We report a case study of an IgG1 with a unique basic charge variant profile caused by C-terminal proline amidation on either one or two heavy chains. The proline amidation was sensitive to copper ion concentration in the production media during cell culture: the higher the Cu2+ ion concentration, the higher the level of proline amidation detected. This conclusion was supported by the analysis of samples that revealed direct correlation between the proline amidation level observed from peptide maps and the level of basic peaks measured by imaged capillary isoelectric focusing and a pH gradient ion-exchange chromatography method. Δ1-pyrroline-5-carboxylate synthetase (P5CS) is a proline biosynthetic pathway enzyme and is known for conferring enhanced salt and drought stress in transgenics carrying this gene in a variety of plant species; however, the wild-type P5CS is subjected to feedback control. Therefore, in the present study, we used a mutagenized version of this osmoregulatory gene-P5CSF129A, which is not subjected to feedback control, for producing transgenic indica rice plants of cultivar Karjat-3 via Agrobacterium tumefaciens. We have used two types of explants for this purpose, namely mature embryo-derived callus and shoot apices. Various parameters for transformation were optimized including antibiotic concentration for selection, duration of cocultivation, addition of phenolic compound, and bacterial culture density. The resultant primary transgenic plants showed more enhanced proline accumulation than their non-transformed counterparts. This proline level was particularly enhanced in the transgenic plants of next generation (T1) under 150 mM NaCl stress. The higher proline level shown by transgenic plants was associated with better biomass production and growth performance under salt stress and lower extent of lipid peroxidation, indicating that overproduction of proline may have a role in counteracting the negative effect of salt stress and higher maintenance of cellular integrity and basic physiological processes under stress.  相似文献

Excised barley (Hordeum vulgare var Larker) leaves were treated with salt solutions or wilted. After the treatment period, the leaves were allowed to recover in a 50 millimolar sucrose and 1 millimolar glutamate solution, and proline, Na+, and K+ were measured at intervals. Na+ and K+ concentrations stayed at a constant high level after the salt treatments, and proline increased to a steady state concentration in response. The relationship between the maximum rate of proline accumulation and the Na+ concentration reached in each experiment was linear. The final steady state proline concentration reached was also directly proportional to the Na+ concentration. For a given Na+ concentration in the leaves, the steady state proline level was greater when 410 millimolar NaCl was added to the leaves than when 205 millimolar NaCl was added. These results are consistent with proline acting as a compatible cytoplasmic solute, balancing an accumulation of salts outside of the cytoplasm.

In contrast to the proline levels in salt-shocked leaves, the concentrations in wilted leaves decreased to near control levels within 24 hours of relief of stress.

The effects of NaCl and mannitol iso-osmotic stresses on calli issued from sugarcane cultivars (cvs.) R570, CP59-73 and NCo310 were investigated in relation to callus growth, water content, ion and proline concentrations. Callus growth and water content decreased under both stresses with the highest reduction under mannitol-induced osmotic stress. The ion concentration was drastically affected after exposure to NaCl and mannitol. Salt stress induced an increase in Na+ and Cl accumulation and a decrease in K+ and Ca2+ concentrations. Under mannitol-induced osmotic stress, K+ and Ca2+ concentrations decreased significantly while Na+ and Cl concentrations remained unchanged. Free proline accumulation occurred under both stresses and was more marked in stress-sensitive cv. than in stress-resistant one. Our results indicated that the physiological mechanisms operating at the plant cell level in response to salt- and osmotic-induced stress in sugarcane cvs. are different. Among the cvs., we concluded that the stress resistance is closely related to the maintain of an adequate water status and a high level of K+ and Ca2+ under both stresses and a low level of Na+ concentration in the presence of NaCl. Thus, sugarcane (Saccharum sp.) can be regarded as a Na+ excluder. We also provided evidence that proline accumulation is a stress-sensitive trait rather than a stress resistance marker.  相似文献

The concentration of proline in shoots of rice (Oryza sativa) seedlings raised in distilled water was about 3.3 times higher than in the seedlings raised in modified B5 medium. The shoots of seedlings raised in B5 medium which was depleted of calcium, iron, magnesium or potassium had a higher concentration of proline than those grown in standard B5 medium. The shoots of seedlings raised in distilled water with iron had a lower level of proline than those in distilled water. These results suggest that iron deficiency leads to high proline accumulation.The electron transport activity of mitochondria from shoots of etiolated seedlings raised in distilled water or iron-depleted B5 medium was significantly lower than those from equivalent seedlings raised with an iron source. As suppression in mitochondrial electron transport leads to an increase in NADH/NAD+ ratio, we propose that the prime cause of the proline accumulation during iron deficiency is to readily maintain NADH/NAD+ ratio.  相似文献

The response of tomato (Solanum lycopersicum L.) to abiotic stress has been widely investigated. Recent physiological studies focus on the use of osmoprotectants to ameliorate stress damage, but experiments at a field level are scarce. Two tomato cultivars were used for an experiment with saline water (6.57 dS m?1) and subsurface drip irrigation (SDI) in a silty clay soil. Rio Grande is a salinity-tolerant cultivar, while Heinz-2274 is the salt-sensitive cultivar. Exogenous application of proline was done by foliar spray at two concentrations (10 and 20 mg L?1) during the flowering stage. Control plants were treated with saline water without proline. Proline at the lower concentration (10 mg L?1) increased dry mass of different plant organs (leaves, stems, and roots) and it improved various chlorophyll a fluorescence parameters compared with controls. Regarding mineral nutrition, K+ and P were higher in different organs, while low accumulation of Na+ occurred. However, Mg2+ was very high in all tissues of Rio Grande at the higher concentration of proline applied. Thus, the foliar spray of proline at 10 mg L?1 increased the tolerance of both cultivars. The growth of aboveground biomass of Heinz-2274 was enhanced by 63.5%, while Rio Grande improved only by 38.9%.  相似文献

The production of malondialdehyde (MDA) was higher in cotyledons from NaCl-raised Brassica juncea seedlings than in control seedlings. Light accelerated the MDA-producing capacity of thylakoids isolated from both control and treated seedlings. When exposed to strong white light (920 mol photons m-2 s-1) the thylakoids from NaCl seedlings produced nearly 5 times more MDA than control thylakoids. In the cotyledons of NaCl seedlings, the proline level was 24-fold higher than in controls. The presence of proline during exposure of thylakoids to white light decreased MDA levels. The reduction in MDA production was higher in the thylakoids of NaCl seedlings than of controls. It is proposed that proline accumulation has an adaptive significance as it lowers the generation of free radicals and thus reduces the lipid peroxidation linked membrane deterioration under stress.  相似文献

The increase in proline induced by ABA, a process stimulated by NaCl or KCl in barley leaves, did not occur when Na+ (or K+) was present in the external medium as the gluconate salt, namely with an anion unable to permeate the plasma membrane. However, proline increase was restored, to different extents, by the addition of various chloride salts but not by ammonium chloride. Moreover, it was shown that the stimulation of the process by NaCl (or KCl) was variously affected by the presence of different salts; all the ammonium salts (10 millimolar NH4+ concentration) inhibited this stimulation almost completely. Inhibition by NH4+ was accompanied by a decreased Na+ influx (−40%). Also, in the case of Na-gluconate, Na+ uptake was reduced and the addition of Cl as the calcium or magnesium salt (but not as ammonium salt) restored both the ion influxes and the increase in proline typical of NaCl treatments. Both 4,4′-diisothiocyano-2,2′-disulfonic acid stilbene (DIDS), an anion transport inhibitor, and tetraethylammonium chloride (TEA), a K+ channels-blocking agent, caused, as well as with a reduction of ion influxes, an inhibition of the proline accumulation. The inhibition was practically total with 1 millimolar DIDS and about 80% with 20 millimolar TEA. A possible role of ion influxes in the process leading to the increase in proline induced by ABA is proposed.  相似文献

In this study, responses of wild species of potato to NaCl stress were investigated in vitro. In S. stoloniferum and S. bulbosum, length of the shoot, fresh and dry weight, photosynthetic pigments, K+ concentration, K+/Na+ ratio, ascorbate pool, anthocyanin, and phenolic and flavonoid compounds were decreased in response to salinity. In these species, salinity increased the level of Na+, lipid peroxidation, proline and ion leakage percentage. In S. acaule, the length of the shoot, and fresh and dry weight were not affected by salinity. Photosynthetic pigments, Na+ concentration, proline, flavonoid and phenolic compounds quantities were increased and K+/Na+ ratio were decreased. K+ concentration, lipid peroxidation, ascorbate pool, anthocyanin and ion leakage were not changed by NaCl stress. Superoxide dismutase, guaiacol peroxidase, ascorbate peroxidase and catalase activities were increased in all species. The results suggest that the non-enzymatic antioxidant capacity in S. acaule (salt tolerant) is more important than the enzymatic antioxidant capacity in comparison with the other species.  相似文献

Salinity causes osmotic stress and negatively impacts plant growth and productivity. Proline is one of the most important osmoprotectants synthesized under stressed conditions. Accumulation of free proline occurs due to enhanced biosynthesis and repressed degradation, and both processes are controlled by feedback regulatory mechanisms. Arbuscular mycorrhizal (AM) fungi are considered to be bioameliorators of salinity stress due to their wide-ranging presence in contaminated soils and their role in modulation of biochemical processes. Chickpea is considered sensitive to salinity. However, reports on AM-induced osmoprotection through regulation of proline biosynthesis in chickpea genotypes are scant. The present study investigated the influence of AM symbiosis on proline metabolism in two chickpea (Cicer arietinum L.) genotypes (PBG-5 and CSG-9505) under salt stress and correlated the same with sodium (Na+) ion uptake. Salinity reduced plant biomass (roots and shoots), with roots being more negatively affected than shoots. Mycorrhizal colonization with Glomus mosseae was much stronger in PBG-5 and was correlated with reduced Na+ ion uptake and higher growth when compared with CSG-9505 under stressed and unstressed conditions. Mycorrhizal symbiosis with chickpea roots boosted proline biosynthesis by significantly increasing pyrroline-5-carboxylate synthetase (P-5-CS) and glutamate dehydrogenase (GDH) activities with a concomitant decline in proline dehydrogenase (ProDH) activity under salt stress. The enhancement of the activity of these enzymes was higher in PBG-5 than in CSG-9505 and could be directly correlated with the percent mycorrhizal colonization and Na+ uptake. The study indicated a strong role of AM symbiosis in enhancing stress tolerance in chickpea by significantly modulating proline metabolism and Na+ uptake.  相似文献

It was shown that callus established from Kosteletzkya virginica (L.) Presl. (Malvaceae) can grow in salinities higher than 200 mM NaCl if previously accomodated stepwise. Callus lines developed from seedlings of different harvests or of the same harvest at different times, all showed the same pattern of growth and sensitiviy to salinity. The absorption of Na+ into the callus increased with increasing external NaCl concentration. In the callus, Na+ was apparently distributed outside and inside a cellular membrane (possibly the plasmalemma). This membrane was, apparently, capable of regulating the Na+ concentration in the protoplast. Outside this membrane Na+ accumulated to concentrations higher than in the external growth medium. Exogenously supplied proline or glycine-betaine did not affect the growth of the callus. Externally applied ABA stimulated growth under saline conditions and increased the accumulation of proline. Growth and proline content were positively correlated in callus exposed to salinity, but in the presence of ABA they were negatively correlated. ABA was involved in both growth and proline accumulation, but there was no clear relationship between these two effects. Root treatments of barley (Hordeum distichum L.) plants with 10-7 to 10-4 M abscisic acid (ABA) caused an increase in proline content, especially at higher concentrations, within 2–3 h. Even 3 h after the removal of ABA from the medium the plants continued to accumulate proline. The higher the concentration of the ABA, the higher was the proline level at 6 h. When the highest ABA concentration, 10-4 M, was tested with polyethylene glycol (PEG) (-5.0 bars) in the medium, the ABA treatment resulted in a higher proline content than in control plants. The treatments PEG alone and PEG + ABA resulted in heavy accumulation of proline, especially, 3 h after releasing the plants from the stress. The proline content in PEG+ABA-treated plants was always higher than plants treated with PGE or ABA alone. In peas (Pisum sativum L. cv Alaska) the same trend occurred although to a lesser degree. The effects of NaCl (0, 50, 100, 150 and 200 mM) on growth, water relations, glycinebetaine, free proline, ion contents, stomata number and size of Kochia prostrata (L.) Schard were determined. Shoot and root fresh and dry matter, root and shoot length, relative growth rate, net assimilation rate, relative water content, water use efficiency, soluble sugars and glycinebetaine contents were not changed at low NaCl concentrations, but they were significantly decreased at 200 mM NaCl. The K+, Mg2+ and Ca2+ contents, water potential, chlorophyll a+b and carotenoides contents, and stomata number and size were reduced already at low concentrations of NaCl. In contrast, the Na+, Cl and proline contents increased several times with increasing NaCl concentration. Kochia prostrata is a salt tolerant species, the optimal growth of this plant occurred up to 150 mM NaCl. The mechanisms of salt tolerance in the plant may be balance among ion accumulation and production of glycinebetaine, proline, soluble sugars for maintenance of pressure potential.  相似文献

Growth and Na+, K+, Cl-, proteins, sugars and proline concentrations were measured in three triticale genotypes M2A, DF99 and Asseret grown on nutrient solution with or without 75 mM NaCl. In saline conditions, leaf area of the three triticales was reduced by 50 % and dry to fresh mass ratio increased. Total protein concentration was diminished by 10 %. K+ concentration decreased whereas Na+ and Cl- accumulated in roots and shoots of salt-stressed plants. This ion accumulation was greater in roots of Asseret than in roots of the other triticales. Soluble sugar concentration increased in M2A and Asseret and decreased in DF99. Proline concentration increased in M2A and DF99 and decreased in Asseret. Osmotic adjustment was essentially realized by Na+ and Cl- uptake. Non-reducing sugars and proline contributed too, but to a lesser extent. The lengths of roots and shoots, fresh and dry matter yield, and the contents of insoluble saccharides and free amino acids were reduced with the rise in NaCl concentration. However, under combination of NaCl with Ca2+ ions, these parameters generally raised. Contents of soluble saccharides, proline and quaternary ammonium compounds increased with increasing NaCl concentration, but under addition of CaCl2 or CaSO4, contents of these compounds were decreased. Low concentrations of NaCl stimulated soluble proteins, production, but higher concentrations decreased the content of soluble proteins. Addition of Ca2+ in the media did not improve the soluble protein production. Insoluble proteins content was increased with the rise of salinity level, but these effects were more pronounced with NaCl and CaCl2 or CaSO4 than with NaCl only.  相似文献

An NaCl-resistant line has been developed from suspension-cultured tobacco cells (Nicotiana tabacum/gossii) by stepwise increases in the NaCl concentration in the medium. Resistance showed stability through at least 24 generations in the absence of added NaCl.

Above an external NaCl concentration of 35 millimolar, proline concentration in the selected cells rose steeply with external NaCl, particularly so above 100 millimolar NaCl. Proline accumulation in the wild type was far slighter. Selected cells which had been grown for 24 generations in the absence of added NaCl accumulated proline strongly on re-exposure to NaCl medium, indicating stability of this character. Proline accumulation was fully reversible with a half-time of about 6 hours. When selected cells were transferred sequentially to lower and lower NaCl concentrations, their proline content fell to the level corresponding to the new NaCl concentration. The NaCl-selected cells responded to water stress (i.e. added mannitol) by accumulating markedly more proline than did the wild type.

The addition of Ca2+ to the growing and rinsing media minimized Na+ and K+ binding in the Donnan free space of cell walls and thus allowed assessment of intracellular Na+ and K+. In both cell types, internal Na+ content rose steadily as a function of external NaCl concentration. In the course of 7 days in NaCl media, the wild type cells lost a considerable part of their K+ content, the extent of the loss increasing with rise in external NaCl concentration. The selected cells, by contrast, lost no K+ at external NaCl concentrations below 50 millimolar external NaCl, and at higher concentrations lost less than the wild type.

The comparative effect of fertilizers (NPK), plant growth regulators (GA3, IAA, Zeatin) and sodium chloride (NaCl) on Cd phytoaccumulation, proline and phenolics production in Cannabis sativa was evaluated. Proline and phenolices were correlated with Cd contents in plant. Cd significantly reduced the plant growth. Fertilizers application (in combination) most significantly increased the growth (19 cm root and 47 cm shoot) on Cd contaminated soil. All treatments increased the Cd contents in plant tissues. This increase was highly significant in fertilizers treated plants (1101, 121 and 544 ppm in roots, stem and leaves respectively). Significantly positive correlation was found between Cd concentration and dry biomass of root (R2 = 0.7511) and leaves (R2 = 0.5524). All treatments significantly increased the proline and total phenolics and maximum was recorded in NaCl treated plants followed by fertilizers. Proline was higher in roots while phenolics in leaves. The correlation between proline and phenolics was positive in leaf (R2 = 0.8439) and root (R2 = 0.5191). Proline and phenolics showed positive correlation with Cd concentration in plant. Conclusively, fertilizers in combination seem to be the better option for Cd phytoextraction. Further investigation is suggested to study the role of phenolics and proline in Cd phytoextraction.  相似文献

The expression of Δ1-pyrroline-5-carboxylate reductase (P5CR) gene was found to be higher in soybean root nodules than in leaves and roots, and its expression in roots appeared to be osmoregulated (AJ Delauney, DPS Verma [1990] Mol Gen Genet 221: 299-305). P5CR was purified to homogeneity as a monomeric protein of 29 kilodaltons by overexpression of a soybean P5CR cDNA clone in Escherichia coli. The pH optimum of the purified P5CR was altered by increasing the salt concentration, and maximum enzyme activity was attainable at a lower pH under high salt (0.2-1 molar NaCl). Kinetic studies of the purified enzyme suggested that nicotinamide adenine dinucleotide phosphate+ inhibited P5CR activity, whereas nicotinamide adenine dinucleotide+ did not. Subcellular fractionation and antibodies raised against purified soybean P5CR were used to investigate location of the enzyme in different parts of soybean as well as in leaves of transgenic tobacco plants synthesizing soybean P5CR. P5CR activity was present in cytoplasm of soybean roots and nodules as well as in leaves, but in leaves, about 15% of the activity was detected in the plastid fraction. The location of P5CR was further confirmed by western blot assay of the proteins from cytosol and plastid fractions of different parts of the plant. Expression of soybean nodule cytosolic P5CR in transgenic tobacco under the control of cauliflower mosaic virus 35S promoter led to the accumulation of this protein exclusively in the cytoplasm, suggesting that the chloroplastic activity may be due to the presence of a plastid form of the enzyme. The different locations of P5CR in root and leaf suggested that proline may be synthesized in different subcellular compartments in root and leaf. Proline concentration was not significantly increased in transgenic plants exhibiting high level P5CR activity, indicating that reduction of P5C is not a rate-limiting step in proline production.  相似文献

The scale of salt tolerance of each plant tissue can be characterized by a “critical” concentration of NaCl. At this concentration the endogenous content of free proline rises. A correlation exists between the endogenous Na+-concentration and this increase of proline. Calcium cations can enhance the NaCl-tolerance by inhibiting the Na+-permeability. Thus, the rise of the proline concentration in this case is produced only by NaCl concentrations higher than the critical concentration. The calcium pretreatment of caryopses during soaking leads to the enhancement of the tolerance under salt stress, too.   相似文献
