By using multinuclear (1H, 13C, 17O, 95Mo, 183W) magnetic resonance spectroscopy (1D and 2D), D-gulonic acid is found to form ten and seven complexes, respectively, with tungsten(VI) and molybdenum(VI), in aqueous solution, depending on pH and metal-ligand molar ratios. Two isomeric 1:2 (metal-ligand) complexes involving the carboxylate and the adjacent OH group are present in the pH range 2-9. At intermediate and high pH, molybdate forms a 2:1 tetradentate complex involving the four secondary hydroxyl groups, whereas tungstate forms one 2:1 terdentate species. At low and intermediate pH values, three 2:1 complexes are found for both metals, involving the carboxylate group and three secondary hydroxyl groups, as well as a 5:2 species involving the carboxylate group and all the secondary hydroxyl groups; the concentration of this species increases in time mainly at the expense of 2:1 and 1:2 complexes. Tungstate can also form two additional species, probably a 5:2 species involving the carboxylate group and all the hydroxyl groups, and a 2:1 pentadentate species involving the carboxylate group and all the secondary hydroxyl groups. In alkaline solutions, tungstate is able to form an additional 2:1 pentadentate complex involving all the hydroxyl groups.  相似文献

This study presents a site-resolved experimental view of backbone C(alpha)H and NH internal motions in the 56-residue immunoglobulin-binding domain of streptococcal protein G, GB1. Using (13)C(alpha)H and (15)NH NMR relaxation data [T(1), T(2), and NOE] acquired at three resonance frequencies ((1)H frequencies of 500, 600, and 800 MHz), spectral density functions were calculated as F(omega) = 2omegaJ(omega) to provide a model-independent way to visualize and analyze internal motional correlation time distributions for backbone groups in GB1. Line broadening in F(omega) curves indicates the presence of nanosecond time scale internal motions (0.8 to 5 nsec) for all C(alpha)H and NH groups. Deconvolution of F(omega) curves effectively separates overall tumbling and internal motional correlation time distributions to yield more accurate order parameters than determined by using standard model free approaches. Compared to NH groups, C(alpha)H internal motions are more broadly distributed on the nanosecond time scale, and larger C(alpha)H order parameters are related to correlated bond rotations for C(alpha)H fluctuations. Motional parameters for NH groups are more structurally correlated, with NH order parameters, for example, being larger for residues in more structured regions of beta-sheet and helix and generally smaller for residues in the loop and turns. This is most likely related to the observation that NH order parameters are correlated to hydrogen bonding. This study contributes to the general understanding of protein dynamics and exemplifies an alternative and easier way to analyze NMR relaxation data.  相似文献

The crystal structure of isothiocyanatotriphenyl- (pyridinium-2-carboxylato)tin(IV) monohydrate is reported. The crystals are monoclinic, space group P21/n, a = 10.349(2), b =12.003(2), c = 19.325(4) Å, β = 97.68(2)°, Z = 4, refined to RF = 0.024 on 4249 observed reflections.The tin(IV) atom is five-coordinate, being bound to three phenyl groups, the isothiocyanato nitrogen atom and an oxygen from the picolinic acid. The geometry around the tin atom is trigonal bipyramidal, with the three phenyl groups occupying the equatorial positions, while the picolinic acid oxygen and the isothiocyanato nitrogen are coordinated axially. The acidic proton of picolinic acid has shifted position in the complex, and is bound to the heterocyclic nitrogen atom. The acid is thus coordinated in the form of a zwitterion. These trigonal bipyramidal units are linked together as dimers by pairs of water molecules, each of which hydrogen- bonds to the non-coordinated carboxylate oxygen atoms of both picolinic acid molecules, plus the heterocyclic nitrogen atom of one picolinic acid molecule. For complex formation with the protonated acid, theheterocyclic nitrogen should be alpha to the carboxylic acid group.  相似文献

Multinuclear ((1)H, (13)C, (17)O, (31)P, (95)Mo, (183)W) magnetic resonance spectroscopy (1D and 2D) has been used to show that 6-phospho-d-gluconic acid forms three complexes with tungsten(VI) and six complexes with molybdenum(VI) in aqueous solution, depending on pH and concentration. Two isomeric 1:2 (metal-ligand) complexes are detected both with tungstate(VI) and molybdate(VI), having MO(2)(2+) centres and involving the carboxylate and the adjacent OH groups in addition to one 2:1 (metal-ligand) complex possessing a M(2)O(5)(2+) centre, with the ligand being coordinated by the carboxylate group and the three consecutive OH groups in positions 2, 3 and 4. Molybdate(VI) forms three additional species, which are not detected with tungstate. One of them is a 2:1 complex with a Mo(2)O(5)(2+) centre, with the ligand being tetradentate via O-3, O-4, O-5 and the phosphate group. The other two are 12:4 species, which can be seen as two 1:2 complexes bound together in a ring through two diphosphomolybdate moieties each derived from heptamolybdate by inclusion of two phosphate groups from the ligands.  相似文献

Well-defined (nearly monodispersed) poly(poly(ethylene glycol)monomethacrylate)-Si hybrids were prepared via surface-initiated atom transfer radical polymerization (ATRP) of the poly(ethylene glycol)monomethacrylate (PEGMA) macromonomer on the hydrogen-terminated Si(111) surface (Si-H surface). Both the active chloride groups at the chain ends (from the ATRP process) and the chloride groups converted from some ( approximately 32%) of the -OH groups of the Si-C bonded PEGMA polymer, or P(PEGMA), brushes were used as leaving groups for the covalent coupling of heparin. For the heparinized P(PEGMA)-Si hybrid surfaces, protein adsorption and platelet adhesion were significantly suppressed. The well-defined and dense P(PEGMA) brushes, prepared from surface-initiated ATRP, had allowed the immobilization of a relatively high concentration of heparin (about 14 mug/cm(2)). The resulting silicon surface exhibited significantly improved antithrombogenecity with a plasma recalcification time (PRT) of about 150 min. The persistence of high bioactivity for the immobilized heparin on the hybrid surfaces can be attributed to the biocompatibility of the PEGMA units, as well as their role as spacers in providing the immobilized heparin with a higher degree of conformational freedom in a more hydrophilic environment. Thus, the heparin-coupled P(PEGMA)-Si hybrids with anti-fouling and antithrombogenic surfaces are potentially useful in silicon-based implantable devices and tissue engineering.  相似文献

The protection of the sugar moiety of a 5-formyluracil nucleoside with acid-labile protecting groups allows for the deprotection of the sugar of a subsequently formed nucleoside possessing a 5-nitrovinyl side-chain. Illus. 1961.—The genus Tridens has been studied from cytological, morphological, anatomical and histological points of view. Species of Tridens can be divided into 2 groups according to the characters of caryopses, stigmas, lemmas, paleas and general habit. In accordance with the morphological disparities, the 2 groups have different appearances of the anatomy of leaf margins and midribs and also of the bicellular microhairs on the epidermis of the leaf blade. Further, the 2 groups have different basic chromosome numbers, x=8 and x=10, and some distinction in karyotype. Somatic chromosomes of 8 species have been examined. The evidence given by the combined studies indicates that Tridens should be regarded as a mixture of 2 phylogenetically distant groups. Based on this conclusion, the 5 species representing one of the 2 groups (T. pilosus, T. avenaceus, T. grandiflorus, T. nealleyi and T. pulchellus) are transferred to the genus Erioneuron which is hereby revived. Nomenclatural changes are made for the species of the revived genus.  相似文献

The importance of Ca2+-signaling for many subcellular processes is well established in higher eukaryotes, whereas information about protozoa is restricted. Recent genome analyses have stimulated such work also with Alveolates, such as ciliates (Paramecium, Tetrahymena) and their pathogenic close relatives, the Apicomplexa (Plasmodium, Toxoplasma). Here we compare Ca2+ signaling in the two closely related groups. Acidic Ca2+ stores have been characterized in detail in Apicomplexa, but hardly in ciliates. Two-pore channels engaged in Ca2+-release from acidic stores in higher eukaryotes have not been stingently characterized in either group. Both groups are endowed with plasma membrane- and endoplasmic reticulum-type Ca2+-ATPases (PMCA, SERCA), respectively. Only recently was it possible to identify in Paramecium a number of homologs of ryanodine and inositol 1,3,4-trisphosphate receptors (RyR, IP3R) and to localize them to widely different organelles participating in vesicle trafficking. For Apicomplexa, physiological experiments suggest the presence of related channels although their identity remains elusive. In Paramecium, IP3Rs are constitutively active in the contractile vacuole complex; RyR-related channels in alveolar sacs are activated during exocytosis stimulation, whereas in the parasites the homologous structure (inner membrane complex) may no longer function as a Ca2+ store. Scrutinized comparison of the two closely related protozoan phyla may stimulate further work and elucidate adaptation to parasitic life. See also "Conclusions" section.  相似文献

In zinc proteins, the Zn2+ cation frequently binds with a tetrahedral coordination to cysteine and histidine side chains, for example, in many DNA-binding proteins, where it plays primarily a structural role. We examine the possibility of thiolate protonation in Cys(x)His(y)-Zn2+ groups, both in proteins and in solution, through a combination of theoretical calculations and database analysis. Seventy-five percent of the thiolate-coordinated zincs in the Cambridge Structural Database are tetrahedral, while di-alkanethiol coordination always involves five or more ligands. Ab initio quantum calculations are performed on (ethanethiol/thiolate)(3)imidazole-Zn2+ complexes in vacuum, yielding geometries and gas phase basicities. Protonating one (respectively two) thiolates increases the Zn-S(thiol) distance by 0.4 A (respectively 0.3 A), providing a structural marker for protonation. The stabilities of the complexes in solution are compared by combining the gas phase basicities with continuum dielectric solvation calculations. In a continuum solvent with permittivity epsilon = 4, 20, or 80, one of three thiolates is predicted to be protonated at neutral pH. By extension, Cys4-Zn2+ groups are expected to be protonated in the same conditions. In contrast, most Cys3His and Cys4 geometries in the Protein Data Bank (PDB) appear consistent with all-thiolate Zn2+ coordination. This apparent discrepancy is resolved by two recent surveys of zinc protein structures, which suggest that these all-thiolate sites are stabilized by charged and polar groups nearby in the protein, thus overcoming their intrinsic instability. However, the experimental resolution is not sufficient in all the PDB structures to rule out a thiol/thiolate mixture, and protonated thiolates may occur in some proteins not solved at high resolution or not represented in the PDB, as suggested by recent mass spectrometry experiments; this possibility should be allowed for in X-ray structure refinement.  相似文献

Phospholipid bilayer interaction of olanzapine (OLZ), a thienobenzodiazepine derivative and an antipsychotic agent, has been studied with (13)C and (31)P solid-state NMR. A dipalmitoyl phosphatidylcholine (60%)/1-palmitoyl-2-oleoyl phosphatidylserine (40%) bilayer (DPPC(60%)/POPS(40%)) with 50 wt.% H(2)O, with and without 10 mol% OLZ have been investigated. The results reveal that both the serine and the choline head groups are affected by OLZ interaction with the bilayer. The OLZ interaction with the serine and the choline head groups appears to be caused by electrostatic attraction to the serine head group carboxyl and repulsion of the choline head group positively charged nitrogen. (31)P MAS NMR experiments show the appearance of two new (31)P resonances both for the PS and the PC phosphorous in the presence of OLZ. Static (31)P NMR spectra demonstrate a decrease in chemical shift anisotropy (CSA) of the OLZ containing bilayer when in the liquid-crystalline phase and an increase in CSA when in the gel state.  相似文献

Speciation and equilibria in the H+-Cd2+-N-(phosphonomethyl)glycine (PMG, H3L) system have been studied in 0.1 M Na(Cl) medium at 25.0 °C. Formation constants for a series of mononuclear complexes, CdHL, CdL, CdL2 4− and CdL(OH)2− were determined from potentiometric titrations. The structures of the predominating species CdL, and CdL2 4− in solution were investigated using EXAFS and IR spectroscopic techniques. In the 1:1 complex bonds are formed between the Cd(II) ion and all three donor groups (amino, carboxylate, phosphonate) of the PMG molecule resulting in two 5-membered chelate rings. At the remaining three of the corners of the distorted Cd(II) octahedra oxygens were found which are replaced by donor groups of a second PMG molecule in the 1:2 complex. Furthermore, a solid phase consisting of Cd9(PMG)6(H2O)12 · 6H2O crystals was synthesized and the crystal structure was determined. The structure consists of six CdL octahedra connected through a seventh Cd-O octahedron in the centre of the entity, with two additional Cd-O octahedra located at the apices of the unit formed.  相似文献

To evaluate the effect of egg age of Spartocera dentiventris (Berg) on the parasitism by Gryon gallardoi (Brethes), groups of 12 eggs of 2, 3, 4, 5, 6, 7, 8, and 12 days old were exposed to one female of G. gallardoi on a test tube, with the wasp observed for 2h subsequently. Later, the eggs were individualised and incubated (25 +/- 1 degrees C; 12h photoperiod) until adult emergence. To identify the preferred age, groups of 12 eggs of 1, 3, 5, 7 and 12 days of age were simultaneously exposed to a parasitoid female on an arena for 2h. These groups were also individualised and incubated until parasitism was confirmed. On all ages tested parasitism was at about 88.5 +/- 2.64%, with a reduction in adult parasitoid emergence with age increasing (P < 0.01). The development time increased with increasing egg age (P < 0.01), with males developing faster. Parasitoid sex ratio for all ages tested was female-biased (0.49:1), with the proportion of males invariant with host age (P > 0.05). When the egg groups were exposed simultaneously, the female preferred eggs one and three days old. The results indicate a preference of G gallardoi for younger eggs of S. dentiventris, with host egg age exerting a negative effect on development time and offspring emergence of the parasitoid.  相似文献

The crystal structure of chlorotriphenyl(quinolinium-2-carboxylato)tin(IV) monohydrate is reported. The crystals are monoclinic, space group C2/c with cell parameters a = 20.048(3) Å, b = 11.724(1) Å, c = 23.291(3) Å, ]gb = 113.42(1), Z = 8, refined to RF = 0.034 on 3331 observed reflections. The tin(IV) atom is five-coordinate, being found to three phenyl groups, the chlorine atom and an oxygen from the quinaldic acid. The geometry around the tin atom is trigonal bipyramidal, with the three phenyl groups occupying the equatorial positions, and the chlorine and quinaldic acid oxygen, the apical ones. The acidic proton of quinaldic acid has shifted position in the complex, and is bound to the heterocyclic nitrogen atom.The acid is thus coordinated in the form of a zwitterion. These trigonal bipyramidal units are linked together as dimers by pairs of water molecules, each of which hydrogen-bonds to the non-coordinated carboxylate oxygen atoms of both quinaldic acid molecules, plus the heterocyclic nitrogen atom of one quinaldic acid molecule. For complex formation with the protonated acid, the heterocyclic nitrogen should be alpha to the carboxylic acid group.  相似文献

In an effort to improve indole-based CB(2) cannabinoid receptor ligands and also to develop SAR for both the CB(1) and CB(2) receptors, 47 indole derivatives were prepared and their CB(1) and CB(2) receptor affinities were determined. The indole derivatives include 1-propyl- and 1-pentyl-3-(1-naphthoyl)indoles both with and without a 2-methyl substituent. Naphthoyl substituents include 4- and 7-alkyl groups as well as 2-, 4-, 6-, 7-methoxy and 4-ethoxy groups. The effects of these substituents on receptor affinities are discussed and structure-activity relationships are presented. In the course of this work three new highly selective CB(2) receptor agonists were identified, 1-propyl-3-(4-methyl-1-naphthoylindole (JWH-120), 1-propyl-2-methyl-3-(6-methoxy-1-naphthoylindole (JWH-151), and 1-pentyl-3-(2-methoxy-1-naphthoylindole (JWH-267). GTPgammaS assays indicated that JWH-151 is a full agonist at CB(2), while JWH-120 and JWH-267 are partial agonists. Molecular modeling and receptor docking studies were carried out on a set of 3-(4-propyl-1-naphthoyl)indoles, a set of 3-(6-methoxy-1-naphthoyl)indoles and the pair of N-pentyl-3-(2-methoxy-1-naphthoyl)indoles. Docking studies indicated that the CB(1) receptor affinities of these compounds were consistent with their aromatic stacking interactions in the aromatic microdomain of the CB(1) receptor.  相似文献

The preparation, spectral properties, and crystal structure of a mononuclear copper(II) complex of acetylsalicylate and pyridine are reported. The complex exists as bis(acetylsalicylato)bis(pyridine)copper(II) both in the solid state and in chloroform solution. The crystal is monoclinic, space group P21/n, with a = 17.823(5), b = 10.903(4), c = 6.598(2) Å, β = 95.74(2)°. The final refinement used 1472 observed reflections and gave an R of 0.046. The copper atom is surrounded by four atoms in a trans square planar arrangement with two short CuO distances of 1.949(3) Å and two CuN distances of 2.003(4) Å. Two longer CuO distances of 2.623(3) Å are made with the remaining oxygen atoms of the aspirin carboxylate groups.  相似文献

A recently developed technique, i.e., two-dimensional infrared (2D IR) correlation spectroscopy, is used to study the thermal behavior of poly(ethylene-co-vinyl alcohol)-graft-poly(epsilon-caprolactone), a new synthesized highly grafted copolymer. The use of the 2D IR approach to analyze temperature-dependent spectra collected in situ during the temperature elevation process effectively enhanced the spectral resolution and revealed details on the hydrogen bonding and conformational change which are not easily detected in the traditional one-dimensional spectra. The sequence of the spectral changes of different OH and CH(2) fundamental vibrations during heating the polymer was inferred from the signs of the asynchronous peaks. Evidence that the conformational changes of the methylene groups precede the loosening of hydrogen bonds is provided.  相似文献

The binding of copper(II) and zinc(II) to oxidized glutathione   总被引:1,自引:0,他引:1  
1H and 13C NMR studies of Zn(II) binding to oxidized glutathione (GSSG) in aqueous solution over the pH range 4-11 show that it forms a complex with a 1:1 Zn:GSSG stoichiometry. At pH values between 6 and 11 the metal ligands are the COO- and NH2 groups of the glutamate residues. Below pH 5 the glycine end of the molecule also binds to the metal ions. EPR and visible absorption spectra of Cu(II) GSSG solutions suggest that similar complexes are formed with Cu(II). The solid products obtained from these solutions are shown by analysis and EPR to be primarily binuclear with Cu2GSSG stoichiometry, although the structures depend on the pH and stoichiometry of the solution from which they were obtained.  相似文献

The crystal structures of four Ag(I) and Hg(II) complexes of the ligand 1,4-bis(1-benzyl-benzimidazol-2-yl)cyclohexane (N-BBzBimCH) have been described, that is, [Hg2(N-BBzBimCH)Cl4] (1), [Hg(N-BBzBimCH)Br2] (2), [Ag(N-BBzBimCH)](NO3)(H2O) (3) and [Ag2(N-BBzBimCH)(CF3OCO)2] (4). All these compounds show 1D polymeric structures in the solid state. In complexes 1 and 4, the chloride ions and the trifluoroacetate groups bridge the [Hg2(N-BBzBimCH)Cl2] and [Ag2(N-BBzBimCH)] fragments, respectively, to generate 1D polymers. While the bromide ions in complex 2 and nitrate groups in complex 3 are only serving as terminal ligands to suffice the coordination geometry of the metal centers. In all cases, weak intermolecular interactions such as C-H?X (X = Cl, Br) contacts, hydrogen bonds, π-π interactions and C-H?π stacking play important roles to extend the 1D chain structures to 2D network. Solid state fluorescence of these compounds was also studied.  相似文献

Two elastin-like poly(pentapeptides), poly(AV1GV2P) and poly(G1V1G2V2P), have been studied in water and in solid state by ATR FTIR and Raman spectroscopy in combination with model ab initio calculations. In aqueous solutions below the transition temperature T(t), a part of the amide groups and of the methyl groups of both polypentapeptides interacts with neighboring water molecules, whereas the other part of amide groups mutually interacts forming a beta-sheetlike structure. Below T(t), poly(AV1GV2P) is dissolved more perfectly, and the water shells around the polymer chains are more closely structured. The suspension of poly(AV1GV2P) formed above T(t) is more compact and, on cooling, resists more to the reverse dissolution, whereas the suspension of poly(G1V1G2V2P) contains more water molecules bound to the carbonyl of amide groups and on backward cooling dissolves fairly reversibly. The measured poly(pentapeptides) tend to form beta-turns due to the conformational transition on the residue between P and V1.  相似文献
