Onion (Allium cepa L.) plants were examined to determine the photosynthetic role of CO2 that accumulates within their leaf cavities. Leaf cavity CO2 concentrations ranged from 2250 L L–1 near the leaf base to below atmospheric (<350 L L–1) near the leaf tip at midday. There was a daily fluctuation in the leaf cavity CO2 concentrations with minimum values near midday and maximum values at night. Conductance to CO2 from the leaf cavity ranged from 24 to 202 mol m–2 s–1 and was even lower for membranes of bulb scales. The capacity for onion leaves to recycle leaf cavity CO2 was poor, only 0.2 to 2.2% of leaf photosynthesis based either on measured CO2 concentrations and conductance values or as measured directly by 14CO2 labeling experiments. The photosynthetic responses to CO2 and O2 were measured to determine whether onion leaves exhibited a typical C3-type response. A linear increase in CO2 uptake was observed in intact leaves up to 315 L L–1 of external CO2 and, at this external CO2 concentration, uptake was inhibited 35.4±0.9% by 210 mL L–1 O2 compared to 20 mL L–1 O2. Scanning electron micrographs of the leaf cavity wall revealed degenerated tissue covered by a membrane. Responses of the rate of net CO2 assimilation (A) to the intercellular partial pressure of CO2 (p i ) were measured on intact spinach (Spinacia oleracea L.) leaves at different irradiances. These responses were analysed to find the value of p i at which the rate of photosynthetic CO2 uptake equalled that of photorespiratory CO2 evolution. At this CO2 partial pressure (denoted ), net rate of CO2 assimilation was negative, indicating that there was non-photorespiratory CO2 evolution in the light. Hence was lower than the CO2 compensation point, . Estimates of were obtained at leaf temperatures from 15 to 30°C, and the CO2/O2 specificity of ribulose 1,5-bisphosphate (RuBP) carboxylase/oxygenase (E.C. 4.1.1.39) was calculated from these data, taking into account changes in CO2 and O2 solubilities with temperature. The CO2/O2 specificity decreased with increasing temperature. Therefore we concluded that temperature effects on the ratio of photorespiration to photosynthesis were not solely the consequence of differential effects of temperature on the solubilities of CO2 and O2. Our estimates of the CO2/O2 specificity of RuBP carboxylase/oxygenase are compared with in-vitro measurements by other authors. The rate of nonphotorespiratory CO2 evolution in the light (R d ) was obtained from the value of A at . At this low CO2 partial pressure, R d was always less than the rate of CO2 evolution in darkness and appeared to decrease with increasing irradiance. The decline was most marked up to about 100 mol quanta m-2 s-1 and less marked at higher irradiances. At one particular irradiance, however, R d as a proportion of the rate of CO2 evolution in darkness was similar in different leaves and this proportion was unaffected by leaf temperature or by [O2] (ambient and greater). Photoautotrophic micropropagation of Russet Burbank Potato   总被引:2,自引:0,他引:2  
The photoautotrophic micropropagation of potato cv. Russet Burbank was investigated. Single node microcuttings were grown for four weeks on Murashige and Skoog (MS) medium with or without sucrose (30 g l–1) in the growth room at 21/19 °C day/night temperature, with 16-h photoperiod at 150 mol m–2 s–1, with or without supplemental CO2 at 1500 l l–1. A 20% increase in the number of nodes per stem (from 7.5 to 9.4) and a 50% increase in stem dry weight were observed in cultures grown on media with sucrose and in CO2 enriched atmosphere comparing to the conventionally micropropagated cultures or the cultures grown photoautotrophically on media without sucrose but in air supplemented with 1500 l l–1CO2. Stems of these cultures (from media with sucrose in CO2 enriched air) almost doubled in length the stems of cultures from the other two treatments. No significant differences were observed between Control (MS medium supplemented with sucrose, 30 g l–1) and photoautotrophic cultures coming from MS medium with no sucrose grown under 1500 l l–1 of CO2. Photoautotrophic cultures produced stems averaging 43.3 mm, with 7 nodes and weighing 9.2 mg (dry weight), similar to conventionally grown in vitro cultures (47.9 mm with 7.5 nodes, 9.7 mg dry weight). Growers may consider photoautotrophic culturing of potato in areas where the high sterility levels are difficult to maintain. Supplementing air in the growth room with 1500 l l–1 of CO2 could be beneficial for potato plantlet production even on media containing sucrose since it significantly improved quality, size and biomass of produced plantlets, speeding up the multiplication.  相似文献

Dry weight and Relative Growth Rate of Lemna gibba were significantly increased by CO2 enrichment up to 6000 l CO2 l–1. This high CO2 optimum for growth is probably due to the presence of nonfunctional stomata. The response to high CO2 was less or absent following four days growth in 2% O2. The Leaf Area Ratio decreased in response to CO2 enrichment as a result of an increase in dry weight per frond. Photosynthetic rate was increased by CO2 enrichment up to 1500 l CO2 l–1 during measurement, showing only small increases with further CO2 enrichment up to 5000 l CO2 l–1 at a photon flux density of 210 mol m–2 s–1 and small decreases at 2000 mol m–1 s–1. The actual rate of photosynthesis of those plants cultivated at high CO2 levels, however, was less than the air grown plants. The response of photosynthesis to O2 indicated that the enhancement of growth and photosynthesis by CO2 enrichment was a result of decreased photorespiration. Plants cultivated in low O2 produced abnormal morphological features and after a short time showed a reduction in growth.  相似文献

Summary In order to document the natural CO2 environment of the moss Hylocomium splendens, and ascertain whether or not the moss was adapted to this, and its interactions with other microenvironmental factors, two studies were carried out. Firstly, the seasonal variations of CO2 concentration, photosynthetically active radiation (PAR), tissue water content and temperature were measured in the natural microenvironment of H. splendens in a subarctic forest during the summer period (July–September). Secondly, the photosynthetic responses of the species to controlled CO2 concentrations, PAR, temperature, and hydration were measured in the laboratory. CO2 concentrations around the upper parts of the plant, when PAR was above the compensation point (30 mol m–2 s–1), were mostly between 400 and 450 ppm. They occasionally increased up to 1143 ppm for short periods. PAR flux densities below saturating light levels for photosynthesis (100 mol m–2 s–1), occurred during 65% (July), 76% (August) and 96% (September) of the hours of the summer period. The temperature optimum of photosynthesis was 20° C: this temperature coincided with PAR above the compensation point during 5%, 6% and 0% of the time in July, August and September, respectively. Optimal hydration of tissues was infrequent. Hence PAR, temperature and water limit CO2 uptake for most of the growing season. Our data suggest that the higher than normal ambient CO2 concentration in the immediate environment of the plant counteracts some of the limitations in PAR supply that it experiences in its habitat. This species already experiences concentrations of atmospheric CO2 predicted to occur over the next 50 years.  相似文献

Summary Photosynthesis and transpiration was measured in the large emergent C4 sedge Cyperus papyrus (papyrus) which occupies wide areas of wetland on the African continent. The maximum observed value of net assimilation was 35 mol CO2 m-2 s-1 at full sunlight but light saturation of photosynthesis did not occur. The quantum yield of photosynthesis obtained from the initial slope of the light response curves (0.06 mol mol-1 incident light) was relatively high and close to previously recorded values for some C4 grasses. Measurements made over two days showed that stomatal conductance was sensitive to the ambient air vapour pressure deficit (VPD) and was consistently lower on the day when VPD's were higher. There was, however, no marked midday closure of the stomata. Photosynthesis was also reduced on the day when VPD's were higher. The relationship between net photosynthesis and stomatal conductance was close to linear over the range of measurement conditions, with the result that intercellular CO2 concentrations (C i ) did not vary markedly. There was some evidence that C i decreased at high VPD's. The regulation of stomatal movement in papyrus appears to minimise excessive water loss while not severely limiting photosynthesis. The significance of this strategy for a wetland species with plentiful supplies of water is discussed.  相似文献

Short-term responses of four carrot (Daucus carota) cultivars: Cascade, Caro Choice (CC), Oranza, and Red Core Chantenay (RCC) to CO2 concentrations (C a) were studied in a controlled environment. Leaf net photosynthetic rate (P N), intercellular CO2 (C i), stomatal conductance (g s), and transpiration rate (E) were measured at C a from 50 to 1 050 mol mol–1. The cultivars responded similarly to C a and did not differ in all the variables measured. The P N increased with C a until saturation at 650 mol mol–1 (C i= 350–400 mol mol–1), thereafter P N increased slightly. On average, increasing C a from 350 to 650 and from 350 to 1 050 mol mol–1 increased P N by 43 and 52 %, respectively. The P N vs. C i curves were fitted to a non-rectangular hyperbola model. The cultivars did not differ in the parameters estimated from the model. Carboxylation efficiencies ranged from 68 to 91 mol m–2 s–1 and maximum P N were 15.50, 13.52, 13.31, and 14.96 mol m–2 s–1 for Cascade, CC, Oranza, and RCC, respectively. Dark respiration rate varied from 2.80 mol m–2 s–1 for Oranza to 3.96 mol m–2 s–1 for Cascade and the CO2 compensation concentration was between 42 and 46 mol mol–1. The g s and E increased to a peak at C a= 350 mol mol–1 and then decreased by 17 and 15 %, respectively when C a was increased to 650 mol mol–1. An increase from 350 to 1 050 mol mol–1 reduced g s and E by 53 and 47 %, respectively. Changes in g s and P N maintained the C i:C a ratio. The water use efficiency increased linearly with C a due to increases in P N in addition to the decline in E at high C a. Hence CO2 enrichment increases P N and decreases g s, and can improve carrot productivity and water conservation.  相似文献

Photosynthetic gas exchange characteristics of two common boreal forest mosses, Sphagnum (section acutifolia) and Pleurozium schreberi, were measured continuously during the time required for the moss to dry out from full hydration. Similar patterns of change in CO2 assimilation with variation in water content occurred for both species. The maximum rates of CO2 assimilation for Sphagnum (approx. 7 mol m–2 s–1) occurred at a water content of approximately 7 (fresh weight/dry weight) while for Pleurozium the maximum rate (approx. 2 mol m–2 s–1) occurred at a water content of approximately 6 (fresh weight/dry weight). Above and below these water contents CO2 assimilation declined. In both species total conductance to water vapour (expressed as a percentage of the maximum rates) remained nearly constant at a water content above 9 (fresh weight/dry weight), but below this level declined in a strong linear manner. Short-term, on-line 13CO2 and C18O16O discrimination varied substantially with changes in moss water content and associated changes in the ratio of chloroplast CO2 to ambient CO2 partial pressure. At full hydration (maximum water content) both Sphagnum and Pleurozium had similar values of 13CO2 discrimination (approx. 15). Discrimination against 13CO2 increased continuously with reductions in water content to a maximum of 27 in Sphagnum and 22 in Pleurozium. In a similar manner C18C16O discrimination increased from approximately 30 at full hydration in both species to a maximum of 150 in Sphagnum and 90 in Pleurozium, at low water content. The observed changes in C18O16O were strongly correlated to predictions of a mechanistic model of discrimination processes. Field measurements of moss water content suggested that photosynthetic gas exchange by moss in the understory of a black spruce forest was regularly limited by low water content.  相似文献

On steep, unvegetated slopes, sands (particle sizes 0.063 to 2 mm) and gravels (2–64 mm) erode from the shoreline of Lake Naivasha (Kenya) and enter the lake basin. This occurred freely where fringing papyrus (Cyperus papyrus) swamp had been cleared in favour of landing jetties or agriculture. Gravel-sized particles have been recovered up to 60 m offshore and sands to 80 m. In an area where papyrus was undisturbed and the swamp margin was 52 m wide, gravel did not enter the lake and sands penetrated to around 35 m. Large particles are much easier to trace to source and to manage than the finer silts and clays (<0.63 m) that form the bulk of Lake Naivasha's sediment. The pattern of mass specific magnetic susceptibilities for the <63 m fraction of lake sediment around the southern shoreline of the lake suggests that fine particles enter open water directly from the shoreline and are not transported from the lake's only perennial inflow, the River Malewa in the north. Such particles originate from a hinterland that supports high intensity horticulture and are therefore a potential source of contamination. Mean ± SD susceptibility (If) immediately offshore papyrus fringe was 0.49 ± 0.08 × 10-6 m3 kg-1 compared with higher values of 1.33 ± 0.14 × 10-6 m3 kg-1 where there was no papyrus barrier (P<0.0001). The value for five sites in the middle parts of the lake was 0.45 ± 0.02 × 10-6 m3 kg-1 with 1.38 ± 0.10 × 10-6 m3 kg-1 near the mouth of the River Malewa. The results of this study are evidence, therefore, that conservation of a continuous papyrus margin of about 50 m width is a priority for intercepting particulate material.  相似文献

The terrestrial blue-green alga (cyanobacterium), Nostoc flagelliforme, was cultured in air at variouslevels of CO2, light and watering to see theireffects on its growth. The alga showed the highestrelative growth rate at the conditions of highCO2 (1500 ppm), high light regime (219–414mol m-2s-1) and twice daily watering,but the lowest rate at the conditions of low light(58–114 mol m-2s-1) and daily twicewatering. Increased watering had little effect ongrowth rate at 350 ppm CO2, but increased byabout 70% at 1500ppm CO2 under high lightconditions. Summary The apparent energy of activation (E a), Michaelis-Menten constant (K mfor oxaloacetate), V max/K mratios and specific activities of NADP+-malate dehydrogenase (NADP+-MDH; EC 1.1.1.82) were analyzed in plants of Barnyard grass from Québec (QUE) and Mississippi (MISS) acclimated to two thermoperiods 28/22°C, 21/15°C, and grown under two CO2 concentrations, 350 l l-1 and 675 l l-1. E avalues of NADP+-MDH extracted from QUE plants were significantly lower than those of MISS plants. K mvalues and V max/K mratios of the enzyme from both ecotypes were similar over the range of 10–30°C but reduced V max/K mratios were found for the enzyme of QUE plants at 30 and 40°C assays. MISS plants had higher enzyme activities when measured on a chlorophyll basis but this trend was reversed when activities were expressed per fresh weight leaf or per leaf surface area. Activities were significantly higher in plants of both populations acclimated to 22/28°C. CO2 enrichment did not modify appreciably the catalytic properties of NADP+-MDH and did not have a compensatory effect upon catalysis or enzyme activity under cool acclimatory conditions. NADP+-MDH activities were always in excess of the amount required to support observed rates of CO2 assimilation and these two parameters were significantly correlated. The enhanced photosynthetic performance of QUE plants under cold temperature conditions, as compared to that of MISS plants, cannot be attributed to kinetic differences of NADP+-malate dehydrogenase among these ecotypes.  相似文献

The interaction of extreme temperature events with future atmospheric CO2 concentrations may have strong impacts on physiological performance of desert shrub seedlings, which during the critical establishment phase often endure temperature extremes in conjunction with pronounced drought. To evaluate the interaction of drought and CO2 on photosynthesis during heat stress, one-year-old Larrea tridentata[DC] Cov. seedlings were exposed to nine days of heat with midday air temperature maxima reaching 53 °C under three atmospheric CO2 concentrations (360, 550 and 700 mol mol–1) and two water regimes (well-watered and droughted). Photosynthetic gas exchange, chlorophyll fluorescence and water potential responses were measured prior to, during and one week following the high temperature stress event. Heat stress markedly decreased net photosynthetic rate (A net), stomatal conductance (g s), and the photochemical efficiency of photosystem II (F v/F m) in all plants except for well-watered L. tridentata grown in 700 mol mol–1 CO2. A net and g s remained similar to pre-stress levels in these plants. In droughted L. tridentata, A net was ca. 2× (in 550 mol mol–1 CO2) to 3× (in 700 mol mol–1 CO2) higher than in ambient-CO2-grown plants, while g s and F v/F m were similar and low in all CO2 treatments. Following heat stress, g s in all well-watered plants rose dramatically, exceeding pre-stress levels by up to 100%. In droughted plants, g s and A net rose only in plants grown at elevated CO2 following release from heat. This recovery response was strongest at 700 mol mol–1 CO2, which returned to A net and g s values similar to pre-heat following several days of recovery. Extreme heat diminished the photosynthetic down-regulation response to growth at elevated CO2 under well-watered conditions, similar to the action of drought. Ambient-CO2-grown L. tridentata did not show significant recovery of photosynthetic capacity (A \max and CE) after alleviation of temperature stress, especially when exposed to drought, while plants exposed to elevated CO2 appeared to be unaffected. These findings suggest that elevated CO2 could promote photosynthetic activity during critical periods of seedling establishment, and enhance the potential for L. tridentata to survive extreme high temperature events.  相似文献

Elevated CO2 has been shown to increase methane emissions in herbaceous wetlands, but it is not clear that this will occur in wetlands dominated by woody plants or in wetlands that are not inundated. We determined the effects of elevated CO2 and water table position on methane emission and oxidation rates from plant-soil microcosms planted with a woody tree, Taxodium distichum, or an emergent aquatic macrophyte, Orontium aquaticum. Experiments were conducted in replicate glasshouses (n = 2) at CO2 concentrations of either 350 or 700 ppmv. Plants were grown from seed and subjected to two water level depths, flooded (+5 cm above the soil surface) and non-flooded (–10 cm for T. distichum and –6 cm for O. aquaticum). Elevated CO2 increased whole-plant photosynthetic rates in both water table treatments. Methane emission rates increased by 62 to 69% in the T. distichum treatment and 27 to 29% in the O. aquaticum treatment. Whole-plant photosynthesis and biomass were strongly correlated with methane emissions (r2 0.75, P 0.01). This relationship provides evidence of a tight coupling between plant and microbial activity and suggests that similar relationships from other wetland studies measured at ambient CO2 can be extrapolated into the future. In the O. aquaticum, non-flooded treatment, methanotrophy consumed 14 and 22% (replicate glasshouses) of the methane produced in the ambient treatment compared to 29 and 36% in the elevated CO2 treatment. However, there was no significant methane oxidation detected in the flooded treatment. We concluded that woody and non-woody wetland ecosystems growing in a future CO2-enriched atmosphere will emit more methane regardless of water table position, but the degree of stimulation will be sensitive to changes in water table position, particularly in forested wetlands.  相似文献

A unicellular marine picoplankton, Nannochloropsis sp., was grown under CO2-enriched photoautotrophic or/and acetate-added mixotrophic conditions. Photoautotrophic conditions with enriched CO2 of 2800 l CO2 l–1 and aeration gave the highest biomass yield (634 mg dry wt l–1), the highest total lipid content (9% of dry wt), total fatty acids (64 mg g–1 dry wt), polyunsaturated fatty acids (35% total fatty acids) and eicosapentaenoic acid (EPA, 20:53) (16 mg g–1 dry wt or 25% of total fatty acids). Mixotrophic cultures gave a greater protein content but less carbohydrates. Adding sodium acetate (2 mM) decreased the amounts of the total fatty acids and EPA. Elevation of CO2 in photoautotrophic culture thus enhances growth and raises the production of EPA in Nannochloropsis sp.  相似文献

Physiological responses to elevated CO2 at the leaf and canopy-level were studied in an intact pine (Pinus taeda) forest ecosystem exposed to elevated CO2 using a free-air CO2 enrichment (FACE) technique. Normalized canopy water-use of trees exposed to elevated CO2 over an 8-day exposure period was similar to that of trees exposed to current ambient CO2 under sunny conditions. During a portion of the exposure period when sky conditions were cloudy, CO2-exposed trees showed minor (7%) but significant reductions in relative sap flux density compared to trees under ambient CO2 conditions. Short-term (minutes) direct stomatal responses to elevated CO2 were also relatively weak (5% reduction in stomatal aperture in response to high CO2 concentrations). We observed no evidence of adjustment in stomatal conductance in foliage grown under elevated CO2 for nearly 80 days compared to foliage grown under current ambient CO2, so intrinsic leaf water-use efficiency at elevated CO2 was enhanced primarily by direct responses of photosynthesis to CO2. We did not detect statistical differences in parameters from photosynthetic responses to intercellular CO2 (A net-C i curves) for Pinus taeda foliage grown under elevated CO2 (550 mol mol–1) for 50–80 days compared to those for foliage grown under current ambient CO2 from similar-sized reference trees nearby. In both cases, leaf net photosynthetic rate at 550 mol mol–1 CO2 was enhanced by approximately 65% compared to the rate at ambient CO2 (350 mol mol–1). A similar level of enhancement under elevated CO2 was observed for daily photosynthesis under field conditions on a sunny day. While enhancement of photosynthesis by elevated CO2 during the study period appears to be primarily attributable to direct photosynthetic responses to CO2 in the pine forest, longer-term CO2 responses and feedbacks remain to be evaluated.  相似文献

CO2 exchange components of a temperate semi-desert sand grassland ecosystem in Hungary were measured 21 times in 2000–2001 using a closed IRGA system. Stand CO2 uptake and release, soil respiration rate (R s), and micrometeorological values were determined with two types of closed system chambers to investigate the daily courses of gas exchange. The maximum CO2 uptake and release were –3.240 and 1.903 mol m–2 s–1, respectively, indicating a relatively low carbon sequestration potential. The maximum and the minimum R s were 1.470 and 0.226 mol(CO2) m–2 s–1, respectively. Water shortage was probably more effective in decreasing photosynthetic rates than R s, indicating water supply as the primary driving variable for the sink-source relations in this ecosystem type.  相似文献

We examined the in situ CO2 gas-exchange of fruits of a tropical tree, Durio zibethinus Murray, growing in an experimental field station of the Universiti Pertanian Malaysia. Day and night dark respiration rates were exponentially related to air temperature. The temperature dependent dark respiration rate showed a clockwise loop as time progressed from morning to night, and the rate was higher in the daytime than at night. The gross photosynthetic rate was estimated by summing the rates of daytime dark respiration and net photosynthesis. Photosynthetic CO2 refixation, which is defined as the ratio of gross photosynthetic rate to dark respiration rate in the daytime, ranged between 15 and 45%. The photosynthetic CO2 refixation increased rapidly as the temperature increased in the lower range of air temperature T c (T c <28.5 °C), while it decreased gradually as the temperature increased in the higher range (T c 28.5 °C). Light dependence of photosynthetic CO2 refixation was approximated by a hyperbolic formula, where light saturation was achieved at 100 mol m–2 s–1 and the asymptotic CO2 refixation was determined to be 37.4%. The estimated gross photosynthesis and dark respiration per day were 1.15 and 4.90 g CO2 fruit–1, respectively. Thus the CO2 refixation reduced the respiration loss per day by 23%. The effect of fruit size on night respiration rate satisfied a power function, where the exponent was larger than unity.  相似文献

Palmaria palmata, which is able to use HCO inf3 sup– as a carbon source for photosynthesis, and Lomentaria articulata, which is dependent on diffusive uptake of dissolved CO2, were grown under constant light and light with sunflecks designed to model wave-induced fluctuations of near-shore underwater light. Both species exhibited significantly increased stable carbon isotope discrimination (more negative values of 13C relative to PDB) when grown with sunflecks. More negative 13C values were associated with decreased growth rate of P. palmata but not of L. articulata. In many temperate-zone ecosystems, seasonal changes in environmental and biological factors influence the dynamics and magnitude of surface–atmosphere exchange. Research was conducted between July and October 2001 to measure growing season surface-layer fluxes of CO2 in a Deyeuxia angustifolia dominated wetland on the Sanjiang Plain in northeastern China. Seasonal fluctuation and daily change in soil-surface CO2 fluxes were measured as well as the edaphic factors controlling CO2 fluxes. Soil-surface CO2 fluxes were measured with a closed-chamber system. The results revealed that there were both seasonal fluctuations and daily change in CO2 fluxes. The ranges of measured soil-surface CO2 flux were 0.208 – 1.265 g CO2m–2h–1. Soil-surface CO2 fluxes averaged 0.620 g CO2 m–2h–1. An analysis of several edaphic factors including soil temperature and soil moisture of the D. angustifolia wetland showed that there was a significant relationship between flux and temperature (R2 = 0.77).  相似文献

Intact cells of the unicellular cyanobacterium Synechococcus UTEX 625 degraded exogenously supplied cyanate (as KOCN) to CO2 and NH3 in a light-dependent reaction. NH3 release to the medium was as high as 80 mol(mgChl)-1h-1 and increased 1.7-fold in the presence of methionine sulfoximine, a glutamine synthetase inhibitor. Cyanate also supporte photosynthetic O2 evolution to a maximum rate of 188 mol O2(mgChl)-1h-1 at pH 8 and 30°C. Cyanate decomposition in cell-free extracts, measured by mass spectrometry as 13CO2 production from KO13CN, occurred in the soluble enzyme fraction, but not in the thylakoid/carboxysome fraction, and was enhanced by HCO3 and inhibited by the dianion oxalate. CO2, rather, than HCO3 , was a product of cyanate decomposition. The ability to decompose cyanate was not dependent upon pre-exposure of cells to cyanate to induce activity. The collective results indicate that Synechococcus UTEX 625 possesses a constitutive, cytosolic cyanase (EC 4.3.99.1), similar in mechanism to that found in some species of heterotrophic bacteria. The reaction catalyzed was: OCN+HCO3+2H+2CO2+NH3. In intact cells, the CO2 produced by the action of cyanase on OCN- was either directly fixed by the Calvin cycle enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase, leading to O2 evolution, or leaked into the medium where it was returned to the cell by the active CO2/HCO3 transport systems for fixation.
