This study investigated the effect on follicular and embryo development of increasing quickly degradable nitrogen (QDN) intake in lactating Holstein dairy cows. Forty mature post-partum cows were fed one of two diets for a minimum of 10 weeks, starting 10 days before first insemination. The Control diet was a high production dairy ration. The High QDN diet comprised the Control ration plus 250 g urea/head/day. Both diets were formulated to ensure that the energy requirements of the cows were satisfied. The High QDN treatment resulted in a significant increase in milk urea, plasma urea and plasma ammonia concentrations. The highest plasma urea (8.2 mmol/l) and ammonia concentrations (120 micromol/l) were recorded within 7 days of the urea supplement being added to the diet. There was no effect of diet on plasma progesterone or glucose concentration. There was also no significant effect of treatment on follicular development or embryo growth. The results from this study suggest that the lactating cow can adapt to increased intakes of QDN if the increase starts at least 10 days before insemination.  相似文献

The objective of this experiment was to examine the effects of an excess intake of quickly degradable nitrogen (QDN) on metabolic and reproductive parameters in lactating dairy cows. Twenty-two lactating dairy cows were fed a total mixed ration once daily. The control diet was a typical ration for high producing cows in the UK (CP = 17.5%, ME = 11.8 MJ/kg DM). The cows were randomly divided into two groups, control diet (control; n = 12) and excess QDN diet (QDN; n = 10). The QDN group was fed an additional 250 g of urea per cow per day, from 10 days before insemination (day 0) until the end of the experiment, 17 days after the second insemination. Ten days before insemination, a synchronized oestrus was induced and the cows inseminated twice, 48 and 72 h after synchronization, with commercial frozen semen from a single sire. Ovaries were scanned using B-mode ultrasonography 10 days before insemination and then daily from 3 days before insemination. Eighteen of the cows (9 control and 9 QDN) were sampled more intensively to determine the pulsatile pattern of secretion of luteinizing hormone (LH) and growth hormone (GH). Cows were slaughtered 17 days after insemination, the reproductive tracts recovered and flushed to retrieve embryos. The excess QDN diet resulted in elevated (P < 0.05) plasma urea concentrations 3 days after starting urea feeding and these were maintained until the end of the experiment. However, the excess QDN diet did not significantly affect daily milk production or plasma concentrations of insulin and IGF-I. The QDN treatment did not significantly affect pulsatile patterns of secretion of LH and GH or the number of small (< 0.5 cm diameter) and medium to large follicles (> 0.5 cm diameter). Twenty cows ovulated following synchronization (control 11/12; QDN 9/10). There were no significant differences between the control and the QDN groups in the peak concentrations of oestradiol during the follicular phase or in the post-ovulatory pattern of plasma and milk progesterone secretion. Embryos and/or foetal membranes were recovered from 10 cows (5 control and 5 QDN). The results of the current study show that feeding excess QDN, as urea, for 27 days commencing 10 days before insemination had no effect on reproductive or metabolic hormonal parameters. Ovulation and the formation and function of the post-ovulatory corpus luteum were also unaffected by excess QDN. These data suggest that the harmful effects of excess intakes of QDN on fertility occur after 17 days following ovulation.  相似文献

High plasma urea nitrogen (PUN) concentrations are associated with decreased fertility in lactating dairy cows. Our objective was to evaluate the quality of embryos flushed from superovulated lactating cows having moderate or high PUN concentrations. Subsequent embryo survival was determined after transfer to recipient heifers with either low or high PUN. Lactating Holstein dairy cows (n = 23; 50-120 days in milk) were randomly assigned to one of two diets designed to result in moderate or high PUN concentrations (15.5 +/- 0.7 and 24.4 +/- 1.0 mg/dl, respectively; P < 0.001) and were fed for 30 days before embryo flushing and recovery. Embryos (n = 94) were evaluated morphologically, frozen and subsequently transferred into synchronized virgin heifers that were fed one of two diets designed to result in either low or high PUN concentrations (7.7 +/- 0.9 and 25.2 +/- 1.5 mg/dl, respectively; P < 0.001; 2 x 2 factorial design). The number, quality and stage of development of recovered embryos were similar for cows with moderate or high PUN. Transfer of embryos from moderate PUN donor cows resulted in a higher pregnancy rate (35%; P < 0.02) than the transfer of embryos from high PUN donor cows (11%). Pregnancy rate was not affected by either recipient diet or the interaction of donor and recipient diets (P > 0.05). These results indicate that high PUN concentrations in lactating dairy cows decrease embryo viability through effects exerted on the oocyte or embryo before recovery from the uterus 7 days after insemination.  相似文献

Initiation of long-term treatment with rbST (Posilac, Monsanto, St. Louis, MO) coincident with first insemination increased pregnancy rates in dairy cattle, but neither the efficacy of using only the initial injection, nor its effects on retention of pregnancy are known. Lactating dairy cows, dairy heifers, and lactating beef cows were assigned at random to treatment (rbST) or control. Dairy cows, dairy heifers, and beef cows received 500 mg rbST (n = 48, 35, 137 inseminations, respectively) at artificial insemination or were left untreated (n = 62, 33, 130 inseminations, respectively). Pregnancy was diagnosed by ultrasonography at 28-36 days. Treatment with rbST at insemination improved conception rates in dairy cows (60.4% versus 40.3%; P < 0.05), but not in dairy heifers or beef cows. Conception rates did not differ in dairy cows at < or =100 days in milk (DIM), but were improved in cows treated with rbST after 100 DIM (64.3% versus 25.8%; P < 0.05). Retention of pregnancy to approximately 60 days and sizes of CL, diameter of follicles > or =5 mm, and crown-rump lengths of embryos were not affected by treatment. The second objective was to examine the effects of rbST at insemination on birth weight and post-natal calf growth in beef cows. However, birth and weaning weights of beef calves were not affected by treatment. In conclusion, a single treatment with rbST at insemination increased conception rates in dairy cows, specifically in those >100 DIM.  相似文献

Wet corn gluten feed (WCGF) is a high moisture feed containing rapidly digestible, non-forage fiber and protein. The objective of this study was to investigate the effect of substituting WCGF and corn stover for alfalfa hay in total mixed ration (TMR) silage on lactation performance and nitrogen balance in dairy cows. Nine multiparous Holstein dairy cows (BW = 532 ± 28.9 kg and day in milk = 136 ± 5.6 d; mean ± SD) were used in a replicated 3 × 3 Latin square design with 21-d periods (14 d of diet adaption and 7 d of sample collection). Groups were balanced for parity, day in milk, and milk production and consumed one of three treatment diets during each period. The treatment diets were fed as TMR and contained similar concentrate mixtures and corn silage but different proportions of roughage and WCGF. The three treatments were: (1) 0% WCGF, 0% corn stover, and 22.1% alfalfa hay (0% WCGF); (2) 6.9% WCGF, 3.4% corn stover, and 11.8% alfalfa hay (7% WCGF); and (3) 13.3% WCGF, 4.9% corn stover, and 3.9% alfalfa hay (13.3% WCGF). Compared to the 0% WCGF diet, the cows fed the 7% and 13.3% WCGF diets had a higher milk yield and concentration of milk fat, protein, lactose, and total solids. Effective degradability of DM was higher in the cows fed the 7% and 13.3% WCGF diets than it was with the 0% WCGF diet. Cows fed the 13.3% WCGF had a higher CP effective degradability and a lower rumen undegraded protein than cows fed the 0% WCGF diet. The concentration of ruminal volatile fatty acids and ammonia-N was higher in cows fed the 7% and 13.3% WCGF diets than cows fed the 0% WCGF diet. The fecal N was lower in cows fed the 7% and 13.3% WCGF diets than it was in cows fed the 0% WCGF diet. Milk N secretion and milk N as a percent of N intake were higher in cows fed the 13.3% WCGF diet than cows fed the 0% and 7% WCGF diets. In conclusion, it appears that feeding a TMR silage containing WCGF and corn stover in combination, replacing a portion of alfalfa hay, may improve lactation performance and nitrogen utilization for lactating dairy cows.  相似文献

In a survey on pregnancy rate and embryonic losses in dairy cattle on 6 Israeli farms, cows (n = 78) were divided into 3 groups on the basis of ultrasonography at 21 d post insemination; pregnancy diagnosis at 40 to 50 d post insemination and blood progesterone (P4) levels at 21 d. The groups were either pregnant (P4 level > 1.0 ng/ mL); not pregnant (P4 < 0.5 ng/mL), or showed early embryo loss (P4 > 1.0 ng/mL and the presence of an embryonic vesicle on D 21 but later returned to estrus or were found not pregnant on D 40 to 50). On the day of insemination, peripheral estrogen was significantly higher (P < 0.05) in the early embryo loss group (15.3 +/- 1.1 pg/mL, n = 27) than in pregnant (9.4 +/- 0.6 pg/mL, n = 26) or not pregnant (9.6 +/- 0.7 pg/mL, n = 25) group. The cows on 3 farms which were fed 1 to 2 kg/d of vetch (Vicia sativa), an estrogenic legume, had higher estrogen concentrations on the day of insemination than cows (2 farms) fed other legumes (13.7 +/- 0.64, n = 58 vs 10.7 +/- 0.8 pg/mL, n = 42; P < 0.01). On one of the 3 farms, vetch was replaced with alfalfa after the first year. Following the cessation of vetch feeding the estrogen concentrations in the blood decreased from 32 +/- 5 pg/mL to 14 +/- 2 pg/mL (n = 9). These data suggest that high peripheral estrogen on the day of insemination is associated with early embryonic loss. These data also indicate that estrogen concentrations on the day of insemination can be influenced by diet.  相似文献

Three ruminally cannulated and multicatheterised lactating dairy cows were used to investigate the effect of different supplement strategies to fresh clover grass on urea and short-chain fatty acid (SCFA) metabolism in a zero-grazing experiment with 24-h blood and ruminal samplings. Fresh clover grass was cut every morning and offered from 0800 to 1500 h. Maize silage was fed at 1530 h. The three treatments, arranged in a Latin square, differed by timing of feeding rolled barley and soya-bean hulls relative to fresh clover grass. All diets had the same overall composition. Treatments were soya-bean hulls fed at 0700 h and barley fed at 1530 h (SAM), barley fed at 0700 h and soya-bean hulls fed at 1530 h (BAM), and both soya-bean hulls and barley fed at 1530 h (SBPM). The grass had an unexpectedly low content of crude protein (12.7%) and the cows were severely undersupplied with rumen degradable protein. The treatment effects were numerically small; greater arterial ammonia concentration, net portal flux of ammonia and net hepatic flux of urea during part of the day were observed when no supplementary carbohydrate was fed before grass feeding. A marked diurnal variation in ruminal fermentation was observed and grass feeding increased ruminal concentrations of propionate and butyrate. The net portal fluxes of propionate, butyrate, isovalerate and valerate as well as the net hepatic uptake of propionate, butyrate, valerate and caproate increased after feeding at 0700 h. The hepatic extraction of butyrate showed a relatively large depression with grass feeding with nadir at 1200 to 1330 h. The increased net portal absorption and the decreased hepatic extraction resulted in an approximately six-fold increase in the arterial blood concentration of butyrate. The gut entry rate of urea accounted for 70 ± 10% of the net hepatic production of urea. Saliva contributed to 14% of the total amount of urea recycled to the gut. Urea recycling to the gut was equivalent to 58% of the dietary nitrogen intake. Despite the severe undersupply of rumen degradable protein, the portal-drained viscera did not extract more than 4.3% of the urea supplied with arterial blood. This value is in line with the literature values for cows fed diets only moderately deficient in rumen degradable protein and indicates that cows maximise urea transfer across gut epithelia even when the diet is moderately deficient in rumen degradable protein.  相似文献

The effect of human chorionic gonadotropin (HCG) on corpus luteum (CL) function in lactating dairy cows and its subsequent effect on pregnancy rates achieved with artificial insemination was studied in two parts. In Part 1, four IM injections at intervals of seven days or two IM injections at a 14 day interval of HCG (10,000 IU) were given to two groups of six cows each, groups A and B respectively, beginning on days 9, 10 or 11 of the estrous cycle. Ten control cows were given 10 ml of isotonic saline solution on days 9, 10 or 11. Interestrous intervals were prolonged by an average of 8.1 and 3.4 days for groups A and B respectively over controls. In Part 2, 200 lactating dairy cows on nine farms were assigned, on an alternate basis at insemination, to control or treatment groups to study the effect of CL prolongation via HCG on pregnancy rate. A single injection of HCG (10,000 IU) was given 10, 11 or 12 days after a first or second insemination. Pregnancy rates for control and treated cows were similar (64 101 = 63% and 58 99 = 59% respectively). Interestrous intervals of treated nonpregnant cows were prolonged by approximately five days. Providing additional time for developing embryos to become established in utero by delaying luteolysis did not improve pregnancy rates achieved with artificial insemination in lactating dairy cows.  相似文献

The objective was to determine if a single measurement of plasma insulin-like growth factor-1 (IGF-1) could predict the number of viable embryos obtained from donors and the likelihood of pregnancy in recipients in multiple ovulation and embryo transfer (MOET) programs in cattle. The embryo yields from 101 embryo recoveries were examined in maiden Holstein heifers (n=75) and multiparous Holstein cows (lactating cows n=20, dry cows n=6). Donors were super stimulated with FSH and embryo recovery was done non-surgically 7 days after artificial insemination. Embryos were classified according to the IETS criteria. Pregnancy rates in 100 maiden Holstein heifer recipients were analysed. Recipients were on day 7+/-1 of the estrous cycle at transfer. Pregnancy diagnosis was carried out at day 30 (PD 30) and rechecked at day 60 (PD 60) after transfer. Blood samples from coccygeal vessels taken at the time of embryo recovery (donors) and transfer (recipients) were analysed for IGF-1, insulin, beta-hydroxybutyrate (beta-OHB), non-esterified fatty acids (NEFA), urea and cholesterol. There was a negative correlation between the number of viable embryos and insulin (r=-0.33, P=0.025) in donor heifers. In donor cows, the number of viable embryos was correlated with IGF-1 (r=0.43, P=0.028) and cholesterol (r=-0.43, P=0.027). In recipients, PD30 and PD 60 were not affected by any of the circulating parameters analysed. Insulin, IGF-1 and cholesterol only explained 8.9, 13.9 and 15.8% of the variation in the production of viable embryos, respectively. Several factors affect MOET programs and under the circumstances of the present study the usefulness of hormonal and metabolic profiles as predictors of the outcome of this biotechnology was limited.  相似文献

Improving reproductive performance is one of the most important factors affecting the profitability of dairy herds. This study investigated the effect of feeding a high starch (HS) diet and body condition score (BCS) at calving on blood metabolites, fertility and ovarian function and milk production in Holstein dairy cows. One hundred seventy-four multiparous cows were fed common close-up and early lactation diets during the first 15 days in milk (DIM). Cows were randomly assigned to 1 of 2 experimental diets from 16 until 50 DIM (n = 87 per group); normal starch (228 g/kg diet DM; NS) or HS (270 g/kg diet DM; HS) diets. Each treatment group was further subdivided based on BCS at calving as normal BCS (BCS ⩽ 3.5; normal BCS (NBCS); n = 45) or high BCS (HBCS) (BCS ⩾ 3.75; HBCS; n = 42). A significant difference was detected for increased milk production (47.24 v. 44.55 kg/day) and decreased milk fat (33.93 v. 36.33 g/kg) in cows fed HS or NS, respectively. Plasma glucose and insulin concentrations were significantly higher in cows fed the HS compared to the NS diet. Diets significantly affected DIM at first artificial insemination (AI, 79.51 ± 3.83 v. 90.40 ± 3.83 days for cows fed HS and NS diets, respectively). High BCS groups had greater milk fat content and elevated plasma nonesterified fatty acids (NEFA), β hydroxybutyrate (BHB) and bilirubin concentrations. In general, feeding higher starch diets to normal BCS cows during the first 50 DIM improved productive and reproductive performance of early-lactating dairy cows.  相似文献

Four lactating dairy cows were used in two experiments to study the effects of the roughage to concentrate ratio in the diet on nitrogen balance, plasma urea, urinary urea, milk urea and urinary purine derivatives. The use of the allantoin to creatinine ratio in spot samples of urine as an index of the urinary allantoin excretion was also evaluated. Four isoenergetic and isonitrogenous diets were formulated according to a 2 × 2 factorial arrangement. Factor I was concentrate content. The roughage to concentrate ratios were 65:35 and 35:65 for the high roughage and high concentrate diets, respectively. Factor II was fat content, which was 2.8% and 5.8% for the low and high fat diets, respectively. In Experiment 1 cows were fed diets with low fat content, and in Experiment 2 cows were fed diets with high fat content. In both experiments, diets were fed according to a change-over design. Nitrogen balance was not affected by the treatments. In cows fed high concentrate diets the amount and the proportion of nitrogen excreted in milk, as well as milk production was higher than in cows fed the high roughage diets. In both experiments, as an overall effect, the urea levels in plasma, urine and morning milk were higher, although the total urinary excretion of urea was lower, for the high concentrate diets. Urinary allantoin excretion was higher, although not significantly in Experiment 1, for the high concentrate diets. The allantoin to creatinine ratio in spot samples of urine showed the same pattern as the total allantoin excretion. Urinary creatinine excretion appeared to be affected by the diet.  相似文献

Our objective was to determine the effects of supplementing methionine and choline during the prepartum and postpartum periods on preimplantation embryos of Holstein cows. Multiparous cows were assigned in a randomized complete-block design into four treatments from 21 days before calving to 30 days in milk (DIM). Treatments (TRT) were MET (n = 9, fed the basal diet + rumen-protected methionine at a rate of 0.08% [w:w] of the dry matter [DM], Smartamine M), CHO (n = 8, fed the basal diet + choline 60 g/d, Reashure), MIX (n = 11, fed the basal diet + Smartamine M and 60 g/d Reashure), and CON (n = 8, no supplementation, fed the close-up and fresh cow diets). Cows were randomly reassigned to two new groups (GRP) to receive the following diets from 31 to 72 DIM; control (CNT, n = 16, fed a basal diet) and SMT (n = 20, fed the basal diet + 0.08% [w:w] of the dry matter intake as methionine). An progesterone intravaginal insert (CIDR) device was inserted in all cows after follicular aspiration (60 DIM) and superovulation began at Day 61.5 using FSH in eight decreasing doses at 12-hour intervals over a 4-day period. On Days 63 and 64, all cows received two injections of PGF2α, and CIDR was removed on Day 65. Twenty-four hours after CIDR removal, ovulation was induced with GnRH. Cows received artificial insemination at 12 hours and 24 hours after GnRH. Embryos were flushed 6.5 days after artificial insemination. Global methylation of the embryos was assessed by immunofluorescent labeling of 5-methylcytosine, whereas lipid content was assessed by staining with Nile red. Nuclear staining was used to count the total number of cells per embryo. There was no difference between TRT, GRP, or their interaction (P > 0.05) for embryo recovery, embryos recovered, embryo quality, embryo stage, or cells per embryo. Methylation of the DNA had a TRT by GRP interaction (P = 0.01). Embryos from cows in CON-CNT had greater (P = 0.04) methylation (0.87 ± 0.09 arbitrary units [AU]) than embryos from cows in MET-CNT (0.44 ± 0.07 AU). The cytoplasmic lipid content was not affected (P > 0.05) by TRT or their interaction, but lipid content was greater (P = 0.04) for SMT (7.02 ± 1.03 AU) than that in CNT (3.61 ± 1.20 AU). In conclusion, cows in MET-CNT had embryos with lower methylation, and SMT cows had a higher lipid content than CNT. Methionine supplementation seems to impact the preimplantation embryo in a way that enhances its capacity for survival because there is strong evidence that endogenous lipid reserves serve as an energy substrate.  相似文献

Embryos were collected non-surgically from the tip of one uterine horn of 23 lactating dairy cows on Day 7 of pregnancy. Embryos were classified on the basis of morphological criteria as normal (n = 12) or abnormal (n = 13). Abnormal embryos were further classified as cleavage stage (n = 9) or morula/blastocyst (n = 4). Cows producing an abnormal embryo did not differ in days post partum at oestrus, age or parity from cows producing a normal embryo. Cows with an abnormal morula/blastocyst also did not differ with respect to days post partum at oestrus from cows with abnormal cleavage-stage embryos but cows with an abnormal morula/blastocyst were significantly older and of greater parity than cows with an abnormal cleavage-stage embryo. Hepes-saline-PVP solution (30 ml) was initially infused into the uterine tip, mixed and then withdrawn with a syringe. Analysis of this fluid revealed that the concentrations of glucose, total protein, calcium, magnesium and potassium were significantly higher in the flushings from the uterus of cows with abnormal embryos than from cows with normal embryos and zinc and phosphorus tended to be higher in the uterine flushings of cows with abnormal embryos. Phosphorus, total protein, calcium and magnesium tended to be higher in the flushings from cows with abnormal morulae/blastocysts than from cows with abnormal cleavage-stage embryos. Plasma progesterone did not differ between cows with normal or abnormal embryos or in cows with abnormal morulae/blastocysts or abnormal cleavage-stage embryos. The objective was to compare conception rates to embryo transfer relative to AI, during summer heat stress, in lactating dairy cows. Holstein cows (n = 180; 50 to 120 d postpartum) were allocated randomly to 1 of 3 groups: artificial insemination (AI, n = 84), embryo transfer using either embryos collected from superovulated donors (ET-DON, n = 48), or embryos produced in vitro (ET-IVF, n = 48). Embryos from superovulated donors were frozen in 10% glycerol and were rehydrated in a 3-step procedure, in decreasing concentrations of glycerol in a sucrose medium before transfer. Embryos produced in vitro were frozen in 1.5 M ethylene glycol, thawed and transferred without rehydration. Blood samples were collected from AI and ET recipients on Days 0, 7 and 22 for measurement of progesterone in plasma. Conception rate was estimated for the three groups at Day 22 (progesterone > 1 ng/mL) and confirmed at Day 42 by palpation per rectum. Conception rate estimates at Day 22 did not differ among groups (AI, 60.7%; ET-DON, 60.4%; ET-IVF, 54.2%), but conception rates at Day 42 differed (AI, 21.4%; ET-DON, 35.4%; ET-IVF, 18.8%; AI versus ET: P > 0.10 and ET-DON versus ET-IVF: P < 0.05). In cows considered pregnant at 22 d but diagnosed open at 42 d, the interestrous intervals were 28.8 +/- 2.2, 35.2 +/- 3.5 and 31.6 +/- 2.9 d, respectively, for AI, ET-DON and ET-IVF groups. Transfer of embryos collected from nonheat-stressed superovulated donors significantly increased conception rates in heat stressed dairy cattle. However, transfer of IVF-derived embryos had no advantage over AI. Where appropriate mechanisms are in place to attenuate the effects of heat stress, embryo transfer using frozen-thawed donor embryos increases conception rates.  相似文献

In this study, a linear programming model was developed to maximize the gross margin of milk production by determining the optimal use of different reproductive technologies in a dairy herd. The model has the potential to vary the use of conventional artificial insemination, insemination with X-sorted sperm, and the use of unselected or sex-selected embryo recovery and transfer. Data from Finnish dairy herd recording systems were used to parameterize the model. This paper presents the results of 6 scenarios for a herd size of 60 dairy cows. In the basic scenario, the optimum economic combination for Finnish conditions was to inseminate 10 heifers and 22 cows with unsorted semen, 8 heifers with X-sorted sperm, and to use 20 cows as embryo donors which was the upper constraint for this technique. The embryo donors were inseminated with conventional semen for both embryo production and their subsequent pregnancy. Without restriction on embryo recovery, the optimum combination was to use all heifers as donors of sex-selected embryos and all cows as donors of unselected embryos. It was more profitable to produce female embryos with X-sorted sperm than by sorting embryos. Embryo recipients were not economically justified in any scenario. In practice, the optimal strategy is herd-specific depending on the input costs, output values and the technical success of each reproductive technology in that herd. This single-year linear programming model adequately differentiates between breeding technologies within a herd, but further research is needed to develop dynamic models to consider genetic improvement and herd expansion.  相似文献

Timed embryo transfer (TET) using in vitro produced (IVP) embryos without estrus detection can be used to reduce adverse effects of heat stress on fertility. One limitation is the poor survival of IVP embryos after cryopreservation. Objectives of this study were to confirm beneficial effects of TET on pregnancy rate during heat stress as compared to timed artificial insemination (TAI), and to determine if cryopreservation by vitrification could improve survival of IVP embryos transferred to dairy cattle under heat stress conditions. For vitrified embryos (TET-V), a three-step pre-equilibration procedure was used to vitrify excellent and good quality Day 7 IVP Holstein blastocysts. For fresh IVP embryos (TET-F), Holstein oocytes were matured and fertilized; resultant embryos were cultured in modified KSOM for 7 days using the same method as for production of vitrified embryos. Excellent and good quality blastocysts on Day 7 were transported to the cooperating dairy in a portable incubator. Nonpregnant, lactating Holsteins (n = 155) were treated with GnRH (100 microg, i.m., Day 0), followed 7 days later by prostaglandin F2alpha (PGF2alpha, 25 mg, i.m.) and GnRH (100 microg) on Day 9. Cows in the TAI treatment (n = 68) were inseminated the next day (Day 10) with semen from a single bull that also was used to produce embryos. Cows in the other treatments (n = 33 for TET-F; n = 54 for TET-V) received an embryo on Day 17 (i.e. Day 7 after anticipated ovulation and Day 8 after second GnRH treatment). The proportion of cows that responded to synchronization based on plasma progesterone concentrations on Day 10 and Day 17 was 67.7%. Pregnancy rate for all cows on Day 45 was higher (P < 0.05) in the TET-F treatment than for the TAI and TET-V treatments (19.0 +/- 5.0,6.2 +/- 3.6, and 6.5 +/- 4.1%). For cows responding to synchronization, pregnancy rate was also higher (P < 0.05) for TET-F than for other treatments (26.7 +/- 6.4, 5.0 +/- 4.3, and 7.4 +/- 4.7%). In the TET-F treatment group, cows producing more milk had lower (P < 0.05) pregnancy rates than cows producing less milk. In conclusion, ET of fresh IVP embryos can improve pregnancy rate under heat stress conditions, but pregnancy rate following transfer of vitrified embryos was no better than that following TAI.  相似文献

The objectives of this study were to identify the level and stage of embryonic mortality that occur in dairy cows during hot and cool seasons of the year. Experimental dairy cows, of varying ages, were artificially inseminated with frozen-thawed semen from proven Holstein sires. Females on each dairy unit were then randomly allocated to one of three experimental groups after partitioning by day of artificial insemination, days post partum, parity, and current milk production level. In Group I and Group II, nonsurgical embryo collection was performed on each cow using Dulbecco's phosphate-buffered saline as the flushing medium. Embryos from cows in Group I were collected on Days 6 or 7 post insemination during the hot (n=93) and cool (n=64) seasons. Embryos from cows in Group II were collected on Days 13 or 14 post insemination during the hot (n=97) and cool (n=63) seasons. In Group III, contemporary control cows were also inseminated during the hot (n=106) and cool (n=106) seasons, and fetal heart beat was evaluated via ultrasound between Days 25 and 35 following insemination. Embryo viability decreased (P<0.05) from 59% at Day 7 to 27% at Day 14 in the hot season, but was not decreased during the cool season (52 vs. 60%). Pregnancy rate at Days 25 to 35 was 21% in the hot season, which was less (P<0.05) than the 36% in the cool season. The percentage of unfertilized ova collected in both the hot and cool seasons suggests that fertilization failure was not affected by season of breeding. In summary, embryonic loss after Day 7 of pregnancy appears to be a problem in this hot, dry climate.  相似文献

Recombinant bovine somatotropin (rbST) has been shown to increase follicular growth in cattle and some studies have demonstrated an increase in superovulatory response for rbST-treated cows. Pregnancy rates have also been shown to increase when rbST was administered around the time of insemination or prior to embryo transfer. The application of rbST for the purpose of increasing superovulatory responses of donor cows and increasing pregnancy rates of recipient heifers was tested in a commercial embryo transfer program. In Experiment 1, embryo donor cows (n = 56) underwent three cycles of control superovulation (two before and one after weaning) and subsequently underwent up to four additional superovulations while being treated with either rbST (500 mg sustained-release rbST; Posilac, Monsanto, St. Louis, MO; n = 28) or excipient (control; n = 28) once every 14 days. In Experiment 2, lactating embryo donor cows (n = 37) underwent a control superovulation and then underwent a superovulation while lactating and being treated with either rbST (n = 16) or excipient (n = 21). In Experiment 3, embryo recipient heifers that were being implanted with either in vitro or in vivo produced embryos were treated with either rbST (n = 146) or excipient (n = 143) at the time of embryo transfer. Treatment of non-lactating (Experiment 1) or lactating (Experiment 2) donor cows with rbST during repeated superovulation did not affect the number of corpora lutea, the sum of transferable embryos, degenerate embryos, and unfertilized oocytes, or the number of transferable embryos. Treatment of recipient heifers with rbST (Experiment 3) did not affect pregnancy rates for either in vitro or in vivo produced embryos. We conclude that superovulatory response and pregnancy rates (respectively) are similar to control for rbST-treated cows undergoing repeated superovulations and rbST-treated recipient heifers treated at the time of embryo transfer.  相似文献

In cattle, increasing early embryonic losses are associated with inadequate progesterone concentrations within the first three weeks of pregnancy. The aim of this study was to investigate the complex relationship between early maternal progesterone concentration and embryo development early within the first week of pregnancy, specifically, on day 5 post-oestrus in dairy cows. Twenty Holstein-Friesian cows at the end of lactation were inseminated at oestrus (day 0) and on day 5 post-oestrus cows were slaughtered and the reproductive tract flushed to determine the presence and stage of embryo development. Three cows that had failed to synchronise correctly were excluded from analysis while in the remaining 17 cows 11 (65%) were pregnant with embryos at the morula (n = 3), 9-16 (n = 3) and 8-cell (n = 5) stages of development. No differences in day 5 plasma progesterone concentrations or corpus luteum (CL) size or progesterone content were observed between pregnant (n = 11) and non-pregnant (n = 6) cows. In cows with embryos beyond the 8-cell stage of development (n = 6) plasma progesterone concentration (P < 0.001) and CL weight (P < 0.01) were higher and plasma insulin concentrations lower (P < 0.001) than in cows with 8-cell embryos (n = 5). In addition there was a negative relationship between plasma progesterone and plasma insulin in pregnant cows (R(2) = 0.65; P < 0.005). In cows with an embryo present in the oviduct, oviductal glucose concentrations were lower (P < 0.05) than in cows with no embryo present. These results confirm progesterone is not only directly associated with embryo development, but that it may indirectly modulate embryo development via changes in the oviductal environment. In summary, the association between maternal progesterone concentration and embryo development exists as early as day 5 of pregnancy in dairy cows.  相似文献

Twin pregnancies represent a management problem in dairy cattle since the risk of pregnancy loss increases, and the profitability of the herd diminishes drastically as the frequency of twin births increases. The aim of this study was to monitor the development of 211 twin pregnancies in high producing dairy cows in order to determine the best time for an embryo reduction approach. Pregnancy was diagnosed by transrectal ultrasonography between 36 and 42 days after insemination. Animals were then subjected to weekly ultrasound examination until Day 90 of gestation or until pregnancy loss. Viability was determined by monitoring the embryonic/fetal heartbeat until Day 50 of pregnancy, and then by heartbeat or fetal movement detection. Eighty-six cows (40.8%) bore bilateral and 125 (59.2%) unilateral twin pregnancies. Embryo death was registered in one of the two embryos in 35 cows (16.6%), 33 of them at pregnancy diagnosis. Pregnancy loss occurred in 22 of these cows between 1 and 4 weeks later. Thus, 13 (6.2% of the total animals) cows, carrying one dead of the two embryos, maintained gestation. Total pregnancy loss before Day 90 of pregnancy (mean 69 +/- 14 days) was registered in 51 (24.2%) cows: 7 (8%) of bilateral pregnancies and 44 (35.2%) of unilateral pregnancies, and it was higher (P = 0.0001) for both right (32.4%, 24/74) and left (39.2%, 20/51) unilateral than for bilateral (8.1%, 7/86) twin pregnancies. The single embryo death rate was significantly (P = 0.02) lower for cows with bilateral twins (9.3%, 8/86) than for total cows with unilateral twins (21.6%, 27/125). By way of overall conclusion, embryo reduction can occur in dairy cattle, and the practical perspective remains that most embryonic mortality in twins (one of the two embryos) occurs around Days 35-40 of gestation, the period when pregnancy diagnosis is generally performed and when embryo reduction could be tried.  相似文献
