High poly(3-hydroxybutyrate) (PHB) content and volumetric productivity were achieved by fed-batch culture of Halomonas boliviensis using a defined medium. Initial shake flask cultivations in a minimal medium revealed that the growth of H. boliviensis was supported only when the medium was supplemented with aspartic acid, glycine, or glutamine. Addition of 0.1% (w/v) glutamine in the medium resulted in the highest cell dry weight (CDW; 3.9 g l−1). Glutamine was replaced by the less expensive monosodium glutamate (MSG) in the medium without any notable change in the final cell density. Effect of initial concentrations of NH4Cl and K2HPO4 on cell growth and PHB accumulation by H. boliviensis was then analyzed using a fed-batch fermentation system. The best conditions for PHB production by H. boliviensis were attained using 0.4% (w/v) NH4Cl and 0.22% (w/v) K2HPO4 and adding MSG intermittently to the fermentor. Poly(3-hydroxybutyrate) content and CDW reached 90 wt.% and 23 g l−1, respectively, after 18 h of cultivation. In order to increase CDW and PHB content, MSG, NH4Cl, and K2HPO4 were initially fed to the fermentor to maintain their concentrations at 2%, 0.4%, and 0.22% (w/v), respectively, and subsequently their feed was suppressed. This resulted in a CDW of 44 g l−1, PHB content of 81 wt.%, and PHB volumetric productivity of 1.1 g l−1 h−1.  相似文献

Summary Pseudomonas 135, a facultative methylotroph, was cultivated on methanol as a sole carbon and energy source for the accumulation of poly--hydroxybutyric acid (PHB). The cells grew fairly well on minimal synthetic medium containing 0.5% (v/v) of methanol at pH 7.0 and 30° C. The maximum specific growth rate was determined to be 0.26–0.28 h–1 with a growth yield of 0.38 in the optimized growth medium. For stimulation of PHB accumulation in the cells, deficiency of nutrients such as NH inf4 sup+ , Mg2+ and PO inf4 sup3– was crucial even though cell growth was significantly suppressed. The PHB content of a 40-h culture was determined to be 37% of the total cell mass in NH inf4 sup+ -limited medium, 42.5% on Mg2+-deficient medium, and 34.5% on PO inf4 sup3– -deficient medium. The maximum content of PHB in the cells could reach 55% in NH inf4 sup+ -limited fed-batch culture. The average relative molecular eight determined by gel permeation chromatography was 3.7 × 105 in NH inf4 sup+ -limited culture, 2.5 × 105 in Mg2+-deficientmedium, and 3.1 × 105 in PO inf4 sup3– -deficient medium. Growth ofS. discophorus in a casamino acids-mineral salts medium was stimulated 3-fold on addition of 0.05% MnSO4·H2O to the medium. Growth was measured by determinations of total nitrogen, protein and DNA on the washed cellular material.Autotrophic growth ofS. discophorus strain 43-R was obtained in an inorganic mineral salts medium supplemented with trace amounts of the essential vitamins, thiamin, biotin and cyanocobalamin and with Mn++ as the sole available source of energy. A gas mixture of 5% CO2-95% air was bubbled continuously through the cultures during incubation. Concomitant with growth, Mn++ disappeared from the cultures and MnO2 was formed.  相似文献

Summary Certain nutritional requirements of one strain ofMyxotrichum uncinatum were established. Growth was measured on a dry weight basis. The approximate optimum medium for growth consisted of 5% KNO3, 4% sucrose, 0.1% K2HPO4, 0.05% MgSO4. 7 H2O, 0.2 mg % biotin and 1.0 mg % uracil. The best initial pH range was 6.0–7.0. Approximately three to five days incubation on a rotary shaker permitted excellent growth.  相似文献

Somatic embryogenesis was obtained in cultures of leaves from young seedlings of Quercus suber L. A two-stage process, in which benzyladenine and naphthaleneacetic acid were added first at high and then at low concentrations, was required to initiate the process. Somatic embryos arose when the explants were subsequently placed on medium lacking plant growth regulators. The embryogenic lines remained productive, by means of secondary embryogenesis, on medium without growth regulators. However, this repetitive induction was influenced by the macronutrient composition of the culture medium. Both low total nitrogen content and high reduced nitrogen concentration decreased the percentage of somatic embryos that showed secondary embryogenesis. Our results suggest that alternate culture on medium that increases embryo proliferation and a low salt medium prohibiting embryo formation will partially synchronize embryo development. Chilling slightly reduced secondary embryogenesis but gave a modest increase in germination. Maturation under light followed by storage at 4 °C for at least 30 days gave the best results in switching embryos from an embryogenic pathway to a germinative one. Under these conditions 15% of embryos showed coordinated root and shoot growth and 35% formed either shoots or mostly roots. These percentages were higher than those of embryos matured in darkness. Seed germination and micropropagation protocols of the medicinal species Maytenus canariensis (Loes.) G. Kunkel & Sunding were optimized. In vitro seed germination occurred (86 to 94.7 %) only after treatment of the seeds with H2SO4, followed by surface sterilization and culture on solid nutrient medium without any growth regulators. Micropropagation failed when explants were taken from mature trees, and browning of the nutrient medium frequently occurred despite testing many growth media. Nonetheless, adventitious shoot regeneration was achieved employing axillary or apical buds taken from 2–2.5 months old plantlets obtained after in vitro germination of seeds, following culture on nutrient media supplemented with benzylaminopurine, kinetin and naphthalenacetic acid (NAA), attaining up to 3.9 shoots per explant, after 4–6 months. Root induction was best on a medium containing 4.0 mg dm−3 NAA, achieving a 100 % induction. After hardening of rooted plants, survival after transfer to soil was 71.43 %.  相似文献

Summary The effect of addition of varying concentrations of certain inorganic compounds in selected media upon growth and pigment elaboration ofEidamella deflexa (Berk.)Benjamin was studied in submerged culture. Preliminary experiments were performed to establish approximate optimum ranges of certain inorganic compounds. Further investigations in a glycine-sucrose basal medium indicated that the following range of concentrations stimulated growth to the greatest extent: 0.2% MgSO4·7H2O, 0.2% K2HPO4, 0.25 mg% CuSO4·5H2O, 0,0% ZnSO4·7H2O and 10.0 mg % FeSO4·7H2O. The degree of pigment production did not seem to be affected by different concentrations of these compounds.The effects of additional molybdenum and zinc upon growth and pigmentation were studied in the following media: lactose-peptone, lactose-glycine, maltose-peptone and maltose-glycine. Molybdenum increased pigmentation slightly in maltose-glycine and maltose-peptone media, but inhibited the amount of growth. Molybdenum decreased both growth and pigmentation in the two lactose media. Zinc had little affect on either growth or pigment elaboration in the two maltose media. However, in the lactose-glycine medium, zinc decreased both growth and pigmentation while in lactose-peptone media zinc slightly increased growth and pigmentation.  相似文献

Growth characteristics ofDeleya halophila (CCM 3662T), were determined using a defined medium.Deleya halophila presented its optimal growth at 7.5% (wt/vol) total salts when it was grwon at incubation temperatures of 32° and 42°C; when the temperature was lowered to 22°C, it had optimal growth at 5% (wt/vol) total salts. This bacterium had an absolute requirement for the Na+ cation; it could not be replaced by other cations. NaBr, Na2SO4, or Na2S2O3 could be substituted for NaCl in the growth medium, but, when MgCl2, KCl, LiCl, NaI, NaF, or NaNO3 was substituted for NaCl, the medium did not support growth. Growth rates of the strain were diverse when NaCl was partially replaced by other sodium salts. Finally,D. halophila suffered loss of viability when the culture was diluted into different low NaCl concentrations (0, 0.5%, and 1%, wt/vol) at various incubation temperatures.  相似文献

Methanobacterium thermoautotrophicum was grown on a defined mineral salts medium under strictly anaerobic conditions with H2 and CO2 as the sole energy and carbon sources, respectively. The cultivation medium was optimized with respect to non-organic components including Se(IV), W(VI), N, Ni(II), Fe(II), Co(II) and Mo(VI). Sulphide concentration in the medium was maintained constant using an on-line regulatory system by the addition of 0.5 M Na2S. A maximum supply rate of 0.6 vvm of a mixture of 80% H2 and 20% CO2 was achieved for the gaseous substrates. Under these conditions a specific growth rate of 0.30 h–1 and a cell concentration of 4.8 g cell dry weight (DW) l–1, representing a 140% increase over previously published results, were obtained. The growth yield of 2.3 g DW mol–1 CH4 was similar to published values. Summary The effect of 17β-estradiol (E2) on growth of GH4C1 rat pituitary tumor cells was investigated under serum-free conditions and with medium containing charcoal-extracted serum. Serum-free TRM-1 medium was a 1∶1 (vol/vol) mixture of F12-DME supplemented with 50 μg/ml gentamicin, 15 mM 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid, 10 μg/ml insulin, 10 μg/ml transferrin, 10 ng/ml selenous acid, 10 nM 3,5,3′-triiodothyronine (T3), 50 μM ethanolamine, and 500 μg/ml bovine serum albumin. The cells grew continuously in TRM-1 but were E2 responsive only when growth was retarded by reducing the T3 concentration to 10 pM (TRM-MOD). Addition of 1 to 10 nM E2 to TRM-MOD increased growth by 0.3 to 0.9 cell population doublings over controls in 9 d. By using medium supplemented with charcoal-extracted sera, basal growth became 1 to 1.5 cell population doublings in 9 d. Addition of 0.1 pM E2 to medium containing charcoal-extracted serum caused a significant increase in cell number whereas pM-nM concentrations stimulated 200 to 570% increases over controls. The effect of steroid hormone was the same in phenol-red-containing and indicator-free medium. The data presented confirm that the major requirements for demonstration of estrogenic effects in culture were optimum concentrations of thyroid hormones and the presence of yet-to-be-characterized serum factors. Summary Bacillus sphaericus (NCIM 2478) produced high levels of penicillin V acylase (100 U / g dry cells) when grown on cornsteep liquor — minerals medium at 25°C for 20 h. Supplementation of the medium either with 1 % (w/v) whole wheat bran or its aqueous extract brought about more than 70 % increase in the total enzyme activity. Summary Explants of sugarcane, a C4 plant, were cultured in vitro for 18d on Floridalite (a solid cube consisting of vermiculite and cellulose fibers) used as supporting material with sugar-free Murashige and Skoog liquid medium with double-strength KH2PO4, MgSO4, FeSO4, and Na2-EDTA in the vessel with enhanced natural ventilation. CO2 concentration in the culture room was kept at 1500 μmol mol−1 (four times the atmospheric CO2 concentration) during the photoperiod. A factorial experiment was designed with two levels of photosynthetic photon flux (PPF) and three levels of N (number of air exchanges of the vessel). The results were compared with those in the control treatment (photomixotrophic culture using sugar-containing agar medium under low PPF and low N). PPF and N showed significant positive effects on the growth of sugarcane plantlets in vitro. In the photoautotrophic (using sugar-free medium) treatments with relatively high PPF (200–400 μmol m−2 s−1) and high N (2–10 h−1), the growth of plantlets was four to seven times greater than that in the control. Also, the culture period for multiplication and rooting was shortened from 30 d in the control to 18 d or less in the photoautotrophic, high PPF, and high N treatments. Use of porous supporting material in photoautotrophic treatments promoted rooting and plantlet growth significantly.  相似文献

Direct differentiation of shoot buds from the collar region of hypocotyl segments of Coffea canephora was obtained on Murashige and Skoog (MS) medium supplemented with 40 μM silver nitrate (AgNO3) and growth regulators indole-3-acetic acid (IAA) and N6 benzyladenine (BA). The highest response to shoot differentiation of 60% frequency and the maximum number of multiple shoots (2–3) per explant were obtained on MS medium containing 8.87 μM BA and 2.85 μM IAA. Apart from this, 70% of hypocotyl explants produced yellow friable embryogenic callus and also globular primary somatic embryos. Subsequent transfer onto the same medium induced secondary somatic embryogenesis. The micro-shoots, upon transfer to the same medium, in the following 6 weeks developed into 4-cm-long shoots with a single root. Further subculturing onto the same medium induced 4–5 roots in a 4-week period. The resulting plantlets were hardened and transferred to micro-pots containing sand:compost mixture (1:2), where 65% of them survived and resumed growth. By using optimal levels of AgNO3, it was possible to obtain effective direct organogenesis and embryogenesis. This system was used for genetic transformation using Agrobacterium tumefaciens. A stable transformation frequency of 2–5% was obtained when both types of explants, i.e., hypocotyl explants with collar region or hypocotyl explants without collar region, were co-cultivated with A. tumefaciens GV 3101 harboring pCAMBIA 1305.2 binary vector. This is the first report of a hypocotyl collar region-based Agrobacterium-mediated transformation protocol for the economically important tropical plant C. canephora.  相似文献

Vibrio parahaemolyticus, the flagellated nonswarming marine bacteria were induced to swarm on solid media under three different conditions: growth at 20–26°C on medium containing 1% NaCl, growth on a medium in a sealed Petridish and growth on H2O2-treated medium. The morphological transformations observed in cells during swarming of V. parahaemolyticus are similar to those found jor the naturally swarming Vibrio alginolyticus. The mechanism of swarming in both species involves massive formation of peritrichous flagella and a negative chemotactive response to metabolic byproducts.  相似文献

Optimization of the medium components which enhance sporulation of the two mating types of the fungus Blakeslea trispora ATCC 14271 and ATCC 14272 (a heterothallic Zygomycota producing carotene) was achieved with the aid of response surface methodology (RSM). Glucose, corn steep liquor, yeast extract, and ammonium sulfate were investigated as carbon and nitrogen sources in a basal medium. RSM was adopted to optimize the medium in order to obtain a good growth of the fungus as a prerequisite for enhanced sporulation. In the second step, the basal medium was supplemented with different trace elements which significantly affect sporulation (i.e. CuSO4·5H2O, FeCl3·6H2O, Co(NO3)2·6H2O, and MnCl2·4H2O). Central composite design proved to be valuable in optimizing a chemically defined solid medium for spore production of B. trispora. The composition of the new solid medium to enhance spore production by B. trispora (ATCC 14271) is as follows (per liter): 7.5 g glucose, 3.2 g corn steep liquor, 1.7 g yeast extract, 4.1 g ammonium sulfate, 6 mg CuSO4·5H2O, 276 mg FeCl3·6H2O, 2 mg Co(NO3)2·6H2O, and 20 g agar (pH 6.0). Practical validation of this optimum medium gave spore number of 1.2 × 108 spores/dish which is 77% higher than that produced in Potato Dextrose Agar (PDA). In the case of B. trispora (ATCC 14272) the new solid substrate for enhanced sporulation consists of (per l) 6.4 g glucose, 3.3 g corn steep liquor, 1.4 g yeast extract, 4.3 g ammonium sulfate, 264 mg CuSO4·5H2O, 485 mg FeCl3·6H2O, 223 mg MnCl2.4H2O, and 20 g agar (pH 6.0). Spore numbers of 2 × 107 spores/dish were obtained on the new medium by B. trispora (ATCC 14272), which is 95% higher than that produced on PDA. The results corroborated the validity and the effectiveness of the models. The new media considerably improved sporulation of both strains of B. trispora compared to the production of spores on PDA, which is the medium usually used for sporulation of the fungus.  相似文献

A highly efficient Agrobacterium-mediated transformation system for Lilium × formolongi was established by modifying the medium used for inoculation and co-cultivation. Meristematic nodular calli of Lilium were inoculated with an overnight culture of A. tumefaciens strain EHA101 containing the plasmid pIG121-Hm harboring an intron-containing β-glucuronidase (GUS), hygromycin phosphotransferase, and neomycin phosphotransferase II genes. The effects of ten different types of media and carbohydrates (sucrose, d-glucose, and l-arabinose) in both inoculation and co-cultivation media were evaluated. Interestingly, a dramatic increase in the frequency of transformation (25.4%) was observed when Murashige and Skoog (MS) medium containing sucrose and lacking KH2PO4, NH4NO3, KNO3, and CaCl2 was used. Hygromycin-resistant transgenic calli were obtained only in medium supplemented with sucrose. The effects of this modified medium were also investigated for Lilium cultivars ‘Acapulco’, ‘Casa Blanca’, and ‘Red Ruby’. The highest frequency of transformation (23.3%) was obtained for cv. Acapulco. Hygromycin-resistant calli were successfully regenerated into plantlets on plant growth regulator-free MS medium. Transgenic plants were confirmed by GUS histochemical assay, polymerase chain reaction (PCR), and Southern blot analyses.  相似文献

Summary To improve proliferation of soybean cultures in liquid medium, the effects of sucrose; total inorganic nitrogen; content of No3 , NH4 +, Ca2+, PO4 3−, K+; NH4 +/NO3 ratio; and medium osmotic pressure were studied using cv. Jack. Sucrose concentration, osmotic pressure, total nitrogen content, and ammonium to nitrate ratio were found to be the major factors controlling proliferation of soybean embryogenic cultures. Growth decreased linearly as sucrose concentration increased from 29.7 mM to 175.3 mM. A sucrose concentration of 29.2 mM, a nitrogen content of 34.9 mM, at 1 to 4 ammonium to nitrate ratio were found to be optimal for the fastest proliferation of soybean embryogenic cultures. There was no significant effect on proliferation of cultures when concentrations of NH4 +, Ca2+, PO4 3−, and K+ were tested in the range of 3.50 to 10.50, 1.02 to 3.06, 0.68 to 2.04, and 22.30 to 36.70 mM, respectively. The relative proliferation of embryogenic cultures of four soybean genotypes was evaluated in Finer and Nagasawa medium and in the new medium formulation. Despite genotype-specific differences in growth, the genotypes tested showed a biomass increase in the new formulation equal to 278, 269, 170, and 251% for Chapman, F138, Jack, and Williams 82, respectively, relative to their growth on standard FN medium. Eine Anfangsbreite der Ph von 5.0–8.0 erlaubte ein optimales Wachstum.

Zusammenfassung In einer Mineralsalzlösung mit 0,1% NH4Cl und 0,01% Hefeextrakt wurden aus Schlammproben unter einer Atmosphäre aus 90% H2 und 10% CO2 im Licht acht verschiedene Stämme von schwefelfreien Purpurbakterien angereichert und isoliert, die zum photoautotrophen Wachstum mit H2-CO2 befähigt sind. Die neuisolierten Stämme wurden aufgrund ihrer mikroskopischen Merkmale und ihrer Fähigkeit zur Verwertung verschiedener organischer C-Verbindungen bestimmt. Drei Rps. capsulata-Stämme benötigen Thiamin als Wachstumsfaktor, zwei R. rubrum-Stämme Biotin und ein Rps. gelatinosa-Stamm benötigt Thiamin und Nicotinsäure. Ein Rps. palustris-Stamm wächst langsam in einem Medium ohne Vitaminzusatz. Der Wachstumsfaktor-Bedarf eines zweiten Rps. palustris-Stammes kann nur durch Hefeextrakt gedeckt werden. Bei allen Stämmen wird das Wachstum durch kleine Mengen (0,01%) Casaminosäuren bzw. Hefeextrakt merklich beschleunigt. Die kürzesten Verdoppelungszeiten beim photoautotrophen Wachstum mit H2 weisen die drei Rps. capsulata-Stämme auf (17 bis 23 h in einem Mineralmedium mit Thiamin, 10,5–11 h in einem Mineralmedium mit 0,01% Hefeextrakt). Die Hydrogenase-Aktivitäten der Stämme sind in starkem Maße abhängig von der Anzucht der Zellen. Die niedrigsten Aktivitäten werden nach photoheterotropher Anzucht in einem Medium mit NH4Cl als N-Quelle gemessen, mittlere Aktivitäten nach photoheterotropher Anzucht in einem Medium mit Glutamat als N-Quelle und die höchsten Aktivitäten nach photoautotropher Anzucht mit H2-CO2. Die höchsten Raten der H2-abhängigen CO2-Fixierung werden ebenfalls nach photoautotropher Anzucht gemessen.
Studies on the photoautotrophic growth of new isolated nonsulfur purple bacteria at the expense of molecular hydrogen
Summary By incubating samples of mud in a mineral salt medium containing 0.1% NH4Cl and 0.01% yeast extract under an atmosphere of 90% H2 and 10% CO2 in the light, eight strains of nonsulfur purple bacteria capable of photoautotrophic growth at the expense of H2-CO2 were enriched and isolated. The bacteria were characterized by their microscopical features and their ability to use various organic compounds for growth. Three strains of Rps. capsulata need thiamine as a growth factor and two strains of R. rubrum biotin. The Rps. gelatinosa strain is dependent on thiamine and nicotinic acid. One of two Rps. palustris strains grows slowly in a medium completely free of growth factors, the other strain is dependent on the presence of 0.01% yeast extract. The growth of all strains is markedly stimulated by small amounts (0.01%) of amino acids or yeast extract. The three Rps. capsulata strains differ significantly from the other strains with regards to their fast photoautotrophic growth (doubling time: 17–23 h in a mineral salt medium with thiamine, 10.5–11 h in a medium with 0.01% yeast extract). The hydrogenase activities of all strains are strongly dependent on the culture conditions. The lowest activities are obtained after photoheterotrophic growth in a medium with NH4Cl as the nitrogen source, moderate activities after photoheterotrophic growth in a medium with glutamate as the nitrogen source and maximum activities after photoautotrophic growth at the expense of H2-CO2. Maximum rates of CO2-fixation are also obtained after photoautotrophic growth.

The positive effect of ventilation of the culture container on in vitro shoot proliferation and quality was already proven for different species. Hereafter we report on the evolution of the headspace during in vitro culture of plantain in a Temporary Immersion Bioreactor (TIB) on the one hand, and culture on semi-solid medium on the other hand. The CO2 and C2H4 concentration reached a maximum of 12% and 0.45 μl l−1, respectively in the control treatment on semi-solid medium, compared to 5.7% CO2 and 0.06 μl l−1 C2H4 in TIB. The minimal O2-concentration on semi-solid medium was 15.1%, compared to 19.3% in TIB. The multiplication rate was best in TIB, 6.4 compared to 4.3 in semi-solid conditions, and this was also the case for shoot height (4.3 cm compared to 3.3 cm), and leaf number (2.6 compared to 1.6). Moreover shoots produced on semi-solid medium showed distorted leaves. A typical day-night pattern in CO2 and O2 concentration was observed in TIB, as well as on semi-solid medium; this is illustrative for the photosynthetic capacity of the plant material produced in both systems.  相似文献
