Previous studies indicate that most natural isolates of Escherichia coli are resistant to most or all colicins (antibiotics produced by E. coli) when assessed in the laboratory. Additionally, resistance to different colicin types appears to arise in a nonindependent manner. One possible mechanism to explain this nonindependence is pleiotropy: Multiple resistances are selected after exposure to a single colicin. This study, which was designed to address the role of pleiotropy in the generation of colicin resistance, revealed that 96% of colicin resistant mutants were resistant to two or more colicins. Mutational class was important because putative translocation mutants (Tol pathway mutants) resisted fewer colicins than putative receptor mutants. To determine whether colicin resistance is costly, the effects of colicin resistance mutations on maximal growth rate in a rich medium were also examined. Relative to the sensitive ancestor, translocation mutations lowered maximal growth rates by 17%, whereas putative receptor mutations did not significantly lower growth rates. Thus, when nutrients are abundant, the most advantageous forms of colicin resistance may not impose a cost. The ecological consequences of pleiotropic colicin resistance could involve population cycling between colicin sensitivity and resistance. Additionally, if the cost of resistance depends on the environment, ecological diversification could result.  相似文献

We have extended our previous study on the suppression of frameshift mutants by Escherichia coli thyA mutants by assaying suppression of 15 rIIB frameshift mutants of bacteriophage T4 on one of our suppressing thyA mutant strains. The majority of insertion mutants were suppressible, whereas none of the deletion mutants tested was suppressible. Frameshift suppression could be inhibited by adding thymidine to the assay medium, but was not affected by the presence of a restrictive rpsL mutation in the host strain. We suggest that the frameshift suppression event occurs at a nonsense codon generated by the frameshift mutation.  相似文献

Bacteriol. 84:1303-1312. 1962.-Recombinant analysis and interrupted mating procedures, in conjunction with molecular hybridization experiments, demonstrated that the genetic homology between Escherichia and Salmonella is incomplete. The most likely explanation of this incomplete homology is imperfect pairing between the deoxyribonucleic acid molecules of the two species. Despite the inhomologies, there is ample evidence that the order and distance of the genetic characters on the Salmonella chromosome are identical to those of Escherichia.  相似文献

1. The rate of inactivation of an anti-coli phage by filtrates of cultures of the homologous bacteria has been studied. 2. The inactivation rate at 37°C. is proportional to phage concentration and filtrate concentration. 3. At 0°C. the rate of phage inactivation becomes proportional to the square root of the filtrate concentration. 4. A reaction scheme to account for these observations is suggested and discussed. 5. This coli-phage is also inactivated by relatively large concentrations of soluble starch, inulin, gum arabic, and acetylated gum arabic. 6. The inactivation is markedly influenced by salt concentration, being rapid at moderate salt concentrations and slow at high or extremely low salt concentrations. 7. SUMMARY: For the determination of Escherichia coli I in sea water lactose broth frequently gave higher presumptive and confirmed counts than MacConkey's broth. In the presumptive count there were 53 cases where lactose broth gave larger numbers than MacConkey's broth, with 11 equal counts, and only 25 cases with smaller counts ( P =0·00137). After confirmation the corresponding numbers of cases were 39, 10 and 27 ( P =0·134).
In the samples giving most probable numbers (MPN) of less than 100 E. coli /100 ml lactose broth was superior to MacConkey's broth ( P =0·021). At higher MPN values both media were satisfactory, but with highly polluted water MacConkey's broth might give better recoveries due to the suppression of high concentrations of non-coli-aerogenes bacteria.
Samples stored for 24 or 48 hr before testing gave higher presumptive recoveries when examined with lactose broth than with MacConkey's broth, the values for P being 0·028 and 0·0027 respectively.
It appears that lactose broth without inhibitory ingredients could be used with advantage in the examination of sea water.  相似文献
