The effects of taurine supplementation on visual discrimination in mice were examined. Taurine, 2-aminoethane-sulphonic acid, found in high concentrations in the central nervous system of mammals and in human milk, has been shown to be essential for development. Male mice were divided into four groups according to taurine supplementation periods. 1) Lifelong: taurine (400 mg/kg/day) was dissolved in distilled water and provided as drinking water. In the prenatal period, taurine was given via the mother. After weaning mice were administered taurine in drinking water. 2) Pre-weaning: mice were exposed to taurine prior to weaning, 3) Post-weaning: mice were exposed to taurine after weaning. 4) Control: no supplementation of taurine. It was shown that the Lifelong group required a longer period of time to acquire visual discrimination than the Control group. Conversely, in the Post-weaning group, mice learned the task faster than Controls. Visual discrimination learning time in the Pre-weaning group showed no significant difference compared with that in the Control group. From these results, we suggest that the perinatal to early postnatal period is a “sensitive period” where taurine supplementation can result in retardation of learning in later life. At the same time, taurine supplementation after weaning improved visual discrimination learning. Thus, timing of taurine supplementation affected learning.  相似文献

Hypolimnetic anoxic water of Lake Onogawa was subjected to aeration experiments. When the samples were agitated by magnetic stirrers for 24 h, dissolved oxygen increased from 0 to more than 7.6 mg l−1, dissolved iron decreased from 98% to about 5% of the initial total iron, and from 32% to 48% of the dissolved organic carbon (DOC) disappeared. On the other hand, when the anoxic waters were left unstirred, dissolved oxygen increased from 0 to 2.2 mg l−1, dissolved iron decreased from 98% to 31%, and 20% of the DOC disappeared within 48 h. Further 24-h incubation had little effect on the DOC loss, although dissolved oxygen increased to 3.9 mg l−1 and dissolved iron decreased to 5%. These rates of DOC disappearance are too large to be explained by bacterial decomposition. It is quite conceivable that a part of the DOC is coprecipitated with iron(III) precipitates. When Fe(II) in the anoxic hypolimnion is oxidized by autumnal water mixing, probably anoxic water is mixed with aerobic water. The anoxic water must receive oxygen from the aerobic water during this mixing and be simultaneously diluted with the aerobic water. Because the present experimental conditions, especially the stirred one, significantly differ from in situ conditions, the present results are thought to be a potential capacity of DOC coprecipitation.  相似文献

Fully hydrated toads, Bufo bufo were acclimated to a simulated terrestrial habitat, with access to shelters and water. To get from the shelters to the water, the toads had to walk across the pan of an Ohaus balance and the body weights were recorded on a computer. Toads were placed inside shelters immediately following injection of human angiotensin II (A II), Thr(8)-saralasin, or Ringer's in the dorsal lymph sac, and their behavior was recorded continuously by video surveillance. The injection doses were 1-100 microg/100 g body weight A II and 100 microg/100 g body weight saralasin dissolved in 0.1 ml Ringer's; control animals received the same volume of Ringer's. The latency from injection to the initiation of water absorption behavior (WR) was significantly shorter in both A-II- and saralasin-injected toads, compared to controls. A-II- and saralasin-injected toads also spent significantly more time in the water than controls. The bladder depots when WR was terminated were significantly larger in A-II- or saralasin-injected toads than in controls. The stimulatory action of Thr(8)-saralasin, an antagonist of A II in mammals, on WR behavior in B. bufo suggests differences in receptor structure and/or receptor distribution between amphibians and mammals.  相似文献

The intracellular sodium concentration and intracellular volume of human red blood cells were determined from 23Na and 1H NMR spectra. It is shown that sodium dissolved in the intracellular water has a concentration higher than that previously published. The intracellular sodium concentration measured was 11.4 +/- 3.1 mM. A comparison of different NMR methods used to determine sodium concentration is given.  相似文献

Reverse process of the coprecipitation of dissolved organic matter with Fe(III) precipitates in a lake is reported. Water containing a slight amount of dissolved oxygen from the hypolimnion of Lake Onogawa was sealed in glass bottles, and some changes in the constituents with time were followed. The water sample contained 0.1 mg l−1 dissolved oxygen at the beginning of the experiment, which decreased to 0.0 mg l−1 within 24 h. In response to the depletion of dissolved oxygen, there were increases in dissolved Fe from 8.4 to 11.4 mg l−1 and dissolved organic carbon (DOC) from 5.1 to 6.9 mg l−1 after 72 h. At the beginning of the experiment, more than 2 mg l−1 of insoluble Fe, which was thought to be Fe(III) precipitates, existed in the water samples. When the water samples became anoxic, the preexisted Fe(III) must have been reduced to soluble Fe(II) over time, resulting in the increase of dissolved Fe. Simultaneous with Fe(III) reduction, coprecipitated DOC with Fe(III) must have been released. The reverse process of the coprecipitation of dissolved organic matter with Fe(III) precipitates found in the present study strongly supports in situ coprecipitation of dissolved organic matter with Fe(III) precipitates. The potential for microscale bacterial Fe redox cycling was investigated in microcosms containing ferrihydrite-coated sand and a coculture of a lithotrophic Fe(II)-oxidizing bacterium (strain TW2) and a dissimilatory Fe(III)-reducing bacterium (Shewanella alga strain BrY). The Fe(II)-oxidizing organism was isolated from freshwater wetland surface sediments which are characterized by steep gradients of dissolved O2 and high concentrations of dissolved and solid-phase Fe(II) within mm of the sediment–water interface, and which support comparable numbers (105–106 mL−1) of culturable Fe(II)-oxidizing and Fe(III)-reducing reducing. The coculture systems showed minimal Fe(III) oxide accumulation at the sand-water interface, despite intensive O2 input from the atmosphere and measurable dissolved O2 to a depth of 2 mm below the sand–water interface. In contrast, a distinct layer of oxide precipitates formed in systems containing Fe(III)-reducing bacteria alone. Examination of materials from the cocultures by fluorescence in situ hybridization indicated close physical juxtapositioning of Fe(II)-oxidizing and Fe(III)-reducing bacteria in the upper few mm of sand. Our results indicate that Fe(II)-oxidizing bacteria have the potential to enhance the coupling of Fe(II) oxidation and Fe(III) reduction at redox interfaces, thereby promoting rapid microscale cycling of Fe. The brain renin-angiotensin system (RAS) is involved in learning and memory, but the actual role of angiotensin II (A(II)) and its metabolites in this process has been difficult to comprehend. This has been so mainly due to procedural issues, especially the use of multi-trial learning paradigms and the utilization of pre-training intracerebroventricular infusion of RAS-acting compounds. Here, we specifically analyzed the action of A(II) in aversive memory retrieval using a hippocampal-dependent, one-trial, step-down inhibitory avoidance task (IA) in combination with stereotaxically localized intrahippocampal infusion of drugs. Rats bilaterally implanted with infusion cannulae aimed to the CA1 region of the dorsal hippocampus were trained in IA and tested for memory retention 24 h later. We found that when given into CA1 15 min before IA memory retention test, A(II), but not angiotensin IV or angiotensin(1-7) induced a dose-dependent and reversible amnesia without altering locomotor activity, exploratory behavior or anxiety state. The effect of A(II) was blocked in a dose-dependent manner by the A(II)-type 2 receptor (AT(2)) antagonist PD123319 but not by the A(II)-type 1 receptor (AT(1)) antagonist losartan. By themselves, neither PD123319 nor losartan had any effect on memory expression. Our data indicate that intra-CA1 A(II) hinders retrieval of avoidance memory through a process that involves activation of AT(2) receptors.  相似文献

Iron (Fe) is ubiquitous in forest ecosystems and its cycle is thought to influence the development of soil, particularly Spodosols (podsolization), and the biogeochemistry of macronutrients such as carbon (C), nitrogen (N), and phosphorus (P), as well as many trace metals. The cycle of Fe in northern hardwood forests remains poorly understood. To address some of these uncertainties, we constructed a biogeochemical budget of Fe for a small catchment at the Hubbard Brook Experimental Forest in the White Mountains of New Hampshire, USA. Horizonal, temporal, and elevational patterns of concentrations and fluxes of oxidized and reduced Fe species were assessed in leaf litter, soil, soil solution, and stream water. The chemistry of dissolved Fe was evaluated in the context of its relationship with dissolved organic carbon, pH, and dissolved oxygen. Soil solution fluxes of Fe were highest in the organic (Oa, 52.5 mol ha?1 year?1) horizon and decreased with depth in the mineral (Bh, 50.5 mol ha?1 year?1, and Bs, 19.7 mol ha?1 year?1) horizons, consistent with podsolization theories predicting immobilization of Fe following downward transport to mineral soils. The export of Fe in stream water (1.8 mol ha?1 year?1) was lower than precipitation input (3.5 mol ha?1 year?1). The low stream flux indicates most Fe in drainage waters was immobilized in the soil and retained in the watershed. The portion of total Fe as Fe(II) was ~10?C60% in soil solutions, seemingly high for soils that are considered to be well-drained, oxidizing environments. Organic complexes likely stabilized Fe(II) in solution under oxidizing conditions that would otherwise promote considerably higher Fe(III)-to-Fe(II) ratios. Our study indicates that there are organic matter-derived sources of dissolved Fe(II) as well as substantial mobilization of Fe(II), possibly the result of the reduction of Fe-bearing soil minerals.  相似文献

The extent that various concentrations of the paramagnetic metal ion manganese [Mn(II)] affect nuclear magnetic resonance (NMR) relaxation times was studied in vitro. Serial dilutions of Mn(II) were prepared in distilled water, 4% human serum albumin, dog plasma, dog gallbladder bile, and dog hepatic bile. T1 and T2 of each were measured at 10.7 M Hz using magnetization recovery and spin-echo radiofrequency sequences, respectively. The results show that relaxation rates (1/T1 and 1/T2) increase in a linear manner with increasing concentration of Mn(II) in all of the solutions tested. Mn(II) dissolved in dog gallbladder and hepatic bile, dog plasma, and 4% human serum albumin reduced relaxation times to a greater extent than Mn(II) in water. T1 times were reduced to a greater extent than T2 values. Thus, in T1 weighted magnetic resonance images, the NMR signal used to produce images would be more sensitive to the presence of Mn(II) in these biological fluids than in water. Furthermore, the magnitude of this in vivo effect of Mn(II) on NMR relaxation parameters depends not only on the concentration of this paramagnetic ion, but also on the constituents comprising the biological fluids (intra- and extracellular water, bile, plasma) and the nature of the chemical molecular interactions between these constituents and Mn(II).  相似文献

溶氧是水环境中一个重要的环境因子,为了探讨水中的溶氧含量水平是否会对陆生植物的耐淹能力造成影响,研究了陆生植物喜旱莲子草(Alternanthera philoxeroides)和牛鞭草(Hemarthria altissima)在遭受不同溶氧含量水体完全淹没后的生长表现、存活情况和非结构碳水化合物的变化。实验结果表明:(1)水体中的溶氧含量显著影响了处于完全水淹环境中的喜旱莲子草和牛鞭草的存活。受高溶氧水体完全水淹的喜旱莲子草和牛鞭草主茎的完好程度和存活叶的数量均显著高于遭受低溶氧水体完全水淹的喜旱莲子草和牛鞭草,喜旱莲子草和牛鞭草在高溶氧水体完全水淹后的生物量比低溶氧水体完全水淹后要高;(2)水体中的溶氧含量显著影响了处于完全水淹环境中的喜旱莲子草和牛鞭草的生长,受高溶氧水体完全水淹的喜旱莲子草主茎伸长生长和不定根生长显著强于受低溶氧水体完全水淹的喜旱莲子草,在不定根的生长上牛鞭草也具有同样的表现。(3)高溶氧水环境有利于减小被完全淹没的喜旱莲子草和牛鞭草的碳水化合物消耗,两种植物在受高溶氧完全水淹后体内具有的非结构性碳水化合物含量均比受低溶氧完全水淹后高。(4)喜旱莲子草比牛鞭草能更好地耐受完全水淹,当处于低溶氧完全水淹时表现得更为明显,本研究表明入侵物种喜旱莲子草比本地物种牛鞭草具有更强的环境适应能力和水淹耐受能力。  相似文献

This study was designed to determine whether magnesium ion in water would influence the colonization of Helicobacter pylori in 2-week-old miniature pigs. Groups A (2 pigs) and B (1 pig) were both fed a milk diet dissolved in drinking water, Group C (2 pigs) was fed a milk diet dissolved in deionized distilled water (DDW), and Group D (1 pig) was fed a milk diet dissolved in DDW supplemented with MgCl2. Groups B, C, and D were all challenged with H. pylori, and Group A was not. Necropsy was performed on the pigs on postinfection Day 5, and biopsy specimens were taken from 16 sites of the stomach. H. pylori were recovered from 11 of 16 sites in Group B, 1 of 32 sites in Group C, and 13 of 16 sites in Group D. On the other hand, the degree of lymphocyte infiltration increased in the order of Group A < Group B < Group C < Group D. These observations suggest that magnesium ion in drinking water is essential for the colonization of H. pylori in the pig stomach. Possible mechanisms for the lymphocyte infiltration are discussed.  相似文献

One nmole of angiotensin II (ANG II) or saralasin, given intracerebroventricularly, failed to alter the motor activity of rats in open field. A combined injection of both peptides caused a significant decrease of the number of crossings and rearings. In the electromagnetic motimeter horizontal activity of animals was changed by neither of the peptides while the vertical activity was increased by ANG II. Again, a combined injection of saralasin and ANG II inhibited both horizontal and vertical activity. Stereotypies evoked by both apomorphine (2 mg/kg) and amphetamine (6.5 mg/kg), given intraperitoneally, were markedly intensified by ANG II and saralasin. A five-fold increase of the re-entry latencies in the passive avoidance situation was observed after pre-test administration of ANG II or saralasin but not the two in combination. These results suggest that ANG II and saralasin may improve processes related to learning and memory through an unspecific mechanism involving central dopamine systems.  相似文献

A promising chemical absorption–biological reduction integrated process has been proposed. A major problem of the process is oxidation of the active absorbent, ferrous ethylenediaminetetraacetate (Fe(II)EDTA), to the ferric species, leading to a significant decrease in NO removal efficiency. Thus the biological reduction of Fe(III)EDTA is vitally important for the continuous NO removal. Oxygen, an oxidizing agent and biological inhibitor, is typically present in the flue gas. It can significantly retard the application of the integrated process. This study investigated the influence mechanism of oxygen on the regeneration of Fe(II)EDTA in order to provide insight on how to eliminate or decrease the oxygen influence. The experimental results revealed that the dissolved oxygen and Fe(III)EDTA simultaneously served as electron acceptor for the microorganism. The Fe(III)EDTA reduction activity were directly inhibited by the dissolved oxygen. When the bioreactor was supplied with 3% and 8% oxygen in the gas phase, the concentration of initial dissolved oxygen in the liquid phase was 0.28 and 0.68 mg l−1. Correspondingly, the instinct Fe(III)EDTA reduction activity of the microorganism determined under anoxic condition in a rotation shaker decreased from 1.09 to 0.84 and 0.49 mM h−1. The oxidation of Fe(II)EDTA with dissolved oxygen prevented more dissolved oxygen access to the microorganism and eased the inhibition of dissolved oxygen on the microorganisms.  相似文献

To address the relative contribution of central and peripheral angiotensin II (ANG II) type 1A receptors (AT(1A)) to blood pressure and volume homeostasis, we generated a transgenic mouse model [neuron-specific enolase (NSE)-AT(1A)] with brain-restricted overexpression of AT(1A) receptors. These mice are normotensive at baseline but have dramatically enhanced pressor and bradycardic responses to intracerebroventricular ANG II or activation of endogenous ANG II production. Here our goal was to examine the water and sodium intake in this model under basal conditions and in response to increased ANG II levels. Baseline water and NaCl (0.3 M) intakes were significantly elevated in NSE-AT(1A) compared with nontransgenic littermates, and bolus intracerebroventricular injections of ANG II (200 ng in 200 nl) caused further enhanced water intake in NSE-AT(1A). Activation of endogenous ANG II production by sodium depletion (10 days low-sodium diet followed by furosemide, 1 mg sc) enhanced NaCl intake in NSE-AT(1A) mice compared with wild types. Fos immunohistochemistry, used to assess neuronal activation, demonstrated sodium depletion-enhanced activity in the anteroventral third ventricle region of the brain in NSE-AT(1A) mice compared with control animals. The results show that brain-selective overexpression of AT(1A) receptors results in enhanced salt appetite and altered water intake. This model provides a new tool for studying the mechanisms of brain AT(1A)-dependent water and salt consumption.  相似文献

In recent years great advances in the understanding of photosystem II have been achieved. The process of photochemical charge separation seems to be fairly well understood, while the identity of the water oxidizing enzyme in photosystem II has remained uncertain. In the first part of the paper a brief review on structural and functional aspects of photosystem II is given, and in the second part the nature of the elusive water oxidizing enzyme is considered. Two models are discussed. The first model, favoured by the majority of groups working in this area, suggests that the reaction center polypeptide D1 (in association with other known photosystem II polypeptides) is the site of water oxidation. The second model, mainly based on our results with cyanobacteria, predicts that the water oxidizing enzyme is a separate polypeptide in the 30 kDa region, distinct from D1 and D2, in addition to the seven polypeptides so far recognized in minimal O2 evolving photosystem II complexes  相似文献

目的:研究以案例为基础的学习(CBL)教学法结合以问题为基础的学习(PBL)教学法在肝胆外科教学中的应用,为临床教学提供依据。方法:选取2014年1月到2015年6月我院肝胆外科实习生270例为研究对象,按照随机数字表法将实习生分为I组、II组和III组,每组90例,I组给予PBL联合CBL教学法,II组给予PBL教学法,III组给予传统学习(LBL)教学法,比较三组教学效果。结果:I组和II组学习兴趣、自主学习能力以及课堂氛围评分显著高于III组,I组知识理解评分显著高于II组和III组,II组显著高于III组,比较差异具有统计学意义(P0.05);I组基础知识分显著高于II组和III组,I组和II组临床技能评分显著高于III组,比较差异具有统计学意义(P0.05);I组和II组调查满意度显著高于III组,且I组显著高于II组,比较差异具有统计学意义(P0.05)。结论:PBL联合CBL教学法用于肝胆外科教学中能显著提高实习生的知识理解能力和满意度。  相似文献

A method of transferring dissolved H2 to a CO2 headspace and then absorbing out the CO2 to concentrate the H2 before gas chromatographic analysis was developed to measure low concentrations of dissolved H2. A detection limit of 10 pmol of H2 ml-1 of water was achieved. When used ot monitor H2 changes in a bovine rumen, a 10-fold increase in H2 was noted 1 h after feeding and then declined rapidly to the normal steady-state concentration of 1 microM.  相似文献

A membrane-inlet mass spectrometer connected to an open-system cuvette was used for direct measurement of dissolved methane and O2 in bacterial samples of strain OU-4-1, a type II methanotrophic bacterium. A technique was applied for keeping the concentration of dissolved methane or O2 in the sample constant while the concentration of the other dissolved gas was varied. This allowed the reaction mechanism of methane mono-oxygenase to be studied in vivo. The enzyme was found to follow a random bi-reactant mechanism with respect to binding of methane and O2. Binding of one substrate decreased the affinity for the other. The true binding constants were 1 microM for methane and 0.14 microM for O2. Studies of HCN inhibition confirmed the random bi-reactant mechanism. HCN was found to be a non-exclusive inhibitor with a binding constant of 0.4 microM.  相似文献

Renal effects of A II, retention of sodium and water, may be mediated by the stimulation of aldosterone secretion and/or by direct effects of A II on the kidneys. An attempt was made to differentiate between these two possibilities. Methods: Conscious, female beagle dogs were used. The dogs were kept under standardized conditions (metabolic cage, daily sodium intake 4.5 mmol X kg-1 bw, chronically implanted arterial and venous catheters, i.v. hormone substitution after adrenalectomy by a portable pump). A II was infused i.v. over a period of 60 min after 60 min control. (Rate: 1, 4, 20 or 200 ng X min-1 X kg-1 bw). Results: Mean arterial blood pressure (MABP) increased with 20 and 200 ng A II X min-1 X kg-1 bw by an average of 34 mm Hg and 65 mm Hg resp. before and after adrenalectomy. Before adrenalectomy: sodium and water excretion decreased always at 4 and 20 ng A II X min-1 X kg-1 bw, whereas a rate of 200 ng A II X min-1 X kg-1 bw had different effects on renal sodium and water excretion. After adrenalectomy: sodium and water excretion decreased at 4 ng A II X min-1 X kg-1 bw. Whereas a rate of 20 and 200 ng. -As no marked alterations of the glomerular filtration rate occurred, sodium retention observed was mainly due to tubular effects of A II. Determination of biodegradable dissolved organic carbon in waters is of particular importance for the water treatment industry. A simple method for determining biodegradable dissolved organic carbon which is applicable to surface and drinking water is proposed. It consists of sterilizing the water sample, inoculating it with autochthonous bacteria, and measuring the decrease in dissolved organic carbon concentration due to the carbon oxidization by bacteria. The detailed experimental procedure is discussed, and validation of the method is presented. The method has been used for studying river waters and for drinking water treatment plant design.  相似文献
