During the development of female mammals, one of the two X chromosomes is inactivated, serving as a dosage-compensation mechanism to equalize the expression of X-linked genes in females and males. While the choice of which X chromosome to inactivate is normally random, X chromosome inactivation can be skewed in F1 hybrid mice, as determined by alleles at the X chromosome controlling element (Xce), a locus defined genetically by Cattanach over 40 years ago. Four Xce alleles have been defined in inbred mice in order of the tendency of the X chromosome to remain active: Xcea < Xceb < Xcec < Xced. While the identity of the Xce locus remains unknown, previous efforts to map sequences responsible for the Xce effect in hybrid mice have localized the Xce to candidate regions that overlap the X chromosome inactivation center (Xic), which includes the Xist and Tsix genes. Here, we have intercrossed 129S1/SvImJ, which carries the Xcea allele, and Mus musculus castaneus EiJ, which carries the Xcec allele, to generate recombinant lines with single or double recombinant breakpoints near or within the Xce candidate region. In female progeny of 129S1/SvImJ females mated to recombinant males, we have measured the X chromosome inactivation ratio using allele-specific expression assays of genes on the X chromosome. We have identified regions, both proximal and distal to Xist/Tsix, that contribute to the choice of which X chromosome to inactivate, indicating that multiple elements on the X chromosome contribute to the Xce.  相似文献

Female mammals are functional mosaics of their parental X-linked gene expression due to X chromosome inactivation (XCI). This process inactivates one copy of the X chromosome in each cell during embryogenesis and that state is maintained clonally through mitosis. In mice, the choice of which parental X chromosome remains active is determined by the X chromosome controlling element (Xce), which has been mapped to a 176-kb candidate interval. A series of functional Xce alleles has been characterized or inferred for classical inbred strains based on biased, or skewed, inactivation of the parental X chromosomes in crosses between strains. To further explore the function structure basis and location of the Xce, we measured allele-specific expression of X-linked genes in a large population of F1 females generated from Collaborative Cross (CC) strains. Using published sequence data and applying a Bayesian “Pólya urn” model of XCI skew, we report two major findings. First, inter-individual variability in XCI suggests mouse epiblasts contain on average 20–30 cells contributing to brain. Second, CC founder strain NOD/ShiLtJ has a novel and unique functional allele, Xceg, that is the weakest in the Xce allelic series. Despite phylogenetic analysis confirming that NOD/ShiLtJ carries a haplotype almost identical to the well-characterized C57BL/6J (Xceb), we observed unexpected patterns of XCI skewing in females carrying the NOD/ShiLtJ haplotype within the Xce. Copy number variation is common at the Xce locus and we conclude that the observed allelic series is a product of independent and recurring duplications shared between weak Xce alleles.  相似文献

X chromosome inactivation (XCI) is the mammalian mechanism of dosage compensation that balances X-linked gene expression between the sexes. Early during female development, each cell of the embryo proper independently inactivates one of its two parental X-chromosomes. In mice, the choice of which X chromosome is inactivated is affected by the genotype of a cis-acting locus, the X-chromosome controlling element (Xce). Xce has been localized to a 1.9 Mb interval within the X-inactivation center (Xic), yet its molecular identity and mechanism of action remain unknown. We combined genotype and sequence data for mouse stocks with detailed phenotyping of ten inbred strains and with the development of a statistical model that incorporates phenotyping data from multiple sources to disentangle sources of XCI phenotypic variance in natural female populations on X inactivation. We have reduced the Xce candidate 10-fold to a 176 kb region located approximately 500 kb proximal to Xist. We propose that structural variation in this interval explains the presence of multiple functional Xce alleles in the genus Mus. We have identified a new allele, Xcee present in Mus musculus and a possible sixth functional allele in Mus spicilegus. We have also confirmed a parent-of-origin effect on X inactivation choice and provide evidence that maternal inheritance magnifies the skewing associated with strong Xce alleles. Based on the phylogenetic analysis of 155 laboratory strains and wild mice we conclude that Xcea is either a derived allele that arose concurrently with the domestication of fancy mice but prior the derivation of most classical inbred strains or a rare allele in the wild. Furthermore, we have found that despite the presence of multiple haplotypes in the wild Mus musculus domesticus has only one functional Xce allele, Xceb. Lastly, we conclude that each mouse taxa examined has a different functional Xce allele.  相似文献

Schizopygopsis younghusbandi younghusbandi is an endemic species whose distribution is restricted to the middle reaches of the Yarlung Zangbo River, being one of the most important commercial fishes in this area. Age and growth of 606 specimens captured between October 2002 and April 2005 were studied. The range in standard length (L) was 65.7–387.3 mm and total weight (W) was 3.3–772.0 g. The relationship between L and W was W = 0.000909L2.2493 for males and W = 0.000259L2.4781 for females. Age, determined from anal scales and lapillus otoliths, ranged from 3 to 18 years. The parameters of von Bertalanffy growth functions, estimated by back-calculated length, were L = 442.7mm  LL_\infty = 442.7mm\;L, k = 0.0738 year−1 and t 0  = −1.4 year for males, and L = 471.4mm  LL_\infty = 471.4mm\;L, k = 0.0789 year−1 and t 0 = 0.2 year for females. Males and females exhibited statistically significant differences in growth. χ 2-test indicated that von Bertalanffy growth functions could well describe the growth of S. y. younghusbandi. The longevities were 39.2 and 38.2 years for males and females, respectively. Growth inflexion points were 10.2 and 12.0 years for males and females, respectively, but 84.8% of the captures were at the smaller ages. So conservation and management schemes for this population should be considered urgently. In addition, we found that populations from the upstream of the Lhasa River, the downstream of the Lhasa River and the middle reaches of the Yarlung Zangbo River showed statistically significant differences in growth patterns.  相似文献

The growth rate, reproductive aspects, and natural mortality of chimaeras and ratfish are poorly known. In this study, life-history parameters for cockfish Callorhinchus callorhynchus (Holocephali—Callorhinchidae) are estimated, which is an important fish resource exploited in Chile. Specimens were sampled from the artisanal fishery captures, from November 2006 to November 2007. The standard length (SL) of males fluctuated between 20 and 62 cm, and between 21 and 70 cm for females. Von Bertalanffy growth parameters were estimated through length-frequency data analysis using MULTIFAN. The length-weight relationship and von Bertalanffy growth parameters were significantly different for males and females, as well as the length at 50% maturity. For males a model with 5 age-classes was the best, with asymptotic length L  = 52 cm SL, growth coefficient K = 0.473 yr−1, and age at length zero t 0 = −0.690 yrs. For females the best model was represented by 10 age-classes (L  = 70.3 cm SL, K = 0.193 yr−1, t 0 = −1.158 yrs) in the length-frequency data sets. Length at 50% maturity of males was estimated in 43.7 cm SL, and in 50.2 cm SL for females. The natural mortality rate fluctuated between 0.42 and 0.82 yr−1 for males and between 0.12 and 0.37 yr−1 for females, depending upon the method used. It is concluded that C. callorhynchus is a species with life-history parameters significantly different between males and females, and such differences should be taken into account in future population dynamics analysis.  相似文献

 The chromosomes of 31 species of Passiflora, distributed throughout the subgenera Astrophea, Calopathanthus, Distephana, Dysosmia, Passiflora, Plectostemma and Tacsonia were analysed. Three different karyotypes were observed: 2n = 12, 24, 36; 2n = 18, 72 and 2n = 20. The karyotype of these species was almost always constituted of metacentric and submetacentric chromosomes with variable karyotype symmetry. In the group with x = 6, represented by the subgenus Plectostemma, six diploid species with 2n = 12, one tetraploid with 2n = 24 (P. suberosa) and an intraspecific polyploid with 2n = 12, 36 (P. misera) were analysed. P. pentagona (subgenus Astrophea) may also be included in this karyological group since it presents 2n = 24 and may be of polyploid origin, with x = 6. The interphase nuclei in this group were areticulate, except those of P. morifolia and P. pentagona with semi-reticulate characteristics. Two small terminal heterochromatic blocks, positive for chromomycin A3, were identified in the largest chromosome pair of P. capsularis and P. rubra, species very closely related, while P. tricuspis displayed four chromosomes with proximal blocks. In the group with x = 9, represented mainly by subgenus Passiflora, 20 species with 2n = 18 and one with 2n = 72 were studied. They presented chromosomes larger than those species with x = 6 and interphase nuclei of semi-reticulate type, except for P. mixta with areticulate nuclei. Four terminal CMA+ blocks were observed in P. edulis, six blocks in P. caerulea and P. racemosa, while five blocks were observed in the single P. amethystina plant analysed. P. foetida (subgenus Dysosmia), the only species with 2n = 20, exhibited six chromosomes with CMA+ blocks and interphase nuclei of the areticulate type. The meiotic analysis of representatives of the three groups (P. foetida, P. suberosa, P. cincinnata and P. racemosa) always presented regular pairing and regular chromosome segregation, except in P. jilekii where a tetravalent was observed. The analysis of the chromosome variation within the genus and the family suggests that the base number of Passiflora may be x1 = 6 or x1 = 12, whereas x2 = 9 is only an important secondary base number. ETA subtype selective antagonists constitute a novel and potentially important class of agents for the treatment of pulmonary hypertension, heart failure, and other pathological conditions. In this paper, 60 benzodiazepine derivatives displaying potent activities against ETA and ETB subtypes of endothelin receptor were selected to establish the 3D-QSAR models using CoMFA and CoMSIA approaches. These models show excellent internal predictability and consistency, external validation using test-set 19 compounds yields a good predictive power for antagonistic potency. Statistical parameters of models were obtained with CoMFA-ETA (q 2 = 0.787, r 2 = 0.935, r 2 pred  = 0.901), CoMFA-ETB (q 2 = 0.842, r 2 = 0.984, r 2 pred  = 0.941), CoMSIA-ETA (q 2 = 0.762, r 2 = 0.971, r 2 pred  = 0.958) and CoMSIA-ETB (q 2 = 0.771, r 2 = 0.974, r 2 pred  = 0.953) respectively. Field contour maps (CoMFA and CoMSIA) corresponding to the ETA and ETB subtypes reflects the characteristic similarities and differences between these types. The results of this paper provide valuable information to facilitate structural modifications of the title compounds to increase the inhibitory potency and subtype selectivity of endothelin receptor.  相似文献

Spatial and temporal interactions among individual members of populations can have direct applications to habitat management of mountain lions (Puma concolor). Our objectives were to evaluate home range overlap and spatial/temporal use of overlap zones (OZ) of mountain lions in Arizona. We incorporated spatial data with genetic analyses to assess relatedness between mountain lions with overlapping home ranges. We recorded the space use patterns of 29 radio-collared mountain lions in Arizona from August 2005 to August 2008. We genotyped 28 mountain lions and estimated the degree of relatedness among individuals. For 26 pairs of temporally overlapping mountain lions, 18 overlapped spatially and temporally and eight had corresponding genetic information. Home range overlap ranged from 1.18% to 46.38% ( [`(x)] = \text24.\text43 \overline x = {\text{24}}.{\text{43}} , SE = 2.96). Male–male pairs were located within 1 km of each other on average, 0.04% of the time, whereas male–female pairs on average were 3.0%. Two male–male pairs exhibited symmetrical spatial avoidance and two symmetrical spatial attractions to the OZ. We observed simultaneous temporal attraction in three male–male pairs and four male–female pairs. Individuals from Tucson were slightly related to one another within the population (n = 13, mean R = 0.0373 ± 0.0151) whereas lions from Payson (n = 6, mean R = −0.0079 ± 0.0356) and Prescott (n = 9, mean R = −0.0242 ± 0.0452) were not as related. Overall, males were less related to other males (n = 20, mean R = −0.0495 ± 0.0161) than females were related to other females (n = 8, mean R = 0.0015 ± 0.0839). Genetic distance was positively correlated with geographic distance (r 2 = 0.22, P = 0.001). Spatial requirements and interactions influence social behavior and can play a role in determining population density.  相似文献

The dynamics of genome modification that occurred from the initial hybridization event to the stabilization of allopolyploid species remains largely unexplored. Here, we studied inheritance and expression of rDNA loci in the initial generations of Brassica napus allotetraploids (2n = 38, AACC) resynthesized from Brassica oleracea (2n = 18, CC) and B. rapa (2n = 20, AA) and compared the patterns to natural forms. Starting already from F1 generation, there was a strong uniparental silencing of B. oleracea genes. The epigenetic reprogramming was accompanied with immediate condensation of C-genome nucleolar organizer region (NOR) and progressive transgeneration hypermethylation of polymerase I promoters, mainly at CG sites. No such changes were observed in the A-genome NORs. Locus loss and gains affecting mainly non-NOR loci after the first allotetraploid meiosis did not influence established functional status of NORs. Collectively, epigenetic and genetic modifications in synthetic lines resemble events that accompanied formation of natural allopolyploid species.  相似文献

The effect of para-fluorophenylalanine (PFP) on the production of trisomic plants of Agave tequilana Weber var. Azul produced through somatic embryogenesis was investigated. Normal diploid plants with 2n = 2x = 60 were obtained in the control treatment and with 4 mg L−1 PFP exposure, while use of 8 and 12 mg L−1 PFP led to production of trisomics with 2n = 2x = 61. Normal diploid plants showed a bimodal karyotype with five pairs of large chromosomes and 25 pairs of small chromosomes. Trisomic plants also had a bimodal karyotype with a group of three chromosomes in position five of the chromosome set. More than 13 homologous chromosome pairs exhibited structural changes. Differences in chromosome arm ratio (long arm/short arm) were also found in eight chromosome pairs; all these aberrations in the chromosome complement of trisomic plants were probably caused by inversions, deletions, and/or duplications produced by high concentrations of PFP. The gross chromosome structural changes and the presence of a single extra chromosome could have been induced by the effect of PFP on the mitotic spindle by inducing nondisjunction of sister chromatids, resulting in hyperploids (2n + x) and hypoploids (2nx). Flow cytometric analysis of nuclear DNA content was performed using nuclei isolated from young leaves of normal and trisomic plants. The 2C DNA content of 8.635 pg (1Cx = 4,223 Mbp of trisomic plants was different (p < 0.001) than that of normal plants (2C DNA = 8.389 pg (1Cx = 4,102 Mbp). The difference in genome size was correlated with the large structural changes in the trisomic plant genomes.  相似文献

Several growth hypotheses have been tested to investigate why males of the sequential hermaphrodite, Mediterranean rainbow wrasse, Coris julis (Linnaeus, 1758), are relatively larger than females of the same age. Individual growth trajectories were estimated to test these hypotheses. A good linear relationship between otolith size and body size was observed (r 2  = 0.71, n = 609), thus, past somatic growth of any specific fish can be inferred from the longitudinal data described by the width of annual increments in the otolith. These data were successfully analyzed by a non-linear mixed-effect model (von Bertalanffy growth model) using a Bayesian approach. The results obtained suggest that Mediterranean rainbow wrasse secondary males are relatively larger than females because 1) fish that change sex are already the larger individuals in their age group (specifically those with higher growth rate, k secondary males  = 0.199 and k females  = 0.161) and 2) they experience a growth spurt after sex change. The differences in growth observed in this species and in other protogynous hermaphrodites could be related to differences in social organization, which, in turn, are related to differences in the sex change mechanisms.  相似文献

Efficient plant regeneration is essential for successful transformation and in vitro polyploidy induction in mulberry. A high frequency (80%) of plant regeneration from hypocotyls occurred under in vitro conditions in mulberry (Morus multicaulis Poir.). We identified three key factors for enhancing successful regeneration based on earlier work: (1) hypocotyl position, (2) the combination and concentration of growth regulators, and (3) the addition of AgNO3. The highest frequency of shoot regeneration was achieved using hypocotyl segments, which are proximal to apical meristems, and the optimal culture conditions were Murashige and Skoog’s (MS) (Murashige and Skoog, 1962) basal medium supplemented with 3.0 mg l−1 6-benzylamino purine, 0.3 mg l−1 indole-3-acetic acid, 0.1% polyvinypyrrolidone, and 1.0 mg/l silver nitrate (AgNO3) under subdued light at 25 ± 2°C. Treating the shoots with 0.2% colchicine (dipping for 72 h) resulted in a 14% tetraploid frequency, whereas a 20% tetraploid frequency resulted from using a 0.25% colchicine (dripping for 5 d) treatment, as determined by chromosome number counts. The diploid plant chromosome number was 28 (2n = 2x = 28) and that of tetraploid plants was 56 (2n = 4x = 56). Regenerated shoots rooted easily in 8–10 d using half-strength basal MS medium with 0.5 mg l−1 indole-3-butyric acid and were successfully established in the soil.  相似文献

High levels of genetic variation enable species to adapt to changing environments and provide plant breeders with the raw materials necessary for artificial selection. In the present study, six AFLP primer pairs were used to assess the genetic diversity of Desmodium triflorum (L.) DC. from 12 populations in South China. A high percentage of polymorphic loci (P = 76.16%) and high total gene diversity (H T = 0.310) were found, indicating that the genetic diversity of D. triflorum is high at the species level. Genetic diversity was also relatively high at the population level (P = 55.85%, H e = 0.230). The coefficient of gene differentiation among populations (G ST) was 0.255, indicating that while most genetic diversity resided within populations, there was also considerable differentiation among populations. AMOVA also indicated 24.29% of the total variation to be partitioned among populations (ΦST = 0.243). UPGMA clustering analysis based on genetic distances showed that the 12 populations could be separated into three subgroups: an eastern, a western, and a central-southern subgroup. However, a Mantel test revealed no significant correlation (r = 0.286, p = 0.983) between the geographical distances and genetic distances separating these populations; mountain barriers to gene flow and human disturbance may have confounded these correlations. The present study has provided some fundamental genetic data that will be of use in the exploitation of D. triflorum.  相似文献

A preliminary analysis of 175 specimens of the white-streaked grouper, Epinephelus ongus (Serranidae), was undertaken to determine life history characteristics of the species. Sagittal otoliths, stomachs, and a subsample of gonads were removed to determine age at length, diet, and reproductive strategy. The von Bertalanffy growth equation was used to describe growth in this species and yielded the growth parameters L = 438.3, K = 0.04334, and t0 = −8.752. Fish ranged in age from 1 to 20 years. Diet was consistent with other serranid species and included crabs, shrimps, octopi, and fishes. Based on a very limited number of specimens (n = 12), the larger size and older age of males compared to females suggests that E. ongus may be a protogynous hermaphrodite.  相似文献

Twenty-eight specimens of the large notothenioid Dissostichus mawsoni were dissected after capture on a set line near the southern limit of its range in McMurdo Sound, Antarctica. Total length (L T) averaged 127.3 cm (range 90–162 cm) and weight (W ) was 26.7 kg (range 10.4–60.3 kg). The length-weight relationship was W=3.44×10−5 L T 2.85 (n=28, r 2=0.96). Subcutaneous lipid thickness averaged 2.6 mm and showed no difference due to sex, but a significant weak relationship to W and L T. Hepatosomatic index (I H) was 1.6% for females and 1.7% for males; gonadosomatic index (I G) was 0.9% for females and 0.2% for males. Although specimens were large and sexually mature, the histology indicated that the gonads of this November sample were in a resting stage. Testes lacked spermatids and spermatozoa. Oocytes were in the previtellogenic stage (91.2%) or in the first stage of vitellogenesis (8.8%). A few atretic oocytes and empty follicles indicated that some females in this sample had spawned previously. A summary of the life-cycle is also presented. Accepted: 27 September 1999  相似文献

We studied the energy flow from C3 and C4 plants to higher trophic levels in a central Amazonian savanna by comparing the carbon stable-isotope ratios of potential food plants to the isotope ratios of species of different consumer groups. All C4 plants encountered in our study area were grasses and all C3 plants were bushes, shrubs or vines. Differences in δ13C ratios among bushes ( = −30.8, SD = 1.2), vines ( = −30.7, SD = 0.46) and trees ( = −29.7, SD = 1.5) were small. However the mean δ13C ratio of dicotyledonous plants ( = −30.4, SD = 1.3) was much more negative than that of the most common grasses ( = −13.4, SD = 0.27). The insect primary consumers had δ13C ratios which ranged from a mean of −29.5 (SD = 0.47) for the grasshopper Tropidacris collaris to a mean of −14.7 (SD = 0.56) for a termite (Nasutitermes sp.), a range similar to that of the vegetation. However, the common insectivorous and omnivorous vertebrates had intermediate values for δ13C, indicating that carbon from different autotrophic sources mixes rapidly as it moves up the food chain. Despite this mixing, the frogs and lizards generally had higher values of δ13C ( = −21.7, SD = 1.6;  = −21.9, SD = 1.8, respectively) than the birds ( = −24.8, SD = 1.8) and the only species of mammal resident in the savanna ( = −25.4), indicating that they are generally more dependent on, or more able to utilise, food chains based on C4 grasses. Spotted Halibut (Verasper variegatus) is a commercially important marine fish species. In the present study, to isolate sex-specific markers in Spotted Halibut, we screened the genomes of Spotted Halibut by AFLP technique with 64 different primer combinations. Two primer combinations, MseI-CAG/EcoRI-ACC and MseI-CAT/EcoRI-AGG, produced a female-specific fragment in all females (n = 88) and in no males (n = 60, except 3 individuals), respectively. Both fragments were excised from the gel, cloned and characterized. The first fragment (named VevaF533) was 533 bp long, while the length of the second one (named VevaF218) was 218 bp. The two sequences showed no similarity to each other, and to the known sequences existing in the GenBank database using BLASTn. Cross-species amplification showed that the marker VevaF218 is a species-specific marker which is present in Spotted Halibut females but absent in Barfin Flounder (Verasper moseri). So this marker could be used for discriminating unambiguously between Spotted Halibut females and Barfin Flounder. Examination of the patterns of inheritance of VevaF218 in an interspecific hybrid family (V. variegatus ♂×V. moseri ♀) showed a female-specific pattern of inheritance from mother to daughter, implying that the marker VevaF218 is located on the female sex chromosome. This study provides a reliable AFLP-based genetic sexing of Spotted Halibut that could be useful for genetic mapping of the sex chromosomes and identification of sex-linked genes.  相似文献

We assessed genetic variation in a total of 22 populations of the tree species Cordia africana Lam. in Ethiopia and analyzed the country-wide impact of population history, forest disturbance and alteration of land use on extant intraspecific diversity. Amplified Fragment Length Polymorphisms (AFLPs) and chloroplast microsatellite markers were investigated. The analyses of the AFLP data revealed high diversity in all investigated populations: the percentage of polymorphic loci (PPL) ranged from 62.2% to 92.2% and Nei's gene diversity from 0.220 to 0.320 within the populations. The mean PPL and the mean diversity within populations were 85.7% and 0.287, respectively. The analysis of molecular variance revealed a moderate level of differentiation (ΦST = 0.07, p < 0.001) among the populations. The Mantel test proved a significant but low correlation (r = 0.31, p < 0.001) between the geographic distance and the genetic differentiation matrices. Only three different cpDNA haplotypes were observed; no more than two haplotypes were found in any population. The dominant haplotype with an overall frequency of 81% was observed in all populations. The level of differentiation among the populations was comparatively low at chloroplast DNA (G ST = 0.18, R ST, N ST = 0.22). The observed patterns and levels of genetic variation within and among the populations indicate that efficient gene flow via pollen and seed is likely to be the main factor contributing to the maintenance of genetic diversity in natural and disturbed conditions.  相似文献
