The distribution of free and protein amino acids during seed development is described for three chestnut varieties. Asparagine accumulation was shown to correlate with protein biosynthesis. Accumulation of free amino acids takes place before that of proteins in ripening seeds. A temporary decrease was generally accompanied by protein biosynthesis and followed by final accumulation of both protein and free amino acids. 相似文献

Thirty-one seaweeds were collected from the warmer KwaZulu-Natal coast and the cooler Cape waters (South Africa). Plant material was extracted with 70% ethanol supplemented with deuterium labelled standards of all known isoprenoid cytokinins. The samples were then centrifuged and purified by combined DEAE-Sephadex×octadecylsilica column and immunoaffinity chromatography and finally analysed for cytokinins by HPLC-linked mass spectrometry and a photodiode array detector. The cytokinin profiles were similar in all the macroalgae regardless of their taxonomy and growing locality. The main type of isoprenoid cytokinins present were zeatins with cis forms being more common than trans forms and isopentenyladenine (iP) derivatives. Only a few dihydrozeatin-type cytokinins were detected at very low levels in only nine species. Aromatic cytokinins were also present but at lower levels and were represented by benzyladenine (BA) and ortho- and meta-topolin derivatives. The topolins were present in greater diversity and concentrations than BA. For all the cytokinin types, the free bases, O-glucosides and nucleotides were the most common with no N-glucosides being detected and ribosides present at very low levels. The results suggest that different pathways for regulating cytokinin concentrations operate in macroalgae than in higher plants. 相似文献

Aim Chestnuts (Castanea spp.) are ecologically and economically important species. We studied the general biology, distribution and climatic limits of seven chestnut species from around the world. We provided climatic matching of Asiatic species to North America to assist the range‐wide restoration of American chestnut [C. dentata (Marsh.) Borkh.] by incorporating blight‐resistant genes from Asiatic species. Location North America, Europe and East Asia. Methods General chestnut biology was reviewed on the basis of published literature and field observations. Chestnut distributions were established using published range maps and literature. Climatic constraints were analysed for the northern and southern distribution limits and the entire range for each species using principal component analysis (PCA) of fourteen bioclimatic variables. Climatic envelope matching was performed for three Chinese species using Maxent modelling to predict corresponding suitable climate zones for those species in North America. Results Chestnuts are primarily distributed in the warm‐temperate and subtropical zones in the northern hemisphere. PCA results revealed that thermal gradient was the primary control of chestnut distribution. Climatic spaces of different species overlap with one another to different degrees, but strong similarities are shown especially between Chinese species and American species. Climatic envelope matching suggested that large areas in eastern North America have a favourable climate for Chinese species. Main conclusions The general biological traits and climatic limits of the seven chestnut species are very similar. The predictions of Chinese species climatic range corresponded with most of the historical American chestnut range. Thus, a regionally adapted, blight‐resistant, introgressed hybrid American chestnut appears feasible if a sufficiently diverse array of Chinese chestnut germplasm is used as a source of blight resistance. Our study provided a between‐continent climate matching approach to facilitate the range‐wide species restoration, which can be readily applied in planning the restoration of other threatened or endangered species. 相似文献

The existence of cytokinins both as a free form and as a constituent of t-RNA was investigated in young fruits of Moringa pterigosperma Gaertn. Purified methanol extract was separated into butanol insoluble and butanol soluble fractions. The cytokinin(s) in the butanol insoluble fraction was tentatively identified as zeatin nucleotide. The butanol soluble fraction contained cytokinins and was chromatographed on Sephadex LH-20 with 35% ethanol. The two active fractions from LH-20 column coincided with zeatin and zeatin riboside. Cytokinin per g tissue was high in early stages of fruit growth and then remained more or less constant. Alkaline phosphatase hydrolysis of t-RNA hydrolysate of fruit tissue showed considerable cytokinin activity. 相似文献

Castasterone and 6-deoxocastasterone were identified by GC/MS and/or selected ion monitoring not only in the insect gall of chestnut but also in the healthy tissues, including the shoot, leaf and flower bud. In addition, the gall was found to contain a small amount of brassinolide. Brassinone, which had been reported to be present in the gall, could not be detected in these tissues. The contents of castasterone, 6-deoxocastasterone and brassinolide, which were determined by using selected ion monitoring, are discussed. 相似文献

Roots are recognised as the major sites of cytokinin synthesis and shoots receive a continuous supply of cytokinins from the roots. Although reports are available on the xylem mobility of putative free bases and their ribosides, relatively few studies on the phloem mobility of cytokinins have been reported. The origin of phloem-mobile cytokinins is uncertain but there is evidence which implicates a recirculation from the root source. This study is the first report in which zeatin and zeatin riboside from the root pressure exudate and phloem sap of Ricinus have been identified by full-scan GC-MS and quantified by GC-MS selective-ion-monitoring. In this study, the concentration of cytokinins in root pressure exudate was similar, but lower, and in the phloem sap higher than that reported previously. The concentration of cytokinins quantified in the phloem sap confirms their transport in the sieve tubes. The relatively high concentration of zeatin riboside detected in the root pressure exudate and of zeatin detected in the phloem sap indicate a possible vascular recirculation of these hormones. 相似文献

The introduction of the gene for cytokinin biosynthesis into the potato genome led to a manifold increase in the level of cytokinins (zeatin and zeatin riboside) in transgenic plants grown in vitro. The high amount of endogenous cytokinins in 20-day-old plants of clone 1339-3A correlated with high cytokinin-binding capacity of ribosomes that is presumably attribute to a cytokinin receptor. 相似文献

To assess the role of endogenous cytokinins in growth and development of Rosa hybrida , their concentrations in bleeding sap and in roots, stem, leaves, axillary shoots and bottom breaks in three stages of development were quantified. Cytokinins were purified by means of immunoaffinity chromatography and HPLC, and identified by retention time, UV spectrum and GC-MS. The major translocation form in the xylem was zeatin riboside (ZR). In all mature tissues, cytokinins of the zeatin-type were predominant, amounting to 80–90% of the total cytokinin concentration. The stems contained high concentrations of cytokinins, probably caused by lateral movement of ZR from the xylem to adjacent stem tissue and the ability of the stem to metabolize cytokinins. In young leaves the contribution of isopentenyl adenine (iP)-type cytokinins to the total cytokinin pool was about 50%, indicating that these leaves might be capable of de novo synthesis of cytokinins. In older leaves, the concentration of an unidentified cytokinin-like compound increased to more than 50% of total cytokinins. This compound, which was also found in the roots, might be a storage form of cytokinins. In young axillary shoots, about 50% of the cytokinins are iP-compounds, suggesting either import of iP-type cytokinins via the phloem or de novo synthesis of cytokinins. In buds forming bottom breaks, ZR and zeatin riboside monophosphate (ZRMP) are the main cytokinins, indicating that these buds receive their cytokinins from the roots. 相似文献

The biomass structure and the contents of (ABA) and cytokinins (zeatin plus zeatin riboside) were studied in flowering plants of ten herbaceous species of the Middle Urals representing stress-tolerant (S) and ruderal (R) ecological strategies. The plants, which explicitly manifested S- and R-strategies, differed in ABA and cytokinin concentrations and phytohormone partitioning in particular organs. Higher ABA accumulation in the leaves and especially in the reproductive organs (here the ABA concentration exceeded by several times that of cytokinins) was characteristic of the R-plants as compared to the S-plants. In addition, the R-plants manifested higher cytokinin contents in the roots, whereas, in the S-plants, cytokinins dominated in the leaves and the reproductive organs (in the latter, the content of cytokinins exceeded that of ABA). The diverse hormonal status of the R- and S-species is discussed in relation to the patterns of plant growth and development and the photosynthate transport and partitioning characteristic of two ecological strategies. The authors conclude that plant ability to maintain the typical hormonal balance is instrumental in acquiring specific ecological strategies. 相似文献

The Oriental chestnut gall wasp, Dryocosmus kuriphilus Yasumatsu (Hymenoptera: Cynipidae), is a global invasive pest that causes serious damage to almost all chestnut species belonging to the Castanea genus (Fagaceae). Dryocosmus zhuili Liu et Zhu is a recently described sibling species of D. kuriphilus, which induces galls on Castanea henryi (Skan) Rehd. et Wils. There are many indigenous parasitoid species in China which play an important role in the natural regulation of their population dynamics. Wolbachia is a maternally inherited α-proteobacterium widely found in arthropods. This study screened for the presence of Wolbachia in the two chestnut gall wasps and in six parasitoid species from 12 populations, to investigate the prevalence patterns of Wolbachia in the chestnut gall wasp-parasitoid communities. We found that D. zhuili and four parasitoid species were infected with Wolbachia; among them, all individuals of the two populations of Megastigmus sp. had multiple Wolbachia infections. By using multilocus sequence types to characterize bacterial strains, three new sequence types were identified. The Wolbachia strains infecting D. zhuili (ST-507), Torymus sinensis Kamijo (ST-508), and Sycophila variegata (Curtis) (ST-508) belonged to supergroup A, whereas the Wolbachia strain infecting Megastigmus nipponicus Kamijo (ST-503) belonged to supergroup B. Our results also suggested that horizontal transmission of Wolbachia occurs between chestnut gall wasps and their parasitoids. Moreover, multiple Wolbachia infections of Megastigmus sp. may be due to gene recombination and horizontal transmission. 相似文献

利用超薄平板聚丙烯酰胺等电聚焦技术对陕西板栗4个实生居群,8个酶系统的20个位点进行了遗传多样性及遗传结构分析,结果表明:4个居群中以宝鸡居群遗传多样性最高,其P为80.0%,Ho,He分别为0.478和0.343,4个居群的居群遗传分化度Gst为6.2%,平均总遗传多样性为0.3372,总遗传多样性的93.8%属于居群内的遗传变异,聚类分析结果表明,以秦岭为界可以分为秦岭以南与秦岭以北2个略有差异的板栗自然亚分布区。 相似文献

Tentative identification of the cytokinins present in extracts of Dianthus caryophyllus ovaries using High Performance Liquid Chromatography and Radioimmunoassay techniques, revealed the presence of trans-ribosylzeatin, trans-zeatin, dihydrozeatin and N6 (2-isopentenyl)adenine. In addition slow moving compounds (paper chromatography) which could be hydrolysed by -glucosidase were also detected. The study presents the comparative analyses of endogenous contents of auxin (IAA), cytokinins (CKs), polyamines (PAs), and
phenolic acids (PhAs) in apical and basal parts of elm multiplicated shoots with regard to the organogenic potential. The
shoot-forming capacity was higher in the apical part than in the basal part. However, the timing of root formation was in
the apical type of explant significantly delayed (compared with the organogenic potential of basal part). Significantly higher
contents of free bases, ribosides and ribotides of isopentenyl adenine, zeatin and dihydrozeatin that were found in the apical
segments, might be considered as the most important factor affecting in vitro shoot formation. The content of endogenous free IAA was approximately three times higher in the basal shoot parts than in
the apical parts. The amounts of putrescine and spermidine were higher in the apical part which generally contains less differentiated
tissues than the basal part of shoot. The predominant PhA in both types of explants was caffeic acid, and concentrations of
other PhAs decreased in the following order: p-coumaric, ferulic, sinapic, vanillic, chlorogenic, p-hydroxybenzoic and gallic acids. The contents of all determined PhAs in their free forms and higher contents of glycoside-bound
p-coumaric, ferulic and sinapic acids, precursors for lignin biosynthesis, were found in the basal parts. 相似文献
