Though some research exists concerning general behavior and activity patterns of Walruses in zoos or aquariums, less is known about how these patterns change in response to various environmental and temporal contexts. This study presents two studies assessing behavioral changes in relation to feeding period, object enrichment (OE), and season in a social group of four Pacific Walruses at the New York Aquarium. Study 1 examined behavior in relation to feeding context (nonfeed, prefeed, postfeed); data were collected over a three‐week period, resulting in 47 observation sessions for each feeding context. Study 2 examined behavior in relation to OE and season; data were collected in two phases resulting in 12 enrichment and 9 no‐enrichment (NE) observation sessions (Phase 1), and 21 enrichment and 18 NE observation sessions (Phase 2). Study 1 showed that after feeding, oral behavior increased while social behavior and total swim frequency decreased. In Study 2, both swim frequency and social behavior were found to interact with OE and phase, while oral behavior remained constant across all conditions. As in the wild, both studies found all animals to be swimming the majority of the time. Though every animal spent much of its swim time engaged in an Individual Swimming Pattern (ISP), both studies showed that the proportion of ISP (in relation to total time swimming) remained stable across all contexts, suggesting a potential functional role of the ISPs. Hair samples of 15 adult male Atlantic walruses (Odobenus rosmarus rosmarus) collected from anaesthetized individuals at Svalbard, Norway, were analysed for cadmium and total mercury. The mean level of cadmium was 0.860 ± 0.321 μg/g dry weight (median = 0.811, range = 0.349–1.51 μg/g dry weight) and the mean level of mercury was 0.235 ± 0.100 μg/g dry weight (median = 0.251, range = 0.121–0.424 μg/g dry weight). Levels of cadmium and mercury in hair of walruses from other areas are not known. Both cadmium and mercury levels in hair of walruses from Svalbard are relatively low compared to the levels found in the hair of other marine mammal species. It has been documented from a number of marine species, including marine mammals such as ringed seals and polar bears, that both cadmium and mercury levels at Svalbard are lower than in other areas. It is uncertain as to what degree levels in hair reflect levels in internal organs in walruses. In rare and highly endangered species or populations tissue samples can be difficult to collect. In walruses, it is possible to collect hair from anaesthetized individuals or at the haul-out sites during moult, to monitor heavy metal levels of the population. Accepted: 6 December 1998  相似文献

The sustainable use of resources requires that management practices and institutions take into account the dynamics of the ecosystem. In this paper, we explore the role of local ecological knowledge and show how it is used in management practices by a local fishing association in a contemporary rural Swedish community. We focus on the local management of crayfish, a common-pool resource, and also address the way crayfish management is linked to institutions at different levels of Swedish society. Methods from the social sciences were used for information gathering, and the results were analyzed within the framework of ecosystem management. We found that the practices of local fishing association resemble an ecosystem approach to crayfish management. Our results indicate that local users have substantial knowledge of resource and ecosystem dynamics from the level of the individual crayfish to that of the watershed, as reflected in a variety of interrelated management practices embedded in and influenced by institutions at several levels. We propose that this policy of monitoring at several levels simultaneously, together with the interpretation of a bundle of indicators and associated management responses, enhances the possibility of building ecological resilience into the watershed. Furthermore, we found that flexibility and adaptation are required to avoid command-and-control pathways of resource management. We were able to trace the development of the local fishing association as a response to crisis, followed by the creation of an opportunity for reorganization and the recognition of slow ecosystem structuring variables, and also to define the role of knowledgeable individuals in the whole process. We discuss the key roles of adaptive capacity, institutional learning, and institutional memory for successful ecosystem management and conclude that scientific adaptive management could benefit from a more explicit collaboration with flexible community-based systems of resource management for the implementation of policies as experiments. We investigate drivers of hybridization of local ecological knowledge (LEK) and scientific knowledge (SK) in small-scale Atlantic salmon (Salmo salar) fisheries in western Norway through a case study from the Ørsta River. We find three primary drivers of knowledge hybridization in local fishing groups as part of wild Atlantic salmon cultivation activities: facilitating intergenerational knowledge exchange, coping with regulatory change, and improving the perceived validity of local knowledge sets. We also identify three challenges to knowledge hybridization, and discuss how both drivers and challenges relate to once complementary SK and LEK sets that have diverged as SK has become more technical and complex. We examine the processes by which LEK and SK develop, evolve, and are used to facilitate wild salmon conservation in these fisheries and discuss the role hatcheries can play adapting and utilizing large-scale SK and salmon policy to the local environment through hybridization processes. We conclude with recommendations as to how reframing managerial views on hatcheries as facilitators of knowledge production and transfer may improve both the accessibility of SK to local communities and the integration of LEK into Norwegian wild salmon management.  相似文献

Although ecologists commonly talk about the impacts of nonindigenous species, little formal attention has been given to defining what we mean by impact, or connecting ecological theory with particular measures of impact. The resulting lack of generalizations regarding invasion impacts is more than an academic problem; we need to be able to distinguish invaders with minor effects from those with large effects in order to prioritize management efforts. This paper focuses on defining, evaluating, and comparing a variety of measures of impact drawn from empirical examples and theoretical reasoning. We begin by arguing that the total impact of an invader includes three fundamental dimensions: range, abundance, and the per-capita or per-biomass effect of the invader. Then we summarize previous approaches to measuring impact at different organizational levels, and suggest some new approaches. Reviewing mathematical models of impact, we argue that theoretical studies using community assembly models could act as a basis for better empirical studies and monitoring programs, as well as provide a clearer understanding of the relationship among different types of impact. We then discuss some of the particular challenges that come from the need to prioritize invasive species in a management or policy context. We end with recommendations about how the field of invasion biology might proceed in order to build a general framework for understanding and predicting impacts. In particular, we advocate studies designed to explore the correlations among different measures: Are the results of complex multivariate methods adequately captured by simple composite metrics such as species richness? How well are impacts on native populations correlated with impacts on ecosystem functions? Are there useful bioindicators for invasion impacts? To what extent does the impact of an invasive species depend on the system in which it is measured? Three approaches would provide new insights in this line of inquiry: (1) studies that measure impacts at multiple scales and multiple levels of organization, (2) studies that synthesize currently available data on different response variables, and (3) models designed to guide empirical work and explore generalities.  相似文献

Despite growing interest in traditional and local ecological knowledge for conservation and resource management, the role of migrant resource users is largely unexplored. Challenging many assumptions about what constitutes “local knowledge,” migrant and immigrant harvesters of non-timber forest products on the Olympic Peninsula, Washington possess useful ecological knowledge of overstory–understory relationships and how forestry practices affect understory biological and commercial production. Harvesters of salal (Gaultheria shallon), a shrub used in the multi-million dollar floral greens industry, were interviewed in Mason County, Washington in 2001–2003. Interviews revealed that harvesters possess different kinds of resource management knowledge depending on whether they are experienced harvesters or more recent newcomers to the area. These differences may also correlate with differences in their harvesting practices. Understanding how resource management knowledge differs between experienced and newcomer harvesters can inform forest managers in their efforts to develop effective management and permitting policies for floral greens and other non-timber forest resources in the Pacific Northwest of the United States.

The chronic diseases, comorbidities and rapidly changing needs of frail older persons increase the complexity of caregiving. A comprehensive, systematic and structured collection of data on the status of the frail older person is presumed to be essential in facilitating decision-making and thus improving the quality of care provided. However, the way in which an assessment is completed has a substantial impact on the quality and value of the results. This study examines the online completion of interRAI Home Care assessments, the possible causes for incomplete assessments and the consequences of these factors with respect to the quality of care received. Our findings indicate high nurse engagement and poor physician participation. We also observed the poor completion of items in predominantly medically- oriented sections characterized by, first, the fact that the assessors felt incapable of answering certain questions, second, the absence of required data or of a competent person to fill out the data, and third, the lack of tools necessary for essential measurements. The incompleteness of assessments has a clear negative influence on outcome generation. Moreover, without the added value of support outcomes, the improvement of care quality can be impeded and information technology can easily be seen as burdensome by the assessors. We have observed that multidisciplinary cooperation is an important prerequisite to establishing high-quality assessments aimed at improving the quality of care.  相似文献

Among the unexpected chemistries that can be catalyzed by nucleic acid enzymes is photochemistry. We have reported the in vitro selection of a small, cofactor-independent deoxyribozyme, UV1C, capable of repairing thymine dimers in a DNA substrate, most optimally with light at a wavelength of >300 nm. We hypothesized that a guanine quadruplex functioned both as a light antenna and an electron source for the repair of the substrate within the enzyme-substrate complex. Here, we report structural and mechanistic investigations of that hypothesis. Contact-crosslinking and guanosine to inosine mutational studies reveal that the thymine dimer and the guanine quadruplex are positioned close to each other in the deoxyribozyme-substrate complex, and permit us to refine the structure and topology of the folded deoxyribozyme. In exploring the substrate utilization capabilities of UV1C, we find it to be able to repair uracil dimers as well as thymine dimers, as long as they are present in an overall deoxyribonucleotide milieu. Some surprising similarities with bacterial CPD photolyase enzymes are noted.  相似文献

There is substantial research on psychological consequences of disasters. However, most disaster studies to date have focused on acute disasters, ignoring slow-onset chronic hazards, such as extreme climate conditions and pollution. Using a multilevel theoretical framework based on the Conservation of Resources theory (S. E. Hobfoll, The Ecology of Stress, Hemisphere, New York, 1988; Stress, Culture, and Community: The Psychology and Philosophy of Stress, Plenum, New York, 1998) and the “ecological analogy” (see e.g., S. E. Hobfoll and R. S Lilly, Journal of Community Psychology, 21:128–148, 1993; E. J. Trickett, Extreme Stress and Communities: Impact and Intervention, Kluwer, Boston, 1995), this critical review of the current literature is aimed at increasing our understanding of personal and community impacts of drought as a classic example of a natural, slow-onset disaster affecting large numbers of people worldwide. A gap in the current literature was identified concerning appraisal and coping at the individual level. These include psychological coping strategies and the role of resources other than economic resources in explaining vulnerability to negative consequences of drought, such as personal resources (e.g., knowledge, skills, self-sufficiency, mastery, control) and social resources (e.g., social support). Important differences were identified with fast-onset disasters. Most importantly, dealing with drought is generally an integrated part of life for people in drought prone areas. Therefore, individuals may not recognize that their problems are part of a community wide stressor, and raising community awareness during severe and long droughts that deplete community resources needs special attention. Implications for studying drought and effective intervention strategies are given.  相似文献

Objectives

To relate socio-demographic and virological information to phylogenetic clustering in HIV infected patients in a limited geographical area and to evaluate the role of recently infected individuals in the spread of HIV.

Methods

HIV-1 pol sequences from newly diagnosed and treatment-naive patients receiving follow-up between 2008 and 2011 by physicians belonging to a health network in Paris were used to build a phylogenetic tree using neighbour-joining analysis. Time since infection was estimated by immunoassay to define recently infected patients (very early infected presenters, VEP). Data on socio-demographic, clinical and biological features in clustered and non-clustered patients were compared. Chains of infection structure was also analysed.

Results

547 patients were included, 49 chains of infection containing 108 (20%) patients were identified by phylogenetic analysis. analysis. Eighty individuals formed pairs and 28 individuals were belonging to larger clusters. The median time between two successive HIV diagnoses in the same chain of infection was 248 days [CI = 176–320]. 34.7% of individuals were considered as VEP, and 27% of them were included in chains of infection. Multivariable analysis showed that belonging to a cluster was more frequent in VEP and those under 30 years old (OR: 3.65, 95 CI 1.49–8.95, p = 0.005 and OR: 2.42, 95% CI 1.05–5.85, p = 0.04 respectively). The prevalence of drug resistance was not associated with belonging to a pair or a cluster. Within chains, VEP were not grouped together more than chance predicted (p = 0.97).

Conclusions

Most newly diagnosed patients did not belong to a chain of infection, confirming the importance of undiagnosed or untreated HIV infected individuals in transmission. Furthermore, clusters involving both recently infected individuals and longstanding infected individuals support a substantial role in transmission of the latter before diagnosis.  相似文献

Lysolipids and fatty acids are the natural products formed by the hydrolysis of phospholipids. Lysolipids and fatty acids form micelles in solution and acts as detergents in the presence of lipid membranes. In this study, we investigate the detergent strength of a homologous series of lyso-phosphatidylcholine lipids (LPCs) on 1-palmitoyl-2-oleyl-sn-glycerol-3-phosphatidylcholine (POPC) lipid membranes by use of isothermal titration calorimetry and vesicle fluctuation analysis. The membrane partition coefficient (K) and critical micelle concentration (cmc) are determined by isothermal titration calorimetry and found to obey an inverse proportionality relation (cmc·K ∼ 0.05-0.3). The partition coefficient and critical micelle concentration are used for the analysis of the effect of LPCs on the membrane bending rigidity. The dependency of the bending rigidity on LPC membrane coverage has been analyzed in terms of a phenomenological model based on continuum elastic theory, which yields information about the curvature-inducing properties of the LPC molecule. The results reveal: 1), an increase in the partition coefficient with increasing LPC acyl-chain length; and 2), that the degree of acyl-chain mismatch between LPC and POPC determines the magnitude of the membrane mechanical perturbation per LPC molecule in the membrane. Finally, the three-stage model describing detergent membrane interaction has been extended by a parameter DMCI, which governs the membrane curvature stability in the detergent concentration range below the cmc-value of the LPC molecule.  相似文献

Bone allografting is the most common form of allotransplantation in modern medicine. Bone banking is usually the major part of most tissue banks throughout the world. Several years ago, many standards of bone banking were set empirically, and have never been evaluated. One particular parameter or standard was outdating graft materials after 5 years of storage. This study was conducted to evaluate the effect of prolonged cryopreservation on the biomechanical properties of bone allografts and establish whether graft materials become contaminated during long-term storage.Proximal humeral bone allografts were obtained from the bone bank after 1, 3 and 5 years of –80°C cryopreservation. Samples of each humeral head, i.e., cartilage, subchondral bone and spongy bone were histologically examined for inter- and intra-cellular changes. A three-point mechanical bending test was used on identical pieces of cortical bone to compare fresh and cryopreserved materials. Fresh-retrieved cortical bone using identically-sized segments, served as a control. Cultures were taken from each respective sample to determine contamination or sterility.Results of both the histological and mechanical testing showed that there were no significant, qualitative histological, or quantitative mechanical differences among the samples. All the cultures were negative. Therefore, based on this study's parameters, bone allografts can safely be used after a cryopreservation period of over 5 years and should not be discarded.  相似文献

The ultimate goal of most anti-tumor therapies is to kill tumor cells. While most of the attention in cancer therapy has been towards enhancing the death of tumor cells, the effect of dying tumors on the immune system has been less studied. Recent studies have suggested that cell death induced by different agents may have distinct consequences for the immune system. One of the immunogenic signals may be the expression of heat shock proteins on dying tumor cells under certain settings. For example, bortezomib (a proteasome inhibitor) induces the expression of heat shock protein 90 (hsp90) on the surface of dying human myeloma tumor cells. Recognition of such tumor cells by antigen presenting dendritic cells leads to the generation of anti-tumor T cells. Harnessing the properties of some anti-tumor agents to induce immunogenic death of tumor cells may facilitate the recruitment of adaptive immunity and promote the durability of anti-tumor effects.  相似文献

Common property arrangements govern the subsistence harvest of berries in the Gwich’in region of the Northwest Territories, Canada. Some of these arrangements, including rules for resource access, sharing information and harvest sharing, enable the Gwich’in to deal with ecological variability. The rules change in response to year-to-year variations in the abundance and distribution of the species, spatially and temporally across the region. The Protein Data Bank (PDB) is the worldwide repository of 3D structures of proteins, nucleic acids and complex assemblies. The PDB’s large corpus of data (> 100,000 structures) and related citations provide a well-organized and extensive test set for developing and understanding data citation and access metrics. In this paper, we present a systematic investigation of how authors cite PDB as a data repository. We describe a novel metric based on information cascade constructed by exploring the citation network to measure influence between competing works and apply that to analyze different data citation practices to PDB. Based on this new metric, we found that the original publication of RCSB PDB in the year 2000 continues to attract most citations though many follow-up updates were published. None of these follow-up publications by members of the wwPDB organization can compete with the original publication in terms of citations and influence. Meanwhile, authors increasingly choose to use URLs of PDB in the text instead of citing PDB papers, leading to disruption of the growth of the literature citations. A comparison of data usage statistics and paper citations shows that PDB Web access is highly correlated with URL mentions in the text. The results reveal the trend of how authors cite a biomedical data repository and may provide useful insight of how to measure the impact of a data repository.  相似文献

The mechanisms of action of the anticonvulsant topiramate (TPM) are indicative of a potential benefit during cerebral ischemia. TPM was studied in a transient global forebrain ischemia (TGFI) model in gerbils in which 40 mg/kg was administered before or after TGFI. Control groups were administered 0.9% normal saline similarly. The evaluation consisted of neurochemical, histological, and functional analyses. The data obtained indicates that unlike the focal cerebral ischemia model, TPM is not neuroprotective in TGFI. The difference in effect, which may be due to the difference in species or the type of ischemia, points to the need for caution when extrapolating animal data from this drug to humans.  相似文献
