The fucose-containing polysaccharides of the brown alga Dictyota dichotoma were extracted with either trichloroacetic acid or HCl to give both water-soluble and water-insoluble materials. The latter had a high proportion (16 to 11%) of protein, and although all the sugars found in the water-soluble extracts were present, the major sugar in these water-insoluble polysaccharides was glucose. The water-soluble material extracted with HCl was a protein-free sulphated heteropolysaccharide. Complete removal of a glucan from the water-soluble extract was achieved by fractional precipitation with ethanol. The recovered glucan-free sulphated polysaccharide, which was rich in glucuronic acid, galactose, fucose and sulphate, showed high anticoagulating activity.  相似文献

The inducible water-soluble bioemulsifier liposan (M. C. Cirigliano and G. M. Carman, Appl. Environ. Microbiol. 48:747-750, 1984) was purified from the yeast Candida lipolytica. The purification procedure included repeated solvent extractions of a concentrated culture filtrate and Affi-Gel concanavalin A affinity chromatography. The procedure yielded a preparation containing a major band (Mr = 27,600) which stained for protein and carbohydrate upon polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. Liposan is composed of approximately 83% carbohydrate and 17% protein. Acid and enzymatic digestions of the emulsifier revealed that the carbohydrate portion is a heteropolysaccharide consisting of glucose, galactose, galactosamine, and galacturonic acid. Liposan effected and stabilized oil-in-water emulsions with a variety of commercial vegetable oils. Emulsification and stabilization properties of liposan were compared to those of a number of commercial emulsifiers and stabilizers.  相似文献

Extraction of sunflower heads with ammonium oxalate afforded water-soluble pectin material and water-insoluble glycoprotein material, the carbohydrate portion of which consisted of galacturonic acid and xylose residues; the pectin material defied fractionation with cetylpyridinium chloride. Extraction with hydrochloric acid (pH 1.5) afforded water-soluble and water-insoluble polysaccharide materials. The former, when fractionated with cetylpyridinium chloride, gave a glycoprotein, the carbohydrate moiety of which was composed of galacturonic acid, galactose (major), glucose, arabinose, and xylose, and also a rhamnan. The latter was a glycoprotein, the carbohydrate portion of which consisted of galactose (major), glucose, xylose, and rhamnose residues. Extraction of the sunflower heads with water also gave glycoprotein material, which was fractionated by paper electrophoresis into a glyco-protein, the carbohydrate moiety ofwhich was composed of galacturonic acid (minor), galactose, glucose, xylose, arabinose, and rhamnose (major) residues, and a heteropolysaccharide composed of galactose (major), glucose, xylose, and arabinose residues.  相似文献

从猴头菌子实体中分离得到一种新型的水溶性杂多糖HEPF2,分子量大小为1.66′104Da,该多糖由岩藻糖、半乳糖和葡萄糖以1.00:3.69:5.42比例构成,同时也含有微量的3-O-甲基鼠李糖。进一步利用傅立叶变换红外光谱法、糖组成分析、甲基化分析、部分酸水解法和核磁共振法等方法进行结构鉴定,检测结果表明,该杂多糖中包含1→4、1→6结合的葡萄糖和1→6结合的半乳糖残基,连接于主链的侧链残基,包括岩藻糖残基、少数的端基葡萄糖和半乳糖残基。核磁共振法检测结果还表明,1→4结合葡萄糖为β构型,(1→6)结合半乳糖、(1→2,6)结合半乳糖和端基葡萄糖均为α构型。  相似文献

A new sulphated heteropolysaccharide containing glucuronic acid, mannose, galactose, xylose, fucose and a protein moiety has been extracted from Sargassum linifolium. The acid-extractable, water-soluble, sulphated heteropolysaccharide, sargassan, contains residues of D-glucuronic acid, D-mannose, D-galactose, D-xylose, and L-fucose, and a protein moiety. Partial, acid hydrolysis of sargassan and auto-hydrolysis of the free-acid polysaccharide have been studied. Several acidic and neutral oligosaccharides were subsequently isolated. Evidence is advanced for the presence of ester sulphate on some galactose and fucose residues. It is concluded that the carbohydrate moiety of sargassan involves a backbone chain of D-glucuronic acid and D-mannose residues, and side chains involving residues of D-galactose, D-xylose, and L-fucose with sulphate attached to some galactose and fucose residues.  相似文献

A novel acidic heteropolysaccharide (TAPA1) was purified from hot water extracts of Tremella aurantialba fruiting bodies by DEAE-Sepharose Fast Flow and Sephacryl S-500 High-Resolution Chromatography. The heteropolysaccharide had a molecular weight of ca. 1.35 × 106 Da, and a carbohydrate content estimated to be ∼98.7% by the phenol-sulfuric acid method. It was composed mainly of d-mannose, d-xylose, and d-glucuronic acid in the ratio of ca. 5:4:1, along with trace amounts of d-galacturonic acid and d-glucose. Monosaccharide compositional analysis and GC-MS of methylated derivatives, combined with 1H and 13C NMR spectroscopies (including COSY, TOCSY, NOESY, HSQC, and HMBC spectra), revealed TAPA1 to consist of an α-(1→3)-linked mannopyranosyl backbone, partially substituted at position 4 with xylose side chains, and at position 2 with side chains composed of either xylose, mannose, and glucuronic acid or of xylose and mannose. Bioactivity testing showed that TAPA1 stimulated the proliferation of mouse spleen lymphocytes in vitro.  相似文献

The extracellular polysaccharide of Coscinodiscus nobilis, a member of the Coscinodiscaceae, contains a highly branched heteropolysaccharide(s) containing fucose, rhamnose, mannose, d-glucose, xylose, d-glucuronic acid, galactose (trace) and half ester sulphate. The positions of linkages between the monosaccharides have been established and evidence for the linkages between d-glucuronic acid and monosaccharides was obtained. The extracellular polysaccharide contained also a chrysolaminaran, but this may have been derived from dead cells. Fucose and mannose occur also in a separate polymer. The diatom contained polysaccharide material consisting of glucose, mannose, fucose and uronic acid residues.  相似文献

Galactomannan, a water-soluble heteropolysaccharide, was isolated from the seed of Far-Eastern population of ground honeysuckle Lotus corniculatus L. (yield, 1.65%). Analysis of this galactomannan showed that is consists of D-mannose and D-galactose residues (molar ratio, 1.22:1). Its aqueous solutions were characterized by specific rotation [alpha]D = +84.10 and characteristic viscosity [eta] = 559 ml/g. Analysis of this heteropolysaccharide using chemical and enzymatic procedures, as well as IR- and 13C-NMR spectroscopy, showed that its main chain comprises 1,4-beta-D-mannopyranose residues, 95.5% of which are substituted at C-6 with single residues of alpha-D-galactopyranose.  相似文献

A novel water-soluble heteropolysaccharide FVP60-B was extracted from the fruiting bodies of Flammulina velutipes with boiling water and purified by Sephacryl S-300 and S-400, which molecular weight was estimated to be 1.3 × 104 Da by HPLC. It is composed of l-fucose, d-mannose, d-glucose and d-galactose in a ratio of 1.16:0.82:1.00:3.08. Sugar analysis, methylation analysis together with 1H, 13C and 2D NMR spectroscopy disclosed that FVP60-B is consisted of a α-(1 → 6)-d-galactopyranan backbone with a terminal fucosyl, terminal glucosyl and α-(1 → 6)-d-mannopyranan units on O-2 of 2,6-O-substituted-d-galactosyl units.  相似文献

A sulphated heteropolysaccharide (~15% of the acid-extractable material) isolated from the brown alga Dictyota dichotoma contains residues of D-glucuronic acid, D-galactose, D-mannose, D-xylose, and L-fucose1. Partial hydrolysis of the polysaccharide with acid gave one neutral and two acidic oligosaccharides. The behaviour towards periodate of the polysaccharide before and after partial hydrolysis, alkali-treatment, and methanolysis has been studied. Evidence is thereby provided that the polysaccharide is partially sulphated and composed of (1→4)-linked residues of D-glucuronic acid, D-galactose, D-mannose, and D-xylose, and (1→2)-linked L-fucose.  相似文献

The structure of rabbit transferrin was investigated with regard to number, size, and composition of the heteropolysaccharide units and their relative location on the polypeptide chain. The composition and molecular weight of the Pronase glycopeptides revealed that rabbit transferrin contains two heteropolysaccharide units, each composed of 2 sialic acid residues, 2 galactose residues, 3 mannose residues, and 4-N-acetylglucosamine residues. The composition and molecular weight of the tryptic glycopeptides further substantiated the existence of two identical heteropolysaccharide units and revealed that both units have identical amino acid residues in the immediate vicinity of the carbohydrate attachment sites to the polypeptide chain, suggesting a sequence homology surrounding the two glycosylation sites. Characterization of the cyanogen bromide fragments from rabbit transferrin indicated that both heteropolysaccharide units are located within a single polypeptide fragment representing approximately one-third of the molecule.  相似文献

Cora pavonia, a lichen having a basidiomycetous mycobiont, is rich in protein (36%), of which 10% is tyrosine. α,α-Trehalose is present and was isolated in 4.4% yield. The lichen was found to contain polysaccharides typical of basidiomycetes and different from those of ascomycetes and ascomycetous lichens. Isolated and characterized were a β-d-glucan and a heteropolysaccharide containing l-rhamnose, l-fucose, d-xylose, d-mannose, d-glucose, and d-galactose. The β-d-glucan was highly branched with 21% of nonreducing end-groups, contained 3-O-, 6-O-, and 3,6-di-O-substituted β-d-glucopyranosyl units, and had a main chain consisting of (1→3)- and (1→6)-links. The heteropolysaccharide component contained mainly mannose and xylose, having a mannose-containing nucleus and a main chain with preponderant (1→3)-linked α-d-mannopyranosyl residues. These were unsubstituted (10%), and 4-O- (10%) and 2,4-di-O-substituted (10%) with residues of β-d-xylopyranose. On methylation analysis of the heteropolysaccharide, a capillary column of DB-210 proved to be particularly useful for gas-liquid chromatographic resolution of partially O-methylated alditol acetates.  相似文献

Several benthic cyanobacteria were found to produce significant amounts of extracellular flocculants. The macromolecular flocculants produced by Phormidium sp. strain J-1 and Anabaenopsis circularis PCC 6720 were characterized. The Phormidium flocculant is a sulfated heteropolysaccharide to which fatty acids and protein are bound. The polysaccharide backbone is composed of uronic acids, rhamnose, mannose, and galactose. The A. circularis flocculant is also an acidic polysaccharide containing keto acid residues and neutral sugars, but to which no fatty acids, proteins, or sulfates are linked. Both flocculants could be recovered from growth medium by precipitation with cetyltrimethylammonium bromide and were found to bind the cationic dye Alcian-blue in a linear proportion to their concentration in solution. The latter property was used to quantify flocculant concentrations in culture supernatants and natural water samples and to compute their anion densities.  相似文献

A novel water-soluble polysaccharide pMTPS-3, obtained from Melia toosendan Sieb. Et Zucc fruit by hot-water extraction and ethanol precipitation, was fractionated by DEAE-52 cellulose anion-exchange and Sephadex G-100 gel filtration chromatography. Its primary structural features and molecular weight were characterized by Fourier infrared spectrometry (FTIR), gel permeation chromatography (GPC) and gas chromatography (GC). And the antioxidant activities of pMTPS-3 in vitro were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, superoxide radical scavenging assay and hydroxyl radical scavenging assay. The results suggested that pMTPS-3 was a heteropolysaccharide, composed of arabinose, glucose, mannose, and galactose in the molar ratio of 17.3:28.3:41.6:12.6 with molecular weight 26 100 Da. The purified pMTPS-3 was revealed to have notable scavenging activity against DPPH radical in a concentration-dependent manner and present a moderate inhibition of superoxide radicals with an IC50 (5.6 mg/ml), and potent inhibiting power for hydroxyl radical compared with crude polysaccharide. Further, it exhibited strong inhibition effect in vitro on the growth of human gastric cancer BGC-823 cells. It is strongly evidenced that pMTPS-3 purified from the crude polysaccharides of Melia toosendan Sieb. Et Zucc could be explored as a potential antioxidant and therapeutics.  相似文献

Extraction with hydrochloric acid (pH 2.5) of the brown alga Padina tetrastromatica afforded water-soluble and water-insoluble polysaccharides. The water-soluble polysaccharide was fractionated using cetyltritmethyl ammonium bromide and chromatography on DEAE-cellulose and Sephadex G-100. A neutral laminaran like glucan and two new sulphated heteropolysaccharides comprising d-glucuronic acid, l-fucose, l-rhamnose, d-xylose, d-arabinose, d-galactose, d-glucose and half-ester sulphate were obtained. The alginic acid isolated from this brown seaweed was found to be predominantly of poly 1 → 4β-d-mannuronic acid type. The water-soluble sulphated polymer showed high anticoagulant activity.  相似文献

Protoplasts of Aureobasidium pullulans are capable of producing pullulan. Biosynthesis of the polymer pullulan required induction with kinetics similar to those of whole cells. The protoplasts also produced a heteropolysaccharide component containing mannose, glucose, and galactose. The relative proportions of the pullulan and heteropolysaccharide fractions were a function of glucose concentration, with the pullulan content of the total polysaccharide rising from 20% at 2.5 mM glucose to 45% at 20 mM glucose. Elaboration of pullulan by both cells and protoplasts was sensitive to 0.6 M KCl, which was present as the osmotic stabilizer in protoplast experiments. The presence of KCl resulted in a shift in the pH optimum to a more acidic value. The molecular weight of the protoplast-derived pullulan was sharply reduced from the molecular weight of the whole-cell-derived product. Exposure of the protoplasts to proteolytic enzymes had no effect on polysaccharide elaboration.  相似文献

Pectin is a heteropolysaccharide which has been investigated for the development of colon-specific drug delivery systems. Polymers have been associated with pectin to reduce its aqueous solubility and improve the performance of drug delivery systems. Pectin–casein interaction is widely known in food research, but it has not been fully considered by pharmaceutical scientists. Thus, this study investigated the potential of casein–pectin microparticles as a drug delivery system and clarified the impact of cross-linking and drying methods on the in vitro release of indomethacin (IND) or acetaminophen (PCT) from microparticles. Microparticles were prepared by coacervation and dried by spray or spouted bed methods. Drug recovery, in vitro drug release, size, morphology, and the thermal and diffractometric properties of dried microparticles were determined. Spray-dried non-cross-linked microparticles were able to prolong IND release, and pectin was still degraded by pectinolytic enzymes. On the other hand, glutaraldehyde cross-linking prevented the enzymatic breakdown of pectin without improving IND release. Spouted bed drying reduced IND recovery from all microparticles when compared with spray drying, thus the successful spouted bed drying of microparticles depends on the chemical characteristics of both the drug and the polymer. Release data from PCT microparticles suggested that the microparticle formulation should be improved to bring about a more efficient delivery of water-soluble drugs. In conclusion, casein–pectin microparticles show great potential as a drug delivery system because casein reduces the water solubility of pectin. The drying method and cross-linking process had significant effects on the in vitro performance of these microparticles.  相似文献

The bruchid beetles Acanthoscelides obtectus (Say) and Zabrotes subfasciatus Boheman, are major storage pests of the economically important legume Phaseolus vulgaris (haricot bean). In the present study, resistance of a wild line of P. vulgaris G12953, towards these two insects is confirmed. Mature seeds from both resistant and susceptible lines were fractionated and tested in feeding trials in an attempt to elucidate the basis of resistance. The heteropolysaccharide fraction isolated from the resistant line and a susceptible line were each incorporated into artificial beans over a concentration range up to 10% dry wt. At a concentration of 4%, the approximate physiological concentration within the seed, the heteropolysaccharide fraction from the resistant line was very toxic resulting in 80–85% larval mortality of A. obtectus with an LC50 of 2.5%. Furthermore, surviving larvae showed a marked increase in their development period. The corresponding fraction from a susceptible line, on the other hand, even at twice the physiological concentration, had a negligible effect upon larval development both in terms of adult emergence and development period. The effects of albumin proteins and globulin proteins on larval development of A. obtectus were also tested by incorporation into artificial beans at a range of concentrations up to 10%. Whilst those from the susceptible line had little effect upon development both the albumins and globulins from the resistant line were found to cause a slight decrease in adult emergence and a small increase in the median development period. These effects, however, are not sufficient to account for the observed seed resistance of G12953. Mature seeds from the resistant line and nine susceptible lines were also screened for the following secondary plant compounds: proteinase inhibitors, α-amylase inhibitors, lectins (phytohaemagglutinins), saponins, tannins and alkaloids. Although some of these antimetabolites were found to be present none, except for α-amylase inhibitors, were thought to play any significant role in conferring seed resistance. It is concluded that this example of insect resistance in seeds of P. vulgaris, line G12953, is due, at least in part, to the presence of a heteropolysaccharide which has an unusually high arabinose and fucose content. In the absence of other antimetabolic compounds the levels of this heteropolysaccharide present in G12953 are sufficiently high to account for the observed resistance. However, it is possible regarding seed resistance towards Z. subfasciatus, that α-amylase inhibitors may be having a synergistic effect.  相似文献
