6,7-Dimethoxycoumarin was isolated and identified from the peels of Citrus sinensis cv. Pineapple orange, of C. paradisi x C. paradisi x C. reticulata cv. Wekiwa and of C. mitis cv. Clamondin. The coumarin was tentatively identified in two other sweet oranges, C. sinensis cv. Mediterranean Sweet and Waite Parson Brown, and in C. aurantifolia cv. West Indian lime and in C. aurantium cv. In the laboratory, the effect of host fruit species Citrus paradisi, C. aurantium, Prunus persica, and Psidium guajava on A. fraterculus parasitism by Diachasmimorpha longicaudata was studied. The number of ovipositor-probing events and the probing-time were documented to evaluate the role of fruit chemistry, and epicarp and mesocarp thickness, respectively. The relationship between the parasitization rate and fruit size in particular plant species was analyzed by applying a simple regression. Results showed that guava and peach yielded significantly more parasitoids than both Citrus spp. Probably, the parasitization rate of D. longicaudata on A. fraterculus would be influenced in part by chemical and physical factors from fruit species.  相似文献

Amylase polymorphism was studied by electrophoresing crude protein extracts from mature leaves of Citrus and some related genera in polyacrylamide gel slabs. A total of 25 isoenzymes distributed within 2 distinct zones were resolved. Cultivars of C. medica were the least polymorphic with 5 isoenzymes, those of C. paradisi were the most polymorphic with 14 isoenzymes; the other taxa were between these two extremes. Cultivars within species, in general, had no qualitative differences in their isoenzymes profiles with the exception of C. aurantifolia, C. limon, and C. aurantium. Certain isoenzymes showed quantitative differences among taxa. Data provided evidence substantiating the presumed hybrid origin of some taxa. The potential of isoenzyme profiles in taxonomic and genetic studies was discussed.  相似文献

Citrus aurantium extracts, which contain large amounts of p-synephrine, are widely used for weight loss purposes and as appetite suppressants. In the liver, C. aurantium (bitter orange) extracts affect hemodynamics, carbohydrate metabolism, and oxygen uptake. The purpose of the present work was to quantify the action of p-synephrine and also to obtain indications about its mechanism of action, a task that would be difficult to accomplish with C. aurantium extracts due to their rather complex composition. The experimental system was the isolated perfused rat liver. p-Synephrine significantly stimulated glycogenolysis, glycolysis, gluconeogenesis, and oxygen uptake. The compound also increased the portal perfusion pressure and the redox state of the cytosolic NAD+/NADH couple. A Ca2+-dependency for both the hemodynamic and the metabolic effects of p-synephrine was found. p-Synephrine stimulated both cAMP overflow and the initial Ca2+ release from the cellular stores previously labeled with 45Ca2+. The metabolic and hemodynamic actions of p-synephrine were strongly inhibited by α-adrenergic antagonists and moderately affected by β-adrenergic antagonists. The results allow to conclude that p-synephrine presents important metabolic and hemodynamic effects in the liver. These effects can be considered as both catabolic (glycogenolysis) and anabolic (gluconeogenesis), they are mediated by both α- and β-adrenergic signaling, require the simultaneous participation of both Ca2+ and cAMP, and could be contributing to the overall stimulation of metabolism that usually occurs during weight loss periods.  相似文献

Chromium(VI), a very strong oxidant, causes high cytotoxicity through oxidative stress in tissue systems. Our study investigated the potential ability of ethanolic Citrus aurantium L., family Rutaceae extract, used as a nutritional supplement, to alleviate lung oxidative damage induced by Cr(VI). A high-performance liquid chromatography coupled with a mass spectrometer method was developed to separate and identify flavonoids in C. aurantium L. Six flavonoids were identified, as (1) poncirin, (2) naringin, (3) naringenin, (4) quercetin, (5) isosinensetin, and (6) tetramethyl-o-isoscutellarein. Adult Wistar rats, used in this study, were divided into six groups of six animals each: group I served as controls which received standard diet, group II received via drinking water K2Cr2O7 alone (700 ppm), groups III and IV were pretreated for 10 days with ethanol extract of C. aurantium L. at doses of 100 and 300 mg/kg body weight/day, respectively, and then K2Cr2O7 was administrated during 3 weeks, and groups V and VI received during 10 days only C. aurantium L. ethanol extract at doses of 100 and 300 mg/kg/day, respectively. Ethanol extract of C. aurantium L. was administered orally. Rats exposed to Cr(VI) showed in lung an increase in malondialdehyde and protein carbonyl levels and a decrease in sulflydryl content, glutathione, nonprotein thiol, and vitamins C and E levels. Decreases in enzyme activities such as in Na+K+ ATPase, catalase, glutathione peroxidase, and superoxide dismutase were noted. Pretreatment with C. aurantium L. of chromium-treated rats ameliorated all biochemical parameters. Lung histological studies confirmed the biochemical parameters and the beneficial role of C. aurantium L.  相似文献

The pathogenic effects of Pratylenchus coffeae on growth and yield of tangelo (Citrus paradisi × C. reticulata) scions grafted on rough lemon (C jambhiri), sour orange (C. aurantium) and ''Cleopatra'' mandarin (C. reticulata) rootstocks were evaluated under field conditions for 4 years. Pratylenchus coffeae on inoculated trees increased to significantly damaging population densities on rough lemon rootstock the second year, on sour orange the third and on Cleopatra mandarin the fourth year after planting. Mean growth reduction of P. coffeae-infected trees after 4 years was 80, 77 and 49%, respectively, for the three rootstocks. Noninoculated trees on rough lemon and sour orange rootstocks yielded significantly more fruit than comparable inoculated trees. Natural migration of P. coffeae occurred horizontally on roots for a distance of 4.5 m.  相似文献

Distribution of the citrus nematode (Tylenchulus semipenetrans) was studied over 18 months in a 6-year-old orchard of grapefruit (Citrus paradisi cv. Ruby Red) on sour orange (C. aurantium) rootstock. The 1.8-ha orchard was under chemical weed control, no tillage, and flood irrigation. Highest numbers of nematodes were found in the top 15 cm of the soil profile. The nematode population peaked in April and declined to lowest levels in August and September. Numbers of nematodes were negatively correlated (r = -0.95) with soil temperatures above 29 C. Soil populations of nematodes were not correlated with soil moisture. The distribution of the nematode in the field was highly skewed and was described by a negative binomial. In this 1.8-ha block, five soil samples of 12 cores each would provide an estimate within 20% of the true nematode population mean with 95% confidence.  相似文献

Valencia orange (C. sinensis) and Robinson tangerine [(C. paradisi × C. reticulata) × (C. reticulata)] were examined for flavonoids. Thirteen flavonoids were isolated, six of which are new constituents of citrus peel. These are: 3,5,6,7,3′,4′-hexamethoxyflavone, 3,5,7,8,3′,4′-hexamethoxyflavone, 5-hydroxy-3,7,8,3′,4′-pentamethoxyflavone, 5-hydroxy-3,6,7,8,3′,4′-hexamethoxyflavone, 5,7,8,4′-tetramethoxyflavone and 5,7,8,3′,4′-pentamethoxyflavone. The latter three flavonoids are reported for the first time as natural products. A method is described for readily obtaining small quantities of 5,7,8,4′-tetramethoxy and 5,7,8,3′,4′-pentamethoxyflavones from their 5,6,7-trimethoxy analogs.  相似文献

The 1H and 13C NMR spectral analysis of the glucoside renifolin, isolated from Pyrola renifolia, demonstrated the so far unknown binding site of the glucose moiety to be at C-8 and hence its structure as 8-β-D-glucosyloxy-2,7-dimethyl-1,4-dihydronaphthalen-5-ol. The earlier reported structure for the glucoside pirolatin, isolated from Pyrola japonica, was also reconfirmed by the 13C NMR spectral analysis.  相似文献

The aim of this study was to determine the effects of nitrogen (N) fertilization methods on root distribution and mineral element concentrations of White Marsh grapefruit (Citrus paradisi MacFadyen) trees on sour orange (C. aurantium Lush) rootstock on a poorly drained soil. At 0–15 cm depth of soil, root density was significantly greater for trees receiving 112 kg N ha-1 yr-1 as dry granular broadcast than those receiving the same amount of N as fertigation. Of the total roots in the top 60 cm soil, >75% was at 0–15 cm and <10% was at 30–60 cm. Root density was greatest near the emitter. Nitrogen concentration of roots was greater for the trees which received fertigation as compared to the trees which received dry fertilizer broadcast or no N.  相似文献

The juvenile hormone (JH) analog methoprene reduces the amount of time it takes laboratory-reared Anastrepha suspensa (Caribbean fruit fly) males to reach sexual maturity by almost half. Here, we examined if methoprene exerted a similar effect on four other tropical Anastrepha species (Anastrepha ludens, Anastrepha obliqua, Anastrepha serpentina and Anastrepha striata) reared on natural hosts and exhibiting contrasting life histories. In the case of A. ludens, we worked with two populations that derived from Casimiroa greggii (ancestral host, larvae feed on seeds) and Citrus paradisi (exotic host, larvae feed on pulp). We found that the effects of methoprene, when they occurred, varied according to species and, in the case of A. ludens, according to larval host. For example, in the case of the two A. ludens populations the effect of methoprene on first appearance of male calling behavior and number of copulations was only apparent in flies derived from C. greggii. In contrast, males derived from C. paradisi called and mated almost twice as often and females started to lay eggs almost 1 day earlier than individuals derived from C. greggii, but in this case there was no significant effect of treatment (methoprene) only a significant host effect. There were also significant host and host by treatment interactions with respect to egg clutch size. A. ludens females derived from C. paradisi laid significantly more eggs per clutch and total number of eggs than females derived from C. greggii. With respect to the multiple species comparisons, the treatment effect was consistent for A. ludens, occasional in A. serpentina (e.g., calling by males, clutch size), and not apparent in the cases of A. obliqua and A. striata. Interestingly, with respect to clutch size, in the cases of A. ludens and A. serpentina, the treatment effect followed opposite directions: positive in the case of A. ludens and negative in the case of A. serpentina. We center our discussion on two hypotheses (differential physiology and larval-food), and also interpret our results in light of the life history differences exhibited by the different species we compared.  相似文献

Cuticular penetration of five different 14C-labeled chemicals (benzoic acid, bitertanole, carbaryl, epoxiconazole and 4-nitrophenol) into Arabidopsis thaliana leaves was measured and permeances P (ms−1) were calculated. Thus, cuticular barrier properties of A. thaliana leaves have been characterized quantitatively. Epoxiconazole permeance of A. thaliana was 2.79 × 10−8 ms−1. When compared with cuticular permeances measured with intact stomatous and astomatous leaf sides of Prunus laurocerasus, frequently used in the past as a model species studying cuticular permeability, A. thaliana has a 48- to 66-fold higher permeance. When compared with epoxiconazole permeability of isolated cuticles of different species (Citrus aurantium, Hedera helix and P. laurocerasus) A. thaliana permeability is between 17- to 199-fold higher. Co-permeability experiments, simultaneously measuring 14C-epoxiconazole and 3H2O permeability of isolated cuticles of three species (C. aurantium, H. helix and P. laurocerasus) showed that 3H2O permeability was highly correlated with epoxiconazole permeability. The regression equation of this correlation can be used predicting cuticular transpiration of intact stomatous leaves of A. thaliana, where a direct measurement of cuticular permeation using 3H2O is impossible. Water permeance estimated for A. thaliana was 4.55 × 10−8 ms−1, which is between 12- and 91-fold higher than water permeances measured with isolated cuticles of C. aurantium, H. helix and P. laurocerasus. This indicates that cuticular water permeability of the intact stomatous leaves of the annual species A. thaliana is fairly high and in the upper range compared with most P values of perennial species published in the past.  相似文献

We examined some biophysical mechanisms of ion migration across leaf cuticles enzymatically isolated from Acer saccharum L. and Citrus aurantium L. leaves. Diffusion potential measurements were used to calculate the permeabilities of Cl-, Li+, Na+, and Cs+ ions all as a ratio with respect to the permeability of K+ in cuticles. In 2 millimolar ionic strength solutions the permeability sequence from high to low was K = Cs > Na > Li » Cl. When the outer and inner surfaces of cuticles were bathed in artificial precipitation and artificial apoplast, respectively, diffusion potentials ranging from −52 to −91 millivolts were measured (inside negative). The Goldman equation predicted that the measured potentials were enough to increase the driving force on the accumulation of heavy metals by a factor of 4 to 7. Other ions migrate with forces 3 to 10 times less than predicted by the Goldman equation for concentration differences alone. Our analysis showed that Ca2+, and perhaps Mg2+, might even be accumulated against concentration gradients under some circumstances. Their uptake was apparently driven by the diffusion potentials created by the outward migration of monovalent salts. We feel that future models predicting leaching of nutrients from trees during acid rain events must be modified to account for the probable influence of diffusion potentials on ion migration.  相似文献

Infection of Citrus tristeza virus (CTV) in different citrus orchards of New Delhi was detected by direct antigen coated-ELISA and RT-PCR. Sweet orange (Citrus sinensis) orchards were found to be susceptible to CTV with estimated disease incidence up to 39%. Kagzi kalan (C. lemon), Pumello (C. paradisi) and Kinnow mandarin (C. reticulata) orchards did not show CTV infection. Three CTV isolates, D1, D7 and D15 randomly selected from infected sweet orange orchards were considered for biological and molecular characterization. In the host range study, all the Delhi isolates infected Darjeeling mandarin (C. reticulata), Kagzi lime (C. aurantifolia), sour orange (C. aurantium) and sweet orange but not Kinnow mandarin. A fragment of 5??ORF1a and complete coat protein (CP) gene of these three isolates were cloned, sequenced and compared with other Indian and international CTV isolates. Delhi isolates shared 85?C92% sequence identity for 5??ORF1a fragment and 89?C91% for CP gene among them. Phylogenetic analysis segregated three Delhi isolates into three genogroups for each of 5??ORF1a fragment and CP gene, however phylogenetic relationships for both the genomic regions was incongruent. Recombination detecting program RDP3 detected CTV isolate D7 as recombinant, indicating genetic variability in CTV isolates might be the outcome of recombination events between divergent CTV sequences. An attempt was made in present study to characterize CTV isolates biologically and at genetic level, and to determine genetic diversity at farm level and study the recombination of CTV isolates in Delhi region.  相似文献

The structure of sulcatine, a novel protoilludene norsesquiterpene isolated from a culture of Laurilia sulcata, has been assigned on the basis of a detailed study of its 1H NMR and 13C NMR spectra and chemical evidence. The relative configuration was deduced from the observed 1H {1H} nuclear Overhauser effects (NOEs) and from the values of 1H1H coupling constants. The application of the exciton chirality method on the dibenzoate permitted establishment of the absolute configuration of sulcatine as 2S,3R,7S,8S,9R.  相似文献

The characteristic pungency of the liverworts Plagiochila species P. fruticosa, P. hattoriana, P. ovalifolia and P. yokogurensis is due to a new ent-secoaromadendrane-type sesquiterpene hemiacetal, plagiochiline A, which exhibits very strong antifeedant activity against the African army worm, Spodoptera exempta at 1–10ng/cm2. Two new secoaromadendranes, plagiochilide and furanoplagiochilal A, together with the previously known plagiochiline C were isolated from P. yokogurensis. Plagiochilal A, which may be a precursor of plagiochilide and its related hemiacetals, and a bitter principle, plagiochiline B were also isolated from P. hattoriana. P. ovalifolia contained plagiochilines A, B and C. From P. fruticosa, plagiochilide and plagiochilines A, B and C were isolated. The structures of the new secoaromadendrane-type sesquiterpenes were elucidated by extensive 1H NMR and 13CNMR studies.  相似文献

The noncallusing morphogenetic properties of Citrus juice vesicles cultured in vitro are unknown and were herein studied. Juice vesicles isolated from 120–180-day-old fruits are capable of proliferation via adventitious vesicle branching in vitro. Gibberellic acid levels of 1–100 mg/L greatly enhanced adventitious vesicle branching while delaying vesicle senescence as evidenced by the vesicles' ability to retain its green color for up to 4 to 6 mo in culture. Vesicles grown on media without gibberellic acid readily matured, turning opaque within 2 mo in culture and rarely produced adventitious branches. Additions of 0.1 mg/L α-naphthaleneacetic acid or 1.0 mg/L benzylaminopurine to media containing 10 mg/L gibberellic acid further enhanced vesicle branching in some species and cultivars. Adventitious juice vesicles have been induced from a variety of citrus species, including Citrus grandis (L.) Osb., C. hystrix DC., C. limon (L.) Burm. f., C. medica L., C. paradisi Macf., C. reticulata Blanco, and C. sinensis (L.) Osb. Adventitious vesicle branches originate from primordia initiated on the surface of the cultured vesicle. These primordia commonly occurred on the terminal meristem region of the vesicle. However, some species (e.g., C. grandis and C. paradisi) produced adventitious vesicle branches from their bodies and stalks as well. Distinct vesicle branches begin to appear on preformed vesicles after 30–60 days in culture. A survey to determine the natural occurrence of vesicle branching in various Citrus species was also conducted. Natural vesicle branching commonly occurred in C. grandis, C. paradisi, and in some cultivars of C. reticulata, but was absent in most citrus species and cultivars (e.g., C. aurantifolia (Christm.) Swing., C. canaliculata Hort. ex Y. Tan., C. hystrix, C. limon, C. medica, C. sinensis). It was found that some species which failed to exhibit the adventitious branching phenomenon in nature did so in vitro (e.g., C. hystrix, C. medica, and C. sinensis).  相似文献

A simple and efficient protocol for obtaining organogenesis from mature nodal explants of Citrus macrophylla (alemow) and Citrus aurantium (sour orange) has been developed by optimizing the concentrations of the plant growth regulators, the incubation conditions, the basal medium and by the choice of explant. In order to optimize the plant growth regulator balance, explants were cultured in the regeneration medium supplemented with several N 6-benzyladenine (BA) concentrations or with 2 mg?l?1 BA in combination with kinetin (KIN) or 1-naphthaleneacetic acid (NAA). The presence of BA was found to be essential for the development of adventitious buds; the best results were obtained using BA at 3 and 2 mg?l?1 for alemow and sour orange, respectively. The combination of BA with KIN or NAA in the culture medium decreased the regeneration frequency, with respect to the use of BA alone. The effect of three different basal media was rootstock-dependent. For C. macrophylla the best results were obtained with Woody Plant Medium or Driver and Kuniyuki Walnut Medium (DKW). However, for C. aurantium, although high percentages of regenerating explants were obtained independently of the basal medium used, the highest number of buds per regenerating explants was obtained with DKW medium. Attempts were made to identify the type of explants which had a higher regeneration ability using particular regions along the mature shoots of C. macrophylla. When nodal segments, where the buds were completely removed, and internode segments were compared, the highest percentage of responsive explants was obtained with nodal segments. The existence of a morphogenetic gradient along the shoot was observed and the organogenic efficiency was highest when explants from the apical zone were used. Incubation in darkness for 3 or 4 wk was essential for regeneration process in both rootstocks.  相似文献

Fick's second law has been used to predict the time course of electrical conductance change in isolated cuticles following the rapid change in bathing solution (KCI) from concentration C to 0.1 C. The theoretical time course is dependent on the coefficient of diffusion of KCI in the cuticle and the cuticle thickness. Experimental results, obtained from cuticles isolated from sour orange (Citrus aurantium), fit with a diffusion model of an isolated cuticle in which about 90% of the conductance change following a solution change is due to salts diffusing from polar pores in the wax, and 10% of the change is due to salt diffusion from the wax. Short and long time constants for the washout of KCI were found to be 0.11 and 3.8 hours, respectively. These time constants correspond to KCI diffusion coefficients of 1 × 10−15 and 3 × 10−17 square meters per second, respectively. The larger coefficient is close to the diffusion coefficient for water in polar pores of Citrus reported elsewhere (M Becker, G Kerstiens, J Schönherr [1986] Trees 1: 54-60). This supports our interpretation of the washout kinetics of KCI following a change in concentration of bathing solution.  相似文献
