Two new species of freshwater ascomycetes belonging to the genus Lindgomyces (Pleosporales, Dothideomycetes) are described and illustrated from submerged wood in North Carolina, USA. Lindgomyces carolinensis is characterized by immersed to erumpent ascomata, fissitunicate broadly cylindrical to clavate asci, and fusiform ascospores with acute ends surrounded by a large, fusiform gelatinous sheath. Lindgomyces cigarospora morphologically differs from L. carolinensis in that its ascospores are fusiform to cylindrical with rounded ends, without a large fusiform gelatinous sheath. These two new species nest in the family Lindgomycetaceae based on analyses of combined SSU and LSU rDNA sequence data. Phylogenetic analyses using ITS sequence data support the establishment of the new taxa as separate species within Lindgomyces. In addition to the new species, we report new ITS sequence data for L. cinctosporus and L. griseosporus from France, and L. ingoldianus from North Carolina, USA. We report a video exhibiting fissitunicate ascus dehiscence in L. carolinensis showing ascospore discharge and unraveling of the gelatinous sheath in real time. Chemical analysis of the organic extracts of L. carolinensis and L. cigarospora resulted in two known cyclodepsipeptides, Sch 378161 and Sch 217048. The in situ spatial mapping of these secondary metabolites on fungal cultures indicates the presence of both compounds on the surface of mycelia, as well as being exuded into the surrounding agar.  相似文献

在大湄公河次区域的水生真菌调查中,从中国云南和西藏的沉水腐木中分离得到4个菌株。基于LSU、SSU、ITS、TEF1-α和RPB2序列进行多基因系统发育分析,表明4个菌株属于菩提科香港霉属真菌。系统发育分析结果显示4个菌株聚集在一起,并与泰国香港霉形成姐妹支。基于形态学及分子系统学研究,将这4个菌株鉴定为新种棕孢香港霉。棕孢香港霉是香港霉属的第二个有性型物种,它因子囊果的孔口处有棕色至黑色的刚毛,且子囊孢子呈梭形,孢子两端逐渐变窄且钝圆,红棕色至暗棕色,具有多个隔膜而区别于另一个有性型物种泰国香港霉。本研究提供了该真菌新种的描述及图版并比较了该种与其他物种的形态差异。  相似文献

本研究对中国滇西北地区淡水木腐真菌进行了调查,在云南省的苍山、怒江和澜沧江采集到3个菌株。根据形态学特征和多基因系统发育分析(ITS、LSU、SSU、TEF1-α和RPB2)描述了新种——苍山长喙孢Rostriconidium cangshanense。苍山长喙孢的特征是菌体形态与营养菌丝不同,分生孢子梗单生或者松散成束,光滑或粗糙;产孢细胞多芽、深褐色、具有黑色驼峰状的分生孢子脱落痕;分生孢子梭形至倒棒状、长喙状,6-8个横隔,有明显油滴状细胞内含物,有时具顶端粘膜鞘。露兜长喙孢Rostriconidium pandanicola报道为世界淡水环境的新记录。  相似文献

In this paper, we describe and illustrate a new species, Lindgomyces angustiascus, from submerged wood in lotic and lentic habitats from Florida, North Carolina and Wisconsin, USA. The new species is characterized by black, partially immersed, flattened, globose ascomata; fissitunicate, long, slender, obclavate asci; and one-septate, hyaline, fusiform ascospores with bipolar appendages, each covered with a gelatinous cap. Maximum Likelihood and Bayesian analyses of partial 18S nrDNA and 28S nrDNA, as well as the entire nuclear ribosomal internal transcribed spacer (ITS1, 5.8S and ITS2) region support the placement and establishment of this new species in the Lindgomycetaceae, Pleosporales, Dothideomycetes. Chemical analysis of the organic extract of L. angustiascus revealed the presence of 6E,9E-octadecadienoic acid and ergosterol peroxide as major secondary metabolites.  相似文献

The taxonomic placement of freshwater and marine Savoryella species has been widely debated, and the genus has been tentatively assigned to various orders in the Sordariomycetes. The genus is characterized as possessing paraphyses that deliquesce early, elongate, clavate to cylindrical asci with a poorly developed apical ring and versicolored, three-septate ascospores. We performed two combined phylogenetic analyses of different genes: (i) partial small subunit rRNA (SSU), large subunit rRNA (LSU), DNA-dependent RNA polymerase II largest subunit (rpb2) dataset and (ii) SSU rDNA, LSU rDNA, DNA-dependent RNA polymerase II largest subunit (rpb1 and rpb2), translation elongation factor 1-alpha (tef1), the 5.8S ribosomal DNA (5.8S rDNA) dataset. Our results indicate that Savoryella species formed a monophyletic group within the Sordariomycetes but showed no affinity to the Hypocreales, Halosphaeriales (now Microascales), Sordariales and Xylariales, despite earlier assignments to these orders. Savoryella, Ascotaiwania and Ascothailandia (and its anamorph, Canalisporium) formed a new lineage that has invaded both marine and freshwater habitats, indicating that these genera share a common ancestor and are closely related. Because they show no clear relationship with any named order we erect a new order Savoryellales in the subclass Hypocreomycetidae, Sordariomycetes. The genera Savoryella and Ascothailandia are monophyletic, while the position of Ascotaiwania is unresolved. All three genera are phylogenetically related and form a distinct clade similar to the unclassified group of marine ascomycetes comprising the genera Swampomyces, Torpedospora and Juncigera (TBM clade: Torpedospora/Bertia/Melanospora) in the Hypocreomycetidae incertae sedis.  相似文献

A high diversity of pleurostomatid ciliates has been discovered in the last decade, and their systematics needs to be improved in the light of new findings concerning their morphology and molecular phylogeny. In this work, a new genus, Protolitonotus gen. n., and two new species, Protolitonotus magnus sp. n. and Protolitonotus longus sp. n., were studied. Furthermore, 19 novel nucleotide sequences of SSU rDNA, LSU rDNA and ITS1‐5.8S‐ITS2 were collected to determine the phylogenetic relationships and systematic positions of the pleurostomatid ciliates in this study. Based on both molecular and morphological data, the results demonstrated that: (i) as disclosed by the sequence analysis of SSU rDNA, LSU rDNA and ITS1‐5.8S‐ITS2, Protolitonotus gen. n. is sister to all other pleurostomatids and thus represents an independent lineage and a separate family, Protolitonotidae fam. n., which is defined by the presence of a semi‐suture formed by the right somatic kineties near the dorsal margin of the body; (ii) the families Litonotidae and Kentrophyllidae are both monophyletic based on both SSU rDNA and LSU rDNA sequences, whereas Amphileptidae are non‐monophyletic in trees inferred from SSU rDNA sequences; and (iii) the genera Loxophyllum and Kentrophyllum are both monophyletic, whereas Litonotus is non‐monophyletic based on SSU rDNA analyses. ITS1‐5.8S‐ITS2 sequence data were used for the phylogenetic analyses of pleurostomatids for the first time; however, species relationships were less well resolved than in the SSU rDNA and LSU rDNA trees. In addition, a major revision to the classification of the order Pleurostomatida is suggested and a key to its families and genera is provided.  相似文献

Conlarium duplumascospora gen. et. sp. nov. and Jobellisia guangdongensis sp. nov. are described and illustrated from submerged wood collected from Guangdong Province, China. Conlarium duplumascospora is characterized by gregarious, coriaceous and beaked ascomata; cylindrical, unitunicate asci with a bipartite apical ring; biseriate, fusiform, hyaline, 0-5-septate ascospores with or without appendages; and anamorph with muriform conidia. Jobellisia guangdongensis is characterized by globose to subglobose, gregarious and papillate ascomata; three-layered peridium; cylindrical, unitunicate asci with a refractive apical ring; and one-septate, fusiform, greenish brown ascospores. Sequence analyses of partial nuclear large subunit ribosomal DNA (LSU rDNA) were performed to infer the phylogenetic affinities of these new taxa. The teleomorph of Aquaphila albicans was discovered on submerged wood collected in Thailand. Its black, soft-textured, setose ascomata, bitunicate asci and hyaline to pale brown, multiseptate ascospores indicated an affinity to Tubeufiaceae (Dothideomycetes). After morphological or molecular comparisons with related species in Tubeufia, Acanthostigma and Taphrophila, it is described and illustrated as a new species, T. asiana Sivichai & K.M. Tsui, sp. nov. Finding this Tubeufia teleomorph was surprising, given the falcate conidia of its A. albicans anamorph, which superficially resemble the conidia of Fusarium and not the coiled, helicosporous conidia of other species in Tubeufiaceae. We assessed the phylogenetic relationships of A. albicans-T. asiana with ribosomal sequences from SSU and ITS and partial LSU regions by parsimony and Bayesian analysis. An initial set of 40 taxa representing a wide range of ascomycete families and their SSU sequences from GenBank showed A. albicans-T. asiana to be nested within the Tubeufiaceae with 100% bootstrap support. Their placement was inferred with ITS and partial LSU ribosomal sequences. The nearly identical ITS sequences of two isolates of A. albicans and one isolate of Tubeufia asiana united these fungi as a monophyletic group with 100% bootstrap support and further nested them, with 88% bootstrap support, in a clade containing Helicoon gigantisporum and Helicoma chlamydosporum. This is the first molecular phylogenetic study to place a nonhelicosporous species within the Tubeufiaceae and to show that helical conidia were lost at least once within the family.  相似文献

During a latitudinal survey of freshwater ascomycetes, an unidentified fungus with bitunicate asci was found on submerged wood and herbaceous material from Florida and Costa Rica. Based on morphological characteristics and 28S rDNA large subunit (LSU) sequence data, this fungus is described as a new genus and species, Wicklowia aquatica, and placed in the Pleosporales (Pleosporomycetidae, Dothideomycetes). Phylogenetic analyses based on LSU sequences did not resolve the familial placement of W. aquatica within the Pleosporales. The characteristic features of W. aquatica are subglobose, dorsiventrally flattened, ostiolate, immersed to erumpent, black ascomata; a peridial wall composed of 4–5 layers of darkened pseudoparenchymatic cells; cellular pseudoparaphyses immersed in a gel matrix; broadly clavate, bitunicate asci; and cylindrical, hyaline, one-septate ascospores with rounded apices and surrounded by a gelatinous sheath that expands in water; ascospore sheath attached at the ascospore base with a gelatinous curtain extending from the base that fragments into basal filamentous appendages which radiate from the base of the ascospore.  相似文献

Three new species of loculoascomycetes collected from freshwater habitats in North America are described as new species of Jahnula (Jahnulales, Dothideomycetes). All three share these morphological features: hyaline to blackish translucent, membranous ascomata with subtending, wide, septate brown, spreading hyphae; peridia composed of large angular cells; hamathecium of septate pseudoparaphyses; 8-spored, clavate to cylindrical asci; and 1-septate, broadly fusiform, brown, multiguttulate ascospores. Two new genera are established in the Dothideomycetes based on morphological and molecular data (SSU and LSU nuclear ribosomal sequences) to accommodate four ascomycete species collected from woody debris submerged in freshwater habitats. The genus Minutisphaera is represented by a single species, M. fimbriatispora, which was collected from freshwater habitats in temperate forests in North America. It has small, superficial, brown, subglobose, papillate pseudothecia with dark, irregularly twisted hairs around the papillae, fissitunicate asci, septate pseudoparaphyses, and hyaline, one-septate ascospores surrounded by a gelatinous sheath and having spine-like appendages radiating around the ascospore at the septum. Minutisphaera formed a strongly supported clade with Farlowiella carmichaeliana. The second genus, Natipusilla, contains three new species, N. decorospora, N. limonensis and N. naponensis, which were collected from Central and South America. Natipusilla is characterized by small, superficial, light-colored, globose pseudothecia, fissitunicate asci, few or no pseudoparaphyses, and hyaline, one-septate to tardily two- or three-septate ascospores with or without a gelatinous sheath. The three Natipusilla species form a well supported clade, but their relationship to other members of the Dothideomycetes remains unclear.  相似文献

Three new genera are established in the Sordariomycetidae based on morphological and molecular data (SSU and LSU nrDNA) to accommodate five ascomycete species collected from submerged woody debris in freshwater habitats from Costa Rica. The genus Bullimyces contains three new species, B. communis, B. costaricensis and B. aurisporus. Bullimyces is characterized by globose to subglobose, membranous, black, ostiolate ascomata; deliquescent, hyaline, globose cells that fill the center of the centrum; unitunicate asci that deliquesce early in some species; and septate, thick-walled ascospores with or without gelatinous sheaths or appendages. Bullimyces species form a well supported clade with 100% bootstrap support, but the position of the genus in the Sordariomycetidae remains unclear. The second genus, Riomyces, is represented by a single species, R. rotundus. Riomyces is characterized by globose to subglobose, membranous, black, ostiolate ascomata, unitunicate, cylindrical asci, hyaline, globose cells that fill the hamathecium and septate, thick-walled ascospores with a gelatinous sheath. Although Riomyces is morphologically similar to Bullimyces, the two genera did not group together with support in any analysis. The third genus, Hydromelitis, is represented by a single species, H. pulchella. Hydromelitis is characterized by pyriform, membranous, black, ostiolate ascomata, unitunicate asci lacking an apical structure, simple, thin-walled, septate paraphyses and hyaline to golden yellow, multiseptate, thick-walled ascospores with a gelatinous sheath. Bullimyces, Riomyces and Hydromelitis were nested within an unsupported clade consisting of members of the Ophiostomatales, Magnaporthales and freshwater Annulatacaceae sensu lato and sensu stricto.  相似文献

An undescribed ascomycete similar to species in the Aliquandostipitaceae (Jahnulales, Dothideomycetes) was collected from submerged wood in a freshwater swamp in Big Cypress National Preserve, Florida. The characteristic features of the new species are as follows: (i) ascomata are small, sessile, light brown, globose to subglobose, papillate, and anchored to the substrate by wide, brown, septate and subtending hyphae; peridial wall is composed of 1 to 2 layers of large, angular cells with large lumens; (ii) asci are ovoid to broadly clavate, and fissitunicate; (iii) ascospores are one-septate, fusiform, multiguttulate, pale brown, surrounded by a fusiform gelatinous sheath, and equipped with numerous filamentous appendages around the midseptum. The new fungus is most similar to Aliquandostipite crystallinus, from which it differs in overall smaller size and morphology. This new fungus is described and illustrated herein as A. minuta.  相似文献

Fungi play a vital role as decomposers in mangrove ecosystems. A new ascomycete species, Acuminatispora palmarum, inhabiting decayed petioles and rachides of palms in mangrove habitats, is introduced in this paper based on morphological and phylogenetic evidence. Phylogenetic relationships of related taxa were inferred from combined LSU, SSU, TEF1α, and RPB2 sequence data, and the analyses indicate that A. palmarum could be recognized as a distinct group in Pleosporales, but its familial placement needs to be further resolved. The morphological characters of this new taxon are also different from other members in Pleosporales by its deeply immersed ascomata, long pedicellate asci, and biseriate to triseriate, 1-(rarely 3) septate, brown, fusiform ascospores with acute or narrowly pointed ending cells. Acuminatispora gen. nov. (Pleosporales, incertae sedis) is therefore established to accommodate the new taxon A. palmarum. Furthermore, phylogenetic relationships of Acrocordiopsis and Caryospora are discussed with a consideration of morphological observations.  相似文献

Molecular phylogenetic and comparative morphological studies provide evidence for the recognition of a new family, Chorioactidaceae, in the Pezizales. Four genera are placed in the family: Chorioactis, Desmazierella, Neournula, and Wolfina. Based on parsimony, likelihood, and Bayesian analyses of LSU, SSU, and RPB2 sequence data, Chorioactidaceae represents a sister clade to the Sarcosomataceae, to which some of these taxa were previously referred. Morphologically these genera are similar in pigmentation, excipular construction, and asci, which mostly have terminal opercula and rounded, sometimes forked, bases without croziers. Ascospores have cyanophilic walls or cyanophilic surface ornamentation in the form of ridges or warts. So far as is known the ascospores and the cells of the paraphyses of all species are multinucleate. The six species recognized in these four genera all have limited geographical distributions in the northern hemisphere.  相似文献

Lasiobolidium gracile isolated from a soil sample collected from California, USA is described as a new species. This species is characterized by yellowish brown to reddish brown, nonostiolate ascomata with numerous, long straight appendages and a translucent peridium, subglobose to broadly clavate asci, hyaline to pale yellowish brown, ellipsoidal ascospores. It differs from the other species ofLasiobolidium in the straight and narrow appendages and the large ascospores. A key is presented separating the seven known species.  相似文献

Ascomata of a Metarhizium species were collected from Tochigi Prefecture, Japan. The ascomata were similar to M. kalasinense in its host preference for elaterids, olive-green clavate stromata, and obliquely immersed perithecia, but the asci and ascospores were significantly shorter in length than those of the latter species. Furthermore, phylogenetic analyses based on the internal transcribed spacer region of nuclear ribosomal DNA and the elongation factor 1-alpha gene showed that the Japanese species was phylogenetically distinctive in the M. anisopliae lineage, including M. kalasinense. Accordingly, we describe a new species, M. brachyspermum. Isolates of this species produced olive-green conidial masses on the surface of the colony. Also, hyphal anastomoses between two metulae were often observed on conidiophores.  相似文献
