The binding of juvenile hormone (JH) by components from hemolymph of adult female Locusta migratoria was characterized to establish whether hemolymph JH-binding proteins could be distinguished from a protein of fat body (BP-1) that may be a JH receptor. Hemolymph was analyzed by the hydroxyapatite assay, gel separation chromatography, polyacrylamide gel electrophoresis, and density gradient centrifugation. Three fractions that bound JH were separated from whole hemolymph by DEAE cellulose column chromatography, and these differed from all three cytosol-binding components. The major hemolymph component (H-A) showed relatively stable binding of JH, a slight loss of binding capacity after delipidation, and a Kd for JH-I of 16 nM. The Kds for JH-l and JH-lll with unfractionated hemolymph were 26 and 42 nM respectively. The order of effectiveness of competitors for binding of [3H]JH-l was JH-lll > JH-l ? methoprene > hydroprene ? acids of methoprene and hydroprene. The data indicated that unlabeled JH-lll was bound more effectively than its radioactive counterpart. The sedimentation values determined by sucrose density gradient ultracentrifugation were 13-14 S for hemolymph, and the sedimentation value was not altered by the inclusion of 0.4 M KCl throughout the gradient. The data indicated that H-A resembled the specific JH carriers and differed from the putative receptor of fat body cytosol by several criteria.  相似文献

ABSTRACT. Detailed studies of lipoprotein A+ formation during AKH action have been made in Locusta migratoria L. using gel filtration combined with the use of radiolabeled haemolymph protein probes. In addition, we have assessed the quantitative contribution of the CL-proteins to A+ formation by direct measurement of the changes in concentration of free CL-proteins and report some properties of the C-I and C-II proteins: they appear to be glycoproteins of 20,000 and 16,000 MW respectively, but do not bind to concanavalin A. We have confirmed earlier observations (using different techniques) which showed that liproprotein Ayellow is not involved per se in A+ formation during the first 15 min of AKH action. In contrast, the (two) CL-proteins take part in A+ formation without any apparent delay after hormone injection. Our observations show that A+ formation is essentially complete within 30 min of AKH injection, although further CL-protein binding and lipid-loading do occur subsequently. After 30 min there is no further decrease in the Ayellow titre. It is argued that much, if not all, CL-protein is located at the surface of the A+ particle. From the changes in titres which occur in Ayellow and CL-proteins during AKH action we estimate that A+ is formed from 1 mole of Ayellow and approximately 28 moles of CL-proteins. Using these figures we calculate an apparent molecular weight for A+ within the range of 1.65–2.12×106, which is in reasonable agreement with estimates derived from gel exclusion chromatography data. These studies emphasize the dynamic and fully reversible nature of lipoprotein A+ formation and highlight the complex nature of the lipoprotein transformations occurring during hormone-stimulated lipid transport in locusts.  相似文献

Summary At the end of blastokinesis, serosal epitheliae of 4- to 5-day-old embryos of Locusta migratoria contain an immunohistologically detectable cytosolic protein (Mr 240 kDa) which is related to the juvenile hormone carrier-protein in the haemolymph of the same species and which binds tritiated juvenile hormone 3 (JH3) (Kd10–8 M). At this early stage of development the corpora allata of the embryo are not yet fully differentiated and do not synthesize JH3 in organ cultures. The earliest detectable JH3 production by corpora allata in isolated heads is on day 6. On the other hand, serosal epitheliae of 4- to 5-day-old embryos produce JH3 in organ cultures, as has been shown by methylation of (10-3H)-JH3-acid to (10-3H)-JH3, and by incorporation of tritiated CH3 from l-(methyl-3H)-methionine into JH3. Isolated heads and abdomens of the embryos used as donors for the serosal preparations did not show methyl transferase activity responsible for JH3 biosynthesis. The serosal cells represent a hitherto unrecognized source of methyl transferase activity and of JH3 production. ABSTRACT. Tritiated 10,11-epoxyfarnesyl diazoacetate (EFDA), a photoaffinity label, can be covalently attached to the binding site of a JH-III-specific binding protein in the haemolymph of Locusta migratoria migratorioides (R & F). The specificity of the binding of EFDA to the binding protein is verified by displacement with excess unlabelled JH-III, and EFDA can be used to identify the binding protein in native pore-limiting gradient poly(acrylamide) gel electrophoresis (PAGE) and sodium dodecyl sulphate-PAGE. The native binding protein has a molecular weight of 575,000 and is composed of seemingly identical subunits of molecular weight 81,000.
Three other high-molecular weight serum proteins are identified by native PAGE: a lipophorin, composed of two kinds of apolipophorins, a larval storage protein and a cyanoprotein. The molecular weights and subunit structures of these proteins are investigated, but none of these other high-molecular weight proteins bind JH-III to an appreciable extent.  相似文献

To understand the hormonal control of embryonic diapause, juvenile hormone analogs (JHAs), methoprene and hydroprene, and 20-hydroxyecdysone (20E) were applied onto diapause eggs of Locusta migratoria and Oxya yezoensis. These insects enter diapause at the mid-stage of embryogenesis prior to blastokinesis. Topical application of JHAs significantly facilitated diapause termination in both species but JHA-treated embryos underwent abnormal morphogenesis, pigmentation and sclerotization without dorsal closure. The Locusta eggs immersed in the 20E solution for 24h terminated diapause in a dose-dependent manner. We also investigated phosphorylation of extracellular signal-regulated kinase (ERK), a member of mitogen-activated protein kinase (MAPK), during diapause-terminating process of Locusta migratoria and found that ERK was activated either by cold exposure or JHA treatment. The possible involvement of the hormones and ERK in embryonic diapause and the possibility of ecdysteroids synthesis by prothoracic glands of diapause embryo were proposed.  相似文献

ABSTRACT. Incubation of corpora allata (CA) from adult females of Locusta migratoria (L.) in vitro in medium with a potassium concentration of 50 mM results in an elevation of the rate of juvenile hormone (JH) release. This elevation is however delayed, becoming apparent after the glands have been returned to low concentration of potassium but it is also prolonged, lasting up to 270 min. The elevation is initially modest but becomes more marked with time. The response of glands to high concentration of potassium is heterogeneous and appears to some extent to be a function of the initial rate of JH release of the glands. Glands starting with low rates of JH release are stimulated strongly by potassium, those with high rates of JH release much less so. Co-incubation of glands with high concentration of potassium and with calcium channel blockers (both organic and ionic) prevents the elevation of release rates otherwise consequent on high concentrations of potassium.  相似文献

From a Locusta migratoria genomic DNA library, a gene has been isolated that codes for a previously unrecognized hemolymph protein of Mr = 19,000, designated 19k protein. The gene has at least five exons, extending over about 9 kb of DNA. Its polypeptide product, obtained by cell-free translation of mRNA selected from adult fat body RNA by hybridization with the cloned DNA, is precipitated by antiserum against a low molecular weight hemolymph protein fraction. The mature protein product has been purified from locust hemolymph, and an N-terminal sequence of 20 amino acids has been determined. In polyacrylamide gel electrophoresis, this protein comigrates with apolipophorin III, from which it was previously not distinguished, but it is clearly distinct by amino acid composition and sequence. The genomic clone was used as a probe to isolate a fat body cDNA clone of the 19k protein mRNA. The 938-base pair cDNA clone contains a 516-base pair open reading frame. The deduced 172-amino acid polypeptide includes an apparent signal peptide, a sequence of four amino acids that may represent a prosegment, and a sequence identical (with a single exception, which may reflect polymorphism) with the N-terminal sequence of the hemolymph protein. Its mRNA occurs at a low level in late larval fat body, is abundant in the newly eclosed adult, then declines to a low level, and rises again at days 8–10; it is greatly reduced after destruction of the corpora allata with precocene and then is elevated after treatment with methoprene, suggesting stimulation by juvenile hormone. During Locusta migratoria embryogenesis, the yolk is progressively degraded and the resulting metabolites are released in the haemolymph. We researched the organs possibly involved in the uptake of haemolymphatic proteins. Among organs originated from mesoderm, the SOB (suboesophageal bodies) situated in the embryonic head are remarkable by a very early acquisition of differentiated cytological characters, while most other cells of the embryo are undifferentiated. The SOB quite disappear before hatching. Just before katatrepsis stage, the other organs derived from mesoderm begin to differentiate, including the PC (pericardial cells) which take over from the SOB. These cells, situated in thorax and abdomen, are developed during the dorsal close of embryo. The development and the ultrastructural changes of the SOB cells and of the PC were studied during an embryogenesis with diapause. The morphology of embryos which enter diapause is comparable with that of a continuous development at the beginning of katatrepsis. However, the cells of SOB and PC cells suffer from remarkable changes not only physiologically but cytologically. At the beginning of diapause, the proteosynthetic activity practically disappears in the SOB cells and the lysis areas appear. Nevertheless, the exchanges between these cells and the haemolymph still remain important. For the period of cold, which is necessary to the resumption of development, the aspect of the SOB cells changes and in particular the areas of lysis become less wide. When the embryo reopens its development, the SOB cells show a proteosynthetic activity and the areas of lysis disappear. The changes of the SOB cells and of the PC cells are regularized during the resumption of the development: the SOB cells which had again taken a normal activity start to regress from the stage VII on, while the PC cells take over.  相似文献

An experiment was conducted to determine the effects of diet on the chemical composition of migratory locusts (Locusta migratoria L.). Fresh and dry weight and the contents of dry matter, ash, lipid, protein, Ca, K, Mg, Na, P, Cu, Fe, Zn, retinol, lutein, zeaxanthine, cryptoxanthin, carotenes, lycopene and gross energy were determined in penultimate instar and adult locusts, that had been fed three different diets. The locusts received a diet of grass or grass+wheat bran or grass+wheat bran+carrots. Adding wheat bran decreased the protein content and increased fat content (633 vs. 583 and 182 vs. 231 g/kg DM, respectively). Addition of carrots to the diet increased fat content further from 231 to 271 g/kg DM. Mineral concentrations of Ca, K, Mg, and Na, were significantly affected by diet. P, K, Cu, and Fe concentrations were significantly different in penultimate migratory locusts compared with adults. Wheat bran decreased the α-carotene content, which did not change by incorporating carrots in the diet. However, carrots did result in higher β-carotene concentrations. Retinol concentrations were increased by incorporating both wheat bran and carrots in the diet compared with the diet containing only grass. This study shows that the chemical composition of migratory locusts can be manipulated through the diet. As such, it enables nutritionists to adapt the chemical composition of live feeder insects to better meet the nutritional demands of predators.  相似文献

Methoprene, a juvenile hormone (JH) analog, is a widely used insecticide that also accelerates behavioral development in honey bees (Apis mellifera). JH regulates the transition from nursing to foraging in adult worker bees, and treatment with JH or methoprene have both been shown to induce precocious foraging. To determine how methoprene changes honey bee behavior, we compared JH titers of methoprene‐treated and untreated bees. Behavioral observations confirmed that methoprene treatment significantly increased the number of precocious foragers in 3 out of 4 colonies. In only 1 out of 4 colonies, however, was there a significant difference in JH titers between the methoprene‐treated and control bees. Further, in all 4 colonies, there was no significant differences in JH titers between precocious and normal‐aged foragers. These results suggest that methoprene did not directly affect the endogenous JH secreted by corpora allata. Because methoprene caused early foraging without changing workers’ JH titers, we conclude that methoprene most likely acts directly on the JH receptors as a substitute for JH.  相似文献

In the Savio strain of Locusta migratoria an imaginai diapause is induced by long daylength. In diapausing females, the haemolymph level of juvenile hormone (JH) was undetectable during the first 3-wk of imaginai life and later rose only slightly to about 20 ng/JH3IR per ml. Only peripheral cells of the corpora aliata (CA) were active. In nondiapausing animals, or after the termination of diapause, the JH level was high (140–200 ng/ml) and the ultrastructure of the gland exhibited signs of activity. CA severance in 3-wk-old diapausing females terminated diapause as a result of activation of the CA. CA disconnection in the fifth larval instar or at the imaginai moult in long daylength animals did not break diapause and the CA stayed inactive. The lateral cells of the protocerebrum exert a jdual effect: at the end of larval life they bring about CA maturation and render them active, whereas during the imaginai diapause they inhibit CA activity. The median neurosecretory cells of the pars intercerebralis support CA activity during vitellogenesis.  相似文献
