The response of the planktonic, sediment, and epilithic bacterial communities to increasing concentrations of heavy metals was determined in a polluted river. None of the communities demonstrated a pollution-related effect on bacterial numbers (viable and total), heterotrophic activity, resistance to Pb or Cu, or species diversity as determined by either the Shannon-Wiener diversity index or rarefaction. The lack of correlation between concentrations of heavy metals and resistance in the sediment bacterial community was investigated and found to be due at least in part to the high pH of the river water and the resultant reduction in heavy metal toxicity. The three different communities demonstrated characteristic profiles based on the relative abundances of bacterial strains grouped according to functional similarities.  相似文献

Measurements of microbial biomass, bacterial numbers, and microbial production were determined for three small woodland streams located in the Adirondack Mountain region of New York State, USA. These streams exhibited spatial and temporal gradients in water pH ranging from a high of 7.0 to a low of 4.5. Twelve sites along these streams were used for comparative analyses of the effects of pH and related water chemistry parameters on the planktonic, sedimentary, and epilithic bacterial communities. The planktonic bacterial communities were not influenced by water pH or related water chemistry parameters. For sedimentary populations, the organic content of the sediment was more important than the chemistry of the overlying water. The epilithic bacterial communities, however, were influenced significantly by the pH of the water column, showing decreased bacterial production at lower pH.  相似文献

A microcosm containing resuspended river sediment was used to investigate the effect of anionic surfactants on the distribution of bacteria between planktonic and attached populations. Freshwater river sediment containing viable bacteria was preequilibrated in the microcosm, which was subsequently supplemented with biodegradable or recalcitrant surfactants and a non-surface-active carbon and energy source. Population dynamics of both free-living and attached bacteria were measured by epifluorescence microscopy with simultaneous analysis of the residual solution concentration of the xenobiotic carbon source. The addition of the readily biodegradable anionic surfactants sodium decyl sulfate and sodium dodecyl sulfate in separate experiments caused an increase in the number of attached bacteria and a concomitant decrease in the number of free-living bacteria. As biodegradation of the surfactants progressed, these trends reversed and the bacterial populations had returned to their preaddition values by the time when biodegradation was completed. In contrast, sodium tetradecyl sulfate or sodium dodecane sulfonate did not stimulate bacterial association with sediment, nor were they biodegraded in the microcosm. Sodium pyruvate, a non-surface-active carbon and energy source, was readily utilized but caused no bacterial attachment to the sediment. These results indicate that for an anionic surfactant to induce bacterial attachment to river sediment, it must be biodegradable. The bacterial attachment to the sediment appears to be reversible and may be dependent on the accumulation of the surfactant at the surface or as a result of alteration of the surface free energies.  相似文献

Epilithic bacteria were isolated nonselectively from riverbed stones and examined by gel zymography for their ability to produce alkylsulfatase (AS) enzymes and thus to metabolize alkyl sulfate surfactants such as sodium dodecyl sulfate. The percentages of AS+ isolates from stone epilithon at five sites from the source to the river mouth were measured on five sampling days spread over 1 year. The results showed that (i) the prevalence of epilithic AS+ strains (as a percentage of all isolates) was much higher at polluted sites than at the source; (ii) when averaged over the whole river, percentages of AS+ strains were significantly higher at the end of summer compared with either the preceding or the following winter; (iii) analysis of site-sampling time interactions indicated that water quality factors (e.g., biochemical oxygen demand and dissolved oxygen concentration) rather than climatic factors determined the distributions of epilithic AS+ isolates; (iv) constitutive strains were the most prevalent (7.2% of all isolates), with smaller numbers of isolates with inducible (4.5%) and repressible (1.7%) enzymes.  相似文献

Riverine biofilms remain one of the least-studied habitats despite the significant increase in the examination of aquatic microbial communities in recent years. In this study, the dynamics of epilithic biofilm communities native on rocks from a low-order upland stream were examined over a period of 3 years. Spatial and temporal variations in bacterial communities were assessed using terminal restriction fragment length polymorphism, based on analysis of the 16S rRNA gene. In total, 108 epilithic biofilm samples were analysed and 170 different ribotypes were detected. A strong temporal gradient in ribotype composition was noticed, especially between samples collected in 2001 and those collected in 2002 and 2003, most likely reflecting interannual differences in weather conditions, such as temperature. A spatial gradient in ribotype composition, from upstream sites to the low-lying sites, was also evident and interpreted as an environmental variation gradient along the river course. Distinct biofilm communities consistently occurred at the first site along the river, which was significantly correlated to low pH. Temporal factors explained the highest degree of variation within the epilithic biofilms. Recurrent blooms of certain bacteria were noted within the system. Phylogenetic relationships of bacteria at one point in the river were determined using a cloning and sequencing approach, with Alphaproteobacteria dominating the community, followed by Cyanobacteria, Bacteroidetes and Betaproteobacteria.  相似文献

The biodegradability potentials of three detergent products with the trade names Omo, Teepol and sodium dodecyl sulfate (SDS) by the native bacteria of the Lagos lagoon was carried out using the lagoon die-away method. Physicochemical parameters of the water samples showed that the lagoon in Apapa was more polluted than at theUniversity of Lagos. In 12 days, approximately 30, 60 and 97% of Omo, Teepol and SDS respectively were degraded. SDS with an alkyl sulfate moiety as surfactant supported the highest growth of the detergent-utilizing organisms, indicating that the components of Omo and Teepol are more resistant to microbial attack. The detergent-utilizing bacteria identified were mainly Gram-negative and of the following genera:Vibrio, Klebsiella, Flavobacterium, Pseudomonas, Escherichia, Enterobacter, Proteus, Shigella andCitrobacter. Vibrio was the most frequently encountered organism whileProteus was the rarest. Results of this investigation had shown that detergents made in Nigeria may still contain components that are recalcitrant to biodegradation.  相似文献

The rate of development and degree of survival of Asellus aquaticus eggs outside the marsupium of ovigerous females are affected by water quality and temperature. Eggs were maintained in polluted river water and relatively clean canal water. Developmental rates increase with increased temperature, but survival decreases. Eggs from polluted site ovigerous females survive better in clean water than in polluted water. Eggs from clean site ovigerous females maintained in polluted water have significantly lower survival rates than eggs from the polluted site at all temperatures tested. The developmental rate of clean site eggs is increased significantly in polluted water at 10–25 °C, possibly as a response to the stress imposed upon them.It is suggested that the method outlined might form the basis of a useful bioassay technique for measuring water quality.  相似文献

Rhizodegradation of organic pollutants, such as polycyclic aromatic hydrocarbons, is based on the effect of root-produced compounds, known as exudates. These exudates constitute an important and constant carbon source that selects microbial populations in the plant rhizosphere, modifying global as well as specific microbial activities. We conducted an experiment in two-compartment devices to show the selection of bacterial communities by root exudates and phenanthrene as a function of distance to roots. Using direct DNA extraction, PCR amplification, and thermal gradient gel electrophoresis screening, bacterial population profiles were analyzed in parallel to bacterial counts and quantification of phenanthrene biodegradation in three layers (0 to 3, 3 to 6, and 6 to 9 mm from root mat) of unplanted-polluted (phenanthrene), planted-polluted, and planted-unpolluted treatments. Bacterial community differed as a function of the distance to roots, in both the presence and the absence of phenanthrene. In the planted and polluted treatment, biodegradation rates showed a strong gradient with higher values near the roots. In the nonplanted treatment, bacterial communities were comparable in the three layers and phenanthrene biodegradation was high. Surprisingly, no biodegradation was detected in the section of planted polluted treatment farthest from the roots, where the bacterial community structure was similar to those of the nonplanted treatment. We conclude that root exudates and phenanthrene induce modifications of bacterial communities in polluted environments and spatially modify the activity of degrading bacteria.  相似文献

The response of river water microbial communities to chemical compounds was monitored under laboratory conditions using aniline as a model. Bacteria were collected from unpolluted and polluted sites. Bacterial abundance (plate and total direct counting) and its relation to aniline biodegradation was examined. Colony hybridization with 16S rRNA oligonucleotide probes was used to study the changes in microbial community structure during biodegradation of aniline. The changes in bacterial abundance and community structure were related to biodegradation of aniline. Burkholderia–Pseudomonas (rRNA group III), an authentic Alcaligenes group became dominant despite the initial differences in the microbial communities, suggesting that these genera are the main aniline degraders in the aquatic environment.  相似文献

Microbial communities associated with Arctic fjord ice polluted with petroleum oils were investigated in this study. A winter field experiment was conducted in the Van Mijen Fjord (Svalbard) from February to June 2004, in which the ice was contaminated with a North Sea paraffinic oil. Holes were drilled in the ice and oil samples frozen into the ice at the start of the experiment. Samples, including cores of both oil-contaminated and clean ice, were collected from the field site 33, 74, and 112 days after oil application. The sampled cores were separated into three sections and processed for microbiological and chemical analyses. In the oil-contaminated cores, enumerations of total prokaryotic cells by fluorescence microscopy and colony-forming units (CFU) counts of heterotrophic prokaryotes both showed stimulation of microbial growth, while concentrations of oil-degrading prokaryotes remained at similar levels in contaminated and clean ice. Analysis of polymerase chain reaction (PCR)-amplified bacterial 16S rRNA gene fragments by denaturing gradient gel electrophoresis (DGGE) revealed that bacterial communities in oil-contaminated ice generated fewer bands than communities in clean ice, although banding patterns changed both in contaminated and clean ice during the experimental period. Microbial communities in unpolluted ice and in cores contaminated with the paraffinic oil were examined by cloning and sequence analysis. In the contaminated cores, the communities became predominated by Gammaproteobacteria related to the genera Colwellia, Marinomonas, and Glaciecola, while clean ice included more heterogeneous populations. Chemical analysis of the oil-contaminated ice cores with determinations of n-C17/Pristane and naphthalene/phenanthrene ratios indicated slow oil biodegradation in the ice, primarily in the deeper parts of the ice with low hydrocarbon concentrations.  相似文献

We isolated 400 aerobic heterotrophic bacteria from the sediment of unpolluted and polluted sites in a fast-flowing south Wales river. Isolates were subjected to taxonomic tests and screened for the presence of plasmid DNA by alkaline lysis and agarose gel techniques. There were no significant differences between sites in either the total percentage of isolates containing plasmids (unpolluted site, 9.4%; polluted site, 15%) or in the percentage of non-Pseudomonas-like isolates containing plasmids (unpolluted site, 15%; polluted site, 10%). There were significantly more Pseudomonas-like isolates with plasmids at the polluted site than at the unpolluted site (unpolluted site, 7%; polluted site, 18%). This presumably reflected a response of the nutritionally versatile Pseudomonas-like isolates to conditions at that site. The majority (86%) of the plasmids detected had molecular masses between 35 and 312 megadaltons. These plasmids were large enough to carry genes for conjugal transfer, suggesting the possibility of such transfer in this environment.  相似文献

A SCUBA diver, using a bar-clamp sampler, collected quantitative epilithic algae samples from artificial substrates (slate and acrylic plates) and river stones at two stations on the Susquehanna River bottom in 1974. At Falls, Pennsylvania, algal colonization on monthly plates (submerged for ca. 30 day periods) was most rapid between September and October. In winter, when algal density on cumulative plates (submerged 2–13 mo) and on stones present in the river peaked at 18,100 and 21,900 units/ mm2, respectively, colonization of clean monthly plates nearly ceased. Similar colonization patterns were observed in a portion of the river polluted by coal mine drainages (SSES), but expansion of the algae population at SSES was inhibited by iron in the effluents, and densities never exceeded 2,400 units/mm2 on stones or plates. Diatoms composed 95% of the overall standing crop at Falls and 65% at SSES.  相似文献

In a 29 month study of bacterial populations at three sites on the Welsh River Dee, the aerobic heterotrophic bacteria increased from an average basal level of about 1.2 times 104 colony-forming units (cfu)/ml near the source of the river, to 2 times 105 cfu/ml in the lower reaches. The ratio, total bacterial cell count: viable count, decreased from 70 in the upland reaches to 10 in the lower parts of the river. There was no apparent seasonal variation in bacterial numbers but on occasions the bacterial populations in both upland and lowland reaches of the river increased simultaneously. Fluctuations in bacterial numbers over a 50-fold range were observed in this study. Bacterial isolates from both upland and lowland sites were predominated by two groupings of bacteria, the Pseudomonas—Agrobacterium—Alcaligenes group and the Flavobacterium—Cytophaga—Flexibacter group. Results suggested that the latter group may have been part of the autochthonous population. Seasonal variation in heterotrophic potential ( V max) for acetate uptake was shown to occur over a 30-fold range in the lowland reaches of the River Dee. Peaks in activity at the lowland site occurred during the summer months, the range of V max values for acetate ranged from 0.2 to 30 μg/1/h. Fluctuations in V max values from the upland site were not seasonal but were instead linked to faecal pollution, V max values from this site range from 0.04 to 3 μg/1/h.  相似文献

2011年夏、秋季在上海市全境主要河流的83个断面进行大型底栖动物的采样, 共获取底栖动物20个分类单位(种), 其中软体动物、环节动物和节肢动物分别占50%、30% 和20%。选取常见的28个生物指数分别进行计算, 并采用非参数统计检验方法筛选出8个敏感生物指数。通过记分法对8种敏感生物指数统一量纲后, 获得数值范围为8-40的综合生物指数, 运用四分法划分了判别河道水环境质量的生物基准: 32-40, 较好(Good); 24-31, 一般(Fair); 16-23, 污染(Poor); 8-15, 严重污染(Very poor)。9个断面未发现活体生物, 定为超严重污染(Super poor)。依照生物基准比较上海市41个代表性断面近5年主要水质理化指标, 基本反映出水质理化指标的空间差异性, 该综合生物指数及判别基准也可适用于评价平原河网地区的河道水质状况。    相似文献

Pheromone trapping was used to monitor populations of the moth Helicoverpa armigera at five cotton‐based agro‐ecological sites – river, vegetable, orchard, forest and clean cultivation (areas under only cotton cultivation) – in the Bahawalpur district, Pakistan. Three locations at each site were chosen and three pheromone traps at each location were installed in cotton fields. Moth catches were recorded at 15–20 day intervals from 24 October 2004 to 19 December 2006. In 2004, the river sites showed the maximum trapped population of H. armigera (0.22/trap) followed by 0.165 per trap at the vegetable sites. Orchard, clean cultivation and forest sites had zero moth catches. In 2005, the river sites again showed the highest trapped population (0.57/trap), followed by clean cultivation (0.45/trap), vegetable (0.44/trap), orchard (0.40/trap) and forest (0.29/trap). The moths appeared during July to December and March to May. In 2006, sites showed non‐significant difference, with a population range of 0.47 to 0.97 moths per trap. On average, river sites peaked at 0.49 per trap, followed by vegetable (0.38), clean cultivation (0.47), orchard (0.35) and forest (0.25) sites. The peak was observed on 3 April 2006, and moths appeared during February to July and October to December. The minimum temperature in river, forest and clean cultivation sites; the maximum temperature in orchard sites; and the average temperature in river, orchard, forest and clean cultivation sites showed significant positive correlations with trapped moth populations. Relative humidity showed significant negative correlation with population at the orchard sites in 2005. All weather factors during 2004 and 2006 showed non‐significant correlations with the moth populations. No model was found to be best fit by multiple linear regression analysis; however, relative humidity, minimum temperature, maximum temperature, minimum temperature and maximum temperature contributed 8.40, 10.23, 2.43, 4.53 and 2.53% to the population fluctuation of the moth at river, vegetable, orchard, forest and clean cultivation sites, respectively.  相似文献

The biodegradation of South Louisiana (SL) crude oil and the effects of nitrogen, phosphorus, and iron supplements on this process were compared in a polluted (10,900 oil degraders per liter) and in a relatively clean (750 oil degraders per liter) littoral seawater sample taken along the New Jersey coast. Without supplements, the biodegradation of SL crude oil was negligible in both seawater samples. Addition of nitrogen and phosphorus allowed very rapid biodegradation (72% in 3 days) in polluted seawater. Total iron in this seawater sample was high (5.2 muM), and the addition of iron did not increase the biodegradation rate further. In the less polluted and less iron-rich (1.2 muM) seawater sample, biodegradation of SL crude oil was considerably slower (21% in 3 days) and the addition of chelated iron had a stimulating effect. Ferric octoate was shown to have a similar stimulating effect on SL crude oil biodegradation as chelated iron. Ferric octoate, in combination with paraffinized urea and octylphosphate, is suitable for treatment of floating oil slicks. We conclude that spills of SL crude and similar oils can be cleaned up rapidly and efficiently by stimulated biodegradation, provided the water temperatures are favorable.  相似文献

The potential for biodegradation of polycyclic aromatic hydrocarbons (PAHs) at low temperature and under anaerobic conditions is not well understood, but such biodegradation would be very useful for remediation of polluted sites. Biodegradation of a mixture of 11 different PAHs with two to five aromatic rings, each at a concentration of 10 μg/ml, was studied in enrichment cultures inoculated with samples of four northern soils. Under aerobic conditions, low temperature severely limited PAH biodegradation. After 90 days, aerobic cultures at 20°C removed 52 to 88% of the PAHs. The most extensive PAH degradation under aerobic conditions at 7°C, 53% removal, occurred in a culture from creosote-contaminated soil. Low temperature did not substantially limit PAH biodegradation under nitrate-reducing conditions. Under nitrate-reducing conditions, naphthalene, 2-methylnaphthalene, fluorene, and phenanthrene were degraded. The most extensive PAH degradation under nitrate-reducing conditions at 7°C, 39% removal, occurred in a culture from fuel-contaminated Arctic soil. In separate transfer cultures from the above Arctic soil, incubated anaerobically at 7°C, removal of 2-methylnaphthalene and fluorene was stoichiometrically coupled to nitrate removal. Ribosomal intergenic spacer analysis suggested that enrichment resulted in a few predominant bacterial populations, including members of the genera Acidovorax, Bordetella, Pseudomonas, Sphingomonas, and Variovorax. Predominant populations from different soils often included phylotypes with nearly identical partial 16S rRNA gene sequences (i.e., same genus) but never included phylotypes with identical ribosomal intergenic spacers (i.e., different species or subspecies). The composition of the enriched communities appeared to be more affected by presence of oxygen, than by temperature or source of the inoculum.  相似文献
