N-Feruloylglycyl-l-phenylalanine was obtained from barley globulins by partial hydrolysis with 4 N HCl. It was isolated by means of preparative and ‘multiple elimination’ TLC (METC) and further identified by TLC comparison with a synthetic sample. Additional proof for its identity was obtained by UV, fluorescence and IR spectroscopy and by the action of carboxypeptidase A. The possible role of N-feruloylglycine as a starter in protein biosynthesis in the barley seed is discussed. 相似文献

Basic c-type cytochromes homologous with plant and animal mitochondrial cytochrome c have been isolated and purified from Rhodymenia palmata and Porphyra umbilicalis. The N-terminal regions have been analysed using a Beckman 890C automatic sequencer. When compared to animal cytochrome c, the Rhodymenia cytochrome c has an unblocked N-terminal tail of 10 amino acids, whereas Porphyra has an unblocked N-terminal tail of only a single amino acid. 相似文献

The amino acid sequence of plastocyanin from dock has been completed. It is a single polypeptide chain of 99 residues which is closely related to other plant plastocyanins. The amino acid sequence of the ferredoxin from Sambucus nigra consists of a single polypeptide chain of 97 amino acid residues, 5 of which are cysteine. The positions of the 4 cysteine residues which bind the iron atoms of the active centre are identical to those of other ferredoxins. Due to difficulties of obtaining pure protein, residues 87–90 have only been identified from the amino acid analysis of peptide C 10 and by homology with other higher plant ferredoxins. 相似文献

The amino acid sequence of cytochrome c from niger-seed has been determined by sequence analysis of chymotryptic and tryptic peptides using the dansyl-phenylisothiocyanate method and by qualitative analysis of peptide composition by the dansyl method. Although the spectral ratios indicated the protein was not completely pure, no indication of impurity was found during the sequence analysis and no peptides in addition to those given here were obtained. In certain cases the alignment of peptides was by homology with other cytochromes c. Four residues in the proposed sequence, alanine-1, cysteine-25, histidine-26 and lysine-61 were identified only from peptide compositions. The amino-terminus of the protein is acetylated. The sequence contains two residues of ϵ-N-trimethyllysine. 相似文献

Proposed amino acid sequence of cytochrome c from Enteromorpha intestinalis is presented. The cytochrome is a basic protein, homologous with higher plant and animal cytochromes c of mitochondrial origin. Peptides from chymotryptic and tryptic digests were analysed by the dansyl-phenylisothiocyanate method and aligned by comparison with other homologous cytochromes c. The proposed sequence consists of a single polypeptide chain of 111 residues which is acetylated at its N-terminus. The amino acid sequence of plastocyanin from lettuce (Lactuca sativa L.) was determined by using a Beckman 890C automatic sequencer and by dansyl-phenylisothiocyanate analysis of peptides obtained by enzymic digestion of purified CNBr fragments. The protein consists of a single polypeptide chain of 99 residues, and shows close homology with other higher plant plastocyanins. The complete amino acid sequence of one of the major wheat protein iso-inhibitors of α-amylase was determined. The sequence was deduced from analysis of fragments and peptides derived from the protein by cleavage with cyanogen bromide and by digestion with trypsin, chymotrypsin, thermolysin and the Staphylococcus aureus V8 protease. The molecule consists of a single polypeptide chain of 123 residues. Both serine and alanine were found in position 65, and further minor examples of microheterogeneity were observed in four other residues. 相似文献

The decarboxylation of (2S)-glutamic acid to yield γ-aminobutyric acid catalysed by L-glutamic acid decarboxylase (EC 4.1.1.15) from Hordeum vulgare proceeds with net retention. The result is interpreted in terms of a single progenitor hypothesis of the pyridoxal phosphate enzymes and confirms that not only bacteria and animals but also plant decarboxylases catalyse the biosynthesis of biogenic amines from amino acids with net retention. 相似文献

The amino acid sequence of Epiteromorpha cytochrome c has been added to an affinity tree relating the cytochrome c sequences of animals, plants, fungi, protozoans and one bacterium, cytochrome c2 from Rhodospirillum. The Enteromorpha sequence lies on the line of descent of the higher plant sequences; it is not closely related to the cytochrome c of the photosynthetic protozoan, Euglena. A new C-glycosylflavone isolated from corn silk inhibits the growth and development of the corn earworm, Heliothis zea. This new compound was shown to be a 2″-O-α-l-rhamnosyl-6-C-(6-deoxy-xylo-hexos-4-ulosyl)luteolin. Ferredoxin from Arctium lappa consists of a single polypeptide chain of 97 residues, four of which are cysteine. These residues, which are in the active centre, are in identical positions to those of other ferredoxins. Barley grains contain hydrocarbons, including a material indistinguishable from ent-kaurene by GLC, and which after appropriate chemical conversions contain material behaving like ent-kauran-16,17-diol, ent-kaurene norketone and ent-17-nor-kaurane on TLC and GLC. The presence of ent-kaurene was confirmed by conversion to ent-kauran-16-ol and, following formation of acetate-[3H], recrystallization to constant specific activity with unlabelled carrier. In the initial ca. 15 hr of germination, preceding the rise in endogenous gibberellins, the level of ent-kaurene falls. Exogenous ent-kaurene-[14C] was not metabolized by intact barley grains. ent-Kauran-16,17-epoxide was formed non-enzymically by boiled extracts. Unboiled homogenates also formed ent-kauran-17-ol and ent-kauran-16,17-diol. The diol appeared to be formed from the epoxide, but the ent-kauran-17-ol was not. No recognized gibberellin precursors were detected. Nevertheless, endogenous ent-kaurene may be the stored biosynthetic precursor of gibberellins in germinating barley grains. 相似文献

Gibberellic acid (GA3) and 13-deoxy-gibberellic acid (GA7) were identified in extracts of germinating barley as their 14C-methyl esters. The maximal level of GA3 was estimated by an isotopic dilution procedure to be 1·5 ng per grain. Germinating barley incorporated 2-14C-mevalonic acid into several terpenes, whose specific radioactivities were measured, but incorporation into GA3 could not be detected. Cell-free embryo extracts from germinating barley converted 2-14C-mevalonic acid into isopentenol, dimethylallyl alcohol, farnesol and squalene, while 14C-isopentenyl pyrophosphate was incorporated into geraniol, farnesol, geranylgeraniol and squalene. There was no detectable incorporation into the gibberellin intermediate ent-kaurene. 相似文献
