Large-conductance Ca2+-activated K+ (BK) channels are reported to be essential for NADPH oxidase-dependent microbial killing and innate immunity in leukocytes. Using human peripheral blood and mouse bone marrow neutrophils, pharmacological targeting, and BK channel gene-deficient (BK–/–) mice, we stimulated NADPH oxidase activity with 12-O-tetradecanoylphorbol-13-acetate (PMA) and performed patch-clamp recordings on isolated neutrophils. Although PMA stimulated NADPH oxidase activity as assessed by O2 and H2O2 production, our patch-clamp experiments failed to show PMA-activated BK channel currents in neutrophils. In our studies, PMA induced slowly activating currents, which were insensitive to the BK channel inhibitor iberiotoxin. Instead, the currents were blocked by Zn2+, which indicates activation of proton channel currents. BK channels are gated by elevated intracellular Ca2+ and membrane depolarization. We did not observe BK channel currents, even during extreme depolarization to +140 mV and after elevation of intracellular Ca2+ by N-formyl-L-methionyl-L-leucyl-phenylalanine. As a control, we examined BK channel currents in cerebral and tibial artery smooth muscle cells, which showed characteristic BK channel current pharmacology. Iberiotoxin did not block killing of Staphylococcus aureus or Candida albicans. Moreover, we addressed the role of BK channels in a systemic S. aureus and Yersinia enterocolitica mouse infection model. After 3 and 5 days of infection, we found no differences in the number of bacteria in spleen and kidney between BK–/– and BK+/+ mice. In conclusion, our experiments failed to identify functional BK channels in neutrophils. A compartment model to estimate the different phosphorus metabolicrates in Daphnia galeata is presented. The model has three compartments:gut, metabolic pool and structural pool. Existing two-compartmentmodels used for carbon and phosphorus turnover in Daphnia donot allow estimation of ingestion and egestion rates. We extendedexisting two-compartment models with one more compartment, thegut, which allowed us to estimate both the ingestion and theegestion rates. Parameters of the model are estimated from asingle experiment of feeding unlabelled Daphnia with 32P-labelledScenedesmus obliquus. Separate experiments with juvenile andadult daphnids were carried out in order to compare their metabolisms.This model permits a reliable estimation of the different metabolicrates of Daphrna in a single experiment and discriminates clearlybetween animals of different sizes.  相似文献

Blooms of the toxic red tide phytoplankton Heterosigma akashiwo(Raphidophyceae) are responsible for substantial losses withinthe aquaculture industry. The toxicological mechanisms of H.akashiwoblooms are complex and to date, heavily debated. One putativetype of ichthyotoxin includes the production of reactive oxygenspecies (ROS) that could alter gill structure and function,resulting in asphyxiation. In this study, we investigated thepotential of H.akashiwo to produce extracellular hydrogen peroxide,and have investigated which cellular processes are responsiblefor this production. Within all experiments, H.akashiwo producedsubstantial amounts of hydrogen peroxide (up to 7.6 pmol min–1104 cells–1), resulting in extracellular concentrationsof ~0.5 µmol l–1 H2O2. Measured rates of hydrogenperoxide production were directly proportional to cell density,but at higher cell densities, accuracy of H2O2 detection wasreduced. Whereas light intensity did not alter H2O2 production,rates of production were stimulated when temperature was elevated.Hydrogen peroxide production was not only dependent on growthphase, but also was regulated by the availability of iron inthe medium. Reduction of total iron to 1 nmol l–1 enhancedthe production of H2O2 relative to iron replete conditions (10µmol l–1 iron). From this, we collectively concludethat production of extracellular H2O2 by H.akashiwo occurs througha metabolic pathway that is not directly linked to photosynthesis.  相似文献

The experimentally measured oxygen consumption rate by the cladoceran,Ceriodaphnia dubia, showed a linear increase between 5 and 20°C.Oxygen consumption rates of C. dubia were estimated in situfrom respiratory electron transport system (ETS) activity inLake Rotongaio during summer stratification and winter mixing.Oxygen consumption was 0.002 µl O2 animal–1 h–1in the hypolimnion and 0.076 µl O2 animal–1 h–1in the epilimnion during stratification. Phagotrophic protists are major components of pelagic food webs,both as consumers of bacterial and phytoplankton cells, andas regenerators of inorganic nutrients. In this study, we estimatedthe efficiency of ammonium regeneration by protists feedingon bacteria within natural plank-tonic assemblages, using a15N tracer method, in which the excretion of 15N-labeled ammoniumdue to grazing on 15N pre-labeled bacteria was followed overtime. We tested this approach in experiments based on the additionof heat-killed 15N-labeled bacteria to laboratory cultures andto samples of coastal seawater. During two experiments, variationin abundance of bacterivores and bacterioplankton resulted innon-constant grazing rates. Deterministic computer models thatused abundance of bacteria and protists as variables were developedto estimate best-fit values of grazing mortality (g, h–1)and of ammonium regeneration efficiency (RE, fraction of theinitial 15N label in added bacteria which is released as ammonium).Estimated ammonium RE were 0.30–0.35 for one trophic linksystems with both a monospecific culture and a mixed speciesassemblage of bacterivorous flagellates. RE was higher for multi-trophicstep food webs: 0.60 for 5 µm pre-screened coastal seawaterand 0.90 for whole coastal seawater.  相似文献

In vitrorates of gross and net oxygen production were measuredas a function of light intensity in some plankton communitiescollected from Bedford Basin, Nova Scotia, and in a monoclonalculture of Synechococcus. The rate of gross oxygen productionwas measured by a technique in which the stable oxygen isotope,18O, serves as a photosynthetic tracer Net oxygen productionwas measured by automated Winkler technique. The rate of communityrespiration in the light was then determined by the differencebetween gross and net rates of oxygen production. In the naturalpopulations examined, neither gross nor net oxygen productionrates were significantly inhibited at the highest light intensitymeasured (500–800 µE m–2 s–1) In a samplein which the dark respiration rate was small relative to themaximal rate of production [Pmax;sensu Platt et al (1980) JMar. Res., 38, 687–701] the rates of ‘light’respiration were 3 times greater. In two other communities,with high rates of dark respiration relative to Pmaxthe ratesof ‘light’ respiration were closer to rates of darkrespiration. In the Synechococcus clone, both gross and netoxygen production rates were inhibited at high light intensities.Rates of ‘light’ respiration were found to varyas a function of light intensity. The greatest rates of respirationwere measured in samples incubated at light intensities thatwere just saturating (100 µE m–2 s–1). The stimulus for these experiments came from a recent seriesof papers which have suggested that the 14C technique may underestimateprimary production by as much as 10-fold. We evolved the followingstrategy to attempt to verify the 14C technique in nearshorewaters: (i) to examine the validity of in vitro (i.e., bottleincubation) measurements by comparing observed in situ oxygenchanges in a large enclosed natural ecosystem against thosedetermined in vitro and if no evidence of containment effectswere indicated, (ii) compare 14C and oxygen measurements insimultaneous in vitro incubations. The first step essentiallytests the containment problem, the second the physiologicaland calibration problems of the 14C technique. The first experimentwas run with nitrate as the main source of inorganic nitrogen,the second with ammonia. PQs for converting the 14C measurementsto oxygen values were calculated from the equation PQ = PQc+ 2/(C/NO3) where PQc is the ‘carbon PQ’ (takenas 1.25) and (C/NO3) is the molar carbon to nitrate assimilationratio. Although there appear to be some minor residual problemsin the interpretation of the data when nitrate was the dominantnitrogen source, the overriding conclusions were: first, thatthe close agreement between the changes in in situ and in vitrodissolved oxygen concentration during the photoperiod gave noevidence for any notable containment effect upon photosynthesis.Secondly, the in vitro rates of 14CO2-determined photosyntheticproduction and gross photosynthetic oxygen production agreed,within the precision of the two techniques. The experiment furtherdemonstrated the need to determine soluble as well as particulateorganic production and to pay attention to the potential effectof the nitrogen nutrient upon the PQ. Thus it was concludedthat our data give no evidence for marked errors in the 14C-techniqueof measuring primary organic production for coastal waters. Laboratory experiments were carried out on the holoplanktonicscyphomedusa Pelagia noctiluca, which exhibits population explosionssome years in the Mediterranean Sea. Feeding experiments performedon small, laboratory-reared medusae showed specific daily rationsof 13 and 35% at 5 and 20 Anemia nauplii prey 1–1 respectively,while basal NH+4 excretion reached 3.9% (specific elementalmetabolic rate) for adults at 22°C. These values appearto be consistent with those previously observed by several authorsfor other planktonic coelenterates. Assuming that such laboratoryphysiological rates are indicative of food requirements in naturalenvironments, and given the high abundance of P. noctiluca observedregionally, we conclude that P. noctiluca can have a strongpredatory impact in the Ligurian Sea (north-west Mediterranean),particularly during the summer and early fall. 3Present address: ORSTOM B.P. 2528, Bamako, Mali  相似文献

In the stoichiometric study of phyto–zooplankton interactions,a controversy exists about how Daphnia species regulate theirfeeding rate when submitted to low-quality food (i.e. high dietarycarbon:phosphorus [C:P] ratio). In this study, we gathered dataover 3 years on in situ clearance rates of a Daphnia galeatapopulation, by conducting grazing experiments from April 1998to October 2000 in the Esch-sur-Sûre reservoir (Grand-Duchyof Luxembourg). Observed clearance rates (2.5–13.5 mLindividual–1 day–1, mean 7.0 mL individual–1day–1) were correlated with population and environmentalvariables. Mean body size of Daphnia individuals was the bestpredictor of clearance rate (r2 = 0.639), followed by watertemperature (r2 = 0.262) and P concentration in the seston (r2  相似文献

Daphnia can suppress ciliates and rotifers through predationand interference competition, but it is not known whether thisproduces any direct benefit to Daphnia. We conducted survivorshipand cohort lifetable experiments to determine whether Daphniacan utilize ciliates and rotifers as food. Three species ofoligotrich ciliates (Halteria grandinella, Strobilidium gyransand Strobilidiumvelox) and one rotifer (Keratella cochlearis)were used. Lifetable experiments were conducted with a basallevel of algae (Cryptomonas sp.), plus either ciliates or rotifers,while survivorship experiments had only the rotifers or ciliates.Densities of 30 H.grandinella ml–1, 50 S.gyrans ml–1and 15 S.velox ml–1 enhanced Daphnia pulex's populationgrowth rate 35–50% over controls with only algae. TenS.gyrans ml–1 did not produce a significant change inDaphnia's growth rate. Densities of 100 and 300 K.cochlearis–1 increased Daphnia population growth rates by II and10%, respectively. Both 10 and 50 S.gyrans ml–1 enhancedDaphnia's survivorship compared to starved controls, but neither100 nor 300 K.cochlearis l–1 enhanced its survivorship.The amount of enhancement of Daphnia growth rates by rotifersand ciliates is roughly proportional to the death rates imposedby Daphnia. The death rate imposed by Daphnia on rotifers isa function of both algal density and Daphnia size. The heterotrophic dinoflagellate Noctiluca scintillans has anegligible swimming ability and feeds predominantly on immobileprey. How, then, does it encounter prey? Noctiluca scintillansis positively buoyant and, therefore, we hypothesized that itintercepts prey particles during ascent and/or that microscaleshear brings it into contact with prey. Noctiluca scintillanshas a specific carbon content 1–2 orders of magnitudeless than that typical for protists and, thus, an inflated volume.It also has a density slightly less than that of the ambientwater and therefore ascends at high velocities (-1 m h–1).In stagnant water, clearance rates of latex spheres (5–80µm) increased approximately with prey particle size squared.This scaling is consistent with N.scintillans being an interceptionfeeder. However, absolute clearance rates were substantiallylower than those predicted by modeling N.scintillans both asa spherical and as a cylindrical collector. The latter modelassumes that prey particles are collected on the string of mucusthat may form at the tip of the tentacle. Feeding, growth andprey selection experiments all demonstrated that diatoms arecleared at substantially higher rates than latex beads and otherphytoplankters, particularly dinoflagellates. We propose thatdiatoms stick more efficiently than latex beads to the mucusof N.scintillans and that dinoflagellates reduce fatal contactbehaviorally. We conclude that N.scintillans is an interceptionfeeder and that the high ascent velocity accounts for encounterswith prey. However, the flow field around the cell-mucus complexis too complicated to be described accurately by simple geometricmodels. Fluid shear (0.7–1.8 s–1 had a negativeimpact on feeding rates, which were much less than predictedby models. Noctiluca scintillans can survive starvation forlong periods (>3 weeks), it can grow at low concentrationsof prey (-15 µg C l–1), but growth saturates onlyat very high prey concentrations of 500–1000 µgC l–1 or more. We demonstrate how the functional biologyof N.scintillans is consistent with its spatial and seasonaldistribution, which is characterized by persistence in the plankton,blooms in association with high concentrations of diatoms, andsurface accumulation during quiescent periods or exponentialdecline in abundance with depth during periods of turbulentmixing.  相似文献

An improved and fast version of a closed loop system for measuringnet CO2 exchange rates (NCER) in the field using a portableinfrared gas analyser (IRGA) is described. An electronic timer-differentiatordevice measures and displays very accurately the time periodnecessary to bridge over a constant and preselected CO2 concentrationdifference. The time required for each measurement from startof the measurement to display of final results is c. 9 s forNCER of 24 µmol m–2 s–1 The measurement interval is sufficiently short that no environmentalcontrol in the perspex mini-cuvette is needed. Some satisfactorydata of NCER measurements obtained under field and glasshouseconditions on poplar, sugar bcct and winter wheat are presented. Measurements of biomass and growth rate of two axenic algalcultures were carried out using three different methodologicalapproaches: the specific 14C-labelling of chlorophyll a, [3H]adenineincorporation into DNA and net organic carbon assimilation.Time-course experiments revealed that the specific activitiesof chlorophyll a were significantly higher than the specificactivity of total algal carbon in six of seven experiments.When the specific activity of chlorophyll a is used to calculatethe carbon biomass and growth rate, the carbon biomass of thealgae will thus be underestimated and the specific growth ratewill be too high. Determination of growth rates from incorporationof [3H]adenine gave lower values than those obtained from netorganic carbon assimilation and from 14C incorporation intochlorophyll a. Problems with adenine saturation are suggested.When [3H]adenine is used to measure growth rates in dense algalcultures, additions of >1 µM [3H]adenine are oftenrequired to maximally label the extracellular and intracellularadenine pools and hence DNA.  相似文献

Selective feeding on large algae by copepods involves remotedetection of individual particles and subsequent active captureresponses In this study we use radiotracer experiments to quantifythe clearance rates of five coexisting freshwater copepods andto investigate the relative merits of the chemoreception andmechanoreception hypotheses of remote detection Tropocyclopsand three diaptomid copepods exhibited relatively high clearancerates when feeding on low concentrations of large algae, suggestingthat most previous studies with freshwater copepods have underestimatedmaximal clearance rates and the degree of size selectivity.All five species of copepods exihibited strong selection foran intermediate-sized flagellate (25 µm Cartena) or alarge-sized nonmotile alga (80 µ.m Pediastrum) over asmall-sized flagellate (6 µ.m Chlamydomonas). The weight-specificclearance rate for Tropocylops prasmus feeding on motile Cartena(271 ml mg1 h1) was about twice that of threediaptomid copepods and more than an order-of-magnitude higherthan the estimate for Epischura lacustris feeding on its preferredalga, Pediastrum Assuming that distance chemoreception (‘smell’)is important in remote detection, we predicted that the additionof high concentrations of ‘algal odors’ would obscureany chemical gradients emanating from individual algal cellsand would thereby hinder the remote detection and active captureof large algae Contrary to this hypothesis, the addition ofamino acids, sucrose, and algal extracts had no effect on theclearance rates and selectivity of Diaptomus birgei. These results,together with recent cinematographic studies (Vanderploeg etal.,1990), suggest that mechanoreception is the primary mechanismfor the remote detection of large particles by diaptomid copepods.A raptorial cyclopoid, Tropocyclops prasinus, exhibited strongpreferences for motile algae, whereas a suspension-feeding calanoid,D birgei, did not select between motile and nonmotile cells.Motility appears to be an important factor in algal detectionfor small cyclopoid copepods but not for suspension-feedingdiaptomids  相似文献

The objective of this study was to quantify the respiratoryrequirements and oxygen consumption rates for seasonal hydromedusaepopulations in the North Inlet estuary, Georgetown, SC, USA.Respiration rates were determined for hydromedusae collectedin January 2006, 2007 and July 2006 using microrespirometry.The numerically dominant gelatinous predators were Nemopsisbachei (January 2006 and 2007) and Bougainvillia muscus (July2006). Seasonal oxygen consumption rates were low (0.80–1.27µmol O2 g–1 h–1). Q10 values were high duringJanuary 2006 and July 2006 (4.7), but low during January 2007(0.5), suggesting hydromedusae were sensitive to the exposuretemperatures (7–34°C) in this study and well adaptedto the temperature range encountered in this estuary. Moreover,the oxygen consumption rates during July 2006 showed significantdifferences between the two species. The allometric relationshipbetween respiration rate and hydromedusae weight differed seasonally.Our findings suggest hydromedusae can persist in hypoxic conditionsand may partially explain the frequently observed increase inthese cnidarians under conditions of low dissolved oxygen concentrations.  相似文献

Growth rates, ingestion rates and grazer yields (grazer volumeproduced/prey volume consumed) were measured for six protozoanspecies (ciliates: Favella sp., Strombidinopsis acuminatum,Uronema sp.; heterotrophic dinoflagellates: Amphidinium sp.,Gymnodinium sp., Noctiluca scintillans) in laboratory batchculture experiments. Comparative growth data indicate that theprymnesiophyte Isochrysis galbana, the prasinophyte Mantoniellasquamata, two cryptophyte species and several autotrophic dinoflagellatespecies were suitable foods for these grazers. When grown onoptimized diets at 13C, maximum ciliate growth rates (range0.77–1.01 day–1 uniformly exceeded maximum heterotrophicdioflagellate growth rates (range 0.41–0.48 day–1).A compilation of published data demonstrates that this growthrate difference persists across a range of ciliate and dinoflagellatetaxa and cell sizes. Comparison of volume-specific ingestionrates and yields for the six species studied here showed thatthere was no single explanation for this growth rate disparity.Heterotrophic dinoflagellates exhibited both low ingestion ratesand, in one case, low yields; ciliates were able to achievehigher growth rates via either higher ingestion rates or higheryields, depending on ciliate species. Volume yield increasedover time throughout the exponential growth phase in nearlyall experiments, suggesting variation in response to changingfood concentrations or long-term acclimation to culture conditions.Higher maximum ciliate growth rates mean that these grazershave the potential to exercise tighter control over incipientblooms of their prey than do heterotrophic dinoflagellates.  相似文献

The growth rates of four saline-lake diatom taxa were measuredunder varying conditions of salinity (5, 8 and 11), brine type(sulfate- versus bicarbonate-dominated) and nitrogen form (NH4+versus NO3), using a full factorial design. With NO3as the nitrogen source, Cyclotella quillensis, Cymbella pusillaand Anomoeoneis costata exhibited lower growth rates in thesulfate versus bicarbonate media. The strain of Chaetoceroselmorei used in these experiments, isolated from a sulfate-dominatedlake, was unable to grow on NO3 alone. In the NH4+ treatments,neither salinity nor brine type affected the growth rates ofC.quillensis or C.elmorei. When supplied with NH4+, C.pusillaand A.costata had higher growth rates in the bicarbonate versussulfate media, although for C.pusilla the difference on NH4+was not as great as on NO3. The impact of brine typeon NO3 use is consistent with the theory that sulfateinhibits molybdate uptake, as molybdenum is required for NO3use but not NH4+. Cymbella pusilla was the only taxon affectedby changes in salinity. The four taxa used in these experimentsare frequently found in saline lakes and saline-lake sediments,hence they are used in paleoclimate reconstructions; the resultspresented here provide additional information that may enhancethese diatom-based reconstructions.  相似文献

Detached Kalancho leaves were placed in the dark and changesin the amounts of total soluble N, asparagine, glutamine, aminoN, ammonia, and titratable acid were followed during the periodsof acidification and deacidification. Abundant starch was presentthroughout the experiments. The amount of acid which accumulatedin the initial period was increased by placing the leaves in5 per cent. CO2 and also by decreasing the temperature from25 to 10°. In neither case were the nitrogen fractions affectedin a similar way to the acid levels. No consistent relationshipcould be found between the amount or change in amount of acidand the amounts of the nitrogen fractions. Over prolonged periodsthe drifts in the amounts of the nitrogen fractions were verysimilar to those known in non-succulent leaves. It is concludedthat, despite the large amounts of free acid and starch present,the nitrogen metabolism of detached Kalancho leaves in the darkis similar to that of non-succulent leaves. A possible explanationis suggested for the incorporation of 14C from 14CO2 into nitrogenoussubstances in Kalancho leaves during dark acidification. Changesin the amounts of glutamine, which seemed to be related to oxygenuptake rather than to acid fluctuations, were measured in comparableleaf samples showing different rates of oxygen uptake. The rateof oxygen uptake of leaves and leaf disks in the dark was controlledby placing them in an atmosphere of nitrogen, or 5, 20, or 100per cent, oxygen, by the addition of 10–3 M. cyanide,or by varying the temperature. At the end of a given periodthe amount of glutamine was always greater in the samples whichhad shown the higher rate of oxygen uptake during this perod.This correlation was found irrespective of whether the amountof glutamine had increased or decreased. With the assumptionsthat glutamine production and consumption were proceeding simultaneouslyand that the rate of consumption was unaffected by the rateof oxygen uptake, the experimental values are consistent withthe hypothesis that the rate of glutamine production and therate of oxygen uptake were directly related. Oxygen- and carbon-14-based primary production estimates from9–16 h in vitro incubations were compared in lakes Huronand Michigan. For surface mixing layer compansons, gross O2/14Cphotosynthetic quotients (gross PQ) averaged 2.2, and net O2/14Cphotosynthetic quotients (net PQ) averaged 1.4. The mean grossPQ is consistent with a theoretical P0 based on the CO2 andNO3 assimilation ratio. However, within the deep chlorophylllayer, gross PQ and net PQ averaged 4.9 and 2.8 respectively.These higher values were likely due to excess NO3 reductionat the expense of CO2 uptake. Thus, during short experimentsunder low light conditions, oxygen evolution and CO2 uptakemay not be tightly coupled. In vitro and in situ O2 productionestimates were compared in four diurnal (dawn to dusk) experimentsin Lake Huron. In situ production estimates were determinedby measuring water-mass oxygen changes and oxygen transfer acrossthe air-water interface. In situ production estimates were approximatelytwice in vitro production estimates for both surface mixinglayer and deep chlorophyll layer comparisons. The differencebetween estimates was attributable to containment effects manifestin 13–16 h bottle incubations. Short-term (1–2 h)in vitro production was also compared to diurnal in vitro production.Rates of short-term production were {small tilde}1.6 times higherthan rates of diurnal production, suggesting that short-termin vitro production experiments may provide reasonable estimatesof in situ primary production.  相似文献

The ndhB and psaE mutants of the cyanobacteriumSynechocystis sp. PCC 6803 are partly deficient in PSI-drivencyclic electron transport. We compared photoinhibition in thesemutants to the wild type to test the hypothesis that PSI cyclicelectron transport protects against photoinhibition. Photoinhibitorytreatment greatly accelerated PSI cyclic electron transportin the wild type and also in both the mutants. The psaEmutant showed rates of PSI cyclic electron transport similarto the wild type under all conditions tested. The ndhBmutant showed much lower rates of PSI cyclic electron transportthan the wild type following brief dark adaptation but exceededwild type rates after exposure to photoinhibitory light. Thewild type and both mutants showed similar rates of photoinhibitiondamage and photoinhibition repair at PSII. Photoinhibition atPSI was much slower than at PSII and was also similar betweenthe wild type and both mutants, despite the known instabilityof PSI in the psaE mutant.
