Groundnuts grown in the postrainy season under terminal drought stress imposed by withholding irrigation, or under a water-deficit gradient created by line-source sprinkler irrigation, were examined for preharvest aflatoxin contamination. High levels of aflatoxin B1 were found in damaged seeds in both situations. When grown under continuous drought-stress, toxin levels in damaged seed samples ranged from 1480 to 2467 ?g/kg in the 1990/91, and 1.3 to 2000 ?g/kg in the 1991/92 postrainy seasons. Aflatoxin B1 contamination in all damaged seed samples increased with increasing water deficit; toxin levels ranged from 26 to 850 ?g/kg across the water deficit gradient. Aflatoxin was either absent or almost negligible (1–2 ?g/kg) in apparently undamaged seed samples. Low risk of aflatoxin contamination in apparently undamaged seeds of groundnuts grown in postrainy seasons is indicated, even when there is terminal drought stress.  相似文献

Aflatoxin contamination in five varieties of pearl millet (ICMH-451, ICMP-50I, ICTP-8203, WCC-75 and ICMV-155) was studied from field and storage conditions in three districts of Andhra Pradesh State, India and the inter-relationships between various parameters such as stage of grain maturation in the field and insect pest infestation in storage in relation to aflatoxin production were evaluated. Aflatoxin contamination was more frequent in the seed samples collected from the fields during rainy season than winter season. All major aflatoxins were isolated from one or the other varieties of pearl millet, whereas aflatoxin G2 was not commonly observed in the seed samples collected during winter. Among all the varieties tested, ICMH-451 was vulnerable to aflatoxin contamination whereas ICMV-155 was the least susceptible variety. The higher amount of aflatoxins was observed in the matured seed samples followed by pre-matured and milky stage. Among all the toxins reported in the field, aflatoxin B1 was found in higher concentration (185 (μg/kg) followed by B2 (105 μg/kg). The four major types of aflatoxins with higher levels (35, 40, 140, 190 μg/kg of G1, G2, B2, B1 were reported in the rainy season seed samples after six months of storage, whereas aflatoxin G1 was not observed in any variety of stored seed sample from winter. Statistical analysis revealed that the aflatoxin incidence in relation to different parameters studied was significantly different for each factor. The relationship between aflatoxin contamination and insect damaged-grain clearly indicated that the seed samples with 16-40% of insect damage contained higher amounts of aflatoxins (758 μg/kg). A survey of aflatoxin contamination in selected Colombian foods was conducted over a 12-month period on a total of 248 samples. Samples were collected in supermarkets, retail stores and stock centres and were grouped into five categories: (1) corn and corn products, (2) cereal grains, (3) rice and rice products, (4) legume seeds; and (5) snacks and breakfast cereals. Aflatoxins were identified and quantitated using a liquid chromatographic technique with a limit of detection of 1 ng/g for each aflatoxin. Aflatoxins were detected in 14 of 109 samples of corn and corn products, 4 of 40 samples of rice and rice products, 2 of 30 samples of legume seeds, and 2 of 11 samples of snacks and breakfast cereals. None of the cereal grains samples analysed contained detectable levels of aflatoxins. Twelve of the total of 22 positive samples exceeded the maximum tolerable level of aflatoxin B1 adopted in most countries (5 ng/g); 10 of these 12 samples corresponded to corn and corn products. The results of the present study indicate that aflatoxin B1 contamination in certain foods in Colombia is a major public health concern. Continuous monitoring of aflatoxin B1 levels in Colombian foods is advised.  相似文献

Thirty-nine species and 16 fungal genera were isolated from Egyptian cotton seeds, cotton seed meal and cotton seed cake on 1% glucose-Czapek's agar medium incubated at 28 °C. Aspergillus was the most frequent genus and it emerged in 87–100% of the samples contributing 70–98% of total fungi in the three substrates tested. The most common species were A. niger, A. flavus, A. fumigatus, A. terreus and Rhizopus stolonifer; A. niger, A. fumigatus and Penicillium corylophilum; and A. niger, A. flavus, A. terreus, A. nidulans and Rhizopus stolonifer, respectively. Cotton seeds and cotton seed products were naturally contaminated by aflatoxin B1 and B2. About 16% of the different substrates tested were positive for aflatoxin contamination. No citrinin, ochratoxin A, patulin, sterigmatocystin, diacetoxyscirpenol, T-2 toxin or zearalenone were detected in the samples assayed.  相似文献

Aflatoxin contamination and biodeterioration were examined in 302 samples of dry cowpeas and pigeon peas that were randomly purchased from 9 districts of the Southern Region of Malawi during July and November 2015. Further, the impact of flotation/washing on aflatoxin levels on the pulses was elucidated. Aflatoxin analyses involved immunoaffinity column (IAC) clean-up and HPLC quantification with fluorescence detection (FLD) while legume biodeterioration assessments were done by visual inspection. Aflatoxins were frequently detected in cowpea (24%, max., 66 μg/kg) and pigeon pea (22%, max., 80 μg/kg) samples that were collected in the month of July. Lower aflatoxin incidence of 15% in cowpeas (max., 470 μg/kg) and 14% in pigeon peas (max., 377 μg/kg) was recorded in the November collection. Overall, aflatoxin levels were significantly higher in the pulses that were collected in November. However, there were no significant differences in the total aflatoxin (aflatoxin B1 (AFB1) + AFB2 + AFG1 + AFG2) levels between the two types of pulses. Remarkably, in 76.2% of the aflatoxin positive cowpea and in 41.7% of the aflatoxin positive pigeon pea samples, aflatoxin G1 concentration exceeded aflatoxin B1. Insect damage percentage averaged at 18.1 ± 18.2% (mean ± SD) in the cowpeas and 16.1 ± 19.4% in pigeon peas. Mean discolouration percentage (number of pulses) of the cowpeas and pigeon peas was found to be at 6.7 ± 4.9 and 8.7 ± 6.2%, respectively. Washing and discarding the buoyant fraction was highly efficient in reducing aflatoxin levels; only 5.2 ± 11.1% of the initial aflatoxin level was found in the cleaned samples. In conclusion, cowpeas and pigeon peas sold on the local market in Malawi may constitute a hazard especially if floatation/washing step is skipped.  相似文献

Inhibition in seed germination, seedling growth, chlorophyll, protein and nucleic acid syntheses was found to be due to aflatoxin B1. A survey was carried out to obtain data on the occurrence of mycotoxins and the mycotoxin-producing potential of fungi isolated from nuts (almonds, peanuts, hazelnuts, pistachio nuts) and sunflower seeds in Spain. Thin-layer chromatography was used to separate the toxins. Aflatoxins were detected in one sample of almonds (95 ppb aflatoxin B1 and 15 ppb aflaxtoxin B2) and in one sample of peanuts at a level below 10 ppb of aflatoxin B1. 100% of samples showed variable incidence of fungal contamination. The predominant fungi present in samples were Penicillium spp, Aspergillus niger, A. flavus, A. glaucus and Rhizopus spp. The results showed that isolates of different species were able to produce aflatoxins B1, B2, G1, and G2, sterigmatocystin, ochratoxin A, patulin, citrinin, penicillic acid, zearalenone, and griseofulvin.  相似文献

The contamination of cow’s milk at the farm level with aflatoxin M1 was investigated in South Africa. Samples of feeds, forage, maize and milk were taken at nine dairy farms, and at the same time samples of the processed milk (retail milk) were collected from the respective dairies to which the farms delivered their milk. The feeds were analysed for aflatoxin B1 and the milk samples for aflatoxin M1 using high performance liquid chromatography (HPLC) and fluorescence detection. All milk samples from the dairy farms were positive for aflatoxin M1, ranging from 0.02 μg/l to 1.5 μg/l. Retail milk was also frequently contaminated with AFM1, at levels of 0.01-3.1 μg/l. High AFB1 levels in feed materials on the farms supplying the raw milk indicate that various sources account for this contamination frequency in milk.  相似文献

Natural occurrence of fumonisins B1 (FB1) and B2 (FB2), a promoter for hepato-carcinogenesis, was investigated in corn and corn — based products sampled in Japan, Nepal, and China by high — performance liquid chromatographic method. From the 9 imported corn kernel and 6 gluten feed samples, FB1 was detected in 8 corn (0.6 ~ 4.1μg/g) and all gluten feed (0.3 ~ 2.4μg/g) samples, while FB2 was found in the same corn (0.3 ~ 10μg/g) and 3 gluten feed (0.8 ~ 8.5μg/g) samples. ELISA analysis also revealed the contamination of aflatoxin B1 in 2 corn and all gluten feed samples along with fumonisins. Of 17 corn grit samples, 14 and 5 samples were contaminated with fumonisin B1 and B2, with maximum levels of 2.6 and 2.8μg/g, respectively. As for corn-based foodstuffs marketed in Japan, no significant contamination of fumonisins was observed. Among 24 corn kernel samples in Nepal, 12 and 7 samples were positive for FB1 and FB2, and averaged to 0.6 and 1.6μg/g, respectively. One sample showed the highest fumonisin contents as 4.6 and 5.5μg/g, respectively. In corn samples harvested at Shanghai and Beijing, China, FB1 and FB2 were detected in various concentrations. Mycological survey has also revealed the presence of a fumonisin — producing fungus in a crop field of Japan. These findings have for the first time demonstrated high levels of contamination of fumonisins in corn and corn — based products in Asian countries. Natural co — occurrence of fumonisins and aflatoxin B1 was also detected in raw materials for mixed feed.  相似文献

In Zambia, groundnut products (milled groundnut powder, groundnut kernels) are mostly sold in under-regulated markets. Coupled with the lack of quality enforcement in such markets, consumers may be at risk to aflatoxin exposure. However, the level of aflatoxin contamination in these products is not known. Compared to groundnut kernels, milled groundnut powder obscures visual indicators of aflatoxin contamination in groundnuts such as moldiness, discoloration, insect damage or kernel damage. A survey was therefore conducted from 2012 to 2014, to estimate and compare aflatoxin levels in these products (n = 202), purchased from markets in important groundnut growing districts and in urban areas. Samples of whole groundnut kernels (n = 163) and milled groundnut powder (n = 39) were analysed for aflatoxin B1 (AFB1) by competitive enzyme-linked immunosorbent assay (cELISA). Results showed substantial AFB1 contamination levels in both types of groundnut products with maximum AFB1 levels of 11,100 μg/kg (groundnut kernels) and 3000 μg/kg (milled groundnut powder). However, paired t test analysis showed that AFB1 contamination levels in milled groundnut powder were not always significantly higher (P > 0.05) than those in groundnut kernels. Even for products from the same vendor, AFB1 levels were not consistently higher in milled groundnut powder than in whole groundnut kernels. This suggests that vendors do not systematically sort out whole groundnut kernels of visually poor quality for milling. However, the overall contamination levels of groundnut products with AFB1 were found to be alarmingly high in all years and locations. Therefore, solutions are needed to reduce aflatoxin levels in such under-regulated markets.  相似文献

The effect of different concentrations of aflatoxin B1 (100, 250, 500, 1000, 2000 µg/l) was studied on germination index and seedling growth in three varieties of wheat seeds. Inhibition in the above process was directly influenced by the concentration of toxin. Concentration of toxin had highly significant effect (p<0.001) for seed germination rate and radicle and plumule development. Inhibition dose for 50% reduction in germination rate (ID50) determined by probit analysis was maximum for the variety HP-129 (895 µg/l).  相似文献

Contamination of milk and dairy products with aflatoxin M1 (AFM1) presents a risk for human health. The aim of this study was to investigate the presence of AFM1 in pasteurized milk samples in Fariman, located in the province of Khorasan Razavi, Iran, by enzyme-linked immunosorbent assay (ELISA). Forty-five samples of pasteurized milk from different supermarkets were collected during 3 months in summer (July to September, 2012). AFM1 contamination was detected in all of milk samples. The mean concentration of aflatoxin M1 was 27.2 ng/l. The range of AFM1 content was 8.8–64 ng/l. Thirteen (28.8 %) of the samples had AFM1 levels exceeding the maximum levels (50 ng/l) accepted by the European Union. Due to the fact that milk is used by all the age groups including infants and children in Fariman city, it is necessary to minimize the health risk from AFM1 contamination in milk. For this reason, the level of its precursor, aflatoxin B1 (AFB1), in dairy feeds must be reduced, requiring constant aflatoxin monitoring of relevant agricultural commodities.  相似文献

A survey of red chilli (Capsicum indicum) for contamination with aflatoxins was performed on different samples comprising whole, crushed and powdered red chilli collected from various stores located in the city of Karachi, Pakistan. Red chilli required rather rigorous clean-up procedure for removal of adulterants and interference resulting from various types of compounds. A modified Romer method followed by bi-directional thin layer chromatography (TLC) was used for the detection of aflatoxins and confirmatory tests were performed by spraying the TLC plates with 50% sulphuric acid and making the derivative with trifluoroacetic acid. Of all the 176 samples of red chilli examined, 66% were found to be contaminated with aflatoxin B1. Generally, samples of red chilli exammined were found to be fairly low in aflatoxin B1 content, whereas only seven samples were found to contain concentrations greater than 25 μg/kg of aflatoxin B1.  相似文献

A study was undertaken to evaluate aflatoxin B1 contamination in coffee beans. 41 samples of green coffee were collected from large lots of material by representative sampling. The raw samples were analyzed and showed no detectable levels of aflatoxin B1. In order to establish the heat stability of the toxin, 3 artificially contaminated samples (average level 10/μg/kg) were roasted atca 200°C for different operation times periods so as to reproduce light and dark roasting procedures. Each sample was roasted both electrically and by gas. The percentage of toxin destruction was up to 93% for light roasted and 99% for dark roasted coffee with a slightly higher rate up to 100% for the electrically roasted coffee for light and dark roasting. In order to evaluate the potential migration of the aflatoxin B1 into the coffee beverage, 1 sample found contaminated after roasting treatment (0.8/°g/kg) was extracted using each of the 3 most common types of coffee makers. Additional destruction of the toxin was observed (up to 99%) in two cases while only 75% of fate was obtained in the third. The process from raw coffee beans to beverage showed a meaningful destruction of aflatoxin B1, ranging from 97 to 100% depending on the extraction technique adopted in the preparation of the beverages.  相似文献

Aim: The aim of this work was to evaluate the effect of Planococcus ficus infection in red wine grapes on Aspergillus section Nigri and ochratoxin A (OTA) contamination. Methods and Results: During 2006/2007 and 2008/2009 vintages, Merlot, Malbec and Cabernet Sauvignon varieties divided into two categories of grape samples (undamaged and damaged by P. ficus) were evaluated. Regardless of the grape variety and the harvest season evaluated, Aspergillus section Nigri incidence and the mean OTA concentration in damaged berries were significantly higher than that in the undamaged ones (P < 0·05; P < 0·001). The Merlot variety showed the highest level of black aspergilli contamination in damaged grapes during the 2006/2007 vintage (53·5% of infection), whereas Malbec presented the highest incidence during the 2008/2009 vintage (57·6% of infection). The Cabernet Sauvignon variety showed the highest OTA levels, ranging from 0·1 to 140 μg kg?1. Conclusions: The presence of P. ficus in vineyards increased the risk of OTA occurrence in grapes, suggesting the need to implement insect control at preharvest stage to reduce the entry of OTA in the wine production chain. Significance and Impact of the Study: This study is the first report on the influence of P. ficus on the potential risk of OTA contamination in grapes.  相似文献

During a 5-year period from 2010 to 2014, n = 919 samples of feed and raw materials were analyzed for aflatoxin B1 (AFB1) contamination using accredited ELISA screening methods. Only 0.76 % of these samples were non-compliant with maximum levels set by the European Union Regulation 32/2002. Non-compliant samples were mainly from the province of Bari (n = 3 samples, mean AFB1 value 7.03 μg/kg), although the highest AFB1 levels were found in two samples from the provinces of Foggia and Brindisi, at 32.6 ± 3.6 μg/kg and 31.0 ± 4.0 μg/kg, respectively. Mean AFB1 levels in samples contaminated but compliant with the limits ranged from 1.4 to 2.2 μg/kg. Considering the great importance of climate conditions in mycotoxins production, during crops production and during the critical phases of materials storage and/or transport, to better understand the variability in contamination levels, the analytical results were reviewed in term of temperature and relative environmental humidity in the sampling areas. Correlations between aflatoxin B1 levels in feed and these climate factors might explain seasonal and annual variations in contamination levels. The data from the present study provide useful suggestions for the organization of targeted monitoring plans and the protection of consumers, as well as for improvement in the quality standards of zootechnological activities and feed industry.  相似文献

It is important to determine the factors prompting seed dispersal because for plant species seed dispersal is the only opportunity to disperse into a new habitat. Previous studies showed that the maternal stress, such as high density and low nutrient levels, induces the adaptive plastic increase of the dispersal ability in seed heteromorphic plants. In this study, we examined whether herbivory can change the relative proportion of dispersal-related seed heteromorphism (floating or non floating seeds) in an invasive weed Ambrosia artemisiifolia. Because A. artemisiifolia often distributes in the riparian habitat, floating seeds might contribute to the long distance dispersal by hydrochory. Floating ability and seed weight were compared between plants damaged by a specialist herbivore Ophraella communa and undamaged plants. The damaged plants produced lighter and more likely floating seeds than the undamaged plants. However, multi-regression analysis revealed that the probability of floating was affected by seed weight but was not affected by herbivore treatment (damaged vs. undamaged plants). These results suggest that the increased proportion of floating seeds was not a direct response to the herbivore signal but an indirect response through the herbivore's effect on the reduction of seed weight. Plants damaged by herbivores might not only decrease seed production and quality but also increase the dispersal ability. These responses in dispersal ability against the herbivores might contribute to the spread of invasive plants.  相似文献

Five different concentrations (100, 250, 500, 1000 and 2000 μg/L of aflatoxin B1 were found to be inhibitory to seed germination and seedling growth (root and shoot lengths) of mustard seeds (variety Pusa bold). These also lowered the levels of chlorophyll and carotenoids in the emerging leaves during seedling growth. The inhibitory effect was correlated with the concentration of applied toxin.  相似文献

The occurrence of spoilage fungi and Aspergillus section Flavi populations, the aflatoxins incidence, the role of insects as vectors of mycotoxin-producing fungi and the AFs-producing ability of the isolated species throughout the peanut (Arachis hypogaea L.) storage period were evaluated. Analyses of fungal populations from 95 peanut seed samples did not demonstrate significant differences between the incidences in each sampling period. Aspergillus section Flavi were isolated during all incubation periods. Cryptolestes spp. (Coleoptera: Cucujidae) were collected in August, September and October with 18, 16 and 28% of peanut samples contaminated, respectively. Insects isolated during August showed 69% of Aspergillus section Flavi contamination. A. flavus was the most frequently isolated (79%) from peanut seeds and from insect (59%). The greater levels of AFB1 were detected in September and October with a mean of 68.86 μg/kg and 69.12 μg/kg respectively. The highest proportion of A. flavus toxigenic strains (87.5%) was obtained in June. The presence of Aspergillus section Flavi and insect vectors of aflatoxigenic fungi presented a potential risk for aflatoxin production during the peanut storage period. The staple crops, maize, sorghum, bambara nut, groundnut, and sunflower common in semi-arid agro-pastoral farming systems of central Tanzania are prone to aflatoxin contamination. Consumption of such crop produce, contaminated with high levels of aflatoxin B1 (AFB1), affects growth and health. In this paper, aflatoxin contamination in freshly harvested and stored crop produce from central Tanzania was examined, including the efficacy of aflatoxin mitigation technologies on grain/kernal quality. A total of 312 farmers were recruited, trained on aflatoxin mitigation technologies, and allowed to deploy the technologies for 2 years. After 2 years, 188 of the 312 farmers were tracked to determine whether they had adopted and complied with the mitigation practices. Aflatoxigenic Aspergillus flavus and aflatoxin B1 contamination in freshly harvested and stored grains/kernels were assessed. A. flavus frequency and aflatoxin production by fungi were assayed by examining culture characteristics and thin-layer chromatography respectively. AFB1 was assayed by enzyme-linked immunosorbent assay. The average aflatoxin contamination in freshly harvested samples was 18.8 μg/kg, which is above the acceptable standard of 10 μg/kg. Contamination increased during storage to an average of 57.2 μg/kg, indicating a high exposure risk. Grains and oilseeds from maize, sorghum, and sunflower produced in aboveground reproductive structures had relatively low aflatoxin contamination compared to those produced in geocarpic structures of groundnut and bambara nut. Farmers who adopted recommended post-harvest management practices had considerably lower aflatoxin contamination in their stored kernels/grains. Furthermore, the effects of these factors were quantified by multivariate statistical analyses. Training and behavioral changes by farmers in their post-harvest practice minimize aflatoxin contamination and improve food safety. Moreover, if non-trained farmers receive mitigation training, aflatoxin concentration is predicted to decrease by 28.9 μg/kg on average.  相似文献
