Experiments were conducted on five chronically instrumented unanesthetized sheep to determine the effects of sustained hypoproteinemia on lung fluid balance. Plasma total protein concentration was decreased from a control value of 6.17 +/- 0.019 to 3.97 +/- 0.17 g/dl (mean +/- SE) by acute plasmapheresis and maintained at this level by chronic thoracic lymph duct drainage. We measured pulmonary arterial pressure, left atrial pressure, aortic pressure, central venous pressure, cardiac output, oncotic pressures of both plasma and lung lymph, lung lymph flow rate, and lung lymph-to-plasma ratio of total proteins and six protein fractions for both control base-line conditions and hypoproteinemia base-line conditions. Moreover, we estimated the average osmotic reflection coefficient for total proteins and the solvent drag reflection coefficients for the six protein fractions during hypoproteinemia. Hypoproteinemia caused significant decreases in lung lymph total protein concentration, lung lymph-to-plasma total protein concentration ratio, and oncotic pressures of plasma and lung lymph. There were no significant alterations in the vascular pressures, lung lymph flow rate, cardiac output, or oncotic pressure gradient. The osmotic reflection coefficient for total proteins was found to be 0.900 +/- 0.004 for hypoproteinemia conditions, which is equal to that found in a previous investigation for sheep with a normal plasma protein concentration. Our results suggest that hypoproteinemia does not alter the lung filtration coefficient nor the reflection coefficients for plasma proteins. Possible explanations for the reported increase in the lung filtration coefficient during hypoproteinemia by other investigators are also made.  相似文献

Mammalian lung has a high concentration of dipalmitoyl phosphatidylcholine and other phospholipids in which both fatty acid ester chains are saturated, as opposed to the usual asymmetric phospholipid (one saturated fatty acid and one unsaturated fatty acid). The acyl transferase system in dog lung microsomes was studied by determining the reactivities of various acyl CoA derivatives with 1-lyso-2-acyl- and 1-acyl-2-lyso-phosphatidylcholine. The 16:0 derivative had equal reactivity for both the 1- and 2-lyso positions. The 18:0 derivative also exhibited marked reactivity toward both positions, although the specific activity of the enzyme when palmitoyl CoA was used was approximately twice that compared to when stearoyl CoA was used. The 16:1 derivative showed approximately the same reactivity toward the 1-lyso position as did 16:0 but both 16:1 and 18:1 were more active with the 2-lyso position. These results suggest that acyl transferases may be important in the lung to insure that sufficient amounts of dipalmitoyl phosphatidylcholine will always be present for use in pulmonary surfactant biosynthesis. It is also conceivable that the acyl transferase system described acts on 1- and 2-lyso-palmitoyl phosphatidylcholine (produced by phospholipase hydrolysis of dipalmitoyl phosphatidylcholine) in order to produce phosphatidylcholine species needed for cellular purposes other than surfactant function.  相似文献

Quantification of surfactant phospholipids in the dog lung   总被引:1,自引:0,他引:1  
We quantified total phospholipid (PL), total and disaturated phosphatidylcholine (PC and DSPC), phosphatidylglycerol (PG), and total protein in alveolar washings and lung tissue in 22 dog lungs. Quantitative recovery of alveolar material and assessment of its possible contamination by blood lipids were important determinants of methodology. To remove blood, the vessels of half the lungs were perfused with a fluorocarbon emulsion before lavage. The volume of blood removed by perfusion and the quantity and fatty acid patterns of its whole blood and plasma PL and PC were determined. Washings of unperfused lungs contained means of 21% more PL and 24% more PC than those of perfused lungs. Although this excess could be accounted for by the PL and PC in pulmonary blood, the hemoglobin and total protein content of washings and their PC fatty acid patterns indicated that blood lipids were not a major source of the excess lipid in washings of unperfused lungs. Using more recent morphometric estimates rather than the indirect ones previously used by others, the quantity of alveolar DSPC (1 mg/g lung) is calculated to be 1.8 times the amount necessary to form a packed monolayer on the internal surface of the lung at functional residual capacity.  相似文献

We have previously reported that lymph from the chronic sheep lung lymph preparation contains 25-60% lymph from nonpulmonary sources. In subsequent studies we found that the lymph flow rate from cannulated lymph vessels depends on the resistance and position of the lymph cannula. Because we did not account for these factors in our estimate of the amount of nonpulmonary lymph in the sheep lung lymph preparation, our data were not accurate. We probably overestimated the amount of nonpulmonary lymph. However, the presence of nonpulmonary lymph remains a potentially serious problem with the sheep lung lymph preparation.  相似文献
