In vitro germinability and membrane integrity (as revealed bythe fluorochromatic reaction (FCR) test) of pollen grains ofCrotalaria retusa L. stored in various organic solvents forsix months at –20±2 °C were studied and correlatedwith leaching of lipids, phospholipids, sugars and free aminoacids from pollen grains into organic solvents during storage.Pollen grains stored in organic solvents with low dielectricconstants (a measure of their non-polar nature), such as hexane,cyclohexane and diethyl ether, showed high scores for germinationand FCR and very little leaching of phospholipids, sugars andamino acids. Pollen grains stored in solvents with high dielectricconstants (a measure of their polar nature) such as isopropanoland methanol did not show germination or positive FCR scores,but showed extensive leaching of phospholipids, sugars and freeamino acids. The viability of pollen grains stored in organicsolvents seems to be determined largely by the effect of theorganic solvents on pollen phospholipid composition, which inturn affects membrane integrity and consequently pollen viability. Summary Methods currently available for evaluating pollen quality in vitro include, (a) tests of germinability; (b) tests of the stainability of the vegetative cell contents; (c) tests for enzyme activity, and (d) the fluorochromatic procedure (FCR), which tests principally the integrity of the plasmalemma of the vegetative cell. Using germinability in vitro as a standard, a comparison has been made between histochemical methods of classes (b), (c) and (d) in application to various pollens, immature, mature, and treated in ways known to affect viability and membrane state. Predictably, the lowest correlation was obtained with tests of stainability. The highest was given by the FCR, which generally provided an excellent guide to potential germinability. The FCR procedure is subject to various limitations, however, (a) A high correlation between FCR and germinability can only be expected when mature, ripe pollen is used; with immature pollen, the FCR will predict excessively high potential germinability. (b) The FCR may also predict a higher potential level of pollen function than in vitro germinability when the germination medium is sub-optimal. In this situation, however, it will generally give a better guide to fertilising capacity, (c) The FCR is not a test of pollen viability. Like germinability in vitro, it can yield a negative score with pollen which is nevertheless capable of functioning. For example, false negatives will be obtained with some species if the pollen is not properly pre-conditioned by rehydration before testing, an important point in monitoring stored pollen. The effects of osmoconditioning on the germination at 15 and25 °C of pepper (Capsicum annuum L.) seeds were studiedover a 3-year period with respect to temperature of storage.Untreated seeds stored at 5 °C showed high germinabilitythroughout the entire storage period, whereas untreated seedsstored at 25 °C showed a progressive decline in germinability,especially when assayed at 15 °C. Seeds that had been osmoconditionedprior to storage retained a high level of germinability irrespectiveof either storage or germination temperatures. When seeds thathad been stored at 25 °C were osmoconditioned after storage,there was a significantly higher germinability (assayed at 15 °C) in comparison with the corresponding untreated seeds.Seeds that were osmoconditioned twice (prior to and after storage)germinated in a similar way to those that had been osmoconditionedonce only Lactuca saliva L., lettuce, Hordeum oulgare L., barley, seed storage, moisture content, relative humidity, water potential, temperature, oxygen  相似文献

The various pathways of pollen development were investigatedin cultured anthers of Lolium temulentum, Festuca pratensisand the L. multiflorum x F. pratensis hybrid ‘Elmet’.In all three, development from the vegetative cell was the predominantpathway of pollen callus development. However, there were characteristicdifferences in the behaviour of the generative cell. In L. temulentumit remained attached to the pollen wall and degenerated, whereasin F. pratensis it divided. In ‘Elmet’ it detachedfrom the pollen wall and remained undivided. Both polarizedand unpolarized partitioned calluses were observed. Developmentof the fusion product of the vegetative and generative nucleiwere also observed in anthers of L. temulentum. Anomalous grainswere not found to be major source of pollen calluses. Sections of anthers of L. temulentum were used to investigatethe origin of S pollen grains, the small pale-staining grainswhich denote pollen dimorphism. Such grains form out of contactwith the tapetum and are therefore determined before or duringmeiosis (i.e. before harvest of anthers for culture). Sectionswere also used to demonstrate the influence of the durationof pretreatment on the development of the middle layer of theanther wall. Generative cell division in tricellular pollen grains of Sambucusnigra L. (Caprifoliaceae) has been examined with light and electronmicroscopy. During division the generative cell is located inthe centre of the pollen grain, near to the nucleus of a surroundingvegetative cell. Conventional mitosis of the generative cellis followed by cytokinesis through centrifugal cell plate formation.Two sister sperm cells remain in spatial contact with each otherand are surrounded, as formerly their progenitor cell was, bythe vegetative cell. From the changes of shape of the generativecell during division and of the sperm cells it may be assumedthat the space between these cells and the vegetative one containsa labile, non-rigid, wall material. No plastids have been observedin the generative cell and its mitochondria appear to be unequallydistributed between the two future sperm cells during division. To determine how the capability of pollen is affected by lengthy exposure to field conditions, we obtained pollen samples (from Erythronium grandiflorum) that were fresh (from newly dehisced anthers) or aged, from 6 to 24 hr. Without pretreating the collected grains, we compared their abilities 1) to retain viability, as indicated by the Heslop-Harrison fluorochromatic reaction (FCR) test; 2) to retain germinability, as indicated by pollen-tube growth in vitro on Brewbaker-Kwack medium; 3) to sire seeds when used as a pollen donor on recipient flowers in the field; and 4) to sire seeds competitively when mixed with another donor pollen in competitive pollinations. FCR scores declined drastically with pollen age. Germinability typically declined with age, but less drastically; the correlation between FCR and germinability ranged from strong to nil in different trials. The sharp decline in FCR was probably due to nonlethal, reversible desiccation. Seed-siring success, both in competitive and noncompetitive trials, bore no relation to pollen age or to germinability. Any loss in seed-siring ability in E. grandiflorum pollen over 24 hr is apparently overwhelmed by other sources of variation. The lack of correlation between seed-siring ability and FCR or germinability scores means that those tests should not be used to estimate overall pollen competence in this species. They should be so used in other species only if experiments have demonstrated their greater reliability.  相似文献

It has been reported that in species of Plumbaginaceae, Chenopodiaceae,Cruciferae and Amaryllidaceae a ‘male germ unit’is formed in which the two male gametes remain inter-connected,with one of the pair linked intimately to the vegetative nucleus.In two species the unit has been shown to remain intact in thepollen tube, and some accounts imply that it is polarized inits movement, the vegetative nucleus leading in the tube. Evidence given in this paper indicates that such a unit is unlikelyto be present in Helleborus foetidus L. (Ranunculaceae). Applicationof an optical sectioning technique has shown that at no timeis there a persistent linkage between the generative cell andthe vegetative nucleus in unhydrated, hydrated and germinatingpollen, nor is one present in the early pollen tube. Furthermore,no inter-connections between the two entities were seen in protoplastsfrom living, hydrated and incipiently germinating grains isolatedmechanically in an osmotically balancing medium. Following germination,the vegetative nucleus leaves the grain in advance of the generativecell in most instances, but in the samples examined the generativecell led in about 30 per cent of the tubes. Assembling a polarisedmale germ unit in these circumstances would require (a) theformation of an inter-connection between the vegetative nucleusand the generative cell or one of the gametes derived from itduring passage through the tube, and (b) where the generativecell initially leads in the tube, an exchange in relative positions.It is considered improbable that these conditions could consistentlybe met. Mature, incipiently germinating pollen of H. foetidus releasesa fibrillar component when extruded into suitable media. Mixed fluorescence/bright field microscopy of Rhododendron pollentubes in the first 72 h after germination reveals a lens-shapedgenerative cell which divides to give two associated spermswithin the original cell boundary. The generative cell is closelyassociated with the vegetative nucleus which precedes it in92 per cent of pollen tubes. Three-dimensional reconstruction from serial thin sections ofa pollen tube fixed 24 h after germination shows that the associationbetween the generative cell and vegetative nucleus is extremelycomplex. Elongated tails of the generative cell physically enfoldthe vegetative nucleus and penetrate into enclaves within it.The association has been clarified by use of the periodic acid-phosphotungsticacid-chromic acid technique to enhance electron contrast ofthe plasma membranes surrounding the generative cell. In thisbicellular system, the male germ unit association is apparentlyinitiated after pollen maturity but prior to generative celldivision. Four clones of Trifolium repens L., originating from Iceland,Denmark and Israel, were cultivated in controlled environmentchambers at 10, 14, 18 and 22°C. Seed set at 10°C was,on average, 45% lower than at 18°C. Generally, seed frequencieswere higher in the stylar ovule positions than in the basal.This polarization became more pronounced at low temperatures.At least 30% of the decrease in seed set was caused by inadequatepollen tube growth. This was further supported by in vitro studieswhich showed that pollen developed significantly shorter pollentubes at low temperatures and a strong positive correlationbetween the capacity of male parents to induce seed set in basalovule positions and the length of pollen tubes in vitro. Femaleage, determined by days relative to anthesis, was also foundto have a significant influence on seed set, i.e. when pollinationwas delayed until the fifth day of flowering, seed set decreasedby an average of 60% when compared to florets pollinated onthe first day of anthesis. Pollen age did not influence seedset. Summary The prolonged exposure of pollen Nicotiana tabacum to high humidity at both room temperature and 38° C did not affect membrane integrity as revealed by the fluorochromatic reaction (FCR) test, but did affect pollen vigour. At room temperature germination was not affected, although tube growth was reduced; at 38° C, there was both a reduction in tube growth and delayed germination. When the pollen was subjected to 1 h hydration followed by 1 h desiccation (up to a maximum of four cycles) at room temperature, a reduction in the FCR, germination and tube length after each desiccation treatment was observed. Subsequent hydration fully restored the FCR, but only partially restored germination and tube growth. At 38° C, however, FCR, germination, and tube growth were drastically reduced. The implications of these results on the relationship between FCR and germinability, the responses of pollen exposed to humidity and temperature stress in the field, and on pollen storage are discussed.  相似文献

Seed survival data for eight diverse species, namely the cerealbarley (Hordeum vulgare L.), the grain legumes chickpea (Cicerarietinum L.), cowpea [Vigna unguiculata (L.) Walp.] and soyabean [Glycine max (L.) Merr.], the timber trees elm (Ulmus carpinifoliaGleditsch.), mahogany (Swietenia humilis Zucc.), and terb (Terminaliabrassii Exell.), and the leaf vegetable lettuce (Lactuca sativaL.) were compared over a wide range of storage environments(temperatures from –13 °C to 90 °C, seed moisturecontents from 1.8 to 25% f. wt) using a viability equation developedpreviously. In accordance with that equation, the effect oftemperature on seed longevity was dependent upon the temperaturerange. The temperature coefficients of the viability equationdid not differ significantly (P > 0.05) among the eight speciesdespite their contrasting taxonomy. Thus the quantitative relationbetween seed longevity and temperature does not vary among diversespecies. The same conclusion was obtained for the coefficientsof a proposed alternative model of the relation between seedlongevity and temperature. The implications of the two temperaturemodels in the viability equation for extrapolations to low andvery low temperatures are discussed. Summary Attempts were made to store pollen grains of Crotalaria retusa L. in a mineral oil (paraffin oil) and two vegetable oils (soybean oil and olive oil). Under laboratory conditions pollen grains not stored in oil lost in vitro germinability within 15–30 days, while those stored in oils maintained some degree of germinability even after 60 days. Pollen samples stored in oils at –20° C did not show any decline in germinability or pollen tube vigour even after 6 months of storage. The results amply demonstrate the feasibility of using oils for short- and long-term pollen storage.  相似文献

The present work deals with the effect of the chelating agentsethylenediamine-di(o-hydroxyphenylacetic acid) [EDDHA] ethylenediamine tetra acetic acid [EDTA] and their iron salts, salicylicacid and ferric citrate on growth and antheridial productionin male clones of three dioecious mosses: Barbula gregaria,Bryum coronatum and Philonotis turneriana., Barbula and Bryumdevelop antheridia in ordinary cultural conditions on basalmedium, whereas Philonotis remains sterile. In Barbula and BryumEDDHA and EDTA enhance the percentage of fertile gametophytes.Fe-EDDHA and Fe-EDTA increase antheridial production as wellas vegetative growth and the former effect is more striking.In this respect Fe-EDDHA is better than Fe-EDTA for Barbula,whereas the reverse is true for Bryum. Salicylic acid inhibitsantheridial production and vegetative growth. Ferric citrateenhances vegetative growth in all three mosses. In Barbula andBryum it also stimulates antheridial production, and this effectis more marked than that on vegetative growth. The survival of seeds of lemon (Citrus limon L.), lime [C. arantifolia(Christm.) Swing.] and sour orange (C. aurantium L.) was examinedunder a wide range of constant moisture contents and temperatures.Seed longevity was increased by decreasing the moisture contentand temperature of the storage environment. Maximum viabilitywas maintained in the combination of storage conditions includingthe lowest moisture content (5 per cent) and lowest temperature(–20 °C) investigated. The microfibrillar polysaccharide component of the pollen intinecan be isolated by progressive chemical digestion of the exineand the cellular contents and the extraction of the matrix materials.The resulting intine ‘ghosts’ reveal various characteristicstructural features. The microfibrils have apparent individualdiameters in the range of 5–15 nm, but they are commonlyassociated laterally to form ribbons, or aggregated in strandsor cables of dimensions great enough to be resolved with theoptical microscope. These often show preferred orientations,which can be associated with pollen grain shape and with thedisposition-of the germination apertures. The apertural intinemay be structurally complex, as in Abutilon hybridum, where,after the removal of the exine, the polysaccharide caps whichoverlie the protein storage sites of the pollen grain wall retainthe elaborate patterning of the original cytoplasmic evaginationsfrom the vegetative cell. Previous studies have demonstrated that pollen of sorghum [Sorghumbicolor (L.) Moench] loses capacity to both germinate in vitroand to set seed in vivo soon after being shed. The current studyevaluates the capacity for dehydrated pollen to effect in vitrogermination, reduce tetrazolium chloride, and set seed on cytoplasmicmale sterile plants. Morphological changes during pollen germinationwere examined by scanning electron microscopy (SEM). Close to70% of the pollen germinated in 5 min, or less, when collectedat 80% relative humidity (RH) and stored in sealed glass vials.Pollen tubes elongated autotropically with atmospheric humidityapparently being a controlling factor in the process. Pollendehydrated at 50% RH and 25°C for 15-30 min neither germinatedin vitro, reduced tetrazolium chloride, nor set seed on malesterile plants. Rehydrating the pollen did not restore the capacityfor germination. SEM micrographs demonstrated that elongatingpollen tubes encircled the pollen grain and were contiguousto the surface. The anther in Ophiorrhiza is dithecous and tetralocular, itsdevelopment being of the dicotyledonous type. The anther wallcomprises epidermis, endothecium, middle layer and secretorytapetum. The pollen grains are tricolpate and triporate. Themicrospore nucleus undergoes division to form a vegetative nucleusand a generative nucleus and protrusions (pollen buds) are formedfrom the germ pores after the first division of the microsporenucleus. The vegetative nucleus moves into one of these budsor first breaks into a number of irregularly sized nuclear particleswhich enter into one, two or into all three buds, where theydegenerate. Then the pollen buds are separated and the generativenucleus divides inside the pollen tube to form two sperms. Seeds of quinoa (Chenopodium quinoa Willd.), sunflower (Helianthusannuus L.) and linseed (Linum usitatissimum L.) showed negativelogarithmic relations between longevity and moisture contentsbetween 4.4 and 15.4, 3.2 and 13.0, and 3.2 and 15.5%, respectively,in hermetic storage at 65 °C. However, between 1.8 and 3.1,1.1 and 1.9, and 1.1 and 2.1%, respectively, longevity did notvary. The critical moisture content, below which further reductionin moisture content no longer increased longevity in hermeticstorage at 65 °C, for each species was 4.1, 2.04 and 2.7%,respectively. Recently it has been reported that maize (Zea mays L.) pollen can be stored up to several hours in a hypertonic aqueous medium at 0°C without losing its germinability (Broglia and Brunori 1994). We found that both release and activity of pollen nucleases are diminished in a cold hypertonic aqueous medium. Nucleases can be washed off while preserving germination ability and thus preserving the possibility of passive uptake of exogeneous DNA into germinating pollen grains. Alternatively, active DNA transfer into non-germinating pollen grains in the storage medium itself may be facilitated, due to the very reduced nuclease activity in this medium.  相似文献
