The oscillation phenomena reported in the preceding article for the anaerobic continuous fermentation of glycerol by Klebsiella pneumoniae are analyzed in terms of metabolic fluxes (metabolic rates and yields) and stoichiometry of pathways. Significant oscillations in the fluxes of CO(2), H(2), formic acid, ethanol, and reducing equivalents are observed which show obvious relationships to each other. Changes in the consumption or production rates of glycerol, acetic acid, 1,3-propanediol, and ATP are irregular and have relatively small amplitudes compared with their absolute values. By comparing the metabolic fluxes under oscillation and steady state that have nearly the same environmental conditions it could be shown that pyruvate metabolism is the main step affected under oscillation conditions. The specific formation rates of all the products originating from pyruvate metabolism (CO(2), H(2), formic acid, ethanol, acetic acid, lactic acid, and 2,3-butanediol) show significant differences under conditions of oscillation and steady state. In contrast, the specific rates of substrate uptake, ATP generation, and formation of products deriving either directly from glycerol (1,3-propanediol) or from the upstream of pyruvate metabolism (e.g., succinic acid) are not, or at least not significantly, affected during oscillation. Stoichiometric analysis of metabolic pathways confirms that other enzyme systems, in addition to pyruvate: formate-lyase, must be simultaneously involved in the pyruvate decarboxylation under both oscillation and steady-state conditions. The results strongly suggest oscillations of activities of these enzymes under oscillation conditions. It appears that the reason for the occurrence of oscillation and hysteresis lies in an unstable regulation of pyruvate metabolism of different enzymes triggered by substrate excess and drastic change(s) of environmental conditions. Sustained oscillations have been observed in cell growth and glycerol metabolism by Klebsiella pneumoniae during long-term continuous cultivations under anaerobic conditions. However, the oscillation behavior of glucose–glycerol co-fermentation under micro-aerobic conditions has yet not been studied. In this study, the unclear mechanism of oscillation was investigated by using different substrates including glycerol, glucose, and mixtures of glycerol and glucose. A large perturbation of substrate concentration was operated to study the dynamic behavior of continuous fermentation by K. pneumoniae DSM 2026 under micro-aerobic conditions. Oscillations occurred when 80 g of glycerol l−1 was used as single carbon source or co-substrate with 35 g of glucose l−1, whereas no oscillation was observed when glucose was used alone. During the oscillation phases, parameters periodically passed maxima and minima at a relatively constant time interval, and the period of oscillation was 10–16 h. The comparison between glycerol and glucose metabolism revealed that a potential mechanism of oscillation lies in the accumulation of 3-hydroxypropionaldehyde, which is toxic to cell growth.  相似文献

The growth and metabolism of Saccharomyces cerevisiae was studied in steady-state chemostat cultures under conditions of scarce oxygen and excess glucose. The specific ethanol productivity and specific glucose uptake rate were stimulated by 50% within a narrow range of air/nitrogen mixtures to the fermentor. Fermentation was inhibited at slightly higher and lower air/nitrogen ratios, confirming similar results by previous investigators. This stimulation could not be caused by obvious mechanisms, such as the Pasteur or Crabtree effects. Since this maximum in the fermentation rate occurred in a steady-state chemostat and at a constant dilution rate, the ATP yield of the culture necessarily attained a minimum. Thus, changes in the energetic efficiency of growth or the degree of wasting of ATP were surmised. The steady-state biomass concentration at various oxygenation rates exhibited hysteresis phenomena. Ignition and extinction of the biomass concentration occurred as critical oxygen feed rates were passed. The hysteresis was prevented by adding yeast extract to or reducing the antifoam concentration in the medium. These medium alterations had the simultaneous effect of stimulating the fermentation rate, suggesting that ATP has a critical role in dictating the biomass concentration in micro-aerobic culture. Silicone polymer antifoam was found to stimulate glycerol production at the expense of ethanol production, having consequences for the energy generation and the biomass concentration of the culture.  相似文献

The appearance of sustained oscillations in bioreactor variables (biomass and nutrient concentrations) in continuous cultures of Saccharomyces cerevisiae indicates the complex nature of microbial systems, the inadequacy of current growth kinetic models, and the difficulties which may arise in bioprocess control and optimization. In this study we investigate continuous bioreactor behavior over a range of operating conditions (dilution rate, feed glucose concentration, feed ammonium concentration, dissolved oxygen, and pH) to determine the process requirements which lead to oscillatory behavior. We present new results which indicate that high feed ammonium concentrations may eliminate oscillations and that under oscillatory conditions ammonium levels are generally low and oscillatory as well. The effects of pH are complex and oscillations were only observed at pH values 5.5 and 6.5; no oscillations were observed at a pH of 4.5. Under our nominal operating conditions (feed glucose concentration 10 g/L, dilution rate 0.145 h(-1), feed ammonium concentration 0.0303M, dissolved oxygen level 50%, pH 5.5, and T = 30 degrees C) we found two possible final bioreactor states depending on the transient used to reach the nominal operating conditions. One of the states was oscillatory and characteristic of oxidative metabolism and the other was nonoscillatory and fermentative.  相似文献

This article addresses the issue of effect of fermentation parameters for conversion of glycerol (in both pure and crude form) into three value-added products, namely, ethanol, butanol, and 1,3-propanediol (1,3-PDO), by immobilized Clostridium pasteurianum and thereby addresses the statistical optimization of this process. The analysis of effect of different process parameters such as agitation rate, fermentation temperature, medium pH, and initial glycerol concentration indicated that medium pH was the most critical factor for total alcohols production in case of pure glycerol as fermentation substrate. On the other hand, initial glycerol concentration was the most significant factor for fermentation with crude glycerol. An interesting observation was that the optimized set of fermentation parameters was found to be independent of the type of glycerol (either pure or crude) used. At optimum conditions of agitation rate (200 rpm), initial glycerol concentration (25 g/L), fermentation temperature (30°C), and medium pH (7.0), the total alcohols production was almost equal in anaerobic shake flasks and 2-L bioreactor. This essentially means that at optimum process parameters, the scale of operation does not affect the output of the process. The immobilized cells could be reused for multiple cycles for both pure and crude glycerol fermentation.  相似文献

Pichia pastoris is a successful system for expressing heterologous proteins and its fermentation pH is always maintained below 7.0. However, particular proteins are unstable under acidic conditions, such as methionine adenosyltransferase (MAT), and thus fermentation under acidic pH conditions is unsuitable because protein activity is lost owing to denaturation. Here, a strategy employing alkaline pH in the late fermentation period was developed to improve MAT production. Initially, P. pastoris KM71 was transformed with the mat gene to overexpress MAT. After 72 h of in vitro incubation at different pH values, the expressed MAT displayed highest stability at pH 8.0; however, pH 8.0 inhibited cell growth and induced cell rupture, thus affecting protein production. To balance MAT stability and Pichia cell viability, different pH control strategies were compared. In strategy A (reference), the induction pH was maintained at 6.0, whereas in strategy B, it was gradually elevated to 8.0 through a 25 h transition period (80 ~ 105 h). MAT activity was 0.86 U/mg (twofold higher than the control). However, MAT content was reduced by 50% when compared with strategy A, because of proteases released upon cell lysis. To improve cell viability under alkaline conditions, glycerol was added in addition to methanol (strategy C). When compared with strategy B, the MAT-specific activity remained nearly constant, whereas the expression level increased to 1.27 g/L. The alkaline pH control strategy presented herein for MAT production represents an excellent alternative for expressing proteins that are stable only under alkaline conditions.  相似文献

Periodic cyclic-AMP pulses control the cell aggregation and differentiation of Dictyostelium discoideum. Another component required for the aggregation and differentiation of these cells appears to be extracellular Ca+ +. Oscillations in extracellular Ca+ + concentration were investigated in suspensions of differentiating cells. We observed spike-shaped and sinusoidal Ca+ + oscillations. In the course of differentiation, spike-shaped Ca+ + oscillations preceded sinusoidal oscillations, and no phase change occurred at the transition from spike-shaped to sinusoidal Ca+ + oscillations. Spike-shaped and sinusoidal Ca+ + oscillations were related to oscillations in (1) the cyclic-AMP and cyclic-GMP content of cells, (2) the light-scattering properties of cells, and (3) the extracellular pH. Spikeshaped Ca+ + oscillations were observed together with cyclic-AMP oscillations. The minima of the extracellular Ca+ + concentration trailed the maxima of the cyclic-AMP concentration by about 30 s. Sinusoidal Ca+ + oscillations were not accompanied by measurable cyclic-AMP oscillations. The amplitudes of the sinusoidal Ca+ + oscillations were smaller than those of the spike-shaped Ca+ + oscillations. A Ca+ + oscillation of small amplitude (instead of a spike-shaped oscillation) was observed when one cyclic-AMP spike was skipped. Our results provide evidence for the existence of a sinusoidal cyclic-AMP-independent Ca+ + oscillation of small amplitude, and they also suggest that spike-shaped Ca+ + oscillations may be superimposed on such small-amplitude oscillations. When D. discoideum cells produce cyclic-AMP spikes, the uptake of additional Ca+ + is induced, resulting in Ca+ + oscillations of a large amplitude.  相似文献

Citrobacter freundii DSM 30040 immobilized on modified polyurethane carrier particles PUR 90/16 was used for continuous glycerol fermentation in an anaerobic fixed bed reactor with effluent recycle and pH control (fixed bed loop reactor). The fermentor was run with buffered mineral medium under growth conditions resulting in the permanent renewal of active biomass. The effects of glycerol concentration in the feed, dilution rate (D), pH and temperature (T) were investigated to optimize the process. With 400 mm glycerol in the feed, pH 6.9, T = 36°C and D = 0.5 h–1 the maximum productivity could be determined as 8.2 g/l per hour of 1,3-propanediol.  相似文献

Synchronous cultures of the budding yeast Candida utilis prepared by continuous-flow size selection showed respiratory oscillations when the energy source was either glucose, acetate or glycerol. The period of the oscillations was about one-third of the cell cycle time (i.e. about 0.5 h). No fluctuations in heat evolution could be detected. In organisms growing with acetate or glycerol, the effects of cyanide, N,N'-dicyclohexylcarbodi-imide and carbonyl cyanide m-chlorophenylhydrazone (maximum inhibition of respiration at respiratory maxima, maximum uncoupling of energy conservation at respiratory minima) suggest that the control mechanism responsible for the oscillations is mitochondrial respiratory control in vivo. The effects of cyanide and N,N'-dicyclohexylcarbodi-imide on the respiration of cultures growing synchronously with glucose were different from those for cultures growing with the non-fermentable substrates; this suggests that the mitochondrial respiratory system interacts with the early reactions of glucose utilization.  相似文献

H(2) and ethanol production from glycerol-containing wastes discharged from a biodiesel fuel production plant by Enterobacter aerogenes NBRC 12010 was demonstrated in bioelectrochemical cells. Thionine as an exogenous electron transfer mediator was reduced by E. aerogenes, and was re-oxidized by a working electrode applied at +0.2 V against a Ag/AgCl reference electrode by a potentiostat (electrode system). At the initial glycerol concentration of 110 mM, 92.9 mM glycerol was consumed in the electrode system with 2 mM thionine after 48 h. On the other hand, the concentration of glycerol consumed was only 50.3 mM under the control conditions without thionine and the electrodes (normal fermentation). There are no differences in the yields of H(2) and ethanol against glycerol consumed between the control conditions and the conditions with the electrode system. A pH of 6.0 was suitable for the H(2) production in the range between pH 6 and pH 7.5 in the electrode system. At pH values of 7.0 and 7.5, H(2) production decreased and formate was remarkably produced in the reaction solution. The rates of both glycerol consumption and the H(2) and ethanol production increased as the thionine concentration and the surface area of the working electrode increased. After 60 h, 154 mM of the initial 161 mM glycerol concentration in the wastes was consumed in the electrode system, which is a 2.6-fold increase compared to the control experiment. Biotechnol. Changes in the intramuscular pH oscillations were examined by the use of an antimony electrode upon perfusing the isolated rat heart under different experimental conditions. The pH oscillations were decreased upon perfusing the hearts with Na+- or Ca2+-free medium and increased upon perfusing with K+-free medium. Increasing the temperature of perfusion medium from 25 to 40 degrees C or omitting glucose from the perfusing medium decreased the magnitude of oscillations. On the other hand, complete interruption of the perfusion flow resulted in an increase in the amplitude of pH oscillation. An initial increase followed by a decrease in the pH oscillation was seen when hearts were perfused with medium containing lactic acid at pH 6.6. These results suggest that pH oscillations reflect fluctuations in myocardial metabolism.  相似文献

Dimethylamine, methylamine, propylamine, and pyrrolidine were the major amines formed by Bacteroides fragilis NCDO 2217 during the active phase of growth in batch culture. Production of these metabolites was strongly pH dependent and was optimal under acidic conditions (pH 6.0). Low pH also favored the formation of pyrrolidine, cadaverine, and dimethylamine by Clostridium perfringens C523, but the reverse was the case with putrescine, butylamine, and propylamine, where production was maximal at neutral pH. B. fragilis was grown in continuous culture under either starch or casein limitation. Amine formation was influenced by carbohydrate availability and was greatest when the bacteria were grown at high growth rates (dilution rate, 0.20/h) under starch limitation, where they constituted about 18% of the total fermentation products measured. Amine production was optimal and increased concomitantly with growth rate when C. perfringens was grown in glucose-limited continuous culture. Under conditions of high growth rate and glucose limitation, amines accounted for approximately 27% of the fermentation products measured. When glucose in the feed medium was increased from 5 to 15 g/liter, amine production was repressed, and under these nutritional conditions the growth rate had little effect on the process.  相似文献

Characteristic steady-state mRNA level oscillations were monitored for the chlorophyll a/b-binding (cab) protein in tomato plants grown under the natural day/night (light/dark) regime as well as under constant environmental conditions. This typical expression pattern was altered when plants were transferred to different light/dark regimes. For example, by shifting the light phase by six hours, a change of the time points of maximum and minimum of expression level was monitored, while the principal oscillation pattern remained the same. It appeared that the transition from dark to light is involved in determining the time points of minima and maxima of mRNA accumulation.After exposing tomato plants to an abnormal light/dark periodicity (e.g. six hours of alternating light/dark) an altered oscillation pattern was determined: within 24 hours two maxima of cab mRNA levels were detected. However, this entrained abnormal rhythm was not manifested at the molecular level and the circadian pattern reappeared under constant environmental conditions (e.g. darkness). This result favours the hypothesis that the oscillation pattern of the cab mRNA in tomato plants is not only endogenous but also hereditary.  相似文献

以从新疆艾丁湖采集的土样中分离出的中度嗜盐菌Salinivibrio YS为研究对象,利用该菌在厌氧条件下生产2,3-丁二醇和琥珀酸,在单因素摇瓶实验基础上,确定影响产物积累的各因素及其相应条件,再利用正交试验确定这些参数的最佳水平,即温度33℃,起始pH为8.0,发酵过程pH为7.0,乙酸添加量为3 g/L,NaCl浓度为l0 g/L.利用优化条件进行3L体系的发酵放大实验,经过108 h的无氧发酵,2,3-丁二醇的产量可达35.05 g/L,而琥珀酸的含量则高达22.46 g/L,且其糖的总转化率高达约50%.首次利用嗜盐菌在厌氧条件下生产2,3-丁二醇和琥珀酸,拓展了嗜盐菌的应用,同时也为生产2,3-丁二醇和琥珀酸提供了新的思路.  相似文献

Oscillations of photosynthesis induced in leaves of Vicia faba L. were accompanied by oscillations not only in the pH of the chloroplast stroma, but also by pH oscillations in the cytosol and in the vacuole of leaf mesophyll cells. Cytosolic pH oscillations were in phase with stromal oscillations, but antiparallel to vacuolar pH oscillations. During maxima of photosynthesis, the cytosolic pH exhibited maxima and the vacuolar pH minima. Vacuolar acidification is interpreted to be the result of energized proton transport from the cytosol into the vacuole. Since the ratio of dihydroxyacetone phosphate to phosphoglycerate is maximal during the peaks of photosynthesis (Stitt et al., 1988, J. Plant Physiol. 133, 133–143; Laisk et al., 1991, Planta 185, 554–562), while the activity of NADP-malic dehydrogenase is highest during minima of photosynthesis (Scheibe and Stitt, 1988, Plant Physiol. Biochem. 26, 473–481), the present data indicate in agreement with earlier observations (Yin et al., 1991, Planta 184, 30–34) that light-dependent cytosolic energization is brought about by the oxidation of dihydroxyacetone phosphate rather than of malate. They also indicate that the over-reduction of the electrontransport chain observed during minima of photosynthesis is relieved not predominantly by oxaloacetate reduction and export of the resulting malate from the chloroplasts but by another reaction, presumably oxygen reduction.Abbreviations CDCF 5-(and 6-)carboxy-2,7-dichlorofluorescein  相似文献

Diastolic mechanical oscillations of right ventricular rabbit papillary muscles investigated at 15 degrees C in the perfusion chamber were analysed using a relaxation model with 6 parameters. From this analysis follows that the first diastolic oscillation amplitude plotted against the driving interval ("interval amplitude curve" of the first diastolic oscillation) shows tow maxima thus differing from the shape of the interval strength curve of the preceding driven contraction. It is concluded that the amplitude of diastolic oscillations is less determined by the Ca-amount released from the sarcoplasmic reticulum during the action potential than by the frequency of processes which effect the Ca release and the Ca-sequestration. The damping ratio varies within the range of tested driving intervals (0.36 to 10 s). After an extrasystole damping ratio and period of diastolic oscillation are diminished compared with the values after regularly driven contractions.  相似文献

Ice-binding proteins (IBPs) can bind to the ice crystal and inhibit its growth. Because this property of IBPs can increase the freeze–thaw survival of cells, IBPs have attracted the attention from industries for their potential use in biotechnological applications. However, their use was largely hampered by the lack of the large-scale recombinant production system. In this study, the codon-optimized IBP from Leucosporidium sp. (LeIBP) was constructed and subjected to high-level expression in methylotrophic Pichia pastoris system. In a laboratory-scale fermentation (7 L), the optimal induction temperature and pH were determined to be 25 °C and 6.0, respectively. Further, employing glycerol fed-batch phase prior to methanol induction phase enhanced the production of recombinant LelBP (rLeIBP) by ~100 mg/l. The total amount of secreted proteins at these conditions (25 °C, pH?6.0, and glycerol fed-batch phase) was ~443 mg/l, 60 % of which was rLeIBP, yielding ~272 mg/l. In the pilot-scale fermentation (700 L) under the same conditions, the yield of rLeIBP was 300 mg/l. To our best knowledge, this result reports the highest production yield of the recombinant IBP. More importantly, the rLeIBP secreted into culture media was stable and active for 6 days of fermentation. The thermal hysteresis (TH) activity of rLeIBP was about 0.42 °C, which is almost the same to those reported previously. The availability of large quantities of rLeIBP may accelerate further application studies.  相似文献

Stoichiometric analysis is applied to continuous glycerol fermentation by Clostridium butyricum to calculate theoretical maximum yields and to predict preferred pathways under different conditions. The upper limits of product concentration and productivity as a function of dilution rate in continuous culture is also predicted from product inhibition kinetic. The theoretical maximum propanediol yield (0.72 mol/mol glycerol) which is calculated for a culture without hydrogen and butyric acid formation agrees well with the experimental maximum value (around 0.71 mol/mol). Comparisons of experimental results (product concentration and productivity) with theoretical calculations and those of the glycerol fermentation by Klebsiella pneumoniae reveal that the production of 1,3-propanediol by C. butyricum is far below the optimum performance available with the present strain. One of the reasons is the relatively high formation of butyric acid under the culture conditions so far applied. The distribution of reducing equivalents to propanediol and hydrogen is also suboptimal. The utilization of the reducing power from pyruvate oxidation for propanediol production is about 60–70% of the theoretical maximum under the present experimental conditions.  相似文献

The fermentation of glycerol by Clostridium pasteurianum was studied with respect to product formation as influenced by the culture conditions. In the majority of batch cultures, butanol was the main fermentation product, but a varying fraction of glycerol was also converted to 1,3-propanediol, butyric and acetic acids and ethanol. More than 60 g/l glycerol was utilized, and up to 17 g/l butanol was produced. Fed-batch cultures did not offer an advantage. When molecular nitrogen was used as a nitrogen source, the fermentation time was prolonged by a factor of 1.5. Fermentations at constant pH values between 4.5 and 7.5 did not reveal significant differences in product formation except for an increase in the ethanol content starting at pH 6.5. Chemostat cultures also yielded predominantly n-butanol, but in some fermentations, the 1,3-propanediol fraction was relatively high. The pH auxostat cultures, which were operated at a glycerol excess, contained 1,3-propanediol as the main product. As a whole, the fermentations were characterized by a certain variability in product formation under seemingly equal or slightly varied conditions.
