The incorporation of [3H]phenylalanine, [3H]tyrosine, and [3H]tryptophan into protein and amino acyl-tRNA was studied in cell-free preparations from rat brain. Tyrosine and tryptophan inhibited the incorporation of phenylalanine into protein, and tyrosine inhibited the incorporation of phenylalanine and tryptophan into amino acyl-tRNAs. In most cases, homogentisate, phenylpyruvate, and phenyllactate inhibited the incorporation of phenylalanine, tyrosine, and tryptophan into protein and amino acyl-tRNAs, and the incorporation of phenylalanine into polyphenylalanine. All other protein amino acids, and phenylacetate, salicylate, and benzoate were wholly ineffectual. The results suggest that the formation of amino acyl-tRNAs may have been the step which was affected most by the inhibitors. The incorporation data at different concentrations of the aromatic amino acids were fitted to the simple Michaelis equation. Homogentisate and phenylpyruvate generally tended to reduce both Km and V in the incorporation of aromatic amino acids into protein and amino acyl-tRNAs, even if V decreased more than Km.  相似文献

Effect of phenylalanine on protein synthesis in the developing rat brain   总被引:5,自引:7,他引:5  
1. Inhibition of the rate of incorporation of [(35)S]methionine into protein by phenylalanine was more effective in 18-day-old than in 8-day-old or adult rat brain. 2. Among the subcellular fractions incorporation of [(35)S]methionine into myelin proteins was most inhibited in 18-day-old rat brain. 3. Transport of [(35)S]methionine and [(14)C]leucine into the brain acid-soluble pool was significantly decreased in 18-day-old rats by phenylalanine (2mg/g body wt.). The decrease of the two amino acids in the acid-soluble pool equalled the inhibition of their rate of incorporation into the protein. 4. Under identical conditions, entry of [(14)C]glycine into the brain acid-soluble pool and incorporation into protein and uptake of [(14)C]acetate into lipid was not affected by phenylalanine. 5. It is proposed that decreased myelin synthesis seen in hyperphenylalaninaemia or phenylketonuria may be due to alteration of the free amino acid pool in the brain during the vulnerable period of brain development. Amyelination may be one of many causes of mental retardation seen in phenylketonuria.  相似文献

IN a previous report (MOKRASCH and MANNER, 1963) some of the characteristics of the incorporation of p4C]amino acids and pT]palmitate into the proteolipid from rat brain slices were detailed. This report deals with a further resolution of the incorporating system in centrifugal subfractions of rat brain homogenate. Some similarities in the reaction condition requirements are seen when the incorporations for slices are compared with those for particle preparations. Differences between the incorporation of p4C]amino acids into ptoteolipid by the system used here and incorporation by microsomal systems ate several. A report with preliminary information on the particulate system has appeared (MOKRASCH, 1963).  相似文献

The effect of undernutrition on the incorporation of [methyl-3H]thymidine into DNA and of 5-[3H]uridine into RNA of cerebral hemispheres, cerebellum, and brain stem was studied in vivo and in vitro in rats. The labeling of DNA from nuclei and mitochondria and of RNA from nuclei, mitochondria, microsomes, and soluble fractions, was also measured in vitro. The results demonstrate that nucleic acid synthesis is impaired and delayed during undernutrition. Specific effects were observed for the different brain regions and subcellular fractions: at 10 days nuclear and mitochondrial DNA and RNA synthesis was impaired, whereas at 30 days only the mitochondrial nucleic acid synthesis was affected.The delay of DNA and RNA labeling, caused by undernutrition, was most evident in the cerebellum, probably due to its intense cell proliferation during postnatal development. The specific sensitivity of mitochondria as compared to other subcellular fractions, may be due to the intense biogenesis and/or turnover of nucleic acids in brain mitochondria not only during postnatal development, but also in the adult animal.  相似文献

Ricin, a protein from the seeds ofRicinus communis which inhibits protein synthesis by eukaryotic ribosomes, is highly toxic when injected intraventricularly to rats, the LD50 being 0.241 g/rat at 72 hr and 0.084 g/rat at 7 days. Poisoned animals showed signs of central depression; they did not die before 20 hr of intoxication. Incorporation of amino acids in vivo into brain total protein and into brain ribosomes was impaired, as was protein synthesis in vitro by microsomes isolated from the brain of poisoned rats.  相似文献

The hydroxylated phenylethylamines p-tyramine, m-tyramine, octopamine, metaraminol and norepinephrine were accumulated by homogenates of rat brain much more vigorously than β-phenethylamine or amphetamine. The affinity concentrations (Km) for initial (5-min) uptake by homogenates of whole brain were 0.5, 3 and 6 μM for DL-norepine-phrine, p-tyramine and DL-octopamine, respectively. The uptake of these three hydroxylated compounds was much more vigorous in striatal tissue than in cortical tissue, and in both tissues the rate of uptake decreased in the sequence: norepinephrine > tyramine > octopamine. The uptake of these three substances was inhibited by reduced temperature, by lack of glucose, by CN- and DNP, and by desmethylimipramine, cocaine and ouabain. The uptake of norepinephrine and octopamine appeared to require Na+. Pretreatment of rats with reserpine or 6-hydroxydopamine decreased the ability of brain to take up norepinephrine or octopamine. Previously accumulated labelled phenylethylamines migrated in sucrose density gradients with a peak of radioactivity corresponding to an equilibrium position of catecholamine-containing nerve endings. The magnitude of the retention of [3H]amine in this synaptosornal peak decreased in the order: norepinephrine > octopamine > tyramine. The accumulated amines were released by sonic, osmotic and thermal stresses which disrupt neuronal membranes. The presence of a β-hydroxyl group appeared to protect amines from destruction by monoamine oxidase, presumably by virtue of uptake in presynaptic storage vesicles. During superfusion, tyramine and metaraminol appeared to displace [3H]norepinephrine from binding sites in brain slices.  相似文献

A growing body of evidence supports the role of free radicals in triggering the functional and metabolic disturbances following transient cerebral ischemia. This study was designed to evaluate whether the extent of reperfusion-induced inhibition of protein synthesis initiation as well as tissue injury can be reduced by Tanakan (Ginkgo biloba extract, EGb 761) (Beaufour-Ipsen Industrie). Rats received Tanakan in the dose of 40 mg/kg/day for 7 days before surgical intervention. Transient forebrain ischemia was induced by 4-vessel occlusion. Rats were subjected to 20 min of ischemia followed by 30 min, 4 h or 7 days of reperfusion. Protein synthesis rate, reinitiation ability and neurodegeneration in the frontal cortex and hippocampus were measured by the incorporation of radioactively labelled leucine into polypeptide chains in postmitochondrial supernatants and by Fluoro-Jade B staining. The protective effect was observed, concerning both the protein synthesis and the number of surviving neurons, in the Tanakan-treated groups. Tanakan significantly reduced the ischemia/reperfusion-induced inhibition of translation in the neocortex as well as in the highly sensitive hippocampus. Our results indicate that free radicals play an important role in the development of reperfusion-induced injury, and the treatment of ischemic and reperfused brain with free radical scavengers may reduce the severity of reperfusion damage.  相似文献

A single dose of L-dihydroxyphenylalanine (L-DOPA, 500 mg/kg, i.p.) that caused massive disaggregation of brain polysomes also suppressed the incorporation of [3H] lysine into trichloracetic acid (TCA)-precipitable proteins of cortex, cerebellum, hypothalamus, brainstem and striatum. The magnitude of this inhibition of [3H] protein synthesis was similar in all brain regions studied, and was not related to changes in the specific activity of the precursor amino acid.  相似文献

The subcellular distribution of histone and phosvitin kinase activities in brain has been studied and the ability of the various fractions to catalyse the phosphorylation of their endogenous proteins (intrinsic protein kinase activity) also examined. Synaptosome membrane fragments have little or no histone or phosvitin kinase activity but contain the highest concentration of cyclic AMP-stimulated intrinsic protein kinase activity. Homogenisation of the membrane fragments in Triton X-100 increased the histone kinase activity but on centrifugation it was all recovered in the supernatant, while the insoluble material contained all the intrinsic protein kinase activity. These results indicate that the intrinsic protein kinase activity of cerebral membrane fragments is due to the presence of a kinase enzyme which is specific to certain membrane proteins. The intrinsic protein kinase activity of synaptosome membrane fragments is a rather slow reaction which takes several minutes to saturate all the acceptor proteins.  相似文献
