Genetic variants of human B component (BF system) and alpha-1-antitrypsin (PI system) in a population from Sardinia

BF- and PI-type determinations have been performed in a population from Sardinia. The corresponding allele frequencies are as follows: BF*S = 0.5783, BF*F = 0.2189, BF*SO7 = 0.0046, BF*F1 = 0.1982 and PI*M1 = 0.5872, PI*M2 = 0.2041, PI*M3 = 0.0459, PI*M4 = 0.0940, PI*S = 0.0619, PI*Z = 0.0046, PI*N = 0.0023. Whereas the BF system shows the originality of the Sardinian population with a very high BF*F1 allele frequency, the PI system does not reveal any characteristic features.     

Localization of the steroid-binding site of the human sex steroid-binding protein of plasma (SBP or SHBG) by site-directed mutagenesis.

The amino-terminal region of the human sex steroid-binding protein of plasma (SBP or SHBG) containing K134 and M139 was found to represent part of the steroid-binding site. This was accomplished by constructing and expressing site-directed mutants having the following replacements: M139L, M139K, M139S, K134A, H235S, and Y57F. The results indicated that M139L and H235S were fully-active, K134A and Y57F were 50 and 67% active, M139K was 7% active, and M139S was inactive. These results support affinity-labeling data indicating that both K134 and M139 are located in or near the site, and suggest that Y57 may play a role in steroid binding. The fully active H235S mutant reveals that H235 is not involved in the steroid-binding process.

Chemical composition and characterization of a galactomannoglucan from Gliocladium viride wall material

The following fractions were obtained from the wall material of Gliocladium viride : F1 (27.5%), a glucan, containing xylose, mannose and galactose, coluble in 1 M NaOH at 20°C; F2 (6.7%), a β-glucan-chitin complex, solubilized with 1 M NaOH at 20°C from the previous residue left overnight at −20°C; F3 (8.1%), a glucan, containing mannose and galactose solubilized with 1 M NaOH at 70°C; and F4, the insoluble residue, a β-glucan-chitin complex similar to F2, amounting to 31.3% of the wall material.
F1 was extracted with distilled water. The soluble material (F1S) was a galactomannoglucan (54.7%) and the inscluble (F1P) a glucan (45.3%). Periodate oxidation revealed the presence of glycerol, erythritol, threitol, ribitol, arabitol, mannose, galactose and glucose in F1S, and glycerol and glucose as the main components in F1P. The fractions obtained when F1S was purified through Sepharose CL6B, were methylated.     

Measles Virus Antibody in Cerebrospinal Fluids from Patients with Multiple Sclerosis

A strong measles-specific gel precipitation reaction was found in the cerebrospinal fluid (C.S.F.) of two patients with multiple sclerosis (M.S.) (total of 15 tested). The serum and C.S.F. specimens from these two patients were tested for measles antibody by six assay methods. The results were compared with those obtained from serum and C.S.F. specimens of a patient with subacute sclerosing panencephalitis (S.S.P.E.). The gel precipitation line produced by the C.S.F. from the M.S. patients was identical with one of the three lines produced by the C.S.F. from the S.S.P.E. patient. The main antigenic component responsible for measles antibody appearing in the C.S.F. of the S.S.P.E. patient and the M.S. patients was also electrophoretically similar, and the corresponding antibody was associated with IgG. The serum/C.S.F. antibody titre ratios with the various assay methods used suggest that the C.S.F. antibodies are mainly to other than envelope components of measles virus. No complement-fixing antibody against 27 other viruses or Mycoplasma pneumoniae was found in the C.S.F. of the two M.S. patients.
