Olive, corn, sesame or peanut oil which have been used as vehicles in the immature rat uterotrophic assay or Hershberger assay, for detection of endocrine disrupting effects of environmental chemicals, was administered to ten immature female rats by subcutaneous injection from postnatal day (PND) 21 for 3 or 7 days, and each oil was also administered to ten male rats from PND 21 for 7 and 10 days. The body weights, and the weights of sex and sex accessory organs in female and male rats were measured. There were no significant differences in body weights of female rats between each oil group and the control group, while the body weight of male rats in the group given peanut or olive oil was significantly increased from 8 or 9 days after administration. There were no changes in the sex and sex accessory organ weights of female or male rats related to the endocrine disrupters. The results of the body weights and organ weights demonstrate that each oil is a suitable vehicle for the immature rat uterotrophic assay. However, each oil is suggested to be unsuitable for the Hershberger assay, because the analysis of changes of sex accessory organ weights in this assay might be confused by the increased body weights.  相似文献

Summary

Electron microscopy was used to examine the morphology of a putative endocrine gland, the juxtaganglionar organ (JO), and its relation to the cerebral ganglia of the hermaphroditic opisthobranch gastropod Aplysia juliana. The JO is a well-vascularized, poorly innervated tissue of glandular cells—rich in mitochondria, lipids, ribosomes, and endoplasmic reticulum, with sparse cilia and membrane-limited secretory granules—within the connective tissue sheath just exterior to the neuronal soma in the dorsal and posterior portions of the cerebral ganglia. The cytology and organization of the JO supports its homology to the dorsal bodies of pulmonate gastropods, which axe endocrine organs known to release one or more female gonadotropic factors.  相似文献

Data from 25 male and 44 female free-ranging dusky leaf monkeys, Presbytis obscura, are analyzed. In a male the mean total testicular weight, 5.82 g, represents 0.078% of the mean adult body weight, 7.39 kg. The mean total ovarian weight, 0.52 g, represents 0.008% of the mean female adult body weight, 6.47 kg. A moderate degree of sexual dimorphism thus exists in this species, female body weight representing 88% that of the male. No seasonal variation in the adult body weight or total gonadal weight of either sex occurs. Only in males is there a strong positive correlation between left and right gonadal weight, and no correlation exists between the total gonadal weight and the estimated age of either male or female adults. Among adults, only females evince a significant positive correlation between total gonadal weight and body weight. No marked change in total ovarian weight occurs during the menstrual cycle, pregnancy, or lactation.  相似文献

The reproductive tract of B. truncatus was investigated histologically in order to study possible effects of castration upon the accessory sex glands. In the female part of the reproductive tract—subdivided into albumen gland, oviduct, muciparous gland, oothecal gland, uterus, vagina and bursa copulatrix—13 histochemically different secretory cell types were distinguished. The majority produce different types of (acid) mucopolysaccharides. The roles of the various parts of the female tract in the production of an egg mass were elucidated by comparing the histochemistry of the egg mass to that of the female tract; the abundance and location of the cell types were also taken into account for this purpose.

The male part appeared to contain 12 histochemically different secretory cell types. These produce mainly (phospho lipoproteins together with some polysaccharides and neutral lipids.

Castration causes an acceleration of the growth of the snails. The volumes of female (albumen gland) and male (prostate gland) accessory sex glands were measured on histological sections. It appeared that growth of the albumen gland is not arrested by castration. This was not established beyond doubt for the prostate gland. The results suggest that the stimulating effects of the dorsal body hormone on the growth and synthetic capacity of the female accessory sex glands—such effects have been established for Lymnaea stagnalis—are not exerted via the ovotestis in B. truncatus.  相似文献

Summary Results obtained with castration and gonad implantation verify that a factor(s) from the gonad is required for development of the female accessory sex organs (ASO) inLimax maximus. In addition, they show that a gonadal factor promotes rapid gonadal growth and sperm production, and gradual or incomplete development of the penis. Although an immature slug that is castrated and then switched to inductive photoperiods fails to exhibit reproductive tract maturation, in two differently designed experimental tests, the brains of such animals were able to promote sexual maturation when subsequently paired with an immature gonad. We interpret the results of these experiments as evidence for photoperiodic induction of secretion of maturation-inducing factor (MH) by the brain in the absence of the gonad. One or more gonadal hormones appear to be both necessary and sufficient for production of sperm and for development of ASO, but they are not required for the photoperiodic stimulation of MH release.Abbreviations ASO accessory sex organs - MH maturation hormone  相似文献

Summary

A study was made of the interaction of the photoperiod and the availability of food in influencing egg laying of the freshwater snail Lymnaea stagnalis. At a long day photoperiod (LD = 16 h light/8 h dark) egg laying of fed snails had increased compared with that of fed animals kept at a medium day photoperiod (MD = 12 h light/12 h dark). In MD snails oviposition ceased within a week of the beginning of a starvation period. This is most probably due to a reduction in the activities of the neuroendocrine caudodorsal cells which secrete an ovulation hormone. In contrast, in starved LD snails a low rate of ovipository activity continued, indicating that a lowered frequency of caudodorsal cell release cycles occurred under these conditions. The decreased mean size (number of eggs) of the egg masses in starved LD snails indicates that the activities of the endocrine dorsal bodies, which control vitellogenesis and synthetic activities in the female accessory sex organs, had decreased.

All MD snails survived, but nearly all LD snails died during the course of the experiment. Determinations of the mantle glycogen stores of LD snails suggest that the high mortality of LD snails is due to exhaustion of the animal's energy reserves.  相似文献

Bi-directional sex change in the deep-water gobiid fish Trimma yanagitai was examined. The gonads of all individuals consisted of ovarian and testicular elements, and an accessory gonadal structure. In no gonads were both testicular and ovarian parts simultaneously active. Bi-directional sex changes occurred during the rearing experiments in aquaria under conditions of which there was co-existence of two males or plural females. The sex of individuals could be determined by their relative body size or social dominance: the largest individuals acting as male and the remainder as female.  相似文献

Summary

Populations of the fresh-water snail Lymnaea peregra from different localities vary in their capacity to lay eggs, responding either to long day or to short day photoperiods. These responses are genetically determined by a single gene locus with the long day allele dominant over the short day allele. Although previtellogenic stages of oogenesis are not affected by an inhibitory photoperiod, vitellogenesis is essentially shut down. The volumes of the dorsal bodies, which are endocrine organs that mediate vitellogenesis, are significantly smaller in snails maintained in an inhibitory photoperiod. Implantation of cerebral ganglia with attached dorsal bodies, both from egg laying animals and from dormant animals that have the potential to lay eggs in the postoperative photoperiod, into snails that were inhibited by insufficient photoperiod resulted in a significant increase in vitellogenesis in the ovotestes of the hosts. An increase in host dorsal body volume was also observed. The photoperiod gene appears to control some aspect of the cerebral ganglion that activates the dorsal body to produce a hormone that stimulates vitellogenesis in the ovotestes. Photoperiodic control of reproduction may be a factor contributing to the diverse life history strategies observed in this species.  相似文献

Treatment of adult male rats with oestradiol benzoate (OB) for 21 days significantly decreased the body, testicular and accessory sex organ weights but increased anterior pituitary weight. OB treatment also significantly suppressed circulating FSH and LH levels as well as plasma and testicular concentrations of testosterone. The seminiferous tubules and interstitial cells were partly atrophied, and there was some effect on spermatogenesis, with step 14 to 19 spermatids being fewer than normal. Rats treated with OB for 21 days were then treated daily with LH-RH analogue ((D-Leu6, des-Gly-NH2(10))-LH-RH-ethylamide), to see if testicular function could be recovered. Circulating gonadotrophins were significantly elevated, testicular histology was normal and testicular and plasma testosterone concentrations and the accessory sex organ weights remained suppressed. These results suggest possible extra-pituitary effects of the LH-RH analogue, including a direct action on the testes and/or accessory sex organs.  相似文献

The sexual differentiation of brain and behavior is reviewed from the findings of sex differences in the vomeronasal pathway. A motivational approach to sex differences in reproductive behavior is stressed by taking into account that sex differences are present in neural networks: from the receptor organ (the vomeronasal organ) to effector nuclei. Sex differences in the brain appear in two morphological patterns. In one, the male presents greater morphological measurements than the female; in the other, the opposite occurs. These two morphological patterns are actively differentiated by gonadal steroids. The functional significance of these two morphological patterns is addressed. Moreover, since the GABAAreceptor is involved in the organization of sex differences in vomeronasal structures such as the accessory olfactory bulb and in maternal behavior, the role of membrane mechanisms, 5α reduced hormones, and neurosteroids in the sexual differentiation process is discussed.  相似文献

Background

Klinefelter syndrome (KS), caused by XXY karyotype, is characterized by low testosterone, infertility, cognitive deficits, and increased prevalence of health problems including obesity and diabetes. It has been difficult to separate direct genetic effects from hormonal effects in human studies or in mouse models of KS because low testosterone levels are confounded with sex chromosome complement.

Methods

In this study, we present the Sex Chromosome Trisomy (SCT) mouse model that produces XXY, XYY, XY, and XX mice in the same litters, each genotype with either testes or ovaries. The independence of sex chromosome complement and gonadal type allows for improved recognition of sex chromosome effects that are not dependent on levels of gonadal hormones. All mice were gonadectomized and treated with testosterone for 3 weeks. Body weight, body composition, and motor function were measured.

Results

Before hormonal manipulation, XXY mice of both sexes had significantly greater body weight and relative fat mass compared to XY mice. After gonadectomy and testosterone replacement, XXY mice (both sexes) still had significantly greater body weight and relative fat mass, but less relative lean mass compared to XY mice. Liver, gonadal fat pad, and inguinal fat pad weights were also higher in XXY mice, independent of gonadal sex. In several of these measures, XX mice also differed from XY mice, and gonadal males and females differed significantly on almost every metabolic measure. The sex chromosome effects (except for testis size) were also seen in gonadally female mice before and after ovariectomy and testosterone treatment, indicating that they do not reflect group differences in levels of testicular secretions. XYY mice were similar to XY mice on body weight and metabolic variables but performed worse on motor tasks compared to other groups.

Conclusions

We find that the new SCT mouse model for XXY and XYY recapitulates features found in humans with these aneuploidies. We illustrate that this model has significant promise for unveiling the role of genetic effects compared to hormonal effects in these syndromes, because many phenotypes are different in XXY vs. XY gonadal female mice which have never been exposed to testicular secretions.

The expression of food-anticipatory activity (FAA) is induced by restricted feeding (RF), and its entrainment requires food-entrainable oscillators, the neuroanatomical basis of which is currently unclear. Although RF impacts various hormones, sex-related differences in FAA are unclear. 'Here, we report significantly more food-anticipatory wheel-running activity in male than in female mice during RF. In parallel with the sex-related difference in FAA, male and female mice display different food intake and body weight in response to RF. Since gonadal hormones could be involved in the sex-specific difference in FAA, we compared sham and gonadectomized male and female wild-type mice. In gonadectomized mice, the sex difference in FAA was abolished, indicating a role for gonadal hormones in FAA. Further, plasma concentrations of the hormone ghrelin were higher in female than in male mice during ad libitum (AL) feeding, and RF induced a temporal advance in its peak in both sexes. RF also shifted the expression peak of the circadian gene mPer1 in the hippocampus and liver, although no sex difference was found in either the level or the cyclic phase of its expression. Per1Brdm1 mutant mice were still sexually dimorphic for FAA, but diminished FAA was noted in both male and female Per2Brdm1 mutant mice. In summary, our results imply that gonadal hormones contribute to the sex difference in FAA, possibly through modulating ghrelin activity.  相似文献

The diversity of inputs that guide sexual fate during development is both intriguing and daunting. In the field of fish biology, the study of sex determination is of great importance. For example, in aquaculture, sexually dimorphic growth rates and overall size leads to one sex being more marketable than the other. Moreover, for breeding purposes it is important to maintain balanced sex ratios. Furthermore, sex determination is sensitive to environmental factors, such as temperature and contaminants, which can lead to skewed sex ratios, intersexes and sterility in wild or farmed fish. The gonad is typically the first organ to exhibit morphological signs of sexual dimorphism and therefore is likely to be the primary organ system whose fate is controlled by the sex determination cues in many fish species. Additionally, the sexual fate of the gonad has been shown to fully or partially control organismal sex differentiation. Thus, understanding the genetic regulation of gonadal sex differentiation is critical in studies of fish sex determination. This review summarizes recent knowledge of genes expressed during gonadal sex differentiation in gonochoristic teleost fish. Three species are discussed, which serve as excellent model systems for probing teleost sex differentiation: the Oreochromis niloticus, Oryzias latipes and Danio rerio. The similarities and differences between gonadal gene expression in these three species and in comparison to mammals suggest conserved roles during vertebrate gonadal sex differentiation. In the future, it will be essential to develop tools to assay the function of genes expressed during gonadal sex differentiation in fish.  相似文献

The aim of this study is to characterize the population biology (sex ratio, size structure, relative growth of reproductive characters and realised fecundity) of the crab Microphrys bicornutusfrom Isla Margarita, Venezuela. Nearly 100 crabs were collected and fixed monthly during 1998. In the laboratory, crabs were sexed and measured: maximum carapace length, chelae length, chelae and abdomen width for females and first pleopod length for males. Additionally, 96 ovigerous females were collected to determine fecundity. The following measurements were taken for each female: body wet weight (BWW), body dry weight (BDW), egg wet weight (EWW), egg dry weight (EDW) and number of eggs (NE). The following determinations were made: relative fecundity (RF) = NE/BDW and reproductive output (RO) = (EDW/ BDW) × 100. Relative size at the onset of maturity (RSOM) was calculated as minimum ovigerous female/female's maximum size. The results indicate that the sex ratio is nearly 1:1 during the entire year. Ovigerous females and moulted males and females were observed throughout the year. Size frequency distribution and size at the onset of sexual maturity, estimated by relative growth of the sexual secondary characters and RSOM value, differed from those estimated in studies done in Jamaica and Buchuaco, Venezuela.  相似文献

We present the first cross-sectional organ weight by body weight reference standards for captive baboons (Papio hamadryas). Organ weight data were obtained from necropsy reports for 634 healthy, pedigreed, captive female and male baboons. From summary statistics we calculated and fit cross-sectional sex-specific percentile curves for: adrenals, brain, eyes, heart, kidneys, liver plus gall bladder, lungs, pancreas, pituitary gland, spleen, and thyroid gland in three kilogram body weight intervals and cross-sectional summary statistics by sex for each organ in one kilogram body weight intervals.  相似文献

Differences between male and female mammals are initiated by embryonic differentiation of the gonad into either a testis or an ovary. However, this may not be the sole determinant. There are reports that embryonic sex differentiation might precede and be independent of gonadal differentiation, but there is little molecular biological evidence for this. To test for sex differences in early-stage embryos, we separated male and female blastocysts using newly developed non-invasive sexing methods for transgenic mice expressing green fluorescent protein and compared the gene-expression patterns. From this screening, we found that the Fthl17 (ferritin, heavy polypeptide-like 17) family of genes was predominantly expressed in female blastocysts. This comprises seven genes that cluster on the X chromosome. Expression analysis based on DNA polymorphisms revealed that these genes are imprinted and expressed from the paternal X chromosome as early as the two-cell stage. Thus, by the time zygotic genome activation starts there are already differences in gene expression between male and female mouse embryos. This discovery will be important for the study of early sex differentiation, as clearly these differences arise before gonadal differentiation.

Androgen assimilation was investigated in a variety of accessory sex organs (seminal vesicles and anterior, dorsal, lateral, and ventral prostates) and in several nonaccessory sex organs in male Wistar rats. After administration of a pulse dose of [3H]testosterone in vivo to intact young (3–4 months old) rats, [3H]testosterone was the primary radioactive steroid recovered from most organs examined, except for the secondary sex glands where the reduced metabolites, [3H]5α-dihydrotestosterone (DHT) and [3H]5α-androstanediol(s), predominated. At longer postinjection times, [3H]DHT was preferentially retained in the accessory sex glands, presumably reflecting intracellular metabolism of [3H]testosterone to this compound and subsequent specific binding of [3H]DHT to receptor proteins. At the longest postinjection interval investigated, the ventral prostate retained greater concentrations of [3H]DHT than the lateral prostate which in turn had a higher [3H]DHT concentration than the seminal vesicles or anterior or dorsal prostates. The latter three glands retained approximately equal concentrations of [3H]DHT. Scatchard plot analyses of cytosol binding in 24-h castrates indicated that with one exception, the level of high affinity DHT binding sites was generally correlated with the retention of [3H]DHT in vivo in intact rats. Specifically, while the affinity for DHT binding in all accessory sex organs was the same, the number of high affinity binding sites per mg wet tissue weight was on the order of ventral prostate > anterior prostate ≥ seminal vesicles ≥ dorsal prostate > lateral prostate. Studies of the influence of aging to 22–26 months revealed no apparent differences in the affinity of the DHT receptor for its ligand in any of the accessory sex glands from 24-h castrates when the receptors were present in levels sufficiently high to quantify. The concentration of available DHT receptors with advancing age remained constant in the anterior and dorsal prostates, increased in the seminal vesicles, and declined in the ventral and lateral prostates. The decreases observed in the ventral prostate were only partial, but the receptors of the lateral prostate declined to nondetectable levels.  相似文献

A SURVEY OF NEUROENDOCRINE PHENOMENA IN NON-ARTHROPOD INVERTEBRATES   总被引:4,自引:0,他引:4
  • 1 Neurosecretory phenomena are apparently ubiquitous among Metazoa.
  • 2 In Hydra, neurosecretory products are probably involved in the control of growth and reproduction.
  • 3 Secretory elements in the central nervous system of turbellarians probably promote fission, regeneration and reproduction.
  • 4 The cerebral ganglia of nemertines are the source of a hormone which exercises an inhibitory influence on maturation of the gonads and the development of somatic sexual characteristics. A principle secreted by the ganglia and/or the associated cerebral organs controls weight regulation. A sex hormone controls sexual differentiation.
  • 5 Secretory neurons may influence the production of exsheathing fluid and ecdysis in nematodes.
  • 6 In nereid polychaetes, a single hormone which originates from the cerebral neuroendocrine system apparently promotes segment proliferation and inhibits maturation. However, in certain other polychaetes, contrasting endocrine mechanisms seem to operate.
  • 7 In lumbricid oligochaetes, a hormone secreted by the cerebral ganglion (and possibly by other nervous centres also) promotes gonadal activity and the differentiation of somatic sexual characters. In some species the ganglion exerts an inhibitory influence on the associated processes of ‘diapause’ and posterior regeneration. The cerebral and suboesophageal ganglia are implicated in the control of osmoregulation. Cerebral neurosecretory cells in limicolous oligochaetes may influence regenerative growth and osmoregulation.
  • 8 The cerebral ganglia of leeches secrete a gonadotrophic hormone.
  • 9 In gastropod molluscs, hormones secreted by the cerebral ganglia, glandular ‘dorsal bodies’ and/or optic tentacles are responsible for the control of the development of the reproductive tract, and the activity of the gonads. Contrasting mechanisms are thought to operate in the different gastropod groups and there is conflicting evidence particularly with respect to the endocrine functions of the optic tentacles and the gonads. The ‘bag cells’ of the abdominal ganglion of ApZysiu secrete a hormone which induces egg-laying. Cells in the pleural and parietal ganglia probably control osmoregulation in Lymnaea.
  • 10 The optic glands of cephalopods secrete a gonadotrophic hormone, but sex hormones are apparently absent. The functional significance of the neurovenous tissues (presumptive neurosecretory complexes of unusual character) is obscure in most cases.
  • 11 The radial nerves of starfish are the source of a hormone which induces the production of I-methyl adenine by the follicle cells of the gonad. This second principle stimulates oocyte maturation and the shedding of male and female gametes.
  • 12 The relevance of such information to the development of certain biological concepts and to various aspects of comparative physiology is briefly discussed.
