ObjectiveWe investigated the levels and antimicrobial activity of antimicrobial proteins and peptides (AMPs) in breast milk consumed by preterm infants, and whether deficiencies of these factors were associated with late-onset neonatal sepsis (LOS), a bacterial infection that frequently occurs in preterm infants in the neonatal period.ResultsLevels of most AMPs and antibacterial activity in preterm breast milk were higher at day 7 than at day 21. Lactoferrin was the only AMP that limited pathogen growth >50% when added to formula at a concentration equivalent to that present in breast milk. Levels of AMPs were similar in the breast milk fed to infants with and without LOS, however, infants who developed LOS consumed significantly less breast milk and lower doses of milk AMPs than those who were free from LOS.ConclusionsThe concentrations of lactoferrin and defensins in preterm breast milk have antimicrobial activity against common neonatal pathogens.  相似文献

The phosphoramidites of the naturally occurring modified nucleotides N 2-methylguanosine and N 6,N 6-dimethyladenosine were synthesized and incorporated into short oligoribonucleotides. ObjectiveTo the assess the iodine status of preterm infants born in an area of iodine sufficiency using the urinary iodine concentration (UIC) and thyroid-stimulating hormone (TSH) levels and compare these values across different feeding practices during the first 7 days of life.MethodsIn this cross-sectional study, 88 preterm infants born at 30 to 34 weeks of gestation and admitted to the neonatal intensive care unit of a referral hospital in Tehran (Iran) were included. The infant UIC and TSH levels and breast milk iodine concentration in mothers who were exclusively breastfeeding were measured.ResultsMedian (interquartile range [IQR]) UIC and TSH levels in the study population were 81 (39-189) μg/L and 1.60 (0.80-2.85) mIU/L, respectively. When preterm infants were stratified by the type of feeding, the median (IQR) UICs were 64 (42-126) μg/L in parenteral nutrition, 125 (41-195) μg/L in exclusively breastfeeding, 57 (28-123) μg/L in formula feeding, and 45 (35-132) μg/L in mixed feeding, with no statistically significant difference between the groups (P = .31). The median (IQR) breast milk iodine concentration was 271 (177-521) μg/L in preterm infants exclusively fed their mothers’ own milk. There was no significant difference in the proportion of the TSH levels of >5 mIU/L between preterm infants who received enteral and parenteral nutrition (P = .27).ConclusionPreterm infants are at risk of iodine deficiency even in an area where the general population has adequate iodine. Only the preterm infants who received exclusively their mothers’ own milk had marginally adequate iodine status. Further studies are warranted to determine the necessity of iodine supplementation for this vulnerable group.  相似文献

It was suggested that the abnormally high N 2 O values found in 130,000–160,000 year-old Vostok ice core samples, characterized by high δ 15 N and low δ 18 O values, resulted from in situ microbial N 2 O production. To substantiate these observations we obtained new geochemical data from the last glacial period and showed the existence of additional small N 2 O anomalies. To test the hypothesis that microbial metabolism could contribute to these anomalies, we developed protocols for examining the ability of Nitrosomonas cryotolerans cells to produce N 2 O at subfreezing temperatures. Our results show that these model, frozen cultures produce N 2 O at temperatures as low as ?32°C.  相似文献

Fluxes of CO2 and N2O were measured from both natural and experimentally augmented snowpacks during the winters of 1993 and 1994 on Niwot Ridge in the Colorado Front Range. Consistent snow cover insulated the soil surface from extreme air temperatures and allowed heterotrophic activity to continue through much of the winter. In contrast, soil remained frozen at sites with inconsistent snow cover and production did not begin until snowmelt. Fluxes were measured when soil temperatures under the snow ranged from –5°C to 0°C, but there was no significant relationship between flux for either gas and temperature within this range. While early developing snowpacks resulted in warmer minimum soil temperatures allowing production to continue for most of the winter, the highest CO2 fluxes were recorded at sites which experienced a hard freeze before a consistent snowpack developed. Consequently, the seasonal flux of CO2 C from snow covered soils was related both to the severity of freeze and the duration of snow cover. Over-winter CO2 C loss ranged from 0.3 g C m−2 season−1 at sites characterized by inconsistent snow cover to 25.7 g C m−2 season−1 at sites that experienced a hard freeze followed by an extended period of snow cover. In contrast to the pattern observed with C loss, a hard freeze early in the winter did not result in greater N2ON loss. Both mean daily N2O fluxes and the total over-winter N2ON loss were related to the length of time soils were covered by a consistent snowpack. Over-winter N2ON loss ranged from less 0.23 mg N m−2 from the latest developing, short duration snowpacks to 16.90 mg N m−2 from sites with early snow cover. These data suggest that over-winter heterotrophic activity in snow-covered soil has the potential to mineralize from less than 1% to greater than 25% of the carbon fixed in ANPP, while over-winter N2O fluxes range from less than half to an order of magnitude higher than growing season fluxes. The variability in these fluxes suggests that small changes in climate which affect the timing of seasonal snow cover may have a large effect on C and N cycling in these environments. Abstract

The objective of this study was to evaluate dry matter (DM), nitrogen yield, N2 fixation (Ndfa) and soil N uptake (Ndfs) in the shrubby medic (Medicago arborea) and saltbush (Atriplex halimus) grown in pots either solely or in a mixture on a salt-affected soil, using 15N dilution method. The combined DM of both species was considerably higher than that of solely grown shrubs. The inclusion of saltbush in the mixed cropping system decreased Ndfs by shrubby medic and enhanced % Ndfa without affecting amounts of N2 fixed. It can be concluded that the use of mixed cropping system of shrubby medic and saltbush could be a promising bio-saline agricultural approach to utilize salt affected soils in terms of forage yield and N2 fixation.  相似文献

Abstract

The N 6-cyclopentyladenosine (CPA) analogue (4) was synthesized in 10 steps starting from glucose. The results of the radioligand binding assays are consistent with the thus far published findings that compounds containing a six-membered moiety at N 9 exhibit extremely weak affinity for adenosine receptors. Replacement of the ribofuranosyl moiety of CPA (2) by a 2-deoxy-D-altrohexitol moiety is sufficient to completely abolish its agonist activity.  相似文献

Five new derivatives of adenosine, N6-[(1-methylethyl)thiomethyl]-(1), N6-methyithiomethyl-(2), N6-phenylthiomethyl-(3), N6-[(3-amino-3-carboxypropyl)thiomethyl]-(4), and N6-[(2-amino-2-carboxyethyl)thiomethyl]adenosine (5), were synthesized and their cytokinin activity was tested in the Amaranthus betacyanin assay and the soybean callus growth.

1, 2, and 3 were active in the former assay and all five compounds were active in the latter assay. The activities of the compounds were, however, weaker than those of the reference derivatives, in which Sulfides were replaced by methylenes, N6-isopentyl-, N6-n-propyl-, N6-benzyl-, and N6-(5-amino-5-carboxypentyl)adenosine. This fact indicates that the sulfide structure introduced into the N6-side chains had the effect of reducing cytokinin activity.  相似文献

1. l-Aspartic acid labeled with N15 was fed to one human adult and six infants, and the total N and N15 were determined in the urine from time to time. 2. The N15 concentration (or isotopic ratio) of urinary N reached its maximum in the adult about 2 hours and in the infants about 4 hours after feeding, then fell off logarithmically. 3. Assuming that the N of aspartic acid readily entered into equilibrium with other N compounds in the pool, the rate of turnover of the N pool was calculated from the rate of fall of the isotopic ratio of urinary N. This rate of turnover of N was about 4 per cent per hour in the adult and 6 to 12 per cent per hour in the infants. 4. The rate of protein synthesis calculated from the rate of turnover of N was 10 mg. N per kilo per hour in the adult and 18 to 27 mg. N per kilo per hour in the infants, with one exception which showed a higher rate of 52 mg. N per kilo per hour. The size of the metabolic pool of N per kilo in non-growing infants was about the same as that in the adult (0.4 to 0.5 gm.) but it was somewhat larger in growing infants (0.5 to 0.8 gm.).  相似文献

IntroductionWhether parenteral nutrition benefits growth of very low birth weight (VLBW) preterm infants in the setting of rapid enteral feeding advancement is unclear. Our aim was to examine this issue using data from Japan, where enteral feeding typically advances at a rapid rate.MethodsWe studied 4005 hospitalized VLBW, very preterm (23–32 weeks'' gestation) infants who reached full enteral feeding (100 ml/kg/day) by day 14, from 75 institutions in the Neonatal Research Network Japan (2003–2007). Main outcomes were weight gain, head growth, and extra-uterine growth restriction (EUGR, measurement <10th percentile for postmenstrual age) at discharge.Results40% of infants received parenteral nutrition. Adjusting for maternal, infant, and institutional characteristics, infants who received parenteral nutrition had greater weight gain [0.09 standard deviation (SD), 95% CI: 0.02, 0.16] and head growth (0.16 SD, 95% CI: 0.05, 0.28); lower odds of EUGR by head circumference (OR 0.66, 95% CI: 0.49, 0.88). No statistically significant difference was seen in the proportion of infants with EUGR at discharge. SGA infants and infants who took more than a week until full feeding had larger estimates.DiscussionEven in infants who are able to establish enteral nutrition within 2 weeks, deprivation of parenteral nutrition in the first weeks of life could lead to under nutrition, but infants who reached full feeding within one week benefit least. It is important to predict which infants are likely or not likely to advance on enteral feedings within a week and balance enteral and parenteral nutrition for these infants.  相似文献

BackgroundThere are no data on the fatty acid (FA) compositions of preterm and term milks for sub-Saharan African populations with advancing lactation. However, it is generally acknowledged that our ancestors evolved in sub-Saharan East-Africa, where they inhabited the land-water ecosystems.MethodsWe compared the FA-compositions of preterm (28–36 weeks) and term (37–42) colostrum (2–5 day), transitional (6–15) and mature (16–56) milks in rural African women with stable dietary habits and lifelong high freshwater fish intakes.ResultsFrom colostrum to mature milk: the median docosahexaenoic acid (DHA) content decreased from 1.11 to 0.75; and arachidonic acid (AA) from 0.93 to 0.69 g% in preterm milk. In term milk, DHA decreased from 0.81 to 0.53 and AA from 1.08 to 0.55 g%. Medium-chain saturated-FA (MCSAFA) increased from 16.9 to 33.7, and 7.92–29.0 g%, while mono-unsaturated FA (MUFA) decreased from 32.5 to 22.6, and 40.0–26.5 g%, in preterm and term milk, respectively. Consistent with the literature, preterm colostrum contained higher DHA and MCSAFA, and lower MUFA compared to term colostrum. These differences vanished rapidly with advancing lactation. MUFA and MCSAFA were inversely related.ConclusionsThe presently found DHA in preterm colostrum and mature milks and AA in premature mature milk proved the highest reported in the literature so far, as derived from analysis with capillary GC-columns. We confirmed the much higher MCSAFA and lower MUFA contents in milk of rural African, compared to Westernized women. The milk FA composition of this traditional population might show us the FA composition on which our species evolved and consequently to which our genome has become adapted to optimally support (infant) health.  相似文献

In a field experiment using microplots, a flooded Crowley silt loam (Typic Albaqualfs) rice soil was fertilized with 15N labelled (60–74 atom %) urea and KNO3. Emission of N2, N2O and CH4 and accumulation in soil were measured for 21 d after fertilizer application.Emission of 15N2-N measured from the urea and KNO3 treated plots ranged from <15 to 570 and from 330 to 3,420 g ha–1 d–1, respectively. Entrapped 15N2-N in the urea treated microplots was significantly lower (<15 g to 2.1 kg ha–1) on all sampling dates compared to the 15N2-N gas accumulation in the KNO3 treated plots (6.4 to 31.5 kg ha–1). Emissions of N2O-N were low and did not exceed 4 g ha–1 d–1. Fluxes of CH4 from the fertilizer and control plots were low and never exceeded 33 g ha–1 d–1. Maximum accumulation of CH4 in the flooded soil measured 460 and 195 g ha–1 for the urea and KNO3 treatments, respectively.  相似文献

Net productions of permanent soil atmosphere gases (N2, CO2, O2) and temporary gases (N2O, NO) were monitored in soil cores using a non-interfering, fully automated measuring technique allowing highly time resolved measurements over prolonged periods. The influence of changes in available organic carbon on CO2, N2O, NO and N2 production was studied by changing the soil carbon content through aerobic preincubations of different length, up to 21 days.The aerobic preincubation caused an increase in NO3 - concentration and a decrease in available carbon content. Available carbon content dominated both CO2 and total N gas (N2+N2O+NO) production during anaerobiosis. Both CO2 and total N gas production rates decreased with increasing length of the previous aerobic preincubation, this in spite of the higher initial NO3 - concentration.Total denitrification rates were closely related to the anaerobic CO2 production rates. No relation was found between water soluble carbon content and total denitrification. The N2O/N2 ratio could be explained by an interaction of carbon availability, NO3 - concentration and enzyme status. Net N2O consumption was monitored. The balance between cumulative total N gas production and NO3 - consumption varied according to the different treatments. Cumulative N2O production exceeded cumulative N2 production for 0 up to 5 days.  相似文献

Summary The 15N/14N ratios of plant and soil samples from Northern California ecosystems were determined by mass spectrometry. The 15N abundance of 176 plant foliar samples averaged 0.0008 atom % 15N excess relative to atmospheric N2 and ranged from-0.0028 to 0.0064 atom % 15N excess relative to atmospheric N2. Foliage from reported N2-fixing species had significantly lower mean 15N abundance (relative to atmospheric N2 and total soil N) and significantly higher N concentration (% N dry wt.) than did presumed non-N2-fixing plants growing on the same sites. The mean difference between N2-fixing species and other plants was 0.0007 atom % 15N. N2-fixing species had lower 15N abundance than the other plants on most sites examined despite large differences between sites in vegetation, soil, and climate. The mean 15N abundance of N2-fixing plants varied little between sites and was close to that of atmospheric N2. The 15N abundance of presumed non-N2-fixing species was highest at coastal sites and may reflect an input of marine spray N having relatively high 15N abundance. The 15N abundance of N2-fixing species was not related to growth form but was for other plants. Annual herbaceous plants had highest 15N abundance followed in decreasing order by perennial herbs, shrubs, and trees. Several terrestrial ferns (Pteridaceae) had 15N abundances comparable to N2-fixing legumes suggesting N2-fixation by these ferns. On sites where the 15N abundance of soil N differs from that of the atmosphere, N2-fixing plants can be identified by the natural 15N abundance of their foliage. This approach can be useful in detecting and perhaps measuring N2-fixation on sites where direct recovery of nodules is not possible.  相似文献

For preterm infants, it is common practice to add human milk fortifiers to native breast milk to enhance protein and calorie supply because the growth rates and nutritional requirements of preterm infants are considerably higher than those of term infants. However, macronutrient intake may still be inadequate because the composition of native breast milk has individual inter- and intra-sample variation. Target fortification (TFO) of breast milk is a new nutritional regime aiming to reduce such variations by individually measuring and adding deficient macronutrients. Added TFO components contribute to the final osmolality of milk feeds. It is important to predict the final osmolality of TFO breast milk to ensure current osmolality recommendations are followed to minimize feeding intolerance and necrotizing enterocolitis. This study aims to develop and validate equations to predict the osmolality of TFO milk batches. To establish prediction models, the osmolalities of either native or supplemented breast milk with known amounts of fat, protein, and carbohydrates were analyzed. To validate prediction models, the osmolalities of each macronutrient and combinations of macronutrients were measured in an independent sample set. Additionally, osmolality was measured in TFO milk samples obtained from a previous clinical study and compared with predicted osmolality using the prediction equations. Following the addition of 1 g of carbohydrates (glucose polymer), 1 g of hydrolyzed protein, or 1 g of whey protein per 100 mL breast milk, the average increase in osmolality was 20, 38, and 4 mOsm/kg respectively. Adding fat decreased osmolality only marginally due to dilution effect. Measured and predicted osmolality of combinations of macronutrients as well as single macronutrient (R2 = 0.93) were highly correlated. Using clinical data (n = 696), the average difference between the measured and predicted osmolality was 3 ± 11 mOsm/kg and was not statistically significant. In conclusion, the prediction model can be utilized to estimate osmolality values after fortification.  相似文献
