Abstract

(E) 5-Alkenyl 2′-deoxyuridines were synthesized with moderate to high yields by the palladium catalyzed coupling of alkenyl-zirconium reagents with 0–3′, 5′-his (trimethylsilyl) deoxyuridine in T H F. Some of these 5-alkenyl-dUrd analogues, i.e. the 1-decenyl (5g) and 2- (1-hydroxycyclopentyl) ethenyl (5f) derivatives, inhibited murine L1210 cell growth at a concentration of about 4 μg/ml, whereas the 5-chloro-1-pentenyl (5c), 5-cyano-1-pentenyl (5d), 5-hexyn-1-enyl (5e) and 2-(1-hydroxycyclopentyl) ethenyl (5f) were inhibitory towards herpes simplex and vaccinia virus within the concentration range of 2–60 μg/ml. However, none of the newly synthesized 5-alkenyl-dUrd analogues proved selective in its antiviral action.  相似文献

The dimeric, trimeric and tetrameric 2′-5′-adenylate phosphothioates were synthesized via the phosphoramidite method using the p-nitrophenylethyl (NPE) group for phosphate protection and followed by sulfur oxidation. Oligodeoxyribonucleotides containing 2′-amino-2′-deoxy-uridine (dU) were synthesized and their ability to form duplexes with complementary DNA or RNA oligonucleotides was studied. Substitution of dU with dU in these oligomers results in lowered Tms of the duplexes.  相似文献

Abstract

The 2′-O-methylisocytidine phosphoramidite synthon 7 and methylphosphonamidite synthon 8 are synthesized from 2′-O-methyluridine. The N2 -(N′, N′-dimethylformamidine) protected 2′-O-methylisocytidine is stable to basic deamination and acidic depyrimidination. Synthon 7 and synthon 8 have been incorporated into oligomers via the automated solid state procedure.

When adenosine (Ia) was allowed to react with 2-methylthio-4H-l,3,2-benzodioxaphosphorin-2-oxide (MTBO) (II), a new phosphorylating agent, in the presence of cyclohexylamine, adenosine 5′-S-methyl phosphorothiolate (IVa) and adenosine 2′: 3′-cyclic phosphate (Va) were obtained.

Ribonucleoside 5′-S-methyl phosphorothiolates (IV) were selectively synthesized directly from borate complexes of ribonucleosides (VI) and MTBO in the presence of cyclohexylamine, followed by removal of metaboric acid by co-distillation with methanol, in 68~86% yields.

Under anhydrous conditions, the phosphorothiolates (IV) were cyclized to give ribonucleoside 3′: 5′-cyclic phosphates (X) by iodine oxidation. Five 3′: 5′-cyclic nucleotides including cyclic AMP were synthesized by this procedure in 38~64% yields after purification.  相似文献

Abstract

The 5′-O-(4,4′-dimethoxytrityl) and 5′-O-(tert-butyldimethylsilyl) derivatives of 2′-,3′-O-thiocarbonyl-6-azauridine and 2′,3′-O-thiocarbonyl-5-chlorouridine were synthesized from the parent nucleosides by reaction with 4, 4′-dimethoxytrityl chloride and tert-butyldimethylsilyl chloride, respectively, followed by treatment with 1,1′-thiocarbonyldiimidazole. Introduction of a 2′-,3′-double bond into the sugar ring by reaction of the 5′-protected 2′-,3′-O-thionocarbonates with 1, 3-dimethyl-2-phenyl-1, 3, 2-diazaphospholidiine was unsuccessful, but could be accomplished satisfactorily with trimethyl phosphite. Reactions were generally more successful with the 5′-silylated than with the 5′-tritylated nucleosides. Formation of 2′-,3′-O-thiocarbonyl derivatives proceeded in higher yield with 5′-protected 6-azauridines than with the corresponding 5-chlorouridines because of the propensity of the latter to form 2,2′-anhydro derivatives. In the reaction of 5′-O-(tert-butyldimethylsilyl)-2′-,3′-O-thiocarbonyl-6-azauridine with trimethyl phosphite, introduction of the double bond was accompanied by N3-methylation. However this side reaction was not a problem with 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-O-thioarbonyl-5-chlorouridine. Treatment of 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-didehydro-2′-,3′-dideoxy-6-azauridine with tetrabutylammonium fluoride followed by hydrogenation afforded 2′-,3′-dideoxy-6-azauridine. Deprotection of 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-didehydro-2′-,3′-dideoxy-5-chlorouridine yielded 2′-,3′-didehydro-2′-,3′-dide-oxy-5-chlorouridine.  相似文献

A series of novel nucleosides bearing a 1,2,3-triazole moiety at the 2′-position of the sugar moiety has been synthesized starting from 2′-azidouridine and using the copper (I)-catalyzed Huisgen–Sharpless–Meldal 1,3-dipolar cycloaddition reaction. The reactions proceeded in overall yield of 52–82% and gave almost exclusively the 1,4-disubstituted 1,2,3-triazoles. The 2′-azidouridine was synthesized from uridine in two steps, and reacted with a variety of differently substituted alkynes to give the desired 2′-triazole-substituted uridine derivatives.  相似文献

The synthesis of 2′-amino-2′-deoxypyrimidine 5′-triphosphates is described. The 2′-amino-2′-deoxyuridine 5′-triphosphate is obtained from uridine in four steps with 25% overall yield. The 2′-amino-2′-deoxycytidine 5′-triphosphate is obtained from uridine in seven steps with 13% overall yield.  相似文献

2′-Deoxy-5-methylisocytidine is widely used in assays to personalize the care of patients infected with HIV, hepatitis C, and other infectious agents. However, oligonucleotides that incorporate 2′-deoxy-5-methylisocytidine are expensive, because of its intrinsic chemical instability. We report here a C-glycoside analog that is more stable and, in oligonucleotides, pairs with 2′-deoxyisoguanosine, contributing to duplex stability about as much as a standard 2′-deoxycytidine and 2′-deoxyguanosine pair.  相似文献

A sensitive and simple fluorometric assay has been developed for detection of pyridoxamine (pyridoxine) 5′-phosphate oxidase. This technique utilizes fluorescent N-(5′-phospho-4′-pyridoxyl)amines as substrates that, upon incubation with the oxidase, release the free fluorescent amine. The substrates were prepared by condensation of pyridoxal 5′-phosphate with fluorescent amines and subsequent hydrogenation of the Schiff bases. Since N-(1-naphthyl)ethylenediamine is 15 times less fluorescent in the intramolecularly quenched substrate than the product amine, the direct increase of fluorescence, as well as selective extraction of more fluorescent product, can be utilized for assay. The apparent Km value for this substrate is 8 μm, which is slightly less than that of pyridoxamine 5′-phosphate; V is larger than the natural substrate value. The greater sensitivity gained by this fluorimetric method allows detection of the oxidase in smaller quantities than can be determined by the conventional colorimetric assay.  相似文献

In this article, we describe the synthesis of 5-nitro-1-(2-deoxy-α-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1-(2-deoxy-β-D-erythro-pentofuranosyl)cytosine (), 5-amino-1-(2-deoxy-α-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1- (2-deoxy-β-D-erythro-pentofuranosyl)cytosine (), 5-nitro-1-(2,3-dideoxy-β- D-ribofuranosyl)uracil (), 5-amino-1-(2,3-dideoxy-α,β-D-ribofuranosyl)uracil (7), 5-nitro-1-(2,3-dideoxy-α,β-D-ribofuranosyl)cytosine (8) and 5-amino-1-(2,3-dideoxy-β-D-ribofuranosyl)cytosine (). The prepared compounds were tested for their activity against HIV and HBV viruses, but they did not show significant activity.  相似文献

Abstract

The diphosphates of a series of 2′-O-allyl-1-β-D-arabinofuranosyl derivatives, previously obtained by us, have been prepared and tested for their inhibitory activity in an in vitro assay using R1 and R2 subunits of the purified recombinant mouse ribonucleotide reductase (RNR). 2′-O-Allyl-araU diphosphate proved to be inhibitory, with an IC50 of 100 μM. The 5′-phosphoramidate pronucleotide of 2′-O-allyl-araU was also prepared and tested for inhibition of tumor cell proliferation.  相似文献

Abstract

The non-exchangeable 1H-NMR signals of the branch core trinucleotide of the lariat branch site (A2′p5′G 3′p5′C), 1) and its derivatives 2 and 3 are completely assigned using one- and two- dimensional NMR techniques including NOE, COSY, NOESY, 1H-1HINADEQUATE and 2D-J-resolved spectroscopy. From the vicinal coupling constants in the individual ribose rings, NOE data and T1 measurements, the following properties of the trimers are deduced.(i)The unique stacking behavior of the trimers is S1′N 3′N, and the sugar rings exist predominantly in the N-conformation (3′-endo-2′-exo).(ii)The sugar-base orientations appear to be anti.(iii) The branched trimers exist in solution as single-stranded right-handed conformations resembling A-RNA with stacking between the adenine and guanine residues in aqueous solution at 21°C and pH 7.2.(iv) The calculated values for the torsion angles εt andγ+ for the trimers are 201–203° and 71–86%, respectively, while the percent β1 values are higher for the guanine (87–92%) than the cytosine residues (73–77%). The computer generated depiction of the triribonucleotide 1 is also shown. These subtle structural features may act as recognition signals for this critical lariat branch site which is essential for the second step in yeast mRNA splicing.  相似文献

Facile synthetic methods of 2′,5′-dideoxy-, 2′,3′-dideoxy- and 3′-deoxy-1,N 6-ethenoadenosine nucleosides by either an enzymatic dideoxyribosyl transfer reaction or a simple chemical reaction were proposed. The synthetic products were isolated and purified by preparative HPLC and their structures were confirmed by1H NMR (500 MHz) and FAB-MS including high resolution mass measurement. These modified nucleoside analogs have not been reported yet. Therefore, these modified nucleoside analogs are of potential value to be studied further for biological activity such as anticancer or antiviral.  相似文献
