镜鲤体长性状的QTL定位分析   总被引:1,自引:0,他引:1

以镜鲤良种后代为祖父母本所培育的杂交F2群体的68个个体为材料,利用553个分子标记(217个SSR和336个SNP标记)对其进行基因型检测,运用JoinMap4.0软件包对遗传连锁图谱进行构建。利用MapQTL5.0区间作图法(Interval mapping)进行QTL检测,通过置换实验(1 000次重复)确定连锁群显著性水平阈值。在对体长的区间定位中共检测到12个与体长性状相关的QTLs区间,分布在BL-1-1(SNP0137-SNP1481)、BL-4-1(SNP0092-HLJ797)、BL-5-1(SNP1268-HLJ423)、BL-7-1(HLJ870-SNP0702)、BL-12-1(SNP0922-HLJ639)、BL-16-1(HLJE351-SNP0674)、BL-25-1(SNP0394-SNP0862)、BL-35-1(HLJ668-SNP0832)、BL-43-1(SNP0389-SNP1425)、BL-47-1(HLJ057-HLJ1113)、BL-47-2(HLJ1439-HLJ1418)等11个连锁群上,解释表型变异范围是13.8%~64.9%,其中贡献率大于20%的主效QTLs有8个,是体长性状的主效QTLs区间。  相似文献

鲤饲料转化率性状的QTL 定位及遗传效应分析   总被引:1,自引:0,他引:1  
数量性状(QTL)定位是实现分子标记辅助育种、基因选择和定位、培育新品种及加快性状遗传研究进展的重要手段。饲料转化率是鲤鱼的重要经济性状和遗传改良的主要目标, 而通过QTL 定位获得与饲料转化率性状紧密连锁的分子标记以及相关基因是遗传育种的重要工具。研究利用SNP、SSR、EST-SSR 等分子标记构建鲤鱼(Cyprinus carpio L.)遗传连锁图谱并对重要经济性状进行QTL 定位。选用174 个SSR 标记、41 个EST-SSR 标记、345 个SNP 标记对德国镜鲤F2 代群体68 个个体进行基因型检测, 用JoinMap4.0 软件包构建鲤鱼遗传连锁图谱。再用MapQTL5.0 的区间作图法(Interval mapping, IM)和多QTL 区间定位法(MQMMapping, MQM)对饲料转化率性状进行QTL 区间检测, 通过置换实验(1000 次重复)确定连锁群显著性水平阈值。结果显示, 在对饲料转化率性状的多QTL 区间定位中, 共检测到15 个QTLs 区间, 分布在9 个连锁群上, 解释表型变异范围为17.70%—52.20%, 解释表型变异最大的QTLs 区间在第48 连锁群上, 为52.20%。HLJE314-SNP0919(LG25)区间标记覆盖的图距最小, 为0.164 cM; 最大的是HLJ1439-HLJ1438(LG39)区间,覆盖图距为24.922 cM。其中区间HLJ1439-HLJ1438、HLJ922 -SNP0711 解释表型变异均超过50.00%, 可能是影响饲料转化率性状的主效QTLs 区间。与饲料转化率相关的15 个QTLs 的加性效应方向并不一致, 有3个区间具有负向加性效应, 平均为?0.027; 12 个正向加性效应, 平均值为0.06。研究检测出的与鲤鱼饲料转化率性状相关的QTL 位点可为鲤鱼分子标记辅助育种和更进一步的QTL 精细定位打下基础。    相似文献

大白×梅山杂交组合肉质性状的数量性状位点定位分析   总被引:4,自引:0,他引:4  
为寻找影响猪肉质数量性状基因位点的染色体区域 ,以 3头英系大白公猪和 7头梅山母猪建立F2 资源家系。随机选留 14 7头F2 代个体 (1998年 81头 ,2 0 0 0年 66头 ) ,经检测均获得肉质性状表型数据。对资源家系内的所有个体位于染色体 1、2、3、4、6和 7上的 48个微卫星位点进行扩增。利用线性模型最小二乘法分别对各年度及两年综合后的肉质性状进行数量性状位点 (QTL)区间定位 ,利用置换法确定显著性阈值。研究结果表明 :在 2 0 0 0年群体中 ,猪 4号染色体 (SSC4)上定位了肌内脂肪QTL ,达到染色体极显著水平 (P <0 0 1)和基因组显著水平 (P <0 0 5) ,解释表型变异为 5 2 4% ,梅山猪具有增加肌内脂肪QTL ;两年度群体综合后 ,在上述 4号染色体同一区间 ,肌内脂肪QTL接近染色体显著水平 ;股二头肌pH值和半棘肌pH值QTL分别定位在SSC1和 3上 ;在 1998年和 2 0 0 0年群体中分别发现 1个和 3个达染色体显著水平 (P <0 0 5)的系水力QTL ;在 1998年群体中 ,肌肉含水量QTL位于SSC6;两年综合群体中 ,SSC2、6和 7上定位了肌肉含水量QTL ,达到染色体显著水平 ,含水量QTL均有印迹效应 ,梅山和大白猪各有增效基因  相似文献

利用苹果栽培品种‘红富士’和新疆野苹果优系‘红肉苹果’杂交的110个F1株系为作图群体,构建了苹果的分子遗传图谱,采用区间作图法对苹果9个叶片相关性状(叶片长度、叶片宽度、叶片厚度、叶柄长度、叶片面积、总叶绿素含量、叶绿素a含量、叶绿素b含量和类胡萝卜素含量)进行了QTL定位分析。结果显示:从110个F1株系中共检测到20个控制叶片相关性状的QTL位点,分布在第1、2、3、4、5、7、8、10、11、12、16、17连锁群上;各QTL位点的LOD值在2.58~3.55之间,其中主效QTL位点2个(LOD≥3.5),可解释11.63%~16.36%的表型变异。获得紧密连锁的特异标记(CH05d11-435m、CH04c06-201m)为进一步进行QTL精细定位提供了参考。  相似文献

利用47个鲤鱼微卫星标记, 对柏氏鲤和荷包红鲤抗寒品系自交F2代的92个个体基因组DNA进行检测, 得到162个等位基因, 各座位等位基因数2~6个, 片段长度100~444 bp, 有效等位基因数1.3069~4.2288, 杂合度0.2361~0.7677, 位点多态信息含量0.5368。利用统计软件SPSS的GLM程序对47个微卫星标记与鲤鱼体重、体长和体高相关性进行了分析, 结果表明HLJ695、HLJ716、HLJ739、HLJ759、HLJ774、K16与体重、体长和体高相关, HLJ776与体高相关。对同一标记不同基因型间进行多重比较, 找到与3种性状相关的基因型。  相似文献

自20世纪60年代农林10号矮秆基因被用于小麦育种以来,矮化育种成为世界范围内势不可挡的趋势,矮秆基因研究被越来越多的育种专家重视,先后鉴定出20余个矮秆基因,并应用其中6,7个基因,培育了大批丰产潜力大的半矮秆品种,应用矮秆冬小麦吕系DN3338(♀)和F390(♂)杂交得到的F2:3群体,研究小麦株高的遗传基础,以控制株高的数量性状基因座进行定位,利用240个F2:3家系,构建了含215个微卫星标记,覆盖3600cM,由21个连锁群组成的遗传 连锁图谱,并对该群体进行了4个环境(2年:2000年和2001年,2点:北京和石家庄)3重复的田间种植;采用区间作图法,对该群体的株高性状进行了QTL分析。结果表明:7个影响株高的QTL分别位于染色体1B,4B(2个),6A(2个),6D和7A上,每个QTL能解释5.2%-50.1%的表型变异,每个环境条件下检测出的所有QTL能解释64.8%-75%,的表型变异,除了7A上的QTL外,其他6个降低株高的QTL均来自ND3338,其效应介于0.94cm-9.33cm之间,且其中的4个在所有的环境下都能被检测出来,具有较高的稳定性,在4BS的Xgwm113标记附近有一主效QTL,其在不同的环境下能降低株高7.91cm-9.33cm,解释27.8%-36.2%,的表型变异,有着同农林10号中Rht-Blb相近的效应;同时在4BS上还发现一个和地点互作的QTL,该QTL在石家庄的两年试验中均被检测到,且有较大的效应值(80cm和7.6cm),因此,认为大部分的QTL能在所有的环境中检测到,这些QTO可以被用于品种改良和分子标记辅助选择育种。  相似文献

大豆耐旱种质鉴定和相关根系性状的遗传与QTL定位   总被引:24,自引:0,他引:24  
从301份黄淮海和长江中下游地区代表性大豆地方品种和育成品种(系)中按根系类型选取59份,在苗期干旱胁迫和非胁迫条件下对地上部和地下部性状进行2年重复鉴定,发现材料间性状隶属函数值具有丰富遗传变异,以株高、叶龄、根干重和茎叶干重隶属函数的算术平均数为抗旱综合指标从中筛选出汉中八月黄、晋豆14,科丰1号,圆黑豆等强耐旱型(1级)和临河大粉青、宁海晚黄豆等干旱敏感型(5级)材料。比根干重、比总根长、比根体积与耐旱隶属函数平均值均呈极显著正相关,可作为耐旱性的根系性状指标。利用“科丰1号×南农1138 2”(1级×4级)衍生的RIL群体为材料,对耐旱相关根系性状采用主基因 多基因混合遗传模型分离分析法进行遗传分析并进行QTL定位。结果表明,该两亲本间比根干重、比总根长、比根体积的遗传均为两对主基因加多基因模型,后两者主基因间有连锁(重组率分别为4.30%和1.93%);主基因遗传率为62.26%~91.81%,多基因遗传率为2.99%~24.75%;耐旱相关根系性状各主要由1对主基因控制,另1对效应较小。QTL分析检测到5、3、5个QTLs分别控制比根重、比根总长、比根体积,位于N6 C2、N8 D1b W、N11 E、N18 K连锁群上。3性状各有1个贡献率大的QTL(Dw1,Rl1,Rv1),而且均位在N6 C2的STAS8_3T STAS8_6T相同距离的区段上,其他QTLs效应均较小。分离分析与QTL定位的结果相对一致。  相似文献

利用GH06×P174组合衍生的183个重组自交系进行种子发芽率遗传分析及种子发芽期间种子生理性状分析。用复合区间作图法对在室温下保存两年的种子(STY)、保存1年的种子(SOY)与新收获种子(FS)发芽率进行QTL定位。另外对保存两年的种子及新收获的种子萌发期间脂肪酶活性、电导率、还原糖含量、总糖含量及根系活力进行了测定, 并对结果进行分析。结果表明3批种子的发芽率QTL位点各不相同, 保存两年的种子在第9、14、17条连锁群上分别检测到1个位点, 保存1年的种子在第5、9条连锁群上各检测到1个位点, 新收获的种子在第4、18条连锁群上分别测检到1个位点。研究发现, 3批种子的发芽率相关性不显著, 发芽率差异达到极显著水平, 同时保存不同年份种子的发芽率QTL各不相同, 这表明甘蓝型油菜发芽率受很多不同因素所控制。保存两年及新收获种子的发芽率与电导率之间的相关性均达到极显著负相关, 表明可以通过电导率的测定估测发芽率, 电导率的研究对种子发芽率的研究具有重要意义。  相似文献

Microsatellite markers in common carp (Cyprinus carpio L.)   总被引:41,自引:0,他引:41  
Microsatellite markers of the poly (CA) type in common carp ( Cyprinus carpio L.) are described. Clones containing a (CA) repeat were isolated from a common carp genomic library and sequenced. The number of repeats found was high compared to mammals but comparable with other teleost fishes. Classification of the repeats (perfect, imperfect and compound) are compared with the Atlantic cod ( Gadus morhua L.), rainbow trout ( Oncorhynchus mykiss ), and Atlantic salmon ( Salmo salar L.). A total of 41 primer sets were designed and tested for polymorphism on a test panel of eight animals (derived from outbred lines, inbred lines and gynogenetic clones). Thirty-two markers were found to be polymorphic. The heterozygosity in the outbred animals was 60·4%, 51·1% in the inbred animals and 0% in the gynogenetic clones. The average number of alleles among the eight animals was 4·7 per marker. Six markers (18·8%) gave an additional polymorphic amplification product besides the polymorphic amplification product in the expected size range. The possibility that these loci are tetraploid is discussed. The polymorphic loci described for common carp will be valuable as genetic markers for use in population, breeding, and evolutionary studies.  相似文献

Ten tri- and tetranucleotide microsatellite DNA markers were isolated and characterized from common carp (Cyprinus carpio L.) to estimate genetic potential. These markers were tested in the samples from two closely related carp populations (Cyprinus carpio var. xingguonensis and Cyprinus carpio var. wananensis). The number of the alleles ranged from three to nine, and observed and expected hererozygosities varied from 0.207 to 1.000 and from 0.499 to 0.900 in each population, respectively. No evidence for linkage disequilibrium was found, indicating that these markers will be useful for population studies.  相似文献

To identify quantitative trait loci (QTL) that affect body shape in common carp Cyprinus carpio, a linkage map, 2159·23 cM long, was constructed with a total of 307 markers covering 51 linkage groups (LG). The map included 167 new single nucleotide polymorphism (SNP) markers derived from expressed sequence tags (EST) together with 140 microsatellite markers reported earlier. A primary genome scan was conducted for QTL for standard length (LS), head length (LH), body height (HB), body width (WB) and tail length (LTAIL) in an F1 line containing 92 offspring. A total of 15 suggestive QTL on six LGs were found to associate with LS, LH, HB, WB and LTAIL which explained 10·7–17·4% of the variance. Five significant QTL were detected for body‐shape related traits and located for LGs (lg1, 12 and 20). These QTL included: one associated with LS (21·1% variance explained), three for HB (almost 20% variance explained) and one for WB (20·7% variance explained).  相似文献

以镜鲤全同胞家系为材料,用246个SSR和306个SNP标记构建了鲤鱼的连锁图谱,利用GridQTL软件对体长(SL)、体高(H)、体厚(BT)和体长/体高(SLH)进行了QTL定位分析。结果显示:共检测到14个相关的QTL,分布在7个连锁群上。其中,7个与体长相关的QTL——LG6、LG17、LG21、LG23和LG35连锁群上的QTL为显著水平(P<0.05),LG1和LG28上达到极显著水平(P<0.01),可解释表型变异为6.6%~12.6%;3个与体高相关的QTL均为极显著水平(P<0.01)位于LG17、LG23和LG28上,可解释表型变异分别为11.6%、12.7%和15.6%;2个与体厚相关的QTL均为显著水平(P<0.05)位于LG23和LG28上,可解释表型变异分别为8.6%和7.2%;2个与体长/体高相关的QTL均为显著水平(P<0.05)位于LG21和LG35上,可解释表型变异均为8.2%。  相似文献

Genome evolution arises from two main ways of duplication and reduction. Fish specific genome duplication (FSGD) may have occurred before the radiation of the teleosts. Common carp (Cyprinus carpio L.) has been considered to be a tetraploid species, because of its chromosome numbers (2n=100) and its high DNA content. Using 69 microsatellite primer pairs, the variations were studied to better understand the genome evolution (genome duplication and diploidization) of common carp from a gynogenetic family. About 48% of primer pairs were estimated to amplify duplicates based on the number of PCR amplification per individual. Segregation patterns in the family suggested a partially duplicated genome structure and disomic inheritance. This indicates that the common carp is tetraploid and polyploidy occurred by allotetraploidy. Two primer pairs (HLJ021 and HLJ332) were estimated to amplify reduction based on the number of PCR amplification per individual. One allele in HLJ002 locus and HLJ332 locus was clearly lost in the gynogenetic family and the same as in six wild populations. Segregation patterns in the family suggested a partially diplodization genome structure. A hypothesis transition (dynamic) and equilibrium (static) were proposed to explain the common carp genome evolution between genome duplication and diploidization.  相似文献

Mapping quantitative trait loci regulating chicken body composition traits   总被引:1,自引:0,他引:1  
Genome scans were conducted on an F2 resource population derived from intercross of the White Plymouth Rock with the Silkies Fowl to detect QTL affecting chicken body composition traits. The population was genotyped with 129 microsatellite markers and phenotyped for 12 body composition traits on 238 F2 individuals from 15 full-sib families. In total, 21 genome-wide QTL were found to be responsible for 11 traits, including two newly studied traits of proventriculus weight and shank girth. Three QTL were genome-wide significant: at 499 c m on GGA1 (explained 3.6% of phenotypic variance, P  < 0.01) and 51 c m on GGA5 (explained 3.3% of phenotypic variance, P  < 0.05) for the shank & claw weight and 502 c m on GGA1 (explained 1.4% of phenotypic variance, P  < 0.05) for wing weight. The QTL on GGA1 seemed to have pleiotropic effects, also affecting gizzard weight at 490 c m , shank girth at 489 c m and intestine length at 481 c m . It is suggested that further efforts be made to understand the possible pleiotropic effects of the QTL on GGA1 and that on GGA5 for two shank-related traits.  相似文献

Body size traits reflect the condition of body development, are always mentioned when a breed is described, and are also targets in breeding programmes. In chicken, there are several reports focused on body size traits, such as shank length, tibia length or bone traits. However, no study was carried out on chest width (CW), chest depth (CD), body slope length (BL) and head width (HW) traits. In this study, genome scans were conducted on an F2 resource population (238 F2 individuals from 15 full‐sib families derived from an intercross of the White Plymouth Rock with the Silkies Fowl) to identify quantitative trait loci (QTL) associated with CW, CD, BL and HW from 7 to 12 weeks of age. In total, 21 significant or suggestive QTL were found that affected four body size traits. Four QTL reached 1% genome‐wide significance level: at 297 cM on GGA3 (associated with CW at 9 weeks of age), between 155 and 184 cM on GGA1 (affecting BL traits at 9 and 10 weeks of age), at 22 cM on GGA2 (related with BL traits at 12 weeks of age) and at 36 cM on GGA1 (for HW trait at 8 weeks of age).  相似文献

The integration pattern and adjacent host sequences of the inserted pMThGH-transgene in the F4 hGH-transgenic common carp were extensively studied. Here we show that each F4 transgenic fish contained about 200 copies of the pMThGH-transgene and the transgenes were integrated into the host genome generally with concatemers in a head-totail arrangement at 4-5 insertion sites. By using a method of plasmid rescue, four hundred copies of transgenes from two individuals of F4 transgenic fish, A and B, were recovered and clarified into 6 classes. All classes of recovered transgenes contained either complete or partial pMThGH sequences. The class I, which comprised 83% and 84.5% respectively of the recovered transgene copies from fish A and B, had maintained the original configuration, indicating that most transgenes were faithfully inherited during the four generations of reproduction. The other five classes were different from the original configuration in both molecular weight and restriction map, indicating that a few transgenes had undergone mutation, rearrangement or deletion during integration and germline transmission. In the five types of aberrant transgenes, three flanking sequences of the host genome were analyzed. These sequences were common carp β-actin gene, common carp DNA sequences homologous to mouse phosphoglycerate kinase-1 and human epidermal keratin 14, respectively.  相似文献

Groups of large (65 -75 g) and small (8-17 g) common carp ( Cyprinus carpio L.) fingerlings were exposed to the bacterium Chromobacterium violaceurn in order to establish whether they could detect and ingest unattached bacteria. Small fish exposed to both bacteria and to cell-free bacterial extracts showed a significant increase in opercular beat rates, thus demonstrating that they are able to detect the presence of unattached bacteria in suspension. Examination of carp gut contents showed that the proportion of small fish ingesting bacteria increased with exposure time although no significant relationship was observed among larger fish. Significant, positive correlations between numbers of viable bacteria isolated from the intestinal tracts and concentration in the environment were observed. Possible mechanisms of bacterial ingestion are discussed.  相似文献
