 肌动蛋白解聚消失, F-肌动蛋白呈“箭头状”, 仅分布于顶体复合体扩张的两翼中。F-肌动蛋白动态变化伴随着细胞核和精子头部的形态改变, F-肌动蛋白的动态装配在精子顶体复合体形态构建和细胞核的形变中起着重要的作用。本研究还发现未成熟的精子尾部有一些富含F-肌动蛋白的细胞质微滴, 与精子形成过程中多余细胞质和细胞器的外排有关。F-肌动蛋白的动态变化研究为进一步阐明细胞骨架蛋白在昆虫精子形成过程中的功能和作用机制奠定了基础。  相似文献

Summary The organisation of the spermatozoa ofAnthopharynx sacculipenis is described, based on electron-microscopical observations. The male gametes are fili-form in shape. They are totally enclosed by cortical microtubules and possess two free cilia. Special features are dot-like dense granules arranged in regular rows and terraced elaborations of the nuclear membrane. Such terraced elaborations are not known in any other species of flatworms whereas dot-like dense granules are described for some other taxa of the Rhabdocoela.
