从内蒙古中东部草原7个不同地理种群的羽茅Achnatherum sibiricum中分离、培养得到484株内生真菌,根据其形态学特征进行了分类学鉴定。结果表明,其中大部分菌株属于Neotyphodium内生真菌,并发现有少量Neotyphodium有性型Epichloё菌株存在。通过与已发表模式菌株的比较,鉴定其中两个种为N.chisosum和N.huerfanum。N.chisosum为羽茅内生真菌的优势种,而N.huerfanum是首次在羽茅中被发现。对其余暂未能确定种的菌株的菌落和显微形态特征进行了详细描述。不同地理种群羽茅内生真菌的分离率有自东向西增加的趋势,而内生真菌的形态多样性有自东向西减少的趋势。  相似文献

Lysergyl peptide synthetase 1 catalyzes the assembly of toxic ergopeptines from activated D-lysergic acid and three amino acids. The gene encoding this enzyme in the endophytic fungus Neotyphodium lolii was analyzed and compared to a homologous gene from the ergot fungus Claviceps purpurea. Each gene contained two introns, which were found in the same relative position within two modules of the gene. The 5' ends of the two genes were unusually divergent. Signature sequences determining substrate specificity were similar in adenylation domains that recognized identical amino acids but differed within the adenylation domain for the amino acid that varies between the major ergopeptines of the two fungi. Homologues were detected in several related endophytic fungi; the tall fescue endophyte Neotyphodium coenophialum contained a divergent, second copy of the gene. Our results provide new information on the structure and distribution of this important peptide synthetase involved in ergot alkaloid biosynthesis.  相似文献

Laboratory experiments were conducted to compare the expression of Diuraphis noxia (Mordvilko) (Homoptera: Aphididae) resistance in four plant introduction (PI) lines of wild barley (Hordeum) infected with different species or strains of endophytic fungi (tribe Balansieae, family Clavicipitaceae, Neotyphodium gen. nov. [formerly Acremonium]). Aphid densities were significantly lower on endophyte-infected plants of PI 314696 (H. bogdanii Wilensky) and PI 440420 (H. brevisubulatum subsp. violaceum (Boissier & Hohenacker)), compared with densities on endophyte-free plants of both PI lines in population growth experiments. This endophyte- associated resistance was the result of antibiosis effects or starvation. In other experiments, endophyte-free plants of PI 269406 and PI 440413 (H. bogdanii) were not superior to endophyte-infected conspecifics as host plants of D. noxia. Our results demonstrate the influence of host plant species/genotype and endophyte species/strain on expression of aphid resistance, provide an explanation of the high levels of D. noxia resistance in PI 314696 and PI 440420 previously reported in the literature, and underscore the potential importance of endophytic fungi in conferring insect resistance in wild barley.  相似文献

综述了国内外近年对禾草 -内生真菌 (N eotyphodium)的研究进展。全世界现已报道 14种禾草内生真菌 ,与 2 3个属的禾草形成共生体 ,我国已在 13属 2 5种天然草地禾草中发现内生真菌 ,其中发草属 (Descampsia)、大麦属 (H ord eum)和赖草属(L eymus)以往在国际文献中未曾报道。内生真菌在禾草体内产生的生物碱 ,致使采食带菌禾草的马、牛、羊、鹿等家畜产生中毒症状 ,每年给美国、新西兰等国造成的经济损失达 6.4亿美元之多 ,对其毒理研究取得了进展。已发现我国醉马草 (Achnatheruminebrians)对家畜的毒性与内生真菌的侵染有关。内生真菌侵染增加可使禾草对 42种害虫的抗性 ,并可增加对某些线虫和病害的抗性。与不带菌禾草相比 ,带菌禾草的另一特点是抗逆性强 ,牧草产量高。国际在该领域的研究主要集中在多年生黑麦草(L olium p erenne)和高羊茅 (Festuca arundinacea)。我国的研究发现 ,带菌布顿大麦草 (H ordeum bodg anii)和圆柱披碱草(Elymus cylind ricus)的牧草产量分别增加 3 3 .3 %和 2 78.8% ,分孽数分别增加 13 6.8%和 84.5%。目前 ,国际研究的重点包括大规模开展内生真菌生物学与生态学特性的研究 ,创造不含对家畜有害毒素的有益禾草 -内生真菌共生体 ,培育带内生真菌的草坪草品种 ,培育抗毒  相似文献

In a previous study, a total of 484 endophytic fungi were isolated and purified from seven populations of Achnatherum sibiricum (L.) Keng collected at six geographical locations in Inner Mongolia, China. Based on growth rates as well as morphological characteristics, the isolates were classified into five morphotypes. Among them, morphotypes A, B and C were ascribed to the same species, Neotyphodium chisosum, based on ITS sequences. Morphotype E was identified as Epichlo? amarillans. In the present study, four morphotypes, A, B, C and E, belonging to two species, were chosen for an in vitro pathogen trial. The results showed that both endophyte colonies and endophyte filtrate of all morphotypes could inhibit the mycelia growth and spore germination of the pathogen fungi tested. The magnitude of inhibition varied not only between species, but also among morphotypes of the same species. Overall, the antifungal ability of E. amarillans (morphotype E) was higher than that of N. chisosum. Within N. chisosum, the antifungal ability was highest in morphotype C, followed by morphotype A, and lowest in morphotype B. This variability suggests that different morphotypes might represent different genotypes of endophyte. The effect of endophyte infection on the host grass should be examined not only on the species level but also on the morphotype level to determine the possible interactions.  相似文献

为研究长白山自然保护区北坡岳桦树(Betula ermanli)的根系内生真菌的种类及其分布,笔者于2011年7~9月分别在长白山自然保护区北坡海拔1700~2100m处采集岳桦树根系组织。分离得到411株内生真菌,经形态学鉴定,分别属于17个属。研究结果表明:除不产孢菌株外,青霉属Penieillium分离频率最高(21.90%),木霉属Trichodmma分离频率次之(17.52%),镰孢菌属Fusarium、伞形霉属Umbelopsis和曲霉属Aspergillus分离频率也相对较高(5.60%、5.35%和4.87%)。通过分析发现,从不同月份和不同海拔高度采集的岳桦树根系分离的内生真菌的种类和数量不同。试验结果表明,长白山自然保护区岳桦树根系的内生真菌种类较多,其种类、数量和分布与采样时间以及海拔等因素存在一定的联系。  相似文献

1. Fast-growing populations of phytophagous insects can be limited by the presence of natural enemies and by alkaloids that are produced by symbiotic associations of many temperate grass species with endophytic fungi. It is unclear if and how acquired plant defences derived from endophytic fungi interact with natural enemies to affect phytophagous insect populations. 2. To assess the relative importance of endophytic fungi compared to that of natural enemies on the population dynamics of phytophagous insects, we carried out a fully factorial field experiment, in which the presence of natural enemies and the presence of endophytic fungi were manipulated simultaneously. Target colonies of aphids were monitored for 8 weeks starting from their natural appearance in the field to the end of the aphid season. 3. We show that on Lolium perenne increased natural enemy densities reduced the individual numbers of two common cereal aphids, Rhopalosiphum padi and Metopolophium festucae. 4. The presence of the endophytic fungi Neotyphodium lolii reduced the number of M. festucae but did not affect the number of R. padi. The reduction in R. padi numbers by predators and parasitoids was not influenced by the presence of endophytes. For adult M. festucae, however, the negative effects of natural enemies were significant only in the absence of endophytes. 5. Over the duration of the experiment, the effect of natural enemies on aphid colony growth was much stronger than the effect of the endophytic fungi N. lolii, presumably because predator and parasitoid action on aphid colonies is much faster than any effects of endophytes. 6. Our results demonstrate that with simultaneous action of acquired endosymbionts and natural enemies, both factors can control aphid colony growth but they generally act independently of each other.  相似文献

Members of Neotyphodium endophytic fungi infecting Lolium perenne L. and Lolium arundinaceum Darb. alter the synthesis of several metabolites. In this study we determined the antioxidative capacity of phenolic compounds from L. perenne and L. arundinaceum infected with Neotyphodium lolii (Latch, Christensen et Samuels) and Neotyphodium coenophialum (Morgan-Jones et Gams) Glenn, Bacon et Hanlin, respectively. The antioxidant capacity was determined by measuring the scavenging capacity of aqueous methanolic extracts to the free-radical DPPH (2,2-diphenyl-1-picrylhydrazyl). L. perenne infected with ‘wild-type’ strain endophyte showed the highest scavenging capacity, whereas endophyte-free showed the least. Those infected with the ‘novel’ strains AR1 and AR37 showed intermediate capacities. L. arundinaceum infected with the ‘novel’ strain AR542 showed a lower scavenging capacity compared with endophyte-free counterparts, regardless of the L. arundinaceum germplasm. The endophyte-free Mediterranean and Continental L. arundinaceum showed a higher capacity to scavenge DPPH when compared with the endophyte-infected Mediterranean and Continental L. arundinaceum. These results suggest that the endophytic fungi alter the antioxidative capacity of the grasses.  相似文献

Presence of Neotyphodium-like endophytes in European grasses   总被引:1,自引:0,他引:1  
In order to improve knowledge of Neotyphodium‐likc fungi in European grasses, microscopic detection of endophytic mycelium was achieved in living grasses and in seeds. Samples of grasses were gathered near permanent pastures or along paths in France (93 samples, 13 genera, 22 species). Typical convoluted mycelium was found inside leaf sheaths of four genera, especially in Festuca and Lolium spp. The results suggest that endophytes were present most often in the southern regions where plants may suffer from summer drought. Endophytes were looked for in 489 seed samples (63 genera, 237 species) received from 24 European botanical gardens. Neotyphodium was found in seeds of only six genera (22 species), especially in Festuca (15 species) and Lolium (three species). No endophytic mycelium was found in the other 215 species, although other work had demonstrated the presence of the fungus in 39 of the species analysed. Neotyphodium‐likc mycelium was detected for the first time in Festuca juncifolia, F. trachyphylla, F. vaginata, F. pseudodalmatica, F. rupicaprina, F. arenaria, Vulpia ciliata and Micropyrum tenellum. Further studies are needed for the correct identification of the Neotyphodium species observed.  相似文献

Plants growing in natural environments experience myriad interactions with a diverse assemblage of pathogens, parasites and mutualists. Many of these interactions involve symbiotic bacteria and fungi, but they also include macroparasitic plants. In this study, we investigated the interactions among a host grass (Lolium pratense, ex., Festuca pratensis), its symbiotic endophytic fungus (Neotyphodium uncinatum), a root hemiparasitic plant (Rhinanthus serotinus) of the host grass and a generalist herbivore (aphid Aulacorthum solani) of the hemiparasite. We demonstrate that the hemiparasitic plant acquires defending mycotoxins produced by the endophytic fungus living within their shared host grass. The uptake of defensive mycotoxins from the endophyte‐infected host grass enhances the resistance of the hemiparasitic plant to the generalist aphid herbivore. Endophyte infection increases the performance of the hemiparasitic plant, but reduces the growth of the host grass. In other words, the mutualistic endophytic fungus becomes parasitic in the presence of the hemiparasitic plant. Our results suggest that the outcomes of grass–endophyte interactions are conditional on the complexity of community‐level interactions; thus, the outcome of multispecies interactions may not be predictable from pair‐wise combinations of species.  相似文献

采用RAPD技术对分离自我国的13株Epichlo?spp.和9株Neotyphodiumspp.的禾本科植物内生真菌进行了遗传多样性分析,同时对其中的4株进行了rDNA-ITS序列分析及系统发育研究。RAPD分析结果显示:原产自我国的菌株与原产自欧洲的菌株N.uncinatum之间亲缘关系较远;我国的21个菌株之间也存在一定的遗传多样性。rDNA-ITS序列分析表明:原产自我国的菌株聚为一枝,表明我国的Neotyphodium属真菌很有可能直接由我国的Epichlo?属真菌演化而来的新的类群;我国的Epichlo?属真菌有可能和国外推测的一个未确定的Neotyphodium属真菌的杂交进化起源(LAC)有关。  相似文献

However, this model of apical growth is incompatible with endophyte colonization of grasses by the symbiotic Neotyphodium and Epichlo? species. These fungi are transmitted through host seed, and colonize aerial tissues that develop from infected shoot apical meristems of the seedling and tillers. We present evidence that vegetative hyphae of Neotyphodium and Epichlo? species infect grass leaves via a novel mechanism of growth, intercalary division and extension. Hyphae are attached to enlarging host cells, and cumulative growth along the length of the filament enables the fungus to extend at the same rate as the host. This is the first evidence of intercalary growth in fungi and directly challenges the centuries-old model that fungi grow exclusively at hyphal tips. Symbiotic associations between grasses and vertically transmitted endophytic fungi are widespread in nature. Within grass populations, changes in the frequency of infected plants are driven by influence of the endophyte on the fitness of their hosts and by the efficiency of endophyte transmission from parent plants to their offspring. During the seed stage, the endophyte might influence the fitness of its host by affecting the rate of seed viability loss, whereas the efficiency of endophyte transmission is affected by losses of viability of the fungus within viable seeds. We assessed the viability losses of Lolium multiflorum seeds with high and low level of infection of the endophyte Neotyphodium occultans, as well as the loss of viability of the fungus itself, under accelerated seed ageing and under field conditions. Starting with high endophyte-infected accessions of L. multiflorum, we produced their low endophyte-infected counterparts by treating seeds with a fungicide, and subsequently multiplying seeds in adjacent plots allowing pollen exchange. In our accelerated ageing experiments, which included three accessions, high endophyte-infected seeds lost viability significantly faster than their low endophyte-infected counterpart, for only one accession. High endophyte-infected seeds of this particular accession absorbed more water than low endophyte-infected seeds. In contrast, the endophyte lost viability within live seeds of all three accessions, as the proportions of viable seeds producing infected seedlings decreased over time. In our field experiment, which included only one accession, high endophyte-infected seed lost viability significantly but only slightly faster than low endophyte-infected seed. In contrast, the loss of viability of the endophyte was substantial as the proportions of viable seeds producing infected seedlings decreased greatly over time. Moving the seeds from the air to the soil surface (simulating seed dispersion off the spikes) decreased substantially the rate of seed viability loss, but increased the rate of endophyte viability loss. Our experiments suggest that, in ageing seed pools, endophyte viability loss and differential seed mortality determine decreases in the proportions of endophyte-infected seeds in L. multiflorum. Endophyte viability loss within live seeds contributes substantially more to infection frequency changes than differential viability losses of infected and non-infected seeds.  相似文献

红树林作为一种特殊的植物群落具有丰富的内生真菌资源,目前已分离鉴定的红树林真菌超过200种,成为海洋真菌的第二大类群,已报道的红树林内生真菌主要类群是链格孢霉(Alternaria)、曲霉(Aspergillus)、芽枝霉(Cladosporium)、炭疽菌(Colletotrichum)、镰孢霉(Fusarium)、拟青霉(Paecilomyces)、拟盘多毛孢(Pestalotiopsis)、青霉(Penicillium)、茎点霉(Phoma)、拟茎点霉(Phomopsis)、叶点霉(Phyllosticta)和木霉(Trichoderma)等.大部分红树林内生真菌具有较宽的宿主范围,极少数只有单一的宿主,不同红树林植物的内生真菌区系及优势种群有很大差异.红树林内生真菌的定殖因宿主植物不同部位、植株的年龄及季节和环境的变化明显不同.红树林内生真菌能产生多种代谢产物,具有抗菌、抗肿瘤等药用价值.红树林植物内生真菌的研究和开发具有重要意义.本文综述了红树林内生真菌的生物多样性及其分布、生物学功能和次生代谢产物等方面的研究进展.  相似文献

Arbuscular mycorrhizal (AM) fungi are ubiquitous root symbionts that form intimate associations with the majority of plants growing in aerobic soil; fungal endophytes live internally, either intercellularly or intracellularly, and asymptomatically within plant tissues. Their presence is correlated with an increased response to biotic and abiotic stress. The populations of AM and of endophytic fungi were studied in the roots of different rice varieties grown in aerobic condition, in experimental fields in Vercelli, North Italy. All the rice varieties resulted colonised by AM fungi with a percentage of arbuscularisation ranging between 4% and 28%. Preliminary molecular analyses on some rice varieties showed that the AM population was composed of fungi identified as Glomus intraradices , on the basis of 18S ribosomal gene. All the varieties analysed but one resulted in colonisation by endophytic fungi. About 300 fungal isolates were obtained, belonging mainly to the genera Neotyphodium , Stagonospora and Penicillium .  相似文献

The symbiosis between grasses and endophytic fungi is a common phenomenon and can affect herbivore performance through acquired, chemical plant defence by fungal alkaloids. In laboratory experiments, two species of common grass aphids, Rhopalosiphum padi and Metopolophium dirhodum were tested, in a population experiment (on four plant cultivars) and individually (on one plant cultivar) for the effects of the endophyte, Neotyphodium lolii, that forms symbiotic associations with perennial ryegrass Lolium perenne. In the population experiment that lasted for four aphid generations both aphid species showed decreased population sizes when feeding on each of the four endophyte-infected cultivars. Individuals of R. padi tested individually showed reduced adult life span and fecundity when feeding on infected plants. Individuals of M. dirhodum showed no response in any of the traits measured. This suggests that R. padi individuals are more sensitive to endophyte infection than M. dirhodum individuals. However, all infected grass cultivars reduced population sizes of both aphid species over four generations. Therefore, fungal endophytes can reduce populations of aphid herbivores independent of plant cultivars.  相似文献
