Ticks represent a large group of pathogen vectors that blood feed on a diversity of hosts. In the Americas, the Ixodidae ticks Amblyomma cajennense are responsible for severe impact on livestock and public health. In the present work, we present the isolation and molecular identification of a group of culturable bacteria associated with A. cajennense eggs from females sampled in distinct geographical sites in southeastern Brazil. Additional comparative analysis of the culturable bacteria from Anocentor nitens, Rhipicephalus sanguineus and Ixodes scapularis tick eggs were also performed. 16S rRNA gene sequence analyses identified 17 different bacterial types identified as Serratia marcescens, Stenotrophomonas maltophilia, Pseudomonas fluorescens, Enterobacter spp., Micrococcus luteus, Ochrobactrum anthropi, Bacillus cereus and Staphylococcus spp., distributed in 12 phylogroups. Staphylococcus spp., especially S. sciuri, was the most prevalent bacteria associated with A. cajennense eggs, occurring in 65% of the samples and also frequently observed infecting A. nitens eggs. S. maltophilia, S. marcescens and B. cereus occurred infecting eggs derived from specific sampling sites, but in all cases rising almost as pure cultures from infected A. cajennense eggs. The potential role of these bacterial associations is discussed and they possibly represent new targets for biological control strategies of ticks and tick borne diseases. 相似文献

Abstract. The effects of adults of the bont tick, Amblyomma hebraeum on the milk production of Sanga and Sanga x zebu (Brahman) cattle were measured over a period of 11 weeks in the lowveld of Zimbabwe in the summer of 1986. Four groups of lactating cows, consisting of two breeds, each divided into a high and low tick treatment, were exposed to very low or high challenges of ticks and their milk production measured by weighing their calves before and after suckling. The liveweight gains (LWG) of the calves were also measured.
Tick burdens on the infested groups averaged around fourteen engorging females of A.hebraeum per day, which amounted to infestations of about 150 adult ticks. That is greater than most observed field infestations. This caused no significant reduction in milk yield or calf growth over the whole period, provided the teats of the dams had not been damaged by ticks. Mismothering occurred when teats were damaged. No breed differences were observed so all data was pooled for further analysis. Average calf LWGs of the high tick groups were reduced by 2.2kg (P < 0.01) during one 4-week period but overall the 3.9 kg difference in LWG of the tick treatment groups was not quite significant (P < 0.10). Although there was a poor relationship between tick numbers and reduced milk yield or calf LWG, the effects were always in the direction expected. The effects averaged 6 ± lOg reduction of milk and 2.6 ± 1.8g loss of LWG of calves for every female tick that engorged. It was concluded that milk production is not an important consideration when estimating the losses in production caused by A.hebraeum on Brahman x Sanga or Sanga breeds of cattle. The tick vectors of heartwater (Cowdria ruminantium infection) in Zimbabwe, Amblyomma hebraeum and Amblyomma variegatum, historically were believed to be confined to the low-lying regions of the south and north-west of the country. However, country-wide surveys performed in 19751980 and 19881991 demonstrated that both species were also established in western parts of the highveld plateau and had started to encroach on the predominantly heartwater-free central and eastern highveld regions. To determine the current distributions of both the vectors and evaluate the potential threat of heartwater to animals in the highveld, a survey of ticks infesting cattle was performed in 1996 at 2994 locations in small-holder and large-scale commercial farming areas throughout Zimbabwe. Amblyomma hebraeum was collected at 1329 locations, A. variegatum at 72 locations and both A. hebraeum and A. variegatum at 13 locations. The results demonstrated that A. hebraeum was present, as previously recorded, throughout the southern half of the country and appeared to have undergone further limited spread into the central and eastern highveld regions. Only the northern-most region of the country appeared to be free of this species. Amblyomma variegatum was collected mainly in the north-west, as previously recorded, but was also found at isolated locations across the central highveld region and along the eastern border with Mozambique. This species was, however, still absent from the southern half and the northern-most regions of the country. An overlap of the distributions of the two species existed within a zone along the southern and eastern regions of the distribution of A. variegatum. Ecdysteroid titer in whole extracts of female O. gammarellus varies during the course of the moult cycle: it is minimal at the beginning of the intermoult cycle (stages A, B and Ca) and in D0, and maximal in D2. In hemolymph, the hormone content presents a single peak level in D2; this peak is constituted by ecdysterone, as revealed by HPLC. Ovaries, at the end of vitellogenesis, contain ecdysterone and low polarity substances. Ecdysteroids are also present in eggs after laying. Variations in ecdysteroid levels seem to influence oocyte growth; vitellogenin synthesis, however, is independent of hormonal fluctuations. 相似文献

In order to understand the composition and quantitative variation of ecdysteriods in the adults and eggs of Opogona sacchari (Bojer), an invasive alien pest, we analyzed the ecdysteroid composition and titers in the adult and egg of this pest. On day 4 after eclosion, the titer of ecdysteriods in the male adult was 0.080 ng/adult, much lower than 5.978 ng/adult in the female adult. During the development of ovaries, the titer of ecdysteriods was low on the first two days, and high in the late period, with the peak (10.48 ng/ovary) appearing on day 3. During the development of eggs, the titer of ecdysteriods was about 0.010 ng/egg from day 1 to day 3, and then decreased to 0.006 ng/egg on day 4. In both adults and eggs, three main components of ecdysteriods were found by identification of HPLC/RIA. They were 20-hydroxyecdysone, 26-hydroxyecdysone, and an unidentified component. 相似文献

Ecdysteroid levels in the developing eggs of Schistocerca gregaria were determined, at daily intervals, using gas chromatography and electron capture detection of ecdysteroid derivatives. Ecdysone and 20-hydroxyecdysone were present both as the free ecdysteroid and as polar conjugates. Total ecdysteroids reached a maximum of 40 ng/egg with ecdysone contributing the greater part. 相似文献

Octopamine elicited a dose-related secretory response by salivary glands isolated from the feeding female tick Amblyomma americanum. Half-maximal stimulation occurred at about 60 μM. Phentolamine (10 μM) failed to inhibit the octopamine-mediated response; however, thioridazine (50 μM) inhibited both octopamine (1,000 μM) and dopamine-stimulated (0.1 μM) secretion. Maximal stimulation by dopamine (1.0 μM) showed no further increase in the rate of secretion after adding octopamine (1,000 or 0.1 μM). Glands responded to octopamine (100 μM) with rates significantly lower than controls following exposure to amphetamine (1,000 μM). Octopamine receptors do not appear to mediate the secretory response, and octopamine may stimulate secretion by releasing catecholamines from presynaptic neurons. These results support the hypothesis that dopamine is the natural transmitter mediating fluid secretion in the feeding tick salivary gland. 相似文献

Aim To offer an objective approach to some of the problems associated with the development of logistic regression models: how to compare different models, determination of sample size adequacy, the influence of the ratio of positive to negative cells on model accuracy, and the appropriate scale at which the hypothesis of a non‐random distribution should be tested. Location Test data were taken from Southern Africa. Methods The approach relies mainly on the use of the AUC (Area under the Curve) statistic, based on ROC (threshold Receiver Operating Characteristic) plots, for between‐model comparisons. Data for the distribution of the bont tick Amblyomma hebraeum Koch (Acari: Ixodidae) are used to illustrate the methods. Results Methods for the estimation of minimum sample sizes and more accurate hypothesis‐testing are outlined. Logistic regression is robust to the assumption that uncollected cells can be scored as negative, provided that the sample size of cells scored as positive is adequate. The variation in temperature and rainfall at localities where A. hebraeum has been collected is significantly lower than expected from a random sample of points across the data set, suggesting that within‐site variation may be an important determinant of its distribution. Main conclusions Between‐model comparisons relying on AUCs can be used to enhance objectivity in the development and refinement of logistic regression models. Both between‐site and within‐site variability should be considered as potentially important factors determining species distributions. 相似文献

We studied the ultrastructural organization of the ovarian follicles in a placentotrophic Andean lizard of the genus Mabuya. The oocyte of the primary follicle is surrounded by a single layer of follicle cells. During the previtellogenic stages, these cells become stratified and differentiated in three cell types: small, intermediate, and large globoid, non pyriform cells. Fluid‐filled spaces arise among follicular cells in late previtellogenic follicles and provide evidence of cell lysis. In vitellogenic follicles, the follicular cells constitute a monolayered granulosa with large lacunar spaces; the content of their cytoplasm is released to the perivitelline space where the zona pellucida is formed. The oolemma of younger oocytes presents incipient short projections; as the oocyte grows, these projections become organized in a microvillar surface. During vitellogenesis, cannaliculi develop from the base of the microvilli and internalize materials by endocytosis. In the juxtanuclear ooplasm of early previtellogenic follicles, the Balbiani's vitelline body is found as an aggregate of organelles and lipid droplets; this complex of organelles disperses in the ooplasm during oocyte growth. In late previtellogenesis, membranous organelles are especially abundant in the peripheral ooplasm, whereas abundant vesicles and granular material occur in the medullar ooplasm. The ooplasm of vitellogenic follicles shows a peripheral band constituted by abundant membranous organelles and numerous vesicular bodies, some of them with a small lipoprotein core. No organized yolk platelets, like in lecithotrophic reptiles, were observed. Toward the medullary ooplasm, electron‐lucent vesicles become larger in size containing remains of cytoplasmic material in dissolution. The results of this study demonstrate structural similarities between the follicles of this species and other Squamata; however, the ooplasm of the mature oocyte of Mabuya is morphologically similar to the ooplasm of mature oocytes of marsupials, suggesting an interesting evolutionary convergence related to the evolution of placentotrophy and of microlecithal eggs. Two tick species Aponomma hydrosauri and Amblyomma limbatum that infest large reptiles have an abrupt parapatric boundary near Mt Mary in South Australia. A previous model has suggested that the boundary is maintained at population density troughs resulting from habitat heterogeneity along a gradual environmental gradient. This paper describes the dynamics of the boundary on three transects over 17 years, 1982-98. Over the last seven years of that period there has been a significant increase in rainfall. At the same time, the boundary position has moved 1-2 km on the transects, with the more mesic adapted Ap. hydrosauri advancing into the distribution of the more xeric adapted Amb. limbatum. Also over the same time the density of ticks on lizards in regions flanking the boundary zone has increased for Ap. hydrosauri and decreased for Amb. limbatum. These results suggest that the environmental gradient has been altered, perhaps by increased rainfall, to favour Ap. hydrosauri, which has been able to colonize more successfully across the density troughs and extend its distribution. 相似文献

A study of acquired resistance in guinea-pigs, guinea-fowl and tortoises to larvae of the ticks Amblyomma hebraeum Koch and A.marmoreum Koch (Acari: Ixodidae) showed that repeated infestations of the laboratory host resulted in a 29.3-49.4% reduction respectively in weight of engorged larvae. No resistance was induced in guinea-fowl, the natural host for larvae of both species. A small decline in the mean weight of engorged larvae was observed for both species of ticks after three successive infestations on tortoises. The reduction in weight was more pronounced in A.hebraeum (14.5%) which does not naturally feed on tortoises than for A.marmoreum (8.4%), a common tick on this host. The occurrence of acquired resistance in natural tick host relationships as opposed to unnatural tick host relationships is discussed. 相似文献

Sublethal doses of the organophosphate Dursban were tested on non-infected and tick-borne encephalitis virus (TBEV)-infected nymphs of Amblyomma hebraeum and Ixodes persulcatus. It was shown that contact with this pesticide enhanced the survival of non-infected ticks, whereas TBEV infection reduced survival. This effect of TBEV infection was greater in I. persulcatus than in A. hebraeum ticks. This phenomenon may be the result of a virus-specific action on nymphs, whereby the virus may enhance activity and thus food reserves may deplete faster. Our data emphasize that the acaricidal action on non-infected ticks and those infected by different tick-borne pathogens may be different. 相似文献

Titers of juvenile hormone (JH) III and free ecdysteroids were studied in the hemolymph of the ovoviviparous Argentinian cockroach, Blaptica dubia, related to fat body depletion and reproduction. Adult females were analyzed during the first (days 5–25) and second vitellogenic cycle (days 80–100) and during the periods of gestation. Body weight changes of adult females were closely related to ovarian growth, ootheca formation, ootheca deposition, and hatching of the nymphs. Biochemical analysis of the fat body revealed lipids as the main storage compounds, followed by glycogen, proteins, and free carbohydrates. Changes in the fat body weight and in the chemical constituents of the fat body correlated with the processes of vitellogenesis and gestation. Concentrations of JH and free ecdysteroids in the hemolymph were measured by high pressure liquid chromatography-mass spectrometry. JH III was the only JH homolog found. JH III titers were high during vitellogenesis as well as toward the end of the gestation period. Changes in the concentrations of ecdysone and 20-hydroxyecdysone were less clear. The results reveal JH III as the major gonadotropic hormone in adult females of B. dubia. 相似文献

Sexual dimorphism and the female reproductive cycle were studied in a population of the viviparous lizard Trachylepis vittata at 2000 m a.s.l. on Mount Sannine, Lebanon. Females have larger body sizes than males and males have relatively larger heads than females. Females reach maturity at 56 mm snout-vent length. They spend at least six months in hibernation, from October to March. Adult females emerge from hibernation in April. Fertilization occurs mid-May and gestation lasts for 8-10 weeks. All females collected on the same date had embryos at the same embryonic developmental stage. Females produced 1-4 embryos. There is a significant positive relationship between female body size and number of embryos. Parturition lasts for two weeks and new-borns achieve adult size within about ten months. 相似文献

The simulation model LSTSIM was revised and adapted to estimate the effects of different management strategies on populations of the lone star tick, Amblyomma americanum (Linnaeus), in non-agricultural areas. The control technologies evaluated were area-wide acaricide application, acaricide self-treatment of white-tailed deer, vegetation reduction and reductions in the density of white-tailed deer. Estimated changes in tick populations using these technologies compared favorably with results from actual integrated tick management studies at five different geographic locations. Area-wide acaricide application and vegetation reduction, as well as combinations of the two technologies proved to be useful for short-term, seasonal management of ticks in small non-agricultural areas and residential sites. Self-treatment of deer with acaricide, applied topically or as a systemic, appears to be the most cost-effective technology for use in area-wide, long-term programs for tick control. The results of simulations suggest that reductions in deer density should be considered as a component of any tick management program. 相似文献

A case of tick bite was found in the inguinal region of a 74-year-old Korean woman. She was attacked by the tick while working in her vegetable garden in the vicinity of mountain located in Suncheon City, the southern coastal area of the Korean Peninsula. On admission she complained of mild discomfort and itching around the bite area. The causative tick was 23 mm long and had slender pedipalps. The scutum was quite ornate and had eyes at the edge. The genital aperture was located anterior to the level of the coxa II. The spiracular plate was comma-shaped and the anus was surrounded posteriorly by the anal groove. The coxa I had subequal 2 spurs; the external one slightly larger. The spur of coxa IV was slightly longer than those of coxae II and III. The tarsus IV had 2 distinct subapical ventral spurs. It was identified as the fully engorged adult female of Amblyomma testudinarium. This is the first human case of Amblyomma bite in Korea. 相似文献

At the beginning of the 1999 rainy season, three traditional cattle herds were monitored for 48 days while grazing in the bushy savannah of southwestern Burkina Faso. Cattle in each herd were caught on several occasions each day and the attached ticks were counted. This confirmed that Amblyomma variegatum Fabricius (Acari: Ixodidae) adults picked up in the pastures mainly attach to the interdigital areas (87% of the 791 ticks captured), and reach the predilection sites later (chest and udder/inguinal area) when the animals lie down. As many females as males attached to the hosts, but the seasonal distribution was very heterogeneous, with only a few females attaching as long as the humidity rate remained low. It is suggested that this prevents eggs from being laid when conditions are not optimal for their survival and that of the larvae. Ticks attached all day but the number picked up hourly and daily varied greatly according to their density on the pasture. As a general trend, confirmed by another study carried out in 2005, the number of ticks picked up daily increased from less than one tick/animal/day, before the onset of the rainy season, to 6.5 (+/- 1.5) ticks/animal/day on average during the infestation peak, which lasted 6-8 weeks, until early or mid-July. The number then decreased to less than one tick/animal/day from the end of July onwards. The infestation on the predilection sites followed the same trend, with a daily tick burden increase of three to five A. variegatum adults, depending on herd and year, during the infestation peak. 相似文献

A perianal tick and the surrounding skin were surgically excised from a 73-year-old man residing in a southwestern costal area of the Korean Peninsula. Microscopically a deep penetrating lesion was formed beneath the attachment site. Dense and mixed inflammatory cell infiltrations occurred in the dermis and subcutaneous tissues around the feeding lesion. Amorphous eosinophilic cement was abundant in the center of the lesion. The tick had Y-shaped anal groove, long mouthparts, ornate scutum, comma-shaped spiracular plate, distinct eyes, and fastoons. It was morphologically identified as a fully engorged female Amblyomma testudinarium. This is the third human case of Amblyomma tick infection in Korea. 相似文献

Amblyomma americanum (L.) is a three‐host tick that spends most of its life off‐host and is an important vector of pathogens in the eastern United States. Our objectives were to develop a predictive statistical model that describes the number of active, off‐host larvae from 2007 to 2011 and to determine the environmental variables associated with this pattern. Data used in this study came from monitoring conducted in northeast Missouri in which off‐host ticks were collected from a permanent plot in a forest and an old field habitat every other week from approximately February to December. Variables examined were day length, degree days, total precipitation prior to sampling, wind speed, saturation deficit, number of adults prior to sampling, and collection site. Of the four regression models tested, the negative binomial model was selected. Fitted candidate models were compared relative to one another using values of eight model selection criteria and model averaging was used to develop a predictive model. The residual plots indicated that the weighted average model performs well in describing the number of larvae. Of the variables considered, the number of larvae was most strongly associated with increasing degree days, the number of active adults prior to sampling, and the forested site. 相似文献
