During a chemosystematic survey of 38 representative species of the Juncaceae for leaf and stem flavonoids, the 5-methyl ether of luteolin was discovered for the first time in plants. It occurs both free and as the 7-glucoside; its identity was confirmed by synthesis. Flavone sulphates were also found in the family and the 7-glucosidesulphates of luteolin and chrysoeriol were characterised for the first time. 7,3′,4′-Trihydroxyflavone and its 7-glucoside, not previously reported in the monocotyledons, were found in three species. The presence of luteolin 5-methyl ether or its glucoside in 70% of the species surveyed serves to distinguish the Juncaceae from the morphologically related Centrolepidaceae, Restionaceae and Thurniaceae. Flavone C-glycosides, common in grasses and sedges, were found only in Prionium, a genus which on anatomical grounds is anomalous in the Juncaceae. Among other phenolics detected during the survey, the uncommon 7,8-dihydroxycoumarin, daphnetin, was identified in Juncus effusus and its 8-methyl ether in four Luzula species. Taken together, these chemical findings show that the Juncaceae are very distinctive in their phenolic pattern and confirm the correctness of assigning them an isolated position in a separate order, the Juncales. The results indicate that the Juncaceae are chemically specialized, in spite of the facts that the family has been regarded as ancestral to the Cyperaceae and Gramineae and that they have been assigned a low advancement index by Sporne.  相似文献

Daphnetin, a plant-derived dihydroxylated derivative of coumarin, is an effective compound extracted from a plant called Daphne Korean Nakai. Coumarin derivates were known for their antithrombotic, anti-inflammatory, and antioxidant activities. The present study was aimed to determine the immunosuppressive effects and the underlying mechanisms of daphnetin on concanavalin A (ConA) induced T lymphocytes in mice. We showed that, in vitro, daphnetin suppressed ConA-induced splenocyte proliferation, influenced production of the cytokines and inhibited cell cycle progression through the G0/G1 transition. The data also revealed that daphnetin could down-regulate activation of ConA induced NF-κB and NFAT signal transduction pathways in mouse T lymphocyte. In vivo, daphnetin treatment significantly inhibited the 2, 4- dinitrofluorobenzene (DNFB) -induced delayed type hypersensitivity (DTH) reactions in mice. Collectively, daphnetin had strong immunosuppressive activity both in vitro and in vivo, suggesting a potential role for daphnetin as an immunosuppressive agent, and established the groundwork for further research on daphnetin.  相似文献

A partially purified UDP-glucose: o-dihydroxycoumarin glucosyltransferase from tobacco cell culture required an intact coumarin ring system with o-dihydroxy groups for highest activity. The enzyme exhibited strict position specificity towards the 7-OH group of both esculetin and daphnetin with the formation of cichoriin and daphnin, respectively. The apparent Km values were 111 and 95 μM, while the Vmax for esculetin was 26.4 pkat/mg protein.  相似文献

Protein kinases play key roles in the control of cell proliferation, differentiation and metabolism. In this work, we studied the effect of coumarin and its derivatives, including daphnetin, esculin, 2-OH-coumarin, 4-OH-coumarin and 7-OH-coumarin, on the activity of protein kinases. It was found that, in these compounds, only daphnetin was a protein kinase inhibitor. This compound inhibited tyrosine-specific protein kinase, EGF receptor (IC(50) = 7.67 microM), and serine/threonine-specific protein kinases, including cAMP-dependent protein kinase (PKA) (IC(50) = 9.33 microM) and protein kinase C (PKC) (IC(50) = 25.01 microM) in vitro. The inhibition of EGF receptor tyrosine kinase by daphnetin was competitive to ATP and non-competitive to the peptide substrate. The inhibition of EGF-induced tyrosine phosphorylation of EGF receptor by daphnetin was not observed in human hepatocellular carcinoma HepG2 cells. The structural comparison of daphnetin with coumarin and other coumarin derivatives suggests that the hydroxylation at C8 may be required for daphnetin acting as a protein kinase inhibitor.  相似文献

Twelve flavonoids including one new sulfate were isolated from Neurolaena lobata, and six known flavonoids were obtained from N. macrocephala. The new compound isolated from N. lobata is 6-hydroxykaempferol 3-methyl ether 7-sulfate, and the known flavonoids are 6-hydroxykaempferol 3,7-di-dimethyl ether, 6-hydroxykaempferol, 3-methyl ether 7-glucoside, 6-hydroxykaempferol 7-glucoside, quercetagetin and its 7-glucoside, quercetagetin 3,6- and 3,7-dimethyl ethers, quercetagetin 3-methyl ether 7-glucoside and 7-sulfate, 6-hydroxyluteolin 3′-methyl ether and 6-hydroxyluteolin 7-glucoside. The known flavonoids identified from N. macrocephala are quercetagetin 3,6- and 3, 7-dimethyl ethers, quercetagetin 6-methyl ether 7-glucoside, quercetagetin 3,6-dimethyl ether 7-glucoside, quercetagetin 7-glucoside and quercetagetin 3-methyl ether 7-sulfate.  相似文献

Six new and nine known flavonoids were obtained from Neurolaena oaxacana. The known flavonoids are 6-hydroxykaempferol 3,7-dimethyl ether, quercetagetin 3,7-dimethyl ether, quercetin 3-methyl ether, axillarin, nodifloretin, 6-hydroxyluteolin 7-glucoside, kaempferol 3-glucoside, quercetagetin 7-glucoside and patulitrin. Daphnetin, a coumarin derivative extracted from Daphne odora var., was reported to possess a neuroprotective effect. Recently, it has been demonstrated that daphnetin attenuates ischemia/reperfusion (I/R) injury. However, the role of daphnetin in cerebral I/R injury and the potential mechanism have not been fully understood. The present study aimed to explore the regulatory roles of daphnetin on oxygen-glucose deprivation/reoxygenation (OGD/R)–induced cell injury in a model of hippocampal neurons. Our results demonstrated that daphnetin improved cell viability and reduced the lactate dehydrogenase leakage in OGD/R–stimulated hippocampal neurons. In addition, daphnetin inhibited oxidative stress and cell apoptosis in hippocampal neurons after OGD/R stimulation. Furthermore, daphnetin significantly enhanced the nuclear translocation of the nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) expression in hippocampal neurons exposed to OGD/R. Knockdown of Nrf2 blocked the protective effect of daphnetin on OGD/R–induced hippocampal neurons. In conclusion, these findings demonstrated that daphnetin attenuated oxidative stress and neuronal apoptosis after OGD/R injury through the activation of the Nrf2/HO-1 signaling pathway in hippocampal neurons. Thus, daphnetin may be a novel therapeutic agent for cerebral I/R injury.  相似文献

Two new 3-hydroxyisoflavanones, (S)-3,4′,5-trihydroxy-2′,7-dimethoxy-3′-prenylisoflavanone (trivial name kenusanone F 7-methyl ether) and (S)-3,5-dihydroxy-2′,7-dimethoxy-2″,2″-dimethylpyrano[5″,6″:3′,4′]isoflavanone (trivial name sophoronol-7-methyl ether) along with two known compounds (dalbergin and formononetin) were isolated from the stem bark of Dalbergia melanoxylon. The structures were elucidated using spectroscopic techniques. Kenusanone F 7-methyl ether showed activity against Mycobacterium tuberculosis, whereas both of the new compounds were inactive against the malaria parasite Plasmodium falciparum at 10 μg/ml. Docking studies showed that the new compounds kenusanone F 7-methyl ether and sophoronol-7-methyl ether have high affinity for the M. tuberculosis drug target INHA.  相似文献

Ten flavonoid compounds, including three new natural products, were isolated from a dichloromethane extract of Wyethia glabra. The known compounds are: orobol 7-methyl ether, orobol 3′-methyl ether, naringenin 7-methyl ether, eriodictyol, 8-C-prenyleriodictyol, 6-C-prenyleriodictyol and 8-C-prenylnaringenin. Eriodictyol 7-methyl ether, 2′,4′,6′-trihydroxy-4-methoxychalcone and 6-C-prenylnaringenin are new natural products. Osteoporosis is a common orthopedic disease which is associated with hyper-activated osteoclastogenesis. Daphnetin is a natural coumarin derivative isolated from Genus Daphne, which possesses antiarthritis effect. However, the role of daphnetin in osteoclastogenesis has not been illustrated. This study aimed to investigate the effects of daphnetin on receptor activator of NF-κB ligand (RANKL)-induced osteoclastogenesis in vitro. Our results showed that the osteoclast formation was significantly suppressed by daphnetin treatment in bone marrow-derived macrophages (BMMs), which was illustrated by reduced number of tartrate-resistant acid phosphatase positive multinucleated osteoclasts and decreased expression levels of tumor necrosis factor receptor-associated factors (TRAF6), c-Fos, nuclear factor of activated T cells c1, and cathepsin K. RANKL caused significant induction effects in reactive oxygen species (ROS) generation and nicotinamide adenine dinucleotide phosphate oxidase activity, whereas the induction was dramatically reduced after pretreatment with daphnetin. In addition, daphnetin prevented the RANKL-induced activation of NF-κB and Akt/GSK-3β pathways in BMMs. These findings indicated that daphnetin exhibited an inhibitory effect on RANKL-induced osteoclastogenesis in vitro. The effect of daphnetin might be mediated by inhibiting ROS signal transduction, as well as preventing the activation of NF-κB and Akt/GSK-3β signaling pathways. These findings indicated that daphnetin might be considered as a new therapeutic approach for the osteoporosis treatment.  相似文献

基于网络药理学预测瑞香素抗恶性胶质瘤、肝癌和三阴性乳腺癌的共同靶点及可能机制,并对其进行体外实验验证。利用Swiss Target Prediction和GeneCards等数据库检索瑞香素与恶性胶质瘤、肝癌和三阴性乳腺癌的共同靶点。使用Cytoscape构建瑞香素-三种肿瘤蛋白质相互作用网络图(PPI)并筛选出核心靶点,并对核心靶点进行GO及KEGG富集分析;通过AutoDock Tools对瑞香素与核心靶点进行分子对接。体外实验验证:采用CCK-8和Western blot法行体外实验验证不同浓度瑞香素对U-251 MG、HepG-2和MDA-MB231细胞系细胞抑制率和P53、RRM2蛋白的表达水平的影响。共筛选出瑞香素抗三种肿瘤核心靶点56个,富集分析显示靶点富集在P53通路和癌症通路,参与细胞周期调节、细胞凋亡、DNA生物合成和修复等生物过程;分子对接结果显示瑞香素与P53、RRM2有较好的结合作用。体外验证实验显示,与对照组比较,瑞香素能显著抑制U-251 MG、HepG-2、MDA-MB231的增殖(P<0.01),并显著上调其P53蛋白及下调RRM2蛋白的表达,且...  相似文献

The methylation of daphnetin (7,8-dihydroxycoumarin) to its 8-methyl derivative is catalyzed by a wheat (Triticum aestivum L.) O-methyltransferase (TaOMT1). This enzyme is regulated by cold and photosystem II excitation pressure (plastid redox state). Here, we investigated the biological significance of this methylation and its potential role in modulating the activity of kinases in wheat. To identify the potential kinases that may interact with daphnetin in wheat, the soluble protein extract from aerial parts of cold-acclimated wheat was purified by DEAE-cellulose separation and affinity chromatography on a daphnetin derivative (7,8-dihydroxy-4-coumarin acetic acid)-EAH sepharose column. Mass spectrometric analysis indicated that wheat phosphoribulokinase (TaPRK) is the major kinase that binds to daphnetin. This TaPRK plays an important role in regulating the flow of carbon through the Calvin cycle, by catalyzing the final step in the regeneration of ribulose 1,5-bisphosphate from ribulose-5-phosphate (Ru5P) and ATP. The activities of TaPRK, endogenous or recombinant, are inhibited by daphnetin in a specific and dose-dependent manner, but not by its monomethyl derivative (7-methyl, 8-hydroxycoumarin). Furthermore, HPLC-MS analysis of wheat extracts reveals that 7,8-dimethoxycoumarin is more abundant than its monomethyl derivative. The results also show that cold acclimation does not alter the level of TaPRK mRNA or its enzyme activity, and thus ensures the stable generation of ribulose 1,5-biphosphate.  相似文献

Hemigossypol-6-methyl ether, reported to be present in the root bark of Bombax malabaricum, has been shown to be isohemigossypol-1-methyl ether. Isohemigossypol-1,2-dimethyl ether, 8-formyl-7-hydroxy-5-isopropyl-2-methoxy-3-methyl-1,4-naphthaquinone, 7-hydroxycadalene and an unidentified phenolic compound have also been isolated. Long range couplings in the 1H NMR spectrum of isohemigossypol-1-methyl ether have been established by decoupling experiments.  相似文献

Eleven O-methylated derivatives of kaempferol, quercetin and quercetagetin were isolated from the dichloromethane leaf-wash of Balsamorhiza deltoidea. Four of these compounds represent new reports from either Balsamorhiza or Wyethia: 6-hydroxykaempferol 7-O-methyl ether, quercetin 3′,4′-O-dimethylether, quercetagetin 7-O-methyl ether, and quercetagetin 3,6,7-O-trimethyl ether. We also confirmed the presence of two isoflavones, santal and orobol 3′-O-methyl ether, in W. mollis. The 8-C-prenylated derivatives of naringenin, eriodictyol, and dihydroisorhamnetin were also identified as constituents of W. mollis. The vacuolar flavonoid fraction of Balsamorhiza deltoidea and Wyethia helenioides was shown to consist of simple mono and diglycosides of kaempferol and quercetin.  相似文献
