Diamine oxidase was purified separately from cotyledon and embryo of pea seedlings germinated for 6 days. The Km of the cotyledon enzyme for putrescine was 1.6 × 10?4M while that for the embryo enzyme was 9 × 10?5M. On heating for 15 min at 70° the embryo enzyme retained about 90% activity whereas the cotyledon enzyme retained only 20% activity. The electrophoretic mobility of the cotyledon enzyme was ca twice that of the enzyme from embryo.  相似文献

The activity of cotyledon and embryo diamine oxidase was reduced by feeding ethrel and chloroethanol to the seedlings. The inhibitory effect of 2,4-D on the activity of enzyme in the cotyledon which may be mediated through ethylene was reversed by exposure of seeds to red light.  相似文献

Biochemical, electrophoretic and immunological studies were made among peroxisomal enzymes in three organs of soybean [Glycine max (L.) Merr. cv. Centennial] to compare the enzyme distribution and characteristics of specialized peroxisomes in one species. Leaves, nodules and etiolated cotyledons were compared with regard to several enzymes localized solely in their peroxisomes: catalase (EC 1.11.1.6), malate synthase (EC 4.1.3.2), glycolate oxidase (EC 1.1.3.1), and urate oxidase (EC 1.7.3.3). Catalase activity was found in all tissue extracts. Electrophoresis on native polyacrylamide gels indicated that leaf catalase migrated more anodally than nodule or cotyledon catalase as shown by both activity staining and Western blotting. Malate synthase activity and immunologically detectable protein were present only in the cotyledon extracts. Western blots of denaturing (lithium dodecyl sulfate) gels probed with anti-cotton malate synthase antiserum, reveal a single subunit of 63 kDa in both cotton and soybean cotyledons. Glycolic acid oxidase activity was present in all three organs, but ca 20-fold lower (per mg protein) in both nodule and cotyledon extracts compared to leaf extracts. Electrophoresis followed by activity staining on native gels indicated one enzyme form with the same mobility in nodule, cotyledon and leaf preparations. Urate oxidase activity was found in nodule extracts only. Native gel electrophoresis showed a single band of activity. Novel electrophoretic systems had to be developed to resolve the urate oxidase and glycolate oxidase activities; both of these enzymes moved cathodally in the gel system employed while most other proteins moved anodally. This multifaceted study of enzymes located within three specialized types of peroxisomes in a single species has not been undertaken previously, and the results indicate that previous comparisons between the enzyme content of specialized peroxisomes from different organisms are mostly consistent with that for a single species, soybean.  相似文献

Diamine oxidase present in the cotyledons of germinating pea seeds is induced by putrescine, spermidine and ornithine. Auxins inhibit enzyme synthesis in cotyledons only in the presence of embryo. Cycloheximide inhibits the synthesis of the cotyledon enzyme but has no effect on the embryo enzyme. 5-Fluorouracil inhibits the synthesis of both cotyledon and embryo enzymes.  相似文献

Summary A difference was observed in the effect of difluoromethlyornithine (DFMO), a specific inhibitor of ornithine decarboxylase, on human and murine granulocyte-macrophage precursor cell (CFU-C) proliferation in vitro, in the presence of fetal bovine serum (FBS) and horse serum (HS). A dose of DFMO which almost totally abolished CFU-C colonies in cultures containing FBS had no effect or very little effect on CFU-C in cultures supplemented with HS. This effect could be reversed by aminoguanidine reacting with diamine oxidase (DAO), which is present in FBS but not in HS. The importance of DAO in the assessment of polyamine effects is also suggested by decreased colony formation in cultures containing HS and DFMO only after the addition of this enzyme. Additionally, Mo T cell line cultures containing DFMO demonstrated a substantially lower intracellular concentration of putrescine in the presence of FBS rather than HS. Endopeptidase activity against azocasein had a higher temperatureoptimum (50°C) in leaf extracts than in cotyledon extracts(37°C). The temperature optima for aminopeptidase (46°C)and for carboxypeptidase (53°C) were similar in leaf andcotyledon extracts. The endopeptidase activity showed an excellentstability in crude extracts from leaves even at 37°C, whilethe endopeptidase in cotyledon extracts was less stable. Carboxypeptidasewas very stable in both leaf and cotyledon extracts. Aminopeptidasewas the least stable of the enzymes investigated and its inactivationrate depended on the source of the extract. A moderate stabilitywas observed in extracts of leaves or of ungerminated seeds,but this enzyme was rapidly inactivated in cotyledon extractsat pH 5.4. At pH 7.5 aminopeptidase remained active longer thanat pH 5.4. From experiments with mixed extracts it could beconcluded that in cotyledons an aminopeptidase inactivatingfactor was formed during germination. This factor was heat sensitive,excluded by Sephadex G-25, precipitated by 75% ammonium sulfateand inhibited by tosyl-L-lysine chloromethyl ketone. These datasuggest that the factor is a protein and considering the similarproperties it appears possible that it is the endopeptidaseformed during germination. Leptosphaeria maculans , the causal agent of blackleg of canola, produces polygalacturonases during infection. Stem extracts of spring and winter canola cultivars contained a water-soluble inhibitor of the polygalacturonase activity of L. maculans . The polygalacturonase inhibitor material had different characteristics dependent upon the cultivar. Some canola cultivars had a polygalacturonase inhibitory compound(s) which was heat liable, low molecular weight and required divalent cations, and other cultivars had a heat stable, low molecular weight compound(s). The cultivar Maluka had a unique polygalacturonase inhibitory compound(s) that was heat labile, low molecular weight and did not need divalent cations. The level of the polygalacturonase inhibitory activity in the stem extracts was significantly related to the resistance of the cultivars to L. maculans as measured by the rate of lesion elongation, but was less related to the rate of stem girdling. The significant correlation between levels of polygalacturonase inhibitor activity and stem resistance in canola cultivars indicates that polygalacturonase inhibitors may be involved in the resistance of stems to blackleg. The two quantitative measures of stem resistance, rate of lesion elongation and rate of stem girdling, were significantly correlated to cotyledon resistance and to each other.  相似文献

利用抑制物生物测定法和酸蚀技术研究了血皮槭种子休眠的原因。血皮槭种子吸水是一个非常缓慢的过程,在140 h以后种子含水量才能达到68%左右。酸蚀处理种子3 h,虽然没有加快种子的吸水速率,但能较好得使果皮变薄,也不影响种子的生活力。种子的各部位(果皮、种皮、子叶、胚根)均含有抑制物质,对小白菜种子的发芽率及胚根生长有很强的抑制作用,子叶各种处理水浸提液的抑制作用最强,果皮和种皮次之。血皮槭种子休眠主要由种壳机械障碍和种胚生理休眠两重因素导致,因此如何克服致密果壳而使激素能接触生理休眠的种胚是打破其种子休眠的关键技术。  相似文献

The inhibitory effect of the polyamines, spermidine and spermine, on the proliferation of human fibroblasts in culture was found to be reversed by the addition of aminoguanidine (AM), a specific and highly effective inhibitor of diamine oxidase (DAO) present in fetal calf serum (FCS). Aminoguanidine itself in concentration as high as 10(-3) M exhibited no effect upon cell proliferation nor did putrescine at similar concentrations. However, at higher concentrations of putrescine, cell proliferation was inhibited and this inhibition was unaffected by the addition of mM concentrations of AM. These studies support earlier hypotheses on the mechanisms of the toxic effects of polyamines on cell proliferation and establish further that the diamine oxidase-catalyzed metabolism of spermine and spermidine is necessary for their toxic effects in cell culture.  相似文献

The activities of adenosylhomocysteinase (EC 3.3.1.1) and adenosine nucleosidase (EC 3.2.2.7) were assayed in extracts from yellow lupin (Lupinus luteus L.) cotyledons at different stages of seed formation and seedling development. Adenosylhomocysteinase activity was demonstrated in all the cotyledon extracts examined. Its lowest level was found in the dry seeds and the highest, in 4-day-old seedling cotyledons. Extracts from the cotyledons of maturating seeds, dry seeds, and seedlings up to the second day of growth exhibited no adenosine nucleosidase activity. Adenosine nucleosidase activity appeared in the cotyledons of 2-day-old seedlings and its highest level was reached in 4-to 5-day-old seedlings. There is no inhibitor of adenosine nucleosidase in the maturating and dry yellow lupin seeds. No activator of a possible zymogen form of adenosine nucleosidase from maturating or dry seeds occurs in the growing seedlings.  相似文献

A Cotyledonary Inhibitor of Root Nodulation in Pisum sativum   总被引:2,自引:0,他引:2  
Root nodule formation was studied in 11-day-old seedlings of Pisum sativum L. cv. Alaska, which were inoculated with Rhizobium leguminosarum at the time of planting. On intact plants an identical nodulation pattern was observed in the lateral roots attached to the two xylem strands connected to the cotyledons. When one cotyledon was removed before germination, however, a highly significant increase in nodultion was observed on the lateral roots attached to the xylem strand which no longer was connected to a cotyledon. Excision of one cotyledon also caused an alteration in the radial location of root nodules on the primary root. Under these conditions there was a distinct promotion of nodular proliferations in the root cortex opposite the primary phloem poles. The fact that removal of one cotyledon increased nodulation on the lateral roots but had no effect on the rhizobial infection of lateral root hairs suggested that a cotyledonary inhibitor acts at a step between the infection process and the appearance of a macroscopic nodule. The data were interpreted in terms of an inhibitor of cortical cell division which is translocated from the cotyledons to the developing root via the phloem.  相似文献

Ethylene production during seed development in Brassica napus occurs first at 20 d after pollination (DAP), while a second greater peak occurs at 35 DAP. Because of the inaccessible location of the embryo within the maternal tissue, microspore-derived embryos (MDEs) of B. napus were used as a model for studying the role of ethylene during embryo development. The MDEs also produced a peak in ethylene evolution at 20 DAC (i.e. the early cotyledonary stage), dropping to minimal levels by 25-30 DAC. At 20 DAC the excised cotyledon evolved 85% of the ethylene found in the whole MDE. To determine the role of ethylene, MDEs were treated with aminoethoxyvinylglycine (AVG, an inhibitor of ethylene biosynthesis), CoCl(2) (an inhibitor of 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase), and silver thiosulphate (STS, an inhibitor of ethylene action). An inhibition in ethylene production or action at 20 DAC resulted in diminished lateral cotyledon expansion, due to a reduction in the lateral expansion of cells within the cotyledon. Recovery to 'control-type' levels of cotyledon cell expansion was achieved by application of ACC (the metabolic precursor of ethylene) to AVG-treated MDEs. Thus, ethylene production at 20 DAP likely controls cotyledon expansion during embryo development.  相似文献

Twelve peptides, including eight conservative amino acid residues in the amino acid sequence of hydrophilic S helix of the alternative oxidase (AOX), were synthesized by solid-phase method. The polypeptide was coupled to αchymotrypsinogen, and the antibodies against this complex were obtained in rabbit. By using these antibodies, which were raised to immunoreact with total proteins of purified mitochondria from different organs of mung bean (Phaseolous radiatus L.) seedlings, it was found that there were two hybridizable AOX fractions in the mitochondria of mung bean seedlings. Their molecular weight was about 35 kD and 38 kD, respectively. Moreover, among the respiratory parameters obtained in hypocotyl, true leaf and cotyledon of mung bean seedlings true leaf had the highest total respiration (Vt), alternative pathway (AP) capacity(Valt) and the activity of AP (ρValt). Hypocotyl Vt and ρValt were the lowest, but its Vt was higher than that of the cotyledon. The activities of total and cyanide-resistant respiration were consistant with the analysis of Western blotting of AOX expression. The highest Vt and ρValt in true leaf were accompanied by two hybridizable polypeptides of AOX protein. The next was cotyledon Vt and ρValt with only one 38 kD hybridizable polypeptide of AOX protein. Hypocotyl Vt and ρValt were the lowest and its immunobloting band was similar to that of the cotyledon, but the expression amount of 38 kD protein was less than that of the cotyledon. The 35 kD AOX may make the main contribution to the true leaf ρValt.  相似文献

The effect of proline, isoleucine, leucine, valine, lysine and ornithine under standard physiological conditions, on purified Vigna catjang cotyledon and buffalo liver arginases was studied. The results showed that V. catjang cotyledon arginase is inhibited by proline at a lower concentration than buffalo liver arginase and the inhibition was found to be linear competitive for both enzymes. Buffalo liver arginase was more sensitive to inhibition by branched-chain amino acids than V. catjang cotyledon. Leucine, lysine, ornithine and valine are competitive inhibitors while isoleucine is a mixed type of inhibitor of liver arginase. We have also studied the effect of manganese concentration which acts as a cofactor and leads to activation of arginase. The optimum Mn2+ concentration for Vigna catjang cotyledon arginase is 0.6 mM and liver arginase is 2.0 mM. The preincubation period required for liver arginase is 20 min at 55 degrees C, the preincubation period and temperature required for activation of cotyledon arginase was found to be 8 min at 35 degrees C. The function of cotyledon arginase in polyamine biosynthesis and a possible role of branched chain amino acids in hydrolysis of arginine in liver are discussed.  相似文献

顽拗性种子脱落时具有较高的含水量和代谢活性, 对脱水高度敏感; 但顽拗性种子脱水敏感性的机理至今仍然不清楚。该文以顽拗性黄皮(Clausena lansium)种子为材料, 研究了种子和胚轴对水分丧失的响应, 在脱水过程中胚轴和子叶的呼吸速率, 胚轴和子叶线粒体的细胞色素c氧化酶(CCO)活性、外膜完整性、CCO和交替氧化酶(AOX)途径以及线粒体活性氧清除酶活性的变化。结果表明, 随着水分的丧失, 种子和胚轴的存活率逐渐下降, 种子的脱水敏感性大于胚轴; 胚轴和子叶的呼吸速率以及线粒体外膜的完整性降低。胚轴和子叶线粒体的CCO途径以及胚轴AOX途径的呼吸速率在脱水初期增加, 随着继续脱水下降, 胚轴线粒体AOX途径的呼吸速率则随着脱水显著下降。胚轴线粒体的超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)和谷胱甘肽还原酶(GR)活性和子叶线粒体的APX活性随着脱水迅速下降; 胚轴线粒体的脱氢抗坏血酸还原酶(DHAR)活性和子叶线粒体的SOD、DHAR和GR活性在脱水初期增加, 然后下降。这些数据表明黄皮种子的脱水敏感性与线粒体的呼吸速率和活性氧清除酶的活性降低密切相关, 也与长期适应热带/亚热带的生境有关。  相似文献

The oxidative deamination of methylated putrescine by a diamine oxidase activity (DAO) is an important step in the biosynthesis of nicotine in tobacco and tropane alkaloids in several Solanaceous plants. A polyclonal rabbit antiserum was previously developed to a purported purified DAO enzyme from Nicotiana tabacum. The antiserum bound to a single 53 kDa protein and immunoprecipitated 80 of DAO activity from tobacco root extracts. In an effort to obtain DAO cDNAs, this antiserum was used to screen a tobacco cDNA expression library and three distinct immunoreactive cDNA clones were isolated. These cDNAs encoded predicted proteins that were either identical or nearly identical to predicted S-adenosylhomocysteine hydrolase (SAHH) from two Nicotiana species. Thus, the rabbit antiserum was not specific to DAO, even though it immunodepleted the majority of DAO activity from root extracts. Alternative hypotheses to explain the DAO immunodepletion results (such as poisoning of DAO activity or that SAHH is a bifunctional enzyme) were tested and ruled out. Therefore, we hypothesize that SAHH associates with DAO as part of a larger multienzyme complex that may function in planta as a nicotine metabolic channel.  相似文献

The Inhibition of Flowering by Non-Induced Cotyledons of Pharbitis nil   总被引:1,自引:0,他引:1  
Inhibitory effects on flowering of a non-induced cotyledon havebeen examined in Pharbitis nil seedlings. The photoperiodicinduction of one cotyledon was accomplished by wrapping it inaluminium foil for 13 to 15 h while the seedling remained inthe light. The presence of the other cotyledon in the lightblocked this inductive stimulus. The timing of its inhibitoryeffect suggested that its action was to block the expressionof the inductive stimulus, presumably at the shoot apex. Byvarying the area of the non-induced cotyledon parallel inhibitoryeffects were shown on export of stimulus and of 14C-labelledassimilate to the apex from the induced cotyledon. Thus, partof the inhibition was by interference with assimilate/stimulusco-transport in the phloem. However, an additional inhibitoryeffect was also evident and for this second component therewas no relationship between assimilate and stimulus transport.This latter inhibition was generated by brief light interruptionsof darkness given to one cotyledon only whilst the other waswrapped. The control treatment, removal of the unwrapped cotyledon,did not alter flowering compared to seedlings with intact, darkenedcotyledons. Thus, these studies show that the brief night interruptionsacted to trigger a photoperiodically sensitive inhibitor notto block induction. The implications of these findings are discussedin relation to models of time measurement in the photoperiodiccontrol of flowering. (Received March 20, 1989; Accepted November 16, 1989)  相似文献

Abstract In non-motile forms of Azospirillum lipoferum isolated from the rhizosphere of rice, polyphenol oxidase activity was observed which correlated with production of a dark-brown pigment. Using a combination of substrate/inhibitor specificity tests, intracellular enzyme extracts of non-motile strains were clearly demonstrated to have a laccase activity by oxidising various o - and p -diphenols. This work is the first report on laccase activity in Azospirillum .  相似文献

It is hypothesized that since protein α-amylase inhibitor (α-AI) and stimulator might be present together in red kidney bean (Phaseolus vulgaris L.) seeds, their in vitro interactions might influence their detection and quantification. Assay of α-AI using extracts from the embryonic axes revealed an unexpected finding in that the extracts stimulated rather than inhibited α-amylase activity. The cotyledon extracts exhibited inhibitory or enhancement effect on α-amylase activity depending on whether prior to the α-amylase assay they had been boiled for 10 min or not. Phytohemagglutinin (PHA-L in particular) is implicated in the present study as a stimulator of α-amylase activity co-extracted with α-AI from red kidney bean cotyledons. The importance of these findings is discussed in relation to the possible widespread occurrence of protein α-amylase stimulator in seeds and other plant parts.  相似文献
