在研究四川省玉米纹枯病菌系时,获得23个RhizoctonizeaeVoorhees菌株,该菌菌丝形态与P.solaniKuhn相似;菌丝细胞含3个以上椭圆形细胞核,隔膜具桶孔,桶孔周围的隔膜肿体(septalswelling)与隔膜板垂直,两端不再膨大外延;菌丝生长较快,在PDA平板26℃下培养,每24小时菌落直径增长33.2~39.1mm;菌丝与苯酚相互作用后,着色较深,呈深褐色,菌核初期为白  相似文献

将来自云南省20多个县市的水稻纹枯病标样130多份,选代表性的标样分离得到54个菌株。按菌丝融合测定法,将54个菌株分为5个菌系:R.solani的AG-1 IA,AG-1 IC,AG-6GW以及以核丝核菌的AG-Bb,AG- ⅠⅡ。经致病性测定表明,该菌系对水稻、玉米、小麦的苗期及成株期的致病性有显著差异。其中AG-1ⅠA,AG-1 IC,AG-Ⅱ的致病力最,AG-6GW的最弱。对这些菌系的酯酶同工酶进行比较研究发现,不同菌系间均存在明显差异,而同菌系不同菌株间却具有一致性。由此说明,按菌丝融合与否区分丝核菌种群较之现行的其它分类法更能反映其遗传本质和亲缘关系,。  相似文献

绿僵菌对小麦纹枯病菌的抑制作用研究   总被引:1,自引:0,他引:1  
在实验室条件下,研究了金龟子绿僵菌(Metarhizium anisopliae)对小麦纹枯病菌(Rhizoctonia cerealis)的拮抗作用及其机理。结果表明,金龟子绿僵菌与小麦纹枯病菌对峙培养以及在培养基中加入金龟子绿僵菌孢子悬浮液,对小麦纹枯病菌菌丝生长均有较好的抑制作用。测定了培养不同天数的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌菌丝生长、菌核产生量及菌核萌发率的影响。结果表明,液体振荡培养25 d的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌的菌丝生长、菌核产生量及菌核萌发具有显著的抑制作用,且Ma55发酵液中的抑菌活性物质具有较好的热稳定性。在光学显微镜下,未观察到Ma55对小麦纹枯病菌的重寄生现象,但发现金龟子绿僵菌与小麦纹枯病菌对峙培养处小麦纹枯病菌营养菌丝的细胞质变稀薄、菌丝部分消解或断裂。上述结果显示,金龟子绿僵菌对小麦纹枯病菌的拮抗机制主要是营养竞争、空间竞争及抗生作用等。  相似文献

[目的]筛选高效拮抗向日葵菌核菌的细菌菌株,为开发防治菌核菌病害、提高向日葵产量的生物菌剂提供菌种资源。[方法]以羧甲基纤维素钠(CMC)、小麦秸秆纤维素为唯一碳源的无机盐培养基,分离高效降解纤维素的细菌菌株;采用纤维素降解菌与菌核菌的平板对峙方法,进一步筛选拮抗菌核菌的菌株;利用16S rDNA序列鉴定菌株、PDYA平板对峙实验检验上述所选拮抗菌株的抑菌谱;采用离体向日葵新鲜叶片、草炭土基质盆栽实验,观察拮抗菌菌株抑制菌核菌生长的能力;温室盆栽和田间试验条件下,研究其防治向日葵菌核菌病害、促进生长和提高产量的效果。[结果]筛选了一株高效抑制菌核菌的细菌YC16,经过16S rDNA序列分析,鉴定为解淀粉芽孢杆菌。YC16菌株能够抑制8种病原真菌生长,包括齐整小核菌、腐皮镰孢菌、尖孢镰刀菌、稻梨孢、辣椒疫霉、镰刀菌、尖镰孢黄瓜专化型和向日葵菌核菌;抑制菌核菌感染叶片,抑制率达到了80.42%;抑制盆栽基质中菌核菌的菌丝生长,基质表面菌丝密度比对照减少了50%以上。盆栽接种YC16的向日葵生物量比对照提高54.9%,田间向日葵接种YC16菌剂对菌核菌引发的盘腐病防治效果达39%-100%,产量提高24.4%-30.2%。[结论]YC16生物菌剂施用于土壤,能够有效防治向日葵的茎腐病和盘腐病,展现了防治向日葵菌核病和提高产量的双重效果,是一株具有良好应用前景的高效菌种资源。  相似文献

杨梅根瘤的横切面是圆形、对称的,含菌组织位于皮层的中部。未成熟根瘤的感染细胞充满内生菌丝。内生菌丝是分枝。具隔膜的Frankia菌。成熟根瘤中,内生菌丝顶端膨大形成无隔膜、表面光滑、球形的泡囊,直径比菌丝直径大,内含有某种内含物。泡囊的大量出现可能是固氮活力提高的标志,成熟壮瘤的固氮活性比幼瘤和衰老瘤的高。  相似文献

将来自川东、南稻区代表性的38个县(市)的水稻纹枯病标样296份,按不同的品种、海拔、土质、前作和症状归粪,选代表性的标样分离得到108个丝核菌菌株。按HCL—Giemsa染色程序和菌丝融合测定法,将108个菌株分为3个菌系:Rhizoctonia solani AG-1和AG-4,以及双核丝核菌的AG—Bb。其比例分别为97%、1%和2%。经致病性测定表明,该各菌系对水稻的致病性有显著差异:R. solani AG-1的大多数菌株最强,双核丝核菌AG—Bb最弱,R. solani AG一4居中。 对上述各菌系的培养性状、非特异性酯酶和过氧化物酶同工酶谱进行比较研究发现,不同菌系间在上述诸方面均存在明显差异,而同菌系不同菌株间却具一致性。由此说明,按菌丝融合与否区分丝核菌种群较之现行的其它分类法更能反映其遗传本质和亲缘关系。  相似文献

Tobacco leaf spot and root rot caused by Rhizoctonia solani Kühn   总被引:1,自引:0,他引:1  
Rhizoctonia solani Kühn is a soil-borne fungal pathogen that causes disease in a wide range of plants worldwide. Strains of the fungus are traditionally grouped into genetically isolated anastomosis groups (AGs) based on hyphal anastomosis reactions. This article summarizes aspects related to the infection process, colonization of the host and molecular mechanisms employed by tobacco plants in resistance against R. solani diseases. TAXONOMY: Teleomorph: Thanatephorus cucumeris (Frank) Donk; anamorph: Rhizoctonia solani Kühn; Kingdom Fungi; Phylum Basidiomycota; Class Agaricomycetes; Order Cantharellales; Family Ceratobasidiaceae; genus Thanatephorus. IDENTIFICATION: Somatic hyphae in culture and hyphae colonizing a substrate or host are first hyaline, then buff to dark brown in colour when aging. Hyphae tend to form at right angles at branching points that are usually constricted. Cells lack clamp connections, but possess a complex dolipore septum with continuous parenthesomes and are multinucleate. Hyphae are variable in size, ranging from 3 to 17 μm in diameter. Although the fungus does not produce any conidial structure, ellipsoid to globose, barrel-shaped cells, named monilioid cells, 10-20 μm wide, can be produced in chains and can give rise to sclerotia. Sclerotia are irregularly shaped, up to 8-10 mm in diameter and light to dark brown in colour. DISEASE SYMPTOMS: Symptoms in tobacco depend on AG as well as on the tissue being colonized. Rhizoctonia solani AG-2-2 and AG-3 infect tobacco seedlings and cause damping off and stem rot. Rhizoctonia solani AG-3 causes 'sore shin' and 'target spot' in mature tobacco plants. In general, water-soaked lesions start on leaves and extend up the stem. Stem lesions vary in colour from brown to black. During late stages, diseased leaves are easily separated from the plant because of severe wilting. In seed beds, disease areas are typically in the form of circular to irregular patches of poorly growing, yellowish and/or stunted seedlings. RESISTANCE: Knowledge is scarce regarding the mechanisms associated with resistance to R. solani in tobacco. However, recent evidence suggests a complex response that involves several constitutive factors, as well as induced barriers controlled by multiple defence pathways. MANAGEMENT: This fungus can survive for many years in soil as mycelium, and also by producing sclerotia, which makes the management of the disease using conventional means very difficult. Integrated pest management has been most successful; it includes timely fungicide applications, crop rotation and attention to soil moisture levels. Recent developments in biocontrol may provide other tools to control R. solani in tobacco.  相似文献

Eighteen isolates from sugar beet roots associated with an unknown etiology were characterized based on observations of morphological characters, hyphal growth at 4-28 C, production of phenol oxidases and sequence analysis of internal transcribed spacer (ITS) and large subunit (LSU) regions of the ribosomal DNA (rDNA). The isolates did not produce asexual or sexual spores, had binucleate hyphal cells with clamp connections, grew 4-22 C with estimated optimal growth at 14.5 C and formed a dark brown pigment on potato dextrose or malt extract agar amended with 0.5% tannic acid. Color changes observed when solutions of gum guiac, guiacol and syringaldzine were applied directly to mycelium grown on these media indicated that all isolates produced phenol oxidases. Sequences of ITS and LSU regions on the rDNA gene from 15 isolates were 99.2-100% identical, and analysis of sequence data with maximum likelihood and maximum parsimony suggest that the isolates from sugar beet roots are phylogenetically related to Athelia bombacina, Granulobasidium vellereum and Cyphella digitalis. High statistical support for both loci under different criteria confirmed that Athelia bombacina was consistently the closest known relative to the sugar beet isolates. Additional taxonomic investigations are needed before species can be clarified and designated for these isolates.  相似文献

This paper presents an in vitro test to screen the pathogenicity of different Rhizoctonia solani isolates on a host range. The level of aggressivity of the different isolates was different for several host plants tested. There were significant differences between the crops and the isolates tested. In general, the disease level was higher on beans, lettuce and cabbage. In carrot and rye grass the level of infection was lower for the isolates of R. solani tested. The potato isolates of R. solani were less aggressive than the isolates coming from maize, fodder beet and sugar beet. The R. solani isolates were also biochemically characterized by pectic zymograms: the isolates Rs0401 (from maize) and Rs0504 (from sugar beet) belong both to the anastomosis group AG2-2.  相似文献

The relative susceptibility of dry beans ( Phaseolus vulgaris ), soybeans and maize to anastomosis group 4 isolates of Rhizoctonia solani was determined in greenhouse experiments. Large variations in virulence were found among 30 field isolates. This variation was not due to differential reductions in isolate virulence during axenic culture. There was considerable variation among isolates from within the same field but variability within isolates was small. Twelve of 30 isolates of R. solani were highly virulent to dry beans and soybeans, while the others were of low virulence. Soybeans were more susceptible than dry beans to both pre-emergence mortality and hypocotyl disease. No isolates were highly virulent to maize. The importance of using isolates with a high level of virulence for testing soybean cultivars for resistance to Rhizoctonia disease is stressed.  相似文献

Potato is planted after rice in several parts of Punjab in India and both crops are attacked by Rhizoctonia solani Kühn. Potato tubers showing black scurf and rice plants affected by sheath blight were collected from different regions of the state and the isolates of R. solani so obtained were studied to determine their variability and to ascertain their cross-infectivity and response to fungicides. Potato isolates of R. solani did not infect rice plants but some rice isolates were weakly pathogenic on potato, the sclerotia being less firmly attached on tuber surface, indicating a possible unsuccessful attempt of rice isolates to infect potato. Rice isolates (66.6%) grew faster (>20 mm colony growth per 24 h) than those of the potato isolates (15–20 mm growth rate per 24 h). Hyphal width of isolates from both hosts varied from 7.2 to 12.1 μm. Colony growth of most potato isolates (61.2%) was appressed, whereas that of most rice isolates (53.3%) was fluffy. Rice isolates (73.3%) formed larger sclerotia (1.5–2.0 mm in diameter) than those of the potato isolates (0.5–1.0 mm in diameter). Anastomosis studies indicated that potato isolates belonged to AG-3 and AG-5 groups while rice isolates belonged to the AG-1-1-A group. Representative R. solani isolates from the two hosts showed significant variation in response to fungicides (i.e. carbendazim, carboxin, pencycuron, propiconazole and validamycin) based on their ED50 and ED90 values.  相似文献

Abstract Eighty-one isolates of Rhizoctonia solani AG4 were obtained from soil samples with diverse geographic origins in Korea. Forty-five out of 81 isolates (56%) contained at least one dsRNA molecule with their sizes ranging from 2.3 to > 23.1 kb. Nine different sizes of dsRNA molecules were found and extensive variation in banding patterns was observed among isolates. By comparing the sizes and combinations of dsRNAs, 21 distinct banding patterns were observed. Cytoplasmic fractions from 3 isolates showed the same dsRNA band patterns as those from the total cell extracts. The dsRNAs were stable through 10 successive hyphal tip cultures and serial transfers onto the potato dextrose agar supplemented with cycloheximide or emetine. The presence or absence of dsRNAs was not apparently correlated with disease severity, phenol oxidase activity, and geographic origin.  相似文献

Antifungal antibiotic from Pseudomonas chlororaphis isolate PA23 was identified as Phenazine using TLC and HPLC. Phenazine recorded the highest inhibition zone of 21?mm with 35.55% percent inhibition of mycelial growth of Pythium aphanidermatum over control. It had a significant effect on the hyphal morphology of P. aphanidermatum and on spore germination of Botryodiplodia theobromae and Alternaria solani. Disorganization of hyphal morphology of P. aphanidermatum includes vacuolization, cell content degeneration and hyphal lysis. Similarly interaction of phenazine with Rhizoctonia solani resulted in abnormal swelling of hyphal tips was noticed in the hyphal tips. Similarly the germination of sclerotia of Macrophomina phaseolina, R. solani and Sclerotium rolfsii were completely inhibited by phenazine at a concentration 50?μl. Incubation of the eggs of the root knot nematode Meloidogyne incognita in 30?μl concentration of phenazine, completely suppressed the hatching of juveniles.  相似文献
