Summary Immunofluorescence and TEM studies of meiosis in two mosses (Bryophyta) provide evidence that the prophasic tetrahedral system of microtubules contributes directly to the metaphase I spindle. Intense staining of tubulin, conspicuously absent around the nuclear envelope, is first seen associated with plastids. By mid-prophase, microtubules radiate from the plastids to the nuclear envelope and become organized into six bands that interconnect the four plastids, forming a tetrahedral cytoskeleton surrounding the nucleus. During transition of prophase to metaphase, the four poles of the tetrahedral microtubule system converge in pairs toward opposite cleavage furrows. Opposite furrows occupy mutually perpendicular planes and the pair of microtubule focal points straddling one furrow lies at right angles to the pair straddling the opposite furrow. Additional microtubules terminate in numerous small clusters in the concave polar regions arching over the cleavage furrows. By early anaphase, the microtubule focal points lie very close to the division axis. We conclude that microtubules recruited from the prophasic quadripolar system are incorporated into the mature metaphase I spindle and the two principal focal points at each pole are those derived from poles of the prophasic quadripolar system.  相似文献

A large increase in surface area must accompany formation of the amphibian embryo first cleavage furrow. The additional membrane for this areal expansion has been thought to be provided entirely from cytoplasmic stores during furrowing. We have radioiodinated surface proteins of fertilized, precleavage Xenopus laevis embryos and followed their redistribution during first cleavage by autoradiography. Near the end of first cleavage, membrane of the outer, pigmented surface of the embryo and a short band of membrane at the leading edge of the furrow displayed a high silver grain density, but the remainder of the furrow membrane was lightly labeled. The membrane of the cleavage furrow is thus mosaic in character; the membrane at the leading edge originates in part from the surface of the zygote, but most of the membrane lining the furrow walls is derived from a source inaccessible to surface radioiodination. The furrow membrane adjacent to the outer, pigmented surface consistently showed a very low silver grain density and was underlain by large membranous vesicles, suggesting that new membrane derived from cytoplasmic precursors is inserted primarily in this location, at least during the later phase of cleavage. Radioiodinated membrane proteins and surface-attached carbon particles, which lie in the path of the future furrow, contract toward the animal pole in the initial stages of cleavage while markers in other regions do not. We suggest that the domain of heavily labeled membrane at the leading edge of the definitive furrow contains the labeled elements that are gathered at the animal pole during the initial surface contraction and that they include membrane anchors for the underlying contractile ring of microfilaments.  相似文献

In the area between the equatorial and the upper vegetal regions of the egg of the newt, Cynops (Triturus) phrrhogaster, strips of cellophane, ordinary filter paper or Millipore filter respectively were inserted transversely at short distances ahead of the advancing cleavage furrow. When a cellophane strip is inserted across the future path of the furrow at 0.5–0.7 mm beyond the furrow tip, the furrow stops on reaching the position of the inserted cellophane strip. If, however, a strip of the Millipore filter is inserted at the same distances ahead of the cleavage furrow, the furrow reaches the one side of the Millipore filter strip and soon appears on the opposite side of it, just as the furrow jumps across the strip, to continue the previous course. Similar tendency is observed in the experiments on insertion of strips of the ordinary filter paper. It is suggested that certain propagating factors which are necessary to induce formation of the cleavage furrow can go through the pores of the Millipore filter to precede the advancing tip of the cleavage furrow.  相似文献

The fact that substrate-anchored Dictyostelium cells undergo cytokinesis in the absence of myosin II underscores the importance of other proteins in enabling the cleavage furrow to constrict. Cortexillins, a pair of actin-bundling proteins, are required for normal cleavage. They are targeted to the incipient furrow in wild-type and, more prominently, in myosin II-null cells. No other F-actin bundling or cross-linking protein tested is co-localized. Green fluorescent protein fusions show that the N-terminal actin-binding domain of cortexillin I is dispensable and the C-terminal region is sufficient for translocation to the furrow and the rescue of cytokinesis. Cortexillins are suggested to have a targeting signal for coupling to a myosin II-independent system that directs transport of membrane proteins to the cleavage furrow.  相似文献

棉铃虫Helicoverpa armigera Hubner雌蛾的性外激素分泌腺是一个完整的上皮环,位于第八、九腹节之间。羽化2天雌蛾的腺体细胞方形,比未分化的上皮细胞大。腺体折皱,表面密布小毛,小毛无孔。表皮可分二层:上表皮和内表皮。上表皮致密,较薄,厚度均匀。内表皮厚度有变化,较厚,由12-14层呈螺旋状排列的几丁微丝组成,有上皮丝穿人,构成孔道。顶部细胞膜组成微绒毛,底部细胞膜有内折。细胞质内有粗面内质网,光面内质网,高尔基氏复合体、脂肪滴、糖原及线粒体等细胞器。大的细胞核位于中下部。  相似文献

Few papers have been devoted to the female pheromone glands, phylogeny and high-level systematics of the Arctiidae. The diversity of these glands is here interpreted in the light of a revised classification. The presence, in the A8-A9 intersegmental membrane, of one or two, distinctly invaginated (‘saccular’), dorsal glands is a characteristic derived trait of the Arctiidae. Lithosiinae typically have an unpaired dorsal gland, anteriorly provided with a pair of short lobes. Non-lithosiine arctiids form a monophyletic group, in which the gland lobes are primarily very long. Several lineages can be characterized by the shape of their dorsal glands, notably the Ctenuchinae (inclusive of Euchromia), the Pericopinae and the Arctiini. Pheromone production is a well-established function of these dorsal glands, while the ventral, intersegmental ‘pouches’ observed in certain taxa may not be pheromone-producing structures. The Thyretini (here allocated to Syntominae) possess ventral saccular glands that bear a marked resemblance to the dorsal pair.  相似文献

The neuronal circuits that generate swimming movements in the leech were simulated by a chain of coupled harmonic oscillators. Our model incorporates a gradient of rostrocaudally decreasing cycle periods along the oscillator chain, a finite conduction delay for coupling signals, and multiple coupling channels connecting each pair of oscillators. The interactions mediated by these channels are characterized by sinusoidal phase response curves. Investigations of this model were carried out with the aid of a digital computer and the results of a variety of manipulations were compared with data from analogous physiological experiments. The simulations reproduced many aspects of intersegmental coordination in the leech, including the findings that: 1) phase lags between adjacent ganglia are larger near the caudal than the rostral end of the leech nerve cord; 2) intersegmental phase lags increase as the number of ganglia in nervecord preparations is reduced; 3) severing one of the paired lateral connective nerves can reverse the phase lag across the lesion and 4) blocking synaptic transmission in midganglia of the ventral nerve cord reduces phase lags across the block.  相似文献

The gross anatomy of muscles, the topography of nerve tissues, and the histology of the pregenital abdominal glands of Nomia melanderi Ckll. are reported in detail. The movable and fixed points of muscle attachment were utilized in establishing a system of nomenclature for a typical abdominal segment. Names of nervules correspond to those of the tissues they innervate. The points of attachment of muscles of the fifth abdominal segment are essentially the same in both Nomia and Apis, except for the second tergo-sternal muscle which, in Nomia, has shifted its point of movable attachment to the membranous integument in front of the intersegmental membrane gland where it helps in relasing glandular secretion. The general plan of the nerves in the fifth abdominal segment in Nomia is more diffuse than in Apis, but there is no difficulty in establishing homology between the nervules of the two species. A pair of intersegmental stretch organs was found in abdominal segments 3–6. Glands of the sixth intersegmental membrane possess a reservoir with peripheral pouches both of which are absent in those of the fifth. Both types of glands have neither closing nor opening mechanisms, and neither is innervated. Release of glandular secretion is accomplished by the action of the tergo-sternal muscles.  相似文献

Non-linear optimisation, such as the type presented by R.D. Crowninshield and R.A. Brand [The prediction of forces in joint structures: Distribution of intersegmental resultants, Exercise Sports Sci. Rev. 9 (1981) 159], has been frequently used to obtain a unique set of muscle forces during human or animal movements. In the past, analytical solutions of this optimisation problem have been presented for single degree-of-freedom models, and planar models with a specific number of muscles and a defined musculoskeletal geometry. Results of these studies have been generalised to three-dimensional problems and for general formulations of the musculoskeletal geometry without corresponding proofs. Here, we extend the general solution of the above non-linear, constrained, planar optimisation problem to three-dimensional systems of arbitrary geometry. We show that there always exists a set of intersegmental moments for which the given static optimisation formulation will predict co-contraction of a pair of antagonistic muscles unless they are exact antagonists. Furthermore, we provide, for a given three-dimensional system consisting of single joint muscles, a method that describes all the possible joint moments that give co-contraction for a given pair of antagonistic muscles.  相似文献

The ultrastructure of the stigma and associated flagellar-microtubular systems in Dinobryon cylindricum var. alpinum is described in detail and compared with observations on comparable photo-kinetic systems in other chrysophycean organisms. The chloroplastidic stigma of D. cylindricum var. alpinum is shown to lie in a particular positional relationship to the flagellar swelling in the anterior furrow and to several other organelles, to consist of a monolayer of c. 40 pigmented granules, each c. 250–500 nm diameter, arranged in a definite pattern, and to be overlain by several membrane systems. Other cytoplasmic pigmented bodies with dense crystalline contents surrounded by a single “unit membrane” aggregate near the anterior furrow on the side opposite the stigma. The swelling on the proximal portion of the smooth flagellum is separated from the plasmalemma of the anterior furrow by a nearly constant distance of 75–100 nm, has a multilamellate substructure that is linked by fine radiating interconnections to the axoneme doublets, and is connected to the plasmalemma by a system of fibrillar interconnections. A transitional helix in the basal body region is described as similar to structures reported in other chrysophycean flagellates. A striated rhizoplast with a periodicity of c. 90 nm extends from basal body I to the nuclear envelope. A seven-stranded microtubular root extends from the same basal body. Other fibrous and microtubular root systems are also described. The inter-relationships and possible functions of the aforementioned structures are discussed.  相似文献

The relative orientation and proximity of the pseudo-symmetrical inner transmembrane helical pairs 5/8 and 2/11 of Glut1 were analyzed by chemical cross-linking of di-cysteine mutants. Thirteen functional di-cysteine mutants were created from a C-less Glut1 reporter construct containing cysteine substitutions in helices 5 and 8 or helices 2 and 11. The mutants were expressed in Xenopus oocytes and the sensitivity of each mutant to intramolecular cross-linking by two homobifunctional thiol-specific reagents was ascertained by protease cleavage followed by immunoblot analysis. Five of 9 mutants with cysteine residues predicted to lie in close proximity to each other were susceptible to cross-linking by one or both reagents. None of 4 mutants with cysteine substitutions predicted to lie on opposite faces of their respective helices was susceptible to cross-linking. Additionally, the cross-linking of a di-cysteine pair (A70C/M420C, helices 2/11) predicted to lie near the exoplasmic face of the membrane was stimulated by ethylidene glucose, a non-transported glucose analog that preferentially binds to the exofacial substrate-binding site, suggesting that the binding of this ligand stimulates the closure of helices at the exoplasmic face of the membrane. In contrast, the cross-linking of a second di-cysteine pair (T158C/L325, helices 5/8), predicted to lie near the cytoplasmic face of the membrane, was stimulated by cytochalasin B, a glucose transport inhibitor that competitively inhibits substrate efflux, suggesting that this compound recruits the transporter to a conformational state in which closure of inner helices occurs at the cytoplasmic face of the membrane. This observation provides a structural explanation for the competitive inhibition of substrate efflux by cytochalasin B. These data indicate that the binding of competitive inhibitors of glucose efflux or influx induce occluded states in the transporter in which substrate is excluded from the exofacial or endofacial binding site.  相似文献

As reported by many workers over the past two centuries, the inner part of the shell of various straight and coiled Palaeozoic to tertiary nautiloid taxa bears a continuous mid-ventral furrow that extends into the phragmocone and the body chamber nearly to the aperture. Study of the early embryonic shell development of Jurassic Nautilida shows that the most apical part of this so-called conchal furrow originates from the inner part of the initial, calcified shell apex, in line with the inner ventral termination of the central linear depression of the cicatrix, the initial site of shell deposition. The conchal furrow corresponds to a morphological feature arising as a developmental by-product. Rare specimens of scattered ammonoid species (and possibly of bactritoids) display a similar feature, whereas their protoconch lacks a cicatrix. However, the protoconch of recent cuttlefish, Sepia officinalis, often displays a longitudinal fold of the primary shell epithelium. A longitudinal groove or a pair of grooves appears connected with this cicatrix-like structure. Although the mid-ventral ridge in ammonoids must probably be viewed as an incidental 'fabricational noise', whether or not it originates from a so far undocumented optional ridge on the protoconch or from some other structure related to shell development remains an open question.  相似文献

A morphofunctional investigation of the different neuronal subpopulations projecting through each of the nerves IV–VI emerging bilaterally from the terminal abdominal ganglion (TAG) was correlated with the octopaminergic activity in the ganglion that controls the ovipositor movements associated with calling behavior in the female gypsy moth Lymantria dispar. Tetramethylrodamine‐dextran backfills from nerve stumps resulted in a relatively low number of TAG projections, ranging from 12 to 13 for nerve pair IV, 12 to 14 for nerve pair V, and 8 to 9 for nerve pair VI. Furthermore, as assessed by electrophysiological recordings, a number of fibers within each of these nerves displays spontaneous tonic activity, also when the ganglion is fully disconnected from the ventral nerve cord (VNC). Octopamine (OA) applications to the TAG strongly enhanced the activity of these nerves, either by increasing the firing rate of a number of spontaneously firing units or by recruiting new ones. This octopaminergic activity affected calling behavior, and specifically the muscle activity leading to cycling extensions of the intersegmental membrane (IM) between segments VIII and IX (ovipositor). Our results indicate that in the female gypsy moth the octopaminergic neural activity of the TAG is coupled with extensions and retractions of IM for the purpose of releasing pheromone, where motor units innervated by nerve pair IV appear antagonistic with respect to those innervated by nerve pair V.  相似文献

Cell lineages during development of ascidian embryos were analyzed by injection of horseradish peroxidase as a tracer enzyme into identified cells at the one-, two-, four-, and eight-cell stages of the ascidians, Halocynthia roretzi, Ciona intestinalis, and Ascidia ahodori. Identical results were obtained with eggs of the three different species examined. The first cleavage furrow coincided with the bilateral symmetry plane of the embryo. The second furrow did not always divide the embryo into anterior and posterior halves as each of the anterior and posterior cell pairs gave rise to different tissues according to their destinies, which became more definitive in the cell pairs at the eight-cell stage. Of the blastomeres constituting the eight-cell stage embryo, the a4.2 pair (the anterior animal blastomeres) differentiated into epidermis, brain, and presumably sense organ and palps. Every descendant cell of the b4.2 pair (the posterior animal blastomeres) has been thought to become epidermis; however, the horseradish peroxidase injection probe revealed that the b4.2 pair gave rise to not only epidermis but also muscle cells at the caudal tip region of the developing tailbud-stage embryos. The A4.1 pair (the anterior vegetal blastomeres) developed into endoderm, notochord, brain stem, spinal cord, and also muscle cells next the caudal tip muscle cells. From the B4.1 pair (the posterior vegetal blastomeres) originated muscle cells of the anterior and middle parts of the tail, mesenchyme, endoderm, endodermal strand, and also notochord at the caudal tip region. These results clearly demonstrate that muscle cells are derived not only from the B4.1 pair, as has hitherto been believed, but also from both the b4.2 and A4.1 pairs.  相似文献

Early authors regarded Pycnogonida (sea spiders) either as aquatic arachnids, ‘degraded’ crustaceans or as some sort of intermediate form between the two. Subsequently, pycnogonids were either placed among the Chelicerata or considered as an isolated group, unrelated to other arthropods. The latter model is untenable under phylogenetic systematics and recent cladistic studies have supported one of two alternative hypotheses. The first is the traditional Chelicerata s.lat. concept, i.e. (Pycnogonida + Euchelicerata). This, however, has only one really convincing synapomorphy: chelate chelicerae. The second hypothesis recognizes (Pycnogonida + all other Euarthropoda) and has been recovered in various ‘total evidence’ studies. Morphologically some characters – the presence of gonopores on the trunk and absence of a labrum, nephridia and intersegmental tendons – support Cormogonida (Euarthropoda excluding pycnogonids). Advances in developmental biology have proposed clear interpretations of segmentation homologies. However, so far there is also a confrontation of the two hypotheses depending on whether the last walking leg segment is considered part of the prosoma. In this case pycnogonids have too many prosomal segments compared with Euchelicerata; perhaps implying they are not sister groups. Alternatively, if part of the postprosomal region, the last leg pair could correspond to the chilarial segment in euchelicerates and its uniramous state could be apomorphic with respect to other euarthropods. Molecular phylogenies need to be more rigorously analysed, better supported by data from different sources and technique‐sensitive aspects need to be explored. Chelicerata s.lat. may emerge as the more convincing model, yet even the putative autapomorphy of chelicerae needs to be treated with caution as there are fossil ‘great appendage’ arthropods in the early Palaeozoic which also have a robust, food‐gathering, pair of head limbs and which may lie on the chelicerate, or even the euarthropod, stem lineage.  相似文献

It is distinguished from the known genera of Alluroididae by having testes in XI, male pores in XIV, and one pair of spermathecae opening laterally at 6/7. Ferritin concentrates, stores, and detoxifies iron in most organisms. The iron is a solid, ferric oxide mineral (< or =4500 Fe) inside the protein shell. Eight pores are formed by subunit trimers of the 24 subunit protein. A role for the protein in controlling reduction and dissolution of the iron mineral was suggested in preliminary experiments [Takagi et al. (1998) J. Biol. Chem. 273, 18685-18688] with a proline/leucine substitution near the pore. Localized pore disorder in frog L134P crystals coincided with enhanced iron exit, triggered by reduction. In this report, nine additional substitutions of conserved amino acids near L134 were studied for effects on iron release. Alterations of a conserved hydrophobic pair, a conserved ion pair, and a loop at the ferritin pores all increased iron exit (3-30-fold). Protein assembly was unchanged, except for a slight decrease in volume (measured by gel filtration); ferroxidase activity was still in the millisecond range, but a small decrease indicates slight alteration of the channel from the pore to the oxidation site. The sensitivity of reductive iron exit rates to changes in conserved residues near the ferritin pores, associated with localized unfolding, suggests that the structure around the ferritin pores is a target for regulated protein unfolding and iron release.  相似文献

Pseudendoclonium basiliense and Trichosarcina polymorpha are essentially identical with regard to the fine structural details of cell division even though one was previously classified in the Chaetophorales and the other in the Ulvales. Cell division in the 2 genera is also shown to be like that in Ulva, as previously suggested might be the case. The combination of mitotic and cytokinetic characteristics common to the 3 genera is distinctive: (1) precocious development of a thick cleavage furrow, (2) centrioles distinctly lateral to polar fenestrae, (3) collapse of the interzonal spindle at telophase, and. (4) a cleavage furrow not associated with microtubules. It is suggested that features of vegetative cell division presently provide the best, characteristics for defining the Ulvaceae and that the use of growth habit should be abandoned. Despite the fact that a phycoplast is not present, in these algae, it is concluded that their affinities lie with genera that do possess a phycoplast.  相似文献
