The synthesis of oleanolic acid 3β-d-glucuronoside from oleanolic acid and UDPGlcA has been demonstrated in cell-free preparations from C. officinalis seedlings. Moreover, the formation of more complex glycosides by successive additions of galactose and glucose to oleanolic acid glucuronoside was observed when cell-free preparations were incubated with UDPGal or UDPGlc. The consecutive steps of oleanolic acid glycosylation are localized in three different cellular compartments. The biosynthesis of the 3-glucuronoside takes place in the microsomes, the elongation of the sugar chain at C-3 of the aglycone proceeds in heavy membrane structures which are probably fragments of the Golgi complex while a cytosol enzyme(s) is involved in glucosylation of the C-17 carboxyl group of oleanolic acid.  相似文献

The biosynthesis of dehydrotremetone in Eupatorium rugosum has been investigated by feeding radioactive precursors to intact plants. The carbon atoms of acetate-[1-14C] and acetate-[2-14C] were identified in dehydrotremetone by degradation of the molecule. From the pattern of labeling it was concluded that the acetophenone moiety was derived from acetate via the polyacetate pathway. From the incorporation of mevalonate it appeared that the furan ring and its side chain were formed from an isoprenoid compound. Potential aromatic intermediates were chemically synthesized and also fed to plants but only tremetone was found to be efficiently incorporated into dehydrotremetone. Neither 4-hydroxyacetophenone nor 4-hydroxy-3[isopenten-(2)-yl]-acetophenone were efficiently incorporated into dehydrotremetone.  相似文献

Administration of 24-methylene-cholesterol-[28-3H] to Withania somnifera, yielded [3H] radioactivity in the isolated withaferin A and withanolide D, whereas administered 24-(R,S)-methyl-cholesterol-[28-3H] was not incorporated into these compounds. 24-Methylene-cholesterol is, therefore, proposed as a sterol precursor of the withanolides. A novel procedure is described for the isolation of withanolides from W. somnifera. This method in conjunction with an improved procedure for administration of labelled sterols and mevalonolactone produces a greatly increased yield of labelled withanolides.  相似文献

Hydrazine, hydrazone and hydrazide derivatives are nitrogen–nitrogen bond containing compounds. Such molecules are relatively scarce in nature and have been isolated from plants, marine organisms and microorganisms. These compounds exhibit remarkable structural diversity and relevant biological activities. The enzymes involved in the formation of the N–N bond are still unknown, but many lines of evidence support the involvement of N-nitrosation and N-hydroxylation activating steps. Beside the challenging N–N bond, N-acylases catalyzing the C–N bond formation contribute to the chemical diversity of N–N-containing natural products (N2NP). This review examines the state of knowledge regarding the biosynthesis of N2NP, for which only two biosynthetic gene clusters have been investigated. Biological properties and chemical synthesis of hydrazines, hydrazones and hydrazides are also reported.  相似文献

Analyses of citrus leaves and fruits, and radioisotope tracer work showed that the leaves could synthesize limonoids and that the trees could translocate limonoids from leaves to the fruit. These results suggest that limonoids in citrus fruit tissues are synthesized in leaves and translocated to the fruit. No evidence was found to support the presence of limonoid biosynthetic systems in the fruit tissues.  相似文献

The incorporation of l-[U-14C]lysine and l-[U-14C]phenylalanine into piperlongumine has been demonstrated in Piper longum. The subsequent stepwise degradation to methyl-(3,4,5-trimethoxyphenyl)-propanoate and δ-aminovaleric acid revealed that the C6-C3 moiety of the alkamide arises from phenylalanine; the heterocyclic ring is biosynthesised from lysine. It has also been shown that dl-[2-14C]tyrosine and [2-14C]sodium acetate are poor precursors of piperlongumine.  相似文献

Two fractions obtained after chromatography of dichloromethane extract of Penicillium brevicompactum culture medium showed anti-juvenile hormone activity. One was active when assayed in vivo against Oncopeltus fasciatus third-instar nymphs, whereas the other showed a strong in vitro inhibition of JH III biosynthesis on Locusta migratoria corpora allata. A subfraction of the latter, constituting 97% of this fraction, is a main component that possessed the juvenile hormone biosynthesis inhibitory activity. Chemical characterization of this compound shows a sesquiterpene-like structure, molecular mass of 278.16185 daltons, corresponding to an empirical formula of C15H22N2O3 named brevioxime. Evidence suggests that the final steps of JH III biosynthesis are the target of the active compound. Its effects may be compared with those of a known inhibitor of P-450-linked epoxidase, KK-42 (1-benzyl-5-[(E)-2,6-dimethyl-1,5-heptadienyl]imidazole), whose effects on L. 抗坏血酸是高等植物中普遍存在的小分子物质,在植物细胞对氧化胁迫的抵抗、细胞分裂和伸长中发挥着重要作用。它在线粒体中合成,然后被转运到其他细胞区间以发挥生理作用。在不同生物膜上可能存在不同的ASC转运体,这些转运体的存在可使ASC及其不同氧化还原态形式根据细胞生理状况的需要而在不同细胞区间转运。ASC的重要生理功能是与其氧化还原状态以及生物合成、代谢、再生和转运的相关酶类活性的变化密切相关的。本文简要综述了近年来人们在植物抗坏血酸生物合成、代谢、转运、逆境响应与植物生长发育中的研究进展。  相似文献

Malonic acid, mevalonic acid, geraniol and nerol were incorporated into tetrahydrocannabinolic acid and cannabichromenic acid in Cannabis sativa. The pathway from cannabigerolic acid to tetrahydrocannabinolic acid via cannabidiolic acid was established by feeding labelled cannabinoid acids. The incorporation of (±)-coclaurine, (±)-norcoclaurine, (±)-N-methylcoclaurine and didehydro-N-methyleoclaurinium iodide into tetrandrine in Cocculus laurifolius has been studied and specific utilization of (±)-N-ethylcoclaurine demonstrated. The evidence indicates that tetrandrine is formed in the plants by oxidative dimerization of N-methylcoclaurine. Double labelling experiment with (±)-N- [14C]-methyl- [1-3H]-coclaurine demonstrated that the hydrogen atom at the asymmetric centre in the 1-benzylisoquinoline precursor is retained in the bioconversion into tetrandrine. Parallel feedings of (+)-(S)- and (?)-(R)-N-methylcoclaurines showed that the stereospecificity is maintained in the biosynthesis of tetrandrine from the 1-benzylisoquinoline precursor.  相似文献

By incubation of germinating soybeans with mevalonate-[2-14C] (MVA), radioactivity was incorporated into four sapogenols which were identified by TLC. Unequivocal evidence for the identity of three of the four sapogenols was provided by co-crystallization to constant specific radioactivity. The partition of incorporated radioactivity into lipid- and water-soluble fractions and the pattern of radioactivity of individual sapogenols varied with the mode of administering labeled substrates to soybean seedlings, such as incubation of germinating soybeans with MVA-[2-14C], immersion of roots into MVA-[2-14C] or foliar application of squalene-[14C]. When alfalfa seedlings were incubated with MVA-[2-14C], about two-thirds of the radioactivity incorporated into the sapogenols was associated with medicagenic acid.  相似文献

Precocene II (6,7-dimethoxy-2,2-dimethylchromene), the most active anti-juvenile hormone isolated from Ageratum houstonianum, has been shown to be hepatotoxic in male Sprague-Dawley rats. A single 300-mg/kg dose of precocene II administered via i.p. injection caused extensive necrosis of parenchymal cells in the hepatic centrolobular areas. Liver functions were markedly affected as shown by the significant increases of glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase in the serum. By means of reversed-phase high pressure liquid chromatography (HPLC), [3H]precocene II was found to be rapidly metabolized in vitro by rat liver microsomes in an NADPH-generating system. Approximately 5% (3.4 nmol/mg protein) of the radioactivity from the [3H]precocene II substrate was covalently bound to the macromolecular pellet at the end of a 15-min incubation period when phenobarbital (PB)-induced microsomes were used. Results obtained from experiments using different incubation systems indicated the involvement of the cytochrome P-450-dependent monooxygenases in the metabolism of precocene II and the concurrent covalent binding. The most predominent metabolite was isolated and accounted for >90% of the radioactivity associated with the ethylacetate-extractable metabolites. Further analysis by mass spectrometry and proton nuclear magnetic resonance (NMR) spectroscopy identified it as a 37 : 63 stereoisomeric mixture of the cis and trans 3,4-dihydrodiols of precocene II. A highly reactive (3,4-epoxy-6,7-dimethyl-2,2-dimethylchromane (precocene-3,4-epoxide) was thus suggested as a crucial metabolic intermediate which may be responsible for the histopathological changes seen in rat liver.  相似文献
