From the hot petrol extract of Piper cubeba ftuits, six lignans were isolated. Two of these, which have been obtained from a natural source for the first time, have been characterized as (2R,3R)-2-(3″,4″,5″-trimethoxybenzyl)-3-(3′,4′-methylenedioxybenzyl)-1,4-butanediol [(?)-dihydroclusin] and (3R,4R)-3,4-bis-(3,4,5-trimethoxybenzyl)tetra-hydro-2-furanol [(?)-cubebinin]. (?)Cubebin, (?)-hinokinin, (?)-clusin and (?)-dihydrocubebin were also found in this plant. Only (?)-cubebin has been reported so far from this source.  相似文献

A series of cis-restricted 1,4- and 1,5-disubstituted 1,2,3-triazole analogs of combretastatin A-4 (1) have been prepared. Cytotoxicity and tubulin inhibition studies showed that 2-methoxy-5-((5-(3,4,5-trimethoxyphenyl)-1H-1,2,3-triazol-1-yl)methyl)aniline (5e) and 2-methoxy-5-(1-(3,4,5-trimethoxybenzyl)-1H-1,2,3-triazol-5-yl)aniline (6e) were two of the most active compounds. Molecular modeling studies revealed that the N-2 and N-3 atoms in the triazole rings in 5e and 6e did not form hydrogen bonds with the amino acids in the anticipated pharmacophore.  相似文献

Of these, three compounds which have been isolated from a natural source for the first time were characterized as (2R,3R)-2-(5″-methoxy-3″,4″-methylenedi [(−)-cubebinone], (2R,3R)-2-(3″,4″-methylenedioxybenzyl)-3-(3′,4′,5′-trimethoxybenzyl)butyrolactone [(−)-isoyatein] and (2R,3R)-2-(3″,4″,5″-trimethoxybenzyl)-3-(3′,4′-dimethoxybenzyl)butyrolactone [(−)-di-O-methyl thujaplicatin methyl ether, i.e. (−)-thujaplicatin trimethyl ether]. The other three compounds were identified as (−)-yatein, (−)-cubebininolide and (2R,3R)-2-(3″,4″-methylenedioxybenzyl)-3-(3′,4′-dimethoxybenzyl) butyrolactone.  相似文献

A novel series of 2-substituted mercapto-3-(3,4,5-trimethoxybenzyl)-4(3H)-quinazolinones 120 was synthesised and evaluated for in vitro antitumour activity. N-(4-Chlorophenyl)-2-[(3-(3,4,5-trimethoxybenzyl)-4(3H)-quinazolinon-2-yl)thio)acetamide (7) and N-(3,4,5 trimethoxybenzyl)-2-[(3-(3,4,5-trimethoxybenzyl)-4(3H)-quinazolinon-2-yl)thio]propanamide (19) exhibited excellent antitumour properties, with mean growth inhibitory concentration (GI50) of 17.90 and 6.33 µΜ, respectively, compared with those of 5-fluorouracil 5-FU, gefitinib, and erlotinib (mean GI50: 18.60, 3.24, and 7.29 µΜ, respectively). Comparison of the GI50 (µM) values of compounds 7 and 19 versus those of 5-FU, gefitinib, and erlotinib against an in vitro subpanel of tumour cells lines showed that compounds 7 and 19 have activities almost equal to or higher than that of those standard drugs, especially against lung, CNS, and breast cancer cells. However, compounds 5, 10, 14, 15, 16, 17, and 20 exhibited effective antitumour activity against the different cell lines tested, with growth inhibition percentage (MGI%) of 19, 24, 19, 17, 16, 15, and 16, respectively. A modelling study was performed for compounds 7 and 19 by docking them into the EGFR kinase enzyme to study their mode of binding with the putative binding site.  相似文献

The aim of this study was to evaluate the cytotoxicity of a series of seven 4′-methylthio-trans-stilbene derivatives against cancer cells: MCF7 and A431 in comparison with non-tumorigenic MCF12A and HaCaT cells. The mechanism of anti-proliferative activity of the most cytotoxic trans-resveratrol analogs: 3,4,5-trimethoxy-4′-methylthio-trans-stilbene (3,4,5-MTS) and 2,4,5-trimethoxy-4′-methylthio-trans-stilbene (2,4,5-MTS) was analyzed and compared with the effect of trans-resveratrol. All the compounds that were studied exerted a stronger cytotoxic effect than trans-resveratrol did. MCF7 cells were the most sensitive to the cytotoxic effect of trans-resveratrol analogs with IC50 in the range of 2.1–6.0 µM. Comparing the cytotoxicity of 3,4,5-MTS and 2,4,5-MTS, a significantly higher cytotoxic activity of these compounds against MCF7 versus MCF12A was observed, whereas no significant difference was observed in cytotoxicity against A431 and HaCaT. In the series of 4′-methylthio-trans-stilbenes, 3,4,5-MTS and 2,4,5-MTS were the most promising compounds for further mechanistic studies. The proapoptotic activity of 3,4,5-MTS and 2,4,5-MTS, estimated with the use of annexin-V/propidium iodide assay, was comparable to that of trans-resveratrol. An analysis of cell cycle distribution showed a significant increase in the percentage of apoptotic cells and G2/M phase arrest in MCF7 and A431 as a result of treatment with 3,4,5-MTS, whereas trans-resveratrol tended to increase the percentage of cells in S phase, particularly in epithelial breast cells MCF12A and MCF7. Both trans-stilbene derivatives enhanced potently tubulin polymerization in a dose-dependent manner with sulfur atom participating in the interactions with critical residues of the paclitaxel binding site of β-tubulin.  相似文献

The trunkwood of Machaerium kuhlmannii contains methyl palmitate, 3-O-acetyloleanolic acid and sitosterol; the benzene derivatives 2,3-dimethoxyphenol, 2,6-dimethoxyphenol, 2-hydroxy-3-methoxyphenol, 2,3-dimethoxybenzaldehyde and methyl 3-(2-hydroxy-4-methoxyphenyl)-propionate; the isoflavonoids formononetin and (6aS,11aS)-medicarpin; the neoflavonoids (R)-3,4-dimethoxydalbergione, (R)-3,4-dimethoxydalbergiquinol, kuhlmanniquinol [(R)-3-(4-hydroxyphenyl)-3-(5-hydroxy-2,3,4-trimethoxyphenyl)-propene], dalbergin, kuhlmannin (6-hydroxy-7,8-dimethoxy-4-phenylcoumarin) and kuhlmannene (6-hydroxy-7,8-dimethoxy-4-phenylchrom-3-ene), as well as the cinnamylphenol kuhlmannistyrene [Z-1-(5-hydroxy-2,3,4-trimethoxybenzyl)-2-(2-hydroxyphenyl)-ethylene]. Five of these compounds, in addition to (R)-4′-hydroxy-3,4-dimethoxydalbergione, were also isolated from a trunkwood extract of M. nictitans. Structural assignments were confirmed by chemical interconversion and by the synthesis of (±)-kuhlmanniquinol.  相似文献

Two new compounds, named as (Z)-7,4′-dimethoxy-6-hydroxy-aurone-4-O-β-glucopyranoside (1), and (1S,3R,4S)-1-(4′-hydroxyl-phenyl)-3,4-dihydro-3,4,5-trimethyl-1H-2-benzopyran-6,8-diol (2), were isolated from the endophytic fungus Penicillium citrinum of Bruguiera gymnorrhiza. Their structures were elucidated on the basis of spectroscopic analysis. Additionally, compound 1 exhibited potent neuroprotective activity in 1-methyl-4-phenylpyridinium-induced oxidative damage in PC12 cells.  相似文献

1. (+)-Mollisacacidin [(+)-3′,4′,7-trihydroxy-2,3-trans-flavan-3,4-trans- diol] is converted by autoclaving into the optically active free phenolic 2,3-trans-3-4-cis (12% yield), 2,3-cis-3,4-trans (11%) and 2,3-cis-3,4-cis (2·8%) diastereoisomers through epimerization at C-2 and C-4. 2. The relative configurations of the epimeric forms were determined by nuclear-magnetic-resonance spectrometry and paper ionophoresis in comparison with synthetic reference compounds, and was confirmed by chemical interconversions. 3. From this a scheme of epimerization is inferred and their absolute configurations are assigned as (2R:3S:4S), (2S:3S:4R) and (2S:3S:4S) respectively from the known absolute configuration (2R:3S:4R) of (+)-mollisacacidin.  相似文献

Two sulfur-containing compounds, (S)-2-amino-5-((R)-1-carboxy-2-((E)-3-(4-hydroxy-3-methoxyphenyl)allylthio)ethyl-amino)-5-oxopentanoic acid (1) and (S)-2-amino-5-((R)-1-(carboxymethylamino)-3-((E)-3-(4-hydroxyphenyl)allylthio)-1-oxopropan-2-ylamino)-5-oxopentanoic acid (2), and one 1H-pyrrole-2-carboxylic acid derivative, 6-(3-(1H-pyrrole-2-carbonyloxy)-2-hydroxypropoxy)-3,4,5-trihydroxy-tetrahydro-2H-pyran-2-carboxylic acid (3), together with eighteen known phenolic compounds, were isolated from the fruits of pineapple. Their structures were elucidated by a combination of spectroscopic analyses. Some of these compounds showed inhibitory activities against tyrosinase. The half maximal inhibitory concentration values of compounds 1, 4, 5, 6, 7 are lower than 1 mM. These compounds may contribute to the well-known anti-browning effect of pineapple juice and be potential skin whitening agents in cosmetic applications.  相似文献

Further investigations on the petrol extract of Piper clusii have afforded four more new lignans.These are 2S,3R,4R,2-ethoxy-3-(3,4,5-trimethoxyphenyl)methyl 4-(1,3-benzodioxol-5-yl) methyl tetrahydrofuranol; 3R,4R,bis-3,4-(3,4,5-trimethoxyphenyl) methyl tetrahydrofuran-2-one; 2R,3R,2-(7-methoxy-1,3-benzodioxol-5-yl) methyl 3-(3,4,5-trimethoxyphenyl)methyl butan-1,4-diol and 2R,3R,2-(1,3-benzodioxol-5-yl)methyl 3-(3,4,5-trimethoxyphenyl) methyl butan-1,4-diol. Two new flavonol tetraglycosides, quercetin 3-O-(4-O-trans-p-coumaroyl)-α-l-rhamnopyranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (krathummuoside A) and quercetin 3-O-(4-O-cis-p-coumaroyl)-α-l-rhamnopyranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (krathummuoside B) were isolated from the leaves of Mitragyna rotundifolia in addition to eight known compounds, quercetin 3-O-α-l-rhamnopuranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside, rutin, (−)-epi-catechin, 3,4,5-trimethoxyphenyl β-d-glucopyranoside, (6S, 9R)-roseoside, 3-O-β-d-glucopyranosyl quinovic acid 28-O-β-d-glucopyranosyl ester, (+)-lyoniresinol 3α-O-β-d-glucopyranoside, and (+)-syringaresinol-4-O-β-d-glucopyranoside. The structure elucidation of these compounds was based on analyses of spectroscopic data including 1D- and 2D-NMR.  相似文献

The aim of this study was to synthesize and evaluate plasmid DNA interaction of new platinum(II) complexes with some 2-substituted benzimidazole derivatives as carrier ligands which may have potent anticancer activity and low toxicity. Twelve benzimidazole derivatives carrying indole, 2-/or 3-/or 4-methoxyphenyl, 4-methylphenyl, 3,4-dimethoxyphenyl, 3,4,5-trimethoxyphenyl, 4-methoxybenzyl, 3,4,5-trimethoxybenzyl, 3,4,5-trimethoxystyryl, 3,4,5-trimethoxybenzylthio or dimethylamino ethyl groups in their position 2 and twelve platinum(II) complexes with these carrier ligands were synthesized. The chemical structure of the platinum complexes have been characterized by their elemental analysis and FIR, 1H NMR and mass spectra and their 1H NMR and FIR spectra were interpreted by comparison with those of the ligands. The interaction of all the ligands and their complexes with plasmid DNA and their restriction endonuclease reactions by BamHI and HindIII enzymes were studied by agarose gel electrophoresis. It was determined that complex 1 [dichloro-di(2-(1H-indole-3-yl)benzimidazole)platinum(II)·2H2O] has stronger interaction than carboplatin and complex 10 [dichloro-di(2-(3,4,5-trimethoxystyryl)benzimidazole)platinum(II)·2H2O] has stronger interaction than both carboplatin and cisplatin with plasmid DNA.  相似文献

Two isomers of megastigmane glycosides, (6R, 9S)-blumenol C 9-O-gentibioside (2) and (6S, 9S)-blumenol C 9-O-gentiobioside (3), and a new 7,9′-dinorlignan glycoside, stepdonorlignoside (4) were isolated from the tubers of Stephania kaweesakii. The structure determinations were considered based on the physical data and spectroscopic evidence. The absolute configurations of two megastigmanes were determined for the first time. Additionally, ten known compounds were isolated: (6R, 9S)-blumenol C 9-O-β-D-glucopyranoside, (+)-isolariciresinol 3a-O-β-D-glucopyranoside, salidroside, N-trans-caffeoyltyramine, (R)-isococlaurine, (R)-isococlaurine 4′-O-β-glucopyranoside, (−)-oblongine, (+)-magnocurarine, fordianoside, and (−)-cyclanoline.  相似文献

The prooxidant effect of resveratrol (3,5,4′-trihydroxy-trans-stibene) and its synthetic analogues (ArOH), that is, 3,4,4′-trihydroxy-trans-stibene (3,4,4′-THS), 3,4,5-trihydroxy-trans-stibene (3,4,5-THS), 3,4-dihydroxy-trans-stibene (3,4-DHS), 4,4′-dihydroxy-trans-stibene (4,4′-DHS), 2,4-dihydroxy-trans-stilbene (2,4-DHS), 3,5-dihydroxy-trans-stilbene (3,5-DHS) and 3,5,4′-trimethoxy-trans-stibene (3,5,4′-TMS), on supercoiled pBR322 plasmid DNA strand breakage and calf thymus DNA damage in the presence of Cu (II) ions has been studied. It was found that the compounds bearing ortho-dihydroxyl groups (3,4-DHS, 3,4,4′-THS, and 3,4,5-THS) or bearing 4-hydroxyl groups (2,4-DHS, 4,4′-DHS, and resveratrol) exhibit remarkably higher activity in the DNA damage than the ones bearing no such functionalities. Kinetic analysis by UV-visible spectra demonstrates that the formation of ArOH-Cu (II) complexes, the stabilization of oxidative intermediate derived from ArOH and Cu (II)/Cu (I) redox cycles, might be responsible for the DNA damage. This study also reveals a good correlation between antioxidant and prooxidant activity, as well as cytotoxicity against human leukemia (HL-60 and Jurkat) cell lines. The mechanisms and implications of these observations are discussed.  相似文献

Conformational modulation of the aryl portion of a set of N,N-bis(cyclohexanol)amine aryl esters (1a-d) that are potent Pgp-dependent MDR inhibitors has been performed. Toward this end the trans-3-(3,4,5-trimethoxyphenyl)acrylic acid present in set 1 was substituted with 3-(3,4,5-trimethoxyphenyl)propanoic and 3-(3,4,5-trimethoxyphenyl)propiolic moieties to give sets 2 and 3, respectively. While the introduction of 3-(3,4,5-trimethoxyphenyl)propanoic moiety resulted in a definite drop in potency and efficacy, esterification with 3-(3,4,5-trimethoxyphenyl)propiolic acid gave four isomers (3a-d) that maintain high potency and possess optimal efficacy. These results are discussed in terms of conformational flexibility of the different sets of compounds.  相似文献

New chiral purinyl and 8-azapurinyl carbanucleoside derivatives based on indanol were synthesized from commercial available (1S,2S)-trans-1-amino-2-indanol and (1R,2R)-trans-1-amino-2-indanol using a linear methodology. The antiviral activity and cytotoxicity of these compounds were evaluated against herpes simplex virus type 1 (HSV-1) in Vero cells, bovine viral diarrhea virus (BVDV) in Mardin-Darby bovine kidney (MDBK) cells and hepatitis B virus (HBV) in HepG2 2.2.15 cell line. Three compounds, showed an inhibition of the HBsAg levels similar to reference drug lamivudine. One chloropurinyl nucleoside, derived from the cis-1-amino-2-indanol, was cytotoxic on MDBK cells and it could be a lead for developing anticancer agents.  相似文献

The trunk wood of an Amazonian Aniba species contains three novel neolignans: (2R, 3R, 3aS, 5R)-3a-allyl-5-methoxy-2-(3,4,5-trimethoxyphenyl)-3-methyl-2,3,3a,4,5,6-hexahydro-6-oxobenzofuran (canellin-D), (2R,3R,3aS,5R)-3a-allyl-5,7-dimethoxy-2-(3-methoxy-4,5-methylenedioxyphenyl)-3-methyl-2,3,3a,4,5,6-hexahydro-6-oxobenzofuran (canellin-E) and (2S,3S,3aS,5R)-3a-allyl-5-methoxy-2-(3-methoxy-4,5-methylenedioxyphenyl)-3-methyl-2,3,3a,4,5,6-hexahydro-6-oxobenzofuran (armenin-C). The absolute stereochemistries of these and of all other known hexahydro-6-oxobenzofurans were determined by CD comparisons with model compounds.  相似文献

The chirality of eschscholtzxanthin (all-trans (3S,3′S)-4′,5′-didehydro-4,5′-retro-β,βcarotene-3,3′-diol) at 3,3′ was assigned from the CD correlation of the natural material and the semi-synthetic carotenoid prepared by (NBS-dehydrogenation of natural zeaxanthin ((3R,3′R)-β,β-carotene-3,3′-diol). The δ6(6′)-trans configuration followed from 1H NMR evidence, including nuclear Overhauser experiments with rhodoxanthin, retrodehydro-carotene (4′,5′-didehydro-4,5′-retro-β,β-carotene) and smaller retro model compounds revealing a general preference for the δ6-trans configuration in retro compounds.
